Acrylamide and pyrazine formation, as influenced by the incorporation of different amino acids, was investigated in sealed low-moisture asparagine-glucose model systems. Added amino acids, with the exception of glycine and cysteine and at an equimolar concentration to asparagine, increased the rate of acrylamide formation. The strong correlation between the unsubstituted pyrazine and acrylamide suggests the promotion of the formation of Maillard reaction intermediates, and in particular glyoxal, as the determining mode of-action. At increased amino acid concentrations, diverse effects were observed. The initial rates of acrylamide formation remained high for valine, alanine, phenylalanine, tryptophan, glutamine, and Ieucine, while a significant mitigating effect, as evident from the acrylamide yields after 60 min of heating at 160 degrees C, was observed for proline, tryptophan, glycine, and cysteine. The secondary amine containing amino acids, proline and tryptophan, had the most profound mitigating effect on acrylamide after 60 min of heating. The relative importance of the competing effect of added amino acids for alpha-dicarbonyls and acrylamide-amino, acid alkylation reactions is discussed and accompanied by data on the relative formation rates of selected amino acid-AA adducts.

An efficient aza-Michael addition of amines to a series of ,-unsaturated ketones, carboxylic esters, nitriles and chalcones has been carried out using perchloric acid supported over silica gel (HClO4-SiO2) at room temperature in high yields under solvent-free reaction conditions.

Nas últimas décadas a quiralidade tornou-se essencial na conceção, descoberta, desenvolvimento e comercialização de novos medicamentos. A importância da quiralidade na eficácia e segurança dos fármacos tem sido globalmente reconhecida tanto pelas indústrias farmacêuticas como pelas agências reguladoras de todo o mundo. De forma a produzir eficazmente medicamentos seguros e dar resposta à demanda da indústria de compostos enantiomericamente puros, a pesquisa de novos métodos de síntese assimétrica, assim como o desenvolvimento estratégico dos métodos já disponíveis tem sido um dos principais objetos de estudo de diversos grupos de investigação tanto na academia como na indústria farmacêutica No primeiro capítulo desta dissertação são introduzidos alguns dos conceitos fundamentais associados à síntese de moléculas quirais e descritas algumas das estratégias que podem ser utilizadas na sua síntese. Apresenta-se ainda uma breve revisão bibliográfica acerca dos antecedentes do grupo de investigação e sobre a ocorrência natural, atividade biológica e métodos de síntese e transformações de compostos do tipo (E,E)-cinamilidenoacetofenona. O segundo capítulo centra-se na adição de Michael enantiosseletiva de diversos nucleófilos a derivados de (E,E)-cinamilidenoacetofenona. Inicialmente descreve-se a síntese de derivados de (E,E)-cinamilidenoacetofenona através de uma condensação aldólica de acetofenonas e cinamaldeídos apropriadamente substituídos. Estes derivados são posteriormente utilizados como substratos na adição de Michael enantiosseletiva de três diferentes nucleófilos: nitrometano, malononitrilo e 2-[(difenilmetileno)amino]acetato de metilo. Nestas reações são utilizados diferentes organocatalisadores de forma a induzir enantiosseletividade nos aductos de Michael para serem utilizados na síntese de compostos com potencial interesse terapêutico. É descrita ainda uma nova metodologia de síntese de Δ1-pirrolinas através de um procedimento one-pot de redução/ciclização/desidratação mediada por ferro na presença de ácido acético de (R,E)-1,5-diaril-3-(nitrometil)pent-4-en-1-onas com bons rendimentos e excelentes excessos enantioméricos. O terceiro capítulo centra-se no estabelecimento de novas rotas de síntese e transformação de derivados do ciclo-hexano. Após uma breve revisão bibliográfica, são descritas três metodologias enantiosseletivas distintas, sendo que a primeira envolve a utilização de organocatalisadores e catalisadores de transferência de fase derivados de alcaloides cinchona. Os derivados do ciclo-hexano foram obtidos a partir da reação entre as (E,E)-cinamilidenoacetofenonas e o malononitrilo com bons rendimentos, mas baixas enantiosseletividades independentemente do catalisador utilizado. De forma a contornar este problema e uma vez que a formação do derivado do ciclo-hexano envolve inicialmente a formação in-situ do aducto de Michael, a segunda e terceira metodologias de síntese envolvem a utilização dos aductos de Michael enantiomericamente puros preparados no segundo capítulo. Assim, a reação do (S,E)-2-(1,5-diaril-1-oxopent-4-en-3-il)malononitrilo com os derivados de (E,E)-cinamilidenoacetofenona organocatalisada pela hidroquinina permitiu obter os compostos pretendidos com excelentes excessos enantioméricos. A utilização de um catalisador de transferência de fase não foi tão eficiente em termos de enantiosseletividades obtidas na reação entre as (R,E)-1,5-diaril-3-(nitrometil)pent-4-en-1-onas e os derivados de (E,E)-cinamilidenoacetofenona, apesar de estes terem sido obtidos em bons rendimentos. A preparação destes derivados levou ainda à idealização de uma nova metodologia de síntese de análogos do ácido γ-aminobutírico (GABA) devido à presença de um grupo nitro em posição gama relativamente a um grupo carboxílico. No entanto, apesar de terem sido testadas várias metodologias, não foi possível obter os compostos pretendidos. No quarto capítulo apresenta-se uma breve revisão bibliográfica acerca da ocorrência natural, atividade biológica e métodos de síntese de derivados de di-hidro- e tetra-hidropiridinas, assim como um enquadramento teórico acerca das reações pericíclicas utilizadas na síntese dos compostos pretendidos. Inicialmente é descrita a preparação de N-sulfonilazatrienos substituídos através da condensação direta de derivados de (E,E)-cinamilidenoacetofenona e sulfonamidas. Estes compostos são posteriormente utilizados na síntese de derivados de 1,2-di-hidropiridinas através de uma aza-eletrociclização-6π por duas metodologias distintas: utilização de organocatalisadores quirais e utilização de complexos metálicos de bisoxazolinas. Na síntese das tetra-hidropiridinas os N-sulfonilazatrienos são utilizados como dienos e o étoxi-eteno como dienófilo numa reação hetero-Diels-Alder inversa utilizando também os complexos metálicos de bisoxazolinas como catalisadores. Todos os novos compostos sintetizados foram caracterizados estruturalmente recorrendo a estudos de espetroscopia de ressonância magnética nuclear (RMN), incluindo espetros de 1H e 13C e estudos bidimensionais de correlação espetroscópica homonuclear e heteronuclear e de efeito nuclear de Overhauser (NOESY). Foram também efetuados, sempre que possível, espetros de massa (EM) e análises elementares ou espetros de massa de alta resolução (EMAR) para todos os novos compostos sintetizados.

The first example of an intramolecular enantioselective Michael addition of nitronates onto conjugated systems utilizing a chiral phase-transfer catalyst is described. A range of five-membered gamma-nitro esters with up to three stereocentres have been prepared and the relative and absolute configurations proven by chemical and crystallographic methods. The products are rapidly obtained and are precursors to five-membered cyclic gamma-amino acids.

Two efficient, regio- and stereo controlled synthetic approaches to the synthesis of racemic analogs of pancratistatin have been accomplished and they serve as the model systems for the total synthesis of optically active 7-deoxy-pancratistatin. In the Diels-Alder approach, an efficient [4+2] cycloaddition of 3,4-methylenedioxyco- nitrostyrene with Danishefsky's diene to selectively form an exo-nitro adduct has been developed as the key step in the construction of the C-ring of the target molecule. In the Michael addition approach, the key step was a conjugate addition of an organic zinc-cuprate to the 3,4-methylenedioxy-(B-nitrostyrene, followed by a diastereocontroUed closure to form the cyclohexane C-ring of the target molecule via an intramolecular nitro-aldol cyclization on a neutral alumina surface. A chair-like transition state for such a cyclization has been established and such a chelation controlled transition state can be useful in the prediction of diastereoselectivity in other related 6-exo-trig nitroaldol reactions. Cyclization of the above products fi^om both approaches by using a Bischler-Napieralski type reaction afforded two lycoricidine derivatives 38 and 50 in good yields. The initial results from the above modeling studies as well as the analysis of the synthetic strategy were directed to a chiral pool approach to the total synthesis of optically active 7-deoxy-pancratistatin. Selective monsilylation and iodination of Ltartaric acid provided a chiral precursor for the proposed key Michael transformation. The outlook for the total synthesis of 7-deoxy-pancratistatin by this approach is very promising.A concise synthesis of novel designed, optically pure, Cz-symmetrical disulfonylamide chiral ligands starting from L-tartaric acid has also been achieved. This sequence employs the metallation of indole followed by Sfj2 replacement of a dimesylate as the key step. The activity for this Cz-symmetric chiral disulfonamide ligand in the catalytic enantioselective reaction has been confirmed by nucleophilic addition to benzaldehyde in the disulfonamide-Ti (0-i-Pr)4-diethylzinc system with a 48% yield and a 33% e.e. value. Such a ligand tethered with a suitable metal complex should be also applicable towards the total synthesis of 7-deoxy-pancratistatin.

The present studies describe our recent progress in target oriented synthesis of complex organic molecules from aromatic precursors. The latest synthetic approaches toward vinca alkaloids are described and include the construction of model substrates for the investigation into Diels-Alder, radical cascade, and tandem Michael addition reactions as possible routes to the family of alkaloids. Also described are the chemoenzymatic syntheses of the natural product (-)-idesolide and unnatural polyhydroxylated pyrrolidines generated from the biotransformation of benzoic acid with Ralstonia eutropha B9.

A simple change in the polarity of the solvent allows both enantiomers of substituted succinimides to be obtained in the enantioselective conjugate addition reaction of aldehydes, mainly α,α-disubstituted, to maleimides catalysed by chiral carbamate-monoprotected trans-cyclohexane-1,2-diamines. Using a single enantiomer of the organocatalyst, both enantiomers of the resulting Michael adducts are obtained in high yields by simply changing the reaction solvent from aqueous DMF (up to 84 % ee) to chloroform (up to 86 % ee). Theoretical calculations are used to explain this uncommon reversal of the enantioselectivity; two transition state orientations of different polarities are differently favoured in polar or nonpolar solvents.

Bifunctional chiral primary amine 8 containing an (S,S)-trans-cyclohexane-1,2-diamine scaffold and a 2-benzimidazole unit is used as a general organocatalyst for the Michael addition of α,α-branched aldehydes to nitroalkenes and maleimides. The reactions take place, with 20 mol % of catalyst in dichloromethane at rt for nitroalkenes and with 15 mol % catalyst loading in toluene at 10 °C for maleimides, in good yields and enantioselectivities. DFT calculations demonstrate the bifunctional character of this organocatalyst activating the aldehyde by enamine formation and the Michael acceptor by double hydrogen bonding.

During the research that it is summarized in the present memory, the activation of enals via iminium ion catalysis in different transformations has been studied. Firstly, a 1,3-dipolar cycloaddition between stable azomethine ylides and a,b-unsaturated aldehydes catalyzed by a chiral imidazolidinone derivative has been optimized. Employing this methodology we have synthesized a large range of densely substituted pyrroloisoquinolines and pyrrolophthalazines with good yields and high values of diastereo- and enantioselectivity. Moreover, a mechanistic study has been carried out based on DFT calculations and experimental data which have allowed us to propose that the (3+2) cycloaddition reaction follows a sequential Michael addition/Mannich cyclization pathway. The formation of the iminium ion as a result of the condensation between the a,b-unsaturated aldehyde and the catalyst plays an essential role, regarding both reactivity and stereoselectivity. On the other hand we have developed a methodology to carry out a cascade Michael/Henry reaction followed by a sequential dehydration. Starting from simple substrates (2-nitromethylacrilates and a,b-unsaturated aldehydes) and employing a prolinol-derivative catalyst a series of quiral nitrocyclohexadienes have been synthesized.

The new readily available sulfamide-amine alcohol 11 was found to be effective in catalyzing enantioselective phenylacetylene addition to aromatic ketones without using another central metal, providing the chiral tertiary propargylic alcohols in good yields (up to 83%) and enantioselectivities (up to 83% e.e.). The NbCl5 being a strong electrophile, is a potential candidate to act as a Lewis acid, and therefore it mediates various organic reactions. For this reason, it has received continuous attention by Brazilian researchers, especially in recent decades, since Brazil holds the largest reserves of niobium, besides being the largest producer of this element. The Michael addition reaction is one of the most widely used for forming carbon-carbon bonds and takes place by the addition of nucleophiles to activated olefins. Although this type of reaction is usually catalyzed by base, there are reports in the literature on the use of various Lewis acids in this type of reaction. The synthesis of enamines based acetilenodicarboxilates and amines, aromatic or alkyl, by Michael addition reaction is quite interesting, since these are valuable synthetic intermediates for the synthesis of heterocyclic and they are used in multicomponent reactions. The derivatives of anilino-fumarate also have a great potential for medical application. The studies conducted during my Phd thesis were focused on two different directions: 1. In one case we tried to face some long standing problems of the asymmetric aminocatalysis as the activation of encumbered carbonyl compounds and the control of the diastereoisomeric ratio in the diastero- and enantioselective construction of all carbon substituted quaternary stereocenters adjacent a tertiary one. In this section (Challenges) was described the asymmetric aziridination of ,-unsaturated ketones, the activation of ,-unsaturated -branched aldehydes and the Michael addition of oxindoles to enals and enones. For the activation via iminium ion formation of sterically demanding substrates, as ,-unsaturated ketones and ,-unsaturated -branched aldehydes, we exploited a chiral primary amine in order to overcome the problem of the iminium ion formation between the catalyst and encumbered carbonylic componds. For the control of diastereoisomeric ratio in the diastero- and enantioselective construction of all carbon substituted quaternary stereocenters adjacent a tertiary one we envisaged that a suitable strategy was the Michael addition to 3 substituted oxindoles to enals activated via LUMO-lowering catalysis. In this synthetic protocol we designed a new bifunctional catalyst with an amine moiety for activate the aldehyde and a tioureidic fragment for direct the approach of the oxindole. This part of the thesis (Challenges) could be considered pure basic research, where the solution of the synthetic problem was the goal itself of the research. 2. In the other hand (Molecules) we applied our knowledge about the carbonylic compounds activation and about cascade reaction to the synthesis of three new classes of spirooxindole in enantiopure form. The construction of libraries of these bioactive compounds represented a scientific bridge between medicinal chemistry or biology and the asymmetric catalysis.

A generalized, odorless, one-pot methodology has been developed for the preparation of 1,2-trans-thioglycosides and thio-Michael addition products of carbohydrate derivatives through triphenyl phosphine mediated cleavage of disulfides and reaction of the thiolate formed in situ with glycosyl bromides and glycosyl conjugated alkenes.
