Isotermas de dessorção de pimenta-de-macaco foram determinadas pelo método gravimétrico estático nas temperaturas de 35, 45 e 55 ºC, com umidade relativa variando de 5,5-81%. Três modelos matemáticos foram aplicados para analisar os dados experimentais. O modelo de GAB modificado apresentou o melhor ajuste aos dados experimentais. O calor isostérico e a entropia diferencial foram determinados pela aplicação das equações de Clausius-Clapeyron e Gibbs-Helmholtz, respectivamente. O calor isostérico e a entropia da isoterma de dessorção apresentaram comportamento similar. A teoria da compensação entalpia-entropia foi aplicada às isotermas indicando que o mecanismo de dessorção de umidade das partes aéreas de pimenta-de-macaco é controlado pela entalpia.

Background: A current challenge in gene annotation is to define the gene function in the context of the network of relationships instead of using single genes. The inference of gene networks (GNs) has emerged as an approach to better understand the biology of the system and to study how several components of this network interact with each other and keep their functions stable. However, in general there is no sufficient data to accurately recover the GNs from their expression levels leading to the curse of dimensionality, in which the number of variables is higher than samples. One way to mitigate this problem is to integrate biological data instead of using only the expression profiles in the inference process. Nowadays, the use of several biological information in inference methods had a significant increase in order to better recover the connections between genes and reduce the false positives. What makes this strategy so interesting is the possibility of confirming the known connections through the included biological data, and the possibility of discovering new relationships between genes when observed the expression data. Although several works in data integration have increased the performance of the network inference methods, the real contribution of adding each type of biological information in the obtained improvement is not clear. Methods: We propose a methodology to include biological information into an inference algorithm in order to assess its prediction gain by using biological information and expression profile together. We also evaluated and compared the gain of adding four types of biological information: (a) protein-protein interaction, (b) Rosetta stone fusion proteins, (c) KEGG and (d) KEGG+GO. Results and conclusions: This work presents a first comparison of the gain in the use of prior biological information in the inference of GNs by considering the eukaryote (P. falciparum) organism. Our results indicates that information based on direct interaction can produce a higher improvement in the gain than data about a less specific relationship as GO or KEGG. Also, as expected, the results show that the use of biological information is a very important approach for the improvement of the inference. We also compared the gain in the inference of the global network and only the hubs. The results indicates that the use of biological information can improve the identification of the most connected proteins.

In this article we introduce a three-parameter extension of the bivariate exponential-geometric (BEG) law (Kozubowski and Panorska, 2005) [4]. We refer to this new distribution as the bivariate gamma-geometric (BGG) law. A bivariate random vector (X, N) follows the BGG law if N has geometric distribution and X may be represented (in law) as a sum of N independent and identically distributed gamma variables, where these variables are independent of N. Statistical properties such as moment generation and characteristic functions, moments and a variance-covariance matrix are provided. The marginal and conditional laws are also studied. We show that BBG distribution is infinitely divisible, just as the BEG model is. Further, we provide alternative representations for the BGG distribution and show that it enjoys a geometric stability property. Maximum likelihood estimation and inference are discussed and a reparametrization is proposed in order to obtain orthogonality of the parameters. We present an application to a real data set where our model provides a better fit than the BEG model. Our bivariate distribution induces a bivariate Levy process with correlated gamma and negative binomial processes, which extends the bivariate Levy motion proposed by Kozubowski et al. (2008) [6]. The marginals of our Levy motion are a mixture of gamma and negative binomial processes and we named it BMixGNB motion. Basic properties such as stochastic self-similarity and the covariance matrix of the process are presented. The bivariate distribution at fixed time of our BMixGNB process is also studied and some results are derived, including a discussion about maximum likelihood estimation and inference. The Hsp70 is an essential molecular chaperone in protein metabolism since it acts as a pivot with other molecular chaperone families. Several co-chaperones act as regulators of the Hsp70 action cycle, as for instance Hip (Hsp70-interacting protein). Hip is a tetratricopeptide repeat protein (TPR) that interacts with the ATPase domain in the Hsp70-ADP state, stabilizing it and preventing substrate dissociation. Molecular chaperones from protozoans, which can cause some neglected diseases, are poorly studied in terms of structure and function. Here, we investigated the structural features of Hip from the protozoa Leishmania braziliensis (LbHip), one of the causative agents of the leishmaniasis disease. LbHip was heterologously expressed and purified in the folded state, as attested by circular dichroism and intrinsic fluorescence emission techniques. LbHip forms an elongated dimer, as observed by analytical gel filtration chromatography, analytical ultracentrifugation and small angle X-ray scattering (SAXS). With the SAXS data a low resolution model was reconstructed, which shed light on the structure of this protein, emphasizing its elongated shape and suggesting its domain organization. We also investigated the chemical-induced unfolding behavior of LbHip and two transitions were observed. The first transition was related to the unfolding of the TPR domain of each protomer and the second transition of the dimer dissociation. Altogether. LbHip presents a similar structure to mammalian Hip, despite their low level of conservation, suggesting that this class of eukaryotic protein may use a similar mechanism of action. This artwork reports on two different projects that were carried out during the three years of Doctor of the Philosophy course. In the first years a project regarding Capacitive Pressure Sensors Array for Aerodynamic Applications was developed in the Applied Aerodynamic research team of the Second Faculty of Engineering, University of Bologna, Forlì, Italy, and in collaboration with the ARCES laboratories of the same university. Capacitive pressure sensors were designed and fabricated, investigating theoretically and experimentally the sensor’s mechanical and electrical behaviours by means of finite elements method simulations and by means of wind tunnel tests. During the design phase, the sensor figures of merit are considered and evaluated for specific aerodynamic applications. The aim of this work is the production of low cost MEMS-alternative devices suitable for a sensor network to be implemented in air data system. The last two year was dedicated to a project regarding Wireless Pressure Sensor Network for Nautical Applications. Aim of the developed sensor network is to sense the weak pressure field acting on the sail plan of a full batten sail by means of instrumented battens, providing a real time differential pressure map over the entire sail surface. The wireless sensor network and the sensing unit were designed, fabricated and tested in the faculty laboratories. A static non-linear coupled mechanical-electrostatic simulation, has been developed to predict the pressure versus capacitance static characteristic suitable for the transduction process and to tune the geometry of the transducer to reach the required resolution, sensitivity and time response in the appropriate full scale pressure input A time dependent viscoelastic error model has been inferred and developed by means of experimental data in order to model, predict and reduce the inaccuracy bound due to the viscolelastic phenomena affecting the Mylar® polyester film used for the sensor diaphragm. The development of the two above mentioned subjects are strictly related but presently separately in this artwork.

The Calabrian-Peloritani arc represents key site to unravel evolution of surface processes on top of subducting lithosphere. During the Pleistocene, in fact the arc uplifted at rate of the order of about 1mm/yr, forming high-standing low-relief upland (figure 2). Our study is focused on the relationship between tectonic and land evolution in the Sila Massif, Messina strait and Peloritani Mts. Landforms reflect a competition between tectonic, climatic, and surficial processes. Many landscape evolution models that explore feedbacks between these competing processes, given steady forcing, predict a state of erosional equilibrium, where the rates of river incision and hillslope erosion balance rock uplift. It has been suggested that this may be the final constructive stage of orogenic systems. Assumptions of steady erosion and incision are used in the interpretation of exhumation and uplift rates from different geologic data, and in the formulation of fluvial incision and hillslope evolution models. In the Sila massif we carried out cosmogenic isotopes analysis on 24 samples of modern fluvial sediments to constrain long-term (~103 yr) erosion rate averaged on the catchment area. 35 longitudinal rivers profiles have been analyzed to study the tectonic signal on the landscape evolution. The rivers analyzed exhibit a wide variety of profile forms, diverging from equilibrium state form. Generally the river profiles show at least 2 and often 3 distinct concave-up knickpoint-bounded segments, characterized by different value of concavity and steepness indices. River profiles suggest three main stages of incision. The values of ks and θ in the lower segments evidence a decrease in river incision, due probably to increasing uplift rate. The cosmogenic erosion rates pointed out that old landscape upland is eroding slowly at ~0.1 mm/yr. In the contrary, the flanks of the massif is eroding faster with value from 0.4 to 0.5 mm/yr due to river incision and hillslope processes. Cosmogenic erosion rates mach linearly with steepness indices and with average hillslope gradient. In the Messina area the long term erosion rate from low-T thermochronometry are of the same order than millennium scale cosmogenic erosion rate (1-2 mm/yr). In this part of the chain the fast erosion is active since several million years, probably controlled by extensional tectonic regime. In the Peloritani Mts apatite fission-track and (U-Th)/He thermochronometry are applied to constraint the thermal history of the basement rock. Apatite fission-track ages range between 29.0±5.5 and 5.5±0.9 Ma while apatite (U-Th)/He ages vary from 19.4 to 1.0 Ma. Most of the AFT ages are younger than the overlying terrigenous sequence that in turn postdates the main orogenic phase. Through the coupling of the thermal modelling with the stratigraphic record, a Middle Miocene thermal event due to tectonic burial is unravel. This event affected a inner-intermediate portion of the Peloritani belt confined by young AFT data (<15 Ma) distribution. We interpret this thermal event as due to an out-of–sequence thrusting occurring in the inner portion of the belt. Young (U-Th)/He ages (c. 5 Ma) record a final exhumation stage with increasing rates of denudation since the Pliocene times due to postorogenic extensional tectonics and regional uplift. In the final chapter we change the spatial scale to insert digital topography analysis and field data within a geodynamic model that can explain surface evidence produced by subduction process.

Das Wachstum von Nervenzellen und deren Verbindungen im zentralen und peripheren Nervensystem wird durch Proteine der extrazellulären Matrix kontrolliert. In dieser Arbeit wurde das Matrixprotein Laminin verwendet, um Netzwerke von Nervenzellen auf künstlichen Substraten in vitro zu erzeugen. Zu diesem Zweck wurden Lamininstrukturen mit Mikrostempeln aus Polydimethylsiloxan auf Zellkultursubstrate übertragen. Die Mikrostempel wurden in einem mehrstufigen Verfahren durch Abformung von photolithographisch hergestellten Masken angefertigt. Nach Vorversuchen mit neuronal differenzierten Zellen der Zellinien MzN und P19 zur Identifizierung geeigneter Abmessungen der Mikrotrukturen, gelang die Realisierung von Linien- und Gitternetzwerken sowie von komplexeren Schaltungen. Eine morphologische Charakterisierung der erzeugten Netzwerke erfolgte durch Phasenkontrast- und Fluoreszenzmikroskopie.Elektrophysiologische Messungen wurden mit der Patch-Clamp Technik an einer Kultur von Nervenzellen aus primär isolierten Hirnschnitten durchgeführt. Der Erhalt des intakten Zellverbundes im Hirnschnitt sollte Bedingungen möglichst nahe zur Situation in vivo schaffen, um die Bildung von Synapsen zu begünstigen. In Patch-Clamp Messungen an bis zu drei Neuronen gleichzeitig, gelang der Nachweis synaptischer Kopplung in strukturierten Netzwerken solcher Hirnschnitt-Kulturen. Sowohl funktionale chemische Synapsen, als auch Ohm'sche Kopplung über Gap-Junctions wurde beobachtet. Es wurde ein elektrisches Kopplungsmodell abgeleitet. Die Signalleitung in den Nervenfasern erfolgt demnach wie in einem zylindrischen, durch die Zellmembran von der Umgebung isolierten Kabel.

In this thesis Marsili back-arc basin and Palinuro Volcanic Complex (Southern Tyrrhenian Sea) have been investigated by using magnetic, bathymetric and gravimetric data. A new velocity model of opening of the Marsili basin has been proposed, highlighting the transition from the horizontal spreading of the back-arc to the vertical accretion of the Marsili seamount. Introducing gravity data, Marsili's internal structure has been modeled and a huge portion of the volcano with low density and vanishing magnetization has been detected. Forward modeling of Palinuro Volcanic Complex showed as Palinuro represents the shallowest evidence of a deep tectonic discontinuity and the possible transition domain between the oceanic crust of Marsili Basin and the continental crust related to the Appenninic chain.

Scopo del presente lavoro di ricerca è quello di comparare due contesti metropolitani, valenciano e bolognese, sulle pratiche di accompagnamento al lavoro rivolte a fasce svantaggiate, in particolare persone con problemi di dipendenza da sostanze psicotrope. L’indagine propone un confronto su alcune tematiche trasversali (tipologia di azioni messe in campo, organizzazione territoriale e governance, profilo degli utenti, inserimento sociale, coinvolgimento del mondo produttivo) e pone in evidenza gli elementi che ci consentono di individuare e segnalare sia delle buone pratiche trasferibili sia delle linee progettuali, partendo dunque dal presupposto che capacitare una persona significa innanzitutto offrirle congrue opportunità di scelta, nel senso seniano e come spiegato dalla stessa Nussbaum, ma soprattutto accompagnarla e sostenerla nel percorso di inserimento lavorativo e, in parallelo, sociale. Il bisogno raccolto è quello di un sostegno, motivazionale e orientativo, che segua un approccio socio educativo capace di fornire, alla persona, una risposta integrata, di unicità, capace dunque di agire sull’autonomia, sull’autostima, sull’elaborazione delle proprie esperienze di vita e lavorative, nonché su elementi anche di contesto quali la casa, le reti amicali e familiari, spesso compromesse. L’elemento distintivo che consente di agire in questa direzione è il lavoro di collaborazione tra i diversi servizi e la co-progettazione del percorso con l’utente stesso. Il tema degli inserimenti lavorativi è un argomento molto complesso che chiama in causa diversi aspetti: i mutamenti sociali e le trasformazioni del lavoro; l’emergere di nuove fasce deboli e il rischio di aggravamento delle condizioni di esclusione per le fasce deboli “tradizionali”; l’importanza del lavoro per la costruzione di percorsi identitari e di riconoscimento; l’impatto delle politiche attive sulle fasce svantaggiate e i concetti di capitazione e attivazione; il ruolo del capitale sociale e l’emergere di nuovi welfare; la rete degli attori coinvolti dal processo di inserimento e il tema della governace territoriale.

Longstanding debates concerning the origin of the Kess Kess Emsian carbonate mounds exposed at Hamar Laghdad Ridge (eastern Anti-Atlas, Morocco) centre around the processes that induced precipitation of carbonate mud and the preservation of steep morphologies. Although in the last years an origin related to hydrothermalism seemed to be more likely, to date the Kess Kess are still considered controversial vent deposits. This study combines in updated research review information from previous work and new detailed field observations coupled with new analytical results to define a consistent framework and some new insight of current knowledge about the origin of these mounds. We obtain a complete minero-petrographic and palaeobiological data set and a detailed geochemical characterization of the different lithologies and facies of the Hamar Laghdad stratigraphic succession, including mounds, and we compared the results with the data from Maïder Basin mounds (Anti-Atlas, Morocco). Our data support the hydrothermal model proposed for the genesis and development of the Kess Kess mounds. The mechanisms linked to the mounds formation and growth are discussed in the light of the new finding of fluid-sediment interaction within a scenario driven by late magmatic fluids circulation. Conical mounds and other fluids related morphologies were also reported from Crommelin crater area (Arabia Terra, Mars). These mounds consist in meter-sized conical buildups hosted in the Equatorial Layered Deposits (ELDs) deposed during a regional groundwater fluid upwelling. Geometries and geological conditions that might have controlled the development of such morphologies were discussed. According to our data the morphological and stratigraphical characteristics of Crommelin area mounds are most consistent with a formation by fluids advection. Then we compare terrestrial and Martian data and examine the geological settings of hydrothermal mound occurrences on Earth in order to describe potential target areas for hydrothermal structures on Mars.

TGF-beta ist ein Schlüsselmolekül zellvermittelter Immuntoleranz. So spielt es neben seiner pleiotropen Rolle in Immunzellen auch bei der Tumorentwicklung eine große Rolle. Das TGF-beta hat bei der Tumorentwicklung eine duale Rolle. So dient es in frühen Phasen als Tumorsuppressor, währenddessen es in späten Phasen der Entwicklung als Tumorpromotor wirkt. Eine strikte Regulation des TGF-beta Signalweges ist daher für ein funktionierendes Immunsystem von essentieller Bedeutung. Die Ubiquitin Ligase Smurf2 ist dabei ein wichtiger negativ Regulator des TGF-beta Signalweges.In der vorliegenden Arbeit konnte eine neue Spleißform des Smurf2 (dE2Smurf2) aus murinen CD4+ T-Zellen isoliert werden, deren Funktion in vitro und in vivo in T-Lymphozyten untersucht worden ist. Für diese Spleißform konnte zudem eine humane Relevanz nachgewiesen werden. Mit Hilfe von Überexpressionen in Cos7 Zellen konnte eine veränderte Lokalisation der Smurf2 Spleißformen (WT und dE2) festgestellt werden. Dabei konnten lysosomale und endosomale Kompartimente bei der Kolokalisation mit dem dE2Smurf2 Konstrukt beobachtet werden. Das Spleißen des Exons2 führte dabei zu Änderungen der Topologie der N-terminalen C2-Domäne, wodurch sich eine veränderte Lokalisation in der Zelle beschreiben ließ. Mit der veränderten intrazellulären Verteilung erfuhr auch die Funktion der dE2Smurf2 Ubiquitin Ligase eine Änderung. So konnte überraschenderweise eine positive Signalinduktion des TGF-beta Signalweges beobachtet werden, was im Gegensatz zum beschriebenen WTSmurf2 stand. Durch eine Überexpression des dE2Smurf2 Proteins in T-Lymphozyten wurde der TGF-beta Signalweg in CD4+ und CD8+ Zellen positiv reguliert, dabei wurde der TGFbetaRII vermehrt exprimiert und gleichzeitig fand eine verstärkte Phosphorylierung der Transkriptionsfaktoren Smad2 und Smad3 nach TGF-beta Stimulation statt. Die transgenen T-Lymphozyten waren somit sensitiver gegenüber TGF-beta. Dies führte zur Hypothese, die durch Western Blot Analyse bestätigt werden konnte, daß das dE2Smurf2 nach Überexpression seine WT-Form bindet und dadurch degradiert. Die Degradation der Ubiquitin Ligase war dabei Smad7 abhängig. Zur Analyse des Einflusses der Ubiquitin Ligase dE2Smurf2 auf die Differenzierung von CD4+ T-Zellen, sowie ihre Rolle bei der T-Zell Proliferation, konnte gezeigt werden, daß durch die höhere Sensitivität gegenüber TGF-beta naive T-Zellen unter Einfluß von TGF-beta und IL6 vermehrt in TH17 Zellen differenzierten. Zudem konnte gezeigt werden, daß die Proliferationsrate transgener naiver CD4+ T-Zellen bei geringen Mengen von TGF-beta starkt vermindert war. Weiterhin konnte gezeigt werden, daß bei einer Differenzierung der naiven CD4+ T-Zellen in TH1 Zellen, diese signifkant weniger das proinflammatorische Zytokin INFγ produzierten.So zeigten in vivo Versuche, daß die transgenen Tiere in der Entwicklung von Kolorektalen Karzinomen protektiert waren. Sowohl im kolitisassiziierten Tumor Modell als auch bei der spontanen Entwicklung von Tumoren im APCmin Modell. Dies konnte zum einen auf eine deutlich verminderte Entzündung (geringere Produktion an Zytokinen durch verminderte Proliferation) des Darms und zum anderen durch eine stärkere Produktion an zytotoxischen Genen, wie Perforin, INFγ und Granzym B erklärt werden. Interessanterweise konnte jedoch im Transfer Kolitis Modell eher eine proinflammatorische Wirkung des dE2Smurf2 Proteins nachgewiesen werden. So wiesen die immundefizienten Mäuse, in denen die transgenen T-Zellen injiziert wurden, eine signifikant stärkere Kolitis auf als die Kontrollen. Dies konnte mit einer Überproduktion an IL17 sezernierenden T-Zellen erklärt werden. Klonierungsexperimente führten zudem zur Identifikation einer bisher nicht beschriebenen nicht kodierenden RNA. Diese zeigte in Kombination mit dem dE2Smurf2 Protein in einer Reportergen Analyse eine Hyperaktivierung des Smad3 Promotors. Diese Daten liefern zum einen ein genaueres Modell über die Regulation des TGF-beta Signalweges sowie wichtige Erkenntnisse zur Pathophysiologie chronisch entzündlicher Darmerkrankung und daraus resultierende Tumorerkrankungen. So entwickelt sich das dE2Smurf2, Teil des TGF-beta Signalweges, als attraktives Zielprotein für die Modulation von chronisch entzündlichen Darmerkrankungen und (kolitisassoziierte) Kolonkarzinomen.

This paper examines the accuracy of software-based on-line energy estimation techniques. It evaluates today’s most widespread energy estimation model in order to investigate whether the current methodology of pure software-based energy estimation running on a sensor node itself can indeed reliably and accurately determine its energy consumption - independent of the particular node instance, the traffic load the node is exposed to, or the MAC protocol the node is running. The paper enhances today’s widely used energy estimation model by integrating radio transceiver switches into the model, and proposes a methodology to find the optimal estimation model parameters. It proves by statistical validation with experimental data that the proposed model enhancement and parameter calibration methodology significantly increases the estimation accuracy.

QUESTIONS UNDER STUDY: This study investigated the use among the Swiss adult population and regional dissemination of various methods of complementary medicine (CM) provided by physicians or therapists in Switzerland. METHODS: Data of the Swiss Health Survey 2007 were used, which comprised a telephone interview followed by a written questionnaire (18,760 and 14,432 respondents, respectively) and included questions about people's state of health, health insurance and usage of health services. Users and non-users of CM were compared using logistic regression models. RESULTS: The most popular CM methods were homeopathy, osteopathy, acupuncture and shiatsu/foot reflexology. 30.5% of women and 15.2% of men used at least one CM method in the 12 months preceding the survey. Lake Geneva region and central Switzerland had more CM users than the other regions. Women, people between 25 and 64 years of age and people with higher levels of education were more likely to use CM. 53.5% of the adult population had a supplemental health insurance for CM treatments. 32.9% of people with such an insurance used CM during the 12 months preceding the survey, and so did 12.0% of people without additional insurance. CONCLUSIONS: Almost one fourth of the Swiss adult population had used CM within the past 12 months. User profiles were comparable to those in other countries. Despite a generally lower self-perceived health status, elderly people were less likely to use CM.
