Loading of the replicative ring helicase onto the origin of replication (oriC) is the final outcome of a well coordinated series of events that collectively constitute a primosomal cascade. Once the ring helicase is loaded, it recruits the primase and signals the switch to the polymerization mode. The transient nature of the helicase-primase (DnaB-DnaG) interaction in the Escherichia coli system has hindered our efforts to elucidate its structure and function. Taking advantage of the stable DnaB-DnaG complex in Bacillus stearothermophilus, we have reviewed conflicting mutagenic data from other bacterial systems and shown that DnaG interacts with the flexible linker that connects the N- and C-terminal domains of DnaB. Furthermore, atomic force microscopy (AFM) imaging experiments show that binding of the primase to the helicase induces predominantly a 3-fold symmetric morphology to the hexameric ring. Overall, three DnaG molecules appear to interact with the hexameric ring helicase but a small number of complexes with two and even one DnaG molecule bound to DnaB were also detected. The structural/functional significance of these data is discussed and a speculative structural model for this complex is suggested.

El presente trabajo de Grado tiene como propsito examinar la incidencia de las sancionesinternacionales en el marco del rgimen de no proliferacin nuclear en el caso de Irn durante el periodo 2006-2015, teniendo en cuenta factores histricos de aos anteriores. Se analiza y explica cmo las sanciones internacionales pueden ser una medida persuasiva por violar ciertos artculos del Tratado de no Proliferacin Nuclear. Finalmente identifica y analiza los tipos de sanciones econmicas, financieras y comerciales que los Estados y el Consejo de Seguridad de las Naciones Unidas le han impuesto a Irn, as como la manera en que estas han incidido en la esfera poltica iran y mundial.

El objetivo de la tesis es identificar una familia de argumentos que comparten una estructura con el principio de dualidad de la geometra proyectiva. Esta familia la denomino "argumentos duales". Para lograr este objetivo, tomo cuatro argumentos importantes de la filosofa analtica e identifico en ellos la estructura que comparten. Los cuatro argumentos son: (i) el acertijo de la induccin de Goodman; (ii) la indeterminacin de la referencia Putnam; (iii) la indeterminacin de la traduccin de Quine; (iv) la paradoja del seguimiento de reglas de Wittgenstein.

Este artculo presenta los resultados de una investigacin realizada al interior de dos contextos. Por un lado, el terico, en el marco de uno de los discursos ms relevantes en los campos de la estrategia organizacional, de la managerial and organizational cognition (MOC) y, en general, de los estudios organizacionales (organization studies): la construccin de sentido (sensemaking). Por el otro, el emprico, en una de las grandes compaas multinacionales del sector automotriz con presencia global. Esta corporacin enfrenta una permanente tensin entre lo que dicta la casa matriz, en relacin con el cumplimiento de metas y estndares especficos, considerando el mundo entero, y los retos que, teniendo en cuenta lo regional y lo local, experimentan los altos directivos encargados de hacer prosperar la empresa en estos lugares. La aproximacin implementada fue cualitativa. Esto en atencin a la naturaleza de la problemtica abordada y la tradicin del campo. Los resultados permiten ampliar el actual nivel de comprensin acerca de los procesos de sensemaking de los altos directivos al enfrentar un entorno estratgico turbulento.

Esse artigo parte da pesquisa que pretende analisar a importncia da ferrovia para a circulao na regio Norte Fluminense do Estado do Rio de Janeiro. A pesquisa, ainda no concluda, est baseada nos preceitos da Geografia Histrica (ver SILVA, 2008). Com isso necessrio analisar o processo maior de implantao ferroviria no Estado do Rio de Janeiro desde o incio da implantao ferroviria, em 1854, da primeira ferrovia brasileira, a Imperial Companhia de Navegao a Vapor e Estrada de Ferro de Petrpolis. A partir da surge a Ferrovia Dom Pedro II, uma das mais importantes do pas e depois a Estrada de Ferro Cantagalo. So ento implantadas diversas ferrovias, sempre atendendo interesses regionais capitaneados por lideranas locais. Assim que a ferrovia chega em Campos dos Goytacazes em 1875. Outra importante ferrovia do estado foi a Estrada de Ferro Leopoldina, que foi incorporando vrias pequenas linhas frreas, at a transformao desta ferrovia em outra empresa a The Leopoldina Railway Company, que comea a funcionar em 1898, substituindo o caos reinante na companhia anterior. O conhecimento do perodo inicial de configurao de uma rede ferroviria no Norte Fluminense passa a ser de fundamental importncia devido importncia da Companhia Leopoldina para a regio Norte Fluminense em perodos posteriores.

Mutations in the FGFR3 gene cause the phenotypic spectrum of FGFR3 chondrodysplasias ranging from lethal forms to the milder phenotype seen in hypochondroplasia (Hch). The p.N540K mutation in the FGFR3 gene occurs in 70% of individuals with Hch, and nearly 30% of individuals with the Hch phenotype have no mutations in the FGFR3, which suggests genetic heterogeneity. The identification of a severe case of Hch associated with the typical mutation c.1620C > A and the occurrence of a c.1150T > C change that resulted in a p.F384L in exon 10, together with the suspicion that this second change could be a modulator of the phenotype, prompted us to investigate this hypothesis in a cohort of patients. An analysis of 48 patients with FGFR3 chondrodysplasia phenotypes and 330 healthy (control) individuals revealed no significant difference in the frequency of the C allele at the c.1150 position (p = 0.34). One patient carrying the combination `pathogenic mutation plus the allelic variant c.1150T > C' had a typical achondroplasia (Ach) phenotype. In addition, three other patients with atypical phenotypes showed no association with the allelic variant. Together, these results do not support the hypothesis of a modulatory role for the c.1150T > C change in the FGFR3 gene.

The objective of the study was to illustrate the applicability and significance of the novel Lewis urothelial cancer model compared to the classic Fisher 344. Fischer 344 and Lewis females rats, 7weeks old, were intravesical instilled N-methyl-N-nitrosourea 1.5mg/kg every other week for a total of four doses. After 15weeks, animals were sacrificed and bladders analyzed: histopathology (tumor grade and stage), immunohistochemistry (apoptotic and proliferative indices) and blotting (Toll-like receptor 2-TLR2, Uroplakin III-UP III and C-Myc). Control groups received placebo. There were macroscopic neoplastic lesions in 20% of Lewis strain and 70% of Fischer 344 strain. Lewis showed hyperplasia in 50% of animals, normal bladders in 50%. All Fischer 344 had lesions, 20% papillary hyperplasia, 30% dysplasia, 40% neoplasia and 10% squamous metaplasia. Proliferative and apoptotic indices were significantly lower in the Lewis strain (p<0.01). The TLR2 and UP III protein levels were significantly higher in Lewis compared to Fischer 344 strain (70.8 and 46.5% vs. 49.5 and 16.9%, respectively). In contrast, C-Myc protein levels were significantly higher in Fischer 344 (22.5%) compared to Lewis strain (13.7%). The innovative Lewis carcinogen resistance urothelial model represents a new strategy for translational research. Preservation of TLR2 and UP III defense mechanisms might drive diverse urothelial phenotypes during carcinogenesis in differently susceptible individuals.

The efficacy of the human papillomavirus type 16 (HPV-16)/HPV-18 AS04-adjuvanted vaccine against cervical infections with HPV in the Papilloma Trial against Cancer in Young Adults (PATRICIA) was evaluated using a combination of the broad-spectrum L1-based SPF10 PCR-DNA enzyme immunoassay (DEIA)/line probe assay (LiPA25) system with type-specific PCRs for HPV-16 and -18. Broad-spectrum PCR assays may underestimate the presence of HPV genotypes present at relatively low concentrations in multiple infections, due to competition between genotypes. Therefore, samples were retrospectively reanalyzed using a testing algorithm incorporating the SPF10 PCR-DEIA/LiPA25 plus a novel E6-based multiplex type-specific PCR and reverse hybridization assay (MPTS12 RHA), which permits detection of a panel of nine oncogenic HPV genotypes (types 16, 18, 31, 33, 35, 45, 52, 58, and 59). For the vaccine against HPV types 16 and 18, there was no major impact on estimates of vaccine efficacy (VE) for incident or 6-month or 12-month persistent infections when the MPTS12 RHA was included in the testing algorithm versus estimates with the protocol-specified algorithm. However, the alternative testing algorithm showed greater sensitivity than the protocol-specified algorithm for detection of some nonvaccine oncogenic HPV types. More cases were gained in the control group than in the vaccine group, leading to higher point estimates of VE for 6-month and 12-month persistent infections for the nonvaccine oncogenic types included in the MPTS12 RHA assay (types 31, 33, 35, 45, 52, 58, and 59). This post hoc analysis indicates that the per-protocol testing algorithm used in PATRICIA underestimated the VE against some nonvaccine oncogenic HPV types and that the choice of the HPV DNA testing methodology is important for the evaluation of VE in clinical trials. Atmospheric carbon dioxide records indicate that the land surface has acted as a strong global carbon sink over recent decades, with a substantial fraction of this sink probably located in the tropics, particularly in the Amazon. Nevertheless, it is unclear how the terrestrial carbon sink will evolve as climate and atmospheric composition continue to change. Here we analyse the historical evolution of the biomass dynamics of the Amazon rainforest over three decades using a distributed network of 321 plots. While this analysis confirms that Amazon forests have acted as a long-term net biomass sink, we find a long-term decreasing trend of carbon accumulation. Rates of net increase in above-ground biomass declined by one-third during the past decade compared to the 1990s. This is a consequence of growth rate increases levelling off recently, while biomass mortality persistently increased throughout, leading to a shortening of carbon residence times. Potential drivers for the mortality increase include greater climate variability, and feedbacks of faster growth on mortality, resulting in shortened tree longevity. The observed decline of the Amazon sink diverges markedly from the recent increase in terrestrial carbon uptake at the global scale, and is contrary to expectations based on models.

The p23 protein is a chaperone widely involved in protein homeostasis, well known as an Hsp90 co-chaperone since it also controls the Hsp90 chaperone cycle. Human p23 includes a -sheet domain, responsible for interacting with Hsp90; and a charged C-terminal region whose function is not clear, but seems to be natively unfolded. p23 can undergo caspase-dependent proteolytic cleavage to form p19 (p231-142), which is involved in apoptosis, while p23 has anti-apoptotic activity. To better elucidate the function of the human p23 C-terminal region, we studied comparatively the full-length human p23 and three C-terminal truncation mutants: p23; p23 and p23. Our data indicate that p23 and p19 have distinct characteristics, whereas the other two truncations behave similarly, with some differences to p23 and p19. We found that part of the C-terminal region can fold in an -helix conformation and slightly contributes to p23 thermal-stability, suggesting that the C-terminal interacts with the -sheet domain. As a whole, our results suggest that the C-terminal region of p23 is critical for its structure-function relationship. A mechanism where the human p23 C-terminal region behaves as an activation/inhibition module for different p23 activities is proposed.

Vitamin C stability and concentration was evaluated in isotonic beverages and B group vitamins (B1, B2, B3, B5 and B6) in power beverages. The amount of vitamins was found to be above of that declared on the labels, even after the shelf life had been exceeded. A small decrease in the amount of B group vitamins was observed during the shelf life of the products. In the case of vitamin C this decrease was slightly higher. The present research shows the need of increased quality control and inspection.

The field of chiral catalysis has experienced explosive growth over the last two decades. By now, many of the classical reactions in organic synthesis can be carried out efficiently in asymmetric manner. As one of the fundamental and powerful C-C bond-forming reactions, enantioselective catalytic allylation (ECA) and crotylation (ECC) of aldehydes has attracted considerable attention. In this article, we present an overview about the importance of chiral Lewis acids and bases in catalytic enantioselective addition of allyl- and crotyl metals to aldehydes and the application of this methodology in the total synthesis of natural and non-natural products.
