Herein, we report results of calculations based on density functional theory (BP86/TZVP) of a set of isatin-Schiff base copper(II) and related complexes, 1-12, that have shown significant pro-apoptotic activity toward diverse tumor cells. The interaction of the copper(II) cation with different ligands has been investigated at the same level of theory. The strength and character of the Cu(II)-L bonding was characterized by metal-ligand bond lengths, vibrational frequencies, binding energies, ligand deformation energies, and natural population analysis. The metal-ligand bonding situation was also characterized by using two complementary topological approaches, the quantum theory of atoms-in-molecules (QTAIM) and the electron localization function (ELF). The calculated electronic g-tensor and hyperfine coupling constants present significant agreement with the EPR experimental data. The calculated parameters pointed to complex 10 as the most stable among the isatin-Schiff base copper(II) species, in good agreement with experimental data that indicate this complex as the most reactive in the series. 2-(Diphenylphosphinomethyl)aniline. H2L1, reacts with [RuCl2(PPh3)(3)] to yield the monomeric complexes [RuCl2(H2L1)(PPh3)(CH3CN)], [RuCl2(H2L1)(2)]and the chloro-bridged dimer [(H2L1)(PPh3)Ru(mu-Cl)(2)Ru(PPh3) (H2L1)] depending on the conditions applied. Exclusively the monochelate [RuCl2 (H2L1)(dmso)(2)] is formed during reactions of H2L1 with [RuCl2(dmso)(4)]. H2L1 acts as a neutral, bidentate ligand in all complexes. Schiff base ligand: N,N'-bis(1-phenylethylidene)ethane-1,2-diamine (L), was derived from acetophenone and ethylenediamine by condensation and its complexes (1-5) were prepared with Pb2+, Ni2+, Co2+, Cu2+ and Cd2+ metal ions. Their structures were characterized by FAB-MS, IR spectra, elemental analyses and molar conductance. The octahedral geometry of the complexes was proposed by electronic spectra and magnetic moment data. The conductivity data showed that the complexes have non-electrolytic nature. The complexes (1-5) have higher in vitro antimicrobial activity than the Schiff base ligand (L). In the nuclease activity, the complexes cleave DNA as compared to control DNA in the presence of H2O2.

Copper complexes with fluorinated beta-diketones were synthesized and characterized in terms of lipophilicity and peroxide-assisted oxidation of dihydrorhodamine as an indicator of redox activity. The biological activity of the complexes was tested against promastigotes of Leishmania amazonensis. Inhibition of trypanosomatid-specific trypanothione reductase was also tested. It was found that the highly lipophilic and redox-active bis(trifluoroacetylacetonate) derivative had increased toxicity towards promastigotes. These results indicate that it is possible to modulate the activity of metallodrugs based on redox-active metals through the appropriate choice of lipophilic chelators in order to design new antileishmanials. Further work will be necessary to improve selectivity of these compounds against the parasite.

Two novel dinuclear complexes involving the antihypertensive drug valsartan and copper(II) ion have been prepared in water and DMSO. The complex compositions were determined as: [Cu(vals)(H(2)O)(3)](2)center dot 6H(2)O and [Cu(vals)(H(2)O)(2)DMSO](2)center dot 2H(2)O. They were thoroughly characterized by elemental and thermal analysis, spectrophotometric titrations and UV-visible, diffuse reflectance, FTIR, Raman and EPR spectroscopies. No effect of the ligand on two tested osteoblastic cell lines in culture (one normal MOT3E1 and one tumoral UMR106) was observed in concentrations up to 100 mu M. Higher concentrations of Valsartan are required to induce cytotoxicity in both cell lines. The antiproliferative effect of the tested complex ([Cu(vals) (H(2)O)(3)](2)center dot 6H(2)O) in a dose-response manner, was higher in the UMR106 osteoblastic cell line than that of the MC3T3E1 normal line at concentrations >= 100 mu M. Die biologische Stickstofffixierung durch Molybdän-haltige Nitrogenasen sowie die Erforschung des zugrundeliegenden komplexen Mechanismus (N2-Aktivierung an Metall-Zentren, 6-fache Protonierung und Reduktion, N–N Bindungsspaltung unter Bildung von Ammoniak) ist von erheblichem Interesse. Insbesondere Molybdän-Komplexe wurden bereits erfolgreich als Modellverbindungen für die Untersuchung elementarer Einzelschritte der N2-Aktivierung eingesetzt. Durch die Verwendung von Triamidoamin-Liganden ist es Schrock et al. sogar gelungen mehrere Katalysezyklen zu durchlaufen und einen Mechanismus zu formulieren. Trotz der sterisch anspruchsvollen Substituenten in den Schrock-Komplexen ist die Umsatzrate dieses homogenen Katalysators, aufgrund Komplex-Deaktivierung infolge intermolekularer Reaktionen wie Dimerisierung und Disproportionierung, limitiert. In der vorliegenden Arbeit wurden einige dieser Herausforderungen angegangen und die aktiven Spezies auf einer Festphase immobilisiert, um intermolekulare Reaktionen durch räumliche Isolierung der Komplexe zu unterdrücken.rnEin Polymer-verankertes Analogon des Schrock Nitrido-Molybdän(VI)-Komplexes wurde auf einem neuen Reaktionsweg synthetisiert. Dieser beinhaltet nur einen einzigen Reaktionsschritt, um die funktionelle Gruppe „MoN“ einzuführen. Protonierung des immobilisierten Nitrido-Molybdän(VI)-Komplexes LMoVIN (L = Polymer-verankerter Triamidoamin-Ligand) mit 2,6-Lutidinium liefert den entsprechenden Imido-Molybdän(VI)-Komplex. Durch anschließende Ein-Elektronen-Reduktion mit Cobaltocen wird der Polymer-angebundene Imido-Molybdän(V)-Komplex erhalten, bewiesen durch EPR-Spektroskopie (g1,2,3 = 1.989, 1.929, 1.902). Durch die Immobilisierung und die effektive räumliche Separation der Reaktionszentren auf der Festphase werden bimolekulare Nebenreaktionen, die oft in homogenen Systemen auftreten, unterdrückt. Dies ermöglicht zum ersten Mal die Darstellung des Imido-Molybdän(V)-Intermediates des Schrock-Zyklus.rnEPR-Spektren des als Spin-Label eingeführten immobilisierten Nitrato-Kupfer(II)-Komplexes wurden unter verschiedenen Bedingungen (Lösungsmittel, Temperatur) aufgenommen, wobei sich eine starke Abhängigkeit zwischen der Zugänglichkeit und Reaktivität der immobilisierten Reaktionszentren und der Art des Lösungsmittels zeigte. Somit wurde die Reaktivität von LMoVIN gegenüber Protonen und Elektronen, welches zur Bildung von NH3 führt, unter Verwendung verschiedener Lösungsmittel untersucht und optimiert. Innerhalb des kugelförmigen Polymers verläuft die Protonierung und Reduktion von LMoVIN stufenweise. Aktive Zentren, die sich an der „äußeren Schale“ des Polymers befinden, sind gut zugänglich und reagieren schnell nach H+/e− Zugabe. Aktive Zentren im „Inneren des Polymers“ hingegen sind schlechter zugänglich und zeigen langsame diffusions-kontrollierte Reaktionen, wobei drei H+/e− Schritte gefolgt von einer Ligandenaustausch-Reaktion erforderlich sind, um NH3 freizusetzen: LMoVIN  LMoVNH  LMoIVNH2  LMoIIINH3 und anschließender Ligandenaustausch führt zur Freisetzung von NH3.rnIn einem weiteren Projekt wurde der Bis(ddpd)-Kupfer(II)-Komplex EPR-spektroskopisch in Hinblick auf Jahn−Teller-Verzerrung und -Dynamik untersucht. Dabei wurden die EPR-Spektren bei variabler Temperatur (70−293 K) aufgenommen. Im Festkörperspektrum bei T < 100 K erscheint der Kupfer(II)-Komplex als gestreckter Oktaeder, wohingegen das EPR-Spektrum bei höheren Temperaturen g-Werte aufzeigt, die einer pseudo-gestauchten oktaedrischen Kupfer(II)-Spezies zuzuordnen sind. Diese Tatsache wird einem intramolekularen dynamischen Jahn−Teller Phänomen zugeschrieben, welcher bei 100 K eingefroren wird.

There is an increasing demand for novel metal-based complexes with biologically relevant molecules in technology and medicine. Three new Cu(II) coordination compounds with antifungal agent isoconazole (L), namely mononuclear complexes CuCl2(L)(2) (1), and Cu(O2CMe)(2)(L)(2)center dot 2H(2)O (2) and coordination polymer Cu(pht)(L)(2)(n) (3) (where H(2)pht - o-phthalic acid) were synthesized and characterized by IR spectroscopy, thermogravimetric analysis and X-ray crystallography. X-ray analysis showed that in all complexes, the isoconazole is coordinated to Cu(II) centres by a N atom of the imidazole fragment. In complex I, the square-planar environment of Cu(II) atoms is completed by two N atoms of isoconazole and two chloride ligands, whereas the Cu(II) atoms are coordinated by two N atoms from two isoconazole ligands and two O atoms from the different carboxylate residues: acetate in 2 and phthalate in 3. The formation of an infinite chain through the bridging phthalate ligand is observed in 3. The biosynthetic ability of micromycetes Aspergillus niger CNMN FD 10 in the presence of the prepared complexes 1-3 as well as the antifungal drug isoconazole were studied. Complexes 2 and 3 accelerate the biosynthesis of enzymes (beta-glucosidase, xylanase and endoglucanase) by this fungus. Moreover, a simplified and improved method for the preparation of isoconazole nitrate was developed.

The synthesis of three bis[(tert-butoxy)carbonyl]-protected (tetramine)dichloroplatinum complexes 2a – c of formula cis-[PtCl2(LL)] and of their cationic deprotected analogs 3a – c and their evaluation with respect to in vitro cytotoxicity, intramolecular stability, DNA binding, and cellular uptake is reported. The synthesis comprises the complexation of K2[PtCl4] with di-N-protected tetramines 1a – c to give 2a – c and subsequent acidolysis, yielding 3a – c. The cytotoxicity of the complexes is in direct relation to the length of the polyamine. Complexes 3a – c display a significant higher affinity for CT DNA as well as for cellular DNA in A2780 cells than cisplatin.

Isolated subcomplexes of photosystem II from spinach (CP47RC), composed of D1, D2, cytochrome b559, CP47, and a number of hydrophobic small subunits but devoid of CP43 and the extrinsic proteins of the oxygen-evolving complex, were shown to reconstitute the Mn4Ca1Clx cluster of the water-splitting system and to evolve oxygen. The photoactivation process in CP47RC dimers proceeds by the same two-step mechanism as observed in PSII membranes and exhibits the same stoichiometry for Mn2+, but with a 10-fold lower affinity for Ca2+ and an increased susceptibility to photodamage. After the lower Ca2+ affinity and the 10-fold smaller absorption cross-section for photons in CP47 dimers is taken into account, the intrinsic rate constant for the rate-limiting calcium-dependent dark step is indistinguishable for the two systems. The monomeric form of CP47RC also showed capacity to photoactivate and catalyze water oxidation, but with lower activity than the dimeric form and increased susceptibility to photodamage. After optimization of the various parameters affecting the photoactivation process in dimeric CP47RC subcores, 18% of the complexes were functionally reconstituted and the quantum efficiency for oxygen production by reactivated centers approached 96% of that observed for reconstituted photosystem II-enriched membranes.
