Introduction - A large number of natural and synthetic compounds having butenolides as a core unit have been described and many of them display a wide range of biological activities. Butenolides from P. malacophyllum have presented potential antifungal activities but no specific, fast, and precise method has been developed for their determination. Objective - To develop a methodology based on micellar electrokinetic chromatography to determine butenolides in Piper species. Methodology - The extracts were analysed in an uncoated fused-silica capillaries and for the micellar system 20 mmol/L SDS, 20% (v/v) acetonitrile (ACN) and 10 mmol/L STB aqueous buffer at pH 9.2 were used. The method was validated for precision, linearity, limit of detection (LOD) and limit of quantitation (LOQ) and the standard deviations were determined from the standard errors estimated by the regression line. Results - A micellar electrokinetic chromatography (MEKC) method for determination of butenolides in extracts gave full resolution for 1 and 2. The analytical curve in the range 10.0-50.0 mu g/mL (r(2) = 0.999) provided LOD and LOQ for 1 and 2 of 2.1/6.3 and 1.1/3.5 mu g/mL, respectively. The RSD for migration times were 0.12 and 1.0% for peak area ratios with 100.0 +/- 1.4% of recovery. Conclusions - A novel high-performance MEKC method developed for the analysis of butenolides 1 and 2 in leaf extracts of P. malacophyllum allowed their quantitative determined within an analysis time shorter than 5 min and the results indicated CE to be a feasible analytical technique for the quantitative determination of butenolides in Piper extracts. Geranylation of benzoic acid derivatives by enzymatic extracts from Piper crassinervium (Piperaceae)

The ability to carry out geranylations on aromatic substrates using enzymatic extracts from the leaves of Piper crassinervium (Piperaceae) was evaluated. A literature analysis pointed out its importance as a source of prenylated bioactive molecules. The screening performed on aromatic acceptors (benzoic acids, phenols and phenylpropanoids) including geranyl diphosphate as prenyl donor, showed the biotransformation of the 3,4-dihydroxybenzoic acid by the crude extract, and the p-hydroxybenzoic acid by both the microsomal fraction and the crude extract, after treating leaves with glucose. The analysis of the products allowed the identification of C- and O-geranylated derivatives, and the protease (subtilisin and pepsin) inhibition performed on the O-geranylated compounds showed weak inhibition. Electrophoretic profiles indicated the presence of bands/spots among 56-58 kDa and pI 6-7, which are compatible with prenyltransferases. These findings show that P. crassinervium could be considered as a source of extracts with geranyltransferase activity to perform biotransformations on aromatic substrates. (C) 2010 Elsevier Ltd. The CH(2)Cl(2) and MeOH extracts from leaves of Piper caldense were subjected to chromatographic separation procedures to afford the new prenylated benzoic acid, caldensinic acid (3-[(2`E,6`E,10`E)-11`-carboxy-3`,7`,15`-trimethylhexadeca-2`,6`,10`,14`-tetraenyl]-4,5-dihydroxybenzoic acid) whose structure was determined by spectral analysis, mainly NMR ((1)H, (13)C, HSQC, HMBC) and ESI-MS. The natural compound and derivatives displayed antifungal activity against the phytopathogenic fungi Cladosporium cladosporioides and C. sphaerospermum by direct bioautography. The bioactivity-guided fractionation of the crude extracts from leaves of Brazilian species Piper aduncum and Piper hostmannianum by means of bioautography using the fungi Cladosporium cladosporioides and C. sphaerospermum afforded prenylated methyl benzoate, chromenes, and dihydrobenzopyran derivatives as antifungal compounds. The isolation and structural elucidation of a new compound methyl 4-hydroxy-3-(2`-hydroperoxy-3`-methyl-3`-butenyl) benzoate were performed by application of chromatographic techniques and spectroscopic analyses. In the scope of our ongoing researchers on antioxidant compounds, twenty four extracts and fractions obtained from Piper arboreum Aublet and Piper tuberculatum Jacq. (Piperaceae) were screened for radical scavenging capacity (RSC) by using DPPH colorimetric assay. The strongest activity was found in ethyl acetate fractions from the leaves of P. arboreum IC(50) = 5.70 mu g/mL) and P. tuberculatum IC(50) = 8.40 mu g/mL). Hydromethanol fractions of the leaves of P. tuberculatum and P. arboreum showed moderate RSC, with values of IC(50) (mu g/mL) of 11.9 and 19.2, respectively. Additionally, a brief phytochemical study of the ethyl acetate fraction of P. arboreum leaves affording quercetin (1) and quercitrin (2), two flavonols with antioxidant activity previously described in the literature.

This study describes the antichagasic potential of five compounds isolated from leaves of Piper crassinervium (Piperaceae). Two prenylated benzoic acid derivatives, one prenylated hydroquinone and two flavanones, were evaluated. The in vitro trypanocidal activity was determined against epimastigote forms of Trypanosoma cruzi (Y strain), the etiologic agent of Chagas disease. The most active compound was the prenylated hydroquinone [1,4-dihydroxy-2-(3(0),7(0)-dimethyl-1(0)-oxo-2(0)-E,6(0)-octadienyl)benzene] with an IC(50) value of 6.10 g mL(-1), which was in the same order of activity if compared with the positive control benznidazole (IC(50) = 1.60 mu g mL(-1)). This is the first report of trypanocidal activity for prenylated hydroquinone and benzoic acid derivatives.

The leaves and twigs of Piper krukoffii, collected in the Carajas National Forest, north Brazil, yielded essential oils (2.0% and 0.8%), the main constituents of which were myristicin (40.3% and 26.7%), apiole (25.4% and 34.1%) and elemicin (2.8% and 3.0%). The antioxidant activities of the oils, methanol extract and its sub-fractions were evaluated. The DPPH EC(50) values varied from the ethyl acetate sub-fraction (73.4 +/- 3.7 mu g/mL) to the methanol extract (24.9 +/- 0.8 mu g/mL), and the ABTS TEAC values ranged in the same order from 265.7 to 349.2 mu Mol TE/g. These results indicated a significant antioxidant activity for the plant. The lignans (-)-kusunokin, yatein, (-)-hinokin and cubebin were identified in the methanol extract. The hydro-methanolic sub-fraction showed a high value for total phenol content (106.5 +/- 0.7 mg GAE/g), as well as (1)H NMR signals for sugar moieties. Crude extracts and sub-fractions were also able to inhibit beta-carotene bleaching, varying from 22.4 to 47.1%. The oils from the leaves and twigs showed strong larvicidal (21.4 and 3.6 mu g/mL) and fungicide (0.5 and 0.1 mu g/mL) activities.

Two new presilphiperfolane sesquiterpenes, 1 and 2, were isolated from the ethyl acetate extract of Xylaria sp., obtained from the leaves of Piper aduncum, along with two known eremophilane sesquiterpenes, phaseolinone (3) and phomenone (4). Chemical structures of 1 and 2 were established by analysis of spectroscopic data. The four compounds were tested in vitro for antifungal and cytotoxicity activities using CHO (Chinese hamster ovary). Compounds 1 and 2 did not show any antifungal and cytotoxic activity. As doenças cerebrovasculares, popularmente conhecidas como derrames, são uma das principais causas de morbidade e mortalidade entre adultos e idosos. Contudo, muitos pacientes que sofrem derrame sobrevivem e experimentam as conseqüências do insulto por muitos anos, muitas vezes a nível emocional, motor ou intelectual. Dentre estas lesões, destaca-se a isquemia cerebral. Existem modelos experimentais de isquemia cerebral in vivo e in vitro. Os modelos in vitro são realizados em culturas ou fatias de tecido cerebral submetidas à Privação de Oxigênio e Glicose (POG), que mimetizam condições traumáticas similares, mas não idênticas às produzidas in vivo. A investigação da atividade de substâncias potencialmente neuroprotetoras a partir da comparação da morte celular entre culturas ou fatias de tecido cerebral controle e tratadas é facilitada neste tipo de modelo experimental. Após a injúria, as culturas são expostas a métodos de avaliação da viabilidade ou dano celular como, por exemplo, o corante fluorescente iodeto de propídeo, que marca seletivamente células mortas ou em curso de morte, ou MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazoliumbromide) que mede a viabilidade mitocondrial possibilitando uma posterior quantificação. As plantas são uma fonte importante de produtos naturais biologicamente ativos, muitos dos quais se constituem em modelos para a síntese de um grande número de fármacos. Um exemplo é a planta kava-kava ou somente kava (Piper Methysticum) a qual chamou a atenção dos pesquisadores devido à sua utilização nas ilhas do Pacífico sul. Foi demonstrada a possibilidade da kava possuir uma variedade de atividades farmacológicas importantes, entre elas a atividade de neuroproteção Esse trabalho tem como objetivo avaliar a atividade neuroprotetora do extrato de kava-kava (Piper methysticum) em modelos in vitro de morte neuronal em fatias de hipocampo de ratos, investigar o envolvimento da proteína de choque térmico HSP27 (Heat Shock Protein) no processo de morte e neuroproteção induzida pela kava, bem como investigar o efeito da lesão induzida por POG sobre o imunoconteúdo da proteína Oxido Nítrico Sintase induzível (iNOS). Os resultados dos experimentos por nós realizados nas culturas organotípicas submetidas à POG por 40 minutos e tratadas com extrato de kava (7µg/ml) demonstraram uma significativa redução, na ordem de 58% na intensidade da morte neuronal na região CA1 do hipocampo em resposta à injúria. Em fatias hipocampais submetidas à POG por 60 minutos a adição do extrato de kava 7µg/ml aumentou em 15% a viabilidade celular, confirmando a atividade neuroprotetora sugerida para esta planta. As culturas expostas à POG e tratadas com kava apresentaram um aumento significativo no imunoconteúdo da proteína HSP27, esse aumento é acompanhado de um aumento da fosforilação, uma vez que a percentagem de proteína fosforilada se mantém igual. As fatias lesionadas tratadas com kava apresentaram uma diminuição significativa no imunoconteúdo da iNOS na ordem de 35 % em relação as fatias lesionadas tratadas com DMSO (Dimetilsulfoxido). Estes dados podem sugerir que um dos mecanismos da neuroproteção observada para o extrato de kava, possa envolver essas proteínas.

A kava kava (Piper methysticum Forst), pertencente à família Piperaceae, é utilizada para diminuir a ansiedade e medo e tratar distúrbios comportamentais. É um fitoterápico utilizado em vários países, entretanto pouco se sabe, sobre seus efeitos no desenvolvimento embrionário. O presente trabalho avaliou o possível efeito teratogênico da formulação fitoterápica contendo Piper methysticum Forst durante o período de organogênese em ratas Wistar. As ratas foram tratadas com 0mg.kg –1 (controle), 5mg.kg –1 ; 35mg.kg –1 e 50mg.kg –1 da preparação fitoterápica (kava kava®), por via oral, do 6° ao 15° dia de prenhez. Os resultados revelaram ausência de toxicidade sistêmica e reprodutiva nas variáveis avaliadas, fundamentados pela ausência de alterações no desenvolvimento ponderal, consumos de ração e água, na massa relativa dos órgãos, nas reabsorções embrionárias, na massa corporal, na vitalidade, no número de fetos por progenitora e nas alterações macroscópicas externas e esqueléticas dos fetos. Adicionalmente as enzimas alanina aminotrasferase (ALT) e fosfatase alcalina (FA) foram determinadas no soro das ratas tratadas, para avaliar o possível efeito hepatotóxico da preparação fitoterápica. Não houve alteração na atividade das enzimas ALT e FA, bem como alterações histopatológicas do fígado das ratas, não confirmando a hepatotoxicidade. Conclui-se que o fitoterápico kava kava, até 35mg.kg –1, não determina o aparecimento de teratogenicidade.

Pearl glands are scattered throughout the lamina of developing leaves and rarely found on adult leaves of Piper regnellii (Piperaceae). The pearl gland is a bicellular secretory trichome composed of a short broad basal cell and a spatula-like, semiglobular apical cell. Four different stages of the pearl grand were determined during its ontogenesis: origin, pre-secretory, secretory and post-secretory. During the pre-secretory stage, mitochondria, ribosomes, dictyosomes, rough endoplasmic reticulum, and plastids with electron dense inclusions were present in the cytoplasm of the apical cell. During the secretory stage, the most remarkable characteristics of the apical cell are the proliferation of dictyosomes and their vesicles, rough endoplasmic reticulum, and modified plastids. At this stage, electron-dense oil drops occur in the plastids as well as scattered within the cytoplasm, proteins and polysaccharides are seen in the plastids, vesicles, and vacuoles. Only polysaccharides are present in the periplasmic space, wall cavities, and on the surface of the apical cell. The polysaccharides are one of the main components of the mucilagenous exudate that covers the developing leaf structures. The apical cell of the senescing trichomes undergoes a progressive degeneration of its cellular components, the plastids being the first organelles to undergo lysis.
