The aim of the present study was to investigate the role of the lateral hypothalamus (LH) and its local glutamatergic neurotransmission in the cardiovascular adjustments observed when rats are submitted to acute restraint stress. Bilateral microinjection of the nonspecific synaptic inhibitor CoCl2 (0.1 nmol in 100 nL) into the LH enhanced the heart rate (HR) increase evoked by restraint stress without affecting the blood pressure increase. Local microinjection of the selective N-methyl-d-aspartate (NMDA) glutamate receptor antagonist LY235959 (2 nmol in 100 nL) into the LH caused effects that were similar to those of CoCl2. No changes were observed in the restraint-related cardiovascular response after a local microinjection of the selective non-NMDA glutamatergic receptor antagonist NBQX (2 nmol in 100 nL) into the LH. Intravenous administration of the muscarinic cholinergic receptor antagonist homatropine methyl bromide (0.2 mg/kg), a quaternary ammonium drug that does not cross the blood-brain barrier, abolished the changes in cardiovascular responses to restraint stress following LH treatment with LY235959. In summary, our findings show that the LH plays an inhibitory role on the HR increase evoked by restraint stress. Present results also indicate that local NMDA glutamate receptors, through facilitation of cardiac parasympathetic activity, mediate the LH inhibitory influence on the cardiac response to acute restraint stress. The bilateral microinjection of the CoCl2 or LY235959 into the LH enhanced the HR increase evoked by restraint stress without affecting the blood pressure increase. Intravenous administration of the homatropine methyl bromide abolished the changes in cardiovascular responses to restraint stress following LH treatment with LY235959. These results suggest that such LH influence is mediated by local NMDA glutamate receptors and involves parasympathetic nervous activation. © 2013 Federation of European Neuroscience Societies and John Wiley & Sons Ltd.

Neural plasticity has been observed in the bed nucleus of the stria terminalis (BNST) following exposure to both cocaine and androgenic-anabolic steroids. Here we investigated the involvement of the BNST on changes in cardiovascular function and baroreflex activity following either single or combined administration of cocaine and testosterone for 10 consecutive days in rats. Single administration of testosterone increased values of arterial pressure, evoked rest bradycardia and reduced baroreflex-mediated bradycardia. These effects of testosterone were not affected by BNST inactivation caused by local bilateral microinjections of the nonselective synaptic blocker CoCl2. The single administration of cocaine as well as the combined treatment with testosterone and cocaine increased both bradycardiac and tachycardiac responses of the baroreflex. Cocaine-evoked baroreflex changes were totally reversed after BNST inactivation. However, BNST inhibition in animals subjected to combined treatment with cocaine and testosterone reversed only the increase in reflex tachycardia, whereas facilitation of reflex bradycardia was not affected by local BNST treatment with CoCl2. In conclusion, the present study provides the first direct evidence that the BNST play a role in cardiovascular changes associated with drug abuse. Our findings suggest that alterations in cardiovascular function following subchronic exposure to cocaine are mediated by neural plasticity in the BNST. The single treatment with cocaine and the combined administration of testosterone and cocaine had similar effects on baroreflex activity, however the association with testosterone inhibited cocaine-induced changes in the BNST control of reflex bradycardia. Testosterone-induced cardiovascular changes seem to be independent of the BNST. © 2013 IBRO.

Aim: This study examines if injection of cobalt chloride (CoCl2) or antagonists of muscarinic cholinergic (atropine), mu(1)-opioid (naloxonazine) or 5-HT1 serotonergic (methiothepin) receptors into the dorsal or ventral portions of the anterior pretectal nucleus (APtN) alters the antinociceptive effects of stimulating the retrosplenial cortex (RSC) in rats. Main method: Changes in the nociceptive threshold were evaluated using the tail flick or incision pain tests in rats that were electrically stimulated at the RSC after the injection of saline, CoCl2 (1 mM, 0.10 mu L) or antagonists into the dorsal or ventral APtN. Key findings: The injection of CoCl2, naloxonazine (5 mu g/0.10 mu L) or methiothepin (3 mu g/0.10 mu L) into the dorsal APtN reduced the stimulation-produced antinociception from the RSC in the rat tail flick test. Reduction of incision pain was observed following stimulation of the RSC after the injection of the same substances into the ventral APtN. The injection of atropine (10 ng/0.10 mu L) or ketanserine (5 mu g/0.10 mu L) into the dorsal or ventral APtN was ineffective against the antinociception resulting from RSC stimulation. Significance: mu(1)-opioid- and 5-HT1-expressing neurons and cell processes in dorsal and ventral APtN are both implicated in the mediation of stimulation-produced antinociception from the RSC in the rat tail flick and incision pain tests, respectively. The medial amygdaloid nucleus (MeA) is a part of the limbic system and is involved in cardiovascular modulation. We previously reported that microinjection of noradrenaline (NA) into the MeA of unanesthetized rats caused pressor and bradycardiac responses, which were mediated by acute vasopressin release into the systemic circulation. In the present study, we tested the possible involvement of magnocellular neurons of the paraventricular (PVN) and/or supraoptic (SON) of the hypothalamus that synthesize vasopressin in the cardiovascular pathway activated by the microinjection of NA into the MeA. Pressor and bradycardiac responses to the microinjection of NA (27 nmol/100 nL) into the MeA were blocked by pretreatment of either the PVN or the SON with cobalt chloride (CoCl2, 1 mM/100 nL), thus indicating that both hypothalamic nuclei mediate the cardiovascular responses evoked by microinjection of NA Into the MeA. Our results suggest that the pressor and bradycardiac response caused by the microinjection of NA into the MeA is mediated by magnocellular neurons in both the PVN and SON. (C) 2012 IBRO. We have previously reported that noradrenaline (NA) microinjected into the lateral septal area (LSA) caused pressor and bradicardic responses that were mediated by vasopressin release into the circulation through the paraventricular nucleus of hypothalamus (PVN). Although PVN is the final structure involved in the cardiovascular responses caused by NA in the LSA, there is no evidence of direct connections between these areas, suggesting that some structures could be links in this pathway. In the present study, we verified the effect of reversible synaptic inactivation of the medial amygdaloid nucleus (MeA), bed nucleus of stria terminalis (BNST) or diagonal band of Broca (DBB) with Cobalt Chloride (CoCl2) on the cardiovascular response to NA microinjection into the LSA of unanesthetized rats. Male Wistar rats had guide cannulae implanted into the LSA and the MeA, BNST or DBB for drug administration, and a femoral catheter for blood pressure and heart rate recordings. Local microinjection of CoCl2 (1 mm in 100 nL) into the MeA significantly reduced the pressor and bradycardic responses caused by NA microinjection (21 nmol in 200 nL) into the LSA. In contrast, microinjection of CoCl2 into the BNST or DBB did not change the cardiovascular responses to NA into the LSA. The results indicate that synapses within the MeA, but not in BNST or DBB, are involved in the cardiovascular pathway activated by NA microinjection into the LSA.

Several studies from our group have indicated that the BNST play an important role in baroreflex modulation. However, the involvement of the BNST in the chemoreflex activity is unknown. Thus, in the present study, we investigated the effect of the local bed nucleus of stria terminalis (BNST) neurotransmission inhibition by bilateral microinjections of the non-selective synaptic blocker cobalt chloride (CoCl2) on the cardiovascular responses to chemoreflex activation in rats. For this purpose, chemoreflex was activated with KCN (i.v.) before and after microinjections of CoCl2 into the BNST. Reversible BNST inactivation produced no significant changes in the magnitude and durations of both pressor and bradycardic responses to intravenous KCN infusion. These findings suggesting that BNST neurotransmission have not influence on both sympathoexcitatory and parasympathoexcitatory components of the peripheral chemoreflex activation. Aseptic loosening of metal implants is mainly attributed to the formation of metal degradation products. These include particulate debris and corrosion products, such as metal ions (anodic half-reaction) and ROS (cathodic half-reaction). While numerous clinical studies describe various adverse effects of metal degradation products, detailed knowledge of metal-induced cellular reactions, which might be important for possible therapeutic intervention, is not comprehensive. Since endothelial cells are involved in inflammation and angiogenesis, two processes which are critical for wound healing and integration of metal implants, the effects of different metal alloys and their degradation products on these cells were investigated. Endothelial cells on Ti6Al4V alloy showed signs of oxidative stress, which was similar to the response of endothelial cells to cathodic partial reaction of corrosion induced directly on Ti6Al4V surfaces. Furthermore, oxidative stress on Ti6Al4V alloy reduced the pro-inflammatory stimulation of endothelial cells by TNF-α and LPS. Oxidative stress and other stress-related responses were observed in endothelial cells in contact with Co28Cr6Mo alloy. Importantly, these features could be reduced by coating Co28Cr6Mo with a TiO2 layer, thus favouring the use of such surface modification in the development of medical devices for orthopaedic surgery. The reaction of endothelial cells to Co28Cr6Mo alloy was partially similar to the effects exerted by Co2+, which is known to be released from metal implants. Co2+ also induced ROS formation and DNA damage in endothelial cells. This correlated with p53 and p21 up-regulation, indicating the possibility of cell cycle arrest. Since CoCl2 is used as an hypoxia-mimicking agent, HIF-1α-dependence of cellular responses to Co2+ was studied in comparison to anoxia-induced effects. Although important HIF-1α-dependent genes were identified, a more detailed analysis of microarray data will be required to provide additional information about the mechanisms of Co2+ action. All these reactions of endothelial cells to metal degradation products might play their role in the complex processes taking place in the body following metal device implantation. In the worst case this can lead to aseptic loosening of the implant and requirement for revision surgery. Knowledge of molecular mechanisms of metal-induced responses will hopefully provide the possibility to interfere with undesirable processes at the implant/tissue interface, thus extending the life-time of the implant and the overall success of metal implant applications.

Übergangsmetallen wie Nickel und Cobalt kommt meist eine große Bedeutung als Cofaktor in Enzymen oder Metallkomplexen im Metabolismus von Lebewesen zu. Da eine sehr geringe Konzentration dieser Übergangsmetalle in einer Zelle für deren Funktionalität ausreicht, ist eine konstante Konzentration der Spurenelemente in einer Zelle angestrebt. Durch meist anthropogene Einflüsse sind Pflanzen und Menschen zunehmend hohen Konzentrationen von Übergangsmetallen ausgesetzt, die in Abhängigkeit von ihrer Spezies, der Konzentration und der Lokalisation unterschiedliche Toxizitäten aufweisen können. Die Speziation von Metallen wurde bisher mittels gängiger Analyseverfahren, wie der ICP-MS und ähnlicher Verfahren, anhand von bulk-Material durchgeführt. Durch die Entwicklung von optischen Sensoren für Metallionen war es möglich, diese Metalle auch in lebenden Zellen mittels Fluoreszenzmikroskopie zu lokalisieren. Ke und Kollegen (2006, 2007) nutzten einen solchen optischen Sensor - Newport Green DCF, um die Aufnahme von Nickel in humane A543 Lungenbronchialepithelzellen nach Inkubation mit dem wasserlöslichen NiCl2 (0,5 mM und 1 mM) sowie den wasserunlöslichen Verbindungen Ni3S2 (0,5 µg/cm2 und 1 µg/cm2) und NiS (2,5 µg/cm2) nachzuweisen und zu lokalisieren und konnten damit eine Akkumulation von Nickel im Zytoplasma und im Zellkern aufzeigen. Dabei war bei wasserlöslichen und wasserunlöslichen Nickelverbindungen Nickel nach 24 h im Zytoplasma und erst nach 48 h im Zellkern zu beobachten.rnrnDa Nickel und Cobalt keine detektierbare Eigenfluoreszenz unter den gegebenen Bedingungen zeigten, wurde für den optischen Nachweis von Nickel und Cobalt mit dem konfokalen Laser-Raster Mikroskop (CLSM) nach der Zugabe der verschiedenen wasserlöslichen und wasserunlöslichen Metallverbindungen NiCl2, NiSO4, Ni3S2 und CoCl2 in einzelnen lebenden humanen Gingiva-Fibroblasten, sowie in Pflanzenzellen in dieser Arbeit ebenfalls der optische Sensor Newport Green DCF genutzt. Korrespondierend zu den Ergebnissen früherer Arbeiten von Ke et al. (2006, 2007), in denen die Nickelaufnahme bei Konzentrationen von >0,5 mM NiCl2 bzw. >0,5 µg/cm2 Ni3S2 gezeigt wurde, wurde Nickel in Fibroblasten in Abhängigkeit von der Spezies mit steigender Metallkonzentration von 100 µM bis 500 µM nach 16 h im Zytoplasma und zunehmend nach 24 h bis 48 h im Zellkern detektiert. Bei der wasserunlöslichen Verbindung Ni3S2 war der Nachweis von Nickel im Zellkern bereits nach 16 h bis 24 h erfolgreich. Zusätzlich wurden weitere Strukturen wie das Endoplasmatische Retikulum, die Mitochondrien und die Nukleoli durch eine starke Fluoreszenz des optischen Sensors bei Colokalisationsexperimenten mit Organell-spezifischen Fluoreszenzfarbstoffen als target für die Nickelbindung vermutet. Die Lokalisation von Cobalt in den Fibroblasten entsprach weitgehend der Lokalisation von Nickel. Im Zellkern war die Cobaltlokalisation jedoch auf die Nukleoli beschränkt. Weiterführende Versuche an humanen Gingiva-Fibroblasten zeigten, dass die Aufnahme der Metalle in die Fibroblasten pH-Wert abhängig war. Niedrige pH-Werte im sauren pH-Bereich verringerten die Aufnahme der Metalle in die Zellen, wobei ein pH-Wert im basischen Bereich keinen bedeutenden Unterschied zum neutralen pH-Bereich aufwies. Im Vergleich zu den Fibroblasten war in Pflanzenzellen zu jedem Zeitpunkt, auch bei geringen Konzentrationen der Metallverbindungen sowie des optischen Sensors, Nickel und Cobalt in den Zellkernen detektierbar. Durch die Eigenschaft der Pflanzenzellen eine Vakuole zu besitzen, war Nickel und Cobalt hauptsächlich in den Vakuolen lokalisiert. Weitere Strukturen wie das Endoplasmatische Retikulum, die Mitochondrien oder auch die Zellwand kamen bei Pflanzenzellen als target in Frage.rnrnDie Fluoreszenz und Lokalisation der Metalle in den Fibroblasten waren unabhängig von der Spezies sehr ähnlich, sodass in den Zellen die Spezies anhand der fluoreszenzmikroskopischen Aufnahmen kaum unterschieden werden konnten. Lambda-Scans in verschiedenen regions of interest (ROI) wurden durchgeführt, um durch die Fluoreszenzspektren Hinweise auf eine charakteristische Beeinflussung der Bindungspartner von Nickel und Cobalt oder dieser Metalle selbst in den Zellen auf den optischen Sensor zu bekommen und diese dadurch identifizieren zu können. Das Ziel der parallelen Detektion bzw. Lokalisation und gleichzeitigen Speziation bestimmter Nickel- und Cobaltpezies in einzelnen lebenden Zellen konnte in dieser Arbeit durch den optischen Sensor Newport Green DCF nicht erreicht werden.

Detached wheat shoots (ear with peduncle and flag leaf) were incubated for 4 d in a solution containing 1 mM RbCl and 1 mM SrCl2 as well as 10, 40 or 160 µM NiCl2 and CoCl2. The phloem of some plants was interrupted by steam-girdling the stem below the ear to distinguish between xylem and phloem transport. The phloem-immobile Sr flowed mainly to the leaf lamina and to the glumes via the xylem. The Sr transport was not sensitive to steam-girdling. In contrast, the phloem-mobile Rb accumulated during the incubation time mainly in the stem and the leaf sheath. The Rb transport to the grains was impaired by steam-girdling as well as by elevated Ni and Co concentrations in the incubation solution indicating that Rb was transported via the phloem to the maturing grains and that this transport was affected by the heavy metals. Ni was removed more efficiently from the xylem in the peduncle than Co (but far less efficiently than Rb). It became evident that the two heavy metals can also be transferred from the xylem to the phloem in the stem of wheat and reach the maturing grains via the phloem.

Bacterial phosphotriesterases are binuclear metalloproteins for which the catalytic mechanism has been studied with a variety of techniques, principally using active sites reconstituted in vitro from apoenzymes. Here, atomic absorption spectroscopy and anomalous X-ray scattering have been used to determine the identity of the metals incorporated into the active site in vivo. We have recombinantly expressed the phosphotriesterase from Agrobacterium radiobacter (OpdA) in Escherichia coli grown in medium supplemented with 1 mM CoCl2 and in unsupplemented medium. Anomalous scattering data, collected from a single crystal at the Fe-K, Co-K and Zn-K edges, indicate that iron and cobalt are the primary constituents of the two metal-binding sites in the catalytic centre (alpha and P) in the protein expressed in E. coli grown in supplemented medium. Comparison with OpdA expressed in unsupplemented medium demonstrates that the cobalt present in the supplemented medium replaced zinc at the beta-position of the active site, which results in an increase in the catalytic efficiency of the enzyme. These results suggest an essential role for iron in the catalytic mechanism of bacterial phosphotriesterases, and that these phosphotriesterases are natively heterobinuclear iron-zinc enzymes.

The ability of SnCl2.2H2O as catalyst to promote the Biginelli three-component condensation reaction from a diversity of aromatic aldehydes, ethyl acetoacetate and urea or thiourea is described. The reaction was carried out in acetonitrile or ethanol as solvents in neutral media and represents an improvement of the classical Biginelli protocol and an advantage in comparison with FeCl3.6H2O, NiCl2.6H2O and CoCl2.6H2O which were used with HCl as co-catalyst. The synthesis of 3,4-dihydropyrimidinones was achieved in good to excelent yields.
