A delivery system containing polymeric (Eudragit) nanoparticles has been developed for encapsulation and controlled release of bioactive flavonoids (quercetin). Nanoparticles were fabricated using a solvent displacement method. Particle size, morphology, and charge were measured by light scattering, electron microscopy and zeta-potential. Encapsulation efficiency (EE) and release profiles were determined using electrochemical methods. Molecular interactions within the particle matrix were characterized by X-ray diffraction, differential scanning calorimetry, and infrared spectroscopy. Antioxidant properties of free and encapsulated quercetin were analyzed by TBARS and fluorescence spectroscopy. Bioaccessibility of quercetin was evaluated using an in vitro digestion model. Relatively small (d a parts per thousand aEuro parts per thousand 370 nm) anionic polymeric nanoparticles were formed containing quercetin in a non-crystalline form (EE a parts per thousand aEuro parts per thousand 67 %). The main interaction between quercetin and Eudragit was hydrogen bonding. Encapsulated quercetin remained stable during 6 months storage and maintained its antioxidant activity. Quercetin bioaccessibility within simulated small intestinal conditions was improved by encapsulation. The knowledge obtained from this study will facilitate the rational design and fabrication of polymeric nanoparticles as oral delivery systems for encapsulation, protection, and release of bioactive compounds.

The anomalies in the anti-Stokes to Stokes intensity ratios in single-molecule surface-enhanced resonance Raman scattering were investigated. Brilliant green and crystal violet dyes were the molecular probes, and the experiments were carried out on an electrochemically activated Ag surface. The results allowed new insights into the origin of these anomalies and led to a new method to confirm the single-molecule regime in surface-enhanced Raman scattering. Moreover, a methodology to estimate the distribution of resonance energies that contributed to the imbalance in the anti-Stokes to Stokes intensity ratios at the electromagnetic hot spots was proposed. This method allowed the local plasmonic resonance energies on the metallic surface to be spatially mapped.

The interaction between the antimicrobial peptide gramicidin (Gr) and dipalmitoylphosphatidylcholine (DPPC)/dioctadecyldimethylammonium bromide (DODAB) 1:1 large unilamellar vesicles (LVs) or bilayer fragments (BFs) was evaluated by means of several techniques. The major methods were: 1) Gr intrinsic fluorescence and circular dichroism (CD) spectroscopy; 2) dynamic light scattering for sizing and zeta-potential analysis; 3) determination of the bilayer phase transition from extrinsic fluorescence of bilayer probes; 4) pictures of the dispersions for evaluation of coloidal stability over a range of time and NaCl concentration. For Gr in LVs, the Gr dimeric channel conformation is suggested from: 1) CD and intrinsic fluorescence spectra similar to those in trifluoroethanol (TFE); 2) KCl or glucose permeation through the LVs/Gr bilayer. For Gr in BFs, the intertwined dimeric, non-channel Gr conformation is evidenced by CD and intrinsic fluorescence spectra similar to those in ethanol. Both LVs and BFs shield Gr tryptophans against quenching by acrylamide but the Stern-Volmer quenching constant was slightly higher for Gr in BFs confirming that the peptide is more exposed to the water phase in BFs than in LVs. The DPPC/DODAB/Gr supramolecular assemblies may predict the behavior of other antimicrobial peptides in assemblies with lipids. (C) 2012 Elsevier B.V. All rights reserved.

The incorporation of the curcumin into poly(lactic-co-glycolic)acid (PLGA) nanospheres by the nanoprecipitation technique, the characterization of the nanoparticles and the schistosomicidal activity of the curcumin-loaded into PLGA nanospheres were reported. The incorporation process occurred with high efficiency and the images of field-emission scanning electron microscopy (FESEM) revealed the production of spherically shaped particles. According to the dynamic light scattering measurements, the particles are nanometric and monodisperse. The curcumin-loaded PLGA nanoparticles (50 and 100 mu M) caused the death of all worms and a separation between 50% and 100% of Schistosoma mansoni couples at concentrations from 30 mu M. Moreover, the curcumin-loaded PLGA nanoparticles also decreased the motor activity and caused partial alterations in the tegument of adult worms. This study marks the first time that schistosomicidal activity has been reported for curcumin-loaded PLGA nanoparticles.

Xyloglucan is a major structural polysaccharide of the primary (growing) cell wall of higher plants. It consists of a cellulosic backbone (beta-1,4-linked glucosyl residues) that is frequently substituted with side chains. This report describes Aspergillus nidulans strain A773 recombinant secretion of a dimeric xyloglucan-specific endo-beta-1,4-glucanohydrolase (XegA) cloned from Aspergillus niveus. The ORF of the A. niveus xegA gene is comprised of 714 nucleotides, and encodes a 238 amino acid protein with a calculated molecular weight of 23.5 kDa and isoelectric point of 4.38. The optimal pH and temperature were 6.0 and 60 degrees C, respectively. XegA generated a xyloglucan-oligosaccharides (XGOs) pattern similar to that observed for cellulases from family GH12, i.e., demonstrating that its mode of action includes hydrolysis of the glycosidic linkages between glucosyl residues that are not branched with xylose. In contrast to commercial lichenase, mixed linkage beta-glucan (lichenan) was not digested by XegA, indicating that the enzyme did not cleave glucan beta-1,3 or beta-1,6 bonds. The far-UV CD spectrum of the purified enzyme indicated a protein rich in beta-sheet structures as expected for GH12 xyloglucanases. Thermal unfolding studies displayed two transitions with mid-point temperatures of 51.3 degrees C and 81.3 degrees C respectively, and dynamic light scattering studies indicated that the first transition involves a change in oligomeric state from a dimeric to a monomeric form. Since the enzyme is a predominantly a monomer at 60 degrees C. the enzymatic assays demonstrated that XegA is more active in its monomeric state. (c) 2012 Elsevier B.V. All rights reserved.

Abstract Background Particulate systems are well known to be able to deliver drugs with high efficiency and fewer adverse side effects, possibly by endocytosis of the drug carriers. On the other hand, cationic compounds and assemblies exhibit a general antimicrobial action. In this work, cationic nanoparticles built from drug, cationic lipid and polyelectrolytes are shown to be excellent and active carriers of amphotericin B against C. albicans. Results Assemblies of amphotericin B and cationic lipid at extreme drug to lipid molar ratios were wrapped by polyelectrolytes forming cationic nanoparticles of high colloid stability and fungicidal activity against Candida albicans. Experimental strategy involved dynamic light scattering for particle sizing, zeta-potential analysis, colloid stability, determination of AmB aggregation state by optical spectra and determination of activity against Candida albicans in vitro from cfu countings. Conclusion Novel and effective cationic particles delivered amphotericin B to C. albicans in vitro with optimal efficiency seldom achieved from drug, cationic lipid or cationic polyelectrolyte in separate. The multiple assembly of antibiotic, cationic lipid and cationic polyelctrolyte, consecutively nanostructured in each particle produced a strategical and effective attack against the fungus cells.

The interest in the use of evaporative light scattering detector (ELSD) for the analysis of different classes of natural products has grown over the years. This is because this detector has become an excellent alternative compared to other types of detectors, such as the refractive index detector and the ultraviolet (UV) detector. This review describes the basic principles of ELSD functioning and discusses the advantages and disadvantages in using an ELSD for the analysis of organic compounds. Additionaly, an overview, covering the last 23 years, of ELSD applications in natural products analysis (saponins, terpenes, carbohydrates, glycosides, alkaloids, steroids, flavonoids, peptides, polyketides, coumarins and iridoids) is presented and discussed.

Chemically resolved submicron (PM1) particlemass fluxes were measured by eddy covariance with a high resolution time-of-flight aerosolmass spectrometer over temperate and tropical forests during the BEARPEX-07 and AMAZE-08 campaigns. Fluxes during AMAZE-08 were small and close to the detection limit (<1 ng m−2 s−1) due to low particle mass concentrations (<1 μg m−3). During BEARPEX-07, concentrations were five times larger, with mean mid-day deposition fluxes of −4.8 ng m−2 s−1 for total nonrefractory PM1 (Vex,PM1 = −1 mm s−1) and emission fluxes of +2.6 ng m−2 s−1 for organic PM1 (Vex,org = +1 mm s−1). Biosphere–atmosphere fluxes of different chemical components are affected by in-canopy chemistry, vertical gradients in gas-particle partitioning due to canopy temperature gradients, emission of primary biological aerosol particles, and wet and dry deposition. As a result of these competing processes, individual chemical components had fluxes of varying magnitude and direction during both campaigns. Oxygenated organic components representing regionally aged aerosol deposited, while components of fresh secondary organic aerosol (SOA) emitted. During BEARPEX-07, rapid incanopy oxidation caused rapid SOA growth on the timescale of biosphere-atmosphere exchange. In-canopy SOA mass yields were 0.5–4%. During AMAZE-08, the net organic aerosol flux was influenced by deposition, in-canopy SOA formation, and thermal shifts in gas-particle partitioning.Wet deposition was estimated to be an order ofmagnitude larger than dry deposition during AMAZE-08. Small shifts in organic aerosol concentrations from anthropogenic sources such as urban pollution or biomass burning alters the balance between flux terms. The semivolatile nature of the Amazonian organic aerosol suggests a feedback in which warmer temperatures will partition SOA to the gas-phase, reducing their light scattering and thus potential to cool the region.

The Large Scale Biosphere Atmosphere Experiment in Amazonia (LBA) is a long-term (20 years) research effort aimed at the understanding of the functioning of the Amazonian ecosystem. The strong biosphere-atmosphere interaction is a key component of the ecosystem functioning. Two aerosol components are the most visible: The natural biogenic emissions of particles and VOCs, and the biomass burning emissions. Two aerosol and trace gases monitoring stations were operated for 4 years in Manaus and Porto Velho, two very distinct sites, with different land use change. Manaus is a very clean and pristine site and Porto Velho is representative of heavy land use change in Amazonia. Aerosol composition, optical properties, size distribution, vertical profiling and optical depth were measured from 2008 to 2012. Aerosol radiative forcing was calculated over large areas. It was observed that the natural biogenic aerosol has significant absorption properties. Organic aerosol dominates the aerosol mass with 80 to 95%. Light scattering and light absorption shows an increase by factor of 10 from Manaus to Porto Velho. Very few new particle formation events were observed. Strong links between aerosols and VOC emissions were observed. Aerosol radiative forcing in Rondonia shows a high -15 watts/m² during the dry season of 2010, showing the large impacts of aerosol loading in the Amazonian ecosystem. The increase in diffuse radiation changes the forest carbon uptake by 20 to 35%, a large increase in this important ecosystem.

In the past decade the study of superparamagnetic nanoparticles has been intensively developed for many biomedical applications such as magnetically assisted drug delivery, MRI contrast agents, cells separation and hyperthermia therapy. All of these applications require nanoparticles with high magnetization, equipped also with a suitable surface coating which has to be non-toxic and biocompatible. In this master thesis, the silica coating of commercially available magnetic nanoparticles was investigated. Silica is a versatile material with many intrinsic features, such as hydrophilicity, low toxicity, proper design and derivatization yields particularly stable colloids even in physiological conditions. The coating process was applied to commercial magnetite particles dispersed in an aqueous solution. The formation of silica coated magnetite nanoparticles was performed following two main strategies: the Stöber process, in which the silica coating of the nanoparticle was directly formed by hydrolysis and condensation of suitable precursor in water-alcoholic mixtures; and the reverse microemulsions method in which inverse micelles were used to confine the hydrolysis and condensation reactions that bring to the nanoparticles formation. Between these two methods, the reverse microemulsions one resulted the most versatile and reliable because of the high control level upon monodispersity, silica shell thickness and overall particle size. Moving from low to high concentration, within the microemulsion region a gradual shift from larger particles to smaller one was detected. By increasing the amount of silica precursor the silica shell can also be tuned. Fluorescent dyes have also been incorporated within the silica shell by linking with the silica matrix. The structure of studied nanoparticles was investigated by using transmission electron microscope (TEM) and dynamic light scattering (DLS). These techniques have been used to monitor the syntetic procedures and for the final characterization of silica coated and silica dye doped nanoparticles. Finally, field dependent magnetization measurements showed the magnetic properties of core-shell nanoparticles were preserved. Due to a very well defined structure that combines magnetic and luminescent properties together with the possibility of further functionalization, these multifunctional nanoparticles are potentially useful platforms in biomedical fields such as labeling and imaging.

Supramolecular self-assembly represents a key technology for the spontaneous construction of nanoarchitectures and for the fabrication of materials with enhanced physical and chemical properties. In addition, a significant asset of supramolecular self-assemblies rests on their reversible formation, thanks to the kinetic lability of their non-covalent interactions. This dynamic nature can be exploited for the development of “self-healing” and “smart” materials towards the tuning of their functional properties upon various external factors. One particular intriguing objective in the field is to reach a high level of control over the shape and size of the supramolecular architectures, in order to produce well-defined functional nanostructures by rational design. In this direction, many investigations have been pursued toward the construction of self-assembled objects from numerous low-molecular weight scaffolds, for instance by exploiting multiple directional hydrogen-bonding interactions. In particular, nucleobases have been used as supramolecular synthons as a result of their efficiency to code for non-covalent interaction motifs. Among nucleobases, guanine represents the most versatile one, because of its different H-bond donor and acceptor sites which display self-complementary patterns of interactions. Interestingly, and depending on the environmental conditions, guanosine derivatives can form various types of structures. Most of the supramolecular architectures reported in this Thesis from guanosine derivatives require the presence of a cation which stabilizes, via dipole-ion interactions, the macrocyclic G-quartet that can, in turn, stack in columnar G-quadruplex arrangements. In addition, in absence of cations, guanosine can polymerize via hydrogen bonding to give a variety of supramolecular networks including linear ribbons. This complex supramolecular behavior confers to the guanine-guanine interactions their upper interest among all the homonucleobases studied. They have been subjected to intense investigations in various areas ranging from structural biology and medicinal chemistry – guanine-rich sequences are abundant in telomeric ends of chromosomes and promoter regions of DNA, and are capable of forming G-quartet based structures– to material science and nanotechnology. This Thesis, organized into five Chapters, describes mainly some recent advances in the form and function provided by self-assembly of guanine based systems. More generally, Chapter 4 will focus on the construction of supramolecular self-assemblies whose self-assembling process and self-assembled architectures can be controlled by light as external stimulus. Chapter 1 will describe some of the many recent studies of G-quartets in the general area of nanoscience. Natural G- quadruplexes can be useful motifs to build new structures and biomaterials such as self-assembled nanomachines, biosensors, therapeutic aptamer and catalysts. In Chapters 2-4 it is pointed out the core concept held in this PhD Thesis, i.e. the supramolecular organization of lipophilic guanosine derivatives with photo or chemical addressability. Chapter 2 will mainly focus on the use of cation-templated guanosine derivatives as a potential scaffold for designing functional materials with tailored physical properties, showing a new way to control the bottom-up realization of well-defined nanoarchitectures. In section 2.6.7, the self-assembly properties of compound 28a may be considered an example of open-shell moieties ordered by a supramolecular guanosine architecture showing a new (magnetic) property. Chapter 3 will report on ribbon-like structures, supramolecular architectures formed by guanosine derivatives that may be of interest for the fabrication of molecular nanowires within the framework of future molecular electronic applications. In section 3.4 we investigate the supramolecular polymerizations of derivatives dG 1 and G 30 by light scattering technique and TEM experiments. The obtained data reveal the presence of several levels of organization due to the hierarchical self-assembly of the guanosine units in ribbons that in turn aggregate in fibrillar or lamellar soft structures. The elucidation of these structures furnishes an explanation to the physical behaviour of guanosine units which display organogelator properties. Chapter 4 will describe photoresponsive self-assembling systems. Numerous research examples have demonstrated that the use of photochromic molecules in supramolecular self-assemblies is the most reasonable method to noninvasively manipulate their degree of aggregation and supramolecular architectures. In section 4.4 we report on the photocontrolled self-assembly of modified guanosine nucleobase E-42: by the introduction of a photoactive moiety at C8 it is possible to operate a photocontrol over the self-assembly of the molecule, where the existence of G-quartets can be alternately switched on and off. In section 4.5 we focus on the use of cyclodextrins as photoresponsive host-guest assemblies: αCD–azobenzene conjugates 47-48 (section 4.5.3) are synthesized in order to obtain a photoresponsive system exhibiting a fine photocontrollable degree of aggregation and self-assembled architecture. Finally, Chapter 5 contains the experimental protocols used for the research described in Chapters 2-4.

Nanopartikel durch Strukturfixierung mizellarer Assoziate aus amphiphilen, endgruppenfunktionalisierten Diblockcopolymeren Zwei unterschiedliche Diblockcopolymersysteme mit Molmassen unterhalb von Mw = 10 000 g/mol wurden über anionische Polymerisation synthetisiert. Ein hetero-telecheles a,w-Poly(dimethylsiloxan)-b-Poly(ethylenoxid) (PDMS-PEO) Diblockcopolymer wurde mit einer Methacrylatendgruppe am PDMS und entweder einer Benzyl-, Hydroxy- oder Carboxylatendgruppe am PEO funktionalisiert. Ein Poly(butadien)-b-Poly(ethylenoxid) (PB-PEO) Diblockcopolymer wurde am PEO ebenfalls entweder mit einer Benzyl-, Hydroxy- oder Carboxylatendgruppe funktionalisiert. In selektiven Lösungsmitteln wie Wasser oder Methanol bilden beide Diblockcopolymersysteme supramolekulare Strukturen mit sphärischer, zylindrischer oder toroider Geometrie aus, die mit statischer und dynamischer Lichtstreuung in Lösung und mit Rasterkraftmikroskopie (AFM) und Transmissionselektronenmikroskopie (TEM) auf der Oberfläche untersucht wurden. Durch Zusatz eines Vernetzers und Initiators wurden die selbstassoziierenden Mizellen des PDMS-PEO Diblockcopolymers permanent durch radikalische Polymerisation mit UV-Licht fixiert. Mizellen des PB-PEO Diblockcopolymers wurden über Bestrahlung mit gamma-Strahlen permanent fixiert. Die Untersuchung der resultierenden Nanopartikel beider Diblockcopolymersysteme mit AFM und TEM zeigte, daß diese sogar in nicht selektiven Lösungsmitteln wie Tetrahydrofuran formstabil bleiben.

Although nickel is a toxic metal for living organisms in its soluble form, its importance in many biological processes recently emerged. In this view, the investigation of the nickel-dependent enzymes urease and [NiFe]-hydrogenase, especially the mechanism of nickel insertion into their active sites, represent two intriguing case studies to understand other analogous systems and therefore to lead to a comprehension of the nickel trafficking inside the cell. Moreover, these two enzymes have been demonstrated to ensure survival and colonization of the human pathogen H. pylori, the only known microorganism able to proliferate in the gastric niche. The right nickel delivering into the urease active site requires the presence of at least four accessory proteins, UreD, UreE, UreF and UreG. Similarly, analogous process is principally mediated by HypA and HypB proteins in the [NiFe]-hydrogenase system. Indeed, HpHypA and HpHypB also have been proposed to act in the activation of the urease enzyme from H. pylori, probably mobilizing nickel ions from HpHypA to the HpUreE-HpUreG complex. A complete comprehension of the interaction mechanism between the accessory proteins and the crosstalk between urease and hydrogenase accessory systems requires the determination of the role of each protein chaperone that strictly depends on their structural and biochemical properties. The availability of HpUreE, HpUreG and HpHypA proteins in a pure form is a pre-requisite to perform all the subsequent protein characterizations, thus their purification was the first aim of this work. Subsequently, the structural and biochemical properties of HpUreE were investigated using multi-angle and quasi-elastic light scattering, as well as NMR and circular dichroism spectroscopy. The thermodynamic parameters of Ni2+ and Zn2+ binding to HpUreE were principally established using isothermal titration calorimetry and the importance of key histidine residues in the process of binding metal ions was studied using site-directed mutagenesis. The molecular details of the HpUreE-HpUreG and HpUreE-HpHypA protein-protein assemblies were also elucidated. The interaction between HpUreE and HpUreG was investigated using ITC and NMR spectroscopy, and the influence of Ni2+ and Zn2+ metal ions on the stabilization of this association was established using native gel electrophoresis, light scattering and thermal denaturation scanning followed by CD spectroscopy. Preliminary HpUreE-HpHypA interaction studies were conducted using ITC. Finally, the possible structural architectures of the two protein-protein assemblies were rationalized using homology modeling and docking computational approaches. All the obtained data were interpreted in order to achieve a more exhaustive picture of the urease activation process, and the correlation with the accessory system of the hydrogenase enzyme, considering the specific role and activity of the involved protein players. A possible function for Zn2+ in the chaperone network involved in Ni2+ trafficking and urease activation is also envisaged.

ZusammenfassungDie selbstkondensierende Gruppenübertragungspolymerisation von 2-[(2-Methyl-1-triethylsiloxy-1-propenyl)oxy]ethyl-methacrylat (MTSHEMA) und die Copolymerisation mit Methylmethacrylat und tert-Butylmethacrylat wurde untersucht. Da MTSHEMA eine polymerisierbare Methacryloyl-Einheit und eine zur Initiierung einer Gruppenübertragungspolymerisation befähigte Silylketenacetal-Einheit besitzt, führt die Homopolymerisation zu hyperverzweigten und die Copolymerisation zu hochverzweigten Polymeren.Bei der Homopolymerisation von MTSHEMA konnten nur niedrige Molekulargewichte erreicht werden. Dies wird auf Nebenreaktionen der aktiven Kettenenden zurückgeführt, welche die Carbonylgruppen nucleophil angreifen und, mit der Doppelbindung Kern-Einheit reagieren. Die Copolymerisation mit Methylmethacrylat verlauft ohne Nebenreaktionen. Durch die Variation des molaren Verhältnisses von MTSHEMA zu den Comonomeren war es möglich, das Molekulargewicht, den Verzweigungsgrad und dadurch die Viskosität in Lösung zu kontrollieren. Die Bestimmung der Molekulargewichtsverteilung sämtlicher Polymere erfolgte durch Kopplung der Gelpermeationschromatographie mit einem Viskositätsdetektor und einem Vielwinkel Lichtstreu-Photometer. Die aus dem Vergleich der Viskositäten und Trägheitsradien ermittelten Schrumpfungspa-rameter lassen Schlüsse auf den Verzweigungsgrad zu.Nach den Ergebnissen der viskoelastischen Spektroskopie folgt das Verhalten der verzweigten Polymere in der Schmelze der Rouse-Theorie und deutet damit auf die Abwesenheit von Verschlaufungen hin.Durch die Copolymerisation mit tert-Butylmethacrylat und MTSHEMA konnte hochverzweigtes Poly(tert-butylmethacrylat) synthetisiert werden. Die Verseifung dieser Polymere ergab verzweigte Polymethacrylsäure.

Zusammenfassung: Es sollten zum einen strukturell fixierte, perlenkettenartige Polyelektrolyte dargestellt werden. Dazu wurde lineares Poly-2-vinylpyridin(PVP) mit vinylfunktionalisierten, hydrophoben Quaternisierungsagentien zu einer Polyseife umgesetzt. Bei der Quaternisierungsreaktion ließ sich der Gehalt an hydrophoben Gruppen variieren, wodurch Polyseifen mit unterschiedlichen Ladungsdichten zugänglich wurden. Trotz vielfältiger Versuche war es dennoch nicht möglich, eine polymerisationsfähige Polyseife herzustellen, welche in wäßriger Lösung intramolekular micellisiert und die Überstruktur einer Perlenkette annimmt. Durch die Herstellung hochreiner PVP-Makromonomere konnten zylindrische Bürsten hergestellt werden. Durch Umsetzung der PVP-Bürsten mit Methyltosylat sind unter milden Reaktionsbedingungen nahezu vollständig umgesetzte positiv geladene Polyelektrolyte zugänglich. Durch eine Sulfonierung von Polystyrol-Polymakromonomeren wurden negativ geladene zylindrische Polyelektrolyte erhalten.Das Verhalten dieser Polyelektrolyte in verdünnter wäßriger Lösung wurde mit der statischen und der dynamischen Lichtstreuung untersucht. Dabei deuten die statischen Messungen darauf hin, daß deren Verhalten in verdünnter wäßriger Lösung maßgeblich durch die osmotische Aktivität der Gegenionen bestimmt wird.Durch eine Quaternisierung der PVP-Bürsten mit langkettigen Reagentien konnten hochverzweigte Polyelektrolytarchitekturen hergestellt werden. Dabei läßt sich die Tatsache, daß eine Quaternisierung mit solchen Reagentien einen nur unwesentlichen Einfluß auf die Struktur der Bürste hat, nicht durch einfache Überlegungen erklären. Dennoch scheinen die langkettigen Seitenketten die Ausbildung geordneter Strukturen innerhalb von Domänen an der Oberfläche zu induzieren.
