Most multimeric lectins are adhesion molecules, promoting attachment and spreading on surface glycodeterminants. In addition, some lectins have counter-adhesion properties, detaching already spread cells which then acquire round or spindle-formed cell shapes. Since lectin-mediated adhesion and detachment is observed in haemocyte-like Drosophila cells, which have haemomucin as the major lectin-binding glycoprotein, the two opposite cell behaviours may be the result of lectin-mediated receptor rearrangements on the cell surface. To investigate oligomeric lectins as a possible extracellular driving force affecting cell shape changes, we examined lectin-mediated reactions in lepidopteran haemocytes after cytochalasin D-treatment and observed that while cell-spreading was dependent on F-actin, lectin-uptake was less dependent on F-actin. We propose a model of cell shape changes involving a dynamic balance between adhesion and uptake reactions. (C) 2004 Elsevier Ltd. All rights reserved.

Insect host-parasitoid interactions provide fascinating examples of evolutionary adaptations in which the parasitoid employs a variety of measures and countermeasures to overcome the immune responses of its host. Maternal factors introduced by the female wasps during egg deposition play an important role in interfering with cellular and humoral components of the host's immune defence. Some of these components actively suppress host immune components and some are believed to confer protection for the developing endoparasitoid by rather passive means. The Venturio conescens/Ephestia kuehniella parrositoid-host system is unique among other systems in that the cellular defence capacity of the host remains virtually intact after parasitization. This system raises some important questions that are discussed in this mini-review: If immune protection of the egg and the emerging larva is achieved by surface properties comprising glycoproteins and virus-like particles (VLPs) produced by the female wasp, why is the prophenoloxidose activating cascade blocked in parasitized caterpillars? Another question is the evolutionary origin of these particles, given that the functional role and structural features of V. canescens VLP proteins are more related to cellular proteins than to viruses.

Lipophorin is the major lipid carrier in insects, but various observations indicate that lipophorin is also involved in immune reactions. To examine a possible role of lipophorin in defence reactions, we mixed hemolymph plasma from Galleria mellonella with LPS and noticed that lipophorin forms detergent-insoluble aggregates, while most other plasma proteins are not affected. Lipophorin particles isolated by low-density gradient centrifugation retained LPS-induced aggregation properties, which suggested to us that these immune-reactive particles are able to recognise LPS and respond by forming insoluble aggregates. Antibodies against LPS-binding proteins, such as immulectin-2 and beta-1,3-glucan binding protein, cross-reacted with proteins associated with purified lipophorin particles. To examine whether LPS-mediated aggregates inactivate LPS, we added LPS-lipophorin mixtures to purified lipophorin particles and monitored aggregate formation. Under these conditions lipophorin did not form insoluble aggregates, which indicates that lipophorin particles sequester LPS into non-toxic aggregates. (c) 2005 Elsevier Ltd. All rights reserved.

Esta dissertação tem por objeto “A Tributação Internacional dos Desportistas na Ótica dos Impostos sobre o Rendimento”, pretendendo-se analisar o objeto e o alcance das convenções fiscais bilaterais, no âmbito da tributação dos desportistas, caraterizando as suas regras, bem como as regras constantes da legislação nacional em sede de Imposto sobre o Rendimento das Pessoas Singulares (IRS) e de Imposto sobre o Rendimento das Pessoas Coletivas (IRC).A evolução da economia portuguesa, e em especial do desporto, fruto da globalização, internacionalização e mobilidade dos fatores de produção, verificada na União Europeia (UE) e a nível mundial, tem originado que cada vez mais fluxos de rendimentos e de capitais circulem de e para Portugal, factos que levam a que a Fiscalidade já não possa ser estudada de uma forma isolada sem se conhecer o regime das convenções de dupla tributação (CDT) celebradas por Portugal relativas aos impostos sobre o rendimento.A diversidade dos rendimentos auferidos, bem como a natureza dos serviços prestados, fazem dos desportistas das pessoas mais móveis do mundo, levantando-se as mais diversas questões no campo tributário, nomeadamente ao nível do conceito de desportista e da qualificação dos rendimentos por eles auferidos, a que pretendemos dar resposta ao longo deste trabalho. A maioria das CDT define como regra geral, no que concerne aos desportistas, a competência cumulativa para a tributação dos rendimentos auferidos pelos mesmos, suscitando desde logo a questão de saber se o modelo de tributação dos desportistas é suficiente ou necessário nos dias de hoje. A esta questão procuraremos dar resposta no final.

This thesis involves two parts. The first is a new-proposed theoretical approach called generalized atoms in molecules (GAIM). The second is a computational study on the deamination reaction of adenine with OH⁻/nH₂O (n=0, 1, 2, 3) and 3H₂O. The GAIM approach aims to solve the energy of each atom variationally in the first step and then to build the energy of a molecule from each atom. Thus the energy of a diatomic molecule (A-B) is formulated as a sum of its atomic energies, EA and EB. Each of these atomic energies is expressed as, EA = Hᴬ + Vₑₑᴬᴬ + 1/2Vₑₑᴬ<>ᴮ EB = Hᴮ + Vₑₑᴮᴮ + 1/2Vₑₑᴬ<>ᴮ where; Hᴬ and Hᴮ are the kinetic and nuclear attraction energy of electrons of atoms A and B, respectively; Vₑₑᴬᴬ and Vₑₑᴮᴮ are the interaction energy between the electrons on atoms A and B, respectively; and Vₑₑᴬ<>ᴮ is the interaction energy between the electrons of atom A with the electrons of atom B. The energy of the molecule is then minimized subject to the following constraint, |ρA(r)dr + |ρB(r)dr = N where ρA(r) and ρB(r) are the electron densities of atoms A and B, respectively, and N is the number of electrons. The initial testing of the performance of GAIM was done through calculating dissociation curves for H₂, LiH, Li₂, BH, HF, HCl, N₂, F₂, and Cl₂. The numerical results show that GAIM performs very well with H₂, LiH, Li₂, BH, HF, and HCl. GAIM shows convergence problems with N₂, F₂, and Cl₂ due to difficulties in reordering the degenerate atomic orbitals Pₓ, Py, and Pz in N, F, and Cl atoms. Further work for the development of GAIM is required. Deamination of adenine results in one of several forms of premutagenic lesions occurring in DNA. In this thesis, mechanisms for the deamination reaction of adenine with OH⁻/nH₂O, (n = 0, 1, 2, 3) and 3H₂O were investigated. HF/6-31G(d), B3LYP/6-31G(d), MP2/6-31G(d), and B3LYP/6-31+G(d) levels of theory were employed to optimize all the geometries. Energies were calculated at the G3MP2B3 and CBS-QB3 levels of theory. The effect of solvent (water) was computed using the polarizable continuum model (PCM). Intrinsic reaction coordinate (IRC) calculations were performed for all transition states. Five pathways were investigated for the deamination reaction of adenine with OH⁻/nH₂O and 3H₂O. The first four pathways (A-D) begin with by deprotonation at the amino group of adenine by OH⁻, while pathway E is initiated by tautomerization of adenine. For all pathways, the next two steps involve the formation of a tetrahedral intermediate followed by dissociation to yield products via a 1,3-hydrogen shift. Deamination with a single OH⁻ has a high activation barrier (190 kJ mol⁻¹ using G3MP2B3 level) for the rate-determining step. Addition of one water molecule reduces this barrier by 68 kJ mol⁻¹ calculated at G3MP2B3 level. Adding more water molecules decreases the overall activation energy of the reaction, but the effect becomes smaller with each additional water molecule. The most plausible mechanism is pathway E, the deamination reaction of adenine with 3H₂O, which has an overall G3MP2B3 activation energy of 139 and 137 kJ mol⁻¹ in the gas phase and PCM, respectively. This barrier is lower than that for the deamination with OH⁻/3H₂O by 6 and 2 kJ mol⁻¹ in the gas phase and PCM, respectively.

La tesi presenta uno studio della libreria grafica per web D3, sviluppata in javascript, e ne presenta una catalogazione dei grafici implementati e reperibili sul web. Lo scopo è quello di valutare la libreria e studiarne i pregi e difetti per capire se sia opportuno utilizzarla nell'ambito di un progetto Europeo. Per fare questo vengono studiati i metodi di classificazione dei grafici presenti in letteratura e viene esposto e descritto lo stato dell'arte del data visualization. Viene poi descritto il metodo di classificazione proposto dal team di progettazione e catalogata la galleria di grafici presente sul sito della libreria D3. Infine viene presentato e studiato in maniera formale un algoritmo per selezionare un grafico in base alle esigenze dell'utente.

The Dot/Icm type IV secretion system (T4SS) of Legionella pneumophila is crucial for the pathogen to survive in protozoa and cause human disease. Although more than 275 effector proteins are delivered into the host cell by the T4SS, the function of the majority is unknown. Here we have characterized the Dot/Icm effector LtpD. During infection, LtpD localized to the cytoplasmic face of the membrane of the Legionella-containing vacuole (LCV). In A549 lung epithelial cells, ectopically expressed LtpD localized to large vesicular structures that contained markers of endosomal compartments. Systematic analysis of LtpD fragments identified an internal 17-kDa fragment, LtpD471-626, which was essential for targeting ectopically expressed LtpD to vesicular structures and for the association of translocated LtpD with the LCV. LtpD471-626 bound directly to phosphatidylinositol 3-phosphate [PtdIns(3)P] in vitro and colocalized with the PtdIns(3)P markers FYVE and SetA in cotransfected cells. LtpD was also found to bind the host cell enzyme inositol (myo)-1 (or 4)-monophosphatase 1, an important phosphatase involved in phosphoinositide production. Analysis of the role of LtpD in infection showed that LtpD is involved in bacterial replication in THP-1 macrophages, the larvae of Galleria mellonella, and mouse lungs. Together, these data suggest that LtpD is a novel phosphoinositide- binding L. pneumophila effector that has a role in intracellular bacterial replication. © 2013, American Society for Microbiology.

Burkholderia phage AP3 (vB_BceM_AP3) is a temperate virus of the Myoviridae and the Peduovirinae subfamily (P2likevirus genus). This phage specifically infects multidrug-resistant clinical Burkholderia cenocepacia lineage IIIA strains commonly isolated from cystic fibrosis patients. AP3 exhibits high pairwise nucleotide identity (61.7%) to Burkholderia phage KS5, specific to the same B. cenocepacia host, and has 46.7% - 49.5% identity to phages infecting other species of Burkholderia. The lysis cassette of these related phages has a similar organization (putative antiholin, putative holin, endolysin and spanins) and shows 29-98% homology between specific lysis genes, in contrast to Enterobacteria phage P2, the hallmark phage of this genus. The AP3 and KS5 lysis genes have conserved locations and high amino acid sequence similarity. The AP3 bacteriophage particles remain infective up to 5 h at pH 4-10 and are stable at 60°C for 30 min, but are sensitive to chloroform, with no remaining infective particles after 24 h of treatment. AP3 lysogeny can occur by stable genomic integration and by pseudo-lysogeny. The lysogenic bacterial mutants did not exhibit any significant changes in virulence compared to wild-type host strain when tested in the Galleria mellonella moth wax model. Moreover, AP3 treatment of larvae infected with B. cenocepacia revealed a significant increase (P < 0.0001) in larvae survival in comparison to AP3-untreated infected larvae. AP3 showed robust lytic activity, as evidenced by its broad host range, the absence of increased virulence in lysogenic isolates, the lack of bacterial gene disruption conditioned by bacterial tRNA downstream integration site, and the absence of detected toxin sequences. These data suggest the AP3 phage is a promising potent agent against bacteria belonging to most common B. cenocepacia IIIA lineage strains.

Introdução: A correção cirúrgica do aneurisma da aorta abdominal (AAA), por Endovascular Aneurysm Repair (EVAR) ou cirurgia convencional (CC), pode agravar a função renal a curto prazo. Esta complicação, mais frequente nos doentes com insuficiência renal crónica (IRC), associa-se a pior prognóstico a longo prazo. O objetivo deste trabalho foi quantificar o agravamento da função renal após reparação do AAA em doentes com IRC prévia e demonstrar o consequente aumento da morbimortalidade. Métodos: Estudo retrospetivo em doentes com IRC estádios Chronic Kidney Disease 3-4 (TFGe 15-59ml/min), submetidos a correção eletiva de AAA entre fevereiro/2011 e fevereiro/2015 numa instituição terciária. Variáveis estudadas: idade, sexo, tipo de intervenção (convencional/EVAR) e estádio CKD. Endpoints: variação da creatinina e taxa de filtração glomerular com a cirurgia, complicações renais pós-operatórias, necessidade de reintervenção cirúrgica e mortalidade. A análise estatística foi realizada em SPSS. Resultados: Foram incluídos 71doentes. Quinze doentes (21%) foram operados por CC e 56 (78%) por EVAR. À data da intervenção, os doentes encontravam-se nos seguintes estádios da DRC: CKD 3 --- 65 (91%) e CKD 4 --- 6 (9%). A variac¸ão da TFG com a cirurgia foi −1,08±18,01mg/dl. Verificou-se IRC agudizada pós-operatória em 22 (31%) doentes e necessidade de diálise em 5 (7%). A mortalidade global foi 8,5%. Os doentes operados por EVAR tinham DRC mais avançada pré-operatoriamente, mas apresentaram menor agravamento da função renal. Variação TFG: EVAR 1,14±16,26ml/min vs. CC 9,40±22,11ml/min (p=0,022); variação creatinina: EVAR 0,17±1,03mg/dl vs. CC 0,81±1,47mg/dl (p=0,02). A agudização da IRC pós-operatória foi superior no grupo CC (53,3 vs. 28,6%; p=0,072), assim como a necessidade de diálise (20 vs. 3,6%, p=0,06). Os 6 doentes que faleceram (EVAR: 3; CC: 3) apresentaram maior agravamento da função renal (variação da creatinina: 1,41±1,63mg/dl vs. 0,20±1,07mg/dl, p=0,001; variação da TFG: −19,0±16,55ml/min; 0,57±17,34ml/min, p=0,007) e necessidade de diálise (50 vs. 3,1%, p=0,003). Conclusão: Os resultados demonstraram uma tendência para uma menor probabilidade de IRA, menor necessidade de diálise pós-operatória e menor mortalidade nos doentes tratados por EVAR. Contudo, o impacto da administração de contraste a médio/longo prazo, decorrente dos programas de vigilância pós-EVAR, deve ser considerado. Julgamos ser possível considerar que a realização de EVAR para o tratamento de doentes com AAA e IRC é um procedimento pelo menos tão seguro como a CC.

El objetivo de este trabajo fue evaluar en diferentes periodos de almacenamiento la calidad, en términos de viabilidad y concentración de conidias, en formulaciones bioplaguicidas a base de Beauveria bassiana, elaboradas con diferentes materiales inertes. Los materiales inertes (tratamientos) evaluados fueron: harina de trigo, arcilla blanca, arcilla verde, sulfato de calcio, aceite de soya, aceite de maní y el testigo fue B. bassiana en sustrato (no formulado). La concentración del hongo, utilizada para las formulaciones fue 8.5 x 109 conidias por gramo. Se utilizó B. bassiana cepa 114, obtenida del cepario del laboratorio de hongos entomopatógenos de la UNA. La reproducción del hongo se hizo mediante el método de producción semi-industrial, utilizando arroz grano entero como sustrato. La caracterización se hizo mediante características macroscópicas y ritmo de crecimiento del hongo en los medios de cultivo: PDA, SDA y EMA. Luego de elaboradas las formulaciones, fueron almacenadas en condiciones de laboratorio (24 oC -28ºC, HR60%) durante seis meses se determinó la concentración y viabilidad de conidias. Para la evaluación de la concentración de conidias se prepararon diluciones seriadas y el conteo de conidias se realizó en una cámara con rayado Neubauer. La evaluación de viabilidad, se hizo en medio Agar-agua 2%. La evaluación de la concentración de conidias así como la evaluación de la viabilidad se hizo cada 15 días durante los primeros tres meses y luego cada 30 días durante los últimos tres meses. El efecto de los materiales inertes sobre la efectividad biológica de B. bassiana fue evaluado en un bioensayo con la especie Galleria mellonella la que se obtuvo del laboratorio de cría de la UNA, usando una suspensión del hongo a una concentración de 1.6 x 108 por ml. Se realizó Análisis de Varianza y separación de medias según Tukey, por cada fecha de monitoreo. Se concluye que el mejor material para formular B. bassiana es harina de trigo, ya que fue el material inerte que presentó menor disminución en la concentración y viabilidad de conidias durante el periodo evaluado. La mayor velocidad promedio de crecimiento se observó en el medio de cultivo PDA. En general, los materiales inertes no afectaron significativamente la efectividad biológica de Beauveria bassiana en larvas de Galleria mellonella.

This paper discusses some didactical aspects of chat usage for instructional purposes. Areas covered range from the choice of course type and student group for using chats, to the preparation and moderation of chats, and discussing how to manage both troublemaking students and systems. In February 1998 I was the chair of a committee at my previous university that was authoring a recommendation list about the Internet to the president of the school. One of our recommendations was to eliminate the IRC server (Internet Relay Chat), since it was only causing traffic and was just being used for games. We stated that we could not envision a use for chatting either for research or for instructional purposes. Five years later here I am: teaching 4 of my 5 courses by way of chat. There has been an enormous growth in the use of chat-based instruction, but also quite a lot of problems encountered. Much of the available literature addresses technical problems, or discusses chats from an educational psychology perspective. This paper will address ten of the didactical issues in chat-based instruction, summarizing the experiences that I have made in many years of chat experience (DIPF/Orig.)
