Desde o primeiro momento, este projeto procurou libertar-se dos cenários tradicionais em que a aprendizagem era encarada como uma atividade solitária e individual; ao longo do processo, cada aluno encontrava-se sozinho perante a tarefa, sob olhar atento do professor (tais posturas eram próprias de uma cultura autoritária e pouco solidária na apropriação do saber). Decorrente dessa vontade de alterar formas de atuação, este projeto assentou os seus pressupostos teóricos na teoria em espiral de Bruner e na teoria das inteligências múltiplas de Gardner. Assim, o aluno é sempre encarado como um ser em desenvolvimento, razão pela qual urge ter uma visão holística e integrada desse desenvolvimento. Como tal, associada a estas teorias aparece a aprendizagem cooperativa e, consequentemente, o conceito de Zona de Desenvolvimento Proximal de Vygotsky e a integração curricular diretamente ligada à metodologia de projeto defendida por autores como Dewey e Kilpatrik, pontos basilares de todo este estudo. Tal como o próprio nome do projeto indica Possibilidades de desenvolvimento integrado pela Expressão Musical: Uma experiência no 1.º CEB, o objetivo principal é investigar as possibilidades de desenvolvimento global do aluno, com a interligação dos vários saberes que lhe permitam construir o seu conhecimento de uma forma integrada, tendo como ponto de partida a expressão musical. Desde a elaboração das planificações, passando pelas atividades desenvolvidas ao longo das aulas, houve sempre a preocupação em promover a participação ativa dos alunos e, como é evidente pelos mapas de conceitos, privilegiar a integração curricular bem como o desenvolvimento global dos alunos. Os dados obtidos através da entrevista efetuada aos alunos e do inquérito final demonstram o reconhecimento, por parte dos alunos, da expressão musical como uma mais3 valia para a sua aprendizagem nas diversas disciplinas, bem como a sua preferência pelo trabalho cooperativo.

Leishmania (Sauroleishmania) tarentolae has biotechnological potential for use as live vaccine against visceral leishmaniasis and as a system for the over expression of eukaryotic proteins that possess accurate post-translational modifications. For both purposes, new systems for protein expression in this non-pathogenic protozoan are necessary. The ribosomal RNA promoter proved to be a stronger transcription driver since its use yielded increased levels of recombinant protein in organisms of both genera Trypanosoma or Leishmania. We have evaluated heterologous expression systems using vectors with two different polypyrimidine tracts in the splice acceptor site by measuring a reporter gene transcribed from L. tarentolae RNA polymerase I promoter. Our data indicate that the efficiency of chloramphenicol acetyl transferase expression changed drastically with homologous or heterologous sequences, depending on the polypyrimidine tract used in the construct and differences in size and/or distance from the AG dinucleotide. In relation to the promoter sequence the reporter expression was higher in heterologous lizard-infecting species than in the homologous L. tarentolae or in the mammalian-infecting L. (Leishmania) amazonensis.

A musically stimulating nvironment is of great significance when considering the opportunities for development and learning that are offered to children in day nurseries. Moreover, the quality of musical activities also has great significance for the musical development of growing children. Young children are always striving to create meaning and understanding; they influence and are influenced by the context in which they are situated. The purpose of this study is to investigate the planned music activities in the musical learning environment of seven toddler’s groups in day nurseries, based on four different aspects: the texts of children’s songs, the melodies of children’s songs, the basic elements of music and the use of instrumentation. The data has been gathered by video observation of the planned music activities. The age of the children studied ranged from 11 months to 3 years and 11 months, and on average there were 13 children in each group. The study involved a total of just over 90 children and nine educators. The methodological approaches are hermeneutic as well as videographic, and they are applied to the analysis as required. The approaches are well suited to an understanding of the planned music activities where the individual’s communications and actions are studied. The results of the study show that educators impart a cultural heritage to the children in the form of old traditional songs, but they also act as intermediaries for newer children's songs. They focus on actively on the text content of the children’s songs in their interaction with the children, but do not address the meaning of the text. Furthermore, while the educators use music sessions for the children to develop, among other things, an understanding of language and social skills, they do not use the sessions to develop the children’s musical skills. The educators could, to a much greater extent than they now do, direct the children's attention to the basic elements of music, and conduct meta-cognitive dialogues with the children. They use a large repertoire of children's songs, which are, however, often too demanding for the children’s voices to cope with. In addition, the educators do not plan, in any significant way, activities for the toddlers in order to help them develop the accuracy of their vocal tones. With regard to the use of instruments, the educators focus, in their interaction with the children, is placed on both the usage of the instruments and knowledge about the instruments. Regarding the use of instruments the study shows that the children’s musical expression takes place in an environment that includes both melody and rhythm instruments. It is clear that children are actively interested in the constructed instrument environment, because for long periods of time they independently play and experiment with instruments.

Ce mémoire traite d'abord du problème de la modélisation de l'interprétation des pianistes à l'aide de l'apprentissage machine. Il s'occupe ensuite de présenter de nouveaux modèles temporels qui utilisent des auto-encodeurs pour améliorer l'apprentissage de séquences. Dans un premier temps, nous présentons le travail préalablement fait dans le domaine de la modélisation de l'expressivité musicale, notamment les modèles statistiques du professeur Widmer. Nous parlons ensuite de notre ensemble de données, unique au monde, qu'il a été nécessaire de créer pour accomplir notre tâche. Cet ensemble est composé de 13 pianistes différents enregistrés sur le fameux piano Bösendorfer 290SE. Enfin, nous expliquons en détail les résultats de l'apprentissage de réseaux de neurones et de réseaux de neurones récurrents. Ceux-ci sont appliqués sur les données mentionnées pour apprendre les variations expressives propres à un style de musique. Dans un deuxième temps, ce mémoire aborde la découverte de modèles statistiques expérimentaux qui impliquent l'utilisation d'auto-encodeurs sur des réseaux de neurones récurrents. Pour pouvoir tester la limite de leur capacité d'apprentissage, nous utilisons deux ensembles de données artificielles développées à l'Université de Toronto.

Cette recherche porte un regard critique sur les interfaces de spatialisation sonore et positionne la composition de musique spatiale, un champ d’étude en musique, à l’avant plan d’une recherche en design. Il détaille l’approche de recherche qui est centrée sur le processus de composition de musique spatiale et les modèles mentaux de compositeurs électroacoustiques afin de livrer des recommandations de design pour le développement d’une interface de spatialisation musicale nommée Centor. Cette recherche montre qu’un processus de design mené à l’intersection du design d’interface, du design d’interaction et de la théorie musicale peut mener à une proposition pertinente et innovatrice pour chacun des domaines d’étude. Nous présentons la recherche et le développement du concept de spatialisation additive, une méthode de spatialisation sonore par patrons qui applique le vocabulaire spectromorphologique de Denis Smalley. C’est un concept d’outil de spatialisation pour le studio qui complémente les interfaces de composition actuelles et ouvre un nouveau champ de possibilités pour l’exploration spatiale en musique électroacoustique. La démarche de recherche présentée ici se veut une contribution au domaine du design d’interfaces musicales, spécifiquement les interfaces de spatialisation, mais propose aussi un processus de design pour la création d’interfaces numériques d’expression artistique.

Ce mémoire de maîtrise porte principalement sur la question de la réappropriation historique et musicale des Tziganes dans le docu-fiction Latcho Drom (1993) de Tony Gatlif. Dans un premier chapitre, il s’agit de comparer l’histoire écrite sur les Tziganes avec leur mise en image afin de déterminer comment le cinéaste apporte dans le langage audiovisuel de Latcho Drom un total renouveau dans le discours dominant. Dans cette perspective, l’appareil cinématographique se révèle être un médium de revendication et de réappropriation de l’être tzigane et de son histoire. Dans un deuxième chapitre, il est question de démontrer avec des études basées sur l’ethnomusicologie comment les musiques tziganes, sont rapidement assimilées au patrimoine culturel des sociétés européennes. Latcho Drom qui traduit avec justesse des expressions musicales très encrées de la vie de ces communautés, s’inscrit en contradiction avec la conception territorialiste de musicologues et ethnomusicologues qui refusent d’accorder à la musique tzigane légitimité et autonomie. Dans un troisième chapitre, il s’agit de déterminer comment le cinéaste cherche à faire entrer son spectateur dans un rapport de proximité avec les communautés de Latcho Drom afin de susciter en lui reconnaissance et empathie.

Sabem que l’educació no es circumscriu només en l’àmbit acadèmic ni dins el marc físic d’un centre docent, sinó que la societat i els productes que genera són elements importants a tenir en compte. En base a aquests supòsits, els nous corrents pedagògics de l’Escola Nova sorgits a començaments del segle XX ja recomanaven obrir les escoles a l’entorn immediat i aprofitar les seves possibilitats tant com a recurs educatiu com a contingut curricular en sí mateix. Aquesta premissa –plenament vigent a l’actualitat– es fa palesa especialment a l’àrea de música, no només per la gran quantitat d’informació –immediata i diversificada- i per les possibilitats didàctiques i educatives de caràcter multimèdia i interactiu de les Noves Tecnologies (NNTT), les Tecnologies de la Informació i la Comunicació (TIC) i les Tecnologies de l’Aprenentatge i la Comunicació (TAC) que configuren l’actual realitat virtual a la que ens veiem abocats, sinó per l’ampli ventall d’estils i gèneres que inclouen les programacions d’espectacles musicals al nostre abast. De la mateixa manera que l’audició de música en directe és del tot imprescindible en aquesta àrea artística i humanística per tal de desenvolupar les destreses que li són pròpies, també ho és la participació activa. En aquest sentit, constatem que la realització d’activitats educatives a nivell col·lectiu permet la interacció entre iguals i la descoberta, desenvolupament o aprofundiment d’actituds com la cooperació, la tolerància, el respecte mutu, la cohesió, etc., valors imprescindibles en una societat democràtica i plural com la nostra. I encara més, que l’activitat col·lectiva realitzada amb l’objectiu de produir un producte propi, afavoreix la satisfacció personal final que conduirà a la veritable integració dels continguts a assimilar. Per tant, la pràctica aplicada d’aquestes actituds i destreses socials es fa imprescindible en els estudis dels futurs docents. L'Espai Europeu de Educació Superior (EEES) ens aporta la incorporació d'un concepte provinent del món professional, les competències, que són un nou referent de rang superior que orienta globalment els estudis universitaris. Els objectius i continguts d'aquests tindran sentit en la mesura que contribueixin a desenvolupar algunes d'aquestes competències, ja que seran l'objecte principal d'avaluació de les titulacions. Pensem que a través d’activitats que es poden fer fora de l'aula com la que presentem en aquesta comunicació, s'ajuda a mobilitzar algunes d'aquestes competències que permetran configurar el perfil de graduat que tots desitgem

The euro crisis has forced member states and the EU institutions to create a series of new instruments to safeguard macro-financial stability of the Union. This study describes the status of existing instruments, the role of the European Parliament and how the use of the instruments impinges on the EU budget also through their effects on national budgets. In addition, it presents a survey of other possible instruments that have been proposed in recent years (e.g. E-bonds and eurobonds), in order to provide an assessment of how EU macro-financial stability assistance could evolve in the future and what could be its impact on EU public finances.

Model of study: Experimental study. Introduction: Recently, stem cell research has generated great interest due to its applicability in regenerative medicine. Bone marrow is considered the most important source of adult stem cells and the establishment of new methods towards gene expression analysis regarding stem cells has become necessary. Thus Differential Display Reverse Transcription Polymerase Chain Reaction (DDRT-PCR) may be an accessible tool to investigate small differences in the gene expression of different stem cells in distinct situations. Aim: In the present study, we investigated the exequibility of DDRT-PCR to identify differences in global gene expression of mice bone marrow cells under two conditions. Methods: First, bone marrow cells were isolated fresh and a part was cultivated during one week without medium replacement. Afterwards, both bone marrow cells (fresh and cultivated) were submitted to gene expression analyses by DDRT-PCR. Results: Initially, it was possible to observe in one week-cultured bone marrow cells, changes in morphology (oval cells to fibroblastic-like cells) and protein profile, which was seen through differences in band distribution in SDS-Page gels. Finally through gene expression analysis, we detected three bands (1300, 1000 and 225 bp) exclusively expressed in the fresh bone marrow group and two bands (400 and 300 bp) expressed specifically in the cultivated bone marrow cell group. Conclusions: In summary, the DDRT-PCR method was proved efficient towards the identification of small differences in gene expression of bone marrow cells in two defined conditions. Thus, we expect that DDRT-PCR can be fast and efficiently designed to analyze differential gene expression in several stem cell types under distinct conditions.

Nowadays communication is switching from a centralized scenario, where communication media like newspapers, radio, TV programs produce information and people are just consumers, to a completely different decentralized scenario, where everyone is potentially an information producer through the use of social networks, blogs, forums that allow a real-time worldwide information exchange. These new instruments, as a result of their widespread diffusion, have started playing an important socio-economic role. They are the most used communication media and, as a consequence, they constitute the main source of information enterprises, political parties and other organizations can rely on. Analyzing data stored in servers all over the world is feasible by means of Text Mining techniques like Sentiment Analysis, which aims to extract opinions from huge amount of unstructured texts. This could lead to determine, for instance, the user satisfaction degree about products, services, politicians and so on. In this context, this dissertation presents new Document Sentiment Classification methods based on the mathematical theory of Markov Chains. All these approaches bank on a Markov Chain based model, which is language independent and whose killing features are simplicity and generality, which make it interesting with respect to previous sophisticated techniques. Every discussed technique has been tested in both Single-Domain and Cross-Domain Sentiment Classification areas, comparing performance with those of other two previous works. The performed analysis shows that some of the examined algorithms produce results comparable with the best methods in literature, with reference to both single-domain and cross-domain tasks, in $2$-classes (i.e. positive and negative) Document Sentiment Classification. However, there is still room for improvement, because this work also shows the way to walk in order to enhance performance, that is, a good novel feature selection process would be enough to outperform the state of the art. Furthermore, since some of the proposed approaches show promising results in $2$-classes Single-Domain Sentiment Classification, another future work will regard validating these results also in tasks with more than $2$ classes.

This chapter maps out central globalizing perspectives that have influenced discourses on revival and related transformational processes of traditional forms of musical expression. Focusing on the Latvian groups Skandinieki and Iļģi and on electric folk in England, the chapter analyzes the extent to which meta-perspectives can help elucidate the impact of global flows on music revivals. As Held et al.’s 2003 globalization study indicates, revival situations can be interpreted from different meta-perspectives. Equating globalization with homogenization, a sceptic perspective perceives revival as an important means of preservation. A hyperglobal approach regards globalization not as a threat, but as a possible access to Western market structures. A transformationalist perspective perceives increasingly dense global networks as the basis for developing new structures, approached from an open perspective. Although supporting a transformationalist perspective, this chapter argues that an adequate modern approach toward revival requires the combination of all three perspectives.

O culto presbiteriano brasileiro se cristalizou de forma peculiar em relação ao seu estilo litúrgico. A exclusão de elementos mais ritualísticos somados ás condições de inserção deram um contorno bem específico a esse culto no Brasil. A sua forma pode ser reduzida a dois pilares de sustentação: a prédica e a música. A música também se desenvolveu de um modo peculiar dentro da denominação. Porém, desde início dos anos 90 essa musica tradicional tem sido abalada com a consolidação do mercado de música gospel. Surgiu então no Brasil um novo tipo de produção musical, relacionada com o louvor congregacional, que pelo aspecto emocional e performático, contraria a produção tradicional. Esse novo modelo de louvor, fomentado pela mídia especializada, pelo mercado gospel e em acordo com as tendências culturais atuais, tem plena aceitação do público jovem e dos novos conversos, dificultando a manutenção da tradição. Muitos são os motivos para tal fato, entre eles a atração que o mercado exerce sobre o jovem presbiteriano e o papel litúrgico da música nesse culto, que sempre gerou um grande clima de insatisfação religiosa de alguns sub grupos do laicato. Assim, a produção e reprodução musical do culto presbiteriano têm sofrido modificações que podem contribuir diretamente para uma das maiores mutações cúlticas que o presbiterianismo já sofreu desde a sua inserção entre nós. Esse novo modelo de louvor propiciou uma ruptura com a hinódia tradicional, que há muito já estava se enfraquecendo. Esta tese discute as mudanças ocorridas na produção e reprodução da música litúrgica do presbiterianismo e a sua conseqüência direta no perfil desse culto, bem como os motivos que sustentam tais mudanças, tanto em uma perspectiva micro como macro social.(AU)
