Topologische Beschränkungen beeinflussen die Eigenschaften von Polymeren. Im Rahmen dieser Arbeit wird mit Hilfe von Computersimulationen im Detail untersucht, inwieweit sich die statischen Eigenschaften von kollabierten Polymerringen, Polymerringen in konzentrierten Lösungen und aus Polymerringen aufgebauten Bürsten mit topologischen Beschränkungen von solchen ohne topologische Beschränkungen unterscheiden. Des Weiteren wird analysiert, welchen Einfluss geometrische Beschränkungen auf die topologischen Eigenschaften von einzelnen Polymerketten besitzen. Im ersten Teil der Arbeit geht es um den Einfluss der Topologie auf die Eigenschaften einzelner Polymerketten in verschiedenen Situationen. Da allerdings gerade die effiziente Durchführung von Monte-Carlo-Simulationen von kollabierten Polymerketten eine große Herausforderung darstellt, werden zunächst drei Bridging-Monte-Carlo-Schritte für Gitter- auf Kontinuumsmodelle übertragen. Eine Messung der Effizienz dieser Schritte ergibt einen Beschleunigungsfaktor von bis zu 100 im Vergleich zum herkömmlichen Slithering-Snake-Algorithmus. Darauf folgt die Analyse einer einzelnen, vergröberten Polystyrolkette in sphärischer Geometrie hinsichtlich Verschlaufungen und Knoten. Es wird gezeigt, dass eine signifikante Verknotung der Polystrolkette erst eintritt, wenn der Radius des umgebenden Kapsids kleiner als der Gyrationsradius der Kette ist. Des Weiteren werden sowohl Monte-Carlo- als auch Molekulardynamiksimulationen sehr großer Ringe mit bis zu einer Million Monomeren im kollabierten Zustand durchgeführt. Während die Konfigurationen aus den Monte-Carlo-Simulationen aufgrund der Verwendung der Bridging-Schritte sehr stark verknotet sind, bleiben die Konfigurationen aus den Molekulardynamiksimulationen unverknotet. Hierbei zeigen sich signifikante Unterschiede sowohl in der lokalen als auch in der globalen Struktur der Ringpolymere. Im zweiten Teil der Arbeit wird das Skalierungsverhalten des Gyrationsradius der einzelnen Polymerringe in einer konzentrierten Lösung aus völlig flexiblen Polymerringen im Kontinuum untersucht. Dabei wird der Anfang des asymptotischen Skalierungsverhaltens, welches mit dem Modell des “fractal globules“ konsistent ist, erreicht. Im abschließenden, dritten Teil dieser Arbeit wird das Verhalten von Bürsten aus linearen Polymeren mit dem von Ringpolymerbürsten verglichen. Dabei zeigt sich, dass die Struktur und das Skalierungsverhalten beider Systeme mit identischem Dichteprofil parallel zum Substrat deutlich voneinander abweichen, obwohl die Eigenschaften beider Systeme in Richtung senkrecht zum Substrat übereinstimmen. Der Vergleich des Relaxationsverhaltens einzelner Ketten in herkömmlichen Polymerbürsten und Ringbürsten liefert keine gravierenden Unterschiede. Es stellt sich aber auch heraus, dass die bisher verwendeten Erklärungen zur Relaxationsverhalten von herkömmlichen Bürsten nicht ausreichen, da diese lediglich den anfänglichen Zerfall der Korrelationsfunktion berücksichtigen. Bei der Untersuchung der Dynamik einzelner Monomere in einer herkömmlichen Bürste aus offenen Ketten vom Substrat hin zum offenen Ende zeigt sich, dass die Monomere in der Mitte der Kette die langsamste Relaxation besitzen, obwohl ihre mittlere Verrückung deutlich kleiner als die der freien Endmonomere ist.

La foca monaca (Monachus monachus, Hermann 1779) è il pinnipede più a rischio d’estinzione del mondo e pochi sono gli studi che hanno indagato l’impatto antropico dato dagli inquinanti chimici su questa specie. Questo studio si propone di analizzare la pressione che elementi in traccia, organoclorurati (OC) e, per la prima volta, idrocarburi policiclici aromatici (IPA) hanno sulla specie, studiando una della popolazioni più numerose presenti nel Mar Mediterraneo, ossia quella greca. Sono stati utilizzati campioni di 59 esemplari deceduti provenienti da diverse colonie residenti in Grecia, di ambedue i sessi, appartenenti a diverse classi d’età e riferibili ad un arco temporali di quasi 20 anni (1994-2013). Per i contaminanti organici (PCB, DDT ed IPA), caratterizzati da un comportamento lipofilico, sono stati utilizzati esclusivamente campioni di tessuto adiposo, mentre per l’analisi di elementi in traccia (essenziali: Fe, Co, Cu, Se, Zn, Mg; tossici: Cr, Ni, As, Cd, Pb, Hg) sono state impiegate diverse matrici, quali fegato, rene, muscolo, adipe, milza, polmone, cuore e pelo. I risultati evidenziano un’importante contaminazione dai OC (125.896,69±289.934,27 ng/g p.s.), in cui DDT e PCB sono presenti rispettivamente al 48,5% e 51,5%, mentre, tra i metalli considerati tossici (As, Cd, Pb, Hg, Cr, Ni), As e Hg sono gli elementi che destano maggiori preoccupazioni per la salute di Monachus monachus, presenti rispettivamente al 36,44% e 12,83%. IPA ed altri elementi in traccia analizzati invece non raggiungono di per sé concentrazioni di rischio per la specie, ma non si escludono possibili effetti sinergici tra i vari inquinanti. L’elaborazione del fingerprint tossicologico ha evidenziato un’eterogeneità nella pressione data dai contaminanti in studio, manifestando che alcune colonie di foca monaca in Grecia sono maggiormente soggette all’azione negativa di organoclorurati, idrocarburi policiclici aromatici o elementi in traccia tossici a seconda del caso.

Lo studio effettuato raccoglie informazioni al fine di svolgere un’analisi globale delle tecniche attualmente in uso, o in fase di studio, nel campo della localizzazione di dispositivi all’interno di un ambiente chiuso, ovvero laddove non è possibile sfruttare la copertura del sistema GPS. La panoramica è frutto dell’analisi e dello studio di paper tecnici pubblicati dai collaboratori dell’IEEE, fruibili all’interno del portale IEEE Xplore. A corredo di questo studio è stata sviluppata una applicazione per dispositivi Android basata sulla tecnica del Wi-Fi fingerprint; l’applicazione, che rappresenta un primo approccio alle tecniche di localizzazione, è a tutti gli effetti un sistema standalone per la localizzazione, consente cioè di costruire sia la mappa per la localizzazione, partendo da un ambiente sconosciuto, sia di ottenere la posizione dell’utente all’interno di una mappa conosciuta. La tesi si conclude con una analisi dei risultati e delle performance ottenute dall’applicazione in un uso comune, al fine di poter valutare l’efficacia della tecnica presa in considerazione. I possibili sviluppi futuri sono analizzati in un capitolo a parte e trovano spazio in ambienti nei quali si vogliono offrire servizi "context-based", ovvero basati sulla posizione esatta dell’utente.

Aerosol particles are important actors in the Earth’s atmosphere and climate system. They scatter and absorb sunlight, serve as nuclei for water droplets and ice crystals in clouds and precipitation, and are a subject of concern for public health. Atmospheric aerosols originate from both natural and anthropogenic sources, and emissions resulting from human activities have the potential to influence the hydrological cycle and climate. An assessment of the extent and impacts of this human force requires a sound understanding of the natural aerosol background. This dissertation addresses the composition, properties, and atmospheric cycling of biogenic aerosol particles, which represent a major fraction of the natural aerosol burden. The main focal points are: (i) Studies of the autofluo-rescence of primary biological aerosol particles (PBAP) and its application in ambient measure-ments, and (ii) X-ray microscopic and spectroscopic investigations of biogenic secondary organic aerosols (SOA) from the Amazonian rainforest.rnAutofluorescence of biological material has received increasing attention in atmospheric science because it allows real-time monitoring of PBAP in ambient air, however it is associated with high uncertainty. This work aims at reducing the uncertainty through a comprehensive characterization of the autofluorescence properties of relevant biological materials. Fluorescence spectroscopy and microscopy were applied to analyze the fluorescence signatures of pure biological fluorophores, potential non-biological interferences, and various types of reference PBAP. Characteristic features and fingerprint patterns were found and provide support for the operation, interpretation, and further development of PBAP autofluorescence measurements. Online fluorescence detection and offline fluorescence microscopy were jointly applied in a comprehensive bioaerosol field measurement campaign that provided unprecedented insights into PBAP-linked biosphere-atmosphere interactions in a North-American semi-arid forest environment. Rain showers were found to trigger massive bursts of PBAP, including high concentrations of biological ice nucleators that may promote further precipitation and can be regarded as part of a bioprecipitation feedback cycle in the climate system. rnIn the pristine tropical rainforest air of the Amazon, most cloud and fog droplets form on bio-genic SOA particles, but the composition, morphology, mixing state and origin of these particles is hardly known. X-ray microscopy and spectroscopy (STXM-NEXAFS) revealed distinctly different types of secondary organic matter (carboxyl- vs. hydroxy-rich) with internal structures that indicate a strong influence of phase segregation, cloud and fog processing on SOA formation, and aging. In addition, nanometer-sized potassium-rich particles emitted by microorganisms and vegetation were found to act as seeds for the condensation of SOA. Thus, the influence of forest biota on the atmospheric abundance of cloud condensation nuclei appears to be more direct than previously assumed. Overall, the results of this dissertation suggest that biogenic aerosols, clouds and precipitation are indeed tightly coupled through a bioprecipitation cycle, and that advanced microscopic and spectroscopic techniques can provide detailed insights into these mechanisms.rn

Hefen der Gattung Saccharomyces und Milchsäurebakterien sind bei der Weinbereitung von besonderer Bedeutung. Neben der alkoholischen Gärung sind Hefen an der Ausbildung von Aromastoffen beteiligt. Milchsäurebakterien spielen eine Rolle beim biologischen Säureabbau (malolaktische Fermentation), können jedoch aufgrund ihrer Stoffwechseleigenschaft weitere Aromamodifikationen bewirken. Die Zusammensetzung der mikrobiellen Flora zu verschiedenen Zeitpunkten der Weinbereitung hat einen direkten Einfluss auf die Qualität der Weine, welche sich sowohl positiv als auch negativ verändern kann. Daher ist die zuverlässige Identifizierung und Differenzierung verschiedener Mikroorganismen auf Art- aber auch Stamm-Ebene während der Vinifikation von Bedeutung.rnDer erste Teil dieser Arbeit beschäftigte sich mit der Differenzierung von Hefearten der Gattung Saccharomyces, welche mit Hilfe konventioneller Methoden nicht eindeutig identifiziert werden können. Unter Verwendung des DNA-Fingerprintverfahrens Specifically Amplified Polymorphic DNA (SAPD)-PCR sowie der Matrix-Assisted-Laser-Desorption/Ionization-Time-Of-Flight-Mass-Spectrometry (MALDI-TOF-MS) war eine Differenzierung dieser taxonomisch sehr nah verwandten Arten möglich. Weiterhin konnten interspezifische Hybridstämme detektiert werden. In diesem Zusammenhang wurde der Hybridcharakter des Stammes NCYC 3739 (S. cerevisiae x kudriavzevii) entdeckt. Um die Elternspezies eines Hybridstamms zuverlässig zu bestimmen, sind weiterführende Genanalysen notwendig. Hierzu konnte eine Restriktionsfragmentlängenpolymorphismus (RFLP)-Analyse verschiedener genetischer Marker erfolgreich herangezogen werden.rnIm Rahmen dieser Arbeit wurde weiterhin ein Schnellidentifizierungssystem zum Nachweis weinrelevanter Milchsäurebakterien entwickelt. Mit Hilfe der Sequence Characterized Amplified Region (SCAR)-Technik konnten artspezifische Primer generiert werden, welche auf der Grundlage charakteristischer Fragmente der SAPD-PCR abgeleitet wurden. Durch die Anwendung dieser Primer in einer Multiplex-PCR-Reaktion war die Detektion verschiedener, einerseits häufig in Wein vorkommender und andererseits potentiell an der Ausbildung von Weinfehlern beteiligter Milchsäurebakterien-Arten möglich. Die ermittelte Nachweisgrenze dieser Methode lag mit 10^4 - 10^5 Zellen/ml im Bereich der Zelltiter, die in Most und Wein anzutreffen sind. Anhand der Untersuchung verschiedener Weinproben von Winzern in Rheinhessen wurde die Praxistauglichkeit dieser Methode demonstriert. rnUm die gesamten Milchsäurebakterien-Population im Verlauf der Weinbereitung zu kontrollieren, kann die Denaturierende Gradienten-Gelelektrophorese herangezogen werden. Hierzu wurden in dieser Arbeit Primer zur Amplifikation eines Teilbereichs des rpoB-Gens abgeleitet, da dieses Gen eine Alternative zur 16S rDNA darstellt. Die DNA-Region erwies sich als geeignet, um zahlreiche weinrelevante Milchsäurebakterien-Arten zu differenzieren. In einigen ersten Versuchen konnte gezeigt werden, dass diese Methode für eine praktische Anwendung in Frage kommt.rnOenococcus oeni ist das wichtigste Milchsäurebakterien während der malolaktischen Fermentation und wird häufig in Form kommerzieller Starterkulturen eingesetzt. Da verschiedene Stämme unterschiedliche Eigenschaften aufweisen können, ist es von Bedeutung, die Identität eines bestimmten Stammes zweifelsfrei feststellen zu können. Anhand der Analyse verschiedener O. oeni-Stämme aus unterschiedlichen Weinbaugebieten konnte gezeigt werden, dass sowohl die nested SAPD-PCR als auch die MALDI-TOF-MS genügend Sensitivität aufweisen, um eine Unterscheidung auf Stamm-Ebene zu ermöglichen, wobei die mittels nSAPD-PCR ermittelten Distanzen der Stämme zueinander mit deren geographischer Herkunft korrelierte.rnDie in der vorliegenden Arbeit entwickelten Methoden können dazu beitragen, den Prozess der Weinherstellung besser zu kontrollieren und so eine hohe Qualität des Endproduktes zu gewährleisten.rn

Hintergrund: Miniaturisierung ist ein häufig beobachtetes Phänomen bei Pflanzen in arktisch-alpinen Lebensräumen und wird als Anpassung an niedrige Jahresmitteltemperaturen und eine kurze Vegetationsperiode interpretiert. Ziele: In der vorliegenden Arbeit wird im Petasites-Clade (Petasites Mill., Endocellion Turcz. ex Herder, Homogyne Cass., Tussilago L.; Asteraceae) und in Soldanella (Primulaceae) die Evolution der Miniaturisierung arktisch-alpiner Arten untersucht. Zudem wird innerhalb von Homogyne untersucht, ob unterschiedliche edaphische Präferenz von H. alpina (variabel) und H. discolor (kalkliebend) genetisch fixiert ist. rnMethoden: Molekulare Phylogenien des Petasites-Clades und von Soldanella wurden mit nukleären und plastidären Markern erstellt, und mit den in den Alpen vorkommenden Soldanella-Arten wurde zudem eine Fingerprint-Studie (AFLPs) gemacht. Zur Datierung der Diversifizierungsereignisse im Petasites-Clade diente eine molekulare Uhr, und die Evolution von Miniaturisierung wurde rekonstruiert. Mit H. alpina und H. discolor wurde ein vergleichendes Kulturexperiment durchgeführt.rnErgebnisse: Miniaturisierung entstand mehrere Male unabhängig voneinander in den arktisch-alpinen Vertretern des Petasites-Clade, aber nicht alle arktisch-alpinen Arten sind klein. Das Alter der arktisch-alpinen Arten deutet darauf hin, dass diese Taxa ihren Ursprung in der arkto-tertiären Flora haben. In Soldanella sind reduzierte Blütenmorphologie sowie Kleinwüchsigkeit der beiden alpinen Arten zweimal parallel entstanden. Homogyne alpina und H. discolor zeigen keine edaphischen Unterschiede hinsichtlich des Keimverhaltens, aber in Kultur zeigt sich, dass die Präferenz von H. discolor für Kalk wahrscheinlich genetisch fixiert ist.rnSchlussfolgerungen: Miniaturisierung von Pflanzen in größerer Höhe und höherer geographischer Breite kann in der Regel beobachtet werden. Allerdings kann die Evolution arktisch-alpiner Arten auch durch Faktoren wie Nährstoffverfügbarkeit, Konkurrenz und Störung beeinflusst werden, die dem Effekt der Temperatur entgegenwirken, so dass nicht alle Pflanzen in arktisch-alpinen Habitaten klein sind. Blütenmorphologische Reduktion in Soldanella kann als Anpassung an einen höheren Grad an Selbstbestäubung interpretiert werden, um eine geringere Bestäuberaktivität im alpinen Lebensraum zu kompensieren.

Questa tesi ha come scopo principale l'analisi delle diverse tecnologie di localizzazione in ambito indoor, analizzando in particolare l'utilizzo del Wifi RSS Fingerprinting. La tecnica del Wifi RSS Fingerprinting è una tecnica per la localizzazione all'interno di ambienti chiusi, che consiste nella definizione di un 'impronta'(fingerprint) in un punto preciso dell'ambiente(definito reference point), andando a inserire in un database i valori di potenza del segnale ricevuto(RSS) da ogni access point rilevato all'interno di quel determinato reference point. Per l'implementazione di questa tecnica è stato sviluppato un applicativo con un architettura client-server. Il client è stato sviluppato in ambiente Android, realizzando una applicazione per la gestione della fase di salvataggio di nuovi fingerprint e per la fase di localizzazione della posizione corrente, tramite l'utilizzo dei vari fingerprint precedentemente inseriti all'interno del DB. Il server, sviluppato in Node.js(framework Javascript), gestirà le diverse richieste ricevute dal client tramite delle chiamate AJAX, prelevando le informazioni richieste direttamente dal database. All'interno delle applicativo sono stati implementati diversi algoritmi per la localizzazione indoor, in modo da poter verificare l'applicabilità di questo sistema in un ambito reale. Questi algoritmi sono stati in seguito testati per valutare l'accuratezza e la precisione di ciascuno, andando ad individuare gli algoritmi migliori da utilizzare in base a scenari diversi.

Questo elaborato studia e analizza il comportamento di tre algoritmi di base per quanto riguarda la localizzazione indoor e in particolare la tecnica del fingerprint. L'elaborato include l'analisi di come l'eterogeneita dei dispositivi possa influenzare gli algoritmi e la loro accuratezza nel produrre il risultato. Si include inoltre l'analisi dello stato dell'arte la progettazione e lo sviluppo di un'applicazione Android e di un web service. L'illustrazione dei test effettuati e le considerazioni finali concludono la tesi.

A new multimodal biometric database designed and acquired within the framework of the European BioSecure Network of Excellence is presented. It is comprised of more than 600 individuals acquired simultaneously in three scenarios: 1) over the Internet, 2) in an office environment with desktop PC, and 3) in indoor/outdoor environments with mobile portable hardware. The three scenarios include a common part of audio/video data. Also, signature and fingerprint data have been acquired both with desktop PC and mobile portable hardware. Additionally, hand and iris data were acquired in the second scenario using desktop PC. Acquisition has been conducted by 11 European institutions. Additional features of the BioSecure Multimodal Database (BMDB) are: two acquisition sessions, several sensors in certain modalities, balanced gender and age distributions, multimodal realistic scenarios with simple and quick tasks per modality, cross-European diversity, availability of demographic data, and compatibility with other multimodal databases. The novel acquisition conditions of the BMDB allow us to perform new challenging research and evaluation of either monomodal or multimodal biometric systems, as in the recent BioSecure Multimodal Evaluation campaign. A description of this campaign including baseline results of individual modalities from the new database is also given. The database is expected to be available for research purposes through the BioSecure Association during 2008.

The NIMH's new strategic plan, with its emphasis on the "4P's" (Prediction, Pre-emption, Personalization, and Populations) and biomarker-based medicine requires a radical shift in animal modeling methodology. In particular 4P's models will be non-determinant (i.e. disease severity will depend on secondary environmental and genetic factors); and validated by reverse-translation of animal homologues to human biomarkers. A powerful consequence of the biomarker approach is that different closely related disorders have a unique fingerprint of biomarkers. Animals can be validated as a highly specific model of a single disorder by matching this 'fingerprint'; or as a model of a symptom seen in multiple disorders by matching common biomarkers. Here we illustrate this approach with two Abnormal Repetitive Behaviors (ARBs) in mice: stereotypies and barbering (hair pulling). We developed animal versions of the neuropsychological biomarkers that distinguish human ARBs, and tested the fingerprint of the different mouse ARBs. As predicted, the two mouse ARBs were associated with different biomarkers. Both barbering and stereotypy could be discounted as models of OCD (even though they are widely used as such), due to the absence of limbic biomarkers which are characteristic of OCD and hence are necessary for a valid model. Conversely barbering matched the fingerprint of trichotillomania (i.e. selective deficits in set-shifting), suggesting it may be a highly specific model of this disorder. In contrast stereotypies were correlated only with a biomarker (deficits in response shifting) correlated with stereotypies in multiple disorders, suggesting that animal stereotypies model stereotypies in multiple disorders.

INTRODUCTION The ATP-binding cassette (ABC) transporter A1 (ABCA1) and ABCG1 are highly expressed in the placenta in various compartments, including the villous syncytiotrophoblast (V-STB) and foetal endothelial cells. Among other not yet characterized functions, they play a role in the foeto-maternal transport of cholesterol and other lipophilic molecules. In humans, preliminary data suggest expressional changes of ABCA1 and ABCG1 in pathologic gestation, particularly under hypoxic conditions, but a systematic expression analysis in common human pregnancy diseases has never been performed. OBJECTIVES The aim of the present study was to characterize ABCA1 and ABCG1 expression in a large series of pathologic placentas, in particular from preeclampsia (PE) and intrauterine growth restriction (IUGR) which are associated with placental hypoxia. METHODS Placentas from 152 pathological pregnancies, including PE and/or HELLP (n=24) and IUGR (n=21), and 20 normal control placentas were assessed for their ABCA1 and ABCG1 mRNA and protein expression with quantitative RT-PCR and semi-quantitative immunohistochemical analysis, respectively. RESULTS ABCA1 protein expression in the V-STB was significantly less extensive in PE compared with normal controls (<10% of V-STB stained for ABCA1 in 58% PE placentas vs. 25% controls; p=0.035). Conversely, it was significantly more wide-spread in IUGR (>75% of V-STB stained in 57% IUGR placentas vs. 15% controls; p=0.009). Moreover, there was an insignificant trend for increased ABCA1 expression in fetal endothelial cells of stem villi in PE (p=0.0588). ABCA1 staining levels in V-STB were significantly associated with placental histopathological features related with hypoxia: they were decreased in placentas exhibiting syncytial knotting (p=0.033) and decidual vasculopathy (p=0.0437) and increased in low weight placentas (p=0.015). The significant and specific alterations in ABCA1 protein expression found at a specific cellular level were not paralleled by changes in ABCA1 mRNA abundance of total placental tissue. ABCG1 staining was universally extensive in the V-STB of normal placentas, always affecting more than 90% of V-STB surface. In comparison, ABCG1 staining of the V-STB was generally often reduced in pregnancy diseases. In particular, less than 90% of V-STB exhibited ABCG1 staining in 26% of PE placentas (p=0.022) and 35% of IUGR placentas (p=0.003). Similarly to ABCA1, ABCG1 mRNA expression in total placental tissue was not significantly different between controls and PE or IUGR. CONCLUSION ABCA1 and ABCG1 proteins are differentially expressed, with either down- or up-regulation, in the V-STB of placentas exhibiting features of chronic hypoxia, such as in PE and IUGR. This suggests that other factors in addition to hypoxia regulate the expression of placental lipid transporters. The specific changes on a cellular level were masked when only total tissue mRNA was analysed underlining the importance of cell specific expression analysis. The potential effects of decreased placental ABCA1 and ABCG1 expression on foetal nutrition and development remain to be elucidated.

Intrauterine growth restriction (IUGR) is defined as a condition in which the fetus does not reach its genetically given growth potential, resulting in low birth weight. IUGR is an important cause of perinatal morbidity and mortality, thus contributing substantially to medically indicated preterm birth in order to prevent fetal death. We subjected umbilical cord blood serum samples either belonging to the IUGR group (n = 15) or to the control group (n = 15) to fractionation by affinity chromatography using a bead system with hydrophobic interaction capabilities. So prepared protein mixtures were analyzed by MALDI-TOF mass spectrometric profiling. The six best differentiating ion signals at m/z 8205, m/z 8766, m/z 13 945, m/z 15 129, m/z 15 308, and m/z 16 001 were collectively assigned as IUGR proteome signature. Separation confidence of our IUGR proteome signature reached a sensitivity of 0.87 and a specificity of 0.93. Assignment of ion signals in the mass spectra to specific proteins was substantiated by SDS-PAGE in conjunction with peptide mass fingerprint analysis of cord blood serum proteins. One constituent of this proteome signature, apolipoprotein C-III(0) , a derivative lacking glycosylation, has been found more abundant in the IUGR cord blood serum samples, irrespective of gestational age. Hence, we suggest apolipoprotein C-III(0) as potential key-marker of the here proposed IUGR proteome signature, as it is a very low-density lipoprotein (VLDL) and high-density lipoprotein (HDL) member and as such involved in triglyceride metabolism that itself is discussed as being of importance in IUGR pathogenesis. Our results indicate that subtle alterations in protein glycosylation need to be considered for improving our understanding of the pathomechanisms in IUGR.

Moraxella catarrhalis is a major mucosal pathogen of the human respiratory tract both in children and in adults. Two subpopulations of this organism have been described that differ in 16S rRNA gene sequence and virulence traits. Three 16S rRNA types have been defined. 2-DE followed by protein identification by MS revealed significant differences in the outer membrane protein (OMP) patterns of each M. catarrhalis 16S rRNA type. Approximately 130 features were detected on the 2-DE map of each M. catarrhalis 16S rRNA type. However, only 50 features were expressed by all strains. Furthermore, direct profiling of isolated OMP using MALDI-TOF MS resulted in a characteristic spectral fingerprint for each 16S rRNA type. Fingerprints remained identical when intact cells instead of isolated OMP were analyzed. This finding suggests that the source of desorbed ions is the outer membrane. Based on the fingerprint we were able to assign 18 well-characterized clinical M. catarrhalis isolates to the correct subpopulation. Therefore, MALDI-TOF of intact M. catarrhalis provides a rapid and robust tool for M. catarrhalis strain typing that could be applied in epidemiological studies.

In Switzerland, a national database with 1028 Campylobacter isolates from poultry, pigs, cats, dogs, cattle, humans, zoo animals and water has been created. The database contains the genetic fingerprint and background information of each Campylobacter isolate. Dominant species could be identified in the different sources with a majority of Campylobacter jejuni in poultry (73%), humans (79%), cattle (95%), zoo animals (40%) and water (100%), of Campylobacter coli in pigs (72%), and of Campylobacter upsaliensis/helveticus in cats and dogs (55%). The comparison of three genotyping methods, amplified fragment length polymorphism (AFLP), pulsed field gel electrophoresis and restriction fragment length polymorphism, revealed that AFLP allows discrimination between the different Campylobacter species and is the most appropriate method to distinguish specific strains within the same species. Genotyping analysis demonstrated that the Campylobacter population is heterogeneous among the different sources and that no dominant clone is spread in the country. Genotyping and the resulting database are useful tools to trace back future Campylobacter infections.

This book will serve as a foundation for a variety of useful applications of graph theory to computer vision, pattern recognition, and related areas. It covers a representative set of novel graph-theoretic methods for complex computer vision and pattern recognition tasks. The first part of the book presents the application of graph theory to low-level processing of digital images such as a new method for partitioning a given image into a hierarchy of homogeneous areas using graph pyramids, or a study of the relationship between graph theory and digital topology. Part II presents graph-theoretic learning algorithms for high-level computer vision and pattern recognition applications, including a survey of graph based methodologies for pattern recognition and computer vision, a presentation of a series of computationally efficient algorithms for testing graph isomorphism and related graph matching tasks in pattern recognition and a new graph distance measure to be used for solving graph matching problems. Finally, Part III provides detailed descriptions of several applications of graph-based methods to real-world pattern recognition tasks. It includes a critical review of the main graph-based and structural methods for fingerprint classification, a new method to visualize time series of graphs, and potential applications in computer network monitoring and abnormal event detection.
