Here we propose a protocol for embryogenic cultures induction, proliferation and maturation for the Brazilian conifer Podocarpus lambertii, and investigated the effect of abscisic acid (ABA) and glutathione (GSH) supplementation on the maturation phase. ABA, zeatin (Z) and salicylic acid (SA) endogenous levels were quantified. Number of somatic embryos obtained in ABA-supplemented treatment was signifi- cant higher than in ABA-free treatment, showing the relevance of ABA supplementation during somatic embryos maturation. Histological analysis showed the stereotyped sequence of developmental stages in conifer somatic embryos, reaching the late torpedo-staged embryo. GSH supplementation in maturation culture medium improved the somatic embryos number and morphological features. GSH 0 mM and GSH 0.1 mM treatments correlated with a decreased ABA endogenous level during maturation, while GSH 0.5 mM treatment showed constantlevels. Alltreatments resulted in decreased Z endogenous levels, supporting the concept that cytokinins are important during the initial cell division but not for the later stages of embryo development. The lowest SA levels found in GSH 0.5 mM treatment were coincident with early embryonic development, and this treatment resulted in the highest development of somatic embryos. Thus, a correlation between lower SA levels and improved somatic embryo formation can be hypothesized

In this work we compare Grapholita molesta Busck (Lepidoptera: Tortricidae) populations originated from Brazil, Chile, Spain, Italy and Greece using power spectral density and phylogenetic analysis to detect any similarities between the population macro- and the molecular micro-level. Log-transformed population data were normalized and AR(p) models were developed to generate for each case population time series of equal lengths. The time-frequency/scale properties of the population data were further analyzed using wavelet analysis to detect any population dynamics frequency changes and cluster the populations. Based on the power spectral of each population time series and the hierarchical clustering schemes, populations originated from Southern America (Brazil and Chile) exhibit similar rhythmic properties and are both closer related with populations originated from Greece. Populations from Spain and especially Italy, have higher distance by terms of periodic changes on their population dynamics. Moreover, the members within the same cluster share similar spectral information, therefore they are supposed to participate in the same temporally regulated population process. On the contrary, the phylogenetic approach revealed a less structured pattern that bears indications of panmixia, as the two clusters contain individuals from both Europe and South America. This preliminary outcome will be further assessed by incorporating more individuals and likely employed a second molecular marker.

Internal browning is an important disorder in pear fruit which can lead to economic losses. Pears (Pyrus communis L. cv. Bartlett) were harvested at early harvest maturity of 90 N from a commercial orchard in southern Brazil. Methyl jasmonate, ethanol, and 1-methylcyclopropene vapor treatments were carried out for 24 hours in order to mitigate the internal browning disorder. Fruit were stored for up to 150 days at 0 ± 1 °C and 90 ± 5 % RH. Pears exhibited internal browning in 37 % of the control samples after 90 days of cold storage. However, no internal browning symptoms were observed in the 1-MCP treatment. The first symptoms in 1-MCP samples were noticed after 120 days of cold storage (12 %) and reached 100 % in five days at room temperature. 1-MCP-treated pears showed flesh firmness values of 82 N after 90 days of cold storage and 18.7 N when they were removed from the cold storage and kept at 20 °C. The greatest acceptance index was attributed to 1- MCP pears after 90 days at 0 ± 1 °C followed by 5 days at 20 ± 1 °C (89.35). High acceptance indexes were attributed to MeJa (77.95) and control pears (76.40) after 30 days in cold storage followed by 5 days at room temperature. 1-MCP (0.3 µL L-1 , 24 hours at 0 ± 1 °C) treatment delays ripening and mitigates the internal browning in early harvested ?Bartlett? pears, that can be stored for up to 90 days at 0 ± 1 °C.

As aplicações sucessivas de fungicidas e caldas via foliar em videiras para o controle preventivo de doenças fúngicas, aumenta o teor de cobre (Cu) e zinco (Zn) nas camadas mais superficiais do solo. O trabalho objetivou avaliar os teores de Cu e Zn no perfil de solos com textura argilosa e com histórico de aplicação de fungicidas cúpricos na região Meio Oeste de SC. Em outubro de 2010, foram selecionados três vinhedos com idades de 4, 6 e 10 anos de idade, no município de Água Doce (SC). Em novembro de 2010 foram coletadas amostras de solo nas camadas de 0-5, 5-10, 10-15, 15-20 e 20-40 cm. Também foi coletado solo em uma área de mata nativa, adjacente aos vinhedos. Os solos foram secos, moídos, passados em peneira e submetidos as análises de Cu e Zn extraídos pelos métodos de EDTA (Na2-EDTA 0,01 mol L-1+acetato de amônio 1,0 mol L-1) com pH ajustado para 7,0. Os teores de Cu e Zn extraídos por HCl 0,1 mol L-1 e EDTA foram maiores nas camadas superficiais do solo, especialmente, no vinhedo mais antigo, onde o Cu migrou até 20 cm e o Zn até 15 cm.

Áreas de produção agropecuária podem ser mapeadas com o uso de receptores GPS, pela medida da posição geográfica de pontos. A coleta de pontos a campo geralmente não segue uma ordem que permita o traçado direto das áreas. O trabalho descreve um algoritmo desenvolvido para ordenar os pontos medidos, usando apenas a identificação da área e setor a qual cada ponto pertence, permitindo criar um mapa das áreas de produção e suas subdivisões (setores de produção contíguos). O algoritmo foi implementado na forma de um programa em linguagem C, sendo testado e validado em mais de 600 propriedades vitícolas.
