In order to investigate the potential of magic angle spinning nuclear magnetic resonance (MAS NMR) in the elucidation of post-mortem metabolism in muscle biopsies, simultaneous H-1 and (31)p MAS NMR measurements were made continuously on postmortem (20 min to 24 h) muscle longissimus samples from rabbits. The animals had either been or not been given adrenaline (0.5 mg kg(-1) 4 h pre-slaughter) to deplete stores of muscle glycogen. The intracellular pH was calculated from H-1 spectra, and the post-mortem rate of formation of lactate was followed and quantified. Comparison of measurements made on muscle samples from rabbits treated with adrenaline with measurements made on muscle samples from untreated' rabbits revealed significant effects of adrenaline treatment on both pH (pH24 h = 6.42 vs. pH24 It = 5.60) and formation of lactate (16 mmol g(-1) vs. 65 mmol g(-1)). The P-31 NMR spectra were used to follow the rate of degradation of ATP and phosphocreatine. The present study clearly shows that MAS NMR has potential for the study of post-mortem energy metabolism.

The influence of a once only administration of a metabolite of vitamin D-3 (HY center dot D-(R)-25-hydroxy vitamin D-3) on myofibrillar meat tenderness in Australian Brahman cattle was studied. Ninety-six Brahman steers of three phenotypes (indo-Brazil, US and US/European) and with two previous hormonal growth promotant (HGP) histories (implanted or not implanted with Compudose((R))) were fed a standard feedlot ration for 70 d. Treatment groups of 24 steers were offered daily 10 g/head HY center dot D-(R) (125 mg 25-hydroxyvitamin D-3) for 6, 4, or 2 d before slaughter. One other group of 24 steers was given the basal diet without HY center dot D-(R). Feed lot performance, blood and muscle samples and carcass quality data were collected at slaughter. Calcium, magnesium, potassium, sodium, iron and Vitamin D-3 metabolites were measured in plasma and longissimus dorsi muscle. Warner-Bratzler (WB) shear force (peak force, initial yield) and other objective meat quality measurements were made on the longissimus dorsi muscle of each steer after ageing for 1, 7 and 14 d post-mortem at 0-2 degrees C. There were no significant effects of HY center dot D-(R) supplements on average daily gain (ADG, 1.28-1.45 kg/d) over the experimental period. HY center dot D-(R) supplements given 6 d prior to slaughter resulted in significantly higher (P < 0.05) initial yield values compared to supplements given 2 d prior to slaughter. Supplementation had no significant effect on meat colour, ultimate pH, sarcomere length, cooking loss, instron compression or peak force. There was a significant treatment (HY center dot D-(R)) by phenotype/HGP interaction for peak force (P = 0.028), in which Indo-Brazil steers without previous HGP treatment responded positively (increased tenderness) to HY center dot D-(R) supplements at 2 d when compared with Indo-Brazil steers previously given HGP. There were no significant effects of treatment on other phenotypes. HY center dot D-(R) supplements did not affect muscle or plasma concentrations of calcium, potassium or sodium, but did significantly decrease plasma magnesium and iron concentrations when given 2 d before slaughter. A restricted maximum likelihood analysis applied to an animal model showed no significant differences (P > 0.05) in pH value of the longissimus dorsi measured at 24 h post-mortem (pH24) between high and low lines of Large White pigs selected over 4 years for post-weaning growth rate on restricted feeding. Genetic and phenotypic correlations between pH24 and production and carcass traits were estimated using all performance testing records combined with the pH24 measurements (5.05–7.02) on slaughtered animals. The estimate of heritability for pH24 was moderate (0.29 ± 0.18). Genetic correlations between pH24 and production or carcass composition traits, except for ultrasonic backfat (UBF), were not significantly different from zero. UBF had a moderate, positive genetic correlation with pH24 (0.24 ± 0.33). These estimates of genetic correlations affirmed that selection for increased growth rate on restricted feeding is likely to result in limited changes in pH24 and pork quality since the selection does not put a high emphasis on reduced fatness.

The objective of this study was to evaluate the effects of the use of cashew bagasse bran (CBB) as food ingredient in qualitative feed restriction programs on the carcass traits, meat quality, organs weight and intestinal morphometry of barrows and gilts. Twenty – four crossbred pigs were used (12 barrows and 12 gilts) with an average initial body weight of 57.93 ± 3.67 kg/LW. The experimental designs was in randomized blocks 3x2 factorial arrangement with three level (0%, 15% e 30% CBB), two genders (barrows and gilts) and four repetition. A total of twenty-four instalments. The treatments were composed of basal diet (BD) formulated with corn, soybean meal and commercial base mix for finishing pigs, being containing different levels of CBB. At the end of the trial period the animals were slaughtered for the evaluation of the meat quality, traits carcass, Absolute Weight (AW) and Relative Weight (RW) of the organs and morphometric study of small intestine fragment. The inclusion of (CBB) in the diets did not affect the traits carcass of gilts, but interfered in the traits carcass of the barrow positively, increasing the yield of meat into cold carcass and reducing the thickness of subcutaneous fat, without affecting the fatty acid profile. However, we observed increased weight of organs and partial volume of absortiva mucosa of gilts. In the comparison between sex was observed a greater liver weight (AW) and (RW), and surface density of absortiva mucosa of barrow. The use of CBB was considered as ingredient to be used in programs of qualitative feed restriction for finishing pigs.

O presente trabalho tem como objetivo testar o efeito do sistema de produção com dieta rica em ácido oleico nas características zootécnicas, de carcaça, físico-químicas e no perfil lipídico dos músculos Longissimus dorsi (LD), Biceps femoris (BF) e Semimembranosus (SM) de suínos de raça Alentejana. Foram utilizados 15 animais, divididos em dois grupos, o grupo IN (n=9) foi mantido em parques individuais abertos ao exterior com 3 m2 e o grupo OUT (n=6) foi mantido numa área de 3 ha com acesso a pastagem. O grupo OUT obteve melhores performances zootécnicas. O rendimento de carcaça foi igual para ambos os grupos, e o rendimento comercial e proporção de peças nobres foi superior nos OUT. Estes obtiveram menores teores de lípidos intramusculares mas maiores teores de PUFA (ácidos gordos polinsaturados) e melhores valores para os rácios PUFA/SFA (ácido gordos saturados) e n-6/n-3. O SM foi menos afetado pelo sistema de produção; Abstract: Effect of the rearing system in the physical-chemical characteristics and the lipid profile of three muscles from Alentejano pigs The present study aims to test the effect of rearing system with oleic acid rich diet on performance, carcass and physical-chemical characteristics, and lipid profile of the muscles Longissimus dorsi (LD), Biceps femoris (BF) and Semimembranosus (SM) from Alentejano pigs. The study used 15 animals, divided into two groups. The Group IN (n = 9) was kept in individual parks open to the exterior with 3 m2, and the OUT (n = 6) was held in an area of 3 ha with access to pasture. The OUT group obtained the best performances. The carcass yield was the same for both groups, meat yield and the proportion of noble parts was superior in the OUT. These obtained lower levels of intramuscular fat but larger PUFA levels (polyunsaturated fatty acids) and best values for the ratios PUFA/SFA (saturated fatty acids) and n-6/n-3. The SM was the least affected by the rearing system

Iron (Fe) is an essential mineral for metabolism and plays a central role in a range of biochemical processes. Therefore, this study aimed to identify differentially expressed (DE) genes and metabolic pathways in Longissimus dorsi (LD) muscle from cattle with divergent iron content, as well as to investigate the likely role of these DE genes in biological processes underlying beef quality parameters. Samples for RNA extraction for sequencing and iron, copper, manganese, and zinc determination were collected from LD muscles at slaughter. Eight Nelore steers, with extreme genomic estimated breeding values for iron content (Fe-GEBV), were selected from a reference population of 373 animals. From the 49 annotated DE genes (FDR<0.05) found between the two groups, 18 were upregulated and 31 down-regulated for the animals in the low Fe-GEBV group. The functional enrichment analyses identified several biological processes, such as lipid transport and metabolism, and cell growth. Lipid metabolism was the main pathway observed in the analysis of metabolic and canonical signaling pathways for the genes identified as DE, including the genes FASN, FABP4, and THRSP, which are functional candidates for beef quality, suggesting reduced lipogenic activities with lower iron content. Our results indicate metabolic pathways that are partially influenced by iron, contributing to a better understanding of its participation in skeletal muscle physiology.

Avaliaram-se os efeitos do fotoperíodo e da castração sobre a composição dos tecidos da paleta e características de qualidade da gordura do lombo e da paleta, de 20 cordeiros mestiços Ideal, distribuídos em esquema fatorial 2 x 2 (dois fotoperíodos - curto (FC), com 12 horas de luz, e longo (FL), com 18 horas de luz, e duas condições sexuais - não castrados (NC) e castrados (C)), com cinco repetições. Os animais foram abatidos aos 37kg de peso corporal. Maior quantidade de gordura total ocorreu nos cordeiros C e mais tecido conjuntivo nos animais NC. A castração influenciou o resíduo mineral fixo (RMF), o extrato etéreo (EE) e a proteína da gordura subcutânea. O efeito da interação entre fotoperíodo longo e castração resultou em aumento do teor de umidade na gordura intermuscular da paleta. A castração elevou o teor de EE e diminuiu o percentual de RMF. Não foi evidenciado efeito do fotoperíodo no EE dos músculos da paleta, e os animais castrados apresentaram gordura intramuscular mais elevada. Os níveis de colesterol da paleta foram mais elevados que os do lombo. Na carne de animais C, verificou-se maior quantidade de ácidos graxos saturados.

O objetivo deste experimento foi comparar as características qualitativas de carcaças de 36 cordeiros terminados em pastagem de coastcross, sendo 13 Corriedale, 14 Bergamácia x Corriedale e 9 Hampshire Down x Corriedale, e 29 cordeiros terminados em confinamento, sendo 10 Corriedale, 11 Bergamácia x Corriedale e 8 Hampshire Down x Corriedale. As médias das variáveis (escala de 1 a 5) condição corporal: 2,70 e 3,13; conformação: 2,65 e 3,35; e cobertura de gordura: 2,43 e 2,95; foram superiores para os cordeiros do confinamento que para cordeiros em pastagem. Na escala de 1 a 3, foram avaliadas a cor da gordura: 1,66 e 1,94; cor da carne: 1,66 e 1,72; e consistência de gordura: 1,62 e 1,83, respectivamente, para os cordeiros em pastagem e em confinamento. A área de olho de lombo foi significativamente (10,21 vs. 9,03 cm²), bem como a espessura de gordura de cobertura: 1,10 vs. 1,70 mm, foi superior para os cordeiros confinados. As médias das porcentagens de músculo no lombo foram de 54,92 e 57,94%; para gordura, 9,29 e 12,95%; e osso, 35,78 e 29,10%, respectivamente, para cordeiros em pastagem e confinamento. Entre os sistemas de terminação, não houve diferenças para porcentagem de proteína (19,71 e 19,10%, respectivamente), mas houve diferença para teor de lipídios, com 5,43% para cordeiros em pastejo e 11,54% para cordeiros confinados. Para as variáveis analisadas não houve diferenças significativas entre os cruzamentos.

Com o objetivo de estudar as características qualitativas da carne de ovinos de diferentes categorias quanto aos músculos dos cortes da carcaça (paleta, lombo e perna), utilizaram-se 18 ovinos ½ Ile de France ½ Ideal (seis cordeiros não-castrados, seis ovelhas de descarte e seis machos adultos castrados). Os animais foram criados em pasto de capim-tifton 85 e receberam suplementação (concentrado). Os cordeiros foram abatidos aos 32 kg, com aproximadamente 5 meses de idade, e as ovelhas e os machos adultos castrados, com 55 kg e aos 60 meses de idade. Não houve diferenças nos valores de pH 45 minutos e pH 24 horas entre as categorias animais e nos músculos dos cortes da carcaça. A idade de abate influenciou a luminosidade da carne de todos os músculos. O teor de vermelho não diferiu entre os animais adultos, mas foi superior aos valores obtidos na carne dos cordeiros. A carne de animais adultos é mais escura que a de cordeiros. As perdas por cocção no músculo Triceps brachii são maiores que no Longissimus lumborum e Semimembranosus. Os valores de pH e a capacidade de retenção de água são similares entre as categorias animais.

RESUMO - Objectivo: As lesões músculo-esqueléticas ligadas ao trabalho (LMELT) são doenças profissionais frequentes. Neste estudo ensaiou-se uma estratégia de diagnóstico do risco e de vigilância da saúde dos trabalhadores numa empresa de abate e desmancha de carne. Métodos: Utilizou-se uma estratégia de obtenção de informação em todos os postos de trabalho e trabalhadores da empresa. Utilizaram-se: (i) adaptação do Questionário Nórdico Músculo-Esquelético, incluindo caracterização da exposição biomecânica; (ii) protocolo de avaliação clínica de LMELT; (iii) filtro RSI e método Strain Index; (iv) instrumentação, como electrogoniometria e sensores de força em postos de risco elevado. Resultados: Identificou-se a presença de sintomas e sinais de LMELT principalmente nos punhos/mãos (n=27) e região lombo-sagrada (n=32), uma importante prevalência de casos relacionados com a actividade de trabalho (30%) e níveis de risco elevados com base nas classificações do Strain Index (n=26 MSDto e n=7 MSEsq). A utilização da instrumentação permitiu obter detalhes da repetitividade, das posturas e dos momentos de aplicação de força, úteis para a intervenção. Conclusões: A prevenção só é possível através da aplicação de programas/estratégias integradas de diagnóstico e gestão do risco de LMELT que sejam eficazes no sentido da intervenção sobre a actividade e as condições de trabalho.--------------------------ABSTRACT – Background: Work-related Musculoskeletal Disorders (WRMSD) are common occupational diseases. The present study aims at examining an integrated perspective of risk assessment and health surveillance at a meatpacking plant. Methods: The strategy adopted was of obtaining information about WRMSDs awareness at all workstations and from all their workers. This was based on: (i) questionnaire application - an adaptation of the Nordic musculoskeletal questionnaire, including a biomechanical item, (ii) WRMSDs clinical protocol (iii) RSI risk filter and Strain Index application, (iv) instrumentation with electrogoniometry and force sensors at previously classified as high risk workstations. Results: WRMSDs signs and symptoms mainly in wrist/hands (n=27) and in lumbar region (n=32) were identified. Results revealed an important prevalence of WRULMSDs associated to meatpacking industry activities (30%) and high risk scores based on Strain Index (n=26 Right UL; n=7 Left UL). Instrumentation showed details of recurrency, of postures and of force, which can be used for intervention.
