Botryosphaeran, a new exopolysaccharide from the endophytic fungus Botryosphaeria rhodina MAMB-05, and algal laminarin were hydrolyzed by partially-fractionated enzymes of the beta-glucanolytic complex from Trichoderma harzianum Rifai. beta-Glucanase fractions (F-I and F-II) separated by gel permeation chromatography presented different modes of attack on botryosphaeran and laminarin. Botryosphaeran was hydrolyzed to the extent of 66% (F-I) and 98% (F-II) within 30 min, and its main hydrolysis products were gluco-oligosaccharides of DP >= 4, with lesser amounts of glucose, di- and tri-saccharides. The action of enzyme fractions I and II on laminarin resulted in 15% conversion to glucose, while the percentage of saccharification was radically different (70% for F-I and 25% for F-II). The different product arrays within the polysaccharide hydrolysates can be explained by the difference in the enzymes' specificities within each enzyme fraction, and the molecular structures of the polysaccharides and their complexity.

The present study was aimed at verifying tocopherols losses and oxidative stability changes in vegetable oils used in discontinuous frying. The frying of frozen cassava chips was carried out in a household electric frying pan, where the oil was heated to a temperature of 180 degrees C for 25 hours, with fresh oil replacement. The results obtained from the analytical determinations were submitted to variance analysis, in a factorial scheme, using a completely randomized design, making it possible to determine the influence of the type of oil and frying times on changes in the oil. The data show that the smallest changes occur in palm oil, which is more saturated. For sunflower and cottonseed oils, which are more unsaturated, there was a clear decrease in both tocopherol concentration and oxidative stability.

Erbium L-3-edge extended x-ray absorption fine structure (EXAFS) measurements were performed on rare earth doped fluorosilicate and fluoroborate glasses and glass ceramics. The well known nucleating effects of erbium ions for the crystallization of cubic lead fluoride (based on x-ray diffraction measurements) and the fact that the rare earth ions are present in the crystalline phase (as indicated by Er3+ emission spectra) seem in contradiction with the present EXAFS analysis, which indicates a lack of medium range structural ordering around the Er3+ ions and suggests that the lead fluoride crystallization does not occur in the nearest neighbor distance of the rare earth ion. Molecular dynamics simulations of the devitrification process of a lead fluoride glass doped with Er3+ ions were performed, and results indicate that Er3+ ions lower the devitrification temperature of PbF2, in good agreement with the experimental results. The genuine role of Er3+ ions in the devitrification process of PbF2 has been investigated. Although Er3+ ions could indeed act as seeds for crystallization, as experiments suggest, molecular dynamics simulation results corroborate the experimental EXAFS observation that the devitrification does not occur at its nearest neighbor distance. Avaliou-se o efeito da adição do fubá de milho no mosto de xarope de cana e o tratamento ácido do pé-de-cuba sobre a microbiota do processo fermentativo, acidez do vinho, grau alcoólico, rendimento e composição da cachaça. O delineamento experimental utilizado foi o de blocos casualizados, no esquema fatorial 2x3 e cinco repetições. A metodologia empregada e as análises foram as recomendadas pelo setor aguardenteiro. Os resultados permitiram concluir que a adição do ácido sulfúrico no pé-de-cuba transferiu a acidez para o vinho, não influenciando na viabilidade das leveduras, rendimento e composição da cachaça. Por outro lado, a acidificação do meio controlou as bactérias láticas no pé-de-cuba. A adição do fubá aumentou a concentração de bactérias lácticas ao final do processo fermentativo e dos álcoois homólogos superiores na cachaça, particularmente, os álcoois propílico e isobutílico.

Cytophotometric observation of corpora allata from workers and queens of A. mellifera showed significant variations in DNA content from the 2nd to the 4th instar of both castes, but the differences between castes are present only in the 2nd instar.The corpora allata are characterized by the presence of diploid, as well as polyploid cells, distributed in classes that are well established since the earliest developmental stages. However the frequency of cell nuclei in the higher classes of ploidy increases with development. This occurrence is responsible for the increase of the average DNA content in older larvae.

MCM-41 material was synthesized starting from hydrogel containing colloidal fumed silica, sodium silicate, cetyltetramethylammonium bromide (CTMABr) as surfactant, and distilled water as solvent. These reactants were mixed to obtain a gel with the following composition: 4SiO(2):1Na(2)O:1CTMABr:200H(2)O. The hydrogel with pH=14 was hydrothermally treated at 100 degreesC, for 4 days. Each day, the pH was measured, and then adjusted to 9.5-10 by using 30% acetic acid solution. Thermogravimetry was the main technique, which was used to monitor the participation of the surfactant on the MCM-41 nanophase, being possible to determine the temperature ranges relative to water desorption as well as the surfactant decomposition and silanol condensation.

Antimicrobial peptides (AMPs) are effector molecules of innate immune systems found in different groups of organisms, including microorganisms, plants, insects, amphibians and humans. These peptides exhibit several structural motifs but the most abundant AMPs assume an amphipathic alpha-helical structure. The alpha-helix forming antimicrobial peptides are excellent candidates for protein engineering leading to an optimization of their biological activity and target specificity. Nowadays several approaches are available and this review deals with the use of combinatorial synthesis and directed evolution in order to provide a high-throughput source of antimicrobial peptides analogues with enhanced lytic activity and specificity.

4-Nitroquinoline 1-oxide (4NQO)-induced rat tongue carcinogenesis is a useful model for studying oral squamous cell carcinoma. The aim of this study was to investigate the expression of bcl-2 and bax during tongue carcinogenesis induced by 4NQO. Male Wistar rats were distributed into three groups of 10 animals each and treated with 50 ppm 4NQO solution through their drinking water for 4, 12 or 20 weeks. Ten animals were used as negative control. Although no histological changes were induced in the epithelium after 4 weeks of carcinogen exposure, bcl-2 and bax were over-expressed (P < 0.01) in all layers of the 'normal' epithelium. The expression levels were the same in all layers of epithelium for both the antibodies used (bcl-2 or bax). In dysplastic lesions at 12 weeks following carcinogen administration, the levels of bcl-2 and bax expression did not increase when compared to negative control with the immunoreactivity for bcl-2 being restricted to the superficial layer of epithelium. In well-differentiated squamous cell carcinoma induced after 20 weeks of treatment with 4NQO, bcl-2 was expressed in some cells of tumour islands. on the other hand, immunostaining for bax was widely observed at the tumour nests. The labelling index for bcl-2 and bax showed an increase (P < 0.05) after only 4 weeks of 4NQO administration. In conclusion, our results suggest that abnormalities in the apoptosis pathways are associated with the development of persistent clones of mutated-epithelial cells in the oral mucosa. Bcl-2 and bax expression appears to be associated with a risk factor in the progression of oral cancer.
