本文采用Cole-Cole,Fuoss-Kirkwood经验相关时间分布模型和构象跳跃,高聚物局部主链运动阻尼取向扩散分子模型,对1,2-聚丁二烯在溶液中的~(13)C-NMR自旋-晶格弛豫时间nT_1和核Overhauser效应(NOE)值进行了数值拟合。用拟合所得分子运动参数讨论了1,2-聚丁二烯微观分子运动对链结构和温度的依赖性。

Polysiphonia urceolata R-phycoerythrin and Porphyridium cruentum B-phycoerythrin were degraded with proteinaseK, and then the nearly native gamma subunits were isolated from the reaction mixture. The process of degradation of phycoerythrin with proteinaseK showed that the gamma subunit is located in the central cavity of (alpha beta)(6) hexamer of phycoerythrin. Comparative analysis of the spectra of the native phycoerythrin, the phycoerythrin at pH 12 and the isolated gamma subunit showed that the absorption peaks of phycoerythrobilins on alpha or beta subunit are at 535 nm (or 545 nm) and 565 nm, the fluorescence emission maximum at 580 nm; the absorption peak of phycoerythrobilins on the isolated gamma subunit is at 589 nm, the fluorescence emission peak at 620 nm which overlaps the absorption maximum of C-phycocyanin and perhaps contributes to the energy transfer with high efficiency between phycoerythrin and phycocyanin in phycobilisome; the absorption maximum of phycourobilin on the isolated gamma subunit is at 498 nm, which is the same as that in native phycoerythrin, and the fluorescence emission maximum at 575 nm.

Porphyra yezoensis Ueda (Rhodophyta) is a seaweed of economic importance with a typical dimorphic life cycle consisting of a leafy gametophyte and a filamentous sporophyte. Recently, it has been recognized as a model system for fundamental and applied studies in marine biological sciences. Conchospore, a major spore linking the two distinct multicellular phases in the life cycle, is most widely used in the breeding of P. yezoensis. In this paper, the early-stage development of conchospore, including the attachment and the cell wall formation, was studied with fluorescent reagents staining and Scanning Electron Microscopy detection. Results displayed: (I) the cell wall began to be generated after culturing for 4 h in the attached conchospores; (2) the initially released conchospores were plastids with some filmy, amorphous substance on the surface, and they attached to the fibers firmly via the actively secreted mucilaginous substances after their touch to the fibers; (3) cellulase and pectolase prohibited the attachment of conchospores in the different ways; and (4) only attached conchospores generated cell walls and developed normally, while the suspending ones could not. It indicated that the cellulose played crucial roles in the permanent attachment as the pectin did in the initial attachment. The conchospore attachment seemed to trigger the cell wall formation and the further development. Affects of light on the development of conchospores were also discussed. The results showed that high intensity (200 mu mol.m(-2).s(-1)) and long-wave (>= 580 nm) light facilitated the division rate of conchospores. (C) 2008 Elsevier B.V. All rights reserved.

Pure C-phycocyanin was prepared from Spirulina platensis using one-step anion-exchange chromatography. The C-PC obtained was with an absorption maximum at 620 nm and a fluorescence emission maximum at 640 nm when excited by 580 nm. SPDP is an excellent heterobifunctional crosslinker for thiolating amines. Different molar ratios of SPDP have remarkable influence on the absorption and fluorescence spectra of C-phycocyanin. The absorption maximum and fluorescence emission maximum both decreased and blue-shifted from 640 run to 630 nm as the molar ratios of SPDP increased. It was found that the molar ratios of SPDP to C-phycocyanin was not more than 100 was appropriate to being conjugated with other biomolecules from the absorption and fluorescence spectra of C-phycocyanin.

R-phycoerythrin was isolated and purified from a red alga, Polysiphonia urceolata Grev, using Streamline column combined with ion-exchange chromatography or hydroxyapatite chromatography. The purity of R-phycoerythrin isolated by Streamline column was up to 1.66 and the yield of R-phycoerythrin could be as high as 0.68 mg/g frozen P. urceolata. All the eluates from Streamline column were divided into two equivalent parts, respectively. One part was pumped into the ion-exchange column loaded with Q-Sepharose and the other was applied to the adsorption column loaded with hydroxyapatite. The purities of R-phycoerythrin purified using these two methods were both up to 3.26, more than 3.2 the commonly accepted criterion. The yield of purified R-phycoerythrin from the ion-exchange chromatography was 0.40 mg/g frozen P. urceolata and that from the hydroxyapatite chromatography could reach 0.34 mg/g frozen P. urceolata. The purified protein had three absorption peaks at 498, 535, and 565 nm and displayed a fluorescence maximum at 580 nm, which was consistent with the typical spectrum of R-phycoerythrin. The purified R-PE was also identified with electrophoresis. Only one single protein band appeared on native-PAGE with silver staining. SDS-PAGE demonstrated the presence of one 20 kDa major subunit, and one low intensity band corresponding to 33 kDa subunit. The results indicate that using the expanded bed adsorption combined with ion-exchange chromatography or hydroxyapatite chromatography, R-phycoerythrin can be purified from frozen P. urceolata on large scale. The F1F0 ATP synthase has been purified from the hyperthermophilic eubacterium Aquifex aeolicus and characterized. Its subunits have been identified by MALDI-mass spectrometry through peptide mass fingerprinting and MS/MS. It contains the canonical subunits alpha, beta, gamma, delta and epsilon of F-1 and subunits a and c of F-0. Two versions of the b subunit were found, which show a low sequence homology to each other. Most likely they form a heterodimer. An electron microscopic single particle analysis revealed clear structural details, including two stalks connecting F-1 and F-0. In several orientations the central stalk appears to be tilted and/or kinked. It is unclear whether there is a direct connection between the peripheral stalk and the 6 subunit. 本论文研究了渤海,胶州湾和东海20-200 μm微型浮游动物(桡足类幼虫、纤毛虫)的数量和生物量(1% Lugol's 试剂固定1000ml,Utermohl法150倍显微镜检查),在渤海和东海用稀释法(Landry and Hassett, 1982; Burkill et al., 1990)估计了微型浮游动物(<200 μm)对浮游植物的摄食压力。对上述海区调查的时间如下:渤海:1997年6,1998年6月(莱州湾),1998年10月;胶州湾1997年8、11月,1998年2、4、8、11月,1999年2月;东海:1998年7月。结果如下:渤海:1997年6月,只发现一种砂壳纤毛虫,数量为0-980个/L。1998年6月(莱州湾),微型浮游动物数量为30-2390个/L,生物量为1.5-25 μg C/L,水体中生物量为0.1-53.2 mgC m~(-2)。共有13种砂壳纤毛虫和一种无壳纤毛虫。纤毛虫的数量为0-2380个/L,生物量为0-24.52 μg C/L。纤毛虫优势种有无壳纤毛虫、Tintinnopsis pallida、T. amoyensis和T. chinglanensis。微型浮游动物的各个类群呈斑块分布。1998年10月,微型浮游动物的数量为0-770个/L,生物量为0-12.3 μg C/L,水体中生物量为0-136 mgC m~(-2)。在两次大面调查间隔的6天内,发生大风。两次大面站调查海区的温盐分布格局没有很大变化,但微型浮游动物的变化却十分显著,这也反映了恶劣天气对海洋生物的影响。在几个连续站对微型浮游动物的连续监测表明,微型浮游动物的分布与水团有很大关系。胶州湾:桡足类幼虫在8月数量最大(850个/L)。共记录两种无壳纤毛虫和7种砂壳纤毛虫。微型浮游动物数量弯化很大,从10-22600个/L。微型浮游动物的分布格局可能是湾内和湾外海水混合的结果:1997年11月,无壳纤毛虫No.12从湾外涌入,其他航次无壳纤毛虫No.11在湾内发生,向湾外扩散。东海:微型浮游动物数量较少,最大数量为1970个/L,绝大多数站位低于300个/L,生物量为0.08-45 μg C/L,水体50m积分生物量为5-283mgC m~(-2)。微型浮游动物有明显的斑块分布,连续监测没有发现明显的变化。根据稀释培养的结果,微型浮游动物对浮游植物摄食压力如下所述:渤海三个航次的微型浮游动物对浮游植物摄食率为0.13-1.01d~(-1),每天摄食浮游植物生物量(现存量)的12-49%,初级生产力的34-267%。东海稀释培养的结果为微型浮游动物地浮游植物的摄食率为0.2-1.2d~(-1),每天摄食浮游动物生物量的20-71%,摄食初级生产力的54-98%。由于稀释培养实验的设计和操作有许多争议之外,本文只将结果列出,对方法不作讨论。

以奥利亚罗非鱼(Oreochromis aureus)为实验对象,设计了3种不同的摄食类型,分别是鲜活饵料组、饥饿3周后饱食投喂组和人工饲料组。鲜活饵料组投喂冰冻赤子爱胜蚓,利用蚯蚓体内丰富的营养成分和活性物质,以期获得奥利亚罗非鱼良好的生长状况;饥饿后饱食组是指饥饿3周后,以人工饲料饱食投喂2周,用于研究饥饿与补偿生长获得快速生长时血液理化指标的变化情况;人工饲料组作为对照组。纯淡水条件下养殖,水温25±2℃。测定了奥利亚罗非鱼在3种摄食类型饲喂下某些血液生理生化指标变化的情况,并将指标变化情况与增重率做相关性分析,试图找出能够反映奥利亚罗非鱼生长性能的血液生理生化指标。 研究结果表明,奥利亚罗非鱼在饥饿3周后获得了补偿生长,补偿生长时的增重率和特定生长率显著高于人工饲料组(P<0.05),高于鲜活饵料组,但差别不显著;相关性分析研究表明血清总蛋白、胆固醇、四碘甲状腺原氨酸(T4)与增重率极显著相关(P<0.01),血红蛋白显著相关(P<0.05),红细胞、白细胞、碱性磷酸酶高度相关(相关系数为0.580、0.551和0.557),因此,建议血清总蛋白、胆固醇和血红蛋白可作为能够反映罗非鱼生长性能的新指标。 根据序列设计引物,PCR反应条件:变性温度:95 ℃,3 min;退火温度:57℃,20 sec;延伸温度:72℃,5 min,共36个循环,从牙鲆、黑鲪和鲈鱼中克隆出胰岛素样生长因子(IGF-Ⅰ)部分序列,首次证实了IGF-Ⅰ在3种海水鱼中的存在。 利用蛋氨酸与ZnSO4•7H2O,在pH 5.5、80℃下,反应1小时,采用蛋氨酸与硫酸锌2:1的配料比,合成出了产物蛋氨酸螯合锌,蛋氨酸螯合锌外观白色,粉状,室温下微溶于水,不溶于乙醇,并用原子吸收光谱法测定其含锌量为15%,螯合率为88.2%。

紫菜是一类海洋藻类研究的模式系统,具有重要的经济价值和理论研究意义。本研究基于紫菜的世代交替生活史,对其壳孢子萌发过程进行了研究,并对丝状孢子体与叶状配子体的世代差异进行了分析,主要包括以下三部分: 1) 对紫菜丝状孢子体与叶状配子体的连接枢纽——壳孢子的发育过程及其光影响进行了研究与讨论,发现壳孢子只有在附着后才能形成细胞壁并发育,说明壳孢子的附着是触发了壳孢子细胞壁的形成以及后期发育的开关,亦即附着是触发紫菜世代交替过程中配子体转录组表达的开关;纤维素酶和果胶酶均能抑制壳孢子附着,但影响机制各不相同,推测果胶质主要介导壳孢子的初始附着,而纤维素则与永久附着相关;波长≥580 nm的高强度(200 μmol•m-2•s-1)可见光有利于壳孢子早期发育。 2) 结合现有藻类数据,对坛紫菜丝状孢子体阶段11000 EST数据进行了大规模的生物信息学分析,结果首次发现坛紫菜丝状孢子体中可能存在PCK型C4光合固碳途径,并筛选出44条在紫菜孢子体中表达上调的代表基因。 3) 结合坛紫菜、条斑紫菜、海带和红毛菜,对红藻和褐藻等大型海藻孢子体与配子体阶段代表基因Rubisco的表达与羧化酶活性差异进行了研究分析,结果表明Rubisco的表达量和初始羧化酶活在其配子体中均显著高于其孢子体世代,即与藻体不同世代的相对复杂度无关,而与染色体倍性相关,说明Rubisco的世代差异极有可能与染色体倍性相连锁,因而可能是海藻世代交替过程中的重要功能基因。

Nutrient dynamics and its influence on the distribution of chlorophyll-a in the upwelling area of the Changjiang (Yangtze) River estuary were investigated in the spring (May) and summer (August) of 2004. In the spring, upwelling was apparent in the region of 122 degrees 20'-123 degrees 00' E, 31 degrees 00'-32 degrees 00' N and was associated with low temperature (16-21 degrees C), high salinity (24-33 practical salinity units [psu]), and low dissolved oxygen (2.5-6.0 mg L-1) in the upper 10 m of the water column. The spring upwelling increased the mixed-layer phosphate, nitrate, and silicate concentrations to roughly 1, 15, and 15 mu mol L-1, respectively, and improved the light transparency in the euphotic zone. This improvement in phytoplankton growing conditions was followed by an increase in chlorophyll-a concentrations. The summer upwelling was weaker and occurred over a smaller geographical area (122 degrees 20'-123 degrees 00' E, 31 degrees 15'-31 degrees 50' N). Strongly influenced by turbid Changjiang diluted water (CDW), it had little impact on the upper 10 m of the water column but instead increased nutrient concentrations at greater depths. The high concentration of particulates in the CDW reduced light transmission in the upper 10 m and, hence, limited phytoplankton growth throughout the water column. Chlorophyll-a concentrations in the summer upwelling area were roughly an order of magnitude lower than in the spring. Water clarity, as influenced by the CDW, appears to be the principal factor limiting the impact of upwelling on phytoplankton biomass in this area.

Object To study the chemical constituents of Euphorbia wallichii.Methods The constituents were repeatedly separated and purified on silica gel column.They were identified on the basis of spectral methods.Results Nine diterpenoids were obtained from the roots of E. wallichii.Among them jolkinol B(I) is lathyrane type;caudicifolin (Ⅱ),helioscopinolides A(Ⅲ),C(Ⅳ),and E(Ⅴ) belong to abietane type;while ent-atisane-3β,16α,17-triol(Ⅵ),ent-16α,17-dihydroxyatisan-3-one(Ⅶ),ent-3β,(13S)-dihydroxyatis-16-en-14-one(Ⅷ),and ent-2-hydroxy-atis-1,16(17)-dien-3,14-dione(Ⅸ) possess an ent-atisane skeleon.Conclusion All of them are isolated from E. wallichii for the first time.

工业无线网络技术是继现场总线之后,工业控制领域的又一个热点技术,是降低工业测控系统成本、提高工业测控系统应用范围的革命性技术。无线HART规范作为第一个开放的工业无线网络国际标准,在满足工业应用苛刻的可靠性、实时性和安全性等关键需求的同时,还提供良好的向后兼容性,具有广阔的应用前景。网络管理器是无线HART网络的核心设备,是无线HART规范研究与开发的重点和难点。 本文在对工业无线网络技术、无线HART协议及其关键技术、无线HART网络管理器进行详细总结的基础上,从核心算法、软件的设计与实现等方面对无线HART网络管理器进行了研究,取得以下成果: (1)提出了一种基于有向图的独立多路径路由算法。算法包括路由发现和路由计算两个过程。路由发现中提出了基于负载均衡和信号强度的邻居过滤,以及邻居冗余等机制,保证了网络可靠性,降低了算法开销。路由计算中提出了基于双向链路筛选的拓扑构建,基于改进Dijkstra算法的最短路径生成,以及基于启发式搜索的独立多路径筛选等机制,满足了工业应用可靠、实时、低开销和低能耗等性能要求。 (2)提出了基于均衡搜索的通信资源分配算法。采用动态的信道黑名单技术,提高了网络的可靠性;保证所分资源均匀分布在搜索空间中,提高了算法应对网络动态性的能力;与主通信资源反序的冗余通信资源搜索过程,使得不同节点的资源均衡,保证了通信资源分配的公平性。此外,该算法还具有轻型、实时等优点。 (3)设计和实现了无线HART网络管理器软件。针对网络管理器功能复杂,性能要求高等问题,本文采用模块化软件设计方法设计和实现了无线HART网络管理器软件,并将其应用于辽宁省一大型纸业有限公司75吨流化床温度与压力监测系统中。网络管理器长期稳定运行、性能良好的事实表明了设计与实现的合理性。

提出了一种基于神经网络的逆控制的水下机器人解耦控制策略,解耦过程无需系统的精确数学模型,将变量间的耦合直接按照干扰处理,通过自适应逆控制中的扰动消除原理进行去祸设计。仿真结果表明,该算法原理简单、鲁棒性强、解耦效果良好。
