Em condições de solução nutritiva foram estudados os seguintes aspectos da nutrição mineral da cv. de arroz de sequeiro L-45, obtida pelo Dr. A. Ando (ESALQ-USP e CENA-USP, Piracicaba, SP, Brasil) através de mutação induzida; acumulação de matéria seca e de nutrientes durante ociclo; exigências nutricionais. Verificou-se que: a velocidade máxima de absorção dos elementos minerais ocorreu entre 64 e 106 dias depois da germinação (perfilhamento e maturação). A exigência nutricional obedeceu à seguinte ordem decrescente: N, K, Ca, P, Mg e S; Mn, Fe, Zn, Cu e B. A exportação como produto colhido obedeceu à ordem, também decrescente: N, K,P, Mg, Ca, S; Mn, Zn, Fe, Cu e B.

O arroz de sequeiro, cv. L-45, foi cultivado em solução nutritiva contendo todos os macro e micronutrientes e com omissão dos primeiros, um de cada vez. A falta dos elementos no substrato afetou o crescimento na seguinte ordem crescente: nitrogênio, fósforo e magnésio. Não se notou influência da omissão dos demais na produção de matéria seca, embora tivesse havido diminuição no teor do elemento na planta quando o mesmo foi fornecido em concentração menor na solução. Verificou-se através da diagnose foliar redução no teor de N, P e K quando os mesmos se achavam deficientes. Resultado análogo foi obtido ao se fazer teste rápido (spot teste) para nitrato, fosfato e potásio solúvel no tecido foliar. A atividade da redutase de nitrato foliar diminuiu como conseqüência de todos os tratamentos, exceto o menos Ca, ca so em que aumentou significativamente.

Foram observadas diferenças significativas na absorção do P32 em função do tempo de contacto para as cultivares L-45, Dourado Precoce e Penatie. Exceção feita no caso da L- 45 as plantas inteiras absorvem mais que as raízes destacadas. 0 mecanismo duplo de absorção atuou no caso das três variedades, uma na faixa de 1-5 umoles de H2PO-4 e outro na de 10² Cn a 10(4) umoles. A presença de uréia na solução aumenta a absorção do P fornecido por via foliar. Quando se empregou fosfato diamônico como fonte de fósforo houve maior transporte do elemento, em presença de uréia, da folha tratada para os outros órgãos da planta. 0 transporte foi maior no caso da cv. Penatie, sendo menor no caso da cv. L-45 e muito pequeno na variedade Dourado Precoce.

BACKGROUND/AIMS: Ligand activation of the mineralocorticoid receptor (MR) induces several post-translational modifications (PTMs). Among the different PTMs, MR is known to be dynamically ubiquitylated with impact on its stability and transcriptional activity. Previously, we have shown that MR is monoubiquitylated at the basal state and that aldosterone stimulation induces monoubiquitylation removal prompting polyubiquitin-dependent destabilization of the receptor and proteasomal degradation. This study investigated the role of the aldosterone induced ubiquitin-specific protease USP2-45 on the ubiquitylation state of MR. METHODS: Renal epithelial cells M1 were co-transfected with MR with or without wild-type or inactive USP2-45. The association of MR with USP2-45 or TSG101 as well as MR ubiquitylation state were determined by immunoprecipitation and immunoblotting. MR transcriptional activity was assessed via a luciferase reporter gene. RESULTS: We show that USP2-45 is able to bind MR and, similarly to aldosterone, induce MR monoubiquitylation removal, disruption of MR/TSG101 association and destabilization of MR at protein level. CONCLUSION: This study provides a novel role for USP2-45 by playing a pivotal role in the regulation of the ubiquitylation state of MR and reveals the existence of a negative feedback loop for limiting the aldosterone induced response.

Monoclonal antibodies (mAbs) and human sera from gametocyte carriers were applied in the bio-assay to test for their transmission-blocking capacity. Competition ELISA's have been developed for the detection of natural transmission blocking antibodies. Approximately 55 of the sera blocking in the bio-assay gave positive results in these competition ELISA's.

Circulating levels of adiponectin, a hormone produced predominantly by adipocytes, are highly heritable and are inversely associated with type 2 diabetes mellitus (T2D) and other metabolic traits. We conducted a meta-analysis of genome-wide association studies in 39,883 individuals of European ancestry to identify genes associated with metabolic disease. We identified 8 novel loci associated with adiponectin levels and confirmed 2 previously reported loci (P = 4.5×10(-8)-1.2×10(-43)). Using a novel method to combine data across ethnicities (N = 4,232 African Americans, N = 1,776 Asians, and N = 29,347 Europeans), we identified two additional novel loci. Expression analyses of 436 human adipocyte samples revealed that mRNA levels of 18 genes at candidate regions were associated with adiponectin concentrations after accounting for multiple testing (p<3×10(-4)). We next developed a multi-SNP genotypic risk score to test the association of adiponectin decreasing risk alleles on metabolic traits and diseases using consortia-level meta-analytic data. This risk score was associated with increased risk of T2D (p = 4.3×10(-3), n = 22,044), increased triglycerides (p = 2.6×10(-14), n = 93,440), increased waist-to-hip ratio (p = 1.8×10(-5), n = 77,167), increased glucose two hours post oral glucose tolerance testing (p = 4.4×10(-3), n = 15,234), increased fasting insulin (p = 0.015, n = 48,238), but with lower in HDL-cholesterol concentrations (p = 4.5×10(-13), n = 96,748) and decreased BMI (p = 1.4×10(-4), n = 121,335). These findings identify novel genetic determinants of adiponectin levels, which, taken together, influence risk of T2D and markers of insulin resistance.

Over the past 50 years organ transplantation has become an established worldwide practice, bringing immense benefits to hundreds of thousands of patients. The use of human organs (hereinafter â?~organsâ?T) for transplantation has steadily increased during the last two decades. Organ transplantation is now the most cost-effective treatment for end-stage renal failure, while for end-stage failure of organs such as the liver, lung and heart it is the only available treatment. This report describes a surface molecule, Tp45, which appears to be involved in interleukin 2 production and Ca2+ mobilization by Jurkat cells. The Tp45 molecule was identified by a monoclonal antibody, MX13, on the surface of either T3/TCR+ or T3/TCR- human T cell lines. Biochemical data showed that mAb MX13 precipitated a single polypeptide chain of 45 kDa both under reduced and nonreduced conditions from lysates of 125I-surface-labeled cells. Sequential immunodepletion experiments using lysates of 125I-labeled T3/TCR+ cells showed that Tp45 was distinct from the alpha chain of the TCR complex. However, incubation of such cells with either anti-T3 or anti-TCR monoclonal antibody induced complete modulation of both the T3/TCR complex and Tp45. Conversely, complete modulation of both Tp45 and the T3/TCR complex was observed after incubation with anti-Tp45 antibody. Functional studies showed that anti-Tp45 antibody induced high levels of interleukin 2 production in Jurkat cells. In addition, incubation of these cells with the antibody resulted in Ca2+ mobilization from internal stores. Anti-Tp45 antibody reacted with 3-19% peripheral blood (E-rosette-positive) T cells in individual donors. The magnitude of the proliferative response elicited by anti-Tp45 antibody for peripheral blood T cells was lower than that induced by an anti-T3 antibody. This observation is compatible with the idea that only a subpopulation of T cells is reactive with anti-Tp45. Multicolor flow cytometry analysis showed that the Tp45+ cells belong preferentially to the T8 subset.
