A microplate assay was modified for the detection of antimicrobial activity in plant extracts. The aim was to develop an in vitro assay that could rapidly screen plant extracts to provide quantitative data on inhibition of microbial growth. A spectrophotometric assay using a microplate with serial dilutions of the plant extract and the bacteria was developed. Two bacteria, Staphylococcus aureus and Escherichia coli, were used for this study. Essential oils, oregano (Origanum vulgare) and lemon myrtle (Backhousia citriodora), and three active components carvacrol, thymol and citral were evaluated. The reproducibility of the assay was high, with correlation coefficients (r aureus and E. coli between 0.9321 and 0.9816. Similarly, r and 0.9814. This assay could also be used to measure antimicrobial activity in plant extracts which vary in pH and color.

In the title Mannich base, C20H21N3O3, an isatin derivative of thymol the O-CH2-C(=O)-N(H)-N fragment connecting the aromatic and fused-ring systems is approximately planar, with the N-N single bond in a Zmconfiguration. The amino H atom of this N-N fragment is intramolecularly hydrogen bonded to the carbonyl O atom of the indolinone fused ring as well as to the phenoxy O atom of the aromat ring. The amino H atom of the indoline fused ring forms a hydrogen bond with the double-bond O atom of an adjacent molecule, this hydrogen bond giving rise to a linear chain motif.

The present paper analyzes the effects of plumes for heat transfer enhancement at solid-liquid interface taking both smooth and grooved surfaces. The experimental setup consists of a tank of dimensions 265 x 265 x 300 (height) containing water. The bottom surface was heated and free surface of the water was left open to the ambient. In the experiments, the bottom plate had either a smooth surface or a grooved surface. We used 90 V-grooved rough surfaces with two groove heights, 10mm and 3mm. The experiment was done with water layer depths of 90mm and 140mm, corresponding to values of aspect ratio(AR) equal to 2.9 and 1.8 respectively. Thymol blue, a pH sensitive dye, was used to visualize the flow near the heated plate. The measured heat transfer coefficients over the grooved surfaces were higher compared that over the smooth surface. The enhanced heat transport in the rough cavities cannot be ascribed to the increase in the contact area, rather it must be the local dynamics of the thermal boundary layer that changes the heat transport over the rough surface.

The diabetic dog represents an excellent model for use in many aspects of diabetic research. The present paper describes, in detail, a reproducible experimental protocol for the successful induction of chemical diabetes in beagles using a combination of the 2 pancreatic beta-cell cytoxic agents alloxan and streptozotocin.

Supercritical fluid extraction (SEE) of the volatile oil from Thymus vulgaris L. aerial flowering parts was performed under different conditions of pressure, temperature, mean particle size and CO2 flow rate and the correspondent yield and composition were compared with those of the essential oil isolated by hydrodistillation (HD). Both the oils were analyzed by GC and GC-MS and 52 components were identified. The main volatile components obtained were p-cymene (10.0-42.6% for SFE and 28.9-34.8% for HD), gamma-terpinene (0.8-6.9% for SFE and 5.1-7.0% for HD), linalool (2.3-5.3% for SFE and 2.8-3.1% for HD), thymol (19.5-40.8% for SFE and 35.4-41.6% for HD), and carvacrol (1.4-3.1% for SFE and 2.6-3.1% for HD). The main difference was found to be the relative percentage of thymoquinone (not found in the essential oil) and carvacryl methyl ether (1.0-1.2% for HD versus t-0.4 for SFE) which can explain the higher antioxidant activity, assessed by Rancimat test, of the SFE volatiles when compared with HD. Thymoquinone is considered a strong antioxidant compound.

A discussion of the most interesting results obtained in our laboratories, during the supercritical CO(2) extraction of bioactive compounds from microalgae and volatile oils from aromatic plants, was carried out. Concerning the microalgae, the studies on Botryococcus braunii and Chlorella vulgaris were selected. Hydrocarbons from the first microalgae, which are mainly linear alkadienes (C(23)-C(31)) with an odd number of carbon atoms, were selectively extracted at 313 K increasing the pressure up to 30.0 MPa. These hydrocarbons are easily extracted at this pressure, since they are located outside the cellular walls. The extraction of carotenoids, mainly canthaxanthin and astaxanthin, from C. vulgaris is more difficult. The extraction yield of these components at 313 K and 35.0 MPa increased with the degree of crushing of the microalga, since they are not extracellular. On the other hand, for the extraction of volatile oils from aromatic plants, studies on Mentha pulegium and Satureja montana L were chosen. For the first aromatic plant, the composition of the volatile and essential oils was similar, the main components being the pulegone and menthone. However, this volatile oil contained small amounts of waxes, which content decreased with decreasing particle size of the plant matrix. For S. montana L it was also observed that both oils have a similar composition, the main components being carvacrol and thymol. The main difference is the relative amount of thymoquinone, which content can be 15 times higher in volatile oil. This oxygenated monoterpene has important biological activities. Moreover, experimental studies on anticholinesterase activity of supercritical extracts of S. montana were also carried out. The supercritical nonvolatile fraction, which presented the highest content of the protocatechuic, vanilic, chlorogenic and (+)-catechin acids, is the most promising inhibitor of the enzyme butyrylcholinesterase. Le parasite Varroa destructor provoque depuis plus de 30 ans la perte de nombreuses colonies à travers le monde. L’utilisation d’acaricides de synthèse s’est avérée inefficace au Canada et ailleurs dans le monde à la suite de la sélection de varroas résistants. Dans ce contexte, il est devenu impératif de trouver de nouveaux moyens pour contrôler cette peste apicole. Ce travail original de recherche a pour but de déterminer les paramètres fondamentaux d’une lutte intégrée contre la varroase fondée sur l’utilisation périodique de différents pesticides organiques (l’acide oxalique, l’acide formique et le thymol) associée à des seuils d’interventions. Les seuils d’intervention ont été déterminés à l’aide de régressions linéaires entre les taux de parasitisme par V. destructor et la formance zootechnique des colonies d’abeilles mellifères (production de miel et force des colonies). Un total de 154 colonies d’abeilles du Centre de recherche en sciences animales de Deschambault (CRSAD) ont été suivies de septembre 2005 à septembre 2006. Les seuils calculés et proposés à la suite de cette recherche sont de 2 varroas par jour (chute naturelle) au début mai, 10 varroas par jour à la fin juillet et de 9 varroas par jour au début septembre. L’efficacité des traitements organiques avec l’acide oxalique (AO), l’acide formique (AF) et le thymol a été vérifiée en mai (avant la première miellée) en juillet (entre deux miellées), en septembre (après la miellée et pendant le nourrissage des colonies) et en novembre (avant l’hivernage). L’acide oxalique a été appliqué en utilisant la méthode d’égouttement (4% d’AO p/v dans un sirop de sucrose 1 :1 p/v). L’acide formique a été appliquée sous forme de MiteAwayII™ (tampon commercial imbibé d’AF 65% v/v placé sur le dessus des cadres à couvain), Mitewipe (tampons Dri-Loc™ 10/15cm imbibés de 35 mL d’AF 65% v/v placés sur le dessus des cadres à couvain) ou Flash (AF 65% coulé directement sur le plateau inférieur d’une colonie, 2 mL par cadre avec abeilles). Le thymol a été appliqué sous forme d’Apiguard™ (gélose contenant 25% de thymol p/v placée sur le dessus des cadres à couvain). Les essais d’efficacité ont été réalisés de 2006 à 2008 sur un total de 170 colonies (98 appartenant au CRSAD et 72 appartenant au privé). Les résultats montrent que les traitements de printemps testés ont une faible efficacité pour le contrôle des varroas qui sont en pleine croissance durant cette période. Un traitement avec l’AF à la mi-été permet de réduire les taux de parasites sous le seuil en septembre mais il y a risque de contaminer la récolte de miel avec des résidus d’AF. Les traitements en septembre avec le MiteAwayII™ suivis par un traitement à l’acide oxalique en novembre (5 mL par égouttement entre chaque cadre avec abeilles, 4% d’AO p/v dans un sirop de sucrose 1 :1 p/v) sont les plus efficaces : ils réduisent les niveaux de varroase sous le seuil de 2 varroas par jour au printemps. Nos résultats montrent également que les traitements réalisés tôt en septembre sont plus efficaces et produisent des colonies plus fortes au printemps comparativement à un traitement réalisé un mois plus tard en octobre. En conclusion, ce travail de recherche démontre qu’il est possible de contenir le développement de la varroase dans les ruchers au Québec en utilisant une méthode de lutte intégrée basée sur une combinaison d’applications d’acaricides organiques associée à des seuils d’intervention.

The combined action of the plant-derived volatile, S-carvone, and mild heat treatment on the food-borne pathogen, Listeria monocytogenes, was evaluated. The viability of exponential phase cultures grown at 8 °C could be reduced by 1·3 log units after exposure to S-carvone (5 mmol l−1) for 30 min at 45 °C, while individual treatment with S-carvone or exposure to 45 °C for 30 min did not result in a loss in viability. Other plant-derived volatiles, namely carvacrol, cinnamaldehyde, thymol and decanal, were also found to reduce the viability of L. monocytogenes in combination with the same mild heat treatment at concentrations of 1·75 mmol l−1, 2·5 mmol l−1, 1·5 mmol l−1 and 2 mmol l−1, respectively. These findings show that essential oil compounds can play an important role in minimally processed foods, and can be used in the concept of Hurdle Technology to reduce the intensity of heat treatment or other individual hurdles.

The Sakaguchi reaction between arginine and hypohalites in the presence of α-phenols (Fig. 1) has been extensively employed for the colorimetric determination of this amino acid [1] M.A Parniak, G Lange and T Viswanatha, Quantitative determination of monosubstituted guanidines: a comparative study of different procedures, J. Biochem. Biophys. Methods 7 (1983), pp. 267–276. Abstract | View Record in Scopus | Cited By in Scopus (8)[1] and [2]. There have been a number of modifications to the reaction for the determination of arginine to improve the color stability and sensitivity. The Sakaguchi reaction is much faster with hypobromite than with hypochlorite, but the colored product fades at a higher rate; however, this can be prevented by adding urea to remove the excess hypobromite [3]. Although 1-naphthol was originally used as the chromogen, other phenols including 2,4-dichloro-1-naphthol [4], 8-quinolinol [5], 5-chloro-7-iodo-8-quinolinol [6], and thymol [2] (Fig. 1) have provided superior analytical figures of merit. We have found that the reaction between arginine and hypobromite is chemiluminescent [7], which has been used to develop an analytical procedure that is rapid, simple, and selective for arginine in the presence of other amino acids.

Toxoplasmosis, a benign disease in normal healthy individuals, can have serious effects in pregnant women and immunocompromised patients. It is a parasitic disease caused by Toxoplasma gondii (Tg), an obligatory intracellular protozoan. The prophylactic and therapeutic arsenal against this parasite is very restricted. Thus, there is an ongoing search for novel drugs and therapeutic strategies. A promising alternative is a rational approach using medicinal plants. This study aimed to standardize methodologies for assessing the toxicological, antiproliferative, antioxidant, antiinflammatory and anti-Toxoplasma effects of Estragole and Thymol compounds isolated from species of plants (Lippia sidoides and Croton zenhtneri) commonly used in the Cariri region of Ceara State, Brazil. First we evaluated in vivo toxicity and conducted a pathological analysis of mice livers. In vivo antiinflammatory activity was assessed using air pouch and paw edema methods. Cytotoxicity assays were performed and antiproliferative, antioxidant and nitric oxide production analyzed. Anti-Toxoplasma activity was evaluated in a congenital experimental model with varying stages of maternal infection using the ME-49 strain and a non- congenital model by using ME-49 and RH strains. The results suggest low to moderate toxicity for both compounds. Thymol was more toxic in vivo and in vitro, having greater pathological repercussion than Estragole. The compounds were inactive for antiproliferative activity. Thymol showed better antioxidant activity, while Estragole stimulated nitric oxide production in macrophages. Both showed significant antiinflammatory activity. In non-congenital Tg infection, both compounds were active only against the ME49 strain. In congenital infection, Estragole (oral route) improved the newborn weight of infected mothers compared with untreated controls. Subcutaneous administration of the two compounds increased the weight of offspring born to infected mothers compared with untreated controls. We concluded that Estragole and Thymol exhibit important biological and anti-Toxoplasma activities. Further studies are needed to elucidate the mechanism of action of these compounds and other possible activities not investigated in the present study

O objetivo deste trabalho foi avaliar os efeitos do ácido oxálico e de óleos essenciais de plantas no controle da infestação pelo ácaro Varroa destructor em colônias de Apis mellifera africanizadas. O experimento foi realizado em delineamento inteiramente casualizado, em 30 colônias, com seis tratamentos e cinco repetições. As colmeias foram tratadas com óleos essenciais de arruda (Ruta graveolens), eucalipto (Eucalyptus spp.) e hortelã (Mentha piperita), além de timol, ácido oxálico e do tratamento controle sem aplicação de produtos. Foram avaliadas a mortalidade de varroas e as taxas de mortalidade de crias e de infestação de varroas em crias e em abelhas adultas, antes e depois da aplicação de cada produto. O ácido oxálico e os óleos de arruda, timol, eucalipto e de hortelã reduziram a mortalidade de crias parasitadas pelo ácaro em 92,1, 83,3, 81,7, 86,4 e 81,3%, respectivamente. O tratamento com ácido oxálico reduziu em 87,4% a infestação de varroas em abelhas adultas. O uso desses produtos é eficiente na redução da mortalidade de crias de A. mellifera parasitadas por V. destructor.

The aim of this study was to evaluate the effect of Cervitec(R) on the abundance of mutans streptococci (MS) in occlusal dental plaque and on 2-year caries increment of partly erupting first permanent molars. Sixteen healthy schoolchildren aged 6-8 years, with at least 2 sound contralateral partly erupted permanent molars, received diet counselling and daily parental supervised toothbrushing with a fluoride dentifrice. Stimulated saliva samples were collected at baseline and after 1 year to evaluate MS levels. In a split-mouth design, Cervitec varnish was applied to one of the teeth at baseline and after 3 and 6 months, while the other tooth in the same jaw was a control. At the 9-month follow-up the teeth were in occlusal contact. At this time, varnish was not applied. At 3 and 6 months after the first application of varnish a significant suppression of MS was observed in plaque. Caries investigations, performed at baseline and every 3 months during the 2 years after the start of the study, showed that all the teeth treated with the varnish were free of caries after 2 years, whereas 8/16 control teeth developed incipient caries. In conclusion, our results suggest that treatment with Cervitec reduces MS in plaque on erupting permanent molars and can lead to a significant decrease in caries incidence. Copyright (C) 2002 S.
