The crystal structure of an enzyme–substrate complex with histidyl-tRNA synthetase from Escherichia coli, ATP, and the amino acid analog histidinol is described and compared with the previously obtained enzyme–product complex with histidyl-adenylate. An active site arginine, Arg-259, unique to all histidyl-tRNA synthetases, plays the role of the catalytic magnesium ion seen in seryl-tRNA synthetase. When Arg-259 is substituted with histidine, the apparent second order rate constant (kcat/Km) for the pyrophosphate exchange reaction and the aminoacylation reaction decreases 1,000-fold and 500-fold, respectively. Crystals soaked with MnCl2 reveal the existence of two metal binding sites between β- and γ-phosphates; these sites appear to stabilize the conformation of the pyrophosphate. The use of both conserved metal ions and arginine in phosphoryl transfer provides evidence of significant early functional divergence of class II aminoacyl-tRNA synthetases.

The crystal structures of the ligand-binding domain (LBD) of the vitamin D receptor complexed to 1α,25(OH)2D3 and the 20-epi analogs, MC1288 and KH1060, show that the protein conformation is identical, conferring a general character to the observation first made for retinoic acid receptor (RAR) that, for a given LBD, the agonist conformation is unique, the ligands adapting to the binding pocket. In all complexes, the A- to D-ring moieties of the ligands adopt the same conformation and form identical contacts with the protein. Differences are observed only for the 17β-aliphatic chains that adapt their conformation to anchor the 25-hydroxyl group to His-305 and His-397. The inverted geometry of the C20 methyl group induces different paths of the aliphatic chains. The ligands exhibit a low-energy conformation for MC1288 and a more strained conformation for the two others. KH1060 compensates this energy cost by additional contacts. Based on the present data, the explanation of the superagonist effect is to be found in higher stability and longer half-life of the active complex, thereby excluding different conformations of the ligand binding domain.

Com esta investigação pretende-se clarificar em que medida os recursos educativos das escolas contribuem para o sucesso escolar dos alunos do 1º Ciclo do Ensino Básico, no agrupamento onde foi realizada. O presente estudo tem como finalidade conhecer os recursos das escolas, assim como a gestão que os docentes fazem dos mesmos, relacionando-os com os resultados escolares dos alunos do 4.º ano de escolaridade, nas áreas de Português e Matemática. Foi efetuado um estudo de natureza mista (quantitativa e qualitativa), recorrendo a uma metodologia de estudo de caso. Foi aplicado inquérito por questionário a todos os docentes do 1º CEB, inquérito por entrevista a duas docentes das escolas em estudo e analisaram-se documentos estruturantes do agrupamento. Após a análise dos resultados, procedeu-se à sustentação teórica dos mesmos, elaboraram-se as considerações finais e sugeriram- -se futuras investigações.

The occurrence of hand grindstones at the Cogotas I archaeological sites is considered to be a common feature. Given that a distant-provenance raw material is frequently involved, determination of its source is a basic factor in the search for a better understanding of resource management and for any Political Economy approach. To progress in these directions an overall study should be planned, using selected grindstones with a view to covering diverse sub-zones of the Cogotas I dispersal area, especially because of its considerable distance from the granite basement source. Such a study may today includes diverse analytical procedures combining successive geographic, petrographic, mineralogical and geochemical criteria. To check the plausibility of the proposed methodology, a preliminary test has been carried out on two granite grindstones, obtained at the archaeological excavation at the Castronuño (Valladolid) Cogotian site, which is fifty km away from an inferred source area that was presumably located at Peñausende (Zamora). The result obtained validates the proposed operational process, yielding a generalizable knowledge to other similar situations.

Research surrounding the transition from II to I millennium cal BC in Eastern Iberian Peninsula has a large and extensive tradition of investigation. However, the chances to do research on this historical process have been limited by the lack of a well contextualized and dated stratigraphic sequence. For this reason, recent studies in this topic have followed the periodic proposals which were developed in closer regions and areas, especially in the South East and North East of the Peninsula. The investigation perspective about the Late Bronze in Eastern Iberian has however now improved, with the development of several archaeological investigations, the increase in the number of sites being dated and more recent studies into the region helping to bring about this change. As such, it is now in the correct state to be able to propose a new periodization and delve into the changes which occurred in the transition between 1500 and 725 cal BC.

Miguel de Unamuno fue un constante lector de poesía épica, en la que Homero siempre tuvo un lugar preferente, y no solo por motivos profesionales. A lo largo de su obra, tanto en prosa como en verso, pueden rastrearse distintas y a veces contradictorias referencias a la Ilíada y la Odisea sobre las que hizo profundas reflexiones, intentando siempre ajustarlas a su propia visión de la vida y de la muerte. Desde esta perspectiva se acerca a escenarios como el ‘prado de asfódelos’, o, a partir del símil homérico, al ‘sucederse de las generaciones de las hojas y de los hombres’, o a figuras míticas como las Sirenas, Helena, Tántalo, Sísifo y Ulises.
