This work reports on the photophysical properties of zinc porphyrins meso-tetrakis methylpyridiniumyl (Zn2+TMPyP) and meso-tetrakis sulfonatophenyl (Zn2+TPPS) in homogeneous aqueous solutions and in the presence of sodium dodecyl sulfate (SDS) and cetyltrimethyl ammonium bromide (CTAB) micelles. The excited-state dynamic was investigated with the Z-scan technique, UV-Vis absorption, and fluorescence spectroscopy. Photophysical parameters were obtained by analyzing the experimental data with a conventional five-energy-level diagram. The interaction of the charged side porphyrin groups with oppositely charged surfactants can reduce the electrostatic repulsion between porphyrin molecules leading to aggregation, which affected the porphyrin characteristics such as absorption cross-sections, lifetimes and quantum yields. The interaction between anionic ZnTPPS with cationic CTAB micelles induced the formation of porphyrin J-aggregates, while this effect was not observed in the interaction of ZnTMPyP with SDS micelles. This difference is, probably, due to the difference in electrostatic repulsion between the porphyrin molecules. The insights obtained by these results are important for the understanding of the photophysical behavior of porphyrins, regarding potential applications in pharmacokinetics as encapsulation of photosensitizer for drug delivery systems and in its interaction with cellular membrane.

Sowohl die Komplexierung von Polyelektrolyten mit anorganischen Salzen, als auch die mit entgegengesetzt geladenen Polymeren wurde von vielen Autoren bereits intensiv untersucht. Doch gerade mit Molekülen die zwischen diesen beiden Extremen liegen, sollte es möglich sein, durch elektrostatische Wechselwirkungen gezielt nanometergroße Teilchen definierter Struktur herzustellen. Ziel dieser Arbeit war es deshalb, die Strukturbildung doppelthydrophiler Blockcopolymere mit mehrwertigen organischen Gegenionen zu untersuchen und insbesondere Parameter für die Bildung supramolekularer Strukturen in wässriger Lösung zu finden. Als Blockcopolymer wurde dabei Polyethylenoxid-b-methacrylsäure mittels anionischer Polymerisation hergestellt und mittels Gelpermeationschromatographie (GPC) und Kernresonanzspektroskopie (NMR)charakterisiert. Die Strukturbildung des Polyelektrolyten mit mehrwertigen organischen Gegenionen wurde in pH = 6- und pH = 7-Pufferlösung mit dynamischer und statischer Lichtstreuung, Kleinwinkelneutronenstreuung und Ultrazentrifugation untersucht. Mit Diaminobenzidin als Gegenion wurden dabei sphärische Komplexe mit einem hydrodynamischen Radius um 100 nm erhalten und mit Ultrazentrifugation der Anteil des Gegenions im Komplex quantifiziert. Die schlechte Löslichkeit des Diaminobenzidins in wässrigem Medium erschwerte allerdings die Interpretation der Ergebnisse. Trotzdem deuten diese darauf hin, dass keine Kolloidbildung des Diaminobenzidins, sondern eine Komplexierung der Einzelmoleküle mit dem Copolymer vorliegt. Um Probleme mit der Löslichkeit zu vermeiden, wurden schliesslich Polyamidoamin-Dendrimere als Gegenionen verwendet. Dabei wurde in pH = 6- und pH = 7-Pufferlösung für Dendrimere der Generation 4 mit steigender Gegenionenkonzentration ein kontinuierlicher Anstieg des hydrodynamischen Radius bis zu einer Größe von 70 nm gefunden. Mit Kleinwinkelneutronenstreuung konnte eine ellipsoidale Struktur dieser Komplexe beobachtet werden. Auch die Größe der Gegenionen spielt für die Bildung supramolekularer Aggregate eine Rolle. So zeigte sich, dass für Polyamidoamin-Dendrimere der Generation 2, analog zu denen der Generation 4, ein Anstieg des hydrodynamischen Radius mit steigender Gegenionenkonzentration zu beobachten ist. Für Generation 0-Dendrimere hingegen wurde ein umgekehrter Verlauf beobachtet, welcher dem für Diaminobenzidin gleicht. Somit kann man annehmen, dass die Aggregation mit kleinen Molekülen zu einer anderen Struktur der Komplexe führt, als die mit größeren Molekülen.

Die elektrostatische Wechselwirkung zwischen entgegengesetzt geladenen Polyelektrolyten führt zur spontanen Bildung von Interpolyelektrolytkomplexen. Besonders im Fokus des akademischen und biotechnologischen Interesses stehen derzeit Komplexe aus DNA und synthetischen Polykationen, da eine Anwendung dieser speziellen Interpolyelektrolytkomplexe in der nicht-viralen Gentherapie vielfältig diskutiert wird. Ziel der vorliegenden Arbeit war es, den Einfluss von Kettensteifheit auf die Bildung von Interpolyelektrolytkomplexen zu untersuchen und dabei Wege aufzuzeigen, die eine Kontrolle der Topologie von Interpolyelektrolytkomplexen ermöglichen. Neben dem topologischen Einfluss wurde untersucht, wie durch Komplexierung hochmolekularer Polyelektrolyte equilibrierbare Strukturen erhalten werden können. Als Modellsystem für diese Untersuchungen wurden zylindrische Bürstenpolymere verwendet, denen als topologischer „Kontrast“ das Komplexierungsverhalten kommerzieller PAMAM-G5-Dendrimere mit kugelförmiger Topologie gegenüber gestellt wurde. Um den Ladungsgrad des Bürstenpolymers beliebig variieren zu können, wurden Bürstenpolymere mit Poly(ethylenimin)-Seitenketten synthetisiert, deren Ladungsdichte über den Protonierungsgrad einstellbar ist. Die vorliegende Arbeit zeigt, wie diese mit Hilfe der Makromonomermethode hergestellt werden konnten. Die Komplexbildung von DNA mit semiflexiblen zylindrischen Bürstenpolymeren mit unterschiedlichen Seitenketten und Ladungsdichten in wässriger Lösung hat gezeigt, dass diese in allen untersuchten Fällen unter kinetischer Kontrolle verläuft und Nicht-Gleichgewichtsstrukturen gebildet werden. Sehr überraschend wurde festgestellt, dass die Größen der mit vorgelegter DNA gebildeten Komplexe ungeachtet des verwendeten Polykations identisch sind und DNA-Komplexe mit einem Radius von 30 bis 50 nm und einer kugelförmigen Topologie resultieren. Diese kinetisch kontrollierte Komplexbildung konnte in nicht-wässriger Lösung durch starke Reduktion der Anzahl wechselwirkender Ladungen verhindert werden, so dass eine thermodynamische Kontrolle möglich wurde. Unter diesen Bedingungen ist es gelungen, aus hochgeladenen Poly(styrolsulfonat)-Bürsten mit modifizierten Poly(ethylenimin)-Bürsten oder PAMAM-Dendrimeren Komplexe zylindrischer Topologie herzu-stellen. Für letztere konnte darüber hinaus postuliert werden, dass diese Komplexe eine dichteste Packung der PAMAM-Dendrimere darstellen, für deren Bildung das Polyanion mit seiner größeren Konturlänge und seiner zylindrischen Topologie als Templat dient.

In this work self-assembling model systems in aqueous solution were studied. The systems contained charged polymers, polyelectrolytes, that were combined with oppositely charged counterions to build up supramolecular structures. With imaging, scattering and spectroscopic techniques it was investigated how the structure of building units influences the structure of their assemblies. Polyelectrolytes with different chemical structure, molecular weight and morphology were investigated. In addition to linear polyelectrolytes, semi-flexible cylindrical bottle-brush polymers that possess a defined cross-section and a relatively high persistence along the backbone were studied. The polyelectrolytes were combined with structural organic counterions having charge numbers one to four. Especially the self-assembly of polyelectrolytes with different tetravalent water-soluble porphyrins was studied. Porphyrins have a rigid aromatic structure that has a structural effect on their self-assembly behavior and through which porphyrins are capable of self-aggregation via π-π interaction. The main focus of the thesis is the self-assembly of cylindrical bottle-brush polyelectrolytes with tetravalent porphyrins. It was shown that the addition of porphyrins to oppositely charged brush molecules induces a hierarchical formation of stable nanoscale brush-porphyrin networks. The networks can be disconnected by addition of salt and single porphyrin-decoratedrncylindrical brush polymers are obtained. These two new morphologies, brush-porphyrin networks and porphyrin-decorated brush polymers, may have potential as functional materials with interesting mechanical and optical properties.

Polyelectrolyte multilayers (PEM) built by layer-by-layer technique have been extensively studied over the last years, resulting in a wide variety of current and potential applications. This technique can be used to construct thin films with different functionalities, or to functionalize surfaces with substantial different properties of those of the underlying substrates. The multilayering process is achieved by the alternate adsorption of oppositely charged polyelectrolytes. In this work we get advantage of the protein resistant property of the Poly (l-lysine)-graft-(polyethyleneglycol) to create protein patterns. Proteins can be immobilized on a surface by unspecific physical adsorption, covalent binding or through specific interactions. The first protein used in this work was laccase, a copper-containing redox enzyme that catalyse the oxidation of a broad range of polyphenols and aromatic substrates, coupled to the reduction of O2 to H2O without need of cofactors. Applications of laccases have been reported in food, pulp, paper, and textile industry, and also in biosensor development. Some uses require the immobilization of the enzyme on solid supports by adsorption, covalent attachment, entrapment, etc, on several substrates. Especially for biosensor development, highly active, stable and reproducible immobilization of laccase is required.

The aim of this work is to investigate the various parameters that could control the encapsulation of lipophilic drugs and investigate the influence of the physical properties of poorly water-soluble drugs on bilayer loading. Initial work investigated on the solubilisation of ibuprofen, a model insoluble drug. Drug loading was assessed using HPLC and UV spectrophotometric analysis. Preliminary studies focused on the influence of bilayer composition on drug loading to obtain an optimum cholesterol concentration. This was followed up by studies investigating the effect of longer alkyl chain lipids, unsaturated alkyl chain lipids and charged lipids. The studies also focused on the effects of pH of the hydration medium and addition of the single chain surfactant a-tocopherol. The work was followed up by investigation of a range of insoluble drugs including flurbiprofen, indomethacin, sulindac, mefenamic acid, lignocaine and progesterone to investigate the influence of drugs properties and functional group on liposomal loading. The results show that no defined trend could be obtained linking the drug loading to the different drug properties including molecular weight, log P and other drug specific characteristics. However, the presence of the oppositely charged lipids improved the encapsulation of all the drugs investigated with a similar effect obtained with the substitution of the longer chain lipids. The addition of the single chain surfactant a-tocopherol resulted in enhancement of drug loading and possibly is governed by the log P of the drug candidate. Environmental scanning-electron microscopy (ESEM) was used to dynamically follow the changes in liposome morphology in real time during dehydration thereby providing a alternative assay of liposome formulation and stability. The ESEM analysis clearly demonstrated ibuprofen incorporation enhanced the stability of PC:Chol liposomes.

Background: It is well known that the Amazon region presents a huge biodiversity; therefore, countless natural resources are being employed in the production of phytocosmetics and phytomedicines. Objective: The purpose of this work was to obtain emulsions produced with Buriti oil and nonionic surfactants. Methods: Two surfactant systems were employed (Steareth-2 associated to Ceteareth-5 and to Ceteareth-20) to produce the emulsions using phase diagram method. Emulsions were obtained by echo-planar imaging method at 75 degrees C. Rheological behavior and zeta potential were evaluated, and accelerated stability tests were performed. Results: All emulsions analyzed presented pseudoplastic behavior. Zeta potential values were obtained between -14.2 and -53.3 mV. The formulations did not show changes in either physical stability, pH, or rheological behavior after accelerated stability tests. Significant differences were observed only after temperature cycling test. Conclusion: Based on these results, the emulsions obtained could be considered as promising delivery systems.

We have grown surfactant-templated silicate films at the air-water interface using n-alkyltrimethylammonium bromide and chloride in an acid synthesis with tetraethyl orthosilicate as the silicate source. The films have been grown with and without added salt (sodium chloride, sodium bromide) and with n-alkyl chain lengths from 12 to 18, the growth process being monitored by X-ray reflectometry. Glassy, hexagonal, and lamellar structures have been produced in ways that are predictable from the pure surfactant-water phase diagrams. The synthesis appears to proceed initially through an induction period characterized by the accumulation of silica-coated spherical micelles near the surface. All syntheses, except those involving C(12)TACl, show a sudden transformation of the spherical micellar phase to a hexagonal phase. This occurs when the gradually increasing ionic strength and/or changing ethanol concentration is sufficient to change the position of boundaries within the phase diagram. A possible mechanism for this to occur may be to induce a sphere to rod transition in the micellar structure. This transformation, as predicted from the surfactant-water phase diagram, can be induced by addition of salts and is slower for chloride than bromide counteranions. The hexagonal materials change in cell dimension as the chain length is changed in a way consistent with theoretical model predictions. All the materials have sufficiently flexible silica frameworks that phase interconversion is observed both from glassy to hexagonal and from hexagonal, to lamellar and vice versa in those surfactant systems where multiple phases are found to exist.

The associated production of a Higgs boson and a top-quark pair, t (t) over barH, in proton-proton collisions is addressed in this paper for a center of mass energy of 13 TeV at the LHC. Dileptonic final states of t (t) over barH events with two oppositely charged leptons and four jets from the decays t -> bW(+) -> bl(+)v(l), (t) over bar -> (b) over barW(-) -> (b) over barl(-)(v) over bar (l) and h -> b (b) over bar are used. Signal events, generated with MadGraph5_aMC@NLO, are fully reconstructed by applying a kinematic fit. New angular distributions of the decay products as well as angular asymmetries are explored in order to improve discrimination of t (t) over barH signal events over the dominant irreducible background contribution, t (t) over barb (b) over bar. Even after the full kinematic fit reconstruction of the events, the proposed angular distributions and asymmetries are still quite different in the t (t) over barH signal and the dominant background (t (t) over barb (b) over bar).

Simultaneous measurements of the tt¯, W+W−, and Z/γ∗→ττ production cross-sections using an integrated luminosity of 4.6 fb−1 of pp collisions at s√=7 TeV collected by the ATLAS detector at the LHC are presented. Events are selected with two high transverse momentum leptons consisting of an oppositely charged electron and muon pair. The three processes are separated using the distributions of the missing transverse momentum of events with zero and greater than zero jet multiplicities. Measurements of the fiducial cross-section are presented along with results that quantify for the first time the underlying correlations in the predicted and measured cross-sections due to proton parton distribution functions. These results indicate that the correlated NLO predictions for tt¯ and Z/γ∗→ττ significantly underestimate the data, while those at NNLO generally describe the data well. The full cross-sections are measured to be σ(tt¯)=181.2±2.8+9.7−9.5±3.3±3.3 pb, σ(W+W−)=53.3±2.7+7.3−8.0±1.0±0.5 pb, and σ(Z/γ∗→ττ)=1174±24+72−87±21±9 pb, where the cited uncertainties are due to statistics, systematic effects, luminosity and the LHC beam energy measurement, respectively.

Hydrophilic nanocarriers formed by electrostatic interaction of chitosan with oppositely charged macromolecules have a high potential as vectors in biomedical and pharmaceutical applications. However, comprehensive information about the fate of such nanomaterials in biological environment is lacking. We used chitosan from both animal and fungal sources to form well-characterized chitosan-pentasodium triphosphate (TPP)//alginate nanogels suitable for comparative studies. Upon exposure of human colon cancer cells (HT29 and CaCo2), breast cancer cells (MDA-MB-231 and MCF-7), glioblastoma cells (LN229), lung cancer cells (A549), and brain-derived endothelial cells (HCEC) to chitosan-(TPP)//alginate nanogels, cell type-, nanogel dosage-, and exposure time-dependent responses are observed. Comparing chitosan-TPP//alginate nanogels prepared from either animal or fungal source in terms of nanogel formation, cell uptake, reactive oxygen species production, and metabolic cell activity, no significant differences become obvious. The results identify fungal chitosan as an alternative to animal chitosan in particular if biomedical/pharmaceutical applications are intended.

We present and analyze exact solutions of the Einstein-Maxwell and Einstein-Maxwell-dilaton equations that describe static pairs of oppositely charged extremal black holes, i.e., black diholes. The holes are suspended in equilibrium in an external magnetic field, or held apart by cosmic strings. We comment as well on the relation of these solutions to brane-antibrane configurations in string and M theory.

In this work we describe the synthesis of a variety of MCM-41 type hexagonal and SBA-1 type cubic mesostructures and mesoporous silicious materials employing a novel synthesis concept based on polyacrylic acid (Pac)-C(n)TAB complexes as backbones of the developing structures. The ordered porosity of the solids was established by XRD and TEM techniques. The synthesis concept makes use of Pac-C(n)TAB nanoassemblies as a preformed scaffold, formed by the gradual increase of pH. On this starting matrix the inorganic precursor species SiO2 precipitate via hydrolysis of TEOS under the influence of increasing pH. The molecular weight (MW) of Pac, as well as the length of carbon chain in C,TAB, determine the physical and structural characteristics of the obtained materials. Longer chain surfactants (C(16)TAB) lead to the formation of hexagonal phase, while shorter chain surfactants (C(14)TAB, C(12)TAB) favor the SBA-1 phase. Lower MW of Pac (approximate to2000) leads to better-organized structures compared to higher MW ( 450,000), which leads to worm-like mesostructures. Cell parameters and pore size increase with increasing polyelectrolyte and/or surfactant chain, while at the same time SEM photography reveals that the particle size decreases. Conductivity experiments provide some insight into the proposed self-assembling pathway. (C) 2003 Elsevier Inc. All rights reserved.
