O presente trabalho surgiu da necessidade de compreender de que modo a inclusão de atividades de natureza investigativa (atividades de investigação ou atividades exploratórias) contribui para a aprendizagem matemática de alunos do secundário. Com este propósito foram formuladas as seguintes questões: Como age o aluno perante este tipo de atividade? Que tipo de conhecimentos mobiliza o aluno nestas atividades? e Que benefícios (para o aluno) são alcançados com este tipo de tarefa? O estudo envolveu os alunos de uma turma do décimo ano e três atividades que foram desenvolvidas ao longo do ano letivo. Estas atividades abrangem duas grandes áreas da Matemática: a geometria e as funções. Tendo em conta que os dados resultantes da aplicação das tarefas eram essencialmente constituídos por pormenores descritivos e de difícil tratamento estatístico, adotei para a sua análise uma abordagem de tipo qualitativo. Embora as atividades aqui apresentadas e desenvolvidas na sala de aula, tenham uma estrutura aberta, o seu grau de complexidade não é muito elevado, pelo que as considerei atividades de exploração. A última tarefa proposta é no entanto menos estruturada do que as duas primeiras, tendo constatado que nesta última alguns alunos alargaram as suas reflexões, o que lhes permitiu aprofundar a investigação. Os diversos materiais utilizados nas atividades tiveram um papel muito importante no desenvolvimento das mesmas e no surgimento de alguns processos matemáticos. Foi também constatado uma melhoria na autonomia dos alunos, à medida que se sucediam as tarefas e ainda a ocorrência de ligações entre diversos temas da Matemática de uma forma coerente e integrada.

A presente dissertação do mestrado em Ciências da Educação - Inovação Pedagógica, tem como objetivo central perceber como o funcionamento do clube escolar Eco-Projeto da Escola Básica e Secundária Gonçalves Zarco pode promover inovação pedagógica, constituindo um indício de quebra do paradigma educativo vigente. Pretende-se assim, compreender em que medida as práticas pedagógicas desenvolvidas neste projeto educativo específico, são práticas construtivistas, centradas no aluno, no desenvolvimento da autonomia, da criatividade, da negociação de conhecimentos entre pares e do desenvolvimento cognitivo e sociocultural dos alunos. O desenvolvimento desta investigação tem como metodologia a investigação qualitativa, baseada na etnografia e na observação participante. O Eco-Projeto, sendo um clube escolar integrado nas atividades extracurriculares, promove uma efetiva aprendizagem cooperativa e significativa, baseada nos pressupostos inculcados pela Educação Ambiental e Desenvolvimento Sustentável defendida pelo programa Eco-Escolas. O programa Eco-Escolas possui uma abrangência internacional, sendo que em Portugal este surge sob a tutela da Associação Bandeira Azul da Europa/Foundation for Environmental Education. Em suma, ao longo desta dissertação é descrita etnograficamente a cultura, o modus vivendi do Eco-Projeto, a forma como os seus membros interagem, solucionam os problemas com que se deparam no dia-a-dia, e como são proporcionadas as aprendizagens, de modo a se verificar um efetivo extrapolar das fronteiras do ensino instrucionista, abrindo um caminho para uma visão inovadora da forma como pode ser encarado o processo de aprendizagem, e a escola na sua globalidade.

A matemática, infelizmente e de uma forma errada, ainda é encarada por muitos como uma ciência que é acessível apenas para alguns “iluminados”. Segundo a frase célebre de Paulo Freire, “Não há saber mais ou saber menos há saberes diferentes” e é este tipo de filosofia que é necessário incutir nas nossas escolas e na sociedade. A meu ver, as atividades investigativas vão ao encontro do pensamento de Paulo Freire, uma vez que, neste tipo de atividades, deve ser dado enfoque aos diferentes percursos e saberes. A matemática deveria ser encarada como uma disciplina que educa para a vida – educação matemática. Neste sentido, ela deveria ser trabalhada em diferentes contextos para que os alunos alarguem o seu campo de visão e utilizem a matemática de uma forma crítica e em prol do seu bem-estar. O presente estudo visou compreender de que forma as atividades investigativas contribuem para a aprendizagem significativa da matemática e qual o seu contributo na formação de alunos autónomos e com competências para pensar e agir. A investigação incidiu sobre a prática matemática dos alunos quando realizavam atividades investigativas sobre a unidade temática das semelhanças, nomeadamente, sobre figuras semelhantes, razão de semelhança e critérios de semelhança de triângulos, onde foi utilizada uma metodologia qualitativa de natureza descritiva através da observação participante. O estudo foi efetuado numa turma do sétimo ano e teve por base três propostas de trabalho, sendo que a primeira foi mais de carácter introdutório. Nestas atividades, foram utilizados materiais manipuláveis e o Microsoft Excel no sentido de motivar os alunos e facilitar a compreensão dos conteúdos trabalhados. Os materiais utilizados despertaram um enorme interesse nos alunos, os quais começaram logo a explorá-los, manuseando-os livremente sem qualquer objetivo pré-definido, isto é, antes de se inteirarem do propósito da atividade. As atividades investigativas aliadas aos materiais manipuláveis são uma maisvalia na aprendizagem dos alunos uma vez que o conhecimento é construído de uma forma natural através de um processo espontâneo e dinâmico, consequentemente, significativo. Com a realização de atividades investigativas, são proporcionados aos alunos momentos de exploração e de investigação aleatórios, culminando numa aprendizagem de improviso, no sentido em que esta não obedece a regras pré-definidas e estimula e desenvolve o raciocínio dos alunos.

A presente dissertação - “A integração das TIC nas Atividades Curriculares sob o Olhar da Inovação Pedagógica” decorre de uma investigação realizada ao longo do ano letivo 2012/2013 numa turma de 4º ano de uma escola de 1º Ciclo do Ensino Básico na Região Autónoma da Madeira. Partindo da formulação da questão - Como é que a integração das TIC na curricular promove inovação pedagógica? – pretendeu-se estudar e compreender como é que acontece a integração curricular das Tecnologias de Informação e Comunicação (TIC) nas atividades curriculares e de que forma a prática pedagógica daí decorrente poderia constituir ou não inovação pedagógica. Num primeiro momento procedeu-se à revisão da literatura relacionada com o tema da investigação. Assim, o debate construiu-se não só em torno da temática da inovação pedagógica, linha orientadora do mestrado mas também da relação existente entre as TIC e a inovação pedagógica, pondo em evidência o modo como estes dois conceitos se cruzam nas práticas pedagógicas. No sentido de contribuir para um enquadramento teórico mais eficaz, foi feita uma breve abordagem à introdução das TIC na Educação, ao enquadramento das TIC nas escolas de 1º Ciclo da Região Autónoma da Madeira bem como a sua integração nas atividades curriculares. Tratando-se da observação de uma turma, a metodologia considerada mais eficaz foi o estudo de caso. Os dados foram recolhidos através da observação participante periférica, traduzida no diário de campo, bem como da análise documental. Assim, através desta investigação, de natureza qualitativa, foi possível observar, descrever e analisar um contexto específico, tornando-se claro que as práticas pedagógicas que aí ocorreram ainda se encontram muito distantes da inovação pedagógica. Sobressaíram, desta forma, práticas enraizadas numa cultura de inovação e tecnologia educativas, não se evidenciando, portanto, a rutura paradigmática preconizada pela inovação pedagógica.

Consciente do lugar de destaque e importância que a educação formal tem na nossa sociedade, propomo-nos a refletir sobre as aprendizagens que decorrem à margem dos saberes dominantes e que para alguns constituem a “fuga” da eterna insatisfação gerada pela rigidez do currículo. Nesta investigação, pretendemos compreender se existe alguma relação entre a frequência das atividades de complemento curricular (ACC) e o aproveitamento escolar dos alunos que as frequentam. Este estudo envolveu simultaneamente as abordagens qualitativa e quantitativa e teve lugar numa escola secundária pública da Região Autónoma da Madeira. Após uma análise dos documentos orientadores do estabelecimento de ensino - Projeto Educativo de Escola, Plano Anual de Atividades e Regulamento Interno - realizámos entrevistas a docentes responsáveis por estas atividades e solicitámos aos alunos o preenchimento de inquéritos por questionário. Constatámos que as ACC são valorizadas por todos os atores educativos, desde as entidades proponentes - Divisão de Gestão de Projetos da Direção Regional de Educação, órgão de gestão da escola, conselho pedagógico e coordenação de ACC até aos alunos que as frequentam e para quem são idealizadas. Foi possível concluir que as ACC são importantes no percurso escolar dos alunos por possibilitarem o desenvolvimento de competências transversais ao currículo, contribuindo desta forma para o processo holístico de educação em particular na sua dimensão pessoal e social. Verificámos que os alunos que frequentam as ACC são oriundos de diferentes cursos e que existe uma predominância dos rapazes na sua frequência.

O estudo incidiu sobre uma turma do décimo primeiro ano de escolaridade do curso de línguas e humanidades. Foram trabalhadas três atividades no âmbito do capítulo Modelos Populacionais. Estas foram desenvolvidas com o auxílio de tecnologia, nomeadamente, calculadora gráfica e computador através da exploração do software Graph 4.4.2, no sentido de motivar os alunos e facilitar a compreensão dos conteúdos abordados. Tendo em conta que um dos objetivos deste estudo é analisar as atitudes reveladas pelos alunos aquando da realização das propostas, os dados obtidos são constituídos essencialmente por pormenores descritivos de tal forma que foi aplicada uma metodologia de natureza qualitativa através da observação participante. The growing importance of tourism in overall economic activity worldwide has favored the intensification of competition among cities that seek to create environments attractive to tourists and potential investors. It has been common practice to import characteristics of the business environment in the public management of cities. The city marketing is a key tool used by public leaders to promote a linkage between the tourism image and urban image and involves, in addition to promoting the image of the city, the planning of interventions in urban space, trying to formulate a positive image of the city able to facilitate the deployment of capital. This research seeks to understand the nature of city marketing as part of contemporary urban management and analyzes how is its application in decisions concerning the promotion of tourism in Natal/RN. The approach of this research is qualitative, exploratory and descriptive, in which respondents were the main leaders of two of the official tourism site, the Empresa Potiguar de Promoção Turística and the Secretaria Municipal de Turismo e Desenvolvimento Econômico. It was found that there is a strong articulation of public power with private enterprise in the design and conduct of the actions of urban marketing, that from the survey data show that the behavior of target markets provide guidelines for taking strategic decisions relating to tourism. Sun and sea are some key elements explored to form the image of Natal and to authorize the sale of the city as a tropical paradise. However, there is an increase in the diversification of tourism products, seeking to increase flow to the segments of ecotourism, adventure, business and culture. It s also growing the use of local culture as a tourism product, however, the cultural representation focuses on superficial values and does not bring to light the social and historical richness that the city has. Public authorities use the city marketing strategies as a means able to maximize the attractiveness of Natal urban space to investors, business groups and tourists. It can be observed that urban managers seek solutions that can continuously increase the tours, which often manifests in interventions that focus the tourist areas of the city, in oposition of those who do not contribute to a positive reading of the city, which ultimately generate the worsening of spacial and social inequalities

Sulfated polysaccharides (PS) are biomolecules with a great biotechnological potential. There are few data about PS from high plants. In addition, pharmacological activities of PS from plants have not been carrying out. The aim of this work was extract PS from the angiosperm Halodule wrightii and study their anticoagulant and antioxidant activities. Histological analysis showed the presence of the PS manly in the roots. A polysaccharide-rich extract was obtained from H. wrightii by proteolysis followed by methanol and TCA precipitation. Chemical, infra-red analysis and agarose gel electrophoresis in 1.3 diaminopropane acetate buffer confirmed the presence of sulfated polysaccharides made by glucose, galactose, xylose and sulfate residues in the proportion 1: 0,9: 1: 1. In addition polyacrilamide electrophoresis have shown that extract is mainly compose by 11kDa sulfated polysaccharides. Pharmacological analysis have shown total antioxidant capacity (CAT) that resulted in 15,21 μg for equivalent of ascorbic acid, scavenging activity of the DPPH radical with 41,36 % of scavenging, activity of reducing power with the maximum of 0,290 nm (50 % of vitamin C activity) and scavenging activity superoxide radical (O2-) with a maximum of 32,23 %. Chelating activity of metal less than 4% and scavenging activity of the radical hydroxyl (OH-) less than 2%. Time of activated partial tromboplastin (aPTT) doubling the time of coagulation from 20μg of and protrombin time (PT) was not present. The data indicate that PS from Halodule wrightii could be considered for future applications in medicine, food production or cosmetic industry

In recent years, sulfated polysaccharides (SP) from marine algae have emerged as an important class of natural biopolymers with potential pharmacology applications. Among these, SP isolated from the cell walls of red algae have been study due to their anticoagulant,antithrombotic and anti-inflammatory activities. In the present study, three sulfated polysaccharides fractions denominated F1.5v, F2.0v and F3.0v were obtained from seaweed G. caudate by proteolysis followed to acetone fractionation. Gel electrophoresis using 0.05 M 1,3-diaminopropane-acetate buffer, pH 9,0, stained with 0.1% toluidine blue, showed the presence of SP in all fractions. The chemical analysis demonstrated that all the fractions are composed mainly of galactose. These compounds were evaluated in anticoagulant, antioxidant and antiproliferative activities. In anticoagulant activity evaluated through aPTT and PT tests, no one fractions presented anticoagulant activity at tested concentrations (0.1 mg/mL; 1.0 mg/mL; 2.0 mg/mL).The antioxidant activities of the three fractions were evaluated by the following in vitro systems: Total antioxidant capacity, superoxide and hydroxyl radical scavenging, ferrous chelating activity and reducing power. The fractions were found to have different levels of antioxidant activity in the systems tested. F1.5v shows the highest activity, especially in the ferrous chelating system, with 70% of ferrous inhibiting at 1.0 mg.mL-1. Finally, all the fractions showed dose-dependent antiproliferative activity against HeLa cells. The fractions F1.5v and F2.0v presented the highest antiproliferative activity at 2.0 mg/mL with 42.7% and 37.0% of inhibition, respectively. Ours results suggests that the sulfated polysaccharides from seaweed G. caudata are promising compounds in antioxidant and/or antitumor therapy

The exopolysaccharides are extracellular compounds produced by some species of fungi and bacteria. It is suggested that these molecules, even when in the form of complex polysaccharide-peptide, are the main bioactive molecules of many fungus. Some of the biological activities displayed by these compounds can be accentuated and others may arise when you add chemically polar or nonpolar groups to polysaccharides. The fruiting body of Pleurotus sajor-caju produces a heteropolysaccharide with antineoplastic and antimicrobial activity, but other biological activities of this polymer have not been evaluated. In this work the exopolysaccharide of Pleurotus sajor-caju was sulfated chemically and structurally characterized. We also evaluated the antiproliferative, antioxidant and anticoagulant activities from native exopolysaccharide (PN) and its sulfated derivated (PS). Polyacrylamide gel electrophoresis, infrared spectroscopy and nuclear magnetic resonance (¹³C) proved successful in sulfation of PN to obtain PS. Analysis by gas chromatography-mass spectroscopy showed that PN and PS are composed of mannose, galactose, 3-O-methyl-galactose and glucose in proportion percentage of 44,9:16,3:19,8:19 and 49, 7:14,4:17,7:18,2, respectively. The percentage of sulfate found in PS was 22.5%. Antioxidants assays revealed that the sulfation procedure affects differently the activities of exopolysaccharides, while the total antioxidant capacity, the scavenging activity of superoxide radical and ferric chelating were not affected by sulfation, on the other hand the chemical modification of PN enhanced the scavenging activity of hydroxyl radical and reducing power. PS also showed anticoagulant activity in a dose-dependent manner and clotting time was 3.0 times higher than the baseline value in APTT at 2 mg/mL. The exopolysaccharide not presented antiproliferative activity against HeLa tumor cells, but PS affects the cellular proliferation in a time-dependent manner. After 72 h, the inhibition rate of PS (2.0 mg/mL) on HeLa cells was about 60%. The results showed that PN sulfation increase some of their activities.

The corn cob is an agricultural by-product still little used, this in part due to the low knowledge of the biotechnological potential of their molecules. Xylan from corn cobs (XSM) is a polysaccharide present in greater quantity in the structure of plant and its biotechnology potential is little known. This study aimed to the extraction, chemical characterization and evaluation of biological activities of xylan from corn cobs. To this end, corncobs were cleaned, cut, dried and crushed, resulting in flour. This was subjected to a methodology that combines the use of alkaline conditions with waves of ultrasound. After methanol precipitation, centrifugation and drying was obtained a yield of 40% (g/g flour). Chemical analysis indicated a high percentage of polysaccharides in the sample (60%) and low contamination by protein (0.4%) and phenolic compounds (> 0.01%). Analysis of monosaccharide composition indicated the presence of xylose:glucose:arabinose:galactose:mannose:glucuronic acid in a molar ratio 50:20:15:10:2.5:2.5. The presence of xylan in the sample was confirmed by nuclear magnetic resonance (¹H and ¹³C) and infrared spectroscopy (IR). Tests were conducted to evaluate the antioxidant potential of XSM. This showed a total antioxidant capacity of 48.45 EAA/g sample. However, did not show scavenging activity of superoxide and hydroxyl radical and also reducing power. But, showing a high capacity chelating iron ions with 70% with about 2 mg/mL. The ability to XSM to influence cell proliferation in culture was also evaluated. This polymer did not influence the proliferation of normal fibroblast cells (3T3), however, decreased the rate of proliferation of tumor cells (HeLa) in a dose-dependent, reaching an inhibition of about 50% with a concentration around 2 mg/mL. Analyzing proteins related to cell death, by immunoblotting, XSM increases the amount of Bax, Bcl-2 decrease, increase cytochrome c and AIF, and reduce pro-caspase-3, indicating the induction of cell death induced apoptosis dependent and independent of caspase. XSM did not show anticoagulant activity in the PT test. However, the test of activated partial thromboplastin time (aPTT), XSM increased clotting time at about 5 times with 600 μg of sample compared with the negative control. The presence of sulfate on the XSM was discarded by agarose gel electrophoresis and IR. After carboxyl-reduction of XSM the anticoagulant activity decreased dramatically. The data of this study demonstrate that XSM has potential as antioxidant, antiproliferative and anticoagulant compound. Future studies to characterize these activities of XSM will help to increase knowledge about this molecule extracted from corn and allow their use in functional foods, pharmaceuticals and chemical industries.

Seaweeds sulfated polysaccharides have been described as having various pharmacological activities. However, nothing is known about the influence of salinity on the structure of sulfated polysaccharides from green seaweed and pharmacological activities they perform. Therefore, the main aim of this study was to evaluate the effect of salinity of seawater on yield and composition of polysaccharides-rich fractions from green seaweed Caulerpa cupressoides var. flabellata, collected in two different salinities beaches of the coast of Rio Grande do Norte, and to verify the influence of salinity on their biological activities. We extracted four sulfated polysaccharides-rich fractions from C. cupressoides collected in Camapum beach (denominated CCM F0.3; F0.5; F1.0; F2.0), which the seawater has higher salinity, and Buzios beach (denominated CCB F0.3; F0.5; F1.0; F2.0). Different from that observed for other seaweeds, the proximate composition of C. cupressoides did not change with increased salinity. Moreover, interestingly, the C. cupresoides have high amounts of protein, greater even than other edible seaweeds. There was no significant difference (p>0.05) between the yield of polysaccharide fractions of CCM and its CCB counterparts, which indicates that salinity does not interfere with the yield of polysaccharide fractions. However, there was a significant difference in the sulfate/sugar ratio of F0.3 (p<0.05) and F0.5 (p<0.01) (CCM F0.3 and CCB F0.5 was higher than those determined for their counterparts), while the sulfate/sugar ratio the F1.0 and F2.0 did not change significantly (p>0.05) with salinity. This result suggested that the observed difference in the sulfate/sugar ratio between the fractions from CCM and CCB, is not merely a function of salinity, but probably also is related to the biological function of these biopolymers in seaweed. In addition, the salinity variation between collection sites did not influence algal monosaccharide composition, eletrophoretic mobility or the infrared spectrum of polysaccharides, demonstrating that the salinity does not change the composition of sulfated polysaccharides of C. cupressoides. There were differences in antioxidant and anticoagulant fractions between CCM and CCB. CCB F0.3 (more sulfated) had higher total antioxidant capacity that CCM F0.3, since the chelating ability the CCM F0.5 was more potent than CCB F0.5 (more sulfated). These data indicate that the activities of sulfated polysaccharides from CCM and CCB depend on the spatial patterns of sulfate groups and that it is unlikely to be merely a charge density effect. C. cupressoides polysaccharides also exhibited anticoagulant activity in the intrinsic (aPTT test) and extrinsic pathway (PT test). CCB F1.0 and CCM F1.0 showed different (p<0,001) aPTT activity, although F0.3 and F0.5 showed no difference (p>0,05) between CCM and CCB, corroborating the fact that the sulfate/sugar ratio is not a determining factor for biological activity, but rather for sulfate distribution along the sugar chain. Moreover, F0.3 and F0.5 activity in aPTT test was similar to that of clexane®, anticoagulant drug. In addition, F0.5 showed PT activity. These results suggest that salinity may have created subtle differences in the structure of sulfated polysaccharides, such as the distribution of sulfate groups, which would cause differences in biological activities between the fractions of the CCM and the CCB

Fungal polysaccharides have received a great deal of attention due to itsbecause of their potential use in a wide rangegreat variety fromof industries. Some studies have demonstrated that polysaccharides extracted offrom basidiomycetes they have presented significant properties as anti-inflammatory, antimicrobial, antioxidant and anti-tumoral properties. In spite of thisDespite these potential properties, these mushrooms have not been insufficiently investigated, and the great number of antibiotics number produced forby these organisms suggests that they canmay be a new source of bioactives composites source. In tThe present work, reports onlated the chemical composition, potential antioxidant, antiinflammatory and citotoxycity of extracted polymers extracted offrom the fruits bodies of the fungiius Geastrum saccatum and Polyporus dermoporus, native mushrooms of the Atlantic forest inof the state of the Rio Grande do Norte, Brazil. The Cchemical analyses had revealed ademonstrated text of total sugar rates of 65% and 49%, and proteins of 7.0% for in extracts of G. saccatum and P. dermoporus extracts, respectively. The analyses ofNMR spectroscopy of RMN had demonstrated that these extracts are composites forof a complex involving β- glucans and- proteins complex. The inhibition of the formation of superoxide radicals formation was of 88.4% in G. saccatum and 83.3% in P. dermoporus, and 75 and 100% for inhibition of hydroxyls radicals inhibition.
