Reliable electronic systems, namely a set of reliable electronic devices connected to each other and working correctly together for the same functionality, represent an essential ingredient for the large-scale commercial implementation of any technological advancement. Microelectronics technologies and new powerful integrated circuits provide noticeable improvements in performance and cost-effectiveness, and allow introducing electronic systems in increasingly diversified contexts. On the other hand, opening of new fields of application leads to new, unexplored reliability issues. The development of semiconductor device and electrical models (such as the well known SPICE models) able to describe the electrical behavior of devices and circuits, is a useful means to simulate and analyze the functionality of new electronic architectures and new technologies. Moreover, it represents an effective way to point out the reliability issues due to the employment of advanced electronic systems in new application contexts. In this thesis modeling and design of both advanced reliable circuits for general-purpose applications and devices for energy efficiency are considered. More in details, the following activities have been carried out: first, reliability issues in terms of security of standard communication protocols in wireless sensor networks are discussed. A new communication protocol is introduced, allows increasing the network security. Second, a novel scheme for the on-die measurement of either clock jitter or process parameter variations is proposed. The developed scheme can be used for an evaluation of both jitter and process parameter variations at low costs. Then, reliability issues in the field of “energy scavenging systems” have been analyzed. An accurate analysis and modeling of the effects of faults affecting circuit for energy harvesting from mechanical vibrations is performed. Finally, the problem of modeling the electrical and thermal behavior of photovoltaic (PV) cells under hot-spot condition is addressed with the development of an electrical and thermal model.

In the present study of sponge-bacterial association, the presence of a marine bacterium which has not seen to be associated previously with the Mediterranean sponge Suberites domuncula was investigated. The marine sponge S. domuncula was chosen as the subject of investigation, for the identification of potential symbiotic microorganisms, since it can be kept under controlled laboratory conditions for over five years. By the use of specialized media assisting in the growth of a metal oxidizing bacterium, the manganese oxidizing bacterium was isolated from the surface of the marine sponge. The bacterium so isolated was characterized for its growth characteristics by microbiological and biochemical techniques, a detailed analysis of which showed that the bacterium followed a life cycle where the culture showed the presence of spore forming bacteria. This was correlated to the manganese oxidation activity of the bacteria and it was found that both stages are interdependent.The action of the protein responsible for carrying out the manganese (Mn) oxidation was studied by an in-gel oxidation assay, and the presence of a multi copper oxidase was confirmed by the use of copper chelators in the buffer. In parallel the effect of addition of copper was observed on the manganese oxidation by the bacteria thus supporting the observations. The manganese oxidation reaction by the bacteria was determined in the culture medium and on the surface of the cells, and it could be concluded that the oxidation was facilitated by the presence of the polysaccharides and proteins on the surface of the cells.Thus the presence of a bacterium capable of oxidizing the manganese from the surroundings was confirmed to be symbiotically associated with the marine sponge S. domuncula by monitoring its growth in axenic cultures. The reasons behind this association were studied.This bacterium displays a crucial role in the physiology/metabolism of the sponge by acting as a reversible Mn store in S. domuncula. According to this view, the presence of SubDo-03 bacteria is required as a protection against higher, toxic concentrations of Mn in the environment; manganese (II) after undergoing oxidation to manganese (IV), becomes an insoluble ion. Since only minute levels of manganese exist in the surrounding seawater a substantial accumulation of manganese has to arise, or a release by the bacterial-precipitated manganese (IV) is implicated to maintain the reversible balance. The other possible benefits provided by the bacterial association to the sponge could be in preventing cellular oxygen toxicity, help in nutrient scavenging and detoxification.

Ozon (O3) ist in der Atmosphäre ein wichtiges Oxidanz und Treibhausgas. Während die höchsten Konzentrationen in der Stratosphäre beobachtet werden und die vor der gefährlichen UV-Strahlung schützende Ozonschicht bilden, können sich signifikante Änderungen der Ozon-Konzentration in der Region der Tropopause auf das Klima der Erde auswirken. Des Weiteren ist Ozon eine der Hauptquellen für das Hydroxylradikal (OH) und nimmt damit entscheidend Einfluss auf die Oxidationskraft der Atmosphäre. Der konvektive Transport von Ozon und seinen Vorläufergasen aus Regionen nahe der Erdoberfläche in die freie Troposphäre beeinflusst das Budget dieser Spezies in der Tropopausenregion.rnDie Datengrundlage der Studie in der vorliegenden Arbeit basiert auf den flugzeuggetragenen Messkampagnen GABRIEL 2005 (Suriname, Südamerika) sowie HOOVER I 2006 und HOOVER II 2007 (beide in Europa). Mit dem zur Verfügung stehenden Datensatz wird das Ozonbudget in der freien, unbelasteten Hintergrundatmosphäre und in der durch hochreichende Konvektion gestörten, oberen Troposphäre untersucht. Anhand der auf in-situ Messungen von O3, NO, OH, HO2 und dem aktinischen Strahlungsfluss basierten Berechnung der Netto-Ozonproduktionsrate (NOPR) werden für das Messgebiet Ozontendenzen in der unbelasteten Troposphäre abgeleitet und mit Simulationen des globalen Chemie-Transport-Modells MATCH-MPIC verglichen. Mit Hilfe zweier Fallstudien in den Tropen in Südamerika und den mittleren Breiten in Europa werden die Auswirkungen von hochreichender Konvektion auf die obere Troposphäre quantifiziert.rnDie Ergebnisse zeigen für die Grenzschicht in niedrigen und mittleren Breiten eine eindeutige Tendenz zur Produktion von Ozon, was für den tropischen Regenwald in der Messregion nicht der allgemeinen Erwartung entsprach, nach der diese Region durch die Zerstörung von Ozon charakterisiert sein sollte. In der oberen Troposphäre ab etwa 7 km wird für die beiden Regionen eine leichte Tendenz zur Ozonproduktion beobachtet. Signifikante Unterschiede zeigen die Ergebnisse für die mittlere Troposphäre. Während die Tropen in dieser Region durch eine eindeutige Tendenz zur Zerstörung von Ozon charakterisiert sind, lässt sich über den mittleren Breiten zwar eine hohe photochemische Aktivität aber keine derart klare Tendenz feststellen. Die hohen Breiten zeichnen sich durch eine neutrale Troposphäre in Bezug auf die Ozontendenz aus und weisen kaum photochemische Aktivität auf. Der Vergleich dieser Ergebnisse mit dem MATCH-MPIC Modell zeigt in weiten Teilen der Messregionen eine grundlegende Übereinstimmung in der Tendenz zur Produktion oder Zerstörung von Ozon. Die absoluten Werte werden vom Modell aber generell unterschätzt. Signifikante Unterschiede zwischen in-situ Daten und Modellsimulationen werden in der Grenzschicht über dem tropischen Regenwald identifiziert.rnDer Einfluss der Konvektion ist durch eine signifikant erhöhte NOPR gekennzeichnet. In dieser Arbeit wird in den Tropen mit einem Median-Wert von 0.20 ppbv h−1 eine um den Faktor 3.6 erhöhte NOPR im Vergleich zur ungestörten oberen Troposphäre abgeschätzt. In den mittleren Breiten führt die um eine Größenordnung höhere NO-Konzentration zu einem Wert von 1.89 ppbv h−1, was einer Überhöhung um einen Faktor 6.5 im Vergleich zum ungestörten Zustand entspricht. Diese Ergebnisse zeigen für beide Regionen in der oberen Troposphäre eine erhöhte Ozonproduktion als Folge konvektiver Aktivität. rnrnHochreichende Konvektion ist zudem ein sehr effektiver Mechanismus für den Vertikaltransport aus der Grenzschicht in die obere Troposphäre. Die schnelle Hebung in konvektiven Wolken führt bei Spurengasen mit Quellen an der Erdoberfläche zu einer Erhöhung ihrer Konzentration in der oberen Troposphäre. Die hochgradig löslichen Spurenstoffe Formaldehyd (HCHO) und Wasserstoffperoxid (H2O2) sind wichtige Vorläufergase der HOx-Radikale. Es wird angenommen, dass sie aufgrund ihrer Löslichkeit in Gewitterwolken effektiv ausgewaschen werden.rnIn der vorliegenden Arbeit wird eine Fallstudie von hochreichender Konvektion im Rahmen des HOOVER II Projekts im Sommer 2007 analysiert. Am 19.07.2007 entwickelten sich am Nachmittag am Südostrand eines in nordöstlicher Richtung ziehenden mesoskaligen konvektiven Systems drei zunächst isolierte konvektive Zellen. Flugzeuggetragene Messungen in der Aus- und der Einströmregion einer dieser Gewitterzellen stellen einen exzellenten Datensatz bereit, um die Auswirkungen von hochreichender Konvektion auf die Verteilung verschiedener Spurengase in der oberen Troposphäre zu untersuchen. Der Vergleich der Konzentrationen von Kohlenstoffmonoxid (CO) und Methan (CH4) zwischen der oberen Troposphäre und der Grenzschicht deutet auf einen nahezu unverdünnten Transport dieser langlebigen Spezies in der konvektiven Zelle hin. Die Verhältnisse betragen (0.94±0.04) für CO und (0.99±0.01) für CH4. Für die löslichen Spezies HCHO und H2O2 beträgt dieses Verhältnis in der Ausströmregion (0.55±0.09) bzw. (0.61±0.08). Dies ist ein Indiz dafür, dass diese Spezies nicht so effektiv ausgewaschen werden wie angenommen. Zum besseren Verständnis des Einflusses der Konvektion auf die Budgets dieser Spezies in der oberen Troposphäre wurden im Rahmen dieser Arbeit Boxmodell-Studien für den Beitrag der photochemischen Produktion in der Ausströmregion durchgeführt, wobei die gemessenen Spezies und Photolysefrequenzen als Randbedingungen dienten. Aus den Budgetbetrachtungen für HCHO und H2O2 wird eine Auswascheffizienz von (67±24) % für HCHO und (41±18) % für H2O2 abgeschätzt. Das für H2O2 überraschende Ergebnis lässt darauf schließen, dass dieses Molekül in einer Gewitterwolke deutlich effektiver transportiert werden kann, als aufgrund seiner hohen Löslichkeit aus der Henry-Konstanten zu erwarten wäre. Das Ausgasen von gelöstem H2O2 beim Gefrieren eines Wolkentropfens, d.h. ein Retentionskoeffizient kleiner 1, ist ein möglicher Mechanismus, der zum beobachteten Mischungsverhältnis dieser löslichen Spezies in der Ausströmregion beitragen kann.

Air quality represents a key issue in the so-called pollution “hot spots”: environments in which anthropogenic sources are concentrated and dispersion of pollutants is limited. One of these environments, the Po Valley, normally experiences exceedances of PM10 and PM2.5 concentration limits, especially in winter when the ventilation of the lower layers of the atmosphere is reduced. This thesis provides a highlight of the chemical properties of particulate matter and fog droplets in the Po Valley during the cold season, when fog occurrence is very frequent. Fog-particles interactions were investigated with the aim to determine their impact on the regional air quality. Size-segregated aerosol samples were collected in Bologna, urban site, and San Pietro Capofiume (SPC), rural site, during two campaigns (November 2011; February 2013) in the frame of Supersito project. The comparison between particles size-distribution and chemical composition in both sites showed the relevant contribution of the regional background and secondary processes in determining the Po Valley aerosol concentration. Occurrence of fog in November 2011 campaign in SPC allowed to investigate the role of fog formation and fog chemistry in the formation, processing and deposition of PM10. Nucleation scavenging was investigated with relation to the size and the chemical composition of particles. We found that PM1 concentration is reduced up to 60% because of fog scavenging. Furthermore, aqueous-phase secondary aerosol formation mechanisms were investigated through time-resolved measurements. In SPC fog samples have been systematically collected and analysed since the nineties; a 20 years long database has been assembled. This thesis reports for the first time the results of this long time series of measurements, showing a decrease of sulphate and nitrate concentration and an increase of pH that reached values close to neutrality. A detailed discussion about the occurred changes in fog water composition over two decades is presented.

The composition of the atmosphere is frequently perturbed by the emission of gaseous and particulate matter from natural as well as anthropogenic sources. While the impact of trace gases on the radiative forcing of the climate is relatively well understood the role of aerosol is far more uncertain. Therefore, the study of the vertical distribution of particulate matter in the atmosphere and its chemical composition contribute valuable information to bridge this gap of knowledge. The chemical composition of aerosol reveals information on properties such as radiative behavior and hygroscopicity and therefore cloud condensation or ice nucleus potential. rnThis thesis focuses on aerosol pollution plumes observed in 2008 during the POLARCAT (Polar Study using Aircraft, Remote Sensing, Surface Measurements and Models, of Climate, Chemistry, Aerosols, and Transport) campaign over Greenland in June/July and CONCERT (Contrail and Cirrus Experiment) campaign over Central and Western Europe in October/November. Measurements were performed with an Aerodyne compact time-of-flight aerosol mass spectrometer (AMS) capable of online size-resolved chemical characterization of non-refractory submicron particles. In addition, the origins of pollution plumes were determined by means of modeling tools. The characterized pollution episodes originated from a large variety of sources and were encountered at distinct altitudes. They included pure natural emissions from two volcanic eruptions in 2008. By the time of detection over Western Europe between 10 and 12 km altitude the plume was about 3 months old and composed to 71 % of particulate sulfate and 21 % of carbonaceous compounds. Also, biomass burning (BB) plumes were observed over Greenland between 4 and 7 km altitude (free troposphere) originating from Canada and East Siberia. The long-range transport took roughly one and two weeks, respectively. The aerosol was composed of 78 % organic matter and 22 % particulate sulfate. Some Canadian and all Siberian BB plumes were mixed with anthropogenic emissions from fossil fuel combustion (FF) in North America and East Asia. It was found that the contribution of particulate sulfate increased with growing influences from anthropogenic activity and Asia reaching up to 37 % after more than two weeks of transport time. The most exclusively anthropogenic emission source probed in the upper troposphere was engine exhaust from commercial aircraft liners over Germany. However, in-situ characterization of this aerosol type during aircraft chasing was not possible. All long-range transport aerosol was found to have an O:C ratio close to or greater than 1 implying that low-volatility oxygenated organic aerosol was present in each case despite the variety of origins and the large range in age from 3 to 100 days. This leads to the conclusion that organic particulate matter reaches a final and uniform state of oxygenation after at least 3 days in the free troposphere. rnExcept for aircraft exhaust all emission sources mentioned above are surface-bound and thus rely on different types of vertical transport mechanisms, such as direct high altitude injection in the case of a volcanic eruption, or severe BB, or uplift by convection, to reach higher altitudes where particles can travel long distances before removal mainly caused by cloud scavenging. A lifetime for North American mixed BB and FF aerosol of 7 to 11 days was derived. This in consequence means that emission from surface point sources, e.g. volcanoes, or regions, e.g. East Asia, do not only have a relevant impact on the immediate surroundings but rather on a hemispheric scale including such climate sensitive zones as the tropopause or the Arctic.

In recent years the hot water treatment (HW) represents an effective and safe approach for managing postharvest decay. This study reported the effect of an HW (60°C for 60 s and 45°C for 10 min) on brown rot and blue mould respectively. Peaches was found more thermotolerant compared to apple fruit, otherwise Penicillium expansum was more resistant to heat with respect to Monilinia spp. In semi-commercial and commercial trials, the inhibition of brown rot in naturally infected peaches was higher than 78% after 6 days at 0°C and 3 days at 20°C. Moreover, in laboratory trials a 100% disease incidence reduction was obtained by treating artificially infected peaches at 6-12 h after inoculation revealing a curative effect of HW. The expression levels of some genes were evaluated by qRT-PCR. Specifically, the cell wall genes (β-GAL, PL, PG, PME) showed a general decrease of expression level whereas PAL, CHI, HSP70 and ROS-scavenging genes were induced in treated peaches compared to the control ones. Contrarily, HW applied on artificially infected fruit before the inoculum was found to increase brown rot susceptibility. This aspect might be due to an increase of fruit VOCs emission as revealed by PTR-ToF-MS analysis. In addition a microarray experiment was conducted to analyze molecular mechanisms underneath the apple response to heat. Our results showed a largest amount of induced Heat shock proteins (HSPs), Heat shock cognate proteins (HSCs), Heat shock transcription factors (HSTFs) genes found at 1 and 4 hours from the treatment. Those genes required for the thermotolerance process could be involved in induced resistance response. The hypothesis was confirmed by 30% of blue mold disease reduction in artificially inoculated apple after 1 and 4 hours from the treatment. In order to improve peaches quality and disease management during storage, an innovative tool was also used: Da-meter.

Among abiotic stresses, high salinity stress is the most severe environmental stress. High salinity exerts its negative impact mainly by disrupting the ionic and osmotic equilibrium of the cell. In saline soils, high levels of sodium ions lead to plant growth inhibition and even death. Salt tolerance in plants is a multifarious phenomenon involving a variety of changes at molecular, organelle, cellular, tissue as well as whole plant level. In addition, salt tolerant plants show a range of adaptations not only in morphological or structural features but also in metabolic and physiological processes that enable them to survive under extreme saline environments. The main objectives of my dissertation were understanding the main physiological and biomolecular features of plant responses to salinity in different genotypes of horticultural crops that are belonging to different families Solanaceae (tomato) and Cucurbitaceae (melon) and Brassicaceae (cabbage and radish). Several aspects of crop responses to salinity have been addressed with the final aim of combining elements of functional stress response in plants by using several ways for the assessment of plant stress perception that ranging from destructive measurements (eg. leaf area, relative growth rate, leaf area index, and total plant fresh and dry weight), to physiological determinations (eg. stomatal conductance, leaf gas exchanges, water use efficiency, and leaf water relation), to the determination of metabolite accumulation in plant tissue (eg. Proline and protein) as well as evaluation the role of enzymatic antioxidant capacity assay in scavenging reactive oxygen species that have been generated under salinized condition, and finally assessing the gene induction and up-down regulation upon salinization (eg. SOS pathway).

The aim of this study is to investigate on some molecular mechanisms contributing to the pathogenesis of osteoarthritis (OA) and in particular to the senescence of articular chondrocytes. It is focused on understanding molecular events downstream GSK3β inactivation or dependent on the activity of IKKα, a kinase that does not belong to the phenotype of healthy articular chondrocytes. Moreover, the potential of some nutraceuticals on scavenging ROS thus reducing oxidative stress, DNA damage, and chondrocyte senescence has been evaluated in vitro. The in vitro LiCl-mediated GSK3β inactivation resulted in increased mitochondrial ROS production, that impacted on cellular proliferation, with S-phase transient arrest, increased SA-β gal and PAS staining, cell size and granularity. ROS are also responsible for the of increased expression of two major oxidative lesions, i.e. 1) double strand breaks, tagged by γH2AX, that associates with activation of GADD45β and p21, and 2) 8-oxo-dG adducts, that associate with increased IKKα and MMP-10 expression. The pattern observed in vitro was confirmed on cartilage from OA patients. IKKa dramatically affects the intensity of the DNA damage response induced by oxidative stress (H2O2 exposure) in chondrocytes, as evidenced by silencing strategies. At early time point an higher percentage of γH2AX positive cells and more foci in IKKa-KD cells are observed, but IKKa KD cells proved to almost completely recover after 24 hours respect to their controls. Telomere attrition is also reduced in IKKaKD. Finally MSH6 and MLH1 genes are up-regulated in IKKαKD cells but not in control cells. Hydroxytyrosol and Spermidine have a great ROS scavenging capacity in vitro. Both treatments revert the H2O2 dependent increase of cell death and γH2AX-foci formation and senescence, suggesting the ability of increasing cell homeostasis. These data indicate that nutraceuticals represent a great challenge in OA management, for both therapeutical and preventive purposes.

In questo lavoro di tesi si è applicato dei materiali a base di ossido di grafene in ambito catalitico ed elettrochimico, dopo uno studio iniziale delle proprietà del materiale di partenza tramite XRD, ATR, Raman e SEM-EDS e la sua evoluzione durante processi di riduzione per ottimizzare il suo utilizzo. In particolare è stato depositato l’ossido di grafene su delle sfere di Al2O3 (γ-Al2O3 e α-Al2O3) per preparare dei catalizzatori eterogenei per la produzione di 2,5-biidrossimetilfurano (BHMF) tramite idrogenazione del 5-idrossimetilfurfurale (HMF). Le prestazioni del catalizzatore costituito di sfere di γ-Al2O3 ricoperte di GO sono state paragonate alle prestazioni di un sistema analogo ma contenente anche Pt 1%wt coimpregnato e alle performance della sospensione di GO non supportata. In ambito elettrochimico è stato preparato ed ottimizzato un sensore a base di ossido di grafene per determinare la concentrazione del glucosio, fondamentale per i soggetti diabetici. Nell’intenzione di ampliare le condizioni di impiego rispetto ai biosensori, sono stati studiati elettrodi modificati con ossido di grafene e composti tipo idrotalciti, i quali operano in ambiente basico sfruttando il fenomeno di scavenging del Ni.

Metallische Nanopartikel und ihre Oxide (z.B. ZnO NP, TiO2 NP und Fe2O3 NP) werden aufgrund ihrer chemischen und physikalischen Eigenschaften häufig als Additive in der Reifenproduktion, in Katalysatoren, Lebensmitteln, Arzneimitteln und Kosmetikprodukten verwendet. Künftig wird ein kontinuierlicher Anstieg der industriellen Anwendung (~ 1663 Tonnen im Jahr 2025) mit gesteigerter Freisetzung in die Umwelt erwartet, was zwangsläufig zu einer vermehrten Aufnahme über das respiratorische Epithel führt. Metalldampffieber ist als gesundheitsschädigender Effekt von Metalloxid-haltigen Aerosolen (z.B. ZnO) nach Inhalation bekannt. Immunreaktionen, wie beispielsweise Entzündungen, werden häufig mit der Entstehung von Sauerstoffradikalen (ROS) in Verbindung gebracht, die wiederum zu DNA-Schäden führen können. Drei mögliche Ursachen der Genotoxität werden angenommen: direkte Interaktion von Nanopartikeln mit intrazellulären Strukturen, Interaktion von Ionen dissoziierter Partikel mit intrazellulären Strukturen sowie die Entstehung von ROS initiiert durch Partikel oder Ionen.rnDie vorliegende Studie befasst sich mit den Mechanismen der Genotoxizität von ZnO Nanopartikeln (ZnO NP), als Beispiel für metallische Nanopartikel, im respiratorischen Epithel. In der Studie wurde gezielt die intrazelluläre Aufnahme und Verteilung von ZnO NP, deren Toxizität, deren DNA schädigendes Potential sowie die Aktivierung der DNA damage response (DDR) analysiert.rnEs konnten kaum internalisierte ZnO NP mittels TEM detektiert werden. Innerhalb der ersten Sekunden nach Behandlung mit ZnO NP wurde spektrofluorometrisch ein starker Anstieg der intrazellulären Zn2+ Konzentration gemessen. In unbehandelten Zellen war Zn2+ in granulären Strukturen lokalisiert. Die Behandlung mit ZnO NP führte zu einer Akkumulation von Zn2+ in diesen Strukturen. Im zeitlichen Verlauf verlagerten sich die Zn2+-Ionen in das Zytoplasma, sowie in Zellkerne und Mitochondrien. Es wurde keine Kolokalisation von Zn2+ mit den frühen Endosomen und dem endoplasmatischen Retikulum beobachtet. Die Vorbehandlung der Zellen mit Diethylen-triaminpentaessigsäure (DTPA), als extrazellulärem Komplexbildner, verhinderte den intrazellulären Anstieg von Zn2+ nach Behandlung mit den Partikeln.rnDie Behandlung mit ZnO NP resultierte in einer zeit- und dosisabhängigen Reduktion der zellulären Viabilität, während die intrazelluläre ROS-Konzentrationen in den ersten 30 min leicht und anschließend kontinuierlich bis zum Ende der Messung anstiegen. Außerdem verringerte sich das mitochondriale Membranpotential, während sich die Anzahl der frühapoptotischen Zellen in einer zeitabhängigen Weise erhöhte. rnDNA Doppelstrangbrüche (DNA DSB) wurden mittels Immunfluoreszenz-Färbung der γH2A.X foci sichtbar gemacht und konnten nach Behandlung mit ZnO NP detektiert werden. Die Vorbehandlung mit dem Radikalfänger N-Acetyl-L-Cytein (NAC) resultierte in stark reduzierten intrazellulären ROS-Konzentrationen sowie wenigen DNA DSB. Die DNA Schädigung wurde durch Vorbehandlung mit DTPA ganz verhindert.rnDie Aktivierung der DDR wurde durch die Analyse von ATM, ATR, Chk1, Chk2, p53 und p21 mittels Western Blot und ELISA nach Behandlung mit ZnO NP überprüft. Der ATR/Chk1 Signalweg wurde durch ZnO NP nicht aktiviert. Die Komplexierung von Zn2+ resultierte in einer verminderten ATM/Chk2 Signalwegaktivierung. Es zeigte sich, dass das Abfangen von ROS keinen Effekt auf die ATM/Chk2 Signalwegaktivierung hatte.rnZusammengefasst wurde festgestellt, dass die Exposition mit ZnO NP in der Entstehung von ROS, reduzierter Viabilität und vermindertem mitochondrialem Membranpotential resultiert, sowie zeitabhängig eine frühe Apoptose initiiert. ZnO NP dissoziierten extrazellulär und wurden schnell als Zn2+ über unbekannte Mechanismen internalisiert. Die Zn2+-Ionen wurden im Zytoplasma, sowie besonders in den Mitochondrien und dem Zellkern, akkumuliert. Die DDR Signalgebung wurde durch ZnO NP aktiviert, jedoch nicht durch NAC inhibiert. Es wurde gezeigt, dass DTPA die DDR Aktivierung komplett inhibierte. Die Behandlung mit ZnO NP induzierte DNA DSB. Die Inhibition von ROS reduzierte die DNA DSB und die Komplexierung der Zn2+ verhinderte die Entstehung von DNA DSB.rnDiese Daten sprechen für die Dissoziation der Partikel und die hierbei freigesetzten Zn2+ als Hauptmediator der Genotoxizität metallischer Nanopartikel. rn

Dynamic core-shell nanoparticles have received increasing attention in recent years. This paper presents a detailed study of Au-Hg nanoalloys, whose composing elements show a large difference in cohesive energy. A simple method to prepare Au@Hg particles with precise control over the composition up to 15 atom% mercury is introduced, based on reacting a citrate stabilized gold sol with elemental mercury. Transmission electron microscopy shows an increase of particle size with increasing mercury content and, together with X-ray powder diffraction, points towards the presence of a core-shell structure with a gold core surrounded by an Au-Hg solid solution layer. The amalgamation process is described by pseudo-zero-order reaction kinetics, which indicates slow dissolution of mercury in water as the rate determining step, followed by fast scavenging by nanoparticles in solution. Once adsorbed at the surface, slow diffusion of Hg into the particle lattice occurs, to a depth of ca. 3 nm, independent of Hg concentration. Discrete dipole approximation calculations relate the UV-vis spectra to the microscopic details of the nanoalloy structure. Segregation energies and metal distribution in the nanoalloys were modeled by density functional theory calculations. The results indicate slow metal interdiffusion at the nanoscale, which has important implications for synthetic methods aimed at core-shell particles.

Here we present stable isotope data for vertical profiles of dissolved molybdenum of the modern euxinic water columns of the Black Sea and two deeps of the Baltic Sea. Dissolved molybdenum in all water samples is depleted in salinity-normalized concentration and enriched in the heavy isotope (δ98Mo values up to + 2.9‰) compared to previously published isotope data of sedimentary molybdenum from the same range of water depths. Furthermore, δ98Mo values of all water samples from the Black Sea and anoxic deeps of the Baltic Sea are heavier than open ocean water. The observed isotope fractionation between sediments and the anoxic water column of the Black Sea are in line with the model of thiomolybdates that scavenge to particles under reducing conditions. An extrapolation to a theoretical pure MoS42− solution indicates a fractionation constant between MoS42− and authigenic solid Mo of 0.5 ± 0.3‰. Measured waters with all thiomolybdates coexisting in various proportions show larger but non-linear fractionation. The best explanation for our field observations is Mo scavenging by the thiomolybdates, dominantly — but not exclusively — present in the form of MoS42−. The Mo isotopic compositions of samples from the sediments and anoxic water column of the Baltic Sea are in overall agreement with those of the Black Sea at intermediate depth and corresponding sulphide concentrations. The more dynamic changes of redox conditions in the Baltic deeps complicate the Black Sea-derived relationship between thiomolybdates and Mo isotopic composition. In particular, the occasional flushing/mixing, of the deep waters, affects the corresponding water column and sedimentary data. δ98Mo values of the upper oxic waters of both basins are higher than predicted by mixing models based on salinity variations. The results can be explained by non-conservative behaviour of Mo under suboxic to anoxic conditions in the shallow bottom parts of the basin, most pronounced on the NW shelf of the Black Sea.

P>1. Proliferative kidney disease (PKD) is a disease of salmonid fish caused by the endoparasitic myxozoan, Tetracapsuloides bryosalmonae, which uses freshwater bryozoans as primary hosts. Clinical PKD is characterised by a temperature-dependent proliferative and inflammatory response to parasite stages in the kidney.;2. Evidence that PKD is an emerging disease includes outbreaks in new regions, declines in Swiss brown trout populations and the adoption of expensive practices by fish farms to reduce heavy losses. Disease-related mortality in wild fish populations is almost certainly underestimated because of e.g. oversight, scavenging by wild animals, misdiagnosis and fish stocking.;3. PKD prevalences are spatially and temporally variable, range from 0 to 90-100% and are typically highest in juvenile fish.;4. Laboratory and field studies demonstrate that (i) increasing temperatures enhance disease prevalence, severity and distribution and PKD-related mortality; (ii) eutrophication may promote outbreaks. Both bryozoans and T. bryosalmonae stages in bryozoans undergo temperature- and nutrient-driven proliferation.;5. Tetracapsuloides bryosalmonae is likely to achieve persistent infection of highly clonal bryozoan hosts through vertical transmission, low virulence and host condition-dependent cycling between covert and overt infections. Exploitation of fish hosts entails massive proliferation and spore production by stages that escape the immune response. Many aspects of the parasite's life cycle remain obscure. If infectious stages are produced in all hosts then the complex life cycle includes multiple transmission routes.;6. Patterns of disease outbreaks suggest that background, subclinical infections exist under normal environmental conditions. When conditions change, outbreaks may then occur in regions where infection was hitherto unsuspected.;7. Environmental change is likely to cause PKD outbreaks in more northerly regions as warmer temperatures promote disease development, enhance bryozoan biomass and increase spore production, but may also reduce the geographical range of this unique multihost-parasite system. Coevolutionary dynamics resulting from host-parasite interactions that maximise fitness in previous environments may pose problems for sustainability, particularly in view of extensive declines in salmonid populations and degradation of many freshwater habitats.

The proposition posed is that the value of amino acid conjugation to the organism is not, as in the traditional view, to use amino acids for the detoxication of aromatic acids. Rather, the converse is more likely, to use aromatic acids that originate from the diet and gut microbiota to assist in the regulation of body stores of amino acids, such as glycine, glutamate, and, in certain invertebrates, arginine, that are key neurotransmitters in the central nervous system (CNS). As such, the amino acid conjugations are not so much detoxication reactions, rather they are homeostatic and neuroregulatory processes. Experimental data have been culled in support of this hypothesis from a broad range of scientific and clinical literature. Such data include the low detoxication value of amino acid conjugations and the Janus nature of certain amino acids that are both neurotransmitters and apparent conjugating agents. Amino acid scavenging mechanisms in blood deplete brain amino acids. Amino acids glutamate and glycine when trafficked from brain are metabolized to conjugates of aromatic acids in hepatic mitochondria and then irreversibly excreted into urine. This process is used clinically to deplete excess nitrogen in cases of urea cycle enzymopathies through excretion of glycine or glutamine as their aromatic acid conjugates. Untoward effects of high-dose phenylacetic acid surround CNS toxicity. There appears to be a relationship between extent of glycine scavenging by benzoic acid and psychomotor function. Glycine and glutamine scavenging by conjugation with aromatic acids may have important psychosomatic consequences that link diet to health, wellbeing, and disease.

Exogenous melatonin is widely used for sleep disorders and has potential value in neuroprotection, cardioprotection and as an antioxidant. Here, a novel method is described for the determination of melatonin and six metabolites in mouse urine by use of LC-MS/MS and GC-MS. LC-MS/MS is used for the measurement of melatonin, N1-acetyl-5-methoxykynuramine (AMK), N1-acetyl-N2-formyl-5-methoxykynuramine (AFMK) and 6-hydroxymelatonin (6-HMEL), while GC/MS is used for the determination of N-[2-(5-methoxy-2-oxo-2,3-dihydro-1H-indol-3-yl)-ethyl]-acetamide (2-OMEL) and cyclic 3-hydroxymelatonin (3-HMEL) with detection limits on column of 0.02-0.5 pmol, depending on the metabolite. Following oral administration of melatonin to mice, a 0-24 hr urine collection revealed the presence of melatonin (0.2% dose), 6-HMEL (37.1%) and NAS (3.1%) comprising >90% of the total metabolites; AMK and AFMK were also detected at 0.01% each; 2-OMEL was found at 2.2% of the dose, which is >100 times more than the AMK/AFMK pathway, and comprises >5% of the melatonin-related material detected in mouse urine. 3-HMEL was largely found as a sulfate conjugate. These studies establish sensitive assays for determination of six melatonin metabolites in mouse urine and confirm the potential for antioxidant activity of melatonin through the identification in vivo of AMK and AFMK, ring-opened metabolites with a high capacity for scavenging reactive oxygen species.
