[ES] The olive ridley sea turtle Lepidochelys olivacea has been recorded in the Cape Verde Islands, but the most recent published data (1998-2000) are of stranded ndividuals and remains only. This article presents new data on olive ridleys recorded during the years 2001-2011 on Boavista and Sal islands. The presence of his species does not appear to be related to nesting activity. The possible geographical origin of these turtles is discussed. In addition, we propose some studies that could help to reinforce the conservation of sea turtles in West Africa.

In questo elaborato è stato discusso il tema della depurazione delle acque reflue, in particolar modo il sistema SBR. L'SBR (Sequencing Batch Reactors) è un sistema di impianti di depurazione composto da una vasca di raccolta del liquame e una o più vasche in parallelo, i reattori, che consistono nel vero e proprio cure dell'impianto. Di fatto le fasi del processo depurativo restano immutate rispetto ad un impianto tradizionale, ma l'SBR introduce come nuovo parametro dimensionale il tempo. infatti, tutti i trattamenti depurativi sono svolti sequenzialmente nel reattore. Un programma gestisce le varie fasi del trattamento esclusivamente sulla base della loro durata (stabilita relativamente alla natura del refluo e quindi ai trattamenti da svolgere in maniera più o meno intensiva). Questa caratteristica rende l'intero impianto molto versatile nel caso di variazioni dei dati in ingresso. Per questo motivo gli impianti SBR sono ottimi per la piccola media impresa, in quanto sono facilmente adattabili alle variazioni stagionali di produzione. Sono inoltre possibili realizzazioni di impianti per il trattamento di ingenti portate, o elevate concentrazioni, ponendo più reattori in parallelo alimentati dalle stessa vasca di accumulo. In questo modo è possibile svolgere un maggiore numero di cicli depurativi al giorno e quindi rispettare i valori normativi in uscita dell'impianto. In conclusione questa tipologia d'impianto presenta notevoli vantaggi fra i quali anche quello di avere bassi costi operativi. La motivazione di ciò sta nel fatto che l'impianto lavora solo se ha effettivamente una portata, per cui se non vi è presenza di refluo il reattore non lavora e quindi non comporta costi.

Sound localisation is defined as the ability to identify the position of a sound source. The brain employs two cues to achieve this functionality for the horizontal plane, interaural time difference (ITD) by means of neurons in the medial superior olive (MSO) and interaural intensity difference (IID) by neurons of the lateral superior olive (LSO), both located in the superior olivary complex of the auditory pathway. This paper presents spiking neuron architectures of the MSO and LSO. An implementation of the Jeffress model using spiking neurons is presented as a representation of the MSO, while a spiking neuron architecture showing how neurons of the medial nucleus of the trapezoid body interact with LSO neurons to determine the azimuthal angle is discussed. Experimental results to support this work are presented.

Lo scopo di questo lavoro di tesi è stato quello di studiare l’efficacia e l’applicabilità dello strumento HERACLES II Flash Gas Chromatography Electronic Nose mediante l’analisi di un set molto ampio di campioni di oli d’oliva vergini reperiti presso un concorso nazionale. Nello specifico, mediante elaborazioni di statistica multivariata dei dati, è stata valutata sia la capacità discriminante del metodo per campioni caratterizzati da un diverso profilo sensoriale, sia la correlazione esistente tra l’intensità delle sensazioni percepite per via sensoriale ed i dati ottenuti mediante l’analisi strumentale. La valutazione delle caratteristiche sensoriali dei campioni è stata realizzata in occasione dello stesso concorso da parte di un gruppo di giudici esperti e secondo le modalità stabilite dai regolamenti comunitari. Ogni campione è stato valutato da almeno 8 assaggiatori che hanno determinato l’intensità di caratteristiche olfattive (eventuali difetti, fruttato e note secondarie positive) e gustative (amaro e piccante). La determinazione dei composti volatili, invece, è stata condotta mediante lo strumento HERACLES II Electronic Nose (AlphaMOS), dotato di due colonne cromatografiche caratterizzate da diversa polarità (MXT-5 con fase stazionaria apolare e MXT-WAX con fase stazionaria polare), ciascuna collegata ad un rivelatore di tipo FID. L’elaborazione multivariata dei dati è stata realizzata mediante il software in dotazione con lo strumento.

L’analisi chimica delle acque di dieci pozzi, di una località posta a N del fiume Po, (44°54’51.10” N; 11°36’42.49”E), ha mostrato la presenza di acque clorurato sodiche, che si suppone derivino da miscelazione (complessa) tra acque di origine marina (acqua di strato) e un’acqua dolce madre (acqua teorica, end member). Si è tentato di calcolare teoricamente la composizione dell’end member, che è risultata un’acqua bicarbonato sodica, e si è proposta la possibile giacitura delle facies idrochimiche riscontrate all’interno dello schema idrostratigrafico regionale Emiliano-Romagnolo. È emerso che le acque analizzate sono presenti ad una profondità anormalmente ridotta, rispetto alle giaciture degli end members di miscelazione: le acque saline di Ambrogio e Diamantina e l’acqua dolce teorica. Considerando la distribuzione geografica delle acque clorurato sodiche nella regione Emilia-Romagna ed il chiaro rapporto che queste hanno con la verticale degli alti strutturali appenninici sepolti si propone una origine influenzata dalla tettonica per la risalita delle acque studiate.

Olive (Olea europaea L.), one of the main crops in the Mediterranean basin, is mainly propagated by cuttings, a classical propagation method that relies on the ability of the cuttings to form adventitious roots. While some cultivars are easily propagated by this technique, some of the most interesting olive cultivars are considered difficult-to-root which poses a challenge for their preservation and commercialization. Therefore, increasing the current knowledge on adventitious root formation is extremely important for species like olive. This research focuses on evaluating the role of free auxins and oxidative enzymes on adventitious root formation of two olive cultivars with different rooting ability - ‘Galega vulgar’ (difficult-to-root) and ‘Cobrançosa’ (easy-to-root). In this context, free auxin levels and enzyme activities were determined in in vitro-cultured ‘Galega vulgar’ microshoots and in semi-hardwood cuttings of cvs. ‘Galega vulgar’ and ‘Cobrançosa’. To attain this goal, an analytical method for the quantification of free indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) was developed, which is based on dispersive liquid-liquid microextraction followed by microwave derivatization (DLLME-MAD) and gas chromatography-mass spectrometry (GC/MS) analysis. The developed method was validated in terms of linearity, recovery, limit of detection (LOD) and limit of quantification (LOQ) and proved to be useful in the analysis of two very different types of plant tissues. The results from auxin quantification in olive samples point at a relationship between free auxin levels and rooting ability of both microshoots and semihardwood cuttings. A defective IBA-IAA conversion, resulting in a peak of free IAA during initiation phase, seems to be associated with low rooting ability. Likewise, differences in the activity of oxidative enzymes also appear to be related with rooting ability. Higher polyphenol oxidases (PPO) activity is likely related with an easyto- root behavior, while the opposite is true for peroxidases (POX) (including IAA oxidase (IAAox)) activity. A possible hypothesis for adventitious root formation in olive microcuttings is presented herein for the first time. Free auxins, oxidative enzymes, alternative oxidase (AOX) and reactive oxygen species (ROS) are some of the factors that may be involved in this highly complex physiological process. Interestingly, while temporal changes in auxin levels were similar between microshoots and semihardwood cuttings, the conclusions obtained from enzyme activity results in microshoots didn’t translate to semi-hardwood tissues, showing the emerging need for adaptation of classical agronomical research studies to modern techniques; Resumo: Procurando compreender o papel das auxinas e enzimas oxidativas na formação de raízes adventícias em cultivares de oliveira (Olea europaea L.) A oliveira (Olea europaea L.) é uma das principais culturas da bacia Mediterrânica e é propagada maioritariamente por estacaria, um processo altamente dependente da capacidade das estacas para formar raízes adventícias. Enquanto algumas cultivares são fáceis de propagar desta forma, algumas das cultivares de oliveira mais interessantes são consideradas difíceis de enraizar, o que dificulta a sua preservação e comercialização e torna extremamente importante aprofundar o conhecimento sobre o enraizamento adventício desta espécie. Este trabalho foca-se na avaliação do papel das auxinas livres e das enzimas oxidativas na formação de raízes adventícias em duas cultivares de oliveira com diferente capacidade de enraizamento - ‘Galega vulgar’ (difícil de enraizar) e ‘Cobrançosa’ (fácil de enraizar). Neste contexto, determinaram-se os níveis de auxinas livres e as actividades de enzimas oxidativas em microestacas de ‘Galega vulgar’ cultivadas in vitro bem como em estacas semi-lenhosas das cvs. ‘Galega vulgar’ e ‘Cobrançosa’. Para tal foi necessário desenvolver uma metodologia analítica para a quantificação de ácido indol-3-acético (IAA) e ácido indol-3-butírico (IBA), baseada em microextracção dispersiva líquido-líquido (DLLME) seguida de derivatização em microondas (MAD) e análise por cromatografia gasosa acoplada a espectrometria de massa (GC/MS). O método desenvolvido foi validado em termos de linearidade, recuperação, limite de detecção (LOD) e limite de quantificação (LOQ), e mostrou-se eficaz na análise de dois tipos de tecidos vegetais bastante diferentes. Os resultados da análise de auxinas em amostras de oliveira apontam para uma possível relação entre os níveis de auxinas livres e a capacidade de enraizamento, tanto em microestacas como em estacas semi-lenhosas. Uma conversão IBA-IAA deficiente, que resulta num pico de IAA durante a fase de iniciação, parece estar associada à baixa capacidade de enraizamento. Por outro lado, a capacidade de enraizamento também parece estar relacionada com diferenças na actividade de enzimas oxidativas. Comportamentos fáceis de enraizar estão associados a actividade mais elevada das polifenoloxidases (PPO), enquanto o oposto é verdade para a actividade das peroxidases (POX) (incluindo a IAA oxidase (IAAox)). Neste trabalho propõe-se pela primeira vez uma possível explicação para o enraizamento adventício em microestacas de oliveira. Auxinas livres, enzimas oxidativas, oxidase alternativa (AOX) e espécies reactivas de oxigénio (ROS) são alguns dos factores envolvidos neste processo fisiológico altamente complexo. Curiosamente, enquanto as alterações temporais nos níveis de auxinas foram semelhantes entre microestacas e estacas semi-lenhosas, o mesmo não se observou relativamente à actividade enzimática, o que mostra a necessidade de adaptação dos estudos agronómicos tradicionais às técnicas correntes.

Aiming to introduce a multiresidue analysis for the trace detection of pesticide residues belonging to organophosphorus and triazine classes from olive oil samples, a new sample preparation methodology comprising the use of a dual layer of “tailor-made” molecularly imprinted polymers (MIPs) SPE for the simultaneous extraction of both pesticides in a single procedure has been attempted. This work has focused on the implementation of a dual MIP-layer SPE procedure (DL-MISPE) encompassing the use of two MIP layers as specific sorbents. In order to achieve higher recovery rates, the amount of MIP layers has been optimized as well as the influence of MIP packaging order. The optimized DL-MISPE approach has been used in the preconcentration of spiked organic olive oil samples with concentrations of dimethoate and terbuthylazine similar to the maximum residue limits and further quantification by HPLC. High recovery rates for dimethoate (95%) and terbuthylazine (94%) have been achieved with good accuracy and precision. Overall, this work constitutes the first attempt on the development of a dual pesticide residue methodology for the trace analysis of pesticide residues based on molecular imprinting technology. Thus, DL-MISPE constitutes a reliable, robust, and sensitive sample preparation methodology that enables preconcentration of the target pesticides in complex olive oil samples, even at levels similar to the maximum residue limits enforced by the legislation.

Harvest efficiency is defined as the percentage of fruits harvested by total production. The percentage of fruits harvested is less than 100% when working with trunk shakers to detach olives. It is important to increase the percentage of fruits harvested in order to increase farmer’s income. This objective can be achieved knowing the evolution of the main factors affecting fruit detachment. Fruit removal force (FRF), fruit weight (P) and the ratio between them are important for harvest efficiency. Field trials took place for two years (2013-2014) in Vilariça Valley, northeast Portugal in an olive orchard with ‘Cobrançosa Transmontana’ cultivar. It was adopted a mechanical harvesting system based on a trunk shaker to detach fruits, and an inverted umbrella to collect fruits. Elementary operation times were measured in seconds to evaluate work rates. FRF and P were measured in the ripening period, to evaluate their evolution. In this paper are presented the preliminary results of the ratio FRF (fruit removal force)/fruit weight evolution during the ripening period (P) and the results of the equipment work rate (trees h-1). The ratio FRF/P has predominantly descendant values in the weeks before harvest, from 140 to 80 as a result of a FRF downward variation from 4.9 to 2.94 N and an upward variation of P from 0.0294 to 0.0637 N. The FRF/P ratio stabilizes the decline in the last week of November just before harvesting, registering in some cases a slight increase in consequence of FRF increase higher than P increase (contrary to the tendency of previous weeks). Equipment work rate showed values between 40 and 57 trees h-1, confirming previous results.

Objective: We evaluated the protective activity of an extract from a by-product such as olive stones, through its ability to inhibit H2O2 induced apoptosis in the SH-SY5Y human neuroblastoma cell line. Material and methods: To such end, 20,000 cells/well were cultivated and differentiation with retinoic acid was initiated. Once the cells were differentiated, apoptosis was induced with and without H2O2 extract. Finally, cDNA extraction was performed, and pro-apoptotic genes Bax and anti-apoptotic genes Bcl-2 were analyzed. Quantification of the gene expression was performed using the GAPDH gene marker. Results: Cell viability with the extract is 97.6% (SD 5.7) with 10 mg/l and 62.8% (SD 1.2) to 50 mg/l, using 10 mg/l for the biomarker assay. The retinoic acid differentiated SH-S cell line (10 µM) shows a clear apoptosis when treated with H2O2 150 µM, with a Bax/Bcl-2 ratio of 3.75 (SD 0.80) in contrast to the differentiated control cells subjected to H2O2 and with extract, which have the same ratio of 1.02 (SD 0.01-0.03). Conclusion: The olive stone extract shows anti-apoptotic activity in the provoked cell death of SH-SY5Y human neuroblastoma cells in their normal state, defending them from oxidative stress which produces a significant increase in the apoptotic gene ratio in contrast to anti-apoptotic genes (Bax/Bcl-2).

"Though olive oil is a perishable product, there is not a European regulation for maximum consumption time after production, in part because its durability depends on the storage conditions. The main objectives of this study were to compare the influence of the type of storage on changes of Portuguese virgin olive oils and to verify whether the addition of Catostylus tagi could increase the oxidative stability of olive oil. Over 12 months, the conservation status of monovarietal and blended olive oils in four contexts possible to be used by the consumer was monitored. The analyzed parameters were chlorophyll content, free acidity, peroxide value, specific extinction coefficients at 232 and 270 nm and delta K. Spaced determinations of iodine index and total tocopherol contents complemented the study. Results showed that at Mediterranean temperatures and normal storage procedure, the mean time to reach maximum peroxides value was 12–13 months. At artificial light storage, C. tagi was effective in reducing peroxides evolution by 11 %."

The activity of oxidative enzymes and the levels of free auxins were determined during adventitious root formation in olive explants. Rooting trials were performed both with in vitro-cultured micro shoots of the cultivar ‘Galega Vulgar’, treated with indole-3-butyric acid (IBA) and with salicylhydroxamic acid(SHAM) + IBA, as well as with semi-hardwood cuttings of the cultivars ‘Galega Vulgar’ (difficult-to-root)and ‘Cobrançosa’ (easy-to-root), treated with IBA. The auxin (IBA) was used in all experiments as a rooting promoter, while SHAM was used in micropropagation trials as rooting inhibitor, providing a negative control. Free indole-3-acetic acid (IAA) and IBA concentrations were determined in microshoots, as well as in semi-hardwood cuttings, throughout the rooting period at pre-established time-points. At the sametime-points, the enzymatic activity of polyphenol oxidases (PPO), peroxidases (POX), and IAA oxidase(IAAox) was evaluated in the microshoots. Microshoots treated with SHAM + IBA revealed higher POX and IAAox activity, as well as lower PPO activity, than those treated only with IBA. IAA levels were higher in IBA-treated microshoots during induction phase, but lower during early initiation phase. Incontrast, free IBA levels were higher in microshoots treated with SHAM + IBA during induction, but lower during initiation. A similar pattern of free auxin levels was observed in semi-hardwood cuttings of the two contrasting cultivars under evaluation. The similarities found on the auxin patterns of microshoots treated with SHAM and those of semi-hardwood cuttings of the difficult-to-root olive cultivar allow considering SHAM a reliable control for when simulation of a difficult-to-root behavior is necessary. The inhibitory effect of SHAM in root formation could be related with 1) the inhibition of alternative oxidase(AOX), leading to a down regulation of phenylpropanoid biosynthetic pathways, which would decrease the concentration of phenolic substrates for PPO; 2) an increase in IAAox activity resulting in lower free IAA levels or; 3) a defective conversion of IBA into IAA.
