Comparação de métodos para extração de dna na reação em cadeia da polimerase para detecção de Salmonella enterica sorovar enteritidis em produtos avícolas

The identification of Salmonella spp. in food samples by microbiological diagnosis is time consuming, with approximately five different stages, requiring about 120 hours until the final result. The utilization of the polymerase chain reaction technique (PCR) can reduce this time, but substances present in samples may affect the reaction. The present work aimed to compare DNA extraction by thermic treatment and by the use of cetyltrimethil ammonium bromide (CTAB), in products originated from poultry houses corresponding to raw material (meat meal) and experimentally contaminated drag swabs. Materials obtained from the extractions were submitted to PCR, utilizing a pair of initiator oligonucleotides for amplification of Sdf 1 gene fragments. Comparing the methods of extraction, it was observed that when CTAB was employed, SE was detected in 70% of meat meal and in 80% of drag swabs, while the thermic treatment method yielded positive results in 20% of meat meal and in 40% of drag swabs. SE was detected under both methods utilized for DNA extraction, but the use of CTAB detected a greater number of positive samples, compared with thermal treatment.

Influência do método de preparação de eletrocatalisadores PtRu/C sobre a atividade catalítica frente à reação de oxidação de etanol em meio ácido

In this work the influence of variations in the borohydrate reduction method on the properties of PtRu/C electrocatalysts was investigated. The electrocatalysts were prepared using 1:1 ; 2:1; 5:1; 50:1 and 250:1 molar ratios of NaBH₄ to metals. The reduction was also performed by dripping or by fast addition of the solution. The results showed that PtRu nanoparticles obtained by fast addition had the smallest crystallite sizes. It was also noted that the catalytic activity increased as the borohydrate:metal molar ratio increased. The PtRu/C electrocatalyst (50:1) obtained by fast addition presented the best catalytic activity for ethanol electro-oxidation.

Efeito da adição de lantânio em catalisadores de Ni/ZrO2 aplicados na reação de reforma a vapor de etanol

The catalytic performance of Ni/ZrO2 catalysts loaded with different lanthanum content for steam reforming of ethanol was investigated. Catalysts were characterized by BET surface area, X-ray diffraction, UV-vis spectroscopy, temperature programmed reduction, and X-ray absorption fine structure techniques. Results showed that lanthanum addition led to an increase in the degree of reduction of both NiO and nickel surface species interacting with the support, due to the higher dispersion effect. The best catalytic performance at 450 ºC was found for the Ni/12LZ catalyst, which exhibited an effluent gaseous mixture with the highest H2 yield.

Componentes sensoriais e atencionais do tempo de reação: efeitos do tamanho, excentricidade e previsibilidade de estímulos visuais

A percepção visual depende do arcabouço sensorial e do processamento atencional. Este trabalho estudou o efeito, sobre o tempo de reação manual (TR), do tamanho, excentricidade e previsibilidade de estímulos visuais. No experimento 1 (n=8), um alvo foi apresentado aleatoriamente em uma de quatro excentricidades diferentes, possuindo três possíveis tamanhos. O experimento 2 (n=12) apresentava configuração similar, porém uma pista indicava o quadrante de maior probabilidade (70%) de apresentação do alvo. Os resultados mostraram um aumento do TR em função da excentricidade do alvo, além de uma diminuição do TR com o aumento do tamanho do alvo e indicação correta da pista. Uma análise das interações sugere uma superposição de mecanismos atencionais e puramente sensoriais compartilhando um estágio comum do processamento visual.

Poli (metil azoteto de glicidila) - GAP. II: mecanismo de reação

O poli (metil azoteto de glicidila) - GAP - é um material energético que pode ser utilizado como aglutinante (binder) e como plastificante energético em compostos explosivos e propulsores de foguetes. O GAP de baixo peso molecular pode ser obtido pela conversão direta da epicloridrina (ECH) a GAP. Neste trabalho, é proposto um possível mecanismo para esta conversão direta, fundamentado em análises de infravermelho de espécies intermediárias.

A short, efficient synthesis of 2'-deoxypseudoisocytidine based on Heck-chemistry

A novel synthesis of 2'-deoxypseudoisocytidine as well as of its phosphoramidite building block for oligonucleotide synthesis is presented. The synthesis is based on Heck-coupling between N-protected pseudoisocytosine and a silyl protected furanoid glycal. With this procedure the corresponding phosphoramidite building block is obtained in 5 steps and an overall yield of 28%.

Synthesis of pyrrolidine C-nucleosides via Heck reaction

A novel method for the synthesis of pyrrolidine C-nucleosides has been developed. The key step of the synthesis is the palladium(0)-mediated coupling of a disubstituted N-protected 2-pyrroline and 5-iodouracil. C-Nucleoside 14 and its N-methyl derivative 15 can easily be converted to the corresponding phosphoramidite building blocks for DNA synthesis
