Pigeon pea ( Cajanus cajan (L.) Millsp.) is a drought tolerant pulse legume, mainly grown for grain in the semi-arid tropics, particularly in Africa. Pigeon pea production in countries like Kenya is faced with a number of challenges, particularly lack of high quality seeds. The objective of this study was to develop an in vitro regeneration system for pigeon pea varieties grown in Kenya, that is amenable to genetic transformation. In vitro regeneration of pigeon pea varieties, KAT 60/8 and ICEAP 00557, commonly grown in Kenya was achieved using leaf explants from in vitro grown seedlings, through callus initiation, followed by shoot and root induction. For callus initiation, MS media supplemented with 0.5-4 mg l-1 2, 4-D and TDZ separately were tested, and IBA at 0.1, 0.5 and 1 mg l-1 was tested for rooting of shoots. Embryogenic calli was obtained on MS containing 2, 4- D; whereas TDZ induced non-embryogenic callus alone or with shoots directly on explants. Indirect shoot regeneration frequency of 6.7 % was achieved using 1 mg l-1 2, 4-D-induced embryogenic callus obtained using KAT 60/8 explants. Whereas direct shoot regeneration frequencies of 20 and 16.7% were achieved using ICEAP 00557 and KAT 60/8 explants, using 0.5 mg l-1 and 2 mg l-1 TDZ, respectively. Optimum rooting was achieved using 0.5 mg l-1 IBA; and up to 92% rooted shoots were successfully established in soil after acclimatisation. Genotype and hormone concentrations had a significant (P<0.05) influence on callus, shoot and root induction. The protocol developed can be optimised for mass production and genetic transformation of KAT 60/8 variety.

Mushrooms are an important source of natural compounds with acknowledged bioactivity. Pleurotus eryngii (DC.) Quél., in particular, is widely recognized for its organoleptic quality and favorable health effects, being commercially produced in great extent. On the other hand, Suillus bellinii (Inzenga) Watling is an ectomycorrhizal symbiont, whose main properties were only reported in a scarce number of publications. Some current trends point toward using the mycelia and the culture media as potential sources of bioactive compounds, in addition to the fruiting bodies. Accordingly, P. eryngii and S. bellinii were studied for their composition in phenolic acids and sterols, antioxidant capacity (scavenging DPPH radicals, reducing power, β-carotene bleaching inhibition and TBARS formation inhibition), anti-inflammatory effect (by down-regulating LPS-stimulated NO in RAW264.7 cells) and anti-proliferative activity (using MCF-7, NCI-H460, HeLa, HepG2 and PLP2 cell lines). Overall, S. bellinii mycelia showed higher contents of ergosterol and phenolic compounds (which were also detected in higher quantity in its fruiting body) and stronger antioxidant activity than P. eryngii. On the other hand, P. eryngii mycelia showed anti-inflammatory (absent in S. bellinii mycelia) and a cytotoxicity similar (sometimes superior) to its fruiting bodies, in opposition to S. bellinii, whose mycelia presented a decreased anti-proliferative activity. Furthermore, the assayed species showed differences in the growth rate and yielded biomass of their mycelia, which should also be considered in further applications.

In vitro selection is one of the most effective and efficient techniques for plant improvement. This is due to its ability to isolate plants with the desired character(s), either by applying a selection agent on the culture media to drive the selection of somaclones with the required character(s), or by establishing particular conditions that change in the genomes of somaclones toward the required character. The objective of this study was to identify a suitable protocol for in vitro selection of Allium white rot disease ( Sclerotium cepivorum ) tolerance in commercial Egyptian onion varieties, namely Giza 20, Giza 6 and Beheri Red. Oxalic acid (OA), the phytotoxin produced by Sclerotium cepivorum, was used as the selective agent. Seeds of the three Egyptian varieties were germinated on four concentrations (0.0, 0.02, 0.2, 2 and 20 mM) of Oxalic acid. Among the tested cultivars, Beheri Red had the highest germination frequency (52%) at all concentrations tested, followed by Giza 20 (42.6%), and Giza 6 at (32%). Cotyledon explants from the varieties were cultured on toxic MSBDK medium, supplemented with 0, 3, 6 and 12 mM OA. The survival of calli on MSBDK free toxic medium was 70.7% for all tested cultivars; however, MSBDK-stressed medium, with 3 mM OA reduced the viable calli to 42.1%. The highest OA concentration (12 mM) completely inhibited calli induction from cotyledons explants. A medium supplement with 3 mM OA retarded 80% of calli growth. Among 156 tested calli of Beheri Red, only 23 calli (14.7%) survived on toxic medium for 45 days. Similarly, there was 15.6% survival for Giza 20 calli, while 40.1% of the Giza 6 calli survived. Plantlets were regenerated from surviving calli and transplanted onto ex vitro, and formed bulb after acclimatisation.

Processed meat products are of worldwide importance and, because of their intrinsic factors as well as the processing methods, they are highly prone to fungal and mycotoxin contamination. Ochratoxin A (OTA) is the most significant mycotoxin in processed meat products. Penicillium nordicum is considered to be responsible for OTA contamination of meat products, as it is highly adapted to salt and protein-rich matrices and is moderately psycrotrophic. However, another OTA-producing fungus, Aspergillus westerdijkiae, adapted to carbon-rich matrices such as cereals and coffee beans, has been recently associated with high levels of OTA in meat products. Several Lactic Acid Bacteria (LAB) and yeasts have been tested as biocontrol agents against P. nordicum growth and OTA production in meat products, with promising results, but none of the studies have considered A. westerdijkiae. The aim of this work was to evaluate in vitro the effect of a commercial starter culture used in sausage fermentation and four yeasts isolated from dry-cured sausage on these two OTA-producing fungi, both in terms of fungal growth and of OTA production, using different meat-based culture media as model systems. The mechanisms underlying the observed effect were also studied. For this purpose, C. krusei, C. zeylanoides, R. mucilaginosa, R. glutinis, a mix of these yeasts and the starter culture were co-inoculated with P. nordicum and A. westerdijkiae in industrial sausage, traditional sausage, and ham-based media, under conditions of water activity, salt concentration and temperature that mimic real conditions at beginning and end of sausage curing process. Fungal growth was determined by measuring colony diameter, and OTA production was quantified by HPLC-FLD after extraction with methanol. Yeasts where found to inhibit significantly the growth of both fungi. P. nordicum was unable to produce detectable OTA in both sausage-based media under any condition. In ham, yeasts reduced OTA production, while the starter culture significantly increased it. Unexpectedly, OTA production by A. westerdijkiae was significantly stimulated in all media tested by all microorganisms. Matrix has a significant effect on OTA production by P. nordicum, but not by A. westerdijkiae, for which only temperature showed to have effect. By testing the mechanisms of action by which starter culture and C. zeylanoides influenced fungal responses, we were able to determine that direct contact and simultaneous growth of test organisms were the mechanisms more significantly involved in the responses. In conclusion, ochratoxigenic fungi do not all respond to antagonistic microorganisms in the same way. The use of biocontrol agents with the intent of reducing fungal growth and mycotoxin production by one fungus can have unexpected effects on others, thus leading to unforeseen safety problems. Further experiments are recommended to properly understand the reasons behind the different effects of microorganisms, to ensure their safe as biocontrol agents.

Este estudo envolve o controlo e a optimização das condições de culturas dos microrganismos: Saccharomyces cerevisiae CCMI 396, S. cerevisiae v. lab., Aspergillus oryzae CCMI 125, Aspergillus japonicus CCMI 443, Fusarium oxysporum CCMI 866, Aspergillus niger CCMI 296 com vista à produção de oligossacáridos. Determinaram-se os parâmetros característicos das culturas de duas diferentes estirpes de Saccharomyces com diferentes fontes de carbono e em diferentes condições ambientais. O perfil de crescimento da S. cerevisiae CCMI 396 foi semelhante nos diferentes meios de cultura estudados, sendo a velocidade específica de crescimento mais elevada no meio com glucose a pH 5 e a 30°C (0,36h-1). A S. cerevisiae v. lab. Teve velocidade específica de crescimento idêntica nas mesmas condições da outra estirpe, no entanto, o perfil de crescimento foi diferente nos outros meios de cultura. Estudou-se o efeito da adição de sumo de laranja ou de tomate ao meio de cultura com sacarose e avaliou-se a evolução glucídica no meio de cultura durante o ensaio por HPLC com detector RI. Determinou-se a frutosiltransferase no sobrenadante e na fracção intracelular e determinou-se a evolução dos oligossacáridos. Numa segunda parte deste trabalho efectuaram-se culturas dos quatro fungos filamentosos com vista a avaliar a capacidade de produção, nomeadamente, de frutooligassacáridos. Os resultados mostraram que a espécie Aspergillus japonicus CCMI 443 originou, nas mesmas condições de cultura, valores superiores, sendo a percentagem de produção FOStotais/GluCtotais de 61% para as enzimas intracelulares e 40% para as enzimas no sobrenadante. ABSTRACT; This study involves control and optimization of the cultures of microorganisms: Saccharomyces cerevisiae CCMI 396, S. cerevisiae v. lab., Aspergillus oryzae CCMI 125, Aspergillus japonicus CCMI 443, Fusarium oxysporum CCMI 866, Aspergillus níger CCMI 296 for oligosaccharides production. Were determined the parameters characteristic of the cultures of two different strains of Saccharomyces with different sources of carbon and in different environmental conditions. The growth profile of S. cerevisiae CCMI 396 was similar in different cultures media, but the highest specific growth was obtained in a medium with glucose, pH 5, at 30°C (0.36h-1). S. cerevisiae v. lab. had similar growth profile in a medium with glucose but with others culture media was different. We studied the effect of adding orange juice or tomato to the culture medium with sucrose and evaluated the evolution glucidic in the culture medium during the test by HPLC with RI detector. Fructosyltransferase was determined in the extracellular and the intracellular fractions and determined the evolution of oligosaccharides. ln the second part of this work were carried out cultures of four filamentous fungi in order to assess production capacity, in particular, fructoligosaccharides. The results showed that the specie Aspergillus japonicus CCMI 443 originated in the same culture conditions, higher values and the percentage of production FOStotal/Guctotal of 61% for intracellular enzymes and 40% for extracellular enzymes.

The acquisition of oligosaccharides from chitosan has been the subject of several studies in the pharmaceutical, biochemical, food and medical due to functional properties of these compounds. This study aimed to boost its production of chitooligosaccharides (COS) through the optimization of production and characterization of chitosanolytic enzymes secreted by microorganisms Paenibacillus chitinolyticus and Paenibacillus ehimensis, and evaluating the antioxidant potential of the products obtained. In the process of optimizing the production of chitosanase were employed strategies Fractional Factorial Experimental Design and Central Composite Rotatable Design. The results identified the chitosan, peptone and yeast extract as the components that influenced the production of chitosanase by these microorganisms. With the optimization of the culture media was possible to obtain an increase of approximately 8.1 times (from 0.043 to 0.35 U.mL U.mL-1) and 7.6 times (from 0.08 U.mL-1 to 0.61 U.mL-1) in the enzymatic activity of chitosanase produced by P. chitinolyticus and P. ehimensis respectively. Enzyme complexes showed high stability in temperature ranges between 30º and 55º C and pH between 5.0 and 9.0. Has seen the share of organic solvents, divalent ions and other chemical agents on the activity of these enzymes, demonstrating high stability of these crude complexes and dependence of Mn2+. The COS generated showed the ability of DPPH radical scavenging activity, reaching a maximum rate of scavenging of 61% and 39% when they were produced with enzymes of P. ehimensis and P. chitinolyticus respectively. The use of these enzymes in raw form might facilitate its use for industrial applications

Wild mushrooms have been extensively studied for their value as sources of high quality nutrients and of powerful physiologically bioactive compounds [1,2]. The present study was designed to evaluate the in vitro development of two wild edible mushroom species: Pleurotus eryngii (DC.) Quél. and Suillus belinii (Inzenga) Watling, by testing different solid (Potato Dextrose Agar medium –PDA and Melin-Norkans medium- MMN) and liquid culture media (Potato dextrose broth- PDB and Melin-Norkans medium- MMN). Each strain of mushroom produces a special type of mycelium and this range of characteristics varies in form, color and growth rate. S. bellinii presents a pigmented and rhizomorphic mycelia, whereas, P. eryngii has depigmented and cottony mycelia. The mycelium isolated and grown in PDA showed a faster radial growth compared to the mycelium isolated and grown in both solid and liquid incomplete MMN medium. P. eryngii exhibited a rapid growth and a higher mycelia biomass in both medium compared to S. belinii. Moreover, the obtained mycelia will be characterized in terms of well-recognized bioactive compounds namely, phenolic acids and mycosterols (mainly ergosterol), by using high performance liquid chromatography coupled to diode array and ultraviolet detectors, respectively. These compounds will be correlated to mycelia bioactivity: i) antioxidant activity, evaluated through free radicals scavenging activity, reducing power and lipid peroxidation inhibition in vitro assays; ii) anti-inflammatory activity, assessed through nitric oxide production inhibition in murine macrophages (RAW 264.7 cell line); iii) cytotoxic activity, evaluated either in human tumor cell lines (MCF-7- breast adenocarcinoma, NCIH460- non-small cell lung cancer, HeLa- cervical carcinoma and HepG2- hepatocellular carcinoma) as also in a non-tumor porcine primary liver cells culture established in-house (PLP2). Overall, our expectation is that the bioactive formulations obtained by in vitro culture can be applied as nutraceuticals or incorporated in functional foods.

En este documento se estima una medida de la incertidumbre inflacionaria. Un modelo de inflación señala incertidumbre cuando los errores de pronóstico son heteroscedásticos. Por medio de la especificación de una ecuación GARCH (Generalized Autoregressive Conditional Heteroscedasticity), para la varianza del término de error de un modelo de inflación, es posible estimar una proxy de incertidumbre inflacionaria. La estimación simultánea del modelo de inflación y de la ecuación GARCH, produce un nuevo modelo de inflación en el cual los errores de pronóstico son homocedásticos. Existe consenso en la literatura económica en que hay una correlación positiva entre incertidumbre inflacionaria y la magnitud de la tasa de inflación, lo cual, como lo señaló Friedman (1977), representa uno de los costos asociados con la persistencia inflacionaria. Esto es porque tal incertidumbre dificulta la toma de decisiones óptimas por parte de los agentes económicos.La evidencia empírica, para el periodo 1954:01-2002:08, apoya la hipótesis de que para el caso de Costa Rica mientras mayor es la inflación mayor es la incertidumbre respecto a esta variable. En los últimos siete años (1997-2002) la incertidumbre presenta la variación media más baja de todo el periodo. Además, se identifica un efecto asimétrico de la inflación sobre la incertidumbre inflacionaria, es decir, la incertidumbre inflacionaria tiende a incrementarse más para el siguiente periodo cuando la inflación pronosticada está por debajo de la inflación actual, que cuando la inflación pronosticada está por arriba de la tasa observada de inflación. Estos resultados tienen una clara implicación para la política monetaria. Para minimizar la dificultad que la inflación causa en la toma óptima de decisiones de los agentes económicos es necesario perseguir no solamente un nivel bajo de inflación sino que también sea estable.AbstractThis paper estimates a measure of inflationary uncertainty. An inflation model signals uncertainty when the forecast errors are heteroskedastic. By the specification of a GARCH (Generalized Autoregressive Conditional Heteroscedasticity) equation, for the variance of the error term of the inflation model, it is possible to estimate a proxy for inflationary uncertainty. By the simultaneous estimation of the inflation model and the GARCH equation, a new inflation model is obtained in which the forecast errors are homocedastic. Most economists agree that there is a positive correlation between inflationary uncertainty and the magnitude of the inflation rate, which, as was pointed out by Friedman (1977), represents one of costs associated with the persistence of inflation. This is because such uncertainty clouds the decision-making process of consumers and investors.The empirical evidence for the period 1954:01-2002:08 confirms that in the case of Costa Rica inflationary uncertainty increases as inflation rises. In the last seven years(1997-2002) the uncertainty present the mean variation most small of the period. In addition, inflation has an asymmetric effect on inflationary uncertainty. That is, when the inflation forecast is below the actual inflation, inflationary uncertainty increases for the next period. The opposite happens when the inflation forecast is above the observed rate of inflation. Besides, the absolute value of the change on uncertainty is greater in the first case than the second. These results have a clear implication for monetary policy. To minimize the disruptions that inflation causes to the economic decision-making process, it is necessary to pursue, not only a low level of inflation, but a stable one as well.

Fungi of the genus Paracoccidioides are responsible for paracoccidioidomycosis. The occurrence of drug toxicity and relapse in this disease justify the development of new antifungal agents. Compounds extracted from fungal extract have showing antifungal activity. Extracts of 78 fungi isolated from rocks of the Atacama Desert were tested in a microdilution assay against Paracoccidioides brasiliensis Pb18. Approximately 18% (5) of the extracts showed minimum inhibitory concentration (MIC) values ≤ 125.0 μg/mL. Among these, extract from the fungus UFMGCB 8030 demonstrated the best results, with an MIC of 15.6 μg/mL. This isolate was identified as Aspergillus felis (by macro and micromorphologies, and internal transcribed spacer, β-tubulin, and ribosomal polymerase II gene analyses) and was grown in five different culture media and extracted with various solvents to optimise its antifungal activity. Potato dextrose agar culture and dichloromethane extraction resulted in an MIC of 1.9 μg/mL against P. brasiliensis and did not show cytotoxicity at the concentrations tested in normal mammalian cell (Vero). This extract was subjected to bioassay-guided fractionation using analytical C18RP-high-performance liquid chromatography (HPLC) and an antifungal assay using P. brasiliensis. Analysis of the active fractions by HPLC-high resolution mass spectrometry allowed us to identify the antifungal agents present in the A. felis extracts cytochalasins. These results reveal the potential of A. felis as a producer of bioactive compounds with antifungal activity.
Pros y contras percibidos del consumo de alcohol y consumo de alcohol en estudiantes de preparatoria

Propósito y Método del Estudio: El propósito fue conocer la relación que existe entre los pros (beneficios) y contras (barreras) percibidos del consumo de alcohol y el consumo de alcohol en estudiantes de una preparatoria técnica de una Universidad Pública ubicada en el Municipio de Guadalupe, Nuevo León. Para el sustento teórico se consideró el Modelo Transteórico ó de Etapas de Cambio (Prochaska y DiClemente, 1986). El diseño del estudio fue descriptivo correlacional. Se realizó un muestreo aleatorio estratificado con asignación proporcional al tamaño del estrato. El tamaño de la muestra se determinó en base a una prueba de correlación, considerando un nivel de significancia de 0.05, Coeficiente de Correlación por hipótesis alterna de .20 y una tasa de no respuesta del 25%, obteniéndose una muestra de n= 333 participantes. Contribución y conclusiones: El 71.8% de los estudiantes ha consumido alcohol alguna vez en la vida, el 55% durante los últimos doce meses y el 17.4% durante los últimos siete días. El 25.2% de los estudiantes presentó consumo sensato de alcohol, el 15.3% consumo dependiente y el 14.4% consumo dañino. La subescala de contras del consumo de alcohol ( = 23.2, DE= 5.1) presentó una media más alta que los pros ( = 12.6, DE= 4.8). Los pros presentaron relación positiva con el consumo de alcohol (AUDIT) (rs= .468, p= .001), consumo sensato (rs= .449, p= .001), consumo dependiente (rs= .345, p= .002) y consumo dañino (rs= .330, p= .001). Los contras presentaron relación negativa con el consumo de alcohol (AUDIT) (rs = - .278, p= .001), consumo sensato (rs= -.335, p= .001) y consumo dependiente (rs = - .222, p= .003). Se identificó que los hombres percibieron mayores pros ( = 13.68, Mdn= 12.00) [U= 9949.00, p=.001] y las mujeres percibieron mayores contras ( = 24.88, Mdn= 26.00) [U= 9123.00, p=.001] hacia el consumo de alcohol. También se encontró que los participantes que estudian y trabajan presentaron mayores pros ( = 15.11, Mdn= 14.00) [U= 5168, p=.001] y los que sólo estudian presentaron mayores contras ( = 23.47, Mdn= 25.00) [U= 6069.50, p=.001] hacia el consumo de alcohol. Además se identificó que el tipo de consumo de alcohol presentó diferencia por sexo, las mujeres reportaron mayor consumo sensato (57.7%) y dependiente (30.8%) y los hombres presentaron mayor consumo dañino de alcohol (37.1%).

Las orquídeas producen abundantes semillas pequeñas, careciendo de endospermo, cotiledones y sustancias de reserva para llevar a cabo su germinación. Es por esto que estratégicamente las semillas establecen una relación simbiótica con un hongo micorrízico que favorece a su germinación y desarrollo. El objetivo de este estudio fue determinar la especificidad del hongo micorrízico (Rhizoctonia sp.) en la germinación de cinco géneros de orquídeas. Se usaron dos medios de cultivo: 1) PhytamaxTM y 2) avena-agar+Rhizoctonia sp. Los resultados evaluados a los 45 y 75 días demuestran que no existe especificidad entre el hongo Rhizoctonia sp. y Trichoceros antenifer, especie de la cual se aisló el hongo. La germinación en tres de los cinco géneros evaluados fue mayor en el tratamiento avena-agar+Rhizoctonia sp., evidenciando también mayor tamaño y vigor las plantas que se desarrollaron en este tratamiento, aunque estos datos no fueron evaluados. Los resultados sugieren también que la planta hospedera del hongo inoculado podría tener una ventaja en cuanto al tiempo requerido para su germinación; sin embargo otras, especies también se ven favorecidas con el inóculo, aunque éste sea obtenido de otra fuente.

Two different culture media, namely CHU12 and inorganic fertilizer NPK (20-5-20) associated with macrophyte (Eichhornia crassipes) extract, were used to evaluate the development of A. gracilis. Growth rate, development, nutritional value and medium water quality were analyzed. A. gracilis in macrophyte+NPK medium had mean cell density (333x10(5) cells/mL) higher than medium CHU12 (302x10(5) cells/mL). Protein rate (12.52% PS), dry weight (397x10(7) pg/cell) and ashes (4.08x10(7) pg/cell) in A. gracilis was higher (p<0.05) than medium macrophyte+NPK. on the other hand, lipids, carbohydrates and fibers had the same rate (p>0.05) in both media. When hydrological variables in the culture medium of A. gracilis are taken into account, dissolved oxygen and free CO2 alone failed to have any significant difference (p>0.05) between the media employed. A. gracilis in the macrophyte+NPK medium had the same nutritional rate as the commercial medium CHU12, with excellent results for alga growth. Macrophyte medium may be used for large scale alga culture in the feed of fish larvae.

O sistema automatizado Bioscreen C foi utilizado para o crescimento de quatro linhagens de Mucor hiemalis, isoladas do solo da Estação Ecológica de Juréia-Itatins (EEJI), estado de São Paulo, em meios líquidos com uma única fonte de carbono (2%) ou de nitrogênio (1%), pH 5,0, a 25ºC, e agitação de 150rpm por 5 dias. O meio com somente uma única fonte de nitrogênio foi adicionado com 2% de glicose. As leituras de densidade óptica foram realizadas a 540nm, em intervalos de 2h, por cinco dias. Os resultados foram analisados estatisticamente com o Teste de Friedman (alfa = 5%). Os melhores crescimentos foram obtidos com as linhagens M1, M2 e M3, que atingiram o início da fase log em 60 horas de cultivo. As melhores fontes de carbono variaram de acordo com a linhagem estudada, e extrato de levedura provou ser a melhor fonte de nitrogênio para todas as linhagens. Acetato de sódio inibiu o crescimento das quatro linhagens, sendo a M3 a mais afetada. O uso do sistema automatizado foi muito conveniente para as culturas em meio liquido, sendo rápido e automático, constituindo em uma boa técnica para a determinação das condições ambientais ótimas para crescimento de fungos filamentosos.
