The wine production in the region of the Sub-middle São Francisco river Valley has started there are few years ago.The region has a climate variability that allows to obtain two or three harvests per year, which influences significantly the composition of the grapes. This work aimed to evaluate the amount of amines present in white and red tropical wines produced in the Northeast of Brazil. The winemaking was done by the traditional method. After stabilizing the wines were bottled and then analyzed for the determination of the amines putrescine, spermidine and spermine, by thin layer chromatography (TLC). The harvests were in June (harvest I) and November (harvest II) 2009. The results showed that the edaphoclimatic conditions of the two harvests had significant influence (p> 0.05) on bioactive amines concentration, with high values for red wines. In addition, tropical wines from Sub-middle São Francisco river Valley showed adequate correlation between bioactive amines as compared to other wines in the world.

The prognostic value of ABC transporters in Ewing sarcoma is still poorly explored and controversial. We described for the first time the impact of various ABCs on Ewing sarcoma prognosis by assessment of their gene expression in two independent cohorts of patients. Unexpected associations with favourable outcomes were observed for two ABCs of the A-subfamily, ABCA6 and ABCA7, whereas no associations with the canonical multidrug ABC transporters were identified. The ABCs of the A-subfamily are involved in cholesterol/phospholipids transportation and efflux from cells. Our clinical data support the drug-efflux independent contribution to cancer progression of the ABCAs, which has been confirmed in PDX-derived cell lines. The impact of these ABCA transporters on tumor progression seems to be mediated by lowering intracellular cholesterol, supporting the role of these proteins in lipid transport. In addition, the gene expression of ABCA6 and ABCA7 is regulated by transcription factors which control lipid metabolism: ABCA6 was induced by the binding of FoxO1/FoxO3a to its promoter and repressed by IGF1R/Akt signaling, whereas the expression of ABCA7 was regulated by p53. The data point to ABCA6 and ABCA7 as potential prognostic markers in Ewing sarcoma and suggest the IGF1/ABCA/lipid axis as an intriguing therapeutic target. Agonist monoclonal antibodies towards ABCA6/7 or inhibitors of cholesterol biosynthesis, such as statins or aminobiphoshonates, may be investigated as therapeutic options in combination with chemotherapy. Considering that no monoclonal antibodies selectively targeting extracellular domains of ABCA6/7 are available, the second part of the project has been dedicated to the generation of human antibody phage-display libraries as tools for selecting monoclonal antibodies. A novel synthetic human antibody phage-display library has been designed, cloned and characterized. The library takes advantages of the high variability of a designed naïve repertoire to be a useful tool for isolating antibodies towards all potential antigens, including the ABCAs.

Zyxin is a phosphoprotein localized at the focal adhesions and on the actin stress fibres, where it regulates the cytoskeleton organization. In addition, zyxin can shift into the nucleus and modulates the gene expression, affecting key cellular processes. Consequently, zyxin is as a crucial factor in the malignancy of several cancers, like Ewing sarcoma (EWS). EWS is a rare tumour of the bones, affecting children and adolescents. The main features of EWS are the presence of a chimeric transcriptional factor, EWS-FLI1 and the high expression of CD99, a glycoprotein necessary for the maintenance of the malignant phenotype. Triggering of CD99 with specific antibodies causes massive cell death, an effect that requires zyxin presence. In EWS zyxin is repressed by EWS-FLI1 and its forced re-expression counteracts the malignant phenotype. In this work we decided to deepen our knowledge on how zyxin affects EWS malignancy. We proved that zyxin is a negative regulator of cell migration, survival and growth in anchorage-independent conditions, confirming the tumour suppressor role of zyxin. Then we focused on the relation between CD99 and zyxin. Loss of function of CD99, by engagement with specific antibodies or use of shRNA, increases zyxin levels and promotes its nuclear translocation. Here, we observed that zyxin impairs the transcriptional activity of the Glioma associated oncogene 1 (Gli1), a member of the Hedgehog signalling pathway, which has a relevant oncogenic function in EWS. To support these evidences, we also reported that the loss of function of CD99 inhibits, trough zyxin mediation, the expression of Gli1 up-regulated target genes, such as NKX2-2, PTCH1 and cyclins, whilst enhances the expression of its down-regulated target GAS1. In conclusion, we presented a more accurate depiction of zyxin role in EWS, which in the future could be further developed in hope to offer new therapeutic approaches.
