A sensor for H2O2 amperometric detection based on a Prussian blue (PB) analogue was developed. The electrocatalytic process allows the determination of hydrogen peroxide at 0.0 V with a limit of detection of 1.3 mu mol L-1 in a flow injection analysis (FIA) configuration. Studies on the optimization of the FIA parameters were performed and under optimal FIA operational conditions the linear response of the method was extended up to 500 mu mol L-1 hydrogen peroxide with good stability. The possibility of using the developed sensor in medium containing sodium ions and the increased operational stability constitute advantages in comparison with PB-based amperometric sensors. The usefulness of the methodology was demonstrated by addition-recovery experiments with rainwater samples and values were in the 98.8 to 103% range.

One method using a solid sampling device for the direct determination of Cr and Ni in fresh and used lubricating oils by graphite furnace atomic absorption spectrometry are proposed. The high organic content in the samples was minimized using a digestion step at 400 degrees C in combination with an oxidant mixture 1.0% (v v(-1)) HNO3+15% (v v(-1)) H2O2+0.1% (m v(-1)) Triton X-100 for the in situ digestion. The 3-field mode Zeeman-effect allowed the spectrometer calibration up to 5 ng of Cr and Ni. The quantification limits were 0.86 mu g g(-1) for Cr and 0.82 mg g(-1) for Ni, respectively. The analysis of reference materials showed no statistically significant difference between the recommended values and those obtained by the proposed methods.

Apomorfina é um potente agonista dopaminérgico D1/D2, utilizada no tratamento da Doença de Parkinson. Em maio de 2001, apomorfina HCl foi aprovada para utilização no tratamento da disfunção erétil, aumentando o número de usuários potenciais deste fármaco. Estudos sugerem que apomorfina e outros agonistas dopaminérgicos podem induzir neurotoxicidade mediada por seus derivados de oxidação semiquinonas e quinonas, os quais levam à formação de espécies reativas de oxigênio. Os objetivos do presente estudo foram de avaliar os possíveis efeitos genotóxicos, antimutagênicos, citotóxicos de apomorfina (APO) e de um produto derivado de sua oxidação, 8-oxo-apomorfina-semiquinona (8-OASQ), utilizando o teste Salmonella/microssoma, Mutoxiteste WP2, ensaio Cometa e teste de sensibilidade em Saccharomyces cerevisiae. Em adição, foram avaliados os efeitos de APO e 8-OASQ sobre a memória e o comportamento em ratos (tarefa de esquiva inibitória, comportamento e habituação ao campo aberto) e o comportamento estereotipado em camundongos. Ambos compostos induziram mutações por erro no quadro de leitura em linhagens de S. typhimurium TA97 e TA98, sendo que 8-OASQ foi cerca de duas vezes mais mutagênico que APO, na ausência de S9 mix. Para linhagens que detectam mutágenos oxidantes, 8-OASQ foi mutagênico, enquanto APO foi antimutagênico, inibindo a mutagenicidade induzida por H2O2 e t-BOOH em linhagens de S. typhimurium e derivadas WP2 de E. coli. O S9 mix inibiu todos os efeitos mutagênicos, provavelmente retardando a oxidação de APO ou devido à conjugação de APO e seus produtos de autoxidação, como 8-OASQ, a proteínas do S9. Em testes de sensibilidade com S. cerevisiae, APO foi citotóxica para algumas linhagens apenas nas doses mais altas. Para 8-OASQ este efeito foi dose-depende para todas as linhagens, sendo que as mutantes deficientes em catalase (ctt1), superóxido dismutase (sod1) e yap1 foram as mais sensíveis. APO protegeu as linhagens de S. cerevisiae contra danos oxidativos induzidos por concentrações altas de H2O2 e t-BOOH, enquanto que 8-OASQ aumentou os efeitos pró-oxidantes e induziu respostas adaptativas para aqueles agentes. APO e 8-OASQ induziram efeitos de prejuízo na memória de curta e de longa duração em uma tarefa de esquiva inibitória em ratos. APO, mas não 8-OASQ, prejudicou a habituação a um novo ambiente de forma dose-dependente. Os efeitos de prejuízo de memória não foram atribuídos à redução da nocicepção ou outra alteração inespecífica de comportamento, visto que nem APO e nem 8-OASQ afetaram a reatividade ao choque nas patas e comportamento durante a exploração ao campo aberto. Os resultados sugerem, portanto, que os produtos de oxidação de dopamina ou de agonistas dopaminérgicos podem induzir deficiências cognitivas.APO, mas não 8-OASQ, induziu comportamento estereotipado em camundongos machos CF-1. A falta da indução deste comportamento por 8-OASQ sugere que a autoxidação de APO causa a perda na sua habilidade de ligar-se a receptores dopaminérgicos. Pelo ensaio Cometa, 8-OASQ provocou danos ao DNA do tecido cerebral de camundongos sacrificados 1 h e 3 h, mas não 24 h após sua administração, enquanto que APO induziu um fraco aumento da freqüência de dano ao DNA 3 h após o tratamento. Esses resultados sugerem que ambos APO e 8-OASQ desempenham uma atividade genotóxica no tecido cerebral.

A atividade física moderada regular apresenta uma série de efeitos benéficos para a fisiologia do organismo, particularmente no tocante à atividade do sistema imunológico. O exercício físico promove aumento da resistência a infecções por agentes virais e bacterianos, reduz a incidência de certos tipos de câncer, reduz enormemente os riscos de doenças cardiovasculares agudas e crônicas e melhora a qualidade de vida de uma maneira geral. Contudo, os mecanismos pelos quais o exercício físico regular melhora a performance do sistema imunológico não estão completamente elucidados. Particularmente em relação às alterações imunológicas observadas em sessões agudas de exercício moderado, pouco se sabe. Neste trabalho foi estudado o efeito de sessões agudas de exercício de natação em ratos (1 h, em água aquecida a 30ºC, com sobrepeso de 5% ligado à cauda) sobre a funcionalidade imunológica e inflamatória de macrófagos obtidos a partir de monócitos circulantes destes animais. Os resultados mostraram que o exercício induz uma intensa ativação na capacidade fagocitária (2,4 vezes maior que nos controles P = 0,0041), na produção basal de NO avaliada pelo acúmulo de NO3-, NO2- e NOx- total em macrófagos incubados por 1 h (até 95,5% de aumento P = 0,0220) e na produção de H2O2 estimulada pelo éster de forbol PMA (9,6 vezes maior que nos controles, P = 0,0022). O aumento na capacidade basal de produção parece estar associado à expressão da isoforma induzível da NO sintase (iNOS = NOS-2), já que o exercício provocou uma expressiva indução na expressão da enzima (P = 0,0319). Embora a indução da expressão da iNOS esteja relacionada à ativação da via do fator nuclear κB (NF-κB) o que, por sua vez, está normalmente associado ao estresse oxidativo, vários parâmetros sugerem que a ativação dos monócitos/macrófagos deva ter ocorrido localmente a despeito de qualquer alteração redox sistêmica, já que o exercício não levou a qualquer alteração na quantidade de espécies reativas ao ácido tiobarbitúrico (TBARS) plasmáticas ou no índice de estado redox eritrocitário, avaliado pela relação entre as concentrações intracelulares de dissulfeto de glutationa e glutationa. Também não houve alteração na expressão das proteínas de choque térmico da família das HSP70, o que sugere que o exercício não tenha provocado nenhuma sorte de estresse pronunciado sobre os macrófagos. Somados, os dados sugerem que o exercício agudo moderado possa desempenhar importante papel na ativação de certos mecanismos imunológicos e inflamatórios em monócitos/macrófagos e que algum fator neural ou humoral, que não o estresse oxidativo, possa mediar esta resposta.

Este trabalho relata a síntese de uma série de novos ligantes quirais (+) e (-)-syn-1,3-aminoálcoois derivados do norbornano. Através da reação de transesterificação enzimática com a lípase da Candida rugosa em acetato de vinila do álcool racêmico 7,7-dimetoxi-1,4,5,6-tetraclorobiciclo[2.2.1]heptan-5-en-2-ol, (±)-3, foram obtidos os álcoois quirais (+)-3 e (-)-3 (Esquema 1). Através da reação de redução e descloração destes álcoois com Na0/NH3/etanol foram obtidos os respectivos álcoois (+)-4 e (-)-4 (Esquema 2). Os álcoois quirais (+)-4 e (-)-4 foram utilizados como produtos de partida para a síntese dos 1,3-aminoálcoois quirais (+)-9 e (-)-9 em 5 etapas. Deste modo, a partir destes aminoálcoois (9), foi possível sintetizar 12 novos compostos (Esquema 2), todos inétidos na literatura. Os 1,3-aminoálcoois 10, 11, 13, 14 e 15 foram empregados como catalisadores quirais na adição enantiosseletiva de ZnEt2 ao benzaldeído. Excelentes rendimentos e excessos enantioméricos (até 91%) foram obtidos. A relação entre a configuração absoluta do 1-fenilpropanol com a configuração do carbono ligado ao grupo hidroxila dos ligantes foi estudada e, de acordo, com a enantiosseletividade observada foi sugerido um mecanismo para a reação Os produtos com esqueleto ciclopentila são importantes compostos com potencial atividade biológica, fazendo parte da estrutura de prostaglandinas, agentes antitumorais e inibidores da glicosidase. Portanto, nós decidimos usar o acetato clorado quiral 2 para preparar ciclopentanóides quirais altamente funcionalizados. Para isso, o acetato clorado quiral 2 foi submetido à oxidação usando uma quantidade catalítica de RuCl3 anidro na presença de NaIO4 obtendo-se a dicetona 16 (Esquema 3). A dicetona 16 foi clivada com H2O2 em meio alcalino fornecendo os diácidos 17a e 17b, que foram esterificados in situ com excesso de CH2N2 para fornecer uma mistura do hidroxi e acetoxi diéster 18 e 19, respectivamente. A redução da mistura 18 e 19 ou da mistura 17a e 17b com BH3.THF fornece a lactona 20 com excelentes rendimentos.

Os alcalóides β-carbolínicos possuem uma ampla distribuição, sendo encontrados em várias famílias de plantas, além de estarem presentes na fumaça do cigarro, bebidas alcoólicas, alimentos excessivamente cozidos e em mamíferos. Estes alcalóides são conhecidos por apresentarem várias ações farmacológicas sobre os sistemas nervoso central, muscular e cardiovascular, causando alucinações, tremores, convulsões, hipotensão e bradicardia. Possuem também atividade antioxidante e imunossupressora, além de ligarem-se a receptores de serotonina, dopamina, benzodiazepina; são também inibidores das enzimas monoamino-oxidase-A (MAO-A) e DNA topoisomerases. Os objetivos do presente estudo foram avaliar os possíveis efeitos antioxidantes, antimutagênicos, antigenotóxicos e neurocomportamentais dos alcalóides β-carbolínicos: harmano, harmina, harmol, harmalol e harmalina, utilizando teste de sensibilidade e antimutagênese em Saccharomyces cerevisiae, ensaio cometa em cultura de células de pulmão de hâmster chinês (V79) e avaliação do comportamento, através da tarefa de reconhecimento de novo objeto, em camundongos, respectivamente. Em testes de sensibilidade, todos os alcalóides protegeram as linhagens deficientes em sistemas de defesa antioxidante de S. cerevisiae contra danos induzidos pelos oxidantes H2O2 e paraquat. De maneira geral, os alcalóides β-carbolínicos protegeram mais as células dos danos gerados pelo H2O2 e o aumento da viabilidade foi mais significativo para as linhagens sod1Δ, sod2Δ e sod1Δsod2Δ. Também, possivelmente por uma ação seqüestradora de espécies reativas de oxigênio, os alcalóides β-carbolínicos mostraram forte atividade contra danos no DNA induzidos pelo H2O2, no teste de antimutagênese com a linhagem N123 de S. cerevisae e no teste de antigenotoxicidade, utilizando o ensaio cometa com células V79. As dihidro-β-carbolinas harmalina e harmalol mostraram um efeito mais pronunciado que as β-carbolinas nos ensaios citados acima. Quanto à avaliação dos efeitos neurocomportamentais em camundongos, a administração sistêmica dos alcalóides β-carbolínicos facilitou a formação de memória de curta duração, no caso da harmina, harmol e harmalina e, também a memória de longa duração (harmalina) em camundongos, verificada com uso da tarefa de reconhecimento de novo objeto.

Marine algae are one of the major sources of biologic compounds. In extracellular matrix of these organisms there are sulfated polysaccharides that functions as structural components and provides protection against dehydration. The fraction 1.0 (F1.0) rich in sulfated galactans obtained from red seaweed Hypnea musciformis was physicochemical characterized and evaluated for pharmacologic activity through antioxidant activity, cytotoxic action on erythrocytes, anticoagulant, stimulatory action under antithrombotic heparan sulfate synthesis and their effects on cell proliferation and cycle cell progression. The main components of F1.0 were carbohydrates (49.70 ± 0.10%) and sulfate (44.59 ± 0.015%), presenting phenolic compounds (4.79 ± 0.016%) and low protein contamination (0.92 ± 0.001%). Fraction 1.0 showed polidisperse profile and signs in infrared analysis in 1262, 1074 and 930, 900 and 850 attributed to sulfate esters S=O bond, presence of a 3,6- anidrogalactose C-O bond, non-sulfated β-D-galactose and a C-O-SO4 bond in galactose C4, respectively. The fraction rich in sulfated galactans exhibited strong antioxidant action under lipid peroxidation assay with IC50 of 0.003 mg/mL. Besides the inhibition of hemolysis induced by H2O2 in erythrocytes treated with F1.0, this fraction did not promote significant cytotoxity under erythrocytes membranes. F1.0 exhibited low anticoagulant activity causing moderate direct inhibition of enzimatic activity of thrombin. This fraction promoted stimulation around of 4.6 times on this synthesis of heparan sulfate (HS) by rabbit aortic endothelial cells (RAEC) in culture when was compared with non treated cells. The fraction of this algae displayed antiproliferative action under RAEC cells causing incresing on cell number on S fase, blocking the cycle cell progression. Thus F1.0 presented cytostatic and no cytotoxic action under this cell lineage. These results suggest that F1.0 from H. musciformis have antioxidant potential which is a great effect for a compound used as food and in food industry which could be an alternative to food industry to prevent quality decay of lipid containing food due to lipid peroxidation. These polysaccharides prevent the lipid peroxidation once the fraction in study exhibited strong inhibitory action of this process. Furthermore that F1.0 present strong antithrombotic action promoting the stimulation of antithrombotic HS synthesis by endothelial cells, being important for thrombosis preventing, by its inhibitory action under reactive oxygen species (ROS) in some in vitro methods, being involved in promotion of hypercoagulability state.

The plants are often exposed to variations in environmental conditions that may trigger metabolic disturbances leading to a consequent loss in productivity of crops. These stressful conditions usually induce an accumulation of reactive oxygen species (ROS) in the cell, a condition known how oxidative stress. Among these species, hydrogen peroxide (H2O2) is an important molecule involved in numerous signaling mechanisms. The present study aimed to understand the relationship between the different enzymatic mechanisms of elimination of H2O2 by catalase (CAT) and ascorbate peroxidase (APX) in leaf tissues of seedlings of the species Vigna unguiculata L. Walp, under conditions of oxidative stress induced by application of CAT inhibitor, 3-amino-1,2,4-triazole (3-AT), and H2O2 itself on the roots. Three experiments were conducted. The first experiment was performed applying the compound 3-AT (5 mM) during the time (hours). In the second experiment, seedlings were exposed to different concentrations of H2O2 (2.5, 5.0, 7.5, 10 mM) for 48 h. The third strategy included the pre-treatment with H2O2 (2.5 mM) for 24 h, followed by subsequent treatment with the inhibitor 3-AT and recovery control condition. Treatment with 3-AT causes a strong inhibition of CAT activity in leaf tissues accompanied by an increase of activity of APX. However a decrease in oxidative damage to lipids is not observed as indicated by TBARS. It was observed that activity of APX is directly linked to the content of peroxide. Inductions in the activities of CAT and APX were observed mainly in the seedlings treated with 2.5 mM H2O2. This can be associated with a decrease in oxidative damage to lipids. In contrast, one same tendency was not observed in treatments with higher concentrations of this ROS. These results suggest that the concentration of 2.5 mM H2O2 can induce responses antioxidants later in seedling cowpea. This concentration when applied as pre-treatment for 24 h promoted an induction systems removers CAT and APX, both in activity and in terms of gene expression. However this increment was not observed in the recovered plants and the plants subsequently subjected to 3-AT. Additionally, the pretreatment was not sufficient to attenuate the inhibition of CAT activity and oxidative damage to lipids caused by the subsequent application of this inhibitor. The results showed that the application of 3-AT and H2O2 in the root systems of seedlings of cowpea promote changes in the parameters analyzed in leaf tissues that indicate a direct response to the presence of these factors or systemic signaling mecanisms. H2O2 appears to activate the responses of two antioxidant systems in this study thar does not promote greater protection in case of additional treatment with 3-AT. This demonstrates the importance of the CAT system. In this work, complete results indicate that there is a difference between the signaling and the effects caused by exposure to H2O2 and by treatment with 3-AT

The β-proteobacterium Chromobacterium violaceum is a Gram-negative, free-living, saprophytic and opportunistic pathogen that inhabits tropical and subtropical ecosystems among them, in soil and water of the Amazon. It has great biotechnological potential, and because of this potential, its genome was completely sequenced in 2003. Genome analysis showed that this bacterium has several genes with functions related to the ability to survive under different kinds of environmental stresses. In order to understand the physiological response of C. violaceum under oxidative stress, we applied the tool of shotgun proteomics. Thus, colonies of C. violaceum ATCC 12472 were grown in the presence and absence of 8 mM H2O2 for two hours, total proteins were extracted from bacteria, subjected to SDS-PAGE, stained and hydrolysed. The tryptic peptides generated were subjected to a linear-liquid chromatography (LC) followed by mass spectrometer (LTQ-XL-Orbitrap) to obtain quantitative and qualitative data. A shotgun proteomics allows to compare directly in complex samples, differential expression of proteins and found that in C. Violaceum, 131 proteins are expressed exclusively in the control condition, 177 proteins began to be expressed under oxidative stress and 1175 proteins have expression in both conditions. The results showed that, under the condition of oxidative stress, this bacterium changes its metabolism by increasing the expression of proteins capable of combating oxidative stress and decreasing the expression of proteins related processes bacterial growth and catabolism (transcription, translation, carbon metabolism and fatty acids). A tool with of proteomics as an approach of integrative biology provided an overview of the metabolic pathways involved in the response of C. violaceum to oxidative stress, as well as significantly amplified understanding physiological response to environmental stress. Biochemical and "in silico" assays with the hypothetical ORF CV_0868 found that this is part of an operon. Phylogenetic analysis of superoxide dismutase, protein belonging to the operon also showed that the gene is duplicated in genome of C. violaceum and the second copy was acquired through a horizontal transfer event. Possibly, not only the SOD gene but also all genes comprising this operon were obtained in the same manner. It was concluded that C. violaceum has complex, efficient and versatile mechanisms in oxidative stress response

Sugar esters are substances which possess surfactant, antifungical and bactericidal actions and can be obtained through two renewable sources of raw materials: sugars and vegetable oils. Their excellent biodegradability, allied to lhe fact that they are non toxic, insipid, inodorous, biocompatible, no-ionic, digestible and because they can resist to adverse conditions of temperature, pH and salinity, explain lhe crescent use of these substances in several sections of lhe industry. The objective of this thesis was to synthesize and characterize surfactants and polymers containing sugar branched in their structures, through enzymatic transesterification of vinyl esters and sugars, using alkaline protease from Bacillus subtilis as catalyst, in organic medium (DMF).Three types of sugars were used: L-arabinose, D-glucose and sucrose and two types of vinyl esters: vinyl laurate and vinyl adipate. Aiming to reach high conversions from substrates to products for a possible future large scale industrial production, a serie of variables was optimized, through Design of Experiments (DOE), using Response Surface Methodology (RSM).The investigated variables were: (1) enzyme concentration; (2) molar reason of substrates; (3) water/solvent rale; (4) temperature and (5) time. We obtained six distinct sugar esters: 5-0-lauroyl L-arabinose, 6-0-lauroyl D-glucose, 1'-O-lauroyl sucrose, 5-0-vinyladipoyl L-arabinose, 6-0-vinyladipoyl D-glucose and 1 '-O-vinyladipoyl sucrose, being lhe last three polymerizable. The progress of lhe reaction was monitored by HPLC analysis, through lhe decrease of sugar concentration in comparison to lhe blank. Qualitative analysis by TLC confirmed lhe formation of lhe products. In lhe purification step, two methodologies were adopted: (1) chromatographic column and (2) extraction with hot acetone. The acylation position and lhe chemical structure were determined by 13C-RMN. The polymerization of lhe three vinyl sugar esters was possible, through chemical catalysis, using H2O2 and K2S2O8 as initiators, at 60°C, for 24 hours. IR spectra of lhe monomers and respective polymers were compared revealing lhe disappearance of lhe vinyl group in lhe polymer spectra. The molar weights of lhe polymers were determined by GPC and presented lhe following results: poly (5-0-vinyladipoyl L-arabinose): Mw = 7.2 X 104; PD = 2.48; poly (6-0-vinyladipoyl D-glucose): Mw = 2.7 X 103; PD = 1.75 and poly (1'-O-vinyladipoyl sucrose): Mw = 4.2 X 104; PD = 6.57. The six sugar esters were submitted to superficial tension tests for determination of the critical micelle concentrations (CMC), which varied from 122 to 167 ppm. Finally, a study of applicability of these sugar esters, as lubricants for completion fluids of petroleum wells was' accomplished through comparative analysis of lhe efficiency of these sugar esters, in relation to three commercial lubricants. The products synthesized in this thesis presented equivalent or superior action to lhe tested commercial products

This project describes a methodology optimization that would allow for a more efficient microwave assisted digestion process for petroleum samples. With the possible chance to vary various factors at once to see if any one factor was significant enough in the answers, experimental planning was used. Microwave assisted digestion allows, through the application of potency, an increasing number of collisions between the HNO3 and H2O2 molecules, favoring sample opening for complex matrixes. For this, a 24 factorial experimental planning was used, varying potency, time and the volumes for HNO3 65% and H2O2 30%. To achieve the desired answers, several elements were monitored (C, Cu, Cr, Fe, Ni, Zn and V) through Inductively coupled plasma atomic emission spectroscopy (ICP-OES). With this initial study it was noticed that the HNO3 was not a significant factor for any of the statistical studies for any of the analytes and the other 3 factors and their interactions showed statistical significance. A Box Behnken experimental planning was used taking in consideration 3 factors: H2O2 volume, time (min) and Potency (W), Nitric Acid kept at 4mL for a mass of 0,1g of petroleum. The results were extremely satisfying showing higher efficiency in the digestion process and taking in a responsibility between the answers for each analyte and the carbon monitoring was achieved in the following conditions: 7mL of H2O2, 700 Watts of potency and a reaction time of 7 minutes with 4mL de HNO3 for a mass of 0,1g of petroleum. The optimized digestion process was applied to four different petroleum samples and the analytes determined by ICP-OES

The genus Saccharum belongs to Poaceae family. Sugarcane has become important monocultures in Brazil due to their products: ethanol and sugar. The production may change between different regions from Brazil. This difference is related to soil, climatic conditions and temperature that promotes oxidative stress that may induce an early flowering. The aim of this work was to identify the effects of oxidative stress. In order to analyse this, sugarcane plants were submitted to oxidative stress using hydrogen peroxide. After this treatment, the oxidative stress were analyzed Then, the plant responses were analyzed under different approaches, using morphophysiological, biochemical and molecular tools. Thus, sugarcane plants were grown under controlled conditions and until two months they were subjected first to a hydroponics condition for 24 hours in order to acclimation. After this period, these plants were submitted to oxidative stresse using 0 mM, 10 mM, 20 mM and 30 mM hydrogen peroxide during 8 hours. The histomorphometric analysis allowed us to verify that both root and leaf tissues had a structural changes as it was observed by the increased in cell volume, lignin accumulation in cell walls. Besides, this observation suggested that there was a change in redox balance. Also, it was analyzed the activity of the SOD, CAT and APX enzymes. It was observed an increase in the SOD activity in roots and it was also observed a lipid peroxidation in leaves and roots. Then, in order to identify proteins that were differently expressed in this conditions it was used the proteomic tool either by bidimensional gel or by direct sequencing using the Q-TOF EZI. The results obtained with this approach identified more than 3.000 proteins with the score ranging from 100-5000 ions. Some of the proteins identified were: light Harvesting; oxygenevolving; Thioredoxin; Ftsh-like protein Pftf precusor; Luminal-binding protein; 2 cys peroxiredoxin e Lipoxygenase. All these proteins are involved in oxidative stress response, photsynthetic pathways, and some were classified hypothetical proteins and/or unknown (30% of total). Thus, our data allows us to propose that this treatment induced an oxidative stress and the plant in response changed its physiological process, it made changes in tissue, changed the redox response in order to survival to this new condition

As maiores limitações para o uso do lodo de esgoto em áreas agrícolas são os riscos de contaminação do solo com metais pesados e sua possível transferência para a cadeia alimentar. Objetivou-se, com este estudo, avaliar os teores de Cd, Cr, Pb e Zn no solo, utilizando-se dois métodos de extração (HNO3 + H2O2 + HCl e HClO4 + HF), o acúmulo destes elementos em plantas de milho cultivadas em Latossolo Vermelho eutroférrico no nono ano de aplicação de lodo de esgoto, bem como seus efeitos na produção de matéria seca e na produtividade de grãos. O experimento foi instalado em condições de campo em Jaboticabal-SP, no delineamento em blocos casualizados, com quatro tratamentos e cinco repetições. Os tratamentos corresponderam a doses acumuladas por nove anos consecutivos de lodo de esgoto (45,0; 90,0 e 127,5 t ha-1, base seca) e um tratamento testemunha que recebeu fertilização mineral com base na análise de solo. O lodo de esgoto foi aplicado manualmente e incorporado ao solo com grade a 0,1 m de profundidade antes da semeadura do milho. Os teores médios de Cd no solo não variaram em função das doses de lodo e dos métodos de extração. Por outro lado, os teores médios de Cr, Pb e Zn no solo obtidos pela digestão com HClO4 + HF foram de 72,4; 31,8 e 62,3 %, respectivamente, superiores àqueles encontrados pela digestão que empregou HNO3 + H2O2 + HCl. Contudo, quando se avaliou apenas o efeito das doses de lodo no solo, em ambos os métodos de extração, verificou-se diferença entre os tratamentos apenas para a concentração de Zn no solo. Exceto o Cd, que não foi detectado, de modo geral, os teores e os acúmulos de Cr, Pb e Zn nas partes das plantas de milho aumentaram com as aplicações de lodo de esgoto. Os teores de Cr, Pb e Zn nos grãos, quando detectados, permaneceram abaixo dos limites máximos estabelecidos para o consumo humano conforme a legislação brasileira. A adição de lodo de esgoto e a fertilização mineral, por longo período, apresentaram efeitos semelhantes na produção de matéria seca e na produtividade de grãos de milho.

With water pollution increment at the last years, so many progresses in researches about treatment of contaminated waters have been developed. In wastewaters containing highly toxic organic compounds, which the biological treatment cannot be applied, the Advanced Oxidation Processes (AOP) is an alternative for degradation of nonbiodegradable and toxic organic substances, because theses processes are generation of hydroxyl radical based on, a highly reactivate substance, with ability to degradate practically all classes of organic compounds. In general, the AOP request use of special ultraviolet (UV) lamps into the reactors. These lamps present a high electric power demand, consisting one of the largest problems for the application of these processes in industrial scale. This work involves the development of a new photochemistry reactor composed of 12 low cost black light fluorescent lamps (SYLVANIA, black light, 40 W) as UV radiation source. The studied process was the photo-Fenton system, a combination of ferrous ions, hydrogen peroxide, and UV radiation, it has been employed for the degradation of a synthetic wastewater containing phenol as pollutant model, one of the main pollutants in the petroleum industry. Preliminary experiments were carrier on to estimate operational conditions of the reactor, besides the effects of the intensity of radiation source and lamp distribution into the reactor. Samples were collected during the experiments and analyzed for determining to dissolved organic carbon (DOC) content, using a TOC analyzer Shimadzu 5000A. The High Performance Liquid Chromatography (HPLC) was also used for identification of the cathecol and hydroquinone formed during the degradation process of the phenol. The actinometry indicated 9,06⋅1018 foton⋅s-1 of photons flow, for 12 actived lamps. A factorial experimental design was elaborated which it was possible to evaluate the influence of the reactants concentration (Fe2+ and H2O2) and to determine the most favorable experimental conditions ([Fe2+] = 1,6 mM and [H2O2] = 150,5 mM). It was verified the increase of ferrous ions concentration is favorable to process until reaching a limit when the increase of ferrous ions presents a negative effect. The H2O2 exhibited a positive effect, however, in high concentrations, reaching a maximum ratio degradation.
