Despite the socioeconomic importance of walnut trees, poor rooting and recalcitrance to in vitro culture have hampered the establishment of high-yield clonal plantations. To improve walnut micropropagation, we introduced several modifications to current methods and evaluated the effects on microshoot performance and acclimatization. Nine selected genotypes (13-year-old trees) of the commercial hybrid Juglans major 209 x J. regia were cultured in vitro on DKW-C medium supplemented with 4.4 M BA and 50 M IBA. A protocol was developed that relies on the use of 0.40 mM phloroglucinol during shoot multiplication, 0.20 mM previous root induction, and 6.81 mg/L Fe3+ (FeEDDHA). Moreover, the addition of 83.2 M glucose during the root expression phase significantly improved plant survival during acclimatization. Phloroglucinol promoted microshoot elongation but inhibited rooting, especially at concentrations above 0.40 mM. Replacing FeEDTA by FeEDDHA diminished chlorotic symptoms and improved rooting, with up to 90% microshoots developing viable roots. Likewise, glucose was more efficient than sucrose or fructose in promoting plant survival. At the proposed working concentrations, neither glucose nor FeEDDHA caused any noticeable deleterious effect on walnut micropropagation. Microscopic analysis revealed the physical continuity between adventitious roots and stem pericycles. Analysis of leaf genomic DNA with eight polymorphic microsatellite markers was supportive of the clonal fidelity and genetic stability of the micropropagated material. Successful clonal plantations (over 5,800 ramets) have been established by applying this protocol.

We have studied the adsorption of two structurally similar forms of hemoglobin (met-Hb and HbCO) to a hydrophobic self-assembled methyl-terminated thiol monolayer on a gold surface, by using a Quartz Crystal Microbalance (QCM) technique. This technique allows time-resolved simultaneous measurements of changes in frequency (f) (c.f. mass) and energy dissipation (D) (c.f. rigidity/viscoelastic properties) of the QCM during the adsorption process, which makes it possible to investigate the viscoelastic properties of the different protein layers during the adsorption process. Below the isoelectric points of both met-Hb and HbCO, the D vs. f graphs displayed two phases with significantly different slopes, which indicates two states of the adsorbed proteins with different visco-elastic properties. The slope of the first phase was smaller than that of the second phase, which indicates that the first phase was associated with binding of a more rigidly attached, presumably denatured protein layer, whereas the second phase was associated with formation of a second layer of more loosely bound proteins. This second layer desorbed, e.g., upon reduction of Fe3+ of adsorbed met-Hb and subsequent binding of carbon monoxide (CO) forming HbCO. Thus, the results suggest that the adsorbed proteins in the second layer were in a native-like state. This information could only be obtained from simultaneous, time-resolved measurements of changes in both D and f, demonstrating that the QCM technique provides unique information about the mechanisms of protein adsorption to solid surfaces.

The crystal structures of cytochrome c oxidase from both bovine and Paracoccus denitrificans reveal two putative proton input channels that connect the heme-copper center, where dioxygen is reduced, to the internal aqueous phase. In this work we have examined the role of these two channels, looking at the effects of site-directed mutations of residues observed in each of the channels of the cytochrome c oxidase from Rhodobacter sphaeroides. A photoelectric technique was used to monitor the time-resolved electrogenic proton transfer steps associated with the photo-induced reduction of the ferryl-oxo form of heme a3 (Fe4+ = O2) to the oxidized form (Fe3+OH). This redox step requires the delivery of a chemical H+ to protonate the reduced oxygen atom and is also coupled to proton pumping. It is found that mutations in the K channel (K362M and T359A) have virtually no effect on the ferryl-oxo-to-oxidized (F-to-Ox) transition, although steady-state turnover is severely limited. In contrast, electrogenic proton transfer at this step is strongly suppressed by mutations in the D channel. The results strongly suggest that the functional roles of the two channels are not the separate delivery of chemical or pumped protons, as proposed recently [Iwata, S., Ostermeier, C., Ludwig, B. & Michel, H. (1995) Nature (London) 376, 660669]. The D channel is likely to be involved in the uptake of both chemical and pumped protons in the F-to-Ox transition, whereas the K channel is probably idle at this partial reaction and is likely to be used for loading the enzyme with protons at some earlier steps of the catalytic cycle. This conclusion agrees with different redox states of heme a3 in the K362M and E286Q mutants under aerobic steady-state turnover conditions.

Incubation of Na/K-ATPase with ascorbate plus H2O2 produces specific cleavage of the subunit. Five fragments with intact C termini and complementary fragments with intact N termini were observed. The subunit is not cleaved. Cleavages depend on the presence of contaminant or added Fe2+ ions, as inferred by suppression of cleavages with nonspecific metal complexants (histidine, EDTA, phenanthroline) or the Fe3+-specific complexant desferrioxamine, or acceleration of cleavages by addition of low concentrations of Fe2+ but not of other heavy metal ions. Na/K-ATPase is inactivated in addition to cleavage, and both effects are insensitive to OH radical scavengers. Cleavages are sensitive to conformation. In low ionic strength media (E2) or media containing Rb ions [E2(Rb)], cleavage is much faster than in high ionic strength media (E1) or media containing Na ions (E1Na). N-terminal fragments and two C-terminal fragments (N-terminals E214 and V712) have been identified by amino acid sequencing. Approximate positions of other cleavages were determined with specific antibodies. The results suggest that Fe2+ (or Fe3+) ions bind with high affinity at the cytoplasmic surface and catalyze cleavages of peptide bonds close to the Fe2+ (or Fe3+) ion. Thus, cleavage patterns can provide information on spatial organization of the polypeptide chain. We propose that highly conserved regions of the subunit, within the minor and major cytoplasmic loops, interact in the E2 or E2(Rb) conformations but move apart in the E1 or E1Na conformations. We discuss implications of domain interactions for the energy transduction mechanism. Fe-catalyzed cleavages may be applicable to other P-type pumps or membrane proteins.

A mechanism of ion transport across membranes is reported. Microbial transport of Fe3+ generally delivers iron, a growth-limiting nutrient, to cells via highly specific siderophore-mediated transport systems. In contrast, iron transport in the fresh water bacterium Aeromonas hydrophila is found to occur by means of an indiscriminant siderophore transport system composed of a single multifunctional receptor. It is shown that (i) the siderophore and Fe3+ enter the bacterium together, (ii) a ligand exchange step occurs in the course of the transport, and (iii) a redox process is not involved in iron exchange. To the best of our knowledge, there have been no other reports of a ligand exchange mechanism in bacterial iron transport. The ligand exchange step occurs at the cell surface and involves the exchange of iron from a ferric siderophore to an iron-free siderophore already bound to the receptor. This ligand exchange mechanism is also found in Escherichia coli and seems likely to be widely distributed among microorganisms.

A Araucaria angustifolia uma confera endmica das regies sul e sudeste do Brasil sendo considerada uma espcie em extino devido ao extenso extrativismo madeireiro. Atualmente, existem inmeros projetos visando o reflorestamento e o uso sustentvel deste pinheiro. Em vista destes pontos, o estudo das propriedades dos componentes das folhas com o intuito da utilizao destes com fins comerciais tornou-se de extrema importncia. As suas folhas foram submetidas extrao com solventes e foram identificados seis biflavonides majoritrios, dentre estes a amentoflavona e a ginkgetina, que so apontados como agentes contra inflamaes e artrites. A frao rica de biflavonides (BFF) extrada da araucaria foi testada frente a sua atividade em proteger contra danos em biomolculas provocadas por espcies reativas de oxignio, capacidade em quelar metais e proteo contra raios UV. A capacidade do BFF em proteger contra danos provocados por espcies reativas de oxignio foi comparado com compostos conhecidamente antioxidantes, como o α-tocoferol, Trolox®, quercetina, rutina e com padres de biflavonides, a amentoflavona e ginkgetina. O BFF demonstrou que possui uma constante de supresso do 1O2 (50 x 106 M-1s-1), superior ao da quercetina (9 x 106 M-1s-1) e foi o mais eficiente na proteo contra quebras de simples fita em DNA plasmidial, provocado por esta espcie reativa. Ainda em relao proteo de DNA plasmidial o BFF foi capaz de proteger tambm contra estes danos provocados atravs da reao de Fenton, apesar de no demonstrar a mesma eficincia da quercetina que mostrou ser um potente protetor destes danos. O BFF protegeu contra lipoperoxidao em lipossomos de fosfatidilcolina induzida por raios UV e reao de Fenton. Em anlises realizadas com espectrometria de massas foi observada a formao de complexos destes biflavonides com ons metlicos como ferro, cobre e alumnio que possuem um papel importante na formao de radicais livres. Em relao capacidade fotoprotetora do BFF, este inibiu a formao de dmeros de pirimidina que so apontados como causadores de cncer de pele induzidos, principalmente por radiao UV-B. Esta ao protetora foi superior quela conferida ao p-metoxicinamato de octila, um conhecido fotoprotetor. Com o intuito de permitir a solubilizao do BFF em solues aquosas e assim, avaliar a ao do BFF em clulas, incorporou-se o BFF em ciclodextrina. Essa incluso favoreceu a incorporao de BFF em clulas CV1-P na concentrao aproximada de 0,4 µg/ml aps 24 horas de incubao. Essa concentrao incorporada no demonstrou ser txica para as clulas no teste com MTT. Assim, o BFF tem despertado grande interesse em relao ao seu potencial na utilizao nas mais variadas reas como cosmtica, alimentos e fitoterpicos.

Catalytically active heterogeneous catalysts have been prepared via microwave deposition of iron oxide nanoparticles (0.51.2 wt%) on MCM-41 type silica materials with different morphologies (particles, helical and spheres). This methodology leads to iron oxide nanoparticles composed by a mixture of FeO and Fe2O3 species, being the Fe(II)/Fe(III) peak ratio near to 1.11 by XPS. DRUV spectroscopy indicates the presence of tetrahedral coordinated Fe3+ in the silica framework of the three catalysts as well as some extraframework iron species in the catalysts with particle and sphere-like morphologies. The loading of the nanoparticles does neither affect the mesopore arrangement nor the textural properties of the silica supports, as indicated by SAXS and nitrogen adsorption/desorption isotherms. A detailed investigation of the morphology of the supports in various microwave-assisted catalyzed processes shows that helical mesostructures provide optimum catalytic activities and improved reusabilities in the microwave-assisted redox (selective oxidation of benzyl alcohol) catalyzed process probably due to a combination of lower particle size and higher acidity in comparison with the supports with particle and sphere morphology.

The electroassisted encapsulation of Single-Walled Carbon Nanotubes was performed into silica matrices (SWCNT@SiO2). This material was used as the host for the potentiostatic growth of polyaniline (PANI) to yield a hybrid nanocomposite electrode, which was then characterized by both electrochemical and imaging techniques. The electrochemical properties of the SWCNT@SiO2-PANI composite material were tested against inorganic (Fe3+/Fe2+) and organic (dopamine) redox probes. It was observed that the electron transfer constants for the electrochemical reactions increased significantly when a dispersion of either SWCNT or PANI was carried out inside of the SiO2 matrix. However, the best results were obtained when polyaniline was grown through the pores of the SWCNT@SiO2 material. The enhanced reversibility of the redox reactions was ascribed to the synergy between the two electrocatalytic components (SWCNTs and PANI) of the composite material.

This study evaluates the application of denim fiber scraps as a precursor for the synthesis of adsorbents for water treatment via pyrolysis and their application in water defluoridation. The best pyrolysis conditions for the synthesis of this novel adsorbent have been identified and a metal doping route with different salts of Al3 +, La3 + and Fe3 + was proposed to improve its fluoride adsorption behavior. Different spectroscopic and microscopic techniques (i.e., FTIR, XPS, XRF, SEM) were used to characterize the precursor and adsorbents, and to analyze the surface interactions involved in the fluoride removal mechanism. Experimental results showed that these adsorbents were effective for fluoride adsorption showing uptakes up to 4.25 mg/g. The Si-OmetalF interactions appear to be highly relevant for the fluoride removal. This study highlights the potential of denim textile waste as a raw material for the production of added-value products, thus minimizing their associated disposal cost. It also shows the performance of denim textile waste as a precursor of adsorbents for addressing relevant environmental concerns such as fluoride pollution.

Major elements, S, F, Cl concentrations and relative proportions of S6+ to total S were analyzed with electron microprobe in sideromelane glass shards from Pleistocene volcaniclastic sediments drilled during ODP Leg 157. Glasses are moderately to strongly evolved and represent a spectrum from alkali basalt, basanite and nephelinite through hawaiite, mugearite and tephrite to phonolitic tephrite. Measured S6+/SumS (0.030.98) and calculated Fe2+/Fe3+ (2.55.8) ratios in the melt yield preeruptive redox conditions ranging from NNO-1.4 to NNO+2.1. The morphology of the glass shards, variations of S and Cl concentrations (0.0100.127 wt% S, 0.0180.129 wt% Cl), calculated preeruptive temperatures (10301200 C) and oxygen fugacities suggest that glasses deposited even within the same ash layers have diverse origin and may have resulted from both submarine and subaerial eruptions. Most vesicle-free glasses are characterized by high concentrations of S and represent undegassed or slightly degassed submarine lavas, whereas vesiculated glasses with low concentrations of S and Cl are strongly degassed and can be ascribed to the eruptions in shallow water or on land. Sideromelane glass shards at Sites 953 are thought to have resulted from submarine eruptions northeast of Gran Canaria, glasses at Site 954 represent mostly volcaniclastic material of shallow water submarine and subaerial eruptions on Gran Canaria and Tenerife, and glasses deposited at Site 956 resulted from submarine or explosive eruptions on Tenerife.

57Fe Mssbauer spectra of 15 oceanic sediment samples collected from Site 612 (Deep Sea Drilling Project Leg 95) were recorded. These spectra showed that most of the iron in the sediments was present as high-spin, paramagnetic Fe2+ and Fe3+. The ferrous iron was mainly distributed in terrigenous clays and biogenic carbonates. The variation of the Mssbauer parameters for Fe2+ with sub-bottom depth suggests that the main Fe2+-bearing component changed with geologic time. The amount of iron in each iron-bearing phase as estimated from the corresponding peak areas in the spectra also changed with depth. These variations in the Mssbauer parameters and peak areas are correlated with lithologic changes in the sediment column.

The compositions of chrome spinels of Costa Rica Rift basalts from Deep Sea Drilling Project Site 505 vary depending on their occurrences as (1) inclusions in olivine crystals, (2) inclusions in Plagioclase crystals, and (3) isolated crystals in variolitic or glassy samples. The variations are a consequence of (1) changes of melt compositions as crystallization proceeds, and (2) contrasting behavior of olivine and Plagioclase in competition with spinels for Al and Mg. Some spinels have skeletal rims compositionally less magnesian than mineral cores; however, the cores do not appear to be xenocrysts, unlike some texturally similar spinels in Mid-Atlantic Ridge basalts.

There has been much recent interest in the origin of silicic magmas at spreading centres away from any possible influence of continental crust. Here we present major and trace element data for 29 glasses (and 55 whole-rocks) sampled from a 40 km segment of the South East Rift in the Manus Basin that span the full compositional continuum from basalt to rhyolite (50-75 wt % SiO2). The glass data are accompanied by Sr-Nd-Pb, O and U-Th-Ra isotope data for selected samples. These overlap the ranges for published data from this part of the Manus Basin. Limited increases in Cl/K ratios with increasing SiO2, La-SiO2 and Yb-SiO2 relationships, and the oxygen isotope data rule out models in which the more silicic lavas result from partial melting of altered oceanic crust or altered oceanic gabbros. Rather, the data form a coherent array that is suggestive of closed-system fractional crystallization and this is well simulated by MELTS models run at 0.2 GPa and QFM (quartz-fayalite-magnetite buffer) with 1 wt % H2O, using a parental magma chosen from the basaltic glasses. Although some assimilation of altered oceanic crust or gabbro cannot be completely ruled out, there is no evidence that this plays an important role in the origin of the silicic lavas. The U-series disequilibria are dominated by 238U and 226Ra excesses that limit the timescale of differentiation to less than a few millennia. Overall, the data point to rapid evolution in relatively small magma lenses located near the base of thick oceanic crust; we speculate that this was coupled with relatively low rates of basaltic recharge. A similar model may be applicable to the generation of silicic magmas elsewhere in the ocean basins.

Volcanic ash layers (1-3 cm thick) are abundant in the North Aoba Basin drill sites but less common at forearc sites. Ash deposited on the forearc slopes is liable to be redistributed as turbidites. In addition, the westerly upper winds also minimize ash-fall on the western (forearc) side of the New Hebrides Island Arc. Crystalline components in the ashes are primarily plagioclase (An90-An44), clinopyroxene (Ca46Mg49Fe5-Ca43Mg33Fe24), olivine (Fo87-Fo62), and titanomagnetite. There are also small amounts of orthopyroxene, magnetite, apatite, and quartz. Glass shards occur in most of the ashes and range in composition from basalt to rhyolite. There is often a variety of glass compositions within a single ash layer. One explanation for this is that the rate of accumulation of ash from several different eruptions or eruptive phases exceeded the background sedimentation rate: there may also have been a certain amount of reworking. The high-K and low-K trends previously recognized in volcanic rocks from the New Hebrides Island Arc are clearly represented in the Leg 134 glasses. All of the ashes investigated here are thought to have originated from the Central Chain volcanoes. The source of the high-K group was probably the Central Basin volcanoes of Santa Maria, Aoba, and Ambrym. The lower-K series includes a distinctive group of dacites and is likely to have originated from the Epi-Tongoa-Tongariki sector of the arc where major pyroclastic eruptions, associated with caldera collapse, have occurred during the Holocene, perhaps as recently as 400 yr ago.

Mssbauer analyses were conducted on a sample of saponite selected from DSDP Leg 69 basalt core. The sample was initially placed within a nitrogen-purged container on-board Glomar Challenger approximately three hours after recovery, where it remained until analysis. The Mssbauer data revealed an original, in situ Fe2O3/FeO ratio of 0.46, with both Fe**2+ and Fe**3+ in octahedral coordination. With controlled exposure to air under ambient laboratory storage conditions, the proportion of Fe**3+ increased from an original 30% to 51% over a period of about 11.5 months. The Fe**3+ thus produced remained in octahedral coordination, and no observable changes occurred in the physical appearance of the sample.
