DETERMINATION OF KINETIC AND THERMODYNAMIC PARAMETERS OF L-CYSTEINE ADSORPTION ONTO GOLD BY THE QCM TECHNIQUE. This article discusses the adsorption kinetics of a L-cysteine monolayer onto a gold surface by means of information obtained through the QCM technique. The results indicate that the adsorption process is rapid and follows the Langmuir isotherm, in which adsorption and desorption are considered. From these measurements the following parameter values were obtained: k(d) = (4.2 +/- 0.4) x 10(-3) s(-1), k(a) = 75 +/- 6 M-1 s(-1), K-eq=(1.8 +/- 0.3) x 10(4) M-1 and Delta G(ads) = -(5.8 +/- 0.2) kcal mol(-1).

Nuclear factor-kappaB regulates genes that control immune and inflammatory responses and are involved in the pathogenesis of several diseases, including AIDS and cancer. It has been proposed that reactive oxygen intermediates participate in NF-kappaB activation pathways, and compounds with putative antioxidant activity such as N-acetyl-L-cysteine (NAC) and pyrrolidine dithiocarbamate (PDTC) have been used interchangeably to demonstrate this point. We examined their effects, separately and combined, on different stages of the NF-kappaB activation pathway, in primary and in transformed T cells. We show that NAC, contrary to its reported role as an NF-kappaB inhibitor, can actually enhance rather than inhibit IkappaB degradation and, most importantly, show that in all cases NAC exerts a dominant antagonistic effect on PDTC-mediated NF-kappaB inhibition. This was observed at the level of IkappaB degradation, NF-kappaB DNA binding, and HIV-LTR-driven reporter gene expression. NAC also counteracted growth arrest and apoptosis induced by dithiocarbamates. Antagonistic effects were further observed at the level of jun-NH2-terminal kinase, p38 and ATF-2 activation. Our findings argue against the widely accepted assumption that NAC inhibits all NF-kappaB activation pathways and shows that two compounds, previously thought to function through a common inhibitory mechanism, can also have antagonistic effects.

The corrosion inhibition of l-cysteine on AA5052 aluminium alloy in 4 mol/L NaOH solution was investigated by hydrogen gas evolution experiment, polarisation curve, galvanostatic discharge, electrochemical impedance spectroscopy measurements and quantum chemical calculations. The adsorption of l-cysteine on aluminium alloy surface obeyed the amended Langmuir's adsorption isotherm. The polarisation curves indicated that l-cysteine acted as a cathodic inhibitor to inhibit cathodic reaction. The inhibition mechanism was dominated by the geometric covering effect. The galvanostatic discharge shows that the additives restrain the hydrogen evolution and increase the anodic utilization rate. Quantum chemical calculations indicated that l-cysteine molecules mainly interacted with on the carboxyl groups on the aluminium alloy surface. A strong hybridization occurred between the s-orbital and p-orbital of reactive sites in the l-cysteine molecule and the sp-orbital of Aluminium.

Background: Alcohol consumption and smoking are the main causes of upper digestive tract cancers. These risk factors account for over 75% of all cases in developed countries. Epidemiological studies have shown that alcohol and tobacco interact in a multiplicative way to the cancer risk, but the pathogenetic mechanism behind this is poorly understood. Strong experimental and human genetic linkage data suggest that acetaldehyde is one of the major factors behind the carcinogenic effect. In the digestive tract, acetaldehyde is mainly formed by microbial metabolism of ethanol. Acetaldehyde is also a major constituent of tobacco smoke. Thus, acetaldehyde from both of these sources may have an interacting carcinogenic effect in the human upper digestive tract. Aims: The first aim of this thesis was to investigate acetaldehyde production and exposure in the human mouth resulting from alcohol ingestion and tobacco smoking in vivo. Secondly, specific L-cysteine products were prepared to examine their efficacy in the binding of salivary acetaldehyde in order to reduce the exposure of the upper digestive tract to acetaldehyde. Methods: Acetaldehyde levels in saliva were measured from human volunteers during alcohol metabolism, during tobacco smoking and during the combined use of alcohol and tobacco. The ability of L-cysteine to eliminate acetaldehyde during alcohol metabolism and tobacco smoking was also investigated with specifically developed tablets. Also the acetaldehyde production of Escherichia coli - an important member of the human microbiota - was measured in different conditions prevailing in the digestive tract. Results and conclusions: These studies established that smokers have significantly increased acetaldehyde exposure during ethanol consumption even when not actively smoking. Acetaldehyde exposure was dramatically further increased during active tobacco smoking. Thus, the elevated aerodigestive tract cancer risk observed in smokers and drinkers may be the result of the increased acetaldehyde exposure. Acetaldehyde produced in the oral cavity during ethanol challenge was significantly decreased by a buccal L-cysteine -releasing tablet. Also smoking-derived acetaldehyde could be totally removed by using a tablet containing L-cysteine. In conclusion, this thesis confirms the essential role of acetaldehyde in the pathogenesis of alcohol- and smoking-induced cancers. This thesis presents a novel experimental approach to decrease the local acetaldehyde exposure of the upper digestive tract with L-cysteine, with the eventual goal of reducting the prevalence of upper digestive tract cancers.

Os pesticidas etileno-bis-ditiocarbamatos da classe dos ditiocarbamatos estão entre os fungicidas mais empregados em todo o mundo para o controle de pragas. Muitos métodos para determinar ditiocarbamatos são baseados na hidrólise ácida em presença de cloreto estanoso e análise do CS2 gerado por diferentes técnicas. Nesse contexto, constituiram em objetivos do presente trabalho, como primeira etapa, o estudo de condições adequadas à estocagem de amostras de solo, e como segunda etapa, a avaliação das taxas de degradação e de lixiviação do fungicida mancozebe num cambissolo distrófico através do método espectrofotométrico. O sítio de estudo foi uma área delimitada de 36 m2, de uma cultura de couve, localizada em São Lourenço no 3 distrito do município de Nova Friburgo-RJ. As análises foram realizadas no laboratório de tecnologia ambiental (LABTAM/UERJ). Na primeira etapa, duas sub-amostras de solo contaminadas com mancozebe foram submetidas a tratamento com cloridrato de L-cisteina e estocadas às temperaturas ambiente e de -20C, sendo posteriormente analisadas em intervalos de 1, 7, 15 e 35 dias após a aplicação do fungicida. Outras duas sub-amostras não tratadas com cloridrato de L-cisteina foram submetidas às mesmas condições de temperatura e analisadas nos mesmos intervalos de tempo. Na segunda etapa, foi efetuada a aplicação do fungicida MANZATE 800 (Dupont Brasil, 80% mancozebe) na dose recomendada de 3,0 Kg ha-1 e coletadas amostras do solo nas profundidades de 0-10, 10-20 e 20-40 cm em intervalos de 2,5,8,12,15,18 e 35 dias após aplicação. As amostras de cada profundidade foram tratadas com cloridrato de L-cisteina e acondicionadas sob temperatura de -20C. Através dos resultados obtidos na primeira etapa, pôde-se concluir que o tratamento com cisteina foi eficaz para conservação do analito, tanto para a amostra mantida a -20C quanto para a amostra mantida à temperatura ambiente. Os dados obtidos na segunda etapa do estudo mostraram que mancozebe apresentou comportamento semelhante ao descrito na literatura, para persistência no solo. Os resultados de lixiviação mostraram que nas condições pelas quais foi conduzido o experimento, resíduos de mancozebe foram detectados em profundidades de até 40 cm, porém através dos modelos de potencial de lixiviação, concluiu-se que o fungicida não oferece risco de contaminação de águas subterrâneas

The temporal expression and secretion of distinct members of a family of virulence-associated cathepsin L cysteine peptidases (FhCL) correlates with the entry and migration of the helminth pathogen Fasciola hepatica in the host. Thus, infective larvae traversing the gut wall secrete cathepsin L3 (FhCL3), liver migrating juvenile parasites secrete both FhCL1 and FhCL2 while the mature bile duct parasites, which are obligate blood feeders, secrete predominantly FhCL1 but also FhCL2.

The helminth parasite Fasciola hepatica secretes cathepsin L cysteine proteases to invade its host, migrate through tissues and digest haemoglobin, its main source of amino acids. Here we investigated the importance of pH in regulating the activity and functions of the major cathepsin L protease FheCL1. The slightly acidic pH of the parasite gut facilitates the auto-catalytic activation of FheCL1 from its inactive proFheCL1 zymogen; this process was approximately 40-fold faster at pH 4.5 than at pH 7.0. Active mature FheCL1 is very stable at acidic and neutral conditions (the enzyme retained approximately 45% activity when incubated at 37 degrees C and pH 4.5 for 10 days) and displayed a broad pH range for activity peptide substrates and the protein ovalbumin, peaking between pH 5.5 and pH 7.0. This pH profile likely reflects the need for FheCL1 to function both in the parasite gut and in the host tissues. FheCL1, however, could not cleave its natural substrate Hb in the pH range pH 5.5 and pH 7.0; digestion occurred only at pH

Il a été suggéré que l’autophagie pouvait participer au processus fibrotique en favorisant la différenciation du fibroblaste en myofibroblaste. La sénescence cellulaire a aussi été montrée comme impliquée dans la réparation tissulaire et la fibrose. Des liens ont été établis entre autophagie et sénescence. Cette étude a pour but d’investiguer les liens possibles entre autophagie, sénescence et différenciation myofibroblastique afin de mieux comprendre les mécanismes moléculaires régulant la réparation tissulaire et la fibrose. Les fibroblastes carencés en sérum pendant quatre jours montrent des ratios LC3B-II/-I élevés et des niveaux de SQSTM1/p62 diminués. L’augmentation de l’autophagie est accompagnée d’une augmentation de l’expression des marqueurs de différenciation myofibroblastique ACTA2/αSMA et collagènes de type 1 et 3 et de la formation de fibres de stress. Les fibroblastes autophagiques expriment les marqueurs de sénescence CDKN1A (p21) et p16INK4a (p16) et montrent une augmentation de l’activité beta-galactosidase associée à la sénescence. L’inhibition de l’autophagie à l’aide de différents inhibiteurs de phosphoinositide 3-kinase de classe I et de phosphatidylinositol 3-kinase de classe III (PtdIns3K) ou par inhibition génique à l’aide d’ARN interférant ATG7 bloquent l’expression des marqueurs de différenciation et de sénescence. L’expression et la sécrétion de CTGF (connective tissue growth factor) sont augmentées chez les fibroblastes autophagiques. L’inhibition de l’expression du CTGF par interférence génique prévient la différenciation myofibroblastique, démontrant l’importance de ce facteur pro-fibrotique pour la différenciation induite par l’autophagie. La phosphorylation de la kinase RPS6KB1/p70S6K, cible du complexe MTORC1, est abolie dans les fibroblastes autophagiques. La phosphorylation d’AKT à la Ser473, une cible du complexe MTORC2, diminue lors de la carence en sérum des fibroblastes mais est suivie d’une rephosphorylation après 2 jours. Ce résultat suggère la réactivation de MTORC2 lors d’une autophagie prolongée. Ceci a été vérifié par inhibition de l’autophagie dans les fibroblastes carencés en sérum. Les inhibiteurs de PtdIns3K et le siRNA ATG7 bloquent la rephosphorylation d’AKT. L’inhibition de la réactivation de MTORC2, et donc de la rephosphorylation d’AKT, est aussi obtenue par exposition des fibroblastes à la rapamycine, le Torin 1 ou par inhibition génique de RICTOR. Ces traitements inhibent l’augmentation de l’expression du CTGF ainsi que des marqueurs de différenciation et de sénescence, démontrant le rôle central joué par MTORC2 dans ces processus. Le stress oxydant peut induire la sénescence et la carence en sérum est connue pour augmenter la quantité de ROS (reactive oxygen species) dans les cellules. Afin d’investiguer le rôle des ROS dans la différenciation et la sénescence induites par l’autophagie, nous avons incubés les fibroblastes carencés en sérum en présence de N-acetyl-L-cysteine (NAC). Le NAC diminue la production de ROS, diminue les marqueurs d’autophagie, de sénescence et de différenciation myofibroblastique. Le NAC inhibe aussi la phosphorylation d’AKT Ser473. L’ensemble de ces résultats identifient les ROS en association avec une autophagie prolongée comme des nouveaux activateurs du complexe MTORC2. MTORC2 est central pour l’activation subséquente de la sénescence et de la différenciation myofibroblastique.

Rat kidney glutamine transaminase K (GTK) exhibits broad specificity both as an aminotransferase and as a cysteine S-conjugate beta-lyase. The beta-lyase reaction products are pyruvate, ammonium and a sulfhydryl-containing fragment. We show here that recombinant human GTK (rhGTK) also exhibits broad specificity both as an aminotransferase and as a cysteine S-conjugate beta-lyase. S-(1,1,2,2-Tetrafluoroethyl)-L-CySteine is an excellent aminotransferase and beta-lyase substrate of rhGTK. Moderate aminotransferase and beta-lyase activities occur with the chemopreventive agent Se-methyl-L-selenocysteine. L-3-(2-Naphthyl)alanine, L-3-(1-naphthyl)alanine, 5-S-L-cysteinyldopamine and 5-S-L-cysteinyl-L-DOPA are measurable aminotransferase substrates, indicating that the active site can accommodate large aromatic amino acids. The alpha-keto acids generated by transamination/L-amino acid oxidase activity of the two catechol cysteine S-conjugates are unstable. A slow rhGTK-catalyzed beta-elimination reaction, as measured by pyruvate formation, was demonstrated with 5-S-L-CysteinyIdopamine, but not with 5-S-L-CySteinyl-L-DOPA. The importance of transamination, oxidation and beta-elimination reactions involving 5-S-L-cysteinyldopamine, 5-S-L-cysteinyt-L-DOPA and Se-methyl-L-selenocysteirte in human tissues and their biological relevance are discussed.
