Rhomboid intramembrane proteases are the enzymes that release active epidermal growth factor receptor (EGFR) ligands in Drosophila and C. elegans, but little is known about their functions in mammals. Here we show that the mammalian rhomboid protease RHBDL4 (also known as Rhbdd1) promotes trafficking of several membrane proteins, including the EGFR ligand TGFα, from the endoplasmic reticulum (ER) to the Golgi apparatus, thereby triggering their secretion by extracellular microvesicles. Our data also demonstrate that RHBDL4-dependent trafficking control is regulated by G-protein coupled receptors, suggesting a role for this rhomboid protease in pathological conditions, including EGFR signaling. We propose that RHBDL4 reorganizes trafficking events within the early secretory pathway in response to GPCR signaling. Our work identifies RHBDL4 as a rheostat that tunes secretion dynamics and abundance of specific membrane protein cargoes.

The marine cycle of calcium carbonate (CaCO3) is an important element of the carbon cycle and co-governs the distribution of carbon and alkalinity within the ocean. However, CaCO3 export fluxes and mechanisms governing CaCO3 dissolution are highly uncertain. We present an observationally constrained, probabilistic assessment of the global and regional CaCO3 budgets. Parameters governing pelagic CaCO3 export fluxes and dissolution rates are sampled using a Monte Carlo scheme to construct a 1000-member ensemble with the Bern3D ocean model. Ensemble results are constrained by comparing simulated and observation-based fields of excess dissolved calcium carbonate (TA*). The minerals calcite and aragonite are modelled explicitly and ocean–sediment fluxes are considered. For local dissolution rates, either a strong or a weak dependency on CaCO3 saturation is assumed. In addition, there is the option to have saturation-independent dissolution above the saturation horizon. The median (and 68 % confidence interval) of the constrained model ensemble for global biogenic CaCO3 export is 0.90 (0.72–1.05) Gt C yr−1, that is within the lower half of previously published estimates (0.4–1.8 Gt C yr−1). The spatial pattern of CaCO3 export is broadly consistent with earlier assessments. Export is large in the Southern Ocean, the tropical Indo–Pacific, the northern Pacific and relatively small in the Atlantic. The constrained results are robust across a range of diapycnal mixing coefficients and, thus, ocean circulation strengths. Modelled ocean circulation and transport timescales for the different set-ups were further evaluated with CFC11 and radiocarbon observations. Parameters and mechanisms governing dissolution are hardly constrained by either the TA* data or the current compilation of CaCO3 flux measurements such that model realisations with and without saturation-dependent dissolution achieve skill. We suggest applying saturation-independent dissolution rates in Earth system models to minimise computational costs.

Drawing on extensive academic research and theory on clusters and their analysis, the methodology employed in this pilot study (sponsored by the Welsh Assembly Government’s Economic Research Grants Assessment Board) seeks to create a framework for reviewing and monitoring clusters in Wales on an ongoing basis, and generate the information necessary for successful cluster development policy to occur. The multi-method framework developed and tested in the pilot study is designed to map existing Welsh sectors with cluster characteristics, uncover existing linkages, and better understand areas of strength and weakness. The approach adopted relies on synthesising both quantitative and qualitative evidence. Statistical measures, including the size of potential clusters, are united with other evidence on input-output derived inter-linkages within clusters and to other sectors in Wales and the UK, as well as the export and import intensity of the cluster. Multi Sector Qualitative Analysis is then designed for competencies/capacity, risk factors, markets, types and crucially, the perceived strengths of cluster structures and relationships. The approach outlined above can, with the refinements recommended through the review process, provide policy-makers with a valuable tool for reviewing and monitoring individual sectors and ameliorating problems in sectors likely to decline further.

Natural selection mediated by pollinators has influenced the evolution of floral diversity of the flowering plants (angiosperms). The scope of this thesis was to study: 1) phenotypic selection, 2) mating systems, and 3) floral shifts involved in plant speciation. Model plant species were Platanthera bifolia and P. chlorantha (Orchidaceae). These orchids are moth-pollinated, strictly co-sexual (bisexual flowers), and produce a spike that displays 10-20 white flowers. I explored the influence of characters on plant fitness by using multiple linear regressions. Pollen removal (male fitness) and fruit set (female fitness) increased with more flowers per plant in three P. bifolia populations. There was selection towards longer spurs in a dry year when average spur length was shorter than in normal-wet years. Female function was sensitive to drought, which enabled an application of the male function hypothesis of floral evolution (Bateman's principle). The results show that selection may vary between populations, years, and sex-functions. I examined inbreeding by estimating levels of geitonogamy (self-pollination between flowers of an individual) with an emasculation method in two P. bifolia populations. Geitonogamy did not vary with inflorescence size. Levels of geitonogamy was 20-40% in the smaller, but non-significant in the larger population. This may relate to lower number of possible mates and pollinator activity in the smaller population. Platanthera bifolia exhibits the ancestral character state of tongue-attachment of pollinia on the pollinator. Its close relative P. chlorantha attaches its pollinia onto the pollinator's eyes. To explore the mechanism of a floral shift, pollination efficiency and speed was compared between the two species. The results showed no differences in pollination efficiency, but P. chlorantha had faster pollen export and import. Efficiency of pollination in terms of speed may cause floral shifts, and thus speciation.

A evolução dos transportes de mercadorias, em Portugal e na União Europeia, assume uma enorme repercussão na economia global, de modo que a combinação dos vários modos de transporte, com vista à obtenção de ganhos ao nÃvel da eficiência, adquire extrema importância. Os programas europeus de apoio aos transportes, enquadrados na rede transeuropeia de transportes e plataformas logÃsticas, também contribuem para a otimização do transporte de mercadorias. A importância do transporte ferroviário de mercadorias no eixo Leixões- Salamanca, nomeadamente para as empresas exportadoras e importadoras da região norte e centro, que utilizam meios alternativos ao ferroviário, constitui o principal objetivo desta dissertação. A revisão bibliográfica inclui uma abordagem aos transportes de mercadorias em geral e de forma mais aprofundada aos modos ferroviário e rodoviário na penÃnsula ibérica, passando pela logÃstica, bem como pela integração de modos: intermodalidade e multimodalidade na rede europeia de transportes e ainda a referência aos portos secos e à s plataformas logÃsticas. Isto permite caraterizar as diferentes empresas operadoras do setor dos transportes de mercadorias e os produtos transacionados, assim como enumerar vantagens e/ou desvantagens do meio de transporte ferroviário face a outros meios, mais concretamente no eixo alvo deste estudo. A metodologia utilizada consiste na análise de informação proveniente de fontes secundárias havendo lugar a uma referência mais detalhada sobre as plataformas logÃsticas de Leixões e Salamanca, o eixo E-80, o corredor ferroviário nº4 e os programas europeus promotores da eficiência no transporte ferroviário de mercadorias: Marathon, Ferremed e Marco Polo. Para a recolha de informação primária o instrumento adotado foi a entrevista semiestruturada, efetuada a dois representantes da empresa CP-Carga e a um representante da empresa KLog, ambas as empresas ligadas ao setor dos transportes e logÃstica. A análise e tratamento de toda a informação recolhida possibilitam, desde logo, evidenciar as potencialidades do eixo Leixões-Salamanca no que se refere ao transporte ferroviário de mercadorias, delinear recomendações para a sua otimização, bem como efetuar uma análise SWOT. As considerações finais revelam que é imperativo adotar medidas, de forma integrada, para que o seu efeito na potenciação do transporte ferroviário de mercadorias, não só no eixo Leixões-Salamanca, mas também a nÃvel europeu, se afirme como uma verdadeira alternativa a outros modos, particularmente ao domÃnio rodoviário.

Foreign trade statistics are the main data source to the study of international trade.However its accuracy has been under suspicion since Morgernstern published hisfamous work in 1963. Federico and Tena (1991) have resumed the question arguing thatthey can be useful in an adequate level of aggregation. But the geographical assignmentproblem remains unsolved. This article focuses on the spatial variable through theanalysis of the reliability of textile international data for 1913. A geographical biasarises between export and import series, but because of its quantitative importance it canbe negligible in an international scale.

Printed electronics is an emerging concept in electronics manufacturing and it is in very early development stage. The technology is not stable, design kits are not developed, and flows and Computer Aided Design (CAD) tools are not fixed yet. The European project TDK4PE addresses all this issues and this PFC has been realized on this context. The goal is to develop an XML-based information system for the collection and management of information from the technology and cell libraries developed in TDK4PE. This system will ease the treatment of that information for a later generation of specific Design Kits (DK) and the corresponding documentation. This work proposes a web application to generate technology files and design kits in a formatted way; it also proposes a structure for them and a database implementation for storing the needed information. The application will allow its users to redefine the structure of those files, as well as export and import XML files, between other formats.

Työssä tutustutaan erilaisiin e-portfoliojärjestelmiin, niiden ominaisuuksiin ja toiminnallisuuksiin sekä vertaillaan valittuja e-portfoliojärjestelmiä keskenään yhteensopivuuden ja -toimivuuden näkökulmasta yliopistomaailmassa. Tavoitteena on valita e-portfoliojärjestelmistä monipuolisin ja opiskelijaystävällisin järjestelmä. Järjestelmän tulee tukea opiskelijan elinikäistä oppimista. E-portfolio on sähköisessä muodossa oleva omien töiden kokoelma ja sitä hallitaan e-portfoliojärjestelmällä. E-portfoliojärjestelmissä tärkeässä roolissa ovat portfoliospesifikaatiot, jotka mahdollistavat e-portfolion siirtämisen sellaisenaan toiseen järjestelmään rajapintojen kautta. Työssä ei keskitytä henkilökohtaiseen oppimisympäristöön, joka on käsitteenä laajempi kuin e-portfolio. Työssä vertailtiin avoimeen lähdekoodiin perustuvia, web-pohjaisia e-portfoliojärjestelmiä ja tutkittiin niiden tarjoamaa mahdollisuutta portfolioiden siirtoon ja tuomiseen. Lisäksi tarkasteltiin järjestelmien integroituvuutta muihin järjestelmiin, kuten virtuaalisiin oppimisympäristöihin. Vertailun tuloksena monipuolisin ja opiskelijaystävällisin e-portfoliojärjestelmä on Mahara ja yleisin portfoliospesifikaatio Leap2A.
