A simple and fast method for the determination of Ca, Cu, Fe, Mg, Mn, Se and Zn in bovine semen by quadrupole inductively coupled plasma spectrometry (q-ICP-MS) is described. Prior to analysis, samples (200 µL) were diluted 1:50 in a solution containing 0.01% v/v Triton® X-100 and 0.5% v/v nitric acid and directly analyzed by ICP-MS. The limits of detection of the method are 0.3, 0.03, 0.2, 0.04, 0.04, 0.03 and 0.03 µg L-1 for 44Ca, 63Cu, 57Fe, 24Mg, 64Zn, 82Se and 55Mn, respectively. For purposes of comparison and method validation, four ordinary bovine semen samples were directly analyzed by ICP-MS and by flame atomic absorption spectrometry (FAAS) or graphite furnace atomic absorption spectrometry (GF AAS), with no statistical difference between the techniques at the 95% level when applying the t-test. Then, the proposed method was applied in the determinations of Ca, Cu, Fe, Mg, Mn, Se and Zn in collected samples of bovine semen from different breeds, which are used in reproduction programs and artificial insemination.

The acute direct action of angiotensin-(1-7) [ANG-(1-7)] on bicarbonate reabsorption (JHCO(3)(-)) was evaluated by stationary microperfusions on in vivo middle proximal tubules in rats using H ion-sensitive microelectrodes. The control JHCO(3)(-) is 2.82 ± 0.078 nmol·cm(-2)·s(-1) (50). ANG-(1-7) (10(-12) or 10(-9) M) in luminally perfused tubules decreases JHCO(3)(-) (36 or 60%, respectively), but ANG-(1-7) (10(-6) M) increases it (80%). A779 increases JHCO(3)(-) (30%) and prevents both the inhibitory and the stimulatory effects of ANG-(1-7) on it. S3226 decreases JHCO(3)(-) (45%) and changes the stimulatory effect of ANG-(1-7) to an inhibitory effect (30%) but does not affect the inhibitory effect of ANG-(1-7). Our results indicate that in the basal condition endogenous ANG-(1-7) inhibits JHCO(3)(-) and that the biphasic dose-dependent effect of ANG-(1-7) on JHCO(3)(-) is mediated by the Mas receptors via the Na(+)/H(+) exchanger 3 (NHE3). The control value of intracellular Ca(2+) concentration ([Ca(2+)](i)), as monitored using fura-2 AM, is 101 ± 2 nM (6), and ANG-(1-7) (10(-12), 10(-9), or 10(-6)M) transiently (3 min) increases it (by 151, 102, or 52%, respectively). A779 increases the [Ca(2+)](i) (25%) but impairs the stimulatory effect of all doses of ANG-(1-7) on it. The use of BAPTA or thapsigargin suggests a correlation between the ANG-(1-7) dose-dependent effects on [Ca(2+)](i) and JHCO(3)(-). Therefore, the interaction of the opposing dose-dependent effects of ANG II and ANG-(1-7) on [Ca(2+)](i) and JHCO(3)(-) may represent an physiological regulatory mechanism of extracellular volume and/or pH changes. However, whether [Ca(2+)](i) modification is an important direct mechanism for NHE3 activation by these peptides or is a side effect of other signaling pathways will require additional studies.

Excitonic dynamics in a hybrid dot-well system composed of InAs quantum dots (QDs) and an InGaAs quantum well (QW) is studied by means of femtosecond pump-probe reflection and continuous wave (cw) photoluminescence (PL) spectroscopy. The system is engineered to bring the QW ground exciton state into resonance with the third QD excited state. The resonant tunneling rate is varied by changing the effective barrier thickness between the QD and QW layers. This strongly affects the exciton dynamics in these hybrid structures as compared to isolated QW or QD systems. Optically measured decay times of the coupled system demonstrate dramatically different response to temperature change depending on the strength of the resonant tunneling or coupling strength. This reflects a competition between purely quantum mechanical and thermodynamical processes.

This study is concerned with speciation and fractionation of the rare earth elements (REE) and calcium (Ca) in aqueous solutions. The aim is to investigate the chemical states and physical sizes in which these elements can be present. The REE (including neodymium) and Ca have contrasting geochemical behavior in aqueous solutions. Ca is a major dissolved element, while the REE are trace components and highly reactive with aquatic particles. The major interests of the five papers included in this thesis are the following: · Papers I and V deal with the behavior of neodymium (Nd) and its isotopes in the Kalix River and some marine waters. · The diffusive gradients in thin-films (DGT) method is developed for measuring Ca and Mg in Paper II. · Paper III presents a speciation and fractionation study of Ca in the Kalix and Amazonian rivers. · The rare earth elements and their carrier phases are investigated in the Kalix river in Paper IV. For most elements a detailed study of speciation and fractionation can not be performed using only one method. This is due to the overall heterogeneity of the material, considering both size and chemical composition, which is present in aquatic solutions. During this project the aquatic geochemistry of the REE and Ca has been studied using mainly three methods; cross-flow filtration (CFF), field-flow fractionation (FFF) and diffusive gradients in thin-films (DGT). Field work has to a large part been conducted in the Kalix River, in northern Sweden, which is one of the last pristine river systems in Europe. Some field work has also been conducted in the Baltic Sea and the Arctic Ocean. Results from Amazonian rivers are also presented. These are the main conclusions from this work: The DGT technique works equally well for measuring Ca and Mg in natural waters as previously reported for trace metal. A significant colloidal phase for Ca could be detected in the Kalix River and in different Amazonian rivers. This was concluded independently using both CFF and FFF. Variations in REE signatures in the Kalix River suggests two different pathways for the REE during weathering and release form soil profiles and transport in the river. No significant variation in Nd-isotopic composition could be detected in the Kalix River although concentrations varied by a factor of ~10. This suggests that there is one major source for Nd in the river although different pathways for the REE may exist. A study of Nd in the Kalix River, the Baltic Sea and the Arctic Ocean showed that the isotopic compositions in the diffusible fractions were similar to water samples. However, the relative amount of diffusible Nd increased with salinity, probably reflecting the lower concentration of colloidal and particulate material in marine waters.

Survival during the early life stages of marine species, including nearshore temperate reef fishes, is typically very low, and small changes in mortality rates, due to physiological and environmental conditions, can have marked effects on survival of a cohort and, on a larger scale, on the success of a recruitment season. Moreover, trade offs between larval growth and accumulation of energetic resources prior to settlement are likely to influence growth and survival until this critical period and afterwards. Rockfish recruitment rates are notoriously variable between years and across geographic locations. Monitoring of rates of onshore delivery of pelagic juveniles (defined here as settlement) of two species of nearshore rockfishes, Sebastes caurinus and Sebastes carnatus, was done between 2003-2009 years using artificial collectors placed at San Miguel and Santa Cruz Island, off Southern California coast. I investigated spatiotemporal variation in settlement rate, lipid content, pelagic larval duration and larval growth of the newly settled fishes; I assessed relationships between birth date, larval growth, early life-history characteristics and lipid content at settlement, considering also interspecific differences; finally, I attempt to relate interannual patterns of settlement and of early life history traits to easily accessible, local and regional indices of ocean conditions including in situ ocean temperature and regional upwelling, sea surface temperature (SST) and Chlorophyll-a (Chl-a) concentration. Spatial variations appeared to be of low relevance, while significant interannual differences were detected in settlement rate, pelagic larval duration and larval growth. The amount of lipid content of the newly settled fishes was highly variable in space and time, but did not differ between the two species and did not show any relationships with early life history traits, indicating that no trade off involved these physiological processes or they were masked by high individual variability in different periods of larval life. Significant interspecific differences were found in the timing of parturition and settlement and in larval growth rates, with S. carnatus growing faster and breeding and settling later than S. caurinus. The two species exhibited also different patterns of correlations between larval growth rates and larval duration. S. carnatus larval duration was longer when the growth in the first two weeks post-hatch was faster, while S. caurinus had a shorter larval duration when grew fast in the middle and in the end of larval life, suggesting different larval strategies. Fishes with longer larval durations were longer in size at settlement and exhibited longer planktonic phase in periods of favourable environmental conditions. Ocean conditions had a low explanatory power for interannual variation in early life history traits, but a very high explanatory power for settlement fluctuations, with regional upwelling strength being the principal indicator. Nonetheless, interannual variability in larval duration and growth were related to great phenological changes in upwelling happened during the period of this study and that caused negative consequences at all trophic levels along the California coast. Despite the low explanatory power of the environmental variables used in this study on the variation of larval biological traits, environmental processes were differently related with early life history characteristics analyzed to species, indicating possible species-specific susceptibility to ocean conditions and local environmental adaptation, which should be further investigated. These results have implications for understanding the processes influencing larval and juvenile survival, and consequently recruitment variability, which may be dependent on biological characteristics and environmental conditions.

ZusammenfassungUntersuchungen zur Speicherung, Laserkühlung und Kristallisation von Ca+-Ionen in einer linearen PaulfalleIn dieser Arbeit wurden grundlegende Untersuchungen vonlasergekühlten Ionen in einer linearen Paulfalledurchgeführt. Diese können in drei Bereiche gegliedertwerden: Dynamik der Ionen in der linearen Paulfalle. Hierzuwurden Speicherbereich, parametrische Anregungen undSchwingungsfrequenzen der Ionen in der linearen Paulfalleauch hinsichtlich der Unterschiede zur traditionellendreidimensionalenuntersucht. Über die genaue Messung der Schwingungsfrequenzenkonnte der Abstand zweier Ionen in einem Kristall bis auf 10-4genau bestimmt werden. Diese Genauigkeit der Abstandsbestimmung ist notwendig, um theoretische Vorhersagen zu kooperativen Effektenüberprüfen zu können. Laserkühlung an einem 3-Niveau-SystemsHierzu wurden für Bereiche außerhalb des Lamb-Dicke Regimes Simulationen zur Laserkühlung an einem einzelnen Ion durchgeführt und soweit möglich experimentellüberprüft. Es wurden Kühlzeiten und Temperaturlimits für verschiedene Verstimmungen der Laser bestimmt.Diese stehen zum Teil im Widerspruch mit den bisherigen Annahmen.Die Kenntnis der Laserverstimmungen für die effektivste Lasekühlung sind notwendig, um bei weiterführenden Kühlmethoden eine hinreichendgute Vorkühlung erreichen zu können.Kristallstrukturen in der linearen Paulfalle.Hier wurden verschiedene theoretisch vorhergesagte Kristallstrukturen der Ionen bei abzählbaren Ionenanzahlenfür dreidimensionalen und quasi-zweidimensionaleSpeicherpotentiale mit experimentellen Werten verglichen. Dabei konnten Abweichungen zur Theoriefestgestellt werden. Ferner wurde die durchden Lichtdruck hervorgerufene Trennung von atomaren Zuständenin großen Ionenkristallen beobachtet.

Cyclooxygenase-2/Carbonic anhydrase-IX up-regulation promotes invasive potential and hypoxia survival in colorectal cancer cells Purpose: Cyclooxygenase-2 (COX-2) is a major mediator of inflammation, playing a pivotal role in colorectal carcinogenesis. Hypoxia is an universal hallmark of solid tumour in vivo. This investigation was prompted by the observation that in colorectal cancer cells the expression of COX-2 protein is positively correlated with that of the hypoxia survival gene Carbonic Anhydrase-IX (CA-IX). Experimental Design: Since COX-2 gene expression and activity is increased in hypoxia, and that CA-IX is expressed also in normoxia in colorectal cancer cells, we tested the hypothesis that COX-2 activity in normoxia, as well as in hypoxia may be functionally linked to that of CA-IX gene. We investigated the role of COX-2 and CA-IX in colorectal cancer cell lines. In this regard, we performed RNA interference to knockdown COX-2 gene in vitro and immunohistochemistry to evaluate the protein expression of COX-2 and CA-IX in human colon cancer tissue specimens ex vivo. Results: We found that COX-2, by PGE2 production, controls CA-IX gene expression in an ERK dependent manner. In line with this finding, we also showed that the COX-2 inhibition by a specific short harpin COX-2 RNA (shCOX-2) or by a specific drug (SC-236), down-regulated CA-IX expression in colon cancer cells. We then exposed colon cancer cells to hypoxia stimuli and found that COX-2/CA-IX interplay promoted hypoxia survival. Moreover, we also report that COX-2/CA-IX interplay triggers Matrix Metalloproteinase 2/9 (MMP-2/9) activation and enhances the invasiveness of colorectal cancer cells. Thus given our above observations, we found that CA-IX and COX-2 protein expressions correlate with more aggressive stage colorectal cancer tissues ex vivo. Conclusions: Taken together these data indicate that COX-2/CA-IX interplay promotes an aggressive phenotype (hypoxia survival and invasiveness) which can be modulated in vitro by COX-2 selective inhibition and which may play a role in determining the biological aggressiveness of colorectal tumours. Moreover, in vitro and ex vivo data also suggest that the signatures of inflammation (COX-2) and hypoxia (CA-IX) may be difficult to be disentangled in colon cancer, being both responsible for the up-regulation of the same pathways.

In Experimenten an lasergekühlten, in einer linearen Paulfalle gespeicherten $Ca^+$-Ionen wurde dieLebensdauer des metastabilen $3D_{5/2}$-Niveaus durch Beobachtung von Quantensprüngen einzelner Ionen zu 1100(18)msbestimmt. Systematische Fehler durch quenchende Stöße oder Stark-Mischen durch das Speicherfeld liegen unterhalb dererreichten Genauigkeit. Abweichungen von früheren Messungen konnten durch eine vernachlässigte Abhängigkeit derLebensdauer von der Laserleistung des Rückpumplasers erklärt werden. Das Endergebnis zeigt gute Übereinstimmung mitneueren theoretischen Werten. In weiteren Messungen an zehn Ionen wurde in einigen Messreihen eine deutliche Reduktionder Lebensdauer gegenüber einem einzelnen Ion festgestellt. Dabei wurden mehr koinzidente Zerfälle von zwei und dreiIonen beobachtet als für unabhängige Teilchen zu erwarten. In einem Ionenkristall wurde eine räumliche Trennung atomarer Zustände erreicht. Dabei wurde ein Teil der Ionen einesKristalls aus einigen hundert Ionen in den metastabilen Zustand gepumpt, der von den Kühllasern vollständig entkoppeltist. Durch sympathetische Kühlung werden diese Ionen weiterhin gekühlt und der Kristall schmilzt nicht. Durch denLichtdruck, den die Kühllaser ausgeüben, werden die Ionen nach atomaren Zuständen sortiert, weil die lasergekühltenIonen einen Rückstoß erfahren, die übrigen aber nicht. Für zukünftige Experimente wurden Verbesserungen des experimentellen Aufbaus auf den Weg gebracht. So wurden Methodenund Komponenten für eine verbesserte Frequenzstabilisierung der Diodenlaser entwickelt.

Ionenkäfige und speziell Penningfallen stellen sich in der Atomphysik als außergewöhnliche Werkzeuge heraus. Zum einen bieten diese 'Teilchencontainer' die Möglichkeit atomphysikalische Präzisionsmessungen durchzuführen und zum anderen stellen Penningfallen schwingungsfähige Systeme dar, in welchen nichtlineare dynamische Prozesse an gespeicherten Teilchen untersucht werden können. In einem ersten Teil der Arbeit wurde mit der in der Atomphysik bekannten Methode der optischen Mikrowellen-Doppelresonanz Spektroskopie der elektronische g-Faktor von Ca+ mit einer Genauigkeit von 4*10^{-8} zu gJ=2,00225664(9) bestimmt. g-Faktoren von Elektronen in gebundenen ionischen Systemen sind fundamentale Größen der Atomphysik, die Informationen über die atomare Wellenfunktion des zu untersuchenden Zustandes liefern. In einem zweiten Teil der Arbeit wurde hinsichtlich der Untersuchungen zur nichtlinearen Dynamik von parametrisch angeregten gespeicherten Elektronen beobachtet, dass ab bestimmten kritischen Teilchendichten in der Penningfalle die gespeicherten Elektronen kollektive Eigenschaften manifestieren. Weiterhin wurde bei der Anregung der axialen Eigenbewegung ein Schwellenverhalten der gemessenen Subharmonischen zur 2*omega_z-Resonanz beobachtet. Dieser Schwelleneffekt lässt sich mit der Existenz eines Dämpfungsmechanismus erklären, der auf die Elektronenwolke einwirkt, so dass eine Mindestamplitude der Anregung erforderlich ist, um diese Dämpfung zu überwinden. Durch Bestimmung der charakteristischen Kurven der gedämpften Mathieuschen Differentialgleichung konnte das beobachtete Phänomen theoretisch verstanden werden.

In linearen Paulfallen gespeicherte und lasergekühlte Ionen stellen in weiten Bereichen der Physik ideale Objekte hinsichtlich störungsfreier und präziser Messungen atomarer Übergangsfrequenzen und der gezielten Manipulation von Quantenzuständen dar. Eine Einschränkung dieser optimalen Bedingungen erfolgt durch Heizmechanismen, die aus Abweichungen des Speicherpotentials von der idealen Quadrupolform resultieren. Höhere Potentialordnungen führen zu einer Kopplung der radialen Bewegungsmoden und bei bestimmten Speicherparametern zu nichtlinearen Resonanzen. Hierbei werden die Ionenbahnen durch eine Energieaufnahme aus dem Speicherfeld destabilisiert. Dieses kann zu Linienverbreiterungen, einer Limitierung der Kohärenzzeiten und unter Umständen zu einem Ionenverlust führen. Die systematische Untersuchung dieser Instabilitäten in einer linearen Paulfalle erfolgt durch Spektroskopie an einer kleinen Anzahl lasergekühlter ^40Ca^+ - Ionen. Der experimentell zugängliche Speicherbereich wird mit hoher Auflösung abgetastet. Durch eine eingehende Quantifizierung der Falleneigenschaften werden die nichtlinearen Resonanzen eindeutig den erzeugenden Potentialtermen zugeordnet. Die Resonanzlinien zeigen eine charakteristische Aufspaltung, deren Größe vom angelegten Axialpotential bestimmt wird. Diese zusätzliche Kopplung der Radialbewegung an die Axialbewegung führt zu einer modifizierten Resonanzbedingung. Nichtlineare Resonanzen treten massenspezifisch auf. Da eine präzise Kontrolle der Axialpotentiale sehr einfach ist, könnten die beobachteten radial-axial koppelnden Resonanzen eine Anwendung in der Massenspektrometrie finden.

Materials that can mold the flow of elastic waves of certain energy in certain directions are called phononic materials. The present thesis deals essentially with such phononic systems, which are structured in the mesoscale (<1 µm), and with their individual components. Such systems show interesting phononic properties in the hypersonic region, i.e., at frequencies in the GHz range. It is shown that colloidal systems are excellent model systems for the realization of such phononic materials. Therefore, different structures and particle architectures are investigated by Brillouin light scattering, the inelastic scattering of light by phonons.rnThe experimental part of this work is divided into three chapters: Chapter 4 is concerned with the localized mechanical waves in the individual spherical colloidal particles, i.e., with their resonance- or eigenvibrations. The investigation of these vibrations with regard to the environment of the particles, their chemical composition, and the influence of temperature on nanoscopically structured colloids allows novel insights into the physical properties of colloids at small length scales. Furthermore, some general questions concerning light scattering on such systems, in dispute so far, are convincingly addressed.rnChapter 5 is a study of the traveling of mechanical waves in colloidal systems, consisting of ordered and disordered colloids in liquid or elastic matrix. Such systems show acoustic band gaps, which can be explained geometrically (Bragg gap) or by the interaction of the acoustic band with the eigenvibrations of the individual spheres (hybridization gap).rnWhile the latter has no analogue in photonics, the presence of strong phonon scatterers, when a large elastic mismatch between the composite components exists, can largely impact phonon propagation in analogy to strong multiple light scattering systems. The former is exemplified in silica based phononic structures that opens the door to new ways of sound propagation manipulation.rnChapter 6 describes the first measurement of the elastic moduli in newly fabricated by physical vapor deposition so-called ‘stable organic glasses’. rnIn brief, this thesis explores novel phenomena in colloid-based hypersonic phononic structures, utilizing a versatile microfabrication technique along with different colloid architectures provided by material science, and applying a non-destructive optical experimental tool to record dispersion diagrams.rn

In this work self-assembling model systems in aqueous solution were studied. The systems contained charged polymers, polyelectrolytes, that were combined with oppositely charged counterions to build up supramolecular structures. With imaging, scattering and spectroscopic techniques it was investigated how the structure of building units influences the structure of their assemblies. Polyelectrolytes with different chemical structure, molecular weight and morphology were investigated. In addition to linear polyelectrolytes, semi-flexible cylindrical bottle-brush polymers that possess a defined cross-section and a relatively high persistence along the backbone were studied. The polyelectrolytes were combined with structural organic counterions having charge numbers one to four. Especially the self-assembly of polyelectrolytes with different tetravalent water-soluble porphyrins was studied. Porphyrins have a rigid aromatic structure that has a structural effect on their self-assembly behavior and through which porphyrins are capable of self-aggregation via π-π interaction. The main focus of the thesis is the self-assembly of cylindrical bottle-brush polyelectrolytes with tetravalent porphyrins. It was shown that the addition of porphyrins to oppositely charged brush molecules induces a hierarchical formation of stable nanoscale brush-porphyrin networks. The networks can be disconnected by addition of salt and single porphyrin-decoratedrncylindrical brush polymers are obtained. These two new morphologies, brush-porphyrin networks and porphyrin-decorated brush polymers, may have potential as functional materials with interesting mechanical and optical properties.

Für viele Anwendungen von Nanomaterialien sind maßgeschneiderte Produkte wün-schenswert, weswegen ein tiefgreifendes und genaues Wissen der Reaktionsabläufe, die zu diesen Produkten führen, unabdingbar ist. Um dies im Fall von SnO2 zu erreichen, behandelt diese Arbeit die kontrollierte Synthese und genaue Charakterisierung von Nanopartikeln von Zinn(IV) Oxid.
