Six Bos taurus (Hereford) steers (body weight 324 22 kg) were used in a 45-day study with a replicated 3 x 3 Latin-square design. Three treatments [ad libitum feeding (ADLIB); limit feeding, 85% of ad libitum (LIMIT); bunk management feeding where steers were only given access to feed from 1600 to 0800 hours the following day (BUNK)] were imposed over 3 periods, with 2 steers assigned to each treatment in each period. Cattle were managed in a temperature-controlled metabolism unit and were exposed to both thermoneutral (17.7degreesC-26.1degreesC) and hot (16.7degreesC-32.9degreesC) environmental conditions. By design, during the thermoneutral period, the ADLIB cattle displayed greater intake (P < 0.05) than the LIMIT group, with the BUNK group being intermediate. However, during the hot period, both the LIMIT and BUNK treatment groups increased feed intake 4-5%, whereas feed intake of the ADLIB treatment group declined nearly 2%. During both periods respiration rate (RR, breath/min) followed the same pattern that was observed for feed intake, with the greatest (P < 0.05) RR found in the ADLIB treatment group (81.09 and 109.55, thermoneutral and hot, respectively) and lowest (P < 0.05) RR in the LIMIT treatment group (74.47 and 102.76, thermoneutral and hot, respectively). Rectal temperature (RT) did not differ among treatments during the thermoneutral period or the first hot day, although during the thermoneutral period the ADLIB treatment group did tend to display a lower RT, possibly as a result of other physiological processes (pulse rate and RR) aiding to keep RT lower. During the hot period, differences in RT were found on Day 5, with the LIMIT cattle having lower (P < 0.10) RT (38.92degreesC) than the ADLIB (39.18degreesC) cattle, with BUNK cattle RT (39.14degreesC) being intermediate. However, when hourly data were examined, the ADLIB cattle had greater(P < 0.05) RT than the BUNK and LIMIT at 1800 hours and greater RT (P < 0.05) than the LIMIT group at 1400, 1500, and 1600 hours. Clearly, a change in diurnal RT pattern was obtained by using the LIMIT and BUNK feeding regimen. Both of these groups displayed a peak RT during the hot conditions, between 2100 and 2200 hours, whereas the ADLIB group displayed a peak RT between 1400 and 1500 hours, a time very close to when peak climatic stress occurs. Based on these results it is apparent that feedlot managers could alleviate the effects of adverse hot weather on cattle by utilising either a limit-feeding regimen or altering bunk management practices to prevent feed from being consumed several hours prior to the hottest portion of the day.

Il sito archeologico di Arslantepe (provincia di Malatya, Turchia) rappresenta un caso di studio di potenziale interesse per l’interazione tra i mutamenti climatici e la storia della civiltà. Il sito, occupato quasi ininterrottamente per un periodo di tempo relativamente lungo (6250-2700 BP), ha fornito una grande quantità di reperti ossei, distribuiti lungo una stratigrafia archeologica relativamente dettagliata e supportata da datazioni al radiocarbonio. Tali reperti, indagati con le tecniche della geochimica degli isotopi stabili, possono costituire degli efficaci proxy paleoclimatici. In questo lavoro è stata studiata la composizione isotopica di 507 campioni di resti ossei umani e animali (prevalentemente pecore, capre, buoi). I rapporti isotopici studiati sono relativi a ossigeno (δ18Ocarb, δ18Oph), carbonio (δ13Ccarb, δ13Ccoll) e azoto (δ15N), misurati nella frazione minerale e organica dell’osso; la variabilità nel tempo di questi parametri, principalmente legati alla paleonutrizione, può essere correlata, direttamente o indirettamente, a cambiamenti dei parametri ambientali quali temperatura e umidità atmosferiche. I risultati indicano che la dieta degli animali selvatici e domestici di Arslantepe era quasi esclusivamente a base di piante a ciclo fotosintetico C3, generalmente tipiche di climi umidi o temperati. La presenza di piante C4, più tipiche di condizioni aride, sembrerebbe essere riconoscibile solamente nella dieta del bue (Bos taurus). La dieta umana era esclusivamente terrestre a base di cereali e carne di caprini con una percentuale esigua o del tutto assente di carne di maiale e bue. Dal punto di vista paleoclimatico il principale risultato del lavoro consiste nel riconoscimento della preservazione di un segnale paleoclimatico a lungo termine (δ18OW, composizione isotopica dell’ossigeno dell’acqua ingerita), che identifica un massimo relativo di umidità attorno al 5000 BP e che si correla, per andamento e ampiezza della variazione a record paleoclimatici di sedimenti lacustri collocati in regioni adiacenti all’area di studio. Sulla base del confronto dei tre segnali isotopici sono state inoltre riconosciute due anomalie climatiche siccitose a breve termine, apparentemente riferibili a due episodi di aridità a scala regionale documentati in letteratura.

La trazabilidad y el correcto etiquetado de los piensos y sus ingredientes son factores esenciales para prevenir fraudes y garantizar la seguridad alimentaria. En el ámbito de la lucha contra las Encefalopatías Espongiformes Transmisibles (EETs), la prohibición de la Unión Europea (UE) de alimentar a rumiantes y otros animales de granja con harinas de carne y huesos derivadas de animales, hace necesaria la disponibilidad de metodologías que permitan identificar el origen de las materias primas e ingredientes presentes en los piensos. El método oficial de análisis microscópico tradicionalmente empleado para este fin presenta limitaciones a la hora de diferenciar entre los huesos de mamíferos y de aves, así como para determinar el origen animal específico de las partículas detectadas. Por ello, una de las prioridades de la UE en los últimos años ha sido potenciar la búsqueda y desarrollo de técnicas analíticas alternativas que permitan la detección específica de todos los componentes que integran los piensos. Teniendo en cuenta estos aspectos, en esta Tesis Doctoral se han desarrollado técnicas de PCR en tiempo real con sondas TaqMan® para el control de autenticidad y trazabilidad de ingredientes de origen animal utilizados en la fabricación de los piensos. Las especies objeto de este trabajo han sido: vaca (Bos taurus), oveja (Ovis aries), cabra (Capra hircos), grupo rumiante, cerdo (Sus scrofa), pollo (Gallus gallos), pavo (Meleagris g-allopavo), pato (Anal platyrhynchos x Cairina moschata), oca (Anser anser), grupo aviar, caballo (Equus caballus), conejo (Oryctolagus cuniculus), liebre (Lepus capensis), grupo lepórido (conejo y liebre) y pescados...

A carne continua a ser a fonte proteica mais comum no quotidiano das pessoas. Além disso, os produtos cárneos processados apresentam-se como uma mais-valia nas suas vidas agitadas. Este tipo de produto torna difícil a diferenciação das carnes utilizadas na sua confecção, sendo por isso propícios a adulteração. A Reacção em Cadeia da Polimerase (PCR) tem ganho cada vez mais importância nos laboratórios de biologia molecular, revelando-se uma técnica de análise rápida, sensível e altamente específica na identificação de espécies em produtos alimentares. No entanto, vários factores podem interferir com o processo de amplificação, pelo que alguns cuidados devem ser implementados desde a aquisição da amostra a analisar, ao seu acondicionamento e posterior extração de ADN. Existem inúmeros protocolos de extração de ADN, devendo para cada estudo avaliar-se e optar-se pelo mais adequado, considerando a finalidade estabelecida para a amostra extraída. O trabalho laboratorial apresentado nesta dissertação baseou-se em três etapas principais. Inicialmente, avaliaram-se diferentes protocolos de extração de ADN, utilizando-se amostras de carne adquiridas num talho. Entre os protocolos testados, o método de Brometo de Cetil-Trimetil-Amónio (CTAB) modificado foi o que permitiu obter amostras de ADN com maior concentração e elevado nível de pureza. Posteriormente, foram testados e optimizados diferentes protocolos de amplificação, por PCR em tempo real, para a detecção das espécies Bos taurus (vaca), Sus scrofa (porco), Equus caballus (cavalo) e Ovis aries (ovelha). Foram empregues primers específicos de espécie para a detecção de genes mitocondriais e genómicos, consoante cada protocolo. Para o caso concreto do porco, foi efectuada a avaliação de dois protocolos, singleplex com EvaGreen® e tetraplex com AllHorse, para possível aplicação dos mesmos na sua quantificação. Os resultados demonstraram elevada especificidade e sensibilidade das reacções para esta espécie, permitindo a sua detecção até um limite de 0,001 ng e 0,1%, respectivamente. Somente a primeira metodologia se mostrou adequada para quantificação. Por último, as metodologias sugeridas foram aplicadas com sucesso na análise de 4 amostras comerciais de hambúrgueres, tendo-se verificado a consistência da rotulagem em todos os casos, no que concerne a composição em termos de espécies animais. O interesse de trabalhos neste âmbito recai na importância da autenticidade dos rótulos de produtos alimentares, principalmente nos produtos cárneos, para segurança dos consumidores e salvaguarda dos produtores.

Resumen Esta revisión compila el desarrollo y la dinámica folicular con una perspectiva histológica y de biología del desarrollo, desde la etapa fetal hasta prepuberal en bovinos (Bos taurus). Se trata de un proceso complejo y coordinado, con cambios fisiológicos y morfológicos que intervienen en la diferenciación y el desarrollo ovocitario. Los folículos son la unidad fundamental del ovario y comandan los procesos reproductivos y las fases del ciclo estral, con dos funciones fundamentales, la producción de hormonas y de ovocitos. La formación de folículos primordiales se conoce como el proceso de ensamblaje folicular, en el cual “nidos” de ovocitos completan su proliferación mitótica y comienzan la meiosis, mientras que muchos de estos folículos se vuelven apoptóticos, favoreciendo la desorganización de los nidos. El proceso de formación de folículos primordiales comienza durante el desarrollo fetal en bovinos, aproximadamente a los 80 días de gestación. Existe gran variación individual en el número de folículos, existiendo un pool de reserva ya al nacimiento. De unos 2.700.000 folículos primordiales constituidos al final del ensamblaje folicular, 90% degeneran, quedando al nacimiento aproximadamente 135.000 células germinales, las cuales declinan rápidamente hasta la pubertad y luego de ella. Los folículos primordiales se desarrollan a folículos primario, secundario, antral y preovularorio (en hembras adultas). Estas transiciones implican cambios citológicos, histológicos y morfológicos que son descritos. En terneras, igual que en animales adultos, ocurren ondas de crecimiento folicular, que aparecen a la segunda semana de vida. En la pubertad se observan importantes cambios endócrinos y ováricos y es influida por múltiples factores. El desarrollo folicular está íntimamente vinculado al futuro desempeño reproductivo de las hembras, por lo cual su conocimiento es fundamental para un adecuado manejo reproductivo de la hembra bovina y para ampliar las investigaciones vinculadas a esta disciplina

In response to uncertainty among cattle producers in Australia regarding the need to treat Bos indices and B. indicus crossbreeds, the scientific literature relating to the productivity effects of Boophilus microplus on cattle of all breeds was reviewed. Estimates of the mean effect of each engorging tick (damage coefficient, d) were made from a simple analysis of the reported data. On average, each engorging female tick is responsible for the loss of 1.37 +/- 0.25 g bodyweight in B. taunts cattle. The comparable value for B. taurus x B. indicus cattle is 1.18 +/- 0.21 g/engorging tick. These values were not statistically significantly different, indicating that if a threshold approach to tick control were taken, then the threshold number of standard ticks would be the same regardless of cattle genotype. No studies provided useable estimates of the effect of tick infestation on pure B. indices cattle. An economic threshold for treatment, below which acaricide application is not beneficial, can be predicted, using known values for the cost of acaricide application and the price of beef. However, the application of a threshold approach to control has not been embraced by government advisers and runs contrary to the accepted principals of strategic control programs. Three experiments were conducted in the dry tropics of north Australia using Bos indicus-cross cows. Cows in mid-late pregnancy were either unsupplemented during the late dry season or offered ad libitum (2 kg/day) molasses with 7.4% urea (w/w) (M8U) or cottonseed meal (1 kg/day) for up to 54 days commencing 2 months before the start of the calving season. Ovaries (n = 140) from 70 mixed-age multiparous, lactating Brahman cross (3/4-7/8 Bos indicus) cows were used to examine the hypothesis that counts of follicles visible on the surface of the ovaries of Bos indicus cows and their classification into diameter size classes, are closely correlated with numbers of follicles in those size classes found by complete dissection of the ovary. immediately after ovariectomy, mean diameters (long and short axes averaged) of all follicles greater than or equal to 2 mm visible on the surface of each ovary were measured. All follicles greater than or equal to 2 mm were dissected from the ovaries, excess stroma removed and follicle diameters measured under a stereomicroscope using an ocular graticule. For each ovary, follicles were classified in either small (8 mm) categories based on either diameters of surface or dissected follicles. Data for numbers of surface and dissected follicles (mean +/- SE) in small, medium, large categories and total follicle numbers, respectively, were 24.4 +/- 1.6 vs. 28.0 +/- 1.9, 1.6 +/- + 0.2 vs. 11.6 +/- 1.0, 0.5 +/- 0.1 vs. 0.7 +/- 0.1 and 26.4 +/- 1.6 vs. 40.4 +/- 2.5. Correlation coefficients (r) for counts of surface and dissected follicles in small, medium, large and total follicle numbers were 0.76, 0.40, 0.69 and 0.79, respectively. Medium size follicles presented only a small translucent area on the surface of the ovary, leading to an underestimate of numbers when categorised by surface evaluation. Counts of follicles visible on the surface of the ovaries of Bos indicus cows and their classification into size classes based on estimated diameter, are closely correlated with numbers of follicles in those size classes found at dissection of the ovary for small (8 mm) and total follicles but not for medium sized (4-8 mm) follicles. Two experiments were conducted to investigate the effects of equine chorionic gonadotropin (eCG) at progestin removal and gonadotropin-releasing hormone (GnRH) at timed artificial insemination (TA!) on ovarian follicular dynamics (Experiment 1) and pregnancy rates (Experiment 2) in suckled Nelore (Bos indicus) cows. Both experiments were 2 x 2 factorials (eCG or No eCG, and GnRH or No GnRH), with identical treatments. In Experiment 1, 50 anestrous cows, 134.5 +/- 2.3 d postpartum, received a 3 mg norgestomet ear implant se, plus 3 mg norgestomet and 5 mg estradiol valerate im on Day 0. The implant was removed on Day 9, with TAI 54 h later. Cows received 400 IU eCG or no further treatment on Day 9 and GnRH (100 mu g gonadorelin) or no further treatment at TAI. Treatment with eCG increased the growth rate of the largest follicle from Days 9 to 11 (means +/- SEM, 1.53 +/- 0.1 vs. 0.48 +/- 0.1 mm/d; P < 0.0001), its diameter on Day 11(11.4 +/- 0.6 vs. 9.3 +/- 0.7 mm; P = 0.03), as well as ovulation rate (80.8% vs. 50.0%, P = 0.02), whereas GnRH improved the synchrony of ovulation (72.0 +/- 1.1 VS. 71.1 +/- 2.0 h). In Experiment 2 (n = 599 cows, 40 to 120 d postpartum), pregnancy rates differed (P = 0.004) among groups (27.6%, 40.1%, 47.7%, and 55.7% for Control. GnRH, eCG, and eCG + GnRH groups). Both eCG and GnRH improved pregnancy rates (51.7% vs. 318%, P = 0.002; and 48.0% vs 37.6%, P = 0.02, respectively), although their effects were not additive (no significant interaction). In conclusion, eCG at norgestomet implant removal increased the growth rate of the largest follicle (LF) from implant removal to TAI, the diameter of the LF at TAI, and rates of ovulation and pregnancy rates. Furthermore, GnRH at TAI improved the synchrony of ovulations and pregnancy rates in postpartum Nelore cows treated with a norgestomet-based TAI protocol. (C) 2010 Elsevier Inc. All rights reserved.

The aim of the present study was to determine the relationship among body weight (BW), body condition score (BCS) and rump fat thickness (RFAT) measured by ultrasonography, and validate the relationship between BCS and RFAT over the time. Two hundred sixty and six Nelore cows had their BW, BCS and RFAT evaluated at five different moments during the production cycle: M1) weaning: M2) parturition, M3) 42 days post-partum; M4) 82 days postpartum and M5) 112 days post-partum. A BCS value was attributed for each cow following a I to 5 points scale. Ultrasonographic images for RFAT measurement were obtained using a 3.5 MHz linear transducer. Images were immediately analyzed as soon as they were formed and frozen. Body condition scores and ultrasound measurements were collected on the same day by a single trained technician. The relationship between BCS and RFAT values was investigated by regression models. The analysis of similarity among the five obtained models was performed using the proc MIXED from SAS and the correlations among variables were analyzed with proc CORR from SAS. The BCS was able to predict RFAT in Nelore cows in all different moments evaluated. Also, it was shown that BCS presented high correlation (r=0.82 to 0.93) and relationship (R(2) = 0.73 to 0.92) with RFAT. However, both BCS and RFAT showed low correlation (r=0.37 to 0.50) and relationship (R(2) = 0.13 to 0.25) with BW. The BCS classification by visual method using a 1 to 5 point scale, was able to predict the RFAT in Nelore cows over the time. (C) 2008 Elsevier B.V. All rights reserved.

Considering, that there is limited information about the preovulatory LH surge in Zebu Cattle (Bos indicus). the purpose of the present work was to assess the LH surge in Nelore cows during the estrous cycle and after ovarian superestimulation of ovarian follicular development with FSH. This information is particularly important to improve superovulatory protocols associated with fixed-time artificial insemination. Nelore cows (n = 12) had their estrus synchronized with all intravaginal device containing progesterone (CIDR-B (R)) associated with estradiol benzoate administration (EB, 2.5 mg, i.m., Day 0). Eight days later all animals were treated with PGF2 alpha (Day 8) in the morning (8:00 h) and at night, when CIDR devices were removed (20:00 11). Starting 38 h after the first PGF2 alpha injection, blood sampling and ovarian ultrasonography took place every 4 h, during 37 consecutive hours. Frequent handling may have resulted in a stress-induced suppression of LH secretion resulting in only 3 of 12 cows having ovulations at 46.7 +/- 4.9 and 72.3 +/- 3.8 h, respectively, after removal of CIDR-B. Thirty days later, the same animals received the described hormonal treatment associated with FSH (Folltropin (R) total dose = 200 mg) administered twice a day, during 4 consecutive days, starting on Day 5. Thirty-six hours after the first injection of PGF2 alpha, to minimize stress. only seven blood samples were collected at 4 h interval each. and ultrasonography was performed every 12 h until ovulation. In 11 of 12 cows (92%) the LH surge and ovulation were observed 34.6 +/- 1.6 and 59.5 +/- 1.9 h. respectively. after removal of progesterone source. The maximum values for LH in those animals were 19.0 +/- 2.6 ng/ml (mean +/- S.E.M.). It is concluded that, in Nelore COWS submitted to a ovarian superstimulation Protocol, the LH surge occurs approximately 35 It after removal of intravaginal device containing progesterone, and approximately 12h before the LH surge observed after an induced estrus without ovarian superstimulation (C) 2008 Elsevier B.V.
