Centronuclear myopathy (CNM) is a genetically heterogeneous disorder associated with general skeletal muscle weakness, type I fiber predominance and atrophy, and abnormally centralized nuclei. Autosomal dominant CNM is due to mutations in the large GTPase dynamin 2 (DNM2), a mechanochemical enzyme regulating cytoskeleton and membrane trafficking in cells. To date, 40 families with CNM-related DNM2 mutations have been described, and here we report 60 additional families encompassing a broad genotypic and phenotypic spectrum. In total, 18 different mutations are reported in 100 families and our cohort harbors nine known and four new mutations, including the first splice-site mutation. Genotype-phenotype correlation hypotheses are drawn from the published and new data, and allow an efficient screening strategy for molecular diagnosis. In addition to CNM, dissimilar DNM2 mutations are associated with Charcot-Marie-Tooth (CMT) peripheral neuropathy (CMTD1B and CMT2M), suggesting a tissue-specific impact of the mutations. In this study, we discuss the possible clinical overlap of CNM and CMT, and the biological significance of the respective mutations based on the known functions of dynamin 2 and its protein structure. The complete surgical removal of disease is a desirable outcome particularly in oncology. Unfortunately much disease is microscopic and difficult to detect causing a liability to recurrence and worsened overall prognosis with attendant costs in terms of morbidity and mortality. It is hoped that by advances in optical diagnostic technology we could better define our surgical margin and so increase the rate of truly negative margins on the one hand and on the other hand to take out only the necessary amount of tissue and leave more unaffected non-diseased areas so preserving function of vital structures. The task has not been easy but progress is being made as exemplified by the presentations at the 2nd Scientific Meeting of the Head and Neck Optical Diagnostics Society (HNODS) in San Francisco in January 2010. We review the salient advances in the field and propose further directions of investigation.

Abstract Background Facilitating the provision of appropriate health care for immigrant and Aboriginal populations in Canada is critical for maximizing health potential and well-being. Numerous reports describe heightened risks of poor maternal and birth outcomes for immigrant and Aboriginal women. Many of these outcomes may relate to food consumption/practices and thus may be obviated through provision of resources which suit the women's ethnocultural preferences. This project aims to understand ethnocultural food and health practices of Aboriginal and immigrant women, and how these intersect with respect to the legacy of Aboriginal colonialism and to the social contexts of cultural adaptation and adjustment of immigrants. The findings will inform the development of visual tools for health promotion by practitioners. Methods/Design This four-phase study employs a case study design allowing for multiple means of data collection and different units of analysis. Phase 1 consists of a scoping review of the literature. Phases 2 and 3 incorporate pictorial representations of food choices (photovoice in Phase 2) with semi-structured photo-elicited interviews (in Phase 3). The findings from Phases 1-3 and consultations with key stakeholders will generate key understandings for Phase 4, the production of culturally appropriate visual tools. For the scoping review, an emerging methodological framework will be utilized in addition to systematic review guidelines. A research librarian will assist with the search strategy and retrieval of literature. For Phases 2 and 3, recruitment of 20-24 women will be facilitated by team member affiliations at perinatal clinics in one of the city's most diverse neighbourhoods. The interviews will reveal culturally normative practices surrounding maternal food choices and consumption, including how women negotiate these practices within their own worldview and experiences. A structured and comprehensive integrated knowledge translation plan has been formulated. Discussion The findings of this study will provide practitioners with an understanding of the cultural differences that affect women's dietary choices during maternity. We expect that the developed resources will be of immediate use within the women's units and will enhance counseling efforts. Wide dissemination of outputs may have a greater long term impact in the primary and secondary prevention of these high risk conditions.

While histopathology of excised tissue remains the gold standard for diagnosis, several new, non-invasive diagnostic techniques are being developed. They rely on physical and biochemical changes that precede and mirror malignant change within tissue. The basic principle involves simple optical techniques of tissue interrogation. Their accuracy, expressed as sensitivity and specificity, are reported in a number of studies suggests that they have a potential for cost effective, real-time, in situ diagnosis.

Problematiche delle infezioni da Citomegalovirus in gravidanza Obiettivi: migliorare la sensibilità dell'ecografia nella diagnosi di infezione da CMV individuando un reperto ecografico cerebrale suggestivo di infezione fetale da Citomegalovirus a 20 settimane di gestazione. Metodi: tra febbraio 1989 e settembre 2009, 721 pazienti afferenti alla nostra Unità di Medicina Materno fetale per infezione primaria da CMV hanno eseguito amniocentesi e sono state sottoposte ad un esame neurosonografico transvaginale a 20-22 settimane di gestazione. Risultati: in 29 feti con infezione congenita sono state evidenziate anomalie ecografiche (17%), di cui in 22 casi a livello cerebrale. In 13 casi l'ecografia transvaginale ha permesso di identificare un alone ecogeno periventricolare a margini ben definiti ad un'epoca gestazionale media di 20.5 settimane (20-22 settimane). Di questi casi 12 pazienti hanno deciso di interrompere la gravidanza. L'unico neonato ha presentato alla nascita un'ipoacusia bilaterale. I riscontri autoptici ottenuti (7/12) hanno mostrato un'infezione citomegalica disseminata ed in 3 casi segni a livello cerebrale. Conclusioni: il limite ecografico della diagnosi di infezione fetale nei casi di infezione primaria da CMV è noto. In pazienti gravide alla 20 settimana di gestazione con infezione recente da Citomegalovirus, il riscontro di un alone ecogeno periventricolare risulta essere un precoce ed attendibile segno di infezione fetale e di possibile danno della sostanza bianca cerebrale. Occorrono però studi aggiuntivi per valutare la possibile manifestazione clinica di questa anomalia cerebrale nei neonati con infezione da CMV.

Zentrales Thema der Arbeit war die Aufklärung von Verwandtschaftsverhältnissen im „Tree of Life“ der vielzelligen Tiere (Metazoa) unter Einsatz großer DNA-Sequenzdatensätze und phylogenomischer Methoden. Zur Untersuchung der internen Phylogenie der Syndermata (= meist freilebende Rädertiere („Rotifera“) + endoparasitische Kratzwürmer (Acanthocephala)) sowie ihrer Position im Metazoen-Stammbaum wurden insgesamt sieben neue mitochondriale (mt) Genome sowie neue Transkriptom-Sequenzdaten von sieben verschiedenen Syndermata-Spezies generiert und/oder analysiert. Die Stammbaumrekonstruktionen auf Grundlage dieser sowie orthologer Sequenzen anderer Spezies in Form von phylogenomischen Datensätzen mit bis zu 82.000 Aminosäurepositionen ergaben folgende Aussagen zur Evolution: (i) Innerhalb der Acanthocephala bilden monophyletische Palaeacanthocephala das Schwestertaxon zu den Eoacanthocephala. Die Archiacanthocephala sind Schwestertaxon zu allen vorgenannten. (ii) Innerhalb der Syndermata bilden die epizoisch lebenden Seisonidea das Schwestertaxon zu den endoparasitischen Acanthocephala (= Pararotatoria), die Bdelloidea sind das Schwestertaxon zu den Pararotatoria (= Hemirotifera) und die Monogononta das Schwestertaxon zu den Hemirotifera. Die klassischen Eurotatoria (= Bdelloidea + Monogononta) sind demnach paraphyletisch. (iii) Innerhalb der Metazoa bilden die Syndermata gemeinsam mit den Gnathostomulida die Gnathifera. Diese sind die Schwestergruppe zu allen anderen Spiralia-Taxa, welche sich in Rouphozoa (= Platyhelminthes + Gastrotricha) sowie die Lophotrochozoa aufspalten. Die Platyzoa (= Gnathifera + Platyhelminthes + Gastrotricha) sind demnach paraphyletisch. Diese phylogenetischen Hypothesen wurden im Hinblick auf ihre Implikationen für die Evolution morphologischer und ökologischer Merkmale interpretiert. Demnach sind während der Evolution dieser Tiergruppen mehrfach sekundäre Verlustereignisse von komplexen morphologischen Merkmalen aufgetreten (laterale sensorische Organe innerhalb der Acanthocephala und das Räderorgan (Corona) innerhalb der Syndermata), was die Verwendung dieser Merkmale im Sinne einer klassisch-morphologischen Phylogenetik kritisch erscheinen lässt. Der Endoparasitismus der Acanthocephala hat sich wahrscheinlich über ein epizoisches Zwischenstadium, wie man es heute noch bei den Seisonidea findet, entwickelt. Der letzte gemeinsame Vorfahre der Spiralia war vermutlich klein und unsegmentiert und besaß keine echte Leibeshöhle (Coelom). Demnach hätten sich Segmentierung und Coelome innerhalb der Metazoa mehrfach unabhängig voneinander (konvergent) entwickelt. Die Arbeit beinhaltete folgende weitere, zum Teil methodische Aspekte: (i) die Analyse der Architektur der mt Genome der Monogononta bestätigte die aberrante Organisation in zwei Subgenomen für die Brachionidae. (ii) Eine Prüfung der Tauglichkeit ribosomaler Proteine für molekular-phylogenetische Arbeiten ergab das Vorhandensein widersprüchlicher phylogenetischer Signale in diesen speziellen Proteinsequenzen. (iii) Es konnte nachgewiesen werden, dass systematische Fehler wie „long-branch attraction“ bei der Positionierung der Syndermata im Stammbaum der Metazoa eine große Rolle spielen und adressiert werden müssen.

Resistance of cancer cells towards chemotherapy is the major cause of therapy failure. Hence, the evaluation of cellular defense mechanisms is essential in the establishment of new chemotherapeutics. In this study, classical intrinsic and acquired as well as new resistance mechanisms relevant in the cellular response to the novel vacuolar H+-ATPase inhibitor archazolid B were investigated. Archazolid B, originally produced by the myxobacterium Archangium gephyra, displayed cytotoxicity in the low nanomolar range on a panel of cancer cell lines. The drug showed enhanced cytotoxic activity against nearly all cancerous cells compared to their non-cancerous pendants. With regards to ABC transporters, archazolid B was identified as a moderate substrate of ABCB1 (P-glycoprotein) and a weak substrate of ABCG2 (BCRP), whereas hypersensitivity was observed in ABCB5-expressing cells. The cytotoxic effect of archazolid B was shown to be independent of the cellular p53 status. However, cells expressing constitutively active EGFR displayed significantly increased resistance. Acquired drug resistance was studied by establishing an archazolid B-resistant MCF-7 cell line. Experiments showed that this secondary resistance was not conferred by aberrant expression or DNA mutations of the gene encoding vacuolar H+-ATPase subunit c, the direct target of archazolid B. Instead, a slight increase of ABCB1 and a significant overexpression of EGFR as well as reduced proliferation may contribute to acquired archazolid B resistance. For identification of new resistance strategies upon archazolid B treatment, omics data from bladder cancer and glioblastoma cells were analyzed, revealing drastic disturbances in cholesterol homeostasis, affecting cholesterol biosynthesis, uptake and transport. As shown by filipin staining, archazolid B led to accumulation of free cholesterol in lysosomes, which triggered sterol responses, mediated by SREBP-2 and LXR, including up-regulation of HMGCR, the key enzyme of cholesterol biosynthesis. Furthermore, inhibition of LDL uptake as well as impaired LDLR surface expression were observed, indicating newly synthesized cholesterol to be the main source of cholesterol in archazolid B-treated cells. This was proven by the fact that under archazolid B treatment, total free cholesterol levels as well as cell survival were significantly reduced by inhibiting HMGCR with fluvastatin. The combination of archazolid B with statins may therefore be an attractive strategy to circumvent cholesterol-mediated cell survival and in turn potentiate the promising anticancer effects of archazolid B.

L'obiettivo di questa tesi è proporre la traduzione di un testo per l'infanzia, Mon petit centre Pompidou, del quale non è stata ancora pubblicata una versione italiana. Il testo è una guida museale per bambini; l'autrice ha raccolto alcune delle opere che si trovano al centre Pompidou sotto forma di albo illustrato. Nell’elaborato, ci si propone di presentare qualche nozione di base sulla letteratura per l’infanzia e sulla sua traduzione, prima di passare direttamente alla proposta di traduzione vera e propria. A seguire un commento alla traduzione, in cui si esporranno le difficoltà incontrate e si darà spiegazione delle scelte effettuate. Le difficoltà principali della proposta di traduzione sono dovute allo specifico lettore target, per cui bisognerà adattare il testo di partenza in maniera da renderlo completamente fruibile e godibile per il bambino. A tal fine, sarà necessario operare certe modifiche al testo di partenza, simile ad una prosa poetica, per adattarlo alle particolarità fonetiche e espressive della lingua italiana.

Gegenstand dieser Arbeit ist die Kinder- und Jugendliteratur im Übersetzungsprozess. Kein Genre wird so sehr von der Arbeit des Übersetzens beeinflusst wie dieses. Am Beispiel von Mario Lodi werden die besonderen Merkmalen und Herausforderungen der Kinder- und Jugendliteratur erläutert, vor allem mit Blick auf den Übersetzungsprozess. Der Übersetzungsvorschlag von Mario Lodis Märchen La Puntura nimmt die zuvor erläuterten Merkmale und Schwierigkeiten wieder auf und erarbeitet Vorschläge und Herangehensweisen für diese an konkreten Beispielen. Im Vordergrund stehen kulturelle Unterschiede, das Beispiel Schule und die Umsetzung von Wortspielen. Im Mittelpunkt steht der Ausdruck LA Puntura und seine mehrfache Bedeutung. Das Resultat der kindlichen Wiederverwendung des originalen Terms.
