Earthen fills and back slopes resulting from highway building and other construction projects pose problems with respect to erosion stabilization and establishing vegetation cover. Sediments from such slopes create stream pollution while the erosion itself results in maintenance problems. Furthermore, adverse conditions aggravated by erosion prevent satisfactory establishing of vegetative cover. A dense vegetative cover is very effective in controlling erosion but even with optimum weather and soil conditions there is a delay of about 10 weeks between seeding and the establishment of a vegetative cover. Under actual field conditions, 3 months to a year may elapse between completion of construction and establishment of a vegetative cover. A research project was initiated early in 1974 to determine the effectiveness of a rock mulch of crushed limestone aggregates in controlling soil losses on highway construction back slopes in Iowa and to find the influence of such treatments on stand establishment of grasses and legumes.

Semi-automatic capillary gas chromatographic method with classical flame ionization detection, which satisfies the conditions for required performance and gave acceptable results within the framework of an interlaboratory certification programme for PAHs in sewage sludge, is described. The interesting feature of the procedure is that it incorporates automatic operations such as sample fractionation by semi-preparative HPLC, fraction collection at signal level recognition and evaporation under nitrogen flow. Multiple injections in the GC capillary column are performed in the on-column mode via an autosampler with temperature-programmable injector. Automatic data acquisition and chromatogram treatment are made via computer software. This partially automatic procedure releases personnel from tedious and time-consuming tasks and its robust character was validated through the certification of reference material for PAHs in sewage sludge, demonstrating its reliable performance.

In forensic pathology routine, fatal cases of contrast agent exposure can be occasionally encountered. In such situations, beyond the difficulties inherent in establishing the cause of death due to nonspecific or absent autopsy and histology findings as well as limited laboratory investigations, pathologists may face other problems in formulating exhaustive, complete reports, and conclusions that are scientifically accurate. Indeed, terminology concerning adverse drug reactions and allergy nomenclature is confusing. Some terms, still utilized in forensic and radiological reports, are outdated and should be avoided. Additionally, not all forensic pathologists master contrast material classification and pathogenesis of contrast agent reactions. We present a review of the literature covering allergic reactions to contrast material exposure in order to update used terminology, explain the pathophysiology, and list currently available laboratory investigations for diagnosis in the forensic setting.

Newly synthesized glucose transporter 4 (GLUT4) enters into the insulin-responsive storage compartment in a process that is Golgi-localized γ-ear-containing Arf-binding protein (GGA) dependent, whereas insulin-stimulated translocation is regulated by Akt substrate of 160 kDa (AS160). In the present study, using a variety of GLUT4/GLUT1 chimeras, we have analyzed the specific motifs of GLUT4 that are important for GGA and AS160 regulation of GLUT4 trafficking. Substitution of the amino terminus and the large intracellular loop of GLUT4 into GLUT1 (chimera 1-441) fully recapitulated the basal state retention, insulin-stimulated translocation, and GGA and AS160 sensitivity of wild-type GLUT4 (GLUT4-WT). GLUT4 point mutation (GLUT4-F5A) resulted in loss of GLUT4 intracellular retention in the basal state when coexpressed with both wild-type GGA and AS160. Nevertheless, similar to GLUT4-WT, the insulin-stimulated plasma membrane localization of GLUT4-F5A was significantly inhibited by coexpression of dominant-interfering GGA. In addition, coexpression with a dominant-interfering AS160 (AS160-4P) abolished insulin-stimulated GLUT4-WT but not GLUT4-F5A translocation. GLUT4 endocytosis and intracellular sequestration also required both the amino terminus and large cytoplasmic loop of GLUT4. Furthermore, both the FQQI and the SLL motifs participate in the initial endocytosis from the plasma membrane; however, once internalized, unlike the FQQI motif, the SLL motif is not responsible for intracellular recycling of GLUT4 back to the specialized compartment. Together, we have demonstrated that the FQQI motif within the amino terminus of GLUT4 is essential for GLUT4 endocytosis and AS160-dependent intracellular retention but not for the GGA-dependent sorting of GLUT4 into the insulin-responsive storage compartment.

Phototropism allows plants to redirect their growth towards the light to optimize photosynthesis under reduced light conditions. Phototropin 1 (phot1) is the primary low blue light-sensing receptor triggering phototropism in Arabidopsis. Light-induced autophosphorylation of phot1, an AGC-class protein kinase, constitutes an essential step for phototropism. However, apart from the receptor itself, substrates of phot1 kinase activity are less clearly established. Phototropism is also influenced by the cryptochromes and phytochromes photoreceptors that do not provide directional information but influence the process through incompletely characterized mechanisms. Here, we show that Phytochrome Kinase Substrate 4 (PKS4), a known element of phot1 signalling, is a substrate of phot1 kinase activity in vitro that is phosphorylated in a phot1-dependent manner in vivo. PKS4 phosphorylation is transient and regulated by a type 2-protein phosphatase. Moreover, phytochromes repress the accumulation of the light-induced phosphorylated form of PKS4 showing a convergence of photoreceptor activity on this signalling element. Our physiological analyses suggest that PKS4 phosphorylation is not essential for phototropism but is part of a negative feedback mechanism.

Os meios semi-sólidos são os mais utilizados em trabalhos de micropropagação, mas há indícios de que o estado físico dos meios de cultura influencia a multiplicação dos cultivos. O objetivo deste trabalho foi estabelecer um protocolo para a multiplicação de material propagativo de batata em meio de cultura líquido. Explantes de batata da cultivar Eliza, com uma gema axilar, foram cultivados em seis diferentes meios de cultura, com (semi-sólido) ou sem (líquido) a adição do solidificante ágar. Após 21 dias de cultivo, o meio de cultura que proporcionou os melhores resultados para crescimento e taxa de multiplicação teve sua composição modificada com o objetivo de melhorar a eficiência de multiplicação de cinco cultivares em meio líquido. Foi também avaliada a necessidade de agitação dos cultivos em meio líquido. Houve ganho na eficiência de multiplicação in vitro da batata, quando se utiliza meio de cultura líquido. Os melhores resultados são obtidos quando o material é cultivado em meio constituído pelos sais de MS na concentração plena, acrescido de ácido pantotênico (5,0 mg L-1), tiamina (1,0 mg L-1), ácido giberélico (0,25 mg L-1) e sacarose (20 g L-1), sob agitação constante.

Soils on gypsum are well known in dry climates, but were very little described in temperate climate, and never in Switzerland. This study aims to describe soils affected by gypsum in temperate climate and to understand their pedogenesis using standard laboratory analyzes performed on ten Swiss soils located on gypsum outcrops. In parallel, phytosociological relevés described the vegetation encountered in gypsiferous grounds. Gypsification process (secondary gypsum enrichment by precipitation) was observed in all soils. It was particularly important in regions where potential evapotranspiration exceed strongly precipitations in summer (central Valais, Chablais under influence of warm wind). Gypsum contents were regularly measured above 20% in deep horizons, and exceeded locally 70%, building a white, indurate horizon. However, the absence of such a gypsic horizon in the top soil hindered the use of gypsosol (according to the Référentiel pédologique, BAIZE & GIRARD 2009), the typical name of soils affected by gypsum, but restricted to dry regions. As all soils had a high content of magnesium carbonates, they were logically classified in the group of DOLOMITOSOLS. However, according to the World Reference Base for Soil Resources (IUSS 2014), five soils can be classified among the Gypsisols, criteria being here less restricting. These soils are characterized by a coarse texture and a particulate brittle structure making a filtering substrate. They allow water to flow easily taking nutrients. They are not retained by clay, which does generally not exceed 1% of the fine material. The saturation of calcium blocks the breakdown of organic matter. Moreover, these soils are often rejuvenated by erosion caused by the rough relief due to gypsum (landslides, sinkholes, cliffs and slopes). Hence, the vegetation is mainly characterized by calcareous and drought tolerant species, with mostly xerothermophilic beech (Cephalanthero-Fagenion) and pine forests (Erico-Pinion sylvestris) in lowlands, or subalpine heathlands (Ericion) and dry calcareous grasslands (Caricion firmae) in higher elevations.

La presència de pesticides en el medi ambient pot comportar efectes nocius pel propi medi i la salut humana, fet que, en alguns casos, converteix en necessària la seva eliminació. Un dels mètodes utilitzats per dur a terme aquesta eliminació és la sorció de contaminats sobre materials adsorbents. Per tal de fer d’aquest mètode un procés sostenible cal investigar nous materials capaços de retenir els contaminants. El suro és la part més externa de l’escorça de l’alzina surera: Quercus Suber L. S’extreu cada 5- 10 anys depenent de la regió i es caracteritza per ser una font natural, renovable i biodegradable amb una heterogènia composició química que el converteix en un material potencialment apte com a adsorbent d’un ampli rang de contaminants. En aquest context, l’objectiu principal d’aquest treball és investigar l’afinitat d’adsorció del suro amb quatre pesticides de diferent hidrofobicitat i estructura química i estudiar el paper que hi juguen els seus compostos químics (extractius, suberina, lignina i polisacàrids) en aquest procés de sorció. Els pesticides investigats han estat: Metamitron: poc hidrofòbic (logKow = 0.83) i de caràcter molecular, Alaclor: moderadament hidrofòbic (logKow = 2.80) i de caràcter iònic (pKa = 0.62), 2,4-D: moderadament hidrofòbic (logKow = 2.81) i de caràcter iònic (pKa = 2.64) i Clorpirifos: molt hidrofòbic (logKow = 4.92) i de caràcter molecular

Síntesis actualizada de los estudios monográficos realizados sobre las cerámicas comunes y ánforas de la ciudad de Tarraco desde finales del s. IV hasta inicios del s. VIII. Se hace especial hincapié en los últimos hallazgos del siglo VII que reflejan una intensificación de la presencia de materiales orientales en la ciudad.

The hepatitis C virus (HCV) NS3-4A protease is not only an essential component of the viral replication complex and a prime target for antiviral intervention but also a key player in the persistence and pathogenesis of HCV. It cleaves and thereby inactivates two crucial adaptor proteins in viral RNA sensing and innate immunity, mitochondrial antiviral signaling protein (MAVS) and TRIF, a phosphatase involved in growth factor signaling, T-cell protein tyrosine phosphatase (TC-PTP), and the E3 ubiquitin ligase component UV-damaged DNA-binding protein 1 (DDB1). Here we explored quantitative proteomics to identify novel cellular substrates of the NS3-4A protease. Cell lines inducibly expressing the NS3-4A protease were analyzed by stable isotopic labeling using amino acids in cell culture (SILAC) coupled with protein separation and mass spectrometry. This approach identified the membrane-associated peroxidase GPx8 as a bona fide cellular substrate of the HCV NS3-4A protease. Cleavage by NS3-4A occurs at Cys 11, removing the cytosolic tip of GPx8, and was observed in different experimental systems as well as in liver biopsies from patients with chronic HCV. Overexpression and RNA silencing studies revealed that GPx8 is involved in viral particle production but not in HCV entry or RNA replication. Conclusion: We provide proof-of-concept for the use of quantitative proteomics to identify cellular substrates of a viral protease and describe GPx8 as a novel proviral host factor targeted by the HCV NS3-4A protease. INTRODUCTION: Osteoset(®) T is a calcium sulphate void filler containing 4% tobramycin sulphate, used to treat bone and soft tissue infections. Despite systemic exposure to the antibiotic, there are no pharmacokinetic studies in humans published so far. Based on the observations made in our patients, a model predicting tobramycin serum levels and evaluating their toxicity potential is presented. METHODS: Following implantation of Osteoset(®) T, tobramycin serum concentrations were monitored systematically. A pharmacokinetic analysis was performed using a non-linear mixed effects model based on a one compartment model with first-degree absorption. RESULTS: Data from 12 patients treated between October 2006 and March 2008 were analysed. Concentration profiles were consistent with the first-order slow release and single-compartment kinetics, whilst showing important variability. Predicted tobramycin serum concentrations depended clearly on both implanted drug amount and renal function. DISCUSSION AND CONCLUSION: Despite the popularity of aminoglycosides for local antibiotic therapy, pharmacokinetic data for this indication are scarce, and not available for calcium sulphate as carrier material. Systemic exposure to tobramycin after implantation of Osteoset(®) T appears reassuring regarding toxicity potential, except in case of markedly impaired renal function. We recommend in adapting the dosage to the estimated creatinine clearance rather than solely to the patient's weight.

The package also includes material specifications for purchasing ETP and specifications for the application of ETP by contract.
