Hookworms are hematophagous nematodes that infect a wide range of mammalian hosts, including humans. There has been speculation for nearly a century as to the identity of the anticoagulant substances) used by these organisms to subvert host hemostasis. Using molecular cloning, we describe a family of potent small protein (75-84 amino acids) anticoagulants from the hookworm Ancylostoma caninum termed AcAP (A. caninum anticoagulant protein). Two recombinant AcAP members (AcAP5 and AcAP6) directly inhibited the catalytic activity of blood coagulation factor Xa (fXa), while a third form (AcAPc2) predominantly inhibited the catalytic activity of a complex composed of blood coagulation factor VIIa and tissue factor (fVIIa/TF). The inhibition of fVIIa/TF was by a unique mechanism that required the initial formation of a binary complex of the inhibitor with fXa at a site on the enzyme that is distinct from the catalytic center (exo-site). The sequence of AcAPc2 as well as the utilization of an exo-site on fXa distinguishes this inhibitor from the mammalian anticoagulant TFPI (tissue factor pathway inhibitor), which is functionally equivalent with respect to fXa-dependent inhibition of fIIa/TF. The relative sequence positions of the reactive site residues determined for AcAP5 with the homologous regions in AcAP6 and AcAPc2 as well as the pattern of 10 cysteine residues present in each of the inhibitors suggest that the AcAPs are distantly related to the family of small protein serine protease inhibitors found in the nonhematophagous nematode Ascaris lumbricoides var. suum.

Vitamin E in the reduced, alpha-tocopherol form shows very modest anticlotting activity. By contrast, vitamin E quinone is a potent anticoagulant. This observation may have significance for field trials in which vitamin E is observed to exhibit beneficial effects on ischemic heart disease and stroke. Vitamin E quinone is a potent inhibitor of the vitamin K-dependent carboxylase that controls blood clotting. A newly discovered mechanism for the inhibition requires attachment of the active site thiol groups of the carboxylase to one or more methyl groups on vitamin E quinone. The results from a series of model reactions support this interpretation of the anticlotting activity associated with vitamin E.

As dietas foram divididas em quatro fases (pré-inicial I 22 a 28 dias; pré-inicial II 29 a 35 dias; inicial I - 36 a 47 dias e inicial II 48 a 63 dias).

Hox genes are essential for the patterning of the axial skeleton. Hox group 10 has been shown to specify the lumbar domain by setting a rib-inhibiting program in the presomitic mesoderm (PSM). We have now produced mice with ribs in every vertebra by ectopically expressing Hox group 6 in the PSM, indicating that Hox genes are also able to specify the thoracic domain. We show that the information provided by Hox genes to specify rib-containing and rib-less areas is first interpreted in the myotome through the regional-specific control of Myf5 and Myf6 expression. This information is then transmitted to the sclerotome by a system that includes FGF and PDGF signaling to produce vertebrae with or without ribs at different axial levels. Our findings offer a new perspective of how Hox genes produce global patterns in the axial skeleton and support a redundant nonmyogenic role of Myf5 and Myf6 in rib formation.

Changes in surface water hydrography in the Southern Ocean (eastern Atlantic sector) could be reconstructed on the basis of isotope-geochemical and micropaleontological studies. A total of 75 high quality multicorer sediment surface samples from the southern South Atlantic Ocean and three Quaternary sediment cores, taken on a meridional transect across the Antarctic Circumpolar Current, have been investigated. The results of examining stable oxygen isotope compositions of 24 foraminiferal species and morphotypes were compared to the near-surface hydrography. The different foraminifera have been divided into four groups living at different depths in the upper water column. The 8180 differences between shallow-living (e.g. G. bulloides, N. pachyderma) and deeper-dwelling (e. g. G. inflata) species reflect the measured temperature gradient of the upper 250 m in the water column. Thus, the 6180 difference between shallow-living and deeper-living foraminifera can be used as an indicator for the vertical temperature gradient in the surface water of the Antarctic Circumpolar Current, which is independent of ice volume. All planktonic foraminifera in the surface sediment samples have been counted. 27 species and morphotypes have been selected, to form a reference data Set for statistical purposes. By using R- and Q-mode principal component analysis these planktonic foraminifera have been divided into four and five assemblages, respectively. The geographic distribution of these assemblages is mainly linked to the temperature of sea-surface waters. The five assemblages (factors) of the Q-mode principal component analysis account for 97.l % of the variance of original data. Following the transferfunction- technique a multiple regression between the Q-mode factors and the actual mean sea-surface environmental parameters resulted in a set of equations. The new transfer function can be used to estimate past sea-surface seasonal temperatures for paleoassemblages of planktonic foraminifera with a precision of approximately ±1.2°C. This transfer function F75-27-5 encompasses in particular the environmental conditions in the Atlantic sector of the Antarctic Circumpolar Current. During the last 140,000 years reconstructed sea-surface temperatures fluctuated in the present northern Subantarctic Zone (PS2076-1/3) at an amplitude of up to 7.5°C in summer and of up to 8.5°C in winter. In the present Polarfrontal Zone (PS1754-1) these fluctuations between glacials and interglacials show lower temperatures from 2.5 to 8.5°C in summer and from 1.0 to 5.0°C in winter, respectively. Compared to today, calculated oxygen isotope temperature gradients in the present Subantarctic Zone were lower during the last 140,000 years. This is an indicator for a good mixing of the upper water column. In the Polarfrontal Zone also lower oxygen isotope temperature gradients were found for the glacials 6, 4 and 2. But almost similar temperature gradients as today were found during the interglacial stages 5, 3 and the Holocene, which implicates a mixing of the upper water column compared to present. Paleosalinities were reconstructed by combining d18O-data and the evaluated transfer function paleotemperatures. Especially in the present Polarfrontal Zone (PS1754-1) and in the Antarctic Zone (PS1768-8), a short-term reduction of salinity up to 4 %o, could be detected. This significant reduction in sea-surface water salinity indicates the increased influx of melt-water at the beginning of deglaciation in the southern hemisphere at the end of the last glacial, approximately 16,500-13,000 years ago. The reconstruction of environmental Parameters indicates only small changes in the position of the frontal Systems in the eastern sector of the Antarctic Circumpolar Current during the last 140,000 years. The average position of the Subtropical Front and Subantarctic Front shifted approximately three latitudes between interglacials and glacials. The Antarctic Polar Front shifted approximately four latitudes. But substantial modifications of this scenario have been interpreted for the reconstruction of cold sea-surface temperatures at 41Â S during the oxygen isotope stages 16 and 14 to 12. During these times the Subtropical Front was probably shified up to seven latitudes northwards.

We examine the quantitative composition of benthic foraminiferal assemblages of Rose Bengal-stained surface samples from 37 stations in the Laptev Sea, and combine this data set with an existing data set along a transect from Spitsbergen to the central Arctic Ocean. Foraminiferal test accumulation rates, diversity, faunal composition and statistically defined foraminiferal associations are analysed for living (Rose Bengal-stained) and dead foraminifers. We compare the results of several benthic foraminiferal diversity indices and statistically defined foraminiferal associations, including Fisher's alpha and Shannon-Wiener diversity indices, Q-mode principal component analysis and correspondence analysis. Diversity and faunal density (standing stock) of living benthic foraminifers are positively correlated to trophic resources. In contrast, the accumulation rate of dead foraminifers (BFAR) shows fluctuating values depending on test disintegration processes. Foraminiferal associations defined by Q-mode principal component analysis and correspondence analysis are comparable. The factor values of the correspondence analysis allow a quantitative correlation between the foraminiferal fauna and the local carbon flux, which may be used as a tool to estimate changes in primary productivity.

Application of quantitative micropaleontologic methods of sea-surface temperature (SST) estimation to the planktic foraminiferal faunas at Hole 548 in the northeast Atlantic between 3.5 and 2.0 Ma provide a detailed record of Pliocene surface temperature changes during the onset of major Northern Hemisphere glaciation. SST estimates indicate a relatively warm interval of "enhanced interglacials (warm peaks)" between 3.2 and 2.8 Ma followed by steady cooling with a significant step-like change toward colder temperatures occurring about 2.4 Ma. This record compares well with faunal and SST records from other North Atlantic sites.

Objective To determine the pharmacokinetics of carboplatin in sulphur-crested cockatoos, so that its use in clinical studies in birds can be considered. Design A pharmacokinetic study of carboplatin, following a single intravenous (IV) or intraosseus (10) infusion over 3 min, was performed in six healthy sulphur-crested cockatoos (Cacatua galerita). Procedure Birds were anaesthetised and a jugular vein cannulated for blood collection. Carboplatin (5 mg/kg) was infused over 3 min by the IV route in four birds via the contralateral jugular vein, and by the 10 route in two birds via the ulna. Serial blood samples were collected for 96 h after initiation of the infusion. Tissue samples from 11 organs were obtained at necropsy, 96 h after carboplatin administration. Total Pt and filterable Pt in plasma and tissue Pt concentrations were assayed by inductively coupled plasma-mass spectrometry. A noncompartmental pharmacokinetic analysis was performed on the plasma data. Results The mean +/- SD for the C-max of filterable Pt was 27.3 +/- 4.06 mg/L and in all six birds occurred at the end of the 3 min infusion, thenceforth declining exponentially over the next 6 h to an average concentration of 0.128 +/- 0.065 mg/L. The terminal half-life (T-1/2) was 1.0 +/- 0.17 h, the systemic clearance (CI) was 5.50 +/- 1.06 mL/min/kg and the volume of distribution (Vss) was 0.378 +/- 0.073 L/kg. The extrapolated area under the curve (AUC(0-x)) was 0.903 +/- 0.127 mg/mL.min; the area extrapolated past the last (6 h) data point to infinite time averaged only 1.25% of the total AUC(0-x). The kidneys had the greatest accumulation of Pt (7.04 +/- 3.006 mug/g), followed by the liver (3.08 +/- 1.785 mug/g DM). Conclusions and clinical relevance Carboplatin infusion in sulphur-crested cockatoos produced mild, transient alimentary tract signs and the Pt plasma concentration was similar whether carboplatin was given intravenously or intraosseously. Filterable plasma Pt concentrations for carboplatin persisted longer than for cisplatin, due mostly to the difference in systemic clearance between these drugs in sulphur-crested cockatoos. The distribution of tissue Pt after carboplatin administration was similar to that reported for cisplatin in sulphur-crested cockatoos. Despite anatomical, physiological and biochemical differences among animal species, the pharmacokinetic disposition of filterable Pt in the sulphur-crested cockatoo shares some features with the kinetics reported previously in other animals and human beings.

The aim of this study was to determine the most informative sampling time(s) providing a precise prediction of tacrolimus area under the concentration-time curve (AUC). Fifty-four concentration-time profiles of tacrolimus from 31 adult liver transplant recipients were analyzed. Each profile contained 5 tacrolimus whole-blood concentrations (predose and 1, 2, 4, and 6 or 8 hours postdose), measured using liquid chromatography-tandem mass spectrometry. The concentration at 6 hours was interpolated for each profile, and 54 values of AUC(0-6) were calculated using the trapezoidal rule. The best sampling times were then determined using limited sampling strategies and sensitivity analysis. Linear mixed-effects modeling was performed to estimate regression coefficients of equations incorporating each concentration-time point (C0, C1, C2, C4, interpolated C5, and interpolated C6) as a predictor of AUC(0-6). Predictive performance was evaluated by assessment of the mean error (ME) and root mean square error (RMSE). Limited sampling strategy (LSS) equations with C2, C4, and C5 provided similar results for prediction of AUC(0-6) (R-2 = 0.869, 0.844, and 0.832, respectively). These 3 time points were superior to C0 in the prediction of AUC. The ME was similar for all time points; the RMSE was smallest for C2, C4, and C5. The highest sensitivity index was determined to be 4.9 hours postdose at steady state, suggesting that this time point provides the most information about the AUC(0-12). The results from limited sampling strategies and sensitivity analysis supported the use of a single blood sample at 5 hours postdose as a predictor of both AUC(0-6) and AUC(0-12). A jackknife procedure was used to evaluate the predictive performance of the model, and this demonstrated that collecting a sample at 5 hours after dosing could be considered as the optimal sampling time for predicting AUC(0-6).

Objective: Sertraline's efficacy and tolerability in treating generalized anxiety disorder were evaluated. Method: Adult outpatients with DSM-IV generalized anxiety disorder and a total score of 18 or higher on the Hamilton Anxiety Rating Scale were eligible. After a 1-week single-blind placebo lead-in, patients were randomly assigned to 12 weeks of double-blind treatment with placebo (N=188, mean baseline anxiety score=25) or flexible doses (50-150 mg/day) of sertraline (N=182, mean anxiety score=25). The primary outcome measure was baseline-to-endpoint change in the Hamilton anxiety scale total score. A secondary efficacy measure was the Clinical Global Impression (CGI) improvement score; response was defined as a score of 2 or less. Results: Sertraline patients had significantly greater improvement than placebo patients on all efficacy measures at week 4. Analysis of covariance of the intent-to-treat group at endpoint (with the last observation carried forward) showed a significant difference in the decrease from baseline of the least-square mean total score on the Hamilton anxiety scale between sertraline (mean=11.7) and placebo (mean=8.0). Significantly greater endpoint improvement with sertraline than placebo was obtained for mean scores on the Hamilton anxiety scale psychic factor (6.7 versus 4.1) and somatic factor (5.0 versus 3.9). The rate of responders, based on CGI improvement and last observation carried forward, was significantly higher for sertraline (63%) than placebo (37%). Sertraline was well tolerated; 8% of patients versus 10% for placebo dropped out because of adverse events. Conclusions: Sertraline appears to be efficacious and well tolerated in the treatment of generalized anxiety disorder.
