Linhagens avançadas do programa de melhoramento do tomateiro (Lycopersicon esculentum) do IAC foram avaliadas em condições de campo em Campinas (SP) para resistência a tospovírus e a potyvírus, nos anos agrícolas 2002/2003 e 2003/2004, respectivamente. No primeiro ano, a única espécie de tospovírus que ocorreu na área experimental foi Tomato chlorotic spot virus (TCSV). As sete linhagens do grupo IAC exibiram baixa porcentagem de plantas sintomáticas em duas avaliações, com médias abaixo de 28%; as cultivares testadas mostraram-se altamente suscetíveis, com médias acima de 85%, à exceção de 'Franco', que apresentou cerca de 55% de infecção. No segundo experimento, conduzido em 2003/2004, dez linhagens do grupo IAC foram comparadas com cinco cultivares de polinização aberta e híbridos F1, além do acesso LA-444-1 de L. peruvianum. Nesse experimento, por meio de testes biológicos e sorológicos, verificou-se ocorrência generalizada de Potato virus Y (PVY). Foi determinado o percentual de plantas com sintomas e avaliada a intensidade dos sintomas mediante uso de escala de notas. Com base nos dois critérios, verificou-se que LA-444-1 apresenta alta resistência a PVY, que 'Tyrade' exibe comportamento intermediário, enquanto todos os demais genótipos demonstram alta suscetibilidade ao vírus. O comportamento dos genótipos avaliados neste trabalho mostra a necessidade de se considerar, nos programas de melhoramento do tomateiro, a introgressão de fatores de resistência não só a vírus de importância atual nas regiões produtoras, como geminivírus, mas também a outros vírus potencialmente nocivos à cultura, como tospovírus e potyvírus.

The black spot of citrus (Citrus sp.) is caused by Guignardia citricarpa with ascospore production depending on temperature, leaf wetness, and rainfall. The number of ascospores produced was monitored using a spore trap and climatic factors were recorded using an automated meteorological station of 'Natal' and 'Valencia' sweet orange (Citrus sinensis) orchards in Mogi Guaçu in the state of São Paulo, Brazil, from November 2000 to March 2001. The fruits were bagged to prevent infection and the bags removed from different sets of fruit for one week during each of the 18 weeks of the season in both orchards. Ascospores were produced during the entire experimental period, from spring through summer, primarily after rain events. In both orchards, ascospore production reached a peak in January and February. Ascospore production was related to leaf wetness only in the Natal orange orchard but was not related to total rainfall or temperature in either orchard. Disease was most severe on fruit exposed the 7th, 8th, and 13th weeks after beginning the experiment in both cultivars as well as after the 16th week for 'Natal'. There was a strong relationship between disease severity and total rainfall for both orchards and a weak correlation between temperature and severity in the 'Natal' block only. There was no relationship between severity and leaf wetness or ascospore numbers.

A qualitative spot-test and tandem quantitative analysis of dipyrone in the bulk drug and in pharmaceutical preparations is proposed. The formation of a reddish-violet color indicates a positive result. In sequence a quantitative procedure can be performed in the same flask. The quantitative results obtained were statistically compared with those obtained with the method indicated by the Brazilian Pharmacopoeia, using the Student's t and the F tests. Considering the concentration in a 100 µL aliquot, the qualitative visual limit of detection is about 5×10-6 g; instrumental LOD ≅ 1.4×10-4 mol L-1 ; LOQ ≅ 4.5×10-4 mol L-1.

Data available in the literature were used to develop a warning system for bean angular leaf spot and anthracnose, caused by Phaeoisariopsis griseola and Colletotrichum lindemuthianum, respectively. The model is based on favorable environmental conditions for the infectious process such as continuous leaf wetness duration and mean air temperature during this subphase of the pathogen-host relationship cycle. Equations published by DALLA PRIA (1977) showing the interactions of those two factors on the disease severity were used. Excell spreadsheet was used to calculate the leaf wetness period needed to cause different infection probabilities at different temperature ranges. These data were employed to elaborate critical period tables used to program a computerized electronic device that records leaf wetness duration and mean temperature and automatically shows the daily disease severity value (DDSV) for each disease. The model should be validated in field experiments under natural infection for which the daily disease severity sum (DDSS) should be identified as a criterion to indicate the beginning and the interval of fungicide applications to control both diseases.

In Brazil, Colletotrichum gloeosporioides is associated with a complex of symptoms in coffee culture. Although this pathogen had its pathogenesis observed and identified, its importance has still been questioned due to its several endophytic forms, raising doubts as to the real importance of the pathosystem. The aim of this study was to demonstrate, by using an isolate transformed with the gene gfp, the infection and colonization capability of C. gloeosporioides in coffee seedlings. After the fourth day of inoculation, manifestation of symptoms as punctual necrosis could be observed, which progressed during the evaluation period, culminating in the death of seedlings. Epifluorescence microscopy confirmed the presence of the pathogen in the seedlings, as well as the visualization of internal colonization of tissues, acervulus formation and conidium production, confirming that it was responsible for the observed symptoms.

ABSTRACT In the present study, the influence of temperature (15, 20, 25, 30 and 35°C) and leaf wetness period (6, 12, 24 and 48 hours) on the severity of Cercospora leaf spot of beet, caused by Cercospora beticola, was studied under controlled conditions. Lesion density was influenced by temperature and leaf wetness duration (P<0.05). Data were subjected to nonlinear regression analysis. The generalized beta function was used for fitting the disease severity and temperature data, while a logistic function was chosen to represent the effect of leaf wetness on the severity of Cercospora leaf spot. The response surface resultant of the product of the two functions was expressed as ES = 0.0001105 * (((x-8)2.294387) * ((36-x)0.955017)) * (0.39219/(1+25.93072 * exp (-0.16704*y))), where: ES represents the estimated severity value (0.1); x, the temperature (ºC) and y, the leaf wetness duration (hours). This model should be validated under field conditions to assess its use as a computational forecast system for Cercospora leaf spot of beet.

High-throughput screening of cellular effects of RNA interference (RNAi) libraries is now being increasingly applied to explore the role of genes in specific cell biological processes and disease states. However, the technology is still limited to specialty laboratories, due to the requirements for robotic infrastructure, access to expensive reagent libraries, expertise in high-throughput screening assay development, standardization, data analysis and applications. In the future, alternative screening platforms will be required to expand functional large-scale experiments to include more RNAi constructs, allow combinatorial loss-of-function analyses (e.g. genegene or gene-drug interaction), gain-of-function screens, multi-parametric phenotypic readouts or comparative analysis of many different cell types. Such comprehensive perturbation of gene networks in cells will require a major increase in the flexibility of the screening platforms, throughput and reduction of costs. As an alternative for the conventional multi-well based high-throughput screening -platforms, here the development of a novel cell spot microarray method for production of high density siRNA reverse transfection arrays is described. The cell spot microarray platform is distinguished from the majority of other transfection cell microarray techniques by the spatially confined array layout that allow highly parallel screening of large-scale RNAi reagent libraries with assays otherwise difficult or not applicable to high-throughput screening. This study depicts the development of the cell spot microarray method along with biological application examples of high-content immunofluorescence and phenotype based cancer cell biological analyses focusing on the regulation of prostate cancer cell growth, maintenance of genomic integrity in breast cancer cells, and functional analysis of integrin protein-protein interactions in situ.

The search for low subjectivity area estimates has increased the use of remote sensing for agricultural monitoring and crop yield prediction, leading to more flexibility in data acquisition and lower costs comparing to traditional methods such as census and surveys. Low spatial resolution satellite images with higher frequency in image acquisition have shown to be adequate for cropland mapping and monitoring in large areas. The main goal of this study was to map the Summer crops in the State of Paraná, Brazil, using 10-day composition of NDVI SPOT Vegetation data for 2005/2006, 2006/2007 and 2007/2008 cropping seasons. For this, a supervised digital classification method with Parallelepiped algorithm in multitemporal RGB image composites was used, in order to generate masks of Summer cultures for each 10-day composition. Accuracy assessment was performed using Kappa index, overall accuracy and Willmott's concordance index, resulting in good levels of accuracy. This methodology allowed the accomplishment, with free and low resolution data, of the mapping of Summer cultures at State level.

Diplomityön tavoitteena oli selvittää etäluentamittareiden mahdollistamia tuotteita ja palveluita sekä niiden toteutettavuutta Mäntsälän Sähkö Oy:ssä. Työssä perehdytään yksityisasiakkaille suunnattuihin palveluihin, joita mittaritekniikka mahdollistaa. Mittarit tulee olla asennettuna 80%:lle verkkoyhtiön sähkönkäyttöpaikoista vuoteen 2014 mennessä. Mäntsälän Sähköllä mittareiden vaihtoprojektin alkamisen vuoksi haluttiin selvittää mahdollisten uusien tuotteiden toteutettavuutta. Työssä esitellään Mäntsälän Sähkö Oy:lle tulevan mittarijärjestelmän. Mittarijärjestelmä käyttää eri tietoliikennetapoja, jotka ovat GPRS, RS485 ja radio. Työ sisältää internet-paneelina suoritetun asiakaskyselyn, jossa selvitettiin asiakkaiden tarpeita ja mielipiteitä mahdollisten uusien tuotteiden osalta. Työssä käsiteltäviä tuotteita ovat kuormanohjaus, kysynnän hintajousto, spot-hinnoittelu, laskutus sekä raportointipalvelu. Kulutuksen hintajousto, kuormanohjaus ja spot-hinnoittelu toimivat toisiaan tukevina tuotteina. Tasasuuruista laskutusta voisi tarjota toteutuneen kulutuksen mukaisen laskutuksen ohessa lisäpalveluna. Raportointipalvelu internetissä on hyvä tapa toimittaa asiakkaalle lain velvoittamat tiedot.
