Abstract Background Chronic Obstructive Pulmonary Disease (COPD) is associated with bronchial epithelial changes, including squamous cell metaplasia and goblet cell hyperplasia. These features are partially attributed to activation of the epidermal growth factor receptor (EGFR). Whereas smoking cessation reduces respiratory symptoms and lung function decline in COPD, inflammation persists. We determined epithelial proliferation and composition in bronchial biopsies from current and ex-smokers with COPD, and its relation to duration of smoking cessation. Methods 114 COPD patients were studied cross-sectionally: 99 males/15 females, age 62 ± 8 years, median 42 pack-years, no corticosteroids, current (n = 72) or ex-smokers (n = 42, median cessation duration 3.5 years), postbronchodilator FEV1 63 ± 9% predicted. Squamous cell metaplasia (%), goblet cell (PAS/Alcian Blue+) area (%), proliferating (Ki-67+) cell numbers (/mm basement membrane), and EGFR expression (%) were measured in intact epithelium of bronchial biopsies. Results Ex-smokers with COPD had significantly less epithelial squamous cell metaplasia, proliferating cell numbers, and a trend towards reduced goblet cell area than current smokers with COPD (p = 0.025, p = 0.001, p = 0.081, respectively), but no significant difference in EGFR expression. Epithelial features were not different between short-term quitters (<3.5 years) and current smokers. Long-term quitters (≥3.5 years) had less goblet cell area than both current smokers and short-term quitters (medians: 7.9% vs. 14.4%, p = 0.005; 7.9% vs. 13.5%, p = 0.008; respectively), and less proliferating cell numbers than current smokers (2.8% vs. 18.6%, p < 0.001). Conclusion Ex-smokers with COPD had less bronchial epithelial remodelling than current smokers, which was only observed after long-term smoking cessation (>3.5 years). Trial registration NCT00158847

Abstract Background The CACTA (also called En/Spm) superfamily of DNA-only transposons contain the core sequence CACTA in their Terminal Inverted Repeats (TIRs) and so far have only been described in plants. Large transcriptome and genome sequence data have recently become publicly available for Schistosoma mansoni, a digenetic blood fluke that is a major causative agent of schistosomiasis in humans, and have provided a comprehensive repository for the discovery of novel genes and repetitive elements. Despite the extensive description of retroelements in S. mansoni, just a single DNA-only transposon belonging to the Merlin family has so far been reported in this organism. Results We describe a novel S. mansoni transposon named SmTRC1, for S. mansoni Transposon Related to CACTA 1, an element that shares several characteristics with plant CACTA transposons. Southern blotting indicates approximately 30–300 copies of SmTRC1 in the S. mansoni genome. Using genomic PCR followed by cloning and sequencing, we amplified and characterized a full-length and a truncated copy of this element. RT-PCR using S. mansoni mRNA followed by cloning and sequencing revealed several alternatively spliced transcripts of this transposon, resulting in distinct ORFs coding for different proteins. Interestingly, a survey of complete genomes from animals and fungi revealed several other novel TRC elements, indicating new families of DNA transposons belonging to the CACTA superfamily that have not previously been reported in these kingdoms. The first three bases in the S. mansoni TIR are CCC and they are identical to those in the TIRs of the insects Aedes aegypti and Tribolium castaneum, suggesting that animal TRCs may display a CCC core sequence. Conclusion The DNA-only transposable element SmTRC1 from S. mansoni exhibits various characteristics, such as generation of multiple alternatively-spliced transcripts, the presence of terminal inverted repeats at the extremities of the elements flanked by direct repeats and the presence of a Transposase_21 domain, that suggest a distant relationship to CACTA transposons from Magnoliophyta. Several sequences from other Metazoa and Fungi code for proteins similar to those encoded by SmTRC1, suggesting that such elements have a common ancestry, and indicating inheritance through vertical transmission before separation of the Eumetazoa, Fungi and Plants.

Neste trabalho, discute-se a fixação de taxas de retorno de concessões no Brasil, com aplicação específica ao caso da metodologia da Agência Nacional de Transportes Terrestres (ANTT). Mostra-se a inadequação da regulamentação vigente, baseada no conceito de taxa interna de retorno (TIR), e não de custo de oportunidade do capital. A partir de um exemplo com dados referentes ao auge da crise financeira internacional (dezembro de 2008), evidencia-se também a falta de lógica decorrente da utilização de retornos e preços passados na estimação de taxas de retorno, um procedimento comum a toda a área de concessões de serviços públicos no Brasil. Propõe-se uma metodologia alternativa cujos resultados são sensíveis às condições correntes de mercado de capitais, que produz resultados coerentes com a situação então vigente.

L’alta risoluzione nel telerilevamento termico (Thermal Remote Sensing) da aereo o satellitare si rivela molto importante nell’analisi del comportamento termico delle superfici, in particolare per lo studio dei fenomeni climatici locali dello spazio urbano. La stato termico dell'ambiente urbano è oggi motivo di grande interesse per ricercatori, organi istituzionali e cittadini. Uno dei maggiori campi di studio del comportamento termico urbano interessa il problema energetico: la riduzione dei consumi e delle emissioni di CO2 è un obiettivo primario da perseguire per uno sviluppo sostenibile, spesso supportato da criteri legislativi e progetti comunitari. Su scala differente e con caratteristiche differenti, un altro degli argomenti che scuote da anni e con notevole interesse la ricerca scientifica, è il fenomeno termico urbano che prende il nome di isola di calore; questa si sviluppa non solo in conseguenza al calore sensibile rilasciato da attività antropiche, ma anche a causa della sempre maggiore conversione del territorio rurale in urbanizzato (inurbamento), con conseguente riduzione del fenomeno dell’evapotraspirazione. Oggetto di questa dissertazione è lo studio del comportamento termico delle superfici in ambito urbano, sperimentato sulla città di Bologna. Il primo capitolo si interessa dei principi e delle leggi fisiche sui quali è basato il telerilevamento effettuato nelle bende spettrali dell’infrarosso termico. Viene data una definizione di temperatura radiometrica e cinematica, tra loro legate dall’emissività. Vengono esposti i concetti di risoluzione (geometrica, radiometrica, temporale e spettrale) dell’immagine termica e viene data descrizione dei principali sensori su piattaforma spaziale per l’alta risoluzione nel TIR (ASTER e Landsat). Il secondo capitolo si apre con la definizione di LST (Land Surface Temperature), parametro del terreno misurato col telerilevamento, e ne viene descritta la dipendenza dal flusso della radiazione in atmosfera e dalle condizioni di bilancio termico della superficie investigata. Per la sua determinazione vengono proposti metodi diversi in funzione del numero di osservazioni disponibili nelle diverse bande spettrali dell’IR termico. In chiusura sono discussi i parametri che ne caratterizzano la variabilità. Il capitolo terzo entra nel dettaglio del telerilevamento termico in ambito urbano, definendo il fenomeno dell’Urban Heat Island su tutti i livelli atmosferici interessati, fornendo un quadro di operabilità con gli strumenti moderni di rilievo alle differenti scale (analisi multiscala). Un esempio concreto di studio multiscala dei fenomeni termici urbani è il progetto europeo EnergyCity, volto a ridurre i consumi energetici e le emissioni di gas serra di alcune città del centro Europa. Il capitolo quarto riporta la sperimentazione condotta sull’isola di calore urbana della città di Bologna tramite immagini ASTER con risoluzione spaziale 90 m nel TIR e ricampionate a 15 m dal VIS. Lo studio dell’isola di calore si è effettuata a partire dal calcolo della Land Surface Temperature utilizzando valori di emissività derivati da classificazione delle superfici al suolo. Per la validazione dei dati, in alternativa alle stazioni di monitoraggio fisse dell’ARPA, presenti nell’area metropolitana della città, si è sperimentato l’utilizzo di data-loggers per il rilievo di temperatura con possibilità di campionamento a 2 sec. installati su veicoli mobili, strumentati con ricevitori GPS, per la misura dei profili di temperatura atmosferica near-ground lungo transetti di attraversamento della città in direzione est-ovest.

L'obiettivo dell'elaborato è definire gli importi delle voci fondamentali in ingresso al metodo di calcolo tramite un'intensa ricerca bibliografica in materia e realizzare un procedimento pratico di riferimento applicato al campo delle infrastrutture stradali. I risultati ottenuti dal calcolo sono stati verificati e confrontati con test di sensitività sulle variabili critiche (Analisi di Sensitività, Analisi di Switch e Analisi di Rischio).

Il presente studio si concentra sulle diverse applicazioni del telerilevamento termico in ambito urbano. Vengono inizialmente descritti la radiazione infrarossa e le sue interazioni con l’atmosfera terrestre, le leggi principali che regolano lo scambio di calore per irraggiamento, le caratteristiche dei sensori e le diverse applicazioni di termografia. Successivamente sono trattati nel dettaglio gli aspetti caratteristici della termografia da piattaforma satellitare, finalizzata principalmente alla valutazione del fenomeno dell'Urban Heat Island; vengono descritti i sensori disponibili, le metodologie di correzione per gli effetti atmosferici, per la stima dell'emissività delle superfici e per il calcolo della temperatura superficiale dei pixels. Viene quindi illustrata la sperimentazione effettuata sull'area di Bologna mediante immagini multispettrali ASTER: i risultati mostrano come sull'area urbana sia riscontrabile la presenza dell'Isola di Calore Urbano, anche se la sua quantificazione risulta complessa. Si procede quindi alla descrizione di potenzialità e limiti della termografia aerea, dei suoi diversi utilizzi, delle modalità operative di rilievo e degli algoritmi utilizzati per il calcolo della temperatura superficiale delle coperture edilizie. Tramite l’analisi di alcune esperienze precedenti vengono trattati l’influenza dell’atmosfera, la modellazione dei suoi effetti sulla radianza rilevata, i diversi metodi per la stima dell’emissività. Viene quindi introdotto il progetto europeo Energycity, finalizzato alla creazione di un sistema GeoWeb di supporto spaziale alle decisioni per la riduzione di consumi energetici e produzione di gas serra su sette città dell'Europa Centrale. Vengono illustrate le modalità di rilievo e le attività di processing dei datasets digitali per la creazione di mappe di temperatura superficiale da implementare nel sistema SDSS. Viene infine descritta la sperimentazione effettuata sulle immagini termiche acquisite nel febbraio 2010 sulla città di Treviso, trasformate in un mosaico georiferito di temperatura radiometrica tramite correzioni geometriche e radiometriche; a seguito della correzione per l’emissività quest’ultimo verrà trasformato in un mosaico di temperatura superficiale.

Survivin, a unique member of the family of inhibitors of apoptosis (IAP) proteins, orchestrates intracellular pathways during cell division and apoptosis. Its central regulatory function in vertebrate molecular pathways as mitotic regulator and inhibitor of apoptotic cell death has major implications for tumor cell proliferation and viability, and has inspired several approaches that target survivin for cancer therapy. Analyses in early-branching Metazoa so far propose an exclusive role of survivin as a chromosomal passenger protein, whereas only later during evolution the second, complementary antiapoptotic function might have arisen, concurrent with increased organismal complexity. To lift the veil on the ancestral function(s) of this key regulatory molecule, a survivin homologue of the phylogenetically oldest extant metazoan taxon (phylum Porifera) was identified and functionally characterized. SURVL of the demosponge Suberites domuncula shares significant similarities with its metazoan homologues, ranging from conserved exon/intron structures to the presence of localization signal and protein-interaction domains, characteristic of IAP proteins. Whereas sponge tissue displayed a very low steady-state level, SURVL expression was significantly up-regulated in rapidly proliferating primmorph cells. In addition, challenge of sponge tissue and primmorphs with cadmium and the lipopeptide Pam3Cys-Ser-(Lys)4 stimulated SURVL expression, concurrent with the expression of newly discovered poriferan caspases (CASL and CASL2). Complementary functional analyses in transfected HEK-293 revealed that heterologous expression of poriferan survivin in human cells not only promotes cell proliferation but also augments resistance to cadmium-induced cell death. Taken together, these results demonstrate both a deep evolutionary conserved and fundamental dual role of survivin, and an equally conserved central position of this key regulatory molecule in interconnected pathways of cell cycle and apoptosis. Additionally, SDCASL, SDCASL2, and SDTILRc (TIR-LRR containing protein) may represent new components of the innate defense sentinel in sponges. SDCASL and SDCASL2 are two new caspase-homolog proteins with a singular structure. In addition to their CASc domains, SDCASL and SDCASL2 feature a small prodomain NH2-terminal (effector caspases) and a remarkably long COOH-terminal domain containing one or several functional double stranded RNA binding domains (dsrm). This new caspase prototype can characterize a caspase specialization coupling pathogen sensing and apoptosis, and could represent a very efficient defense mechanism. SDTILRc encompasses also a unique combination of domains: several leucine rich repeats (LRR) and a Toll/IL-1 receptor (TIR) domain. This unusual domain association may correspond to a new family of intracellular sensing protein, forming a subclass of pattern recognition receptors (PRR).

A unique characteristic of soft matter is its ability to self-assemble into larger structures. Characterizing these structures is crucial for their applications. In the first part of this work, I investigated DNA-organic hybrid material by means of Fluorescence Correlation Spectroscopy (FCS) and Fluorescence Cross-Correlation Spectroscopy (FCCS). DNA-organic hybrid materials, a novel class of hybrid materials composed of synthetic macromolecules and oligodeoxynucleotide segmenta, are mostly amphiphilic and can self-assemble into supramolecular structures in aqueous solution. A hybrid material of a fluorophore, perylenediimide (PDI), and a DNA segment (DNA-PDI) has been developed in Prof. A. Hermann’s group (University of Groningen). This novel material has the ability to form aggregates through pi-pi stacking between planar PDIs and can be traced in solution due to the fluorescence of PDI. I have determined the diffusion coefficient of DNA-PDI conjugates in aqueous solution by means of FCS. In addition, I investigated whether such DNA-PDIs form aggregates with certain structure, for instance dimers. rnOnce the DNA hybrid material self-assemble into supermolecular structures for instance into micelles, the single molecules do not necessarily stay in one specific micelle. Actually, a single molecule may enter and leave micelles constantly. The average residence time of a single molecule in a certain micelle depends on the nature of the molecule. I have chosen DNA-b-polypropylene oxide (PPO) as model molecules and investigated the residence time of DNA-b-PPO molecules in their according micelles by means of FCCS.rnBesides the DNA hybrid materials, polymeric colloids can also form ordered structures once they are brought to an air/water interface. Here, hexagonally densely packed monolayers can be generated. These monolayers can be deposited onto different surfaces as coating layers. In the second part of this work, I investigated the mechanical properties of such colloidal monolayers using micromechanical cantilevers. When a coating layer is deposited on a cantilever, it can modify the elasticity of the cantilever. This variation can be reflected either by a deflection or by a resonance frequency shift of the cantilever. In turn, detecting these changes provides information about the mechanical properties of the coating layer. rnIn the second part of this work, polymeric colloidal monolayers were coated on a cantilever and homogenous polymer films of a few hundred nanometers in thickness were generated from these colloidal monolayers by thermal annealing or organic vapor annealing. Both the film formation process and the mechanical properties of these resulting homogenous films were investigated by means of cantilever. rnElastic property changes of the coating film, for example upon absorption of organic vapors, induce a deflection of the cantilever. This effect enables a cantilever to detect target molecules, when the cantilever is coated with an active layer with specific affinity to target molecules. In the last part of this thesis, I investigated the applicability of suitably functionalized micromechanical cantilevers as sensors. In particular, glucose sensitive polymer brushes were grafted on a cantilever and the deflection of this cantilever was measured during exposure to glucose solution. rn

In this thesis, different complex colloids were prepared by the process of solvent evaporation from emulsion droplets (SEED). The term “complex” is used to include both an addressable functionality as well as the heterogeneous nature of the colloids.Firstly, as the SEED process was used throughout the thesis, its mechanism especially in regard to coalescence was investigated,. A wide variety of different techniques was employed to study the coalescence of nanodroplets during the evaporation of the solvent. Techniques such as DLS or FCS turned out not to be suitable methods to determine droplet coalescence because of their dependence on dilution. Thus, other methods were developed. TEM measurements were conducted on mixed polymeric emulsions with the results pointing to an absence of coalescence. However, these results were not quantifiable. FRET measurements on mixed polymeric emulsions also indicated an absence of coalescence. Again the results were not quantifiable. The amount of coalescence taking place was then quantified by the application of DC-FCCS. This method also allowed for measuring coalescence in other processes such as the miniemulsion polymerization or the polycondensation reaction on the interface of the droplets. By simulations it was shown that coalescence is not responsible for the usually observed broad size distribution of the produced particles. Therefore, the process itself, especially the emulsification step, needs to be improved to generate monodisperse colloids.rnThe Janus morphology is probably the best known among the different complex morphologies of nanoparticles. With the help of functional polymers, it was possible to marry click-chemistry to Janus particles. A large library of functional polymers was prepared by copolymerization and subsequent post-functionalization or by ATRP. The polymers were then used to generate Janus particles by the SEED process. Both dually functionalized Janus particles and particles with one functionalized face could be obtained. The latter were used for the quantification of functional groups on the surface of the Janus particles. For this, clickable fluorescent dyes were synthesized. The degree of functionality of the polymers was found to be closely mirrored in the degree of functionality of the surface. Thus, the marriage of click-chemistry to Janus particles was successful.Another complex morphology besides Janus particles are nanocapsules. Stimulus-responsive nanocapsules that show triggered release are a highly demanding and interesting system, as nanocapsules have promising applications in drug delivery and in self-healing materials. To achieve heterogeneity in the polymer shell, the stimulus-responsive block copolymer PVFc-b-PMMA was employed for the preparation of the capsules. The phase separation of the two blocks in the shell of the capsules led to a patchy morphology. These patches could then be oxidized resulting in morphology changes. In addition, swelling occurred because of the hydrophobic to hydrophilic transition of the patches induced by the oxidation. Due to the swelling, an encapsulated payload could diffuse out of the capsules, hence release was achieved.The concept of using block copolymers responsive to one stimulus for the preparation of stimulus-responsive capsules was extended to block copolymers responsive to more than one stimulus. Here, a block copolymer responsive to oxidation and a pH change as well as a block copolymer responsive to a pH change and temperature were studied in detail. The release from the nanocapsules could be regulated by tuning the different stimuli. In addition, by encapsulating stimuli-responsive payloads it was possible to selectively release a payload upon one stimulus but not upon the other one.In conclusion, the approaches taken in the course of this thesis demonstrate the broad applicability and usefulness of the SEED process to generate complex colloids. In addition, the experimental techniques established such as DC-FCCS will provide further insight into other research areas as well.

Polymerbasierte Kolloide mit Groen im Nanometerbereich werden als aussichts- reiche Kandidaten fur die Verkapselung und den Transport von pharmazeutischen Wirkstoen angesehen. Daher ist es wichtig die physikalischen Prozesse, die die Bil- dung, Struktur und kinetische Stabilitat der polymerbasierten Kolloide beein ussen, besser zu verstehen. Allerdings ist die Untersuchung dieser Prozesse fur nanome- tergroe Objekte kompliziert und erfordert fortgeschrittene Techniken. In dieser Arbeit beschreibe ich Untersuchungen, bei denen Zwei-Farben-Fluoreszenzkreuz- korrelationsspektroskopie (DC FCCS) genutzt wurde, um Informationen uber die Wechselwirkung und den Austausch von dispergierten, nanometergroen Kolloiden zu bekommen. Zunachst habe ich den Prozess der Polymernanopartikelherstellung aus Emul- sionstropfen untersucht, welcher einen der am haugsten angewendeten Prozesse der Nanopartikelformulierung darstellt. Ich konnte zeigen, dass mit DC FCCS eindeutig und direkt Koaleszenz zwischen Emulsionstropfen gemessen werden kann. Dies ist von Interesse, da Koaleszenz als Hauptgrund fur die breite Groenverteilung der nalen Nanopartikel angesehen wird. Weiterhin habe ich den Austausch von Mizellen bildenden Molekulen zwischen amphiphilen Diblock Kopolymermizellen untersucht. Als Modellsystem diente ein Linear-Burste Block Kopolymer, welches Mizellen mit einer dichten und kurzen Korona bildet. Mit Hilfe von DC FCCS konnte der Austausch in verschiedenen Losungsmitteln und bei verschiedenen Temperaturen beobachtet werden. Ich habe herausgefunden, dass in Abhangigkeit der Qualitat des Losungsmittels die Zeit des Austausches um Groenordnungen verschoben werden kann, was eine weitreichende Einstellung der Austauschkinetik ermoglicht. Eine Eigenschaft die all diese Kolloide gemeinsam haben ist ihre Polydispersitat. Im letzten Teil meiner Arbeit habe ich am Beispiel von Polymeren als Modellsystem untersucht, welchen Eekt Polydispersitat und die Art der Fluoreszenzmarkierung auf FCS Experimente haben. Eine Anpassung des klassischen FCS Modells kann die FCS Korrelationskurven dieser Systeme beschreiben. Die Richtigkeit meines Ansatzes habe ich mit dem Vergleich zur Gel-Permeations-Chromatographie und Brownschen Molekulardynamiksimulationen bestatigt.

Nella regione del TIR, le transizioni spettrali vibro-rotazionali della CO2 sono sfruttate per ricavare la distribuzione di P e T negli esperimenti spaziali. Oltre all’importanza di questi due parametri, la loro conoscenza è necessaria per ricavare la distribuzione di qualsiasi molecola dalle sue transizioni spettrali. Per ricavare P e T si assume di conoscere il VMR della CO2. L’accuratezza con cui si ricava la distribuzione della CO2 insieme a quelle di P e T non è sufficiente. Inoltre, il VMR della CO2 aumenta nel corso degli anni. Per questo, in questa tesi si propone una nuova strategia per misurare la CO2 usando uno strumento satellitare a scansione del lembo. L’idea è quella di sfruttare le transizioni rotazionali pure dell’O2 nella regione del FIR per ricavare P e T. Poiché queste transizioni traggono origine da un momento di dipolo magnetico la loro forza di riga è molto bassa. Tuttavia, grazie alla grande abbondanza dell’O2 in atmosfera e alla lunghezza dei cammini ottici, queste transizioni sono tra le più intense nello spettro atmosferico del FIR. Il satellite considerato è posto su un’orbita quasi polare e lo strumento osserva l’emissione del lembo atmosferico in direzione opposta a quella di volo. Lo strumento ipotizzato è uno spettrometro a trasformata di Fourier con due porte di output ospitanti un detector per la regione del FIR e uno per quella del TIR. La risoluzione spettrale è di 0.004 cm-1. Mentre il NESR è di 5 nW. Il campionamento geometrico verticale dell’atmosfera è di 3 Km mentre quello orizzontale è di circa 100 Km. Usando la teoria dell’optimal estimation sono stati selezionati due set di intervalli spettrali da analizzare, uno per la regione del FIR e l’altro per la regione del TIR. Con queste ipotesi sono stati effettuati test di retrieval su osservati simulati per valutare le performance del sistema ipotizzato. Si è dimostrato che le transizioni della CO2 nella regione del TIR non sono sufficienti per ricavare P e T insieme al proprio VMR con precisione soddisfacente e che l’uso dell’informazione derivante dal FIR fa aumentare la qualità del retrieval. Le performance dell’esperimento permettono di ricavare il VMR della CO2 con una precisione di circa 1 ppm tra 10 Km e 60 Km con una risoluzione verticale di 3 Km e una orizzontale di circa 2.5° di latitudine. Si è quindi dimostrato la validità della strategia proposta in questo studio.

The Advanced Spaceborne Thermal Emission and Reflection Radiometer (ASTER) has been used to quantify SO2 emissions from passively degassing volcanoes. This dissertation explores ASTER’s capability to detect SO2 with satellite validation, enhancement techniques and extensive processing of images at a variety of volcanoes. ASTER is compared to the Mini UV Spectrometer (MUSe), a ground based instrument, to determine if reasonable SO2 fluxes can be quantified from a plume emitted from Lascar, Chile. The two sensors were in good agreement with ASTER proving to be a reliable detector of SO2. ASTER illustrated the advantages of imaging a plume in 2D, with better temporal resolution than the MUSe. SO2 plumes in ASTER imagery are not always discernible in the raw TIR data. Principal Component Analysis (PCA) and Decorrelation Stretch (DCS) enhancement techniques were compared to determine how well they highlight a variety of volcanic plumes. DCS produced a consistent output and the composition of the plumes was easy to identify from explosive eruptions. As the plumes became smaller and lower in altitude they became harder to distinguish using DCS. PCA proved to be better at identifying smaller low altitude plumes. ASTER was used to investigate SO2 emissions at Lascar, Chile. Activity at Lascar has been characterized by cyclic behavior and persistent degassing (Matthews et al. 1997). Previous studies at Lascar have primarily focused on changes in thermal infrared anomalies, neglecting gas emissions. Using the SO2 data along with changes in thermal anomalies and visual observations it is evident that Lascar is at the end an eruptive cycle that began in 1993. Declining gas emissions and crater temperatures suggest that the conduit is sealing. ASTER and the Ozone Monitoring Instrument (OMI) were used to determine the annual contribution of SO2 to the troposphere from the Central and South American volcanic arcs between 2000 and 2011. Fluxes of 3.4 Tg/a for Central America and 3.7 Tg/a for South America were calculated. The detection limits of ASTER were explored. The results a proved to be interesting, with plumes from many of the high emitting volcanoes, such as Villarrica, Chile, not being detected by ASTER.

AIM To report a rare case of a spinal WHO grade I meningioma extending through intervertebral foramina C7 to D4 with an extensive mediastinal mass and infiltration of the vertebrae, and to discuss the malignant behavior of a tumor classified as benign. METHODS (Clinical Presentation, Histology, and Imaging): A 54-year-old man suffered from increasing lower back pain with gait difficulties, weakness and numbness of the lower extremities, as well as urge incontinence. CT scan of the thorax and MRI scan of the spine revealed a large prevertebral tumor, which extended to the spinal canal and caused compression of the spinal cord at the levels of C7 to D4 leading to myelopathy with hyperintense signal alteration on T2-weighted MRI images. The signal constellation (T1 with and without contrast, T2, TIR) was highly suspicious for infiltration of vertebrae C7 to D5. Somatostatin receptor SPECT/CT with (111)In-DTPA-D: -Phe-1-octreotide detected a somatostatin receptor-positive mediastinal tumor with infiltration of multiple vertebrae, dura, and intervertebral foramina C7-D4, partially with Krenning score >2. Percutaneous biopsies of the mediastinal mass led to histopathological findings of WHO grade I meningioma of meningothelial subtype. RESULTS (Therapy): C7 to D4 laminoplasty was performed, and the intraspinal, extradural part of the tumor was microsurgically removed. Postoperative stereotactic radiation therapy was done using the volumetric modulated arc therapy (VMAT) technique (RapidArc). No PRRNT with (90)Y-DOTA-TOC was done. CONCLUSIONS Due to the rare incidence and complex presentation of this disease not amenable to complete surgical resection, an individualized treatment approach should be worked out interdisciplinarily. The treatment approach should be based not only on histology but also on clinical and imaging findings. Close clinical and radiological follow-up may be mandatory even for benign tumors.

During senescence, chlorophyll (chl) is metabolized to colorless nonfluorescent chl catabolites (NCCs). A central reaction of the breakdown pathway is the ring cleavage of pheophorbide (pheide) a to a primary fluorescent chl catabolite. Two enzymes catalyze this reaction, pheide a oxygenase (PAO) and red chl catabolite reductase. Five NCCs and three fluorescent chl catabolites (FCCs) accumulated during dark-induced chl breakdown in Arabidopsis (Arabidopsis thaliana). Three of these NCCs and one FCC (primary fluorescent chl catabolite-1) were identical to known catabolites from canola (Brassica napus). The presence in Arabidopsis of two modified FCCs supports the hypothesis that modifications, as present in NCCs, occur at the level of FCC. Chl degradation in Arabidopsis correlated with the accumulation of FCCs and NCCs, as well as with an increase in PAO activity. This increase was due to an up-regulation of Pao gene expression. In contrast, red chl catabolite reductase is not regulated during leaf development and senescence. A pao1 knockout mutant was identified and analyzed. The mutant showed an age- and light-dependent cell death phenotype on leaves and in flowers caused by the accumulation of photoreactive pheide a. In the dark, pao1 exhibited a stay-green phenotype. The key role of PAO in chl breakdown is discussed.
