As a consequence of climate change there is now a more frequent occurrence of extreme rainfall events where, with higher rates of urbanisation, the built environment has become increasingly affected by flooding.. This is of particular importance in relation to the stability of bridge structures that span rivers and canals etc. In November 2009, the UK and Ireland were subjected to extraordinarily severe weather conditions for several days. The rainfall was logged as the highest level of rainfall ever recorded within the UK, and as a direct consequence, unprecedented flooding occurred in Cumbria. This flooding led to the collapse of three road bridges which were generally 19th century masonry arch bridges, with relatively shallow foundations. In the UK, knowledge of the combined effect of bridge scouring and inundation has been not been particularly widely studied. Research carried out by Hamill et al [1] considered the hydraulic analysis of single arch bridges under flood conditions, but no consideration was given towards the likely damage to these structures due to scouring. Prior to this, Bierry and Delleur [2] produced a classic paper in predicting the discharge downstream of an inundated arch, focussing on predicting afflux as opposed to bridge scour. Further work on backwater effects was carried out by Martin-Vide & Prio [3] in semi-circular arch bridges. Both pressurized and free-surface flows at the bridge were investigated. Flows on a mobile bed in clear-water conditions were compared to those with a rigid bed, but no predictive equation for scour under pressurised conditions was considered. This paper will present initial findings from an experimental investigation into the effects of surcharged flow and subsequent scour within the vicinity of single span arch bridges. Velocities profiles will be shown within the vicinity of the arch, in addition to the depth of clear water scour, for a series of flows and model spans. The data will be presented, where results will be correlated to the most recent predictive equations that are proposed.

Taste and odour compounds, especially geosmin (GSM) and 2-methylisoborneol (2-MIB), cause major problems in both drinking water and aquaculture industries world-wide. Aquaculture in particular has experienced significant financial losses due to the accumulation of taint compounds prior to harvest resulting in consumer rejection. UV-TiO2 photocatalysis has been demonstrated to remove GSM and 2-MIB at laboratory scale but the development of a continuous flow reactor suitable for use in water treatment has not been investigated. In this study, a pilot packed bed photocatalytic reactor was developed and evaluated for water treatment with both laboratory and naturally tainted samples. A significant reduction of both 2-MIB and GSM was achieved in both trials using the packed bed reactor unit. With the laboratory spiked water (100ngL-1 of each compound added prior to treatment), detectable levels were reduced by up to 97% after a single pass through the unit. When the reactor was used to treat water in a fish farm where both compounds were being produced in situ (2-MIB: 19ngL-1 and GSM: 14ngL-1) a reduction of almost 90% in taint compounds was achieved. These very encouraging promising results demonstrate the potential of this UV-TiO2 photocatalytic reactor for water treatment in fish rearing systems and other applications.

With the development and deployment of IEC 61850 based smart substations, cybersecurity vulnerabilities of supervisory control and data acquisition (SCADA) systems are increasingly emerging. In response to the emergence of cybersecurity vulnerabilities in smart substations, a test-bed is indispensable to enable cybersecurity experimentation. In this paper, a comprehensive and realistic cyber-physical test-bed has been built to investigate potential cybersecurity vulnerabilities and the impact of cyber-attacks on IEC 61850 based smart substations. This test-bed is close to a real production type environment, and has the ability to carry out end-to-end testing of cyber-attacks and physical consequences. A fuzz testing approach is proposed for detecting IEC 61850 based intelligent electronic devices (IEDs) and validated in the proposed test-bed.

In Europe, maximum journey time for transported sheep is set at 29. h (EC Regulation 1/2005), after which animals must be unloaded, fed and watered in control posts stopping for 24. h, as all other species, before continuing their journey. The industry considers these resting times too general, not taking into account the peculiar differences between species or age classes. Also, loading and unloading have been reported to be detrimental for the animals. Therefore, the industry pushes to reduce the times at control post and avoid unloading the animals from the truck. Since there is little information concerning the effect of resting in a stationary truck after long journeys, the present study aims to evaluate the effect of an 8. h rest stop on the truck for transported ewes compared to being unloaded for resting in a control post for the same amount of time, considering physiological and behavioural measures. Two groups of ewes were transported for 29. h, after which one was unloaded and housed in a pen (P) at the control post while the other was left inside the truck (T). After 8. h stop, a further 6. h travel was headed to the farm of origin. A third group (C) stayed at the farm as control. During the stop, standing, resting, moving and eating behaviour of all groups was recorded. Blood parameters, salivary and faecal cortisol were assessed at different stages. The behaviour of P animals during the resting period was more similar to C than to T ones, where feeding and lying behaviours were restricted by the limited space allowance on the truck. After returning to the farm of origin, both T and P animals showed different parameters' levels as compared to C. P ewes showed a mean loss weight of 2. kg not recorded in group T and showed higher signs of muscular damage compared to C group. It was concluded that, with so short resting times as 8. h, there is no clear advantages in terms of animal welfare for avoiding the unloading and loading of the animals in the control post after long journeys.

Dado o aumento acelerado dos preços dos combustíveis fósseis e as incertezas quanto à sua disponibilidade futura, tem surgido um novo interesse nas tecnologias da biomassa aplicadas à produção de calor, eletricidade ou combustíveis sintéticos. Não obstante, para a conversão termoquímica de uma partícula de biomassa sólida concorrem fenómenos bastante complexos que levam, em primeiro lugar, à secagem do combustível, depois à pirólise e finalmente à combustão ou gasificação propriamente ditas. Uma descrição relativamente incompleta de alguns desses estágios de conversão constitui ainda um obstáculo ao desenvolvimento das tecnologias que importa ultrapassar. Em particular, a presença de elevados conteúdos de matéria volátil na biomassa põe em evidência o interesse prático do estudo da pirólise. A importância da pirólise durante a combustão de biomassa foi evidenciada neste trabalho através de ensaios realizados num reator piloto de leito fluidizado borbulhante. Verificou-se que o processo ocorre em grande parte à superfície do leito com chamas de difusão devido à libertação de voláteis, o que dificulta o controlo da temperatura do reator acima do leito. No caso da gasificação de biomassa a pirólise pode inclusivamente determinar a eficiência química do processo. Isso foi mostrado neste trabalho durante ensaios de gasificação num reator de leito fluidizado de 2MWth, onde um novo método de medição permitiu fechar o balanço de massa ao gasificador e monitorizar o grau de conversão da biomassa. A partir destes resultados tornou-se clara a necessidade de descrever adequadamente a pirólise de biomassa com vista ao projeto e controlo dos processos. Em aplicações de engenharia há particular interesse na estequiometria e propriedades dos principais produtos pirolíticos. Neste trabalho procurou-se responder a esta necessidade, inicialmente através da estruturação de dados bibliográficos sobre rendimentos de carbonizado, líquidos pirolíticos e gases, assim como composições elementares e poderes caloríficos. O resultado traduziu-se num conjunto de parâmetros empíricos de interesse prático que permitiram elucidar o comportamento geral da pirólise de biomassa numa gama ampla de condições operatórias. Para além disso, propôs-se um modelo empírico para a composição dos voláteis que pode ser integrado em modelos compreensivos de reatores desde que os parâmetros usados sejam adequados ao combustível ensaiado. Esta abordagem despoletou um conjunto de ensaios de pirólise com várias biomassas, lenhina e celulose, e temperaturas entre os 600 e 975ºC. Elevadas taxas de aquecimento do combustível foram alcançadas em reatores laboratoriais de leito fluidizado borbulhante e leito fixo, ao passo que um sistema termo-gravimétrico permitiu estudar o efeito de taxas de aquecimento mais baixas. Os resultados mostram que, em condições típicas de processos de combustão e gasificação, a quantidade de voláteis libertada da biomassa é pouco influenciada pela temperatura do reator mas varia bastante entre combustíveis. Uma análise mais aprofundada deste assunto permitiu mostrar que o rendimento de carbonizado está intimamente relacionado com o rácio O/C do combustível original, sendo proposto um modelo simples para descrever esta relação. Embora a quantidade total de voláteis libertada seja estabelecida pela composição da biomassa, a respetiva composição química depende bastante da temperatura do reator. Rendimentos de espécies condensáveis (água e espécies orgânicas), CO2 e hidrocarbonetos leves descrevem um máximo relativamente à temperatura para dar lugar a CO e H2 às temperaturas mais altas. Não obstante, em certas gamas de temperatura, os rendimentos de algumas das principais espécies gasosas (e.g. CO, H2, CH4) estão bem correlacionados entre si, o que permitiu desenvolver modelos empíricos que minimizam o efeito das condições operatórias e, ao mesmo tempo, realçam o efeito do combustível na composição do gás. Em suma, os ensaios de pirólise realizados neste trabalho permitiram constatar que a estequiometria da pirólise de biomassa se relaciona de várias formas com a composição elementar do combustível original o que levanta várias possibilidades para a avaliação e projeto de processos de combustão e gasificação de biomassa.

Wireless mesh networks present an attractive communication solution for various research and industrial projects. However, in many cases, the appropriate preliminary calculations which allow predicting the network behavior have to be made before the actual deployment. For such purposes, network simulation environments emulating the real network operation are often used. Within this paper, a behavior comparison of real wireless mesh network (based on 802.11s amendment) and the simulated one has been performed. The main objective of this work is to measure performance parameters of a real 802.11s wireless mesh network (average UDP throughput and average one-way delay) and compare the derived results with characteristics of a simulated wireless mesh network created with the NS-3 network simulation tool. Then, the results from both networks are compared and the corresponding conclusion is made. The corresponding results were derived from simulation model and real-worldtest-bed, showing that the behavior of both networks is similar. It confirms that the NS-3 simulation model is accurate and can be used in further research studies.

Glioblastoma multiforme (GBM) tumors are the most common malignant primary brain tumors in adults. Although many GBM tumors are believed to be caused by self-renewing, glioblastoma-derived stem-like cells (GSCs), the mechanisms that regulate self-renewal and other oncogenic properties of GSCs are only now being unraveled. Here we showed that GSCs derived from GBM patient specimens express varying levels of the transcriptional repressor repressor element 1 silencing transcription factor (REST), suggesting heterogeneity across different GSC lines. Loss- and gain-of-function experiments indicated that REST maintains self-renewal of GSCs. High REST-expressing GSCs (HR-GSCs) produced tumors histopathologically distinct from those generated by low REST-expressing GSCs (LR-GSCs) in orthotopic mouse brain tumor models. Knockdown of REST in HR-GSCs resulted in increased survival in GSC-transplanted mice and produced tumors with higher apoptotic and lower invasive properties. Conversely, forced expression of exogenous REST in LR-GSCs produced decreased survival in mice and produced tumors with lower apoptotic and higher invasive properties, similar to HR-GSCs. Thus, based on our results, we propose that a novel function of REST is to maintain self-renewal and other oncogenic properties of GSCs and that REST can play a major role in mediating tumorigenicity in GBM. Remote monitoring of a power boiler allows the supplying company to make sure that equipment is used as supposed to and gives a good chance for process optimization. This improves co-operation between the supplier and the customer and creates an aura of trust that helps securing future contracts. Remote monitoring is already in use with recovery boilers but the goal is to expand especially to biomass-fired BFB-boilers. To make remote monitoring possible, data has to be measured reliably on site and the link between the power plant and supplying company’s server has to work reliably. Data can be gathered either with the supplier’s sensors or with measurements originally installed in the power plant if the plant in question is not originally built by the supplying company. Main goal in remote monitoring is process optimization and avoiding unnecessary accidents. This can be achieved for instance by following the efficiency curves and fouling in different parts of the process and comparing them to past values. The final amount of calculations depends on the amount of data gathered. Sudden changes in efficiency or fouling require further notice and in such a case it’s important that dialogue toward the power plant in question also works.

A study has been conducted focusing on how the phosphorus renrx)val efficiency of a constructed wetland (CW) can be optimized through the selective enrichment of the substratum. Activated alumina and powdered iron were examined as possible enrichment compounds. Using packed glass column trials it was found that alumina was not suitable for the renx)val of ortho-phosphate from solution, while mixtures of powdered iron and quartz sand proved to be very efficient. The evaluation of iron/sand mixtures in CWs planted with cattails was performed in three stages; first using an indoor lab scale wetland, then an outdoor lab scale wetland, and finally in a small scale pilot project. For the lab scale tests, three basic configurations were evaluated: using the iron/sand as a pre-filter, in the root bed. and as a post filter. Primary lagoon effluent was applied to the test cells to simulate actual CW conditions, and the total phosphorus and iron concentrations of the influent and effluent were nfK)nitored. The pilot scale trials were limited to using only a post filter design, due to in-progress research at the pilot site. The lab scale tests achieved average renrK>val efficiencies greater than 91% for all indoor configurations, and greater than 97% for all outdoor configurations. The pilot scale tests had an average renK)val efficiency of 60%. This relatively low efficiency in the pilot scale can be attributed to the post filters being only one tenth the size of the lab scale test in terms of hydraulic loading (6 cm/day vs. 60 cm/day).

The effect that plants {Typha latifolia) as well as root-bed medium physical and chemical characteristics have on the treatment of primary treated domestic wastewater within a vertical flow constructed wetland system was investigated. Five sets of cells, with two cells in each set, were used. Each cell was made of concrete and measured 1 .0 m X 1 .0 m and was 1.3 m deep. Four different root-bed media were tested : Queenston Shale, Fonthill Sand, Niagara Shale and a Michigan Sand. Four of the sets contained plants and a single type of root-bed medium. The influence of plants was tested by operating a Queenston Shale set without plants. Due to budget constraints no replicates were constructed. All of the sets were operated independently and identically for twenty-eight months. Twelve months of data are presented here, collected after 16 months of continuous operation. Root-bed medium type did not influence BOD5 removal. All of the sets consistently met Ontario Ministry of Environment (MOE) requirements (<25 mg/L) for BOD5 throughout the year. The 12 month average BOD5 concentration from all sets with plants was below 2.36 mg/L. All of the sets were within MOE discharge requirements (< 25 mg/L) for suspended solids with set effluent concentrations ranging from 1.53 to 14.80 mg/L. The Queenston Shale and Fonthill Sand media removed the most suspended solids while the Niagara Shale set produced suspended solids. The set containing Fonthill Sand was the only series to meet MOE discharge requirements (< Img/L) for total phosphorus year-round with a twelve month mean effluent concentration of 0.23 mg/L. Year-round all of the root-bed media were well below MOE discharge requirements (< 20mg/L in winter and < 10 mg/L in sumnner) for ammonium. The Queenston Shale and Fonthill Sand sets removed the most total nitrogen. Plants had no effect on total nitrogen removal, but did influence how nitrogen was cycled within the system. Plants increased the removal of suspended solids by 14%, BOD5 by 10% and total phosphorus by 22%. Plants also increased the amount of dissolved oxygen that entered the system. During the plant growing season removal of total phosphorus was better in all sets with plants regardless of media type. The sets containing Queenston Shale and Fonthill Sand media achieved the best results and plants in the Queenston Shale set increased treatment efficiency for every parameter except nitrogen. Vertical flow wetland sewage treatment systems can be designed and built to consistently meet MOE discharge requirements year-round for BOD5, suspended solids, total phosphorus and ammonium. This system Is generally superior to the free water systems and sub-surface horizontal flow systems in cold climate situations.

This thesis investigated the subcellular location of skeletal muscle PLIN proteins (PLIN2, PLIN3, and PLIN5) as well as protein interactions with ATGL and HSL at rest and following lipolytic stimulation. In addition, the serine phosphorylation state of PLIN2, PLIN3, and PLIN5 was determined at rest and following lipolytic stimulation. An isolated whole muscle technique was used to study the effects of contraction and epinephrine-induced lipolysis. This method allowed for the examination of the effects of contraction and epinephrine alone and in combination. Further, the soleus was chosen for investigating the role of PLIN proteins in skeletal muscle lipolysis due to its suitability for isolated incubation, and the fact that it is primarily oxidative in nature (~80% type I fibres). It has also been previously shown to have the greatest reliance on lipid metabolism and for this reason is ideal for investigating the role of PLIN proteins in lipolysis. Immunofluorescence microscopy revealed that skeletal muscle lipid droplets are partially co-localized to both PLIN2 and PLIN5 and that contraction does not affect the amount of colocalization, indicating that PLIN5 is not recruited to lipid droplets with contraction (PLIN2 ~65%; PLIN5 ~56%). Results from the immunoprecipitation studies revealed that with lipolysis in skeletal muscle the interaction between ATGL and CGI-58 is increased (study 2: 128% with contraction, p<0.05; study 3: 50% with contraction, 25% epinephrine, 80% contraction + epinephrine, p>0.05). Further PLIN2, PLIN3, and PLIN5 all interact with ATGL and HSL, while only PLIN3 and PLIN5 interact with CGI-58. Among these interactions, the association between PLIN2 and ATGL decreases with lipolytic stimulation (study 2: 21% with contraction, p<0.05). Finally our results demonstrate that PLIN3 and PLIN5 are serine phosphorylated at rest and that the level of phosphorylation remains unchanged in the face of either contractile or adrenergic stimulation. In summary, the regulation of skeletal muscle lipolysis is a complex process involving multiple proteins and enzymes. The skeletal muscle PLIN proteins likely play a role in skeletal muscle lipid droplet dynamics, and the data from this thesis indicate that these proteins may work together in regulating lipolysis by interaction with both ATGL and HSL.

Cette thèse examine le rythme veille-sommeil et son association avec l’instabilité émotionnelle, l’agressivité et l’impulsivité dans le trouble de personnalité limite (TPL) à l’adolescence. Dans un premier temps, la revue de la littérature sur les perturbations objectives du sommeil dans le TPL a mis en lumière plusieurs difficultés similaires, évaluées par polysomnographie, à celles observées dans la dépression adulte. De 1980 à 2010, aucune recherche n’a examiné le rythme veille-sommeil, aucune n’a étudié les adolescents TPL et plusieurs n’ont pas contrôlé l’état dépressif comme facteur de confusion. De ce constat, il s’avérait pertinent de mener une étude sur le rythme veille-sommeil dans le TPL à l’adolescence en l’absence de dépression co-occurrente. L’adolescence comportant plusieurs caractéristiques physiologiques, psychologiques et sociales, tenir compte des aspects développementaux était essentiel. Dans un second temps, un protocole de recherche fût mis en place à la Clinique des troubles de l’humeur et le recrutement a été réalisé auprès d’adolescents souffrant d’un TPL et sans état dépressif actuel. Ils devaient porter pendant plus de neuf jours (période comprenant deux fins de semaine) un actigraphe, appareil non invasif évaluant l’alternance veille-sommeil dans l’environnement naturel. L’abandon précoce au traitement étant prévalent chez les patients TPL, la fiabilité de l’étude a été examinée afin de déterminer les raisons favorisant et celles nuisant au recrutement et à la collecte des données. La réflexion sur les aspects méthodologiques de l’étude actigraphique a permis d’expliquer les limites de ce type de protocole. Dans un troisième temps, le rythme veille-sommeil des adolescents TPL (n=18) a été caractérisé et comparé à celui des jeunes ayant un trouble bipolaire (n=6), trouble psychiatrique partageant plusieurs manifestations communes avec le TPL, et à celui des adolescents sans trouble de santé mentale (n=20). Les résultats suggèrent que l’adolescent TPL passe plus de temps en éveil durant la période de repos que les jeunes appartenant aux deux autres groupes. De plus, les adolescents TPL présentent une plus grande variabilité inter journalière des heures de lever et du temps total de sommeil que les autres adolescents. Ils se réveillent une heure de plus, et dorment donc une heure supplémentaire, que les adolescents sans trouble mental lors des journées sans routine. Dans un quatrième temps, les analyses corrélationnelles entre les données actigraphiques et les scores aux questionnaires auto-rapportés évaluant l’instabilité émotionnelle, l’agressivité et l’impulsivité suggèrent que plus l’adolescent TPL passe du temps éveillé alors qu’il est au lit, plus il déclare présenter des comportements agressifs, surtout physiques, durant le jour. En résumé, cette thèse contribue à la littérature scientifique en explorant pour la première fois le rythme veille-sommeil et son lien avec les manifestations symptomatiques dans le TPL à l’adolescence. Les résultats suggèrent fortement l’importance d’évaluer et de traiter les problèmes du rythme veille-sommeil que présentent ces jeunes lors de la prise en charge.

Problématique : La littérature scientifique ne contient actuellement aucune étude épidémiologique portant sur la relation entre l’exposition à une infestation de punaises de lit (Cimex lectularis) et la santé mentale. L’objectif de cette étude est d’explorer la relation entre le statut d’exposition aux punaises de lit et des mesures de dépression, anxiété et perturbation du sommeil. Méthodes : Cette recherche est une étude transversale basée sur une analyse secondaire des données provenant de N=91 adultes locataires de logements insalubres qui ont répondu à un questionnaire de santé au moment d’interventions médico-environnementales menées par la direction de santé publique de Montréal entre janvier et juin 2010. Le questionnaire de santé inclut le « Questionnaire de santé du patient (QSP-9) », qui est un outil mesurant les symptômes associés à la dépression, l’outil de dépistage d’anxiété généralisée (GAD-7) et les items 1-18 de l’indice de la qualité du sommeil de Pittsburgh (PSQI). L’association entre une exposition autorapportée à une infestation de punaises de lit et le niveau de perturbation du sommeil selon la sous-échelle correspondante du PSQI, les symptômes dépressifs selon le QSP-9 et les symptômes anxieux selon le GAD-7, a été évaluée en utilisant une analyse de régression linéaire multivariée. Des données descriptives relatives aux troubles de sommeil autodéclarés et à des comportements reliés à un isolement social, dus à une exposition aux punaises de lit, sont aussi présentées. Résultats : L’échantillon comprenait 38 hommes et 53 femmes. Parmi les 91 participants, 49 adultes ont signalé une exposition aux punaises de lit et de ce nombre, 53,06% (26/49) ont déclaré des troubles de sommeil et 46,94% (23/49) des comportements d’isolement social, en raison de l’exposition. Les adultes exposés à des punaises de lit ont obtenu des résultats significativement plus élevés sur le QSP-9 (p=0,025), le GAD-7 (p=0,026) et sur la sous-échelle mesurant la perturbation du sommeil (p=0,003) comparativement à ceux qui n’étaient pas exposés. Conclusions : Cette première étude exploratoire met en évidence une association significative entre l’état d’exposition aux punaises de lit et des troubles de sommeil, ainsi que des symptômes anxieux et dépressifs, indiquant que ceux qui sont exposés aux punaises de lit représentent un groupe possiblement à risque de développer des problèmes de santé mentale. Ces résultats viennent en appui aux initiatives des décideurs pour organiser des efforts coordonnés d’éradication au niveau des villes et des provinces.

Background. Sleep-wake disturbances are among the most persistent sequelae after traumatic brain injury (TBI) and probably arise during the hospital stay following TBI. These disturbances are characterized by difficulties sleeping at night and staying awake during the day. Objective. The aim of the present study was to document rest-activity cycle consolidation in acute moderate/severe TBI using actigraphy and to assess its association with injury severity and outcome. Methods. In all, 16 hospitalized patients (27.1 ± 11.3 years) with moderate/severe TBI wore actigraphs for 10 days, starting in the intensive care unit (ICU) when continuous sedation was discontinued and patients had reached medical stability. Activity counts were summed for daytime (7:00-21:59 hours) and nighttime periods (22:00-6:59 hours). The ratio of daytime period activity to total 24-hour activity was used to quantify rest-activity cycle consolidation. An analysis of variance was carried out to characterize the evolution of the daytime activity ratio over the recording period. Results. Rest-activity cycle was consolidated only 46.6% of all days; however, a significant linear trend of improvement was observed over time. Greater TBI severity and longer ICU and hospital lengths of stay were associated with poorer rest-activity cycle consolidation and evolution. Patients with more rapid return to consolidated rest-activity cycle were more likely to have cleared posttraumatic amnesia and have lower disability at hospital discharge. Conclusions. Patients with acute moderate/ severe TBI had an altered rest-activity cycle, probably reflecting severe fragmentation of sleep and wake episodes, which globally improved over time. A faster return to rest-activity cycle consolidation may predict enhanced brain recovery.
