Culturally responsive instruction refers to the identification of relevant cultural aspects of students' lives and infusion of these into the curriculum. This instructional approach assumes that a culturally appropriate curriculum can potentially motivate, engage, and lead students to higher rates of achievement. This quasi-experimental study (N=44) investigated the relationship of culturally responsive instruction and the reading comprehension and attitude of struggling urban adolescent readers. The study incorporated the use of culturally responsive instruction using culturally relevant literature (CRL), the Bluford Series Novels, as authentic texts of instruction. Participants were seventh grade reading students at a Title I middle school in South Florida. After a baseline period, two different classes were taught for 8 weeks using different methods. One class formed the experimental group ( n=22) and the other class formed the comparison group (n =22). The CRI curriculum for the experimental group embraced the socio-cultural perspective through the use of small discussion groups in which students read and constructed meaning with peers through interaction with the Bluford Series Novels; gave written responses to multiple strategies according to SCRAP - Summarize, Connect, Reflect, Ask Questions, Predict; responded to literal and inferential questions, while at the same time validating their responses through evidence from the text. The Read XL (basal reader) curriculum of the comparison group utilized a traditional form of instruction which incorporated the reading of passages followed by responses to comprehension questions, and teacher-led whole group discussion. The main sources of data were collected from the Gates-MacGinitie Reading Tests, the Florida Assessments for Instruction in Reading (FAIR), and the Rhody Secondary Reading Attitude Assessment. Statistical analyses were performed using Repeated Measures ANOVAs. Findings from the study revealed that the experimental participants' reading attitudes and FAIR comprehension scores increased when compared to the comparison group. Overall, the results from the study revealed that culturally responsive instruction can potentially foster reading comprehension and a more positive attitude towards reading. However, a replication of this study in other settings with a larger, more randomized sample size and a greater ethnic variation is needed in order to make full generalizations.

High-resolution sedimentary records of major and minor elements (Al, Ba, Ca, Sr, Ti), total organic carbon (TOC), and profiles of pore water constituents (SO42-, CH4, Ca2+, Ba2+, Mg2+, alkalinity) were obtained for two gravity cores (core 755, 501 m water depth and core 214, 1686 m water depth) from the northwestern Black Sea. The records were examined in order to gain insight into the cycling of Ba in anoxic marine sediments characterized by a shallow sulfate-methane transition (SMT) as well as the applicability of barite as a primary productivity proxy in such a setting. The Ba records are strongly overprinted by diagenetic barite (BaSO4) precipitation and remobilization; authigenic Ba enrichments were found at both sites at and slightly above the current SMT. Transport reaction modeling was applied to simulate the migration of the SMT during the changing geochemical conditions after the Holocene seawater intrusion into the Black Sea. Based on this, sediment intervals affected by diagenetic Ba redistribution were identified. Results reveal that the intense overprint of Ba and Baxs (Ba excess above detrital average) strongly limits its correlation to primary productivity. These findings have implications for other modern and ancient anoxic basins, such as sections covering the Oceanic Anoxic Events for which Ba is frequently used as a primary productivity indicator. Our study also demonstrates the limitations concerning the use of Baxs as a tracer for downward migrations of the SMT: due to high sedimentation rates at the investigated sites, diagenetic barite fronts are buried below the SMT within a relatively short period. Thus, 'relict' barite fronts would only be preserved for a few thousands of years, if at all.

During Ocean Drilling Program Leg 210, a greatly expanded sedimentary sequence of continuous Cretaceous black shales was recovered at Site 1276. This section corresponds to the Hatteras Formation, which has been documented widely in the North Atlantic Ocean. The cored sequence extends from the lowermost Albian, or possibly uppermost Aptian, to the Cenomanian/Turonian boundary and is characterized by numerous gravity-flow deposits and sporadic, finely laminated black shales. The sequence also includes several sedimentary intervals with high total organic carbon (TOC) contents, in several instances of probable marine origin that may record oceanic anoxic events (OAE). These layers might correspond to the Cenomanian-Turonian OAE 2; the mid-Cenomanian event; and OAE 1b, 1c, and 1d in the Albian. In addition, another interval with geochemical characteristics similar to OAE-type layers was recognized in the Albian, although it does not correspond to any of the known OAEs. This study investigates the origin of the organic matter contained within these black shale intervals using TOC and CaCO3 contents, Corg/Ntot ratios, organic carbon and nitrogen isotopes, trace metal composition, and rock-eval analyses. Most of these black shale intervals, especially OAE 2 and 1b, are characterized by low 15N values (<0) commonly observed in mid-Cretaceous black shales, which seem to reflect the presence of an altered nitrogen cycle with rates of nitrogen fixation significantly higher than in the modern ocean.

d37Cl values were determined for Izu Bonin arc magmas erupted 0-44 Ma in order to better understand the time-dependent processing of volatiles in subduction zones. Pristine ash-sized particles (glass, pumice, scoria, and rock fragments) were handpicked from tephra drilled at ODP Site 782. d37Cl values for these particles span a large range from -2.1 to +1.7 per mil (error = ± 0.3 per mil) vs. SMOC (Standard Mean Ocean Chloride, defined as 0 per mil). The temporal data extend the previously reported range of d37Cl values of -2.6 to 0.4 per mil (bulk ash) and -5.4 to -0.1 per mil (volcanic gases) from the Quaternary Izu Bonin-Mariana volcanic front to more positive values. Overall, the temporal data indicate a time-progressive evolution, from isotopically negative Eocene and Oligocene magmas (-0.7 ± 1.1 per mil, n = 10) to Neogene magmas that have higher ?37Cl values on average (+0.3 ± 1.1 per mil; n = 13). The increase is due to the emergence of positive d37Cl values in the Neogene, while minimum d37Cl values are similar through time. The range in d37Cl values cannot be attributed to fractionation during melt formation and differentiation, and must reflect the diversity of Cl present in the arc magma sources. Cl clearly derives from the slab (> 96% Cl in arc magmas), but d37Cl values do not correlate with isotope tracers (e.g. 207Pb/204Pb and 87Sr/86Sr) that are indicative of the flux from subducting sedimentary and igneous crust. Given the steady, high Cl flux since at least 42 Ma, the temporal variability of d37Cl values is best explained by a flux from subducting isotopically positive and negative serpentinite formed in the ocean basins that mingles with and possibly overprints the isotopically negative flux from sediment and igneous crust at arc front depths. The change in the d37Cl values before and after backarc spreading may reflect either a tectonically induced change in the mechanism of serpentinite formation on the oceanic plate, or possibly the integration of isotopically positive wedge serpentinite as arc fluid source during the Neogene. Our study suggests that serpentinites are important fluid sources at arc front depth, and implies the return of isotopically positive and negative Cl from the Earth surface to the mantle.

Analysis of the word lancea, of Hispanic origin after Varro, and of place names, people´s names and personal names derived from it. It confirms that the spear was the most important weapon in the Bronze Age, belonging to the iuventus and used as heroic and divine symbol. This analysis confirms also the personality of the Lusitanians, a people related to the Celts but with more archaic archaeological, linguistic and cultural characteristics originated in the tradition of the Atlantic Bronze in the II millennium BC. It is also relevant to better know the organisation of Broze and Iron Age societies and the origin of Indo-Europeans peoples in Western Europe and of pre-Roman peoples of Iberia.

Este artículo pretende ser complemento y continuación de mis anteriores trabajos sobre la figura de Petrus Hispanus O. P., Auctor Summularum. Comienzo presentando algunos nuevos documentos relacionados con las cuestiones ya examinadas en mis artículos de 1997 y 2001. A continuación, me ocupo de las cuestiones aplazadas en el artículo de 2001: los problemas relativos a la figura de “Petrus Ferrandi” y su posible relación con el “auctor Summularum”, así como los argumentos de Tugwell contra la hipótesis de la posible identidad de estas dos figuras, examinados ahora desde la perspectiva del autor de la Legenda prima. Tras analizar testimonios procedentes de muy diversos ámbitos, afirmo, por una parte, que la hipótesis de la identidad entre “Petrus Ferrandi” y “Petrus Hispanus” podría ser correcta y, por otra parte, que no hay argumentos concluyentes que obliguen a afirmar con seguridad que el autor de la Legenda prima es Pedro Ferrando. Aunque los análisis no permiten por el momento determinar si es “Petrus Alfonsi” o “Petrus Ferrandi” el “auctor Summularum”, los testimonios recogidos y las conexiones establecidas contribuirán, sin duda, a orientar futuras investigaciones en torno a la figura de “Petrus Hispanus”.

El trabajo pretende mostrar los estereotipos de hombre y mujer en la sociedad occidental según los estudios de género para, más tarde, comprobar si dichos estereotipos se reflejan en la fraseología checa y española relativa a animales, es decir, en los zoologismos. El análisis se sustenta en las teorías de la lingüística cognitiva acerca de la metáfora conceptual y del lenguaje figurado convencional. Las conclusiones muestran una clara discriminación de ambos géneros en el lenguaje, siendo el femenino más afectado que el masculino.

Al revisar el corpus teórico sobre la discapacidad nos damos cuenta de que el discurso predominante ha sido el que proviene de las disciplinas científicas propias del modelo médico-patológico. Por ello, se presenta un análisis crítico de la discapacidad aportando perspectivas situadas y contextualizadas política y socialmente que tienen en cuenta la voz y la vivencia de las personas implicadas. Las teorías feministas, enmarcadas en las Ciencias Sociales, resultan claves para desentrañar la construcción ideológica de los discursos que históricamente han servido para dominar y oprimir a las personas con discapacidad, y a su vez, para construir un sujeto anómalo y desviado que había que disciplinar.

Vorliegende Arbeit beschäftigt sich mit den Auswirkungen von selbst-definierten Extensions auf Kompatibilität von SKOS-Thesauri untereinander. Zu diesem Zweck werden als Grundlage zunächst die Funktionsweisen von RDF, SKOS, SKOS-XL und Dublin Core Metadaten erläutert und die verwendete Syntax geklärt. Es folgt eine Beschreibung des Aufbaus von konventionellen Thesauri inkl. der für sie geltenden Normen. Danach wird der Vorgang der Konvertierung eines konventionellen Thesaurus in SKOS dargestellt. Um dann die selbst-definierten Erweiterungen und ihre Folgen betrachten zu können, werden fünf SKOS-Thesauri beispielhaft beschrieben. Dazu gehören allgemeine Informationen, ihre Struktur, die verwendeten Erweiterungen und ein Schaubild, das die Struktur als Übersicht darstellt. Anhand dieser Thesauri wird dann beschrieben wie Mappings zwischen den Thesauri erstellt werden und welche Herausforderungen dabei bestehen.

Human exposure to Bisphenol A (BPA) results mainly from ingestion of food and beverages. Information regarding BPA effects on colon cancer, one of the major causes of death in developed countries, is still scarce. Likewise, little is known about BPA drug interactions although its potential role in doxorubicin (DOX) chemoresistance has been suggested. This study aims to assess potential interactions between BPA and DOX on HT29 colon cancer cells. HT29 cell response was evaluated after exposure to BPA, DOX, or co-exposure to both chemicals. Transcriptional analysis of several cancer-associated genes (c-fos, AURKA, p21, bcl-xl and CLU) shows that BPA exposure induces slight up-regulation exclusively of bcl-xl without affecting cell viability. On the other hand, a sub-therapeutic DOX concentration (40 nM) results in highly altered c-fos, bcl-xl, and CLU transcript levels, and this is not affected by co-exposure with BPA. Conversely, DOX at a therapeutic concentration (4 μM) results in distinct and very severe transcriptional alterations of c-fos, AURKA, p21 and CLU that are counteracted by co-exposure with BPA resulting in transcript levels similar to those of control. Co-exposure with BPA slightly decreases apoptosis in relation to DOX 4 μM alone without affecting DOX-induced loss of cell viability. These results suggest that BPA exposure can influence chemotherapy outcomes and therefore emphasize the necessity of a better understanding of BPA interactions with chemotherapeutic agents in the context of risk assessment.

A produção de proteínas através de microrganismos tornou-se uma técnica muito importante na obtenção de compostos de interesse da indústria farmacêutica e alimentícia. Extratos brutos nos quais as proteínas são obtidas são geralmente complexos, contendo sólidos e células em suspensão. Usualmente, para uso industrial destes compostos, é necessário obtê-los puros, para garantir a sua atuação sem interferência. Um método que vem recebendo destaque especialmente nos últimos 10 anos é o uso da cromatografia de troca iônica em leito expandido, que combina em uma única etapa os passos de clarificação, concentração e purificação da molécula alvo, reduzindo assim o tempo de operação e também os custos com equipamentos para realização de cada etapa em separado. Combinado a este fato, a última década também é marcada por trabalhos que tratam da modelagem matemática do processo de adsorção de proteínas em resinas. Está técnica, além de fornecer informações importantes sobre o processo de adsorção, também é de grande valia na otimização da etapa de adsorção, uma vez que permite que simulações sejam feitas, sem a necessidade de gasto de tempo e material com experimentos em bancada, especialmente se é desejado uma ampliação de escala. Dessa forma, o objetivo desta tese foi realizar a modelagem e simulação do processo de adsorção de bioprodutos em um caldo bruto na presença de células, usando inulinase e C-ficocianina como objeto de estudo e purificar C-ficocianina utilizando resina de troca iônica em leito expandido. A presente tese foi então dividida em quatro artigos. O primeiro artigo teve como objeto de estudo a enzima inulinase, e a otimização da etapa de adsorção desta enzima em resina de troca iônica Streamline SP, em leito expandido, foi feita através da modelagem matemática e simulação das curvas de ruptura em três diferentes graus de expansão (GE). As máximas eficiências foram observadas quando utilizadas maiores concentrações de inulinase (120 a 170 U/mL), e altura de leito entre 20 e 30 cm. O grau de expansão de 3,0 vezes foi considerado o melhor, uma vez que a produtividade foi consideravelmente superior. O segundo artigo apresenta o estudo das condições de adsorção de C-ficocianina em resina de troca iônica, onde foi verificado o efeito do pH e temperatura na adsorção e após construída a isoterma de adsorção. A isoterma de adsorção da C-ficocianina em resina Streamline Q XL feita em pH 7,5 e a 25°C (ambiente), apresentou um bom ajuste ao modelo de Langmuir (R=0,98) e os valores qm (capacidade máxima de adsorção) e Kd (constante de equilíbrio) estimados pela equação linearizada da isoterma, foram de 26,7 mg/mL e 0,067mg/mL. O terceiro artigo aborda a modelagem do processo de adsorção de extrato não clarificado de C-ficocianina em resina de troca iônica Streamline Q XL em coluna de leito expandido. Três curvas de ruptura foram feitas em diferentes graus de expansão (2,0, 2,5 e 3,0). A condição de adsorção de extrato bruto não clarificado de C-ficocianina que se mostrou mais vantajosa, por apresentar maior capacidade de adsorção, é quando se alimenta o extrato até atingir 10% de saturação da resina, em grau de expansão 2,0, com uma altura inicial de leito de 30 cm. O último artigo originado nesta tese foi sobre a purificação de C-ficocianina através da cromatografia de troca iônica em leito expandido. Uma vez que a adsorção já havia sido estudada no artigo 2, o artigo 4 enfoca na otimização das condições de eluição, visando obter um produto com máxima pureza e recuperação. A pureza é dada pela razão entre a absorbância a 620 nm pela absorbância a 280 nm, e dizse que quando C-ficocianina apresenta pureza superior a 0,7 ela pode ser usada em como corante em alimentos. A avaliação das curvas de contorno indicou que a faixa de trabalho deve ser em pH ao redor de 6,5 e volumes de eluição próximos a 150 mL. Tais condições combinadas a uma etapa de pré-eluição com 0,1M de NaCl, permitiu obter C-ficocianina com pureza de 2,9, concentração 3 mg/mL, e recuperação ao redor de 70%.

The β-proteobacterium Chromobacterium violaceum is a Gram-negative, free-living, saprophytic and opportunistic pathogen that inhabits tropical and subtropical ecosystems among them, in soil and water of the Amazon. It has great biotechnological potential, and because of this potential, its genome was completely sequenced in 2003. Genome analysis showed that this bacterium has several genes with functions related to the ability to survive under different kinds of environmental stresses. In order to understand the physiological response of C. violaceum under oxidative stress, we applied the tool of shotgun proteomics. Thus, colonies of C. violaceum ATCC 12472 were grown in the presence and absence of 8 mM H2O2 for two hours, total proteins were extracted from bacteria, subjected to SDS-PAGE, stained and hydrolysed. The tryptic peptides generated were subjected to a linear-liquid chromatography (LC) followed by mass spectrometer (LTQ-XL-Orbitrap) to obtain quantitative and qualitative data. A shotgun proteomics allows to compare directly in complex samples, differential expression of proteins and found that in C. Violaceum, 131 proteins are expressed exclusively in the control condition, 177 proteins began to be expressed under oxidative stress and 1175 proteins have expression in both conditions. The results showed that, under the condition of oxidative stress, this bacterium changes its metabolism by increasing the expression of proteins capable of combating oxidative stress and decreasing the expression of proteins related processes bacterial growth and catabolism (transcription, translation, carbon metabolism and fatty acids). A tool with of proteomics as an approach of integrative biology provided an overview of the metabolic pathways involved in the response of C. violaceum to oxidative stress, as well as significantly amplified understanding physiological response to environmental stress. Biochemical and "in silico" assays with the hypothetical ORF CV_0868 found that this is part of an operon. Phylogenetic analysis of superoxide dismutase, protein belonging to the operon also showed that the gene is duplicated in genome of C. violaceum and the second copy was acquired through a horizontal transfer event. Possibly, not only the SOD gene but also all genes comprising this operon were obtained in the same manner. It was concluded that C. violaceum has complex, efficient and versatile mechanisms in oxidative stress response
