O presente trabalho tem por objetivo a descrição de um estágio realizado na DIE - Direcção de Infra-Estruturas, do Exército Português, com a finalidade de elaborar o Trabalho Final de Mestrado (TFM) de Engenharia Civil na área de especialização de Hidráulica. O objetivo principal do estágio foi conceber, dimensionar e projetar redes de distribuição de água e redes de drenagem de águas residuais domésticas e pluviais em unidades militares. Para além do trabalho de engenharia propriamente dito, o estágio incluiu também os trabalhos de campo (reconhecimento dos locais e levantamentos topográficos) e os trabalhos complementares com vista à adjudicação para execução das obras (medições e preparação de documentação). Este trabalho insere-se no âmbito dos sistemas prediais de abastecimento de água e de drenagem de águas residuais, que são fundamentais para o conforto e bem-estar dos seus utilizadores, tanto em novas construções como em obras de remodelação ou recuperação de edifícios. No decorrer deste trabalho serão descritas as atividades principais e os conhecimentos mais relevantes durante o estágio, assim como todos os pormenores teóricos e práticos considerados de maior importância. A escolha deste tema com recurso a um estágio, para realizar o TFM, deveu-se ao facto de poder simultaneamente desenvolver competências, a nível deste tipo de projetos, e enriquecer a formação profissional através da partilha de conhecimentos na Secção de Projetos da Repartição Técnica de Engenharia (RTE), que elabora projetos para o exército Português há mais de oito décadas.

The purpose of this study was to investigate the presence of beta-lactam-resistant bacteria in six different types of Portuguese cheese. The numbers of ampicillin resistant (AMP(r)) bacteria varied from 4.7 x 10(2) to 1.5 x 10(7) CFU/g. Within 172 randomly selected beta-lactam-resistant bacteria, 44 resistant phenotypes were found and 31.4% were multidrug resistant. The majority (85%) of the isolates identified belonged to the Enterobacteriaceae family. The presence of the bla(TEM) gene was detected in 80.9% of the tested isolates. The results suggest that without thermal processing of the milk and good hygienic practices, cheese may act as a vehicle of transfer of beta-lactam-resistant bacteria to the gastrointestinal tract of consumers.

The principal aim of this study was to investigate the possibility of transference to Escherichia coli of β-lactam resistance genes found in bacteria isolated from ready-to-eat (RTE) Portuguese traditional food. From previous screenings, 128 β-lactam resistant isolates (from different types of cheese and of delicatessen meats), largely from the Enterobacteriaceae family were selected and 31.3% of them proved to transfer resistance determinants in transconjugation assays. Multiplex PCR in donor and transconjugant isolates did not detect bla CTX, bla SHV and bla OXY, but bla TEM was present in 85% of them, while two new TEMs (TEM-179 and TEM-180) were identified in two isolates. The sequencing of these amplicons showed identity between donor and transconjugant genes indicating in vitro plasmid DNA transfer. These results suggest that if there is an exchange of genes in natural conditions, the consumption of RTE foods, particularly with high levels of Enterobacteriaceae, can contribute to the spread of antibiotic resistance.

Studies on microbial characterization of cold-smoked salmon and salmon trout during cold storage were performed on samples available in the Portuguese market. Samples were also classified microbiologically according to guidelines for ready-to-eat (RTE) products. Further investigations on sample variability and microbial abilities to produce tyramine and histamine were also performed. The coefficient of variation for viable counts of different groups of microorganisms of samples collected at retail market point was high in the first 2 wk of storage, mainly in the Enterobacteriaceae group and aerobic plate count (APC), suggesting that microbiological characteristics of samples were different in numbers, even within the same batch from the same producer. This variation seemed to be decreased when storage and temperature were controlled under lab conditions. The numbers of Enterobacteriaceae were influenced by storage temperature, as indicated by low microbial numbers in samples from controlled refrigeration. Lactic acid bacteria (LAB) and Enterobacteriaceae were predominant in commercial products, a significant percentage of which were tyramine and less histamine producers. These results might be influenced by (1) the technological processes in the early stages of production, (2) contamination during the smoking process, and (3) conditions and temperature fluctuations during cold storage at retail market point of sale.

OBJECTIVE: To describe food habits and dietary intakes of athletic and non-athletic adolescents in Switzerland. SETTING: College, high schools and professional centers in the Swiss canton of Vaud. METHOD: A total of 3,540 subjects aged 9-19 y answered a self-reported anonymous questionnaire to assess lifestyles, physical plus sports activity and food habits. Within this sample, a subgroup of 246 subjects aged 11-15 also participated in an in-depth ancillary study including a 3 day dietary record completed by an interview with a dietician. RESULTS: More boys than girls reported engaging in regular sports activities (P<0.001). Adolescent food habits are quite traditional: up to 15 y, most of the respondents have a breakfast and eat at least two hot meals a day, the percentages decreasing thereafter. Snacking is widespread among adolescents (60-80% in the morning, 80-90% in the afternoon). Food habits among athletic adolescents are healthier and also are perceived as such in a higher proportion. Among athletic adolescents, consumption frequency is higher for dairy products and ready to eat (RTE) cereals, for fruit, fruit juices and salad (P<0.05 at least). Thus the athletic adolescent's food brings more micronutrients than the diet of their non-athletic counterparts. Within the subgroup (ancillary study), mean energy intake corresponds to requirements for age/gender group. CONCLUSIONS: Athletic adolescents display healthier food habits than non-athletic adolescents: this result supports the idea that healthy behavior tends to cluster and suggests that prevention programs among this age group should target simultaneously both sports activity and food habits.

El present projecte té com a objecte d’estudi l’anàlisi de la situació ambiental en que es troben els ETR de l’àrea d’influència del Parc del Garraf (PG). En una primera fase, s’inventarien i analitzen els 12 ETR més propers als límits del sistema natural del PG, Parc d’Olèrdola i Foix a partir d’una ecofitxa de criteris ambientals d’àmbit general. Posteriorment, s’estudia la viabilitat d’implantació del Distintiu de Garantia de Qualitat Ambiental (DGQA) en els 6 ETR pilot escollits a partir d’una segona ecofitxa creada a partir dels criteris del distintiu. D’aquest detallat estudi s’extreuen relacions interessants com per exemple, que els ETR més aïllats del nucli urbà fan servir el doble de sistemes per obtenir energia que els establiments ubicats al recinte urbà i que actualment, el preu segueix estant més associat a confort que no pas a compromís ambiental. Finalment, s’elaboren unes propostes de millora i els pressupostos associats per que aquests ETR puguin complir les exigències del distintiu.

This review provides an update on the fruit and vegetable review conducted in 2007. It collates and considers the information available – academic, regulatory, public health – on the health and food safety implications of fruit and vegetables. It focuses on ready-to-eat (RTE) fresh fruit and vegetables which are consumed raw, whether whole or prepared, and includes products packaged under vacuum or in a modified atmosphere that have not undergone any treatment (chemical, physical or biological) to ensure preservation, other than chilling. The key finding was that 75% of Irish consumers are now able to identify that they should eat five portions of fruit and vegetables a day, but only a third (36%) of consumers say they are doing so. The average intake of fruit and vegetables is currently less than half that recommended, at 2.4 portions per day. (Résumé de l'ouvrage) Dürfen Forscherinnen und Forscher Embryonen und Föten für den medizinischen Fortschritt verwenden? Wie soll die Verwendung von medizinischen Daten geregelt werden, welche für andere Zwecke erfasst wurden (z. B. Blutproben)? Worin bestehen die Hoffnungen und Grenzen der klinischen Tests? Wie verhält sich die Politik bzw. die Gesellschaft angesichts dieser brisanten wissenschaftspolitischen Fragen? Das Bundesgesetz über die Forschung am Menschen wird Bereiche regeln, die bisher kaum oder überhaupt nicht gesetzlich erfasst sind. Wegen der Fortschritte in der biomedizinischen Forschung wächst der gesetzliche Regelungsbedarf für Bereiche wie die klinischen Tests, die biomedizinische Forschung an Embryonen und Föten sowie den Umgang mit entnommenem biologischem Material (u. a. Plazenta, Nabelschnurblut, Gewebeproben). Der zweite Band der Reihe «SCIENCE & SOCIETY» enthält Positionen von Wissenschaftlerinnen und Wissenschaftlern aus Natur- und Sozialwissenschaften sowie von Mitgliedern der Eidgenössischen Räte. A numerical procedure for solving the nongray radiative transfer equation (RTE) in two-dimensional cylindrical participating media is presented. Nongray effects are treated by using a narrow-band approach. Radiative emission from CO, CO2, H2O, CH4 and soot is considered. The solution procedure is applied to study radiative heat transfer in a premixed CH4-O2, laminar, flame. Temperature, soot and IR-active species molar fraction distributions are allowed to vary in the axial direction of the flame. From the obtained results it is possible to quantify the radiative loss in the flame, as well as the importance of soot radiation as compared to gaseous radiation. Since the solution procedure is developed for a two-dimensional cylindrical geometry, it can be applied to other combustion systems such as furnaces, internal combustion engines, liquid and solid propellant combustion.

Les temps de réponse dans une tache de reconnaissance d’objets visuels diminuent de façon significative lorsque les cibles peuvent être distinguées à partir de deux attributs redondants. Le gain de redondance pour deux attributs est un résultat commun dans la littérature, mais un gain causé par trois attributs redondants n’a été observé que lorsque ces trois attributs venaient de trois modalités différentes (tactile, auditive et visuelle). La présente étude démontre que le gain de redondance pour trois attributs de la même modalité est effectivement possible. Elle inclut aussi une investigation plus détaillée des caractéristiques du gain de redondance. Celles-ci incluent, outre la diminution des temps de réponse, une diminution des temps de réponses minimaux particulièrement et une augmentation de la symétrie de la distribution des temps de réponse. Cette étude présente des indices que ni les modèles de course, ni les modèles de coactivation ne sont en mesure d’expliquer l’ensemble des caractéristiques du gain de redondance. Dans ce contexte, nous introduisons une nouvelle méthode pour évaluer le triple gain de redondance basée sur la performance des cibles doublement redondantes. Le modèle de cascade est présenté afin d’expliquer les résultats de cette étude. Ce modèle comporte plusieurs voies de traitement qui sont déclenchées par une cascade d’activations avant de satisfaire un seul critère de décision. Il offre une approche homogène aux recherches antérieures sur le gain de redondance. L’analyse des caractéristiques des distributions de temps de réponse, soit leur moyenne, leur symétrie, leur décalage ou leur étendue, est un outil essentiel pour cette étude. Il était important de trouver un test statistique capable de refléter les différences au niveau de toutes ces caractéristiques. Nous abordons la problématique d’analyser les temps de réponse sans perte d’information, ainsi que l’insuffisance des méthodes d’analyse communes dans ce contexte, comme grouper les temps de réponses de plusieurs participants (e. g. Vincentizing). Les tests de distributions, le plus connu étant le test de Kolmogorov- Smirnoff, constituent une meilleure alternative pour comparer des distributions, celles des temps de réponse en particulier. Un test encore inconnu en psychologie est introduit : le test d’Anderson-Darling à deux échantillons. Les deux tests sont comparés, et puis nous présentons des indices concluants démontrant la puissance du test d’Anderson-Darling : en comparant des distributions qui varient seulement au niveau de (1) leur décalage, (2) leur étendue, (3) leur symétrie, ou (4) leurs extrémités, nous pouvons affirmer que le test d’Anderson-Darling reconnait mieux les différences. De plus, le test d’Anderson-Darling a un taux d’erreur de type I qui correspond exactement à l’alpha tandis que le test de Kolmogorov-Smirnoff est trop conservateur. En conséquence, le test d’Anderson-Darling nécessite moins de données pour atteindre une puissance statistique suffisante.

Le principal rôle du corps calleux est d’assurer le transfert de l’information entre les hémisphères cérébraux. Du support empirique pour cette fonction provient d’études investiguant la communication interhémisphérique chez les individus à cerveau divisé (ICD). Des paradigmes expérimentaux exigeant une intégration interhémisphérique de l’information permettent de documenter certains signes de déconnexion calleuse chez ces individus. La présente thèse a investigué le transfert de l’information sous-tendant les phénomènes de gain de redondance (GR), de différence croisé– non-croisé (DCNC) et d’asynchronie bimanuelle chez les ICD et les individus normaux, et a ainsi contribué à préciser le rôle du corps calleux. Une première étude a comparé le GR des individus normaux et des ICD ayant subi une section partielle ou totale du corps calleux. Dans une tâche de détection, le GR consiste en la réduction des temps de réaction (TR) lorsque deux stimuli sont présentés plutôt qu’un seul. Typiquement, les ICD présentent un GR beaucoup plus grand (supra-GR) que celui des individus normaux (Reuter-Lorenz, Nozawa, Gazzaniga, & Hughes, 1995). Afin d’investiguer les conditions d’occurrence du supra-GR, nous avons évalué le GR en présentation interhémisphérique, intrahémisphérique et sur le méridien vertical, ainsi qu’avec des stimuli requérant une contribution corticale différente (luminance, couleur équiluminante ou mouvement). La présence d’un supra-GR chez les ICD partiels et totaux en comparaison avec celui des individus normaux a été confirmée. Ceci suggère qu’une section antérieure du corps calleux, qui perturbe le transfert d’informations de nature motrice/décisionnelle, est suffisante pour produire un supra-GR chez les ICD. Nos données permettent aussi d’affirmer que, contrairement au GR des individus normaux, celui des ICD totaux est sensible aux manipulations sensorielles. Nous concluons donc que le supra-GR des ICD est à la fois attribuable à des contributions sensorielles et motrices/décisionnelles. Une deuxième étude a investigué la DCNC et l’asynchronie bimanuelle chez les ICD et les individus normaux. La DCNC réfère à la soustraction des TR empruntant une voie anatomique « non-croisée » aux TR empruntant une voie anatomique « croisée », fournissant ainsi une estimation du temps de transfert interhémisphérique. Dans le contexte de notre étude, l’asynchronie bimanuelle réfère à la différence de TR entre la main gauche et la main droite, sans égard à l’hémichamp de présentation. Les effets de manipulations sensorielles et attentionnelles ont été évalués pour les deux mesures. Cette étude a permis d’établir une dissociation entre la DCNC et l’asynchronie bimanuelle. Précisément, les ICD totaux, mais non les ICD partiels, ont montré une DCNC significativement plus grande que celle des individus normaux, alors que les deux groupes d’ICD se sont montrés plus asynchrones que les individus normaux. Nous postulons donc que des processus indépendants sous-tendent la DCNC et la synchronie bimanuelle. De plus, en raison de la modulation parallèle du GR et de l’asynchronie bimanuelle entre les groupes, nous suggérons qu’un processus conjoint sous-tend ces deux mesures.

Dans les cas de lymphopénie, les lymphocytes T résiduels prolifèrent exagérément dans un phénomène appelé «expansion homéostatique périphérique» (HPE), qui est efficace pour la régénération des T CD8+, mais inefficace pour les T CD4+. L’interleukine-7 (IL7) est une cytokine homéostatique utilisée afin d’augmenter les comptes lymphocytaires T des patients lymphopéniques. Toutefois, la raison de l’expansion préférentielle des lymphocytes T CD8+ par l’IL7 demeure toujours inconnue. Nous montrons que cette expansion est due au fait que l’IL7 induit une prolifération efficace des T CD8+ périphériques (CD8+PERI) ainsi que des émigrants thymiques CD8+ (CD8+RTEs). Par contre, l’effet prolifératif de l’IL7 est restreint presqu’uniquement aux CD4+RTEs même si les CD4+PERI survivent mieux que les CD4+RTEs. De plus faibles doses d’IL7 sont nécessaires aux CD4+RTEs afin de phosphoryler STAT5 ou de proliférer comparativement aux CD4+PERI et nous démontrons que les contacts TCR/CMHII sont nécessaires à la prolifération induite par l’IL7 des CD4+RTEs en périphérie. De fait, augmenter au Flt3 ligand le nombre de cellules dendritiques périphériques d’une souris donneuse, avant de transférer ses TPERI dans des souris receveuses traitées à l’IL7 induit une prolifération significative des CD4+PERI. Nos résultats indiquent donc que l’abondance des contacts TCR/CMHII reçus dans le thymus semble contrôler la sensibilité à l’IL7 des CD4+RTEs. Finalement, l’observation que les CD8+PERI et CD8+RTEs prolifèrent pareillement pendant la thérapie à l’IL7, alors que la prolifération des T CD4+ est largement restreinte aux RTEs expliquerait pourquoi, dans les cas de lymphopénie, la régénération des T CD4+ est aussi dépendante de la thymopoïèse.

The doubly excited 2s2p ^1P_1 level of Kr^{34+} populated via resonant transfer and excitation (RTE) feeds selectively the metastable ls2s ^1 S_0 state which can only decay via simultaneous emission of two photons to the ground state 1s^2 ^1 S_0. X-ray/X-ray coincidence measurements in heavy ionatom collisions enable the direct measurement of the spectral distribution of the two-photon decay in He-like ions. In addition, we observe strong photon cascades indueed by radiative electron capture.
