A presença de microssatélites no genoma de Echinococcus granulosus foi verificada utilizando-se oito oligonucleotídeos com repetições como sondas (GT15, CT15, AT15, CG15, CAT10, CAA10, CGG10 e CATA10). Experimentos de hibridização revelaram que as repetições GT, CAA, CATA e CT são as mais frequentes no genoma de E. granulosus. As sondas AT e GC não apresentaram sinais de hibridização. Seis lócus contendo repetições CA/GT, quatro lócus contendo repetições GA/CT e oito lócus contendo repetições AAC/GGT foram clonados e sequenciados. O lócus Egmsca1 foi analisado em 73 isolados do Brasil e da Argentina cujas linhagens haviam sido previamente caracterizadas e em 27 isolados provenientes da Etiópia cujas linhagens ainda não foram identificadas. Os isolados brasileiros da linhagem bovina e os isolados argentinos da linhagem do camelo apresentaram-se monomórficos e compartilharam o alelo (CA)7. Isolados argentinos das linhagens da ovelha e da ovelha da Tasmânia compartilharam dois alelos [(CA)8 e (CA)10] com os isolados brasileiros da linhagem da ovelha. O alelo (CA)11 foi encontrado somente em isolados brasileiros da linhagem da ovelha em uma baixa frequência. O alelo (CA)9 ocorreu apenas em um isolado da Etiópia. As populações brasileira e argentina da linhagem da ovelha foram testadas em relação ao equilíbrio de Hardy-Weinberg e somente a primeira estava de acordo com as expectativas. Protoescólices isolados de um único cisto hidático não apresentaram polimorfismo, validando a utilização de protoescólices de um mesmo cisto, agrupados como um isolado, em estudos populacionais. Um microssatélite contendo repetições de pentanucleotídeos, contido no complexo gênico do snRNA U1, também foi isolado. Este estudo descreve pela primeira vez o isolamento e caracterização de microssatélites em E. granulosus.

O gado zebu (Bos indicus) é predominante em todo o mundo. Atualmente, no Brasil, o rebanho de bovinos e zebuínos é composto por aproximadamente 170 milhões de indivíduos, no qual, 80% são zebuínos ou animais de raças cruzadas com zebu. O principal objetivo deste estudo foi determinar a possibilidade da utilização de marcadores moleculares de bovinos na identificação genética de zebuínos. A confirmação desta possibilidade seria de extrema valia, já que a quantidade de informações sobre o genoma zebuíno é limitada e metodologias específicas de identificação de marcadores genéticos para raças zebuínas são raras. Assim, o DNA de 65 zebus, de 4 raças diferentes (Gir, Tabapuã, Nelore e Brahman), foi extraído a partir de sangue aplicado em papel filtro, utilizando o “kit” DNA IQTM SYSTEM (Promega®). Após a extração de DNA, foi feito um PCR Multiplex utilizando o “kit” StockMarks® (Applied Biosystems®), o qual amplifica um total de 11 loci (BM1824, BM2113, ETH10, ETH225, ETH3, INRA023, SPS115, TGLA122, TGLA126, TGLA227 e TGLA53). Os fragmentos gerados pela PCR foram analisados por eletroforese capilar utilizando o analisador genético ABI Prism 3100 (Applied Biosystems®) e o programa GeneScan® v.3.7 (Applied Biosystems®). A freqüência dos marcadores e os índices de variabilidade genética foram calculados utilizando o programa Microsatellite Toolkit v.3.1 e o equilíbrio de Hardy-Weinberg foi determinado através do programa GENEPOP v.3.4. Através destas técnicas foi possível demonstrar que os marcadores moleculares utilizados para identificação genética de bovinos podem também ser utilizados para a realização da técnica de identificação genética de zebuínos Além disso, foi possível demonstrar a freqüência alélica na população de zebuínos analisada como um todo, assim como foi determinada a freqüência alélica para os diferentes marcadores moleculares dentro das 4 raças estudas. Considerando o total de 100 diferentes alelos identificados entre os 11 STR (Short Tandem Repeats) analisados, foi possível observar que a heterozigosidade esperada (He) foi relativamente alta na população analisada, assim como na análise por raça. Estes dados indicam alto grau de diversidade genética nas diferentes raças. Neste sentido, a raça Brahman apresentou a menor diversidade (0,60) e a raça Tabapuã, a maior (0,69). Contudo, estudos complementares são necessários, a fim de confirmar as freqüências alélicas estabelecidas, uma vez que os resultados encontrados no presente trabalho referem-se a um número relativamente pequeno de zebuínos, quando comparados com o total de zebus encontrados no rebanho brasileiro.

O objetivo deste trabalho é entender o processo de institucionalização do curso de graduação em Administração de Empresas da Faculdade de Economia, Administração e Contabilidade da Universidade de São Paulo (FEA-USP), tendo como referencial teórico o modelo de Barley e Tolbert (1997). Explorando aspectos históricos para tentar explicar o “espírito do tempo” (Zeitgeist) e ilustrar o processo de surgimento de uma nova graduação para a USP, a metodologia do estudo de caso (YIN, 2001) foi a escolhida por permitir uma maior gama de recursos de pesquisa. Dessa forma, o trabalho analisou as Atas de Reunião da Congregação da época (1946-1965), entrevistas colhidas com quinze pessoas entre protagonistas e observadores da história desta instituição durante o período analisado, jornais publicados pelos próprios alunos da Faculdade e bibliografia sobre o assunto. Utilizando-se de conceitos presentes em Giddens (1979, 1986), Machado-da-Silva, Fonseca e Crubellate (2005), DiMaggio e Powell (1983), Hardy e Maguire (2008), Khurana (2007), entre outros autores que trabalham com a Teoria Institucional, o processo de institucionalização do curso de Administração de Empresas na FEA-USP é analisado, relacionando-se a literatura disponível com os fatos encontrados em documentos e depoimentos colhidos. Alguns acontecimentos chamam a atenção, como a demora em mais de uma década para a fundação da FEA dentro da USP, a ligação da cadeira de Ciência da Administração com o Instituto de Administração (IA), que teve sua gênese a partir do Departamento do Serviço Público (DSP), órgão ligado ao Governo do Estado de São Paulo. Singularidades à parte, houve pessoas e situações necessárias para que o curso surgisse na USP apenas a partir da década de 1960. Nesse sentido, os papéis desempenhados pelo Prof. Antônio Delfim Netto e pelo Prof. Ruy Aguiar da Silva Leme são explorados com maior detalhamento, tendo em vista o grande impacto que suas decisões tiveram no processo de criação e estabelecimento da carreira de administração. De fato, tem-se que as instituições, com ênfase nesta pesquisa sobre a FEA-USP, são locais propícios para o estudo da mudança e que os atores sociais através de suas ações empreendidas são capazes de gerar modificação ou reprodução do status quo dentro destes estabelecimentos. As instituições constituem possibilidades para aqueles que a compõem ao mesmo tempo em que constrangem ações e são necessárias na sociedade tal qual como é apresentada na modernidade.

A indústria criativa de moda está sendo impactada pela ação de consumidores que utilizam novos meios de comunicação como forma de interação, tais como blogs, Instagram e Twitter. Os consumidores que, nesse contexto, assumiram o papel de empreendedores, não estavam insatisfeitos com o mercado, apenas queriam compartilhar suas ideias e aquisições com outros usuários da rede. Com o decorrer do tempo, no entanto, essa ação, inicialmente despretensiosa, alcançou popularidade e captou a atenção de leitores e de empresas do setor, ganhando escala (DIAMANDIS & KOTLER, 2015) e visibilidade. Sendo assim, a pesquisa terá como objetivo identificar e analisar as estratégias discursivas que conduziram consumidores, que apenas desejavam se comunicar e trocar suas ideias em torno de um tema que lhes interessava, à categoria de empreendedores institucionais (ANSARI & PHILLIPS, 2011; DOLBEC & FISCHER, 2015). A pergunta de pesquisa respondida foi: Quais são as estratégias discursivas utilizadas por consumidores que se tornaram empreendedores institucionais? Para tal foi realizado um estudo exploratório, na forma de estudo de caso, com a blogger Camila Coutinho, do blog Garotas Estúpidas. Foram analisadas as estratégias discursivas da empreendedora, compiladas principalmente por meio de postagens do blog, materiais divulgados na mídia e entrevistas. Os dados foram analisados pelo método de análise de discurso (VAN DIJK, 1997; PHILLIPS & HARDY, 2002). Esta pesquisa contribui para a teoria de empreendedorismo institucional ao identificar e analisar as estratégias discursivas usadas por um empreendedor institucional. Além disso, contribui para a prática gerencial ao identificar as mudanças geradas por empreendedores no campo de mídia virtual.

The aim of this thesis was to evaluate historical change of the landscape of Madeira Island and to assess spatial and temporal vegetation dynamics. In current research diverse “retrospective techniques”, such as landscape repeat photography, dendrochronology, and research of historical records were used. These, combined with vegetation relevés, aimed to gather information about landscape change, disturbance history, and vegetation successional patterns. It was found that landscape change, throughout 125 years, was higher in the last five decades manly driven by farming abandonment, building growth and exotic vegetation coverage increase. Pristine vegetation was greatly destroyed since early settlement and by the end of the nineteenth century native vegetation was highly devastated due to recurrent antropogenic disturbances. These actions also helped to block plant succession and to modify floristical assemblages, affecting as well as species richness. In places with less hemeroby, although significant growth of vegetation of lower seral stages was detected, the vegetation of most mature stages headed towards unbalance between recovery and loss, being also very vulnerable to exotic species encroachment. Recovery by native vegetation also occurred in areas formerly occupied by exotic plants and agriculture but it was almost negligible. Vegetation recovery followed the successional model currently proposed, attesting the model itself. Yet, succession was slower than espected, due to lack of favourable conditions and to recurrent disturbances. Probable tempus of each seral stage was obtained by growth rates of woody taxa estimated through dendrochronology. The exotic trees which were the dominant trees in the past (Castanea sativa and Pinus pinaster) almost vanished. Eucalyptus globulus, the current main tree of the exotic forest is being replaced by other cover types as Acacia mearnsii. The latter, along with Arundo donax, Cytisus scoparius and Pittosporum undulatum are currently the exotic species with higher invasive behaviour. However, many other exotic species have also proved to be highly pervasive and came together with the ones referred above to prevent native vegetation regeneration, to diminish biological diversity, and to block early successional phases delaying native forest recovery.

Sulfated polysaccharides (PS) are biomolecules with a great biotechnological potential. There are few data about PS from high plants. In addition, pharmacological activities of PS from plants have not been carrying out. The aim of this work was extract PS from the angiosperm Halodule wrightii and study their anticoagulant and antioxidant activities. Histological analysis showed the presence of the PS manly in the roots. A polysaccharide-rich extract was obtained from H. wrightii by proteolysis followed by methanol and TCA precipitation. Chemical, infra-red analysis and agarose gel electrophoresis in 1.3 diaminopropane acetate buffer confirmed the presence of sulfated polysaccharides made by glucose, galactose, xylose and sulfate residues in the proportion 1: 0,9: 1: 1. In addition polyacrilamide electrophoresis have shown that extract is mainly compose by 11kDa sulfated polysaccharides. Pharmacological analysis have shown total antioxidant capacity (CAT) that resulted in 15,21 μg for equivalent of ascorbic acid, scavenging activity of the DPPH radical with 41,36 % of scavenging, activity of reducing power with the maximum of 0,290 nm (50 % of vitamin C activity) and scavenging activity superoxide radical (O2-) with a maximum of 32,23 %. Chelating activity of metal less than 4% and scavenging activity of the radical hydroxyl (OH-) less than 2%. Time of activated partial tromboplastin (aPTT) doubling the time of coagulation from 20μg of and protrombin time (PT) was not present. The data indicate that PS from Halodule wrightii could be considered for future applications in medicine, food production or cosmetic industry

Physiological changes induced by the aging process is dynamic and progressive, reducing the adaptability and independence of older people and may be influenced by genetic and environmental factors. Thus the aim of this thesis was to investigate the association between polymorphism of the ACE gene ID and the phenotypes of muscular strength and blood pressure of 62 elderly Brazilian (67.35 ± 5.66 years) during a 16-week program of supervised training. The elderly women were stratified by age, with the group 1 (G1, n = 34) <70 years and group 2 (G2 n = 28) ≥ 70 years, and in three groups by ACE, ACE-II (n = 8) ACE- DD (n = 35) and ACE-ID (n = 19). The level of muscle strength was evaluated by the method of maximum repetitions and measures of blood pressure (BP) were measured before and after training (PAPré1 and PAPós1) and before and after each training session (PAPre2 and PAPós2), in place of training. DNA samples were isolated from peripheral blood leukocytes polymorphism and insertion / deletion (ID) of the ACE gene (rs1800795) was genotyped by polymerase chain reaction (PCR) plus PCR-confirmatory. The genotype distribution of the polymorphism ID attended the prerogatives of Hardy-Weitíherg. There was variation in power levels before and after training and the age between groups (t-test) and the ACE polymorphism (ANOVA) (p <0.05). Depending on the results it was concluded that resistance training helps to reduce SBP and increased muscle strength of upper and lower limbs when considering the age and ACE polymorphism. In this study the Elderly carriers of the D allele were more reactive to changes in BP resistance training. This study was multidisciplinary project involving researchers in the areas Medical, Physical Education, Pharmacy, Nutrition, Gerontology and Statistics. This fulfilled the requirements of the multidisciplinary Graduate Program in Health Sciences

Human population have a significant number of polymorphic loci, whose use and applications range from construction of linkage maps, to study the evolution of populations, through the determination of paternity, forensic medicine and migration. Currently, STRs (Short Tanden Repeats) markers are considered the major markers for human identification, mainly due to its abundance and high variability because of the fact that they are easily amplifiable by PCR (Polymerase Chain Reaction), work with low amounts of DNA and be capable of automation processes involving fluorescence detection. The creation of regional databases containing allele frequencies of population provide subsidies to increase the reliability of the results of determining the genetic link. This paper aims to obtain a database of allele frequencies of 15 polymorphic molecular loci (D8S1179, D21S11, D7S820, CSF1PO, D19S433, vWA, TPOX, D18S51, D3S1358, TH01, D13S317, D16S539, D2S1338, D5S818 e FGA) in a population classifies as born in the State of Rio Grande do Norte, Brazil, totaling 1100 unrelated individuals. To evaluate the frequency, DNA samples were submitted to PCR amplification, followed by capilarry electrophoresis genetic sequencer. The frequencies identified in this study were compared with brazilian population in general and other states in Brazil. Except for the loci D21S11, D19S433 and D2D1338, the genotypes found were in Hardy-Weinberg equilibrium and no significant differences among the frequencies were found in the populations studied. The most informative loci was D2S1338 and D18S51, and the less informative is the locus TPOX

The Nile tilapia, Oreochromis niloticus, is an important omnivorous fish in the reservoirs of the semi-arid region of Brazil. Throughout its growth tilapia s feeding behavior changes from a visual predator of zooplankton to a filter-feeder, collecting suspended particulate matter, including planktonic organisms, through pumping. This feature results in different impacts of tilapia on plankton community as the fish grows. Aiming to quantify the functional response of different sizes of Nile tilapia on zooplankton experiments in microcosms scale in the laboratory and in mesocosm scale in the field were carried out. The data were fitted to four different models of functional response. The best fits were obtained for nonlinear models in laboratory experiments. While the experiments in mesocosms were the best settings for responses of type I (juvenile and adult tilapia) and type III (fry). The Manly's alpha index was used to evaluate the feeding selectivity of tilapia on the three main groups of the zooplankton in the experiments in mesocosms. The results show that: (i) rotifers were the preferred prey of fingerlings,(ii) copepods were rejected by fry and juvenile tilapia and (iii) adult fish fed non-selectively on copepods, cladocerans and rotifers. The functional response models obtained in this research can be applied to population models and help in modeling the dynamics of interactions between Nile tilapia and the planktonic communities in the reservoirs of the semi-arid

The Bola-DRB3 gene participates in the development of the immune response and is highly polymorphic. For these reasons, it has been a candidate gene in studies of the genetic basis of disease resistance and in population genetic analysis. South American native cattle breeds have been widely replaced by improved exotic breeds leading to a loss of genetic resources. In particular South American native breeds have high levels of fertility and disease resistance. This work describes genetic variability in the BoLA-DRB3 gene in native (Caracu, Pantaneiro, Argentinean Creole) and exotic (Holstein, Jersey, Nelore, Gir) cattle breeds in Brazil and Argentina. PCR-RFLP alleles were identified by combining the restriction patterns for the BoLA-DRB3.2 locus obtained with RsaI, BstY, and HaeIII restriction enzymes. Allelic frequencies and deviations from the Hardy-Weinberg equilibrium were also calculated. Analysis of the 24 BoLA-DRB3 PCR-RFLP alleles identified showed differences in the allele distributions among breeds.

We describe the isolation and characterization of ten microsatellite loci from the red-winged tinamou (Rhynchotus rufescens) and also evaluated the cross-amplification of these loci and other ten loci previously developed for the great tinamou (Tinamus major) in other tinamous. Genetic variability was assessed using 24 individuals. Six loci were polymorphic with moderate to high number of alleles per locus (2-12 alleles) and showed expected heterozygosity (HE) ranging from 0.267 to 0.860. All loci conformed to the Hardy-Weinberg expectation and linkage disequilibrium was not significant for any pair of loci. This battery of polymorphic loci showed high paternity exclusion probability (0.986) and low genetic identity probability (4.95 x 10(-5)), proving to be helpful for parentage tests and population analyses in the red-winged tinamou. The cross-amplification was moderate where of the 160 locus/taxon combinations, 46 (28.75%) successfully amplified.

As data on X-chromosomal short tandem repeats (X-STRs) for the Brazilian population are scarse, the aim of this study was to determine the allele frequencies of five X-STRs (DXS6854, DXS7424, DXS101, DXS6808 and DXS7132) in the São Paulo State, Brazil. No deviations from the Hardy-Weinberg equilibrium were observed, with the exception of DXS101. The forensic efficiency parameters demonstrated that DXS101 was the most informative marker. Population comparisons revealed that the X-STR profile sampled in the state of São Paulo was more similar to European and African populations than Asiatic populations reported in this work.

The objective of this study was to identify DNA polymorphisms at the genes leptin, β-lactoglobulin and pituitary-specific transcription factor in three genetic groups of Holstein x Guzerat dairy cows and investigate the relationship between their genotypes and the composition and quality of milk of dairy cows. Samples were collected in August 2009, being 113 blood samples from lactating crossbred cows and 58 milk samples. For analysis of DNA polymorphisms blood samples were collected, analyzed later in the Genetic Laboratory affiliated to the Zootechny Institute of São Paulo and individual milk samples were collected according to standards established by the laboratory of Management Program of Northeast Dairy Herds (PROGEN), at Federal Rural University of Pernambuco (UFRPE) for analysis of milk composition and quality. The characterization of genotypes was performed by PCR-RFLP, for which were designed specific primers for each studied gene and restriction enzymes Kpn2I, HaeIII and HinfI that cut the DNA of the following genes: leptin, β-lactoglobulin and a PIT, respectively. The leptin estimate genotypic frequence were CC 0.112, TT 0.225 and CT 0.661, for β-lactoglobulin were AA 0.136, AB 0.323 and BB 0.539, and for PIT were ++ 0.655, -- 0.311 and +- 0.032. The results show that the population is in Hardy-Weinberg disequilibrium for leptin, β-lactoglobulin and a PIT due to excess of heterozygotes in the population, however, as these genes are associated with the milk production it is considered that the animals have genetic potential for milk production in the Brazilian semi-arid conditions. Through the characterization of the studied herd there were not found implications of the polymorphism of leptin, β-lactoglobulin and PIT in the composition and quality of milk from cows in the different genetic groups 1/2, 3/4 and 7/8 Holstein x Guzerat.
