There is increasing evidence to support the notion that membrane proteins, instead of being isolated components floating in a fluid lipid environment, can be assembled into supramolecular complexes that take part in a variety of cooperative cellular functions. The interplay between lipid-protein and protein-protein interactions is expected to be a determinant factor in the assembly and dynamics of such membrane complexes. Here we report on a role of anionic phospholipids in determining the extent of clustering of KcsA, a model potassium channel. Assembly/disassembly of channel clusters occurs, at least partly, as a consequence of competing lipid-protein and protein-protein interactions at nonannular lipid binding sites on the channel surface and brings about profound changes in the gating properties of the channel. Our results suggest that these latter effects of anionic lipids are mediated via the Trp67–Glu71–Asp80 inactivation triad within the channel structure and its bearing on the selectivity filter.

This dissertation relates job desires and outcomes to the Dark Personality (Psychopathy, Machiavellianism, Narcissism, Low Agreeableness, Low Honesty-Humility) in the United States Army. It purports that individuals high on the Dark Personality desire more power, money, and status, and that they obtain jobs that afford them these luxuries by using manipulation at work. Two pilot studies used samples of United States Army members to create and test index variables: Dark Personality, Total Manipulation in the workplace, Desire for Job Success, and Total Job Success in the Army. Individual personality traits, manipulation tactics, and job desires were examined in secondary analyses. Using a sample of 468 United States Army Members, central analyses indicated that Army members high on the Dark Personality desired Job Success. Likewise, army members higher on the Dark Personality used more Manipulation tactics at work, including the egregious tactics. Yet, using more Manipulation tactics at work predicted lower levels of Job Success in the Army. Most manipulation tactics had a negative impact on Job Success, with the exception of soft tactics like Reason and Responsibility Invocation. Together, these results indicate that selective use of soft manipulation predicted Job Success, but use of more manipulation tactics predicted less Job Success in the Army. Curvilinear results indicated that being either very low or very high on the Dark Personality predicted more Job Success in the Army, whereas having intermediate levels of the Dark Personality predicted less Job Success. Finally, possessing the Dark Personality and using more Manipulation tactics at work, together, predicted less Job Success in the Army. Collectively, the results indicate that army members with intermediate levels of the Dark Personality want more powerful and high paying jobs, yet their strategy of manipulating their coworkers to move up the job ladder does not result in higher ranking, higher paying Army positions. However, Army members highest on the Dark Personality achieved job success, defying the maladaptive influence that antisocial personality traits and manipulative behaviour had on job success for most Army members. Therefore, this dissertation indicates that successful corporate scoundrels exist in the Army, but there are few of them.

Enquadramento - O envolvimento paterno no trabalho de parto tem suscitado o interesse de vários profissionais de saúde, nomeadamente dos enfermeiros obstetras, dado que os benefícios da presença paterna no contexto de parto ainda não são clara e totalmente assumidos por todos os profissionais. Por outro lado reconhece-se que o pai é o acompanhante ideal da grávida durante o trabalho de parto, pelo seu apoio e lugar que ocupa na tríade. Objetivos – Identificar os benefícios e constrangimentos do envolvimento paterno durante o trabalho de parto e parto. Método – Efetuada revisão integrativa da literatura sobre os “Benefícios e constrangimentos do envolvimento paterno durante o trabalho de parto e parto” nas bases de dados: PubMed, LILACS, SciELO, Repositórios Institucionais e Bibliotecas Digitais e EBSCO Host, elaborados entre 2000 e 2014. Selecionaram-se 19 artigos que obedeceram aos critérios de inclusão deste estudo. Dos 19 artigos selecionados, 4 são revisões sistemáticas da literatura, 13 são estudos qualitativos e 2 são estudos quantitativos. Resultados – Evidenciou-se um “novo” pai, que acompanha a grávida desde a gravidez, e que esteve presente no trabalho de parto. É dada grande relevância aos Enfermeiros Especialistas em Saúde Materna e Obstetrícia, pelo seu papel na inclusão do pai na sala de parto e desmitificação de tabus. Após esta pesquisa constatou-se que, ainda há lacunas relativamente à presença do pai parto. São muitos os benefícios do envolvimento paterno no parto, mas ainda são visíveis constrangimentos por parte do pai relativamente ao contexto de parto. Conclusões - A presença do pai na sala de parto permite estreitar os laços mais íntimos, consolidando a união familiar e proporcionando bem-estar à grávida. Realça-se o papel dos profissionais de saúde na integração do pai na maternidade. Caminhamos para um contexto de parto mais alargado e do qual já não faz parte apenas a grávida e o seu filho, mas no qual o pai é também elemento chave de todo o processo. Upper Berriasian to lower Aptian calcareous nannofossil assemblages have been studied from a siliciclastic deep-sea fan complex and a subjacent limestone sequence drilled beneath the lower continental rise in the western North American Basin, 270 miles (435 km) off Cape Hatteras, North Carolina (USA). Sharp lithologic facies changes and reworking by turbidites complicate the biostratigraphic interpretation, but provide an excellent opportunity to better distinguish "nearshore" from open-ocean nannofossil species, and to investigate the introduction of neritic taxa into the deep-see environment, a phenomenon that appears to have been widespread within the circum-North Atlantic during Neocomian times. Well-preserved assemblages in dark, carbonaceous claystones were probably displaced from the oxygen minimum zone along the upper slope or outer shelf. Neritic, continental margin species prevalent in this facies include the holococcolith Zebrashapka vanhintei n. gen., n. sp., Lithraphidites alatus magnus n. spp., Pickelhaube furtiva n. gen., and a host of nannoconids and micrantholiths. A qualitative evaluation of widely used guide fossils suggests that the triad of proposed markers for the base of Roth's Zone NC3 make their first appearances in the following (ascending) order: Diadorhombus rectus, TUbodiscus verenae, Calcicalathina oblongata. Of these, we chose the nominative species for the zone, T. verenae, to mark its base and to approximate the Berriasian/Valangian boundary. Cyclagelosphaera deflandrei is strongly affected by diagenesis and is therefore not a reliable index species for the base of Zone NC4 near the Valanginian/Hauterivian boundary (the last occurrence of T. verenae is also not suitable there). In addition, Lithraphidites bollii, a form apparently confined to the low latitudes of the Tethyan region, was absent at the more temperate Site 603 and not available as a subzonal marker for the upper Hautervian-lower Barremian (mid-NC4 and mid-NC5, respectively). Cruciellipsis cuvillieri, however, provides a reliable datum just below the Hauterivian/Barremian boundary (base of NC5), despite the potential for reworking in this section. Nannoconids tend to be reworked in this section, and do not provide trustworthy forms to mark the Barremian/Aptian boundary (base of NC6). Hayesites irregularis n. comb, probably does provide a useful first appearance datum within the lower Aptian, if it is not confused with a more birefringent and globular form, Rucinolithus terebrodentarius n. sp. Rhagodiscus angustus is mimicked by a similar form (Zeughrabdotusl pseudoangustus n. sp.), which apparently ranges down to the Hauterivian, thus Lithastrinus floralis provides a more useful first appearance datum for the base of the middle-upper Aptian Rhagodiscus angustus Zone (NC7). Aside from the new taxa mentioned above, the following are also described: Cretarhabdusl delicatus n. sp. and Cyclagelosphaera jiangii n. sp.

Children bear disproportionate consequences of armed conflict. The 21st century continues to see patterns of children enmeshed in international violence between opposing combatant forces, as victims of terrorist warfare, and, perhaps most tragically of all, as victims of civil wars. Innocent children so often are the victims of high-energy wounding from military ordinance. They sustain high-energy tissue damage and massive burns - injuries that are not commonly seen in civilian populations. Children have also been deliberately targeted victims in genocidal civil wars in Africa in the past decade, and hundreds of thousands have been killed and maimed in the context of close-quarter, hand-to-hand assaults of great ferocity. Paediatricians serve as uniformed military surgeons and as civilian doctors in both international and civil wars, and have a significant strategic role to play as advocates for the rights and welfare of children in the context of the evolving 'Laws of War'. One chronic legacy of contemporary warfare is blast injury to children from landmines. Such blasts leave children without feet or lower limbs, with genital injuries, blindness and deafness. This pattern of injury has become one of the post-civil war syndromes encountered by all intensivists and surgeons serving in four of the world's continents. The continued advocacy for the international ban on the manufacture, commerce and military use of antipersonnel landmines is a part of all paediatricians' obligation to promote the ethos of the Laws of War. Post-traumatic stress disorder remains an undertreated legacy of children who have been trapped in the shot and shell of battle as well as those displaced as refugees. An urgent, unfocused and unmet challenge has been the increase in, and plight of, child soldiers themselves. A new class of combatant comprises these children, who also become enmeshed in the triad of anarchic civil war, light-weight weaponry and drug or alcohol addiction. The International Criminal Court has outlawed as a War Crime, the conscription of children under 15 years of age. Nevertheless, there remain more than 300 000 child soldiers active and enmeshed in psychopathic violence as part of both civil and international warfare. The typical profile of a child soldier is of a boy between the ages of 8 and 18 years, bonded into a group of armed peers, almost always an orphan, drug or alcohol addicted, amoral, merciless, illiterate and dangerous. Paediatricians have much to do to protect such war-enmeshed children, irrespective of the accident of their place of birth. Only by such vigorous and maintained advocacy can the world's children be better protected from the scourge of future wars.

Hong Kong firms continue to play an important role in the development of mainland China trade. This paper studies the nature of coordinations in triadic interactions between Hong Kong Chinese intermediaries and mainland Chinese sellers, and buyers from the West, and their impact on relationships development. Contrary to popular belief, the results presented here identify that interactions between Hong Kong intermediaries and mainland Chinese sellers tend to include relationships based on price and authority. These interactions tend to be classical in nature, and serve to hinder the development of relationships. On the other hand, interactions between buyers from the West and Hong Kong intermediaries tend to be relational in nature. They include different forms of personal relationships, which tends to be positive for the development of relationships. A new plural form of international business relationship triad is also identified.

A loss of function mutation in growth differentiation factor 9 (GDF9) in sheep causes increased ovulation rate and infertility in a dosage-sensitive manner. Spontaneous dizygotic (DZ) twinning in the human is under genetic control and women with a history of DZ twinning have an increased incidence of multiple follicle growth and multiple ovulation. We sequenced the GDF9 coding region in DNA samples from 20 women with DZ twins and identified a four-base pair deletion in GDF9 in two sisters with twins from one family. We screened a further 429 families and did not find the loss of function mutation in any other families. We genotyped eight single nucleotide polymorphisms across the GDF9 locus in 379 families with two sisters who have both given birth to spontaneous DZ twins (1527 individuals) and 226 triad families with mothers of twins and their parents (723 individuals). Using case control analysis and the transmission disequilibrium test we found no evidence for association between common variants in GDF9 and twinning in the families. We conclude that rare mutations in GDF9 may influence twinning, but twinning frequency is not associated with common variation in GDF9.

The MFG test is a family-based association test that detects genetic effects contributing to disease in offspring, including offspring allelic effects, maternal allelic effects and MFG incompatibility effects. Like many other family-based association tests, it assumes that the offspring survival and the offspring-parent genotypes are conditionally independent provided the offspring is affected. However, when the putative disease-increasing locus can affect another competing phenotype, for example, offspring viability, the conditional independence assumption fails and these tests could lead to incorrect conclusions regarding the role of the gene in disease. We propose the v-MFG test to adjust for the genetic effects on one phenotype, e.g., viability, when testing the effects of that locus on another phenotype, e.g., disease. Using genotype data from nuclear families containing parents and at least one affected offspring, the v-MFG test models the distribution of family genotypes conditional on offspring phenotypes. It simultaneously estimates genetic effects on two phenotypes, viability and disease. Simulations show that the v-MFG test produces accurate genetic effect estimates on disease as well as on viability under several different scenarios. It generates accurate type-I error rates and provides adequate power with moderate sample sizes to detect genetic effects on disease risk when viability is reduced. We demonstrate the v-MFG test with HLA-DRB1 data from study participants with rheumatoid arthritis (RA) and their parents, we show that the v-MFG test successfully detects an MFG incompatibility effect on RA while simultaneously adjusting for a possible viability loss.

Esta pesquisa, Responsabilidade Social, Pregação e Educação: tensões missiológicas no projeto missionário da Igreja Metodista em terras brasileiras, tem como objetivo analisar as ações missionárias da Igreja Metodista do período de sua implantação até o final do período referente ao 18° Concílio Geral (2006-2011). A análise resgatou a tríade Responsabilidade Social, Pregação e Educação como uma marca identitária do movimento wesleyano e que esse princípio missionário nem sempre foi seguido pelo metodismo brasileiro. Notou-se também que a tríade serviu, em momentos decisivos, como elemento norteador do projeto missionário. Isso é percebido no período da implantação, bem como na constituição do Plano para Vida e Missão (PVM). Em que pese que as ações missionárias no metodismo brasileiro nem sempre foram providas de uma reflexão teológica sistemática, a tríade Responsabilidade Social, Pregação e Educação sempre funcionou como eixos essenciais da missão e de forma indissociável. Não foram poucos os momentos em que o projeto missionário metodista enfrentou tensões na sua execução. O ímpeto pelo crescimento numérico e expansão geográfica, muitas vezes, ofuscou a fidelidade à tríade. Nessa trajetória de pesquisa está construída em quatro capítulos, nos quais se busca a superação de uma compreensão histórica equivocada e faz-se necessário que o projeto missionário assuma os elementos essenciais da tríade como marco regulatório da missão metodista. As repetitivas opções equivocadas não colaboram com a nova compreensão de missão. O resultado da pesquisa encoraja futuras análises para que o metodismo assuma o papel de agente de Deus na missio Dei e para que o seu envolvimento missionário mantenha fidelidade à tríade Responsabilidade Social, Pregação e Educação.

O tema central de nossa pesquisa são as interações dos pentecostais com as questões socioambientais. Portanto, trata-se do estudo do grau de envolvimento dos pentecostais com as políticas públicas ambientais e o meio ambiente em periferia urbana - Rio Grande da Serra - Grande ABC Paulista. A intensa presença dos pentecostais nos bairros mais periféricos da cidade permitiu-nos indagar sobre as relações que esses mantêm com a expansão urbana nas áreas a serem protegidas, com os impactos ambientais, com consumo e pós-consumo na região, na participação de políticas públicas, bem como, os discursos e práticas religiosas interferem no comportamento dos religiosos frente a todas essas questões, uma vez que, a cidade está inserida em Área de Proteção e Recuperação de Mananciais, imbricada em um dos biomas terrestres mais ameaçados de extinção - o Bioma da Mata Atlântica. Como procedimentos metodológicos servimo-nos das observações empíricas, registro dos aspectos e impactos ambientais, aplicação de formulários e entrevistas com roteiro semiestruturado, elementos que constituíram uma tríade instrumental analítica. Concluímos que os pentecostais embora sejam atuantes em questões sociais, todavia, não o são quanto às questões ambientais na cidade, pois, não participam de maneira mais efetiva nas políticas públicas ambientais. Entretanto, os pentecostais demonstram uma potencialidade em participar do planejamento urbano participativo e dos processos de gestão urbana a médio e longos prazos, desde que sejam estimulados, primando dessa maneira, pela proteção, defesa e justiça ambiental nessas áreas de vulnerabilidade socioambiental.
