Heterotrimeric G proteinshave been previously linked to plant defense; however a role for the G beta gamma dimer in defense signaling has not been described to date. Using available Arabidopsis (Arabidopsis thaliana) mutants lacking functional G alpha or G beta subunits, we show that defense against the necrotrophic pathogens Alternaria brassicicola and Fusarium oxysporum is impaired in G beta- deficient mutants while G alpha-deficient mutants show slightly increased resistance compared to wild-type Columbia ecotype plants. In contrast, responses to virulent (DC3000) and avirulent (JL1065) strains of Pseudomonas syringae appear to be independent of heterotrimeric G proteins. The induction of a number of defense-related genes in G beta-deficient mutants were severely reduced in response to A. brassicicola infection. In addition, G beta-deficient mutants exhibit decreased sensitivity to a number of methyl jasmonate- induced responses such as induction of the plant defensin gene PDF1.2, inhibition of root elongation, seed germination, and growth of plants in sublethal concentrations of methyl jasmonate. In all cases, the behavior of the G alpha- deficient mutants is coherent with the classic heterotrimeric mechanism of action, indicating that jasmonic acid signaling is influenced by the Gbg functional subunit but not by G alpha. We hypothesize that G beta gamma acts as a direct or indirect enhancer of the jasmonate signaling pathway in plants.

Reforestation in tropical areas is usually attempted by planting seedlings but, direct seeding (the artificial addition or sowing of seed) may be an alternative way of accelerating forest recovery and successional processes. This study investigated the effects of various sowing treatments (designed to create different microsite conditions for seed germination) and seed sizes on the early establishment and growth of directly sown rainforest tree species in a variety of experimental plots at three sites in the wet tropical region of north-cast Queensland, Australia. The different sowing treatments were found to have significant effects on seedling establishment. Broadcast sowing treatments were ineffective and resulted in very poor seedling establishment and high seed wastage. Higher establishment rates occurred when seeds were buried. Seed size was found to be an important factor affecting establishment in relation to micro-site condition. In general, larger seeded species had higher establishment rates at all three sites than species of small and intermediate seed size, but only in sowing treatments where seeds were buried. Overall these results suggest that direct sowing of seed can be used as a too] to accelerate recolonisation of certain rainforest tree species on degraded tropical lands, but initial success will be dependent on the choice of sowing method and its suitability for the seed types selected. The results also indicate that the recruitment of naturally dispersed tree species at degraded sites is likely to be severely limited by the availability of suitable microsites for seed germination. Consequently the natural recovery of degraded sites via seed rain can be expected to be slow and unpredictable, particularly in areas where soil compaction has occurred. (c) 2006 Elsevier B.V. Lepidocaryum tenue Mart. (Arecaceae) is a small, understory palm of terra firme forests of the western and central Amazon basin. Known as irapai, it is used for roof thatch by Amazonian peoples who collect its leaves from the wild and generate income from its fronds and articles fabricated from them. Increasing demand has caused local concern that populations are declining. Cultivation attempts have been unsuccessful. The purpose of this study was to investigate market conditions and quantify population dynamics and demographic responses of harvested and unharvested irapai growing near Iquitos, Peru. ^ Ethnobotanical research included participant surveys to determine movement of thatch tiles, called crisnejas, through Moronacocha Port. I also conducted a seed germination trial, and for four years studied five populations growing in communities with similar topography and soils but different land tenure and management strategies. Stage, survival, leaf production, and reproductive transitions were used to calculate ramet demographic rates and develop population projection matrices. ^ Weavers made an average of 20–30 crisnejas per day (90–130 leaves each), and earned US$0.09 to 0.70 each (US$1.80 to 21.00 per day). Average crisnejas per month sold per vendor was 2,955 with a profit range of US$0.05 to 0.32 per crisneja. Wholesalers worked with capital outlay from US$100 to 400, and an estimated ten to twenty vendors could be found at a given time. Consumers paid between US$0.23 to 1.20 per crisneja. Although differences in demographic rates by location existed, most were not significant enough to attribute to management. ^ After 60 months, mean seed germination rate was 19.5% in all media (37.9% in peat). Seedling survival was less than two percent after twelve months. Annual palm mortality was three percent, and occurred disproportionately in small (<50 cm) palms. Small palms grew more in height. Unharvested palms grew less than harvested palms. Large palms (≥50 cm) produced more leaves, were more likely to reproduce, and collectors harvested them more frequently. Reproductive potentials (sexual and asexual) were low. Population growth rates were greater than or not significantly different from 1.0, indicating populations maintained or increased in size. Current levels of irapai harvest appear sustainable. DNA analysis of stems and recruits is recommended to understand population composition and stage-specific asexual fecundity. ^

Morphological, anatomical and physiological plant and leaf traits of A. distorta, an endemic species of the Central Apennines on the Majella Massif, growing at 2,675 m a.s.l, were analyzed. The length of the phenological cycle starts immediately after the snowmelt at the end of May, lasting 128 ± 10 days. The low A. distorta height  (Hmax= 64 ± 4 mm) and total leaf area (TLA= 38 ± 9 cm2) associated to a high leaf mass area (LMA =11.8±0.6 mg cm−2) and a relatively high leaf tissue density (LTD = 124.6±14.3 mg cm−3) seem to be adaptive traits to the stress factors of the environment where it grows. From a physiological point of view, the high A. distorta photosynthetic rates (PN =19.6 ± 2.3 µmol m−2 s−1) and total chlorophyll content (Chla+b = 0.88 ± 0.13 mg g−1) in July are justified by the favorable temperature. PN decreases by 87% in September at the beginning of plant senescence. Photosynthesis and leaf respiration (RD) variations allow A. distorta to maintain a positive carbon balance during the growing season becoming indicative of the efficiency of plant carbon use. The results could be an important tool for conservation programmes of the A. distorta wild populations.

The strong selection pressure exerted by intensive use of glyphosate in cultivated areas has selected populations of the Rubiaceae weed species Borreria latifolia (Aubl.) K.Shum. (broadleaf buttonweed), Galianthe chodatiana (Standl.) E.L. Cabral (galiante) and Richardia brasiliensis Gomes (Brazilian pusley) with differential sensitivity to this herbicide in the South region of Brazil. The control of these weeds with herbicides is troublesome and signals the need to incorporate management practices of ruderal flora and crops, more sustainable and that results in more efficient control for long-term. Therefore, it is very important to expand the information about their biology and management. This study aimed: (a) select efficient methods to overcome dormancy of B. latifolia and G. chodatiana and determine how they influence the kinetics of seeds germination; (b) analyze the effects of temperature, irradiance, pH, aluminum and salinity on seed germination and initial growth of the B. latifolia, G. chodatiana e R. brasiliensis seedlings; (c) evaluate tolerance to glyphosate levels in biotypes of B. latifolia, G. chodatiana e R. brasiliensis through dose-response curves and compare two methods to evaluate herbicidal control; (d) and evaluated the effectiveness of alternative herbicides in pre-emergence and in early and late post-emergence of the three species. The treatment with KNO3 2%/3h + gibberellic acid 400 ppm resulted in higher percentage of G. chodatiana seed germination. This treatment and also the dry heat (60°C/30 min) + KNO3 2%/3h were more effective in overcoming dormancy of B. latifolia. G. chodatiana and R. brasiliensis tolerate lower temperatures during the germination process, while B. latifolia tolerate higher temperatures. B. latifolia and R. brasiliensis are positive photoblastic while G. chodatiana is indifferent to the photoperiod. B. latifolia shows higher germination and early development in pH 3, while G. chodatiana and R. brasiliensis prefer pH range between 5 and 7. B. latifolia and G. chodatiana were more tolerant to the aluminum during the germination process than R. brasiliensis. Low salt levels were sufficient to reduce the seed germination of the three species. Some biotypes of B. latifolia and R. brasiliensis showed medium-high glyphosate tolerance, not being controlled by higher doses than recommended. The G. chodatiana specie was not controlled with the highest dose used, showing a high glyphosate tolerance. The sulfentrazone, s-metolachlor and saflufenacil herbicides sprayed in pre-emergence showed high efficacy both on B. latifolia and R. brasiliensis, while chlorimuron-ethyl and diclosulan were effective only on R. brasiliensis. In early post-emergence the fomesafen, lactofem and flumioxazin herbicides efficiently controlled plants of all species, while bentazon showed high efficacy only on B. latifolia. Noteworthy the susceptibility of the G. chodatiana specie for applications in early post-emergence, because the control effectiveness and the number of effective herbicides are reduced with increasing the plant age. Many treatments with tank mix or sequencial applications with glyphosate, were effective in controlling B. latifolia and R. brasiliensis plants in advanced stage of development.

Sesbania punicea (Cav.) Benth. é um arbusto que pode atingir de 1-4m de altura, cuja floração ocorre na primavera e no verão, frutificando no outono. O fruto é do tipo legume reto, produzindo em média 10 sementes por fruto. Sua propagação é frequentemente facilitada por perturbações na natureza, sendo bastante tolerante a solos pobres se adaptando a qualquer clima. O presente estudo teve por objetivo verificar os requisitos para germinação de Sesbania punicea sob influência de fatores abióticos. As sementes de Sesbania utilizadas neste estudo foram coletadas na APA da Lagoa Verde, Rio Grande- RS. Para avaliar o investimento reprodutivo da espécie foram realizadas as medidas morfométricas de 200 sementes. Para o teste de germinação, foram utilizados dois tipos de tratamentos: umidade e alagamento, ambos nas condições de luz e escuro, à temperatura de 25ºC, submetidos a quatro potenciais salinos: 0, 5, 10 e 15g de NaClL -1 . No teste em alagamento foram colocadas 100 sementes para cada salinidade em tubos Falcon transparentes, e para umidade, o mesmo número de sementes foi disposta em caixas gerbox transparentes contendo papel germitest, distribuídas de acordo com as respectivas salinidades. Os testes tiveram duração de 30 dias e a germinação foi acompanhada diariamente. Para avaliar o crescimento, 80 sementes foram colocadas em copos de poliestireno transparente de 100 ml, sendo 10 sementes para cada teste e, para análise da concentração de pigmentos, foram utilizadas plântulas com 30 dias de idade, crescendo nas mesmas condições de salinidade em que as sementes germinaram. As amostras de plântulas foram pesadas e logo após maceradas com 100 ml de acetona a 80% de concentração. A seguir, foram realizadas as leituras da concentração dos pigmentos em espectrofotômetro. As sementes de S. punicea utilizadas neste estudo, apresentaram tamanho pequeno (0,6 ± 0,01 cm). Houve respostas positivas de germinação tanto em presença quanto em ausência de luz, embora a germinação no teste em luz tenha sido maior, assim como a germinação no tratamento Alagado/ Luz, em 0g NaCl L-1 . Ao serem realizados os testes adicionando a variável salinidade, as respostas de germinação destas sementes mostraram um comportamento diferenciado, tendo um número menor de sementes germinadas com o aumento da concentração salina. As plântulas cresceram mais nos tratamentos em alagado e o aumento da concentração salina também reduziu o tamanho e o número de folhas das mesmas. No experimento de extração de pigmentos, clorofila a e b, os testes não demonstraram diferenças entre os tratamentos. Embora sementes de Sesbania punicea germinem sob influência de salinidade, o baixo desenvolvimento das plântulas observadas neste estudo sinalizam o cuidado que deve ser tomado ao utilizarmos a espécie para recuperação de áreas degradadas em ambientes com influência de salinidade.

Fumigation with SF did not have any impact on the baking properties of a wholemeal durum flour-commercial flour mix. Therefore, SF is not recommended if the grains are to be used as seeds for agricultural production but for the production of semolina, pasta and bread, SF used under typical fumigation conditions has little to no impact on technological properties of durum wheat.

The bean bruchids, Acanthoscelides obtectus Say and Zabrotes subfasciatus Boheman (Coleoptera: Bruchidae), are cosmopolitan pests of stored dry common beans ( Phaseolus vulgaris L. ), causing damage through reduction of grain quality and seed germination. Biological resistance to these bruchids was definitively established in non-cultivated bean accessions, and has been introgressed into a range of drybean market classes. However, existing resistance to bruchids in Uganda’s common bean germplasm has not been systematically studied. In this study, 45 bean genotypes from the National Bean-Breeding Programme (25 genotypes) and agroecologically diverse bean growing areas in Uganda (20 genotypes), were evaluated for postharvest bruchid resistance. None of the evaluated bean genotypes expressed resistance to either bruchid species, with all the 45 bean genotypes supporting bruchid development, reproduction and feeding. All genotypes were severely damaged by bruchids feeding, resulting in significant (P<0.05) reduction of seed germination. Reduction in seed germination was related to the number of emergence holes and seed size; small bean seeds damaged by up to 2 bruchid emergence holes had a 7.1% reduction in germination, while large bean seeds with a similar number of emergence holes showed a 25% reduction in germination. Whereas this study further confirms bruchids as important storage pests of beans causing direct loss through consumption of the seed and indirect loss through viability deterioration, the resistance to bruchids in the evaluated range of Uganda’s dry bean germplasm is inadequate for direct exploitation in a breeding programme.

Avaliaram-se a germinação e o vigor de sementes de melão (Cucumis melo L.), sob condicoes de ripado, utilizando-se solos com diferentes concentrações salinas. Observou-se que a germinação das sementes manteve-se inalterada ate o nível de 6,27 mmho/cm de condutividade elétrica, enquanto o seu vigor foi afetado por níveis inferiores. Níveis mais altos de salinidade exerceram efeitos bem pronunciados na germinação ao nível de CE 25oC= 9,46 mmho/cm, uma redução superior a 50% e no vigor, uma drástica redução.

Las orquídeas producen abundantes semillas pequeñas, careciendo de endospermo, cotiledones y sustancias de reserva para llevar a cabo su germinación. Es por esto que estratégicamente las semillas establecen una relación simbiótica con un hongo micorrízico que favorece a su germinación y desarrollo. El objetivo de este estudio fue determinar la especificidad del hongo micorrízico (Rhizoctonia sp.) en la germinación de cinco géneros de orquídeas. Se usaron dos medios de cultivo: 1) PhytamaxTM y 2) avena-agar+Rhizoctonia sp. Los resultados evaluados a los 45 y 75 días demuestran que no existe especificidad entre el hongo Rhizoctonia sp. y Trichoceros antenifer, especie de la cual se aisló el hongo. La germinación en tres de los cinco géneros evaluados fue mayor en el tratamiento avena-agar+Rhizoctonia sp., evidenciando también mayor tamaño y vigor las plantas que se desarrollaron en este tratamiento, aunque estos datos no fueron evaluados. Los resultados sugieren también que la planta hospedera del hongo inoculado podría tener una ventaja en cuanto al tiempo requerido para su germinación; sin embargo otras, especies también se ven favorecidas con el inóculo, aunque éste sea obtenido de otra fuente.

RESUMO: O objetivo deste estudo foi verificar a influência de Apis mellifera na produção de grãos e qualidade de sementes da soja transgênica Glycine max (L.) Merrill Roundup ReadyTM e convencional. A soja transgênica foi plantada intercalada com a convencional, em 18 parcelas, em três tratamentos: gaiolas com abelhas A. mellifera, gaiolas sem abelhas e áreas descobertas, com livre visitação de insetos. Na soja transgênica, em três parcelas de cada tratamento, foi aplicado glifosato, 30 dias após a emergência. Os parâmetros analisados foram: produção de grãos; número de vagens por planta; peso de 100 sementes e porcentagem de germinação das sementes. Não houve diferença entre as cultivares, entretanto a produção de 2.757,40 kg ha-1 obtida na área coberta por gaiola com abelhas, e 2.828,47 kg ha-1 na área livre para visitação de insetos foram superiores a 2.000,53 kg ha-1 da área coberta por gaiola sem abelhas. O número de vagens por planta foi maior na área coberta por gaiola com abelhas (38,28) e área livre (32,65), quando comparado com o da área coberta por gaiola sem abelhas (21,19). O peso médio de 100 sementes e a germinação das sementes não foram diferentes entre as cultivares e nem entre os tratamentos. Conclui-se que, para as cultivares estudadas, houve benefício na produção de grãos de 37,84%, quando foi permitida a visita de abelhas. ABSTRACT. Pollination by Apis mellifera in transgenic soy (Glycine max (L.) Merrill) Roundup Ready? cv. BRS 245 RR and conventional cv. BRS 133. This research was carried out to evaluate the influence of Africanized honeybees in grain production and seed quality of Glycine max (L.) Merrill Roundup Ready? transgenic soy, as well as of conventional soy. Transgenic soy was interposed with conventional soybean, in 18 plots and three treatments: covered area with Africanized honeybees, covered area without honeybees, and uncovered area with free insect visitation. The herbicide Glyphosate was applied on three plots of each treatment of transgenic soy, 30 days after emergence. Grain production, number of pods/plant, weight per 100 seeds, and seed germination percentage were evaluated. There was no difference among cultivars; however, the production in the covered area with honeybees (2757.4 kg ha-1) and in the uncovered area (2828.47 kg ha-1) were higher than in the covered area without honeybees (2000.53 kg ha-1). The number of pods/plant was greater than in the covered area with honeybees (38.28) and in the uncovered area (32.65) as compared to the covered area without honeybees (21.19). The weight per 100 seeds seed germination did not differ among cultivars or treatments. It can be concluded that, for these cultivars, there was a rise in grain production of 37.84% when honeybee visits were allowed.

The brazilian-plum (Spondias tuberosa, His) is a tropical fruit tree that has been consolidated in the market for agribusiness processing, due to its characteristic flavor of fruit. Accordingly, studies to optimize the propagation of plants are necessary for production of seedlings with agronomic and quality assurance measures. This study aimed at determining the efficient techniques for uniform seed germination, as brazilian-plum seed present mechanical dormancy, and establish optimal culture media for multiplication of shoots from the in vitro micropropagation. Firstly, in a greenhouse at the Universidade Federal do Rio Grande do Norte, was evaluated the influence of different methods of breaking dormancy in the emergence of seedlings of brazilian-plum and speed of germination (IVG) of seeds. After 60 days of cultivation, it was found that splay in the distal portion of the seed was the best treatment, with rates of 85.33% in germinability and 3.415 of IVG, compared with the treatment of seed-soaking in water for 12h + humus and the control group. Subsequently, new sources of seedling explants were obtained in studies of tissue culture. Laboratory of Plant Biotechnology that the university, was used stem apex, nodal segments and internodes in search of decontamination with various concentrations of calcium hypochlorite [Ca(OCl)2] and micropropagation, inoculating them in half WPM (1980) with various concentrations of 6-benzylaminopurine (BAP). We used 10 sample units with three replications for different concentrations of [Ca(OCl)2], BAP and explants type. After thirty days, which was observed for the control of contamination, during the establishment in vitro, concentrations of [Ca(OCl)2] between 0.5% and 2.0% were effective in combating exogenous contamination of the apex. In nodal segments and internodes, concentrations of [Ca(OCl)2] between 1.0% and 2.0% and 1.5% and 2.0% were respectively, sufficient to reduce the percentage of losses in these infestations explants. For micropropagation, the culture medium supplemented with 0.1 mg.L-1 BAP promotes better development of multiple shoots per explants from nodal segment. However, success does not get to shoot training in internodal segment
