Resistance of Plasmodium falciparum to the usual antimalarials, as well as their adverse effects and high cost, has led to the search of new drugs against malaria. Several of these have been developed from medicinal plants based on ethnopharmacology, including the most widely used antimalarials today: quinine and artemisinin. In the present study schizonticide activity of extracts and fractions of a number of medicinal plants from the Caatinga and Amazon biomes were assessed based on ethnopharmacological and chemosystematic information. These included Ximenia americana, Maytenus rigida, Sideroxylon obtusifolium, Stryphnodendro coriaceum, Bowdichia virgiliodes, Schinopis brasiliensis and Picrolemma sprucei, the last, an Amazon species. Antimalarial tests of blood schizonticides were conducted in Swiss mice infected with P. berghei and in vitro against P. falciparum. In vitro cytotoxicity studies were carried out using HeLa, CHO, 3T3, Raw and HEPG2 cell lines. Except for X. americana, all species exhibited in vivo or in vitro antimalarial activity, inhibiting parasitic growth by up to 79%. Extracts exhibited moderate toxicity with dosedependent kinetics. In this sense, ethnopharmacological and chemosystematic approaches were shown to be useful and promising tools in the search of new drugs. These findings represent a significant contribution to scientific knowledge of the antimalarial potential of Brazilian flora, thereby opening perspectives for the development of new antimalarials

Studies have shown that Casearia sylvestris compounds protect DNA from damage both in vitro and in vivo. Complementarily, the aim of the present study was to assess the chemopreventive effect of casearin B (CASB) against DNA damage using the Ames test, the comet assay and the DCFDA antioxidant assay. The genotoxicity was assessed by the comet assay in HepG2 cells. CASB was genotoxic at concentrations higher than 0.30μM when incubated with the FPG (formamidopyrimidine-DNA glycosylase) enzyme. For the antigenotoxicity comet assay, CASB protected the DNA from damage caused by H2O2 in the HepG2 cell line in concentrations above 0.04μM with post-treatment, and above 0.08μM with pre-treatment. CASB was not mutagenic (Ames test) in TA 98 and TA 102. In the antimutagenicity assays, the compound was a strong inhibitor against aflatoxin B1 (AFB) in TA 98 (>88.8%), whereas it was moderate (42.7-59.4%) inhibitor against mytomicin C (MMC) in TA 102. Additionally, in the antioxidant assay using DCFDA, CASB reduced reactive oxygen species (ROS) generated by H2O2. In conclusion, CASB was genotoxic to HepG2 cells at high concentrations; was protective of DNA at low concentrations, as shown by the Ames test and comet assay; and was also antioxidant. © 2012 Elsevier Ltd.

Hymenoptera venoms are constituted by a complex mixture of chemically or pharmacologically bioactive agents, such as phospholipases, hyaluronidases and mastoparans. Venoms can also contain substances that are able to inhibit and/or diminish the genotoxic or mutagenic action of other compounds that are capable of promoting damages in the genetic material. Thus, the present study aimed to assess the effect of the venom of Polybia paulista, a neotropical wasp, by assays with HepG2 cells maintained in culture. The cytotoxic potential of the wasp venom, assessed by the methyl thiazolyl tetrazolium assay (MTT assay), was tested for the concentrations of 10μg/mL, 5μg/mL and 1μg/mL. As these concentrations were not cytotoxic, they were used to evaluate the genotoxic (comet assay) and mutagenic potential (micronucleus test) of the venom. In this study, it was verified that these concentrations induced damages in the DNA of the exposed cells, and it was necessary to test lower concentrations until it was found those that were not considered genotoxic and mutagenic. The concentrations of 1ng/mL, 100pg/mL and 10pg/mL, which did not induce genotoxicity and mutagenicity, were used in four different treatments (post-treatment, pre-treatment, simultaneous treatment with and without incubation), in order to evaluate if these concentrations were able to inhibit or decrease the genotoxic and mutagenic action of methyl methanesulfonate (MMS). None of the concentrations was able to inhibit and/or decrease the MMS activity. The genotoxic and mutagenic activity of the venom of P. paulista could be caused by the action of phospholipase, mastoparan and hyaluronidase, which are able to disrupt the cell membrane and thereby interact with the genetic material of the cells or even facilitate the entrance of other compounds of the venom that can act on the DNA. Another possible explanation for the genotoxicity and mutagenicity of the venom can be the presence of substances able to trigger inflammatory process and, consequently, generate oxygen reactive species that can interact with the DNA of the exposed cells. © 2013 Elsevier Ltd.

Die S-adenosyl-L-Homocysteinhydrolase (AHCY)-Defizienz ist eine seltene autosomal rezessive Erbkrankheit, bei der Mutationen im AHCY-Gen die Funktionsfähigkeit des kodierten Enzyms beeinträchtigen. Diese Krankheit führt zu Symptomen wie Entwicklungsverzögerungen, mentaler Retardierung und Myopathie. In der vorliegenden Arbeit wurde der Einfluss der AHCY-Defizienz auf die Methylierung der DNA in Blutproben und Fibroblasten von Patienten mit AHCY-Defizienz, sowie in HEK293- und HepG2-Zelllinien mit AHCY-Knockdown untersucht. Der gesamtgenomische Methylierungsstatus wurde mit Hilfe des MethylFlash ™ Methylated DNA Quantification Kit (Epigentek) bei drei Patienten-Blutproben festgestellt. In den Blutproben von sieben Patienten und Fibroblasten von einem Patienten wurde die Methylierung von DMRs sieben geprägter Gene (GTL2, H19, LIT1, MEST, NESPAS, PEG3, SNRPN) und zwei repetitiver Elemente (Alu, LINE1) mittels Bisulfit-Pyrosequenzierung quantifiziert und durch High Resolution Melting-Analyse bestätigt. Zusätzlich wurde eine genomweite Methylierungsanalyse mit dem Infinium® HumanMethylation450 BeadChip (Illumina) für vier Patientenproben durchgeführt und die Expression von AHCY in Fibroblasten mittels Expressions-qPCR und QUASEP-Analyse untersucht. Die Methylierungsanalysen ergaben eine Hypermethylierung der gesamtgenomischen DNA und stochastische Hypermethylierungen von DMRs geprägter Gene bei einigen Patienten. Die HEK293- und HepG2-Zelllinien wiesen dagegen hauptsächlich stochastische Hypomethylierungen an einigen DMRs geprägter Gene und LINE1-Elementen auf. Die genomweite Methylierungsarray-Analyse konnte die Ergebnisse der Bisulfit-Pyrosequenzierung nicht bestätigen. Die Expressionsanalysen der AHCY-defizienten Fibroblasten zeigten eine verminderte Expression von AHCY, wobei beide Allele etwa gleich stark transkribiert wurden. Die Ergebnisse deuten darauf hin, dass die AHCY-Defizienz eine gute Modellerkrankung für die Untersuchung biologischer Konsequenzen von Methylierungsstörungen im Rahmen der Epigenetik-Forschung sein könnte. Sie ist unseres Wissens die erste monogene Erkrankung mit symptomaler DNA-Hypermethylierung beim Menschen.

BACKGROUND ; AIMS: Hints, histidine triad nucleotide-binding proteins, are adenosine monophosphate-lysine hydrolases of uncertain biological function. Here we report the characterization of human Hint2. METHODS: Tissue distribution was determined by real-time quantitative polymerase chain reaction and immunoblotting, cellular localization by immunocytochemistry, and transfection with green fluorescent protein constructs. Enzymatic activities for protein kinase C and adenosine phosphoramidase in the presence of Hint2 were measured. HepG2 cell lines with Hint2 overexpressed or knocked down were established. Apoptosis was assessed by immunoblotting for caspases and by flow cytometry. Tumor growth was measured in SCID mice. Expression in human tumors was investigated by microarrays. RESULTS: Hint2 was predominantly expressed in liver and pancreas. Hint2 was localized in mitochondria. Hint2 hydrolyzed adenosine monophosphate linked to an amino group (AMP-pNA; k(cat):0.0223 s(-1); Km:128 micromol/L). Exposed to apoptotic stress, fewer HepG2 cells overexpressing Hint2 remained viable (32.2 +/- 0.6% vs 57.7 +/- 4.6%), and more cells displayed changes of the mitochondrial membrane potential (87.8 +/- 2.35 vs 49.7 +/- 1.6%) with more cleaved caspases than control cells. The opposite was observed in HepG2 cells with knockdown expression of Hint2. Subcutaneous injection of HepG2 cells overexpressing Hint2 in SCID mice resulted in smaller tumors (0.32 +/- 0.13 g vs 0.85 +/- 0.35 g). Microarray analyses revealed that HINT2 messenger RNA is downregulated in hepatocellular carcinomas (-0.42 +/- 0.58 log2 vs -0.11 +/- 0.28 log2). Low abundance of HINT2 messenger RNA was associated with poor survival. CONCLUSION: Hint2 defines a novel class of mitochondrial apoptotic sensitizers down-regulated in hepatocellular carcinoma.

Vitamin K antagonists such as warfarin inhibit the vitamin K-dependent γ-glutamyl carboxylation during protein processing and block the secretion of under-γ-carboxylated prothrombin (FII) in the rat but not in the human or bovine. Under-γ-carboxylated prothrombin is also secreted from warfarin-treated human (HepG2) cell cultures but is degraded in the endoplasmic reticulum in warfarin-treated rat (H-35) cell cultures. This differential response to warfarin has been shown to be determined by the structural difference in the proteins rather than by the origin of the cell line. When recombinant rat prothrombin (rFII) and human prothrombin (hFII) were expressed in a transformed human kidney cell line (HEK293), secretion of rFII but not hFII was drastically decreased in response to warfarin. To determine the structural signal required for this differential response, chimeric cDNAs with the propeptide/Gla domains, kringle domain, and serine protease domain exchanged between rFII and hFII were generated (FIIRHH and FIIHRR, FIIRRH and FIIHHR, FIIRHR and FIIHRH) and expressed in both warfarin-treated HEK293 cells and HepG2 cells. The presence of the hFII kringle domain changed the stability of rFII to that of hFII, and the rFII kringle domain changed the stability of hFII to that of rFII. The kringle domain therefore is critical in determining the metabolic fate of under-γ-carboxylated prothrombin precursors during processing. Prothrombin contains two kringle structures, and expression of additional rFII/hFII chimeras (FIIHrhH and FIIHhrH, FIIRrhR, and FIIRhrR) was used to determine that the first of the two kringles plays a more important role in the recognition process.

The thymidine analog fialuridine deoxy-2-fluoro-beta-D-arabinofuranosyl)-5-iodouracil (FIAU) was toxic in trials for chronic hepatitis B infection. One mechanism postulated that defective mtDNA replication was mediated through inhibition of DNA polymerase-gamma (DNA pol-gamma), by FIAU triphosphate (FIALTP) or by triphosphates of FIAU metabolites. Inhibition kinetics and primer-extension analyses determined biochemical mechanisms of FIAU, 1-(2-deoxy-2-fluoro-beta-D-arabinofuranosyl) -5-methyluracil (FAU), 1-(2-deoxy-2-fluoro-beta-D-arabinofuranosyl)uracil triphosphate (TP) inhibition of DNA pol-gamma. dTMP incorporation by DNA pol-gamma was inhibited competitively by FIAUTP, FMAUTP, and FAUTP (K1=0.015, 0.03, and 1.0 microM, respectively). By using oliginucleotide template-primers. DNA pol-gamma incorporated each analog into DNA opposite a single adenosine efficiently without effects on DNA chain elongation. Incorporation of multiple adjacent analogs at positions of consecutive adenosines dramatically impaired chain elongation by DNA pol-gamma. Effects of FIAU, FMAU, and FAU on HepG2 cell mmtDNA abundance and ultrastructure were determined. After 14 days, mtDNA decreased by 30% with 20 microM FIAU or 20 microM FMAU and decreased less than 10% with 100 microM FAU. FIAU and FMAU disrupted mitochondria and caused accumulation of intracytoplasmic lipid droplets. Biochemical and cell biological findings suggest that FIAU and its metabolites inhibit mtDNA replication, most likely at positions of adenosine tracts, leading to decreased mtDNA and mitochondrial ultrastructural defects.

De nouveaux modèles cellulaires in vitro par transfert de milieu et par coculture ont été mis au point afin d’évaluer la capacité des HDL à éliminer l’excès de cholestérol des tissus périphériques et de le transporter vers le foie afin d’être excrété par le foie, un processus nommé le transport inverse du cholestérol (TIC). Le système cellulaire par transfert in vitro où des macrophages J774 sont gorgés de LDL acétylées et marqués au 3H-cholestérol a été préalablement établi afin de mesurer par scintillation l’efflux de cholestérol marqué vers le milieu de culture contenant des accepteurs de cholestérol. Ce milieu conditionné est transféré sur des cellules HepG2 afin d’étudier l’influx du cholestérol marqué. Ce dernier nous permet d’observer un transport de cholestérol de 25 % hors des J774 et un transport de 39 000 cpm dans les HepG2 en utilisant un milieu contenant 2 % de sérums humains mis en commun. Une stimulation des cellules J774 par l’AMPc augmente l’efflux et l’influx d’environ 45 %. Des tests de preuve de concept ont été effectués sur le système cellulaire par co-culture qui utilise des chambres de Boyden où les J774 sont localisées au fond d’un puits et les HepG2 dans un insert, et où le milieu est partagé entre les deux types cellulaires. On a déterminé qu’une confluence densité de 60 000 cellules/cm2 sur un insert constitué d’une membrane de polyester avec des pores de 3,0 μm, sans autre revêtement, permet d’observer un influx spécifique au sérum d’environ 6 000 cpm associés aux cellules HepG2, où 50 % des comptes radioactifs sont dans les cellules et l’autre moitié présente à la surface cellulaire.
