Mitochondria are central organelles for cell survival with particular relevance in energy production and signalling, being mitochondrial fatty acid β–oxidation (FAO) one of the metabolic pathways harboured in this organelle. FAO disorders (FAOD) are among the most well studied inborn errors of metabolism, mainly due to their impact in health. Nevertheless, some questions remain unsolved, as their prevalence in certain European regions and how pathophysiological determinants combine towards the phenotype. Analysis of data from newborn screening programs from Portugal and Spain allowed the estimation of the birth prevalence of FAOD revealing that this group of disorders presents in Iberia (and particularly in Portugal) one of the highest European birth prevalence, mainly due to the high birth prevalence of medium chain acyl-CoA dehydrogenase deficiency. These results highlight the impact of this group of genetic disorders in this European region. The characterization of mitochondrial proteome, from patients fibroblasts with FAOD, namely multiple acyl-CoA dehydrogenase deficiency (MADD) and long chain acyl-CoA dehydrogenase deficiency (LCHADD), provided a global perspective of the mitochondrial proteome plasticity in these disorders and highlights the main molecular pathways involved in their pathogenesis. Severe MADD forms show an overexpression of chaperones, antioxidant enzymes (MnSOD), and apoptotic proteins. An overexpression of glycolytic enzymes, which reflects cellular adaptation to energy deficiency due to FAO blockage, was also observed. When LCHADD fibroblasts were analysed a metabolic switching to glycolysis was also observed with overexpression of apoptotic proteins and modulation of the antioxidant defence system. Severe LCHADD present increased ROS alongside with up regulation of MnSOD while moderate forms have lower ROS and down-regulation of MnSOD. This probably reflects the role of MnSOD in buffering cellular ROS, maintain them at levels that allow cells to avoid damage and start a cellular response towards survival. When ROS levels are very high cells have to overexpress MnSOD for detoxifying proposes. When severe forms of MADD were compared to moderate forms no major differences were noticed, most probably because ROS levels in moderate MADD are high enough to trigger a response similar to that observed in severe forms. Our data highlights, for the first time, the differences in the modulation of antioxidant defence among FAOD spectrum. Overall, the data reveals the main pathways modulated in FAOD and the importance of ROS levels and antioxidant defence system modulation for disease severity. These results highlight the complex interaction between phenotypic determinants in FAOD that include genetic, epigenetic and environmental factors. The development of future better treatment approaches is dependent on the knowledge on how all these determinants interact towards phenotype.!

Enquadramento: O ‘Physiosensing’ (‘PhyS’) é um dispositivo médico destinado ao treino do controlo postural nas posições de sentado e de pé, bem como no levantar e sentar, possibilitando também a avaliação do desempenho a este nível. Este trabalho teve como objetivo avaliar a fiabilidade e a validade da plataforma ‘Physiosensing’ na avaliação do equilíbrio em pessoas com deficiência intelectual (PCDI). Métodos: Para o grupo experimental (GE) foram recrutados 47 indivíduos com deficiência intelectual e para o grupo de controlo (GC) 39 indivíduos sem deficiência intelectual, provenientes da região do BaixoMondego. A avaliação da fiabilidade incluiu as análises da concordância entre observadores, reprodutibilidade temporal e consistência interna. A análise fatorial exploratória analisou os pressupostos de subdomínios propostos pelos autores. A validade discriminante foi analisada através da comparação de resultados entre o GE e o GC, e a validade concorrente pela análise dos valores de associação entre os resultados do ‘PhyS’ com a Escala de Equilíbrio de Berg (EEB). Resultados: O subconjunto de exercícios relacionados com a transferência de peso lateralmente (TPL) apresentou os resultados mais elevados a nível da concordância entre observadores (0,40 ≤ CCI > 0,75) e na reprodutibilidade intemporal (CCI ≥ 0,75). O instrumento apresenta uma consistência interna fraca (α = 0,63) quando considerados todos os exercícios, tendo-se obtido o melhor resultado para o subconjunto de exercícios TPL (α = 0,81). A análise fatorial exploratória devolveu quatro fatores, explicando 76,4% da variância, agrupando no primeiro fator o subconjunto de exercícios TPL. Foram encontradas diferenças estatisticamente significativas entre os resultados dos participantes com e sem deficiência intelectual, em dez dos onze exercícios que compõem a configuração da plataforma. Seis exercícios, que incluem os exercícios TPL, apresentam valores de associação estatisticamente significativos com a EEB. Conclusão: Alguns exercícios da plataforma ‘PhyS’ não se mostram adequados para medir o equilíbrio em PCDI, não podendo ser incluídos numa medida global. No entanto, os exercícios TPL poderão constituir um indicador global do equilíbrio em pessoas com PCDI. Recomenda-se a definição de procedimentos de medição de forma a melhorar os índices de fiabilidade, o aprofundamento da configuração de exercícios para a avaliação do equilíbrio e o estudo do potencial da plataforma em programas de intervenção para o treino das funções do equilíbrio.

Portugal has strong musical traditions, which have been perpetrated by decades through folkloristic activities. In folk groups from Alto Minho (north of Portugal), folk singing is mostly performed by cantadeiras, amateur female solo singers who learn this style orally. Their vocal characteristics are distinctive when compared with other regions of the country; however, deep understanding of these vocal practices is still missing. The present work aims at studying Alto Minho cantadeira’s vocal performance in a multidimensional perspective, envisioning social, cultural and physiological understanding of this musical style. Thus, qualitative and quantitative data analyses were carried out, to: (i) describe current performance practices, (ii) explore existent perceptions about most relevant voice features in this region, (iii) investigate physiological and acoustic properties of this style, and (iv) compare this style of singing with other non-classical singing styles of other countries. Dataset gathered involved: 78 groups whose members were telephone interviewed, 13 directors who were asked to fill in a questionnaire on performance practices, 1 cantadeira in a pilot study, 16 cantadeiras in preliminary voice recordings, 77 folk group members in listening tests, and 10 cantadeiras in multichannel recordings, including audio, ELG, air flow and intra-oral pressure signals. Data were analysed through thematic content analysis, descriptive and inferential statistics, hierarchical principal components, and multivariate linear regression models. Most representative voices have a high pitched and loud voice, with a bright timbre, predominance of chest register without excessive effort, and good text intelligibility with regional accent. High representativeness levels were obtained by few cantadeiras; these sing with high levels of subglottal pressure and vocal fold contact quotient, predominance of high spectrum energy and vocal loudness, corroborating indications of prevalence of pressed phonation. These vocal characteristics resemble belting in musical theatre and share similarities with country (USA) and ojikanje (Croatia) singing. Strategies that may contribute to the preservation of this type of singing and the vocal health of current cantadeiras are discussed, pointing at the direction of continuous education among folk groups, following practices that are already adopted elsewhere in Europe.

Bioactive glasses and glass–ceramics are a class of biomaterials which elicit special response on their surface when in contact with biological fluids, leading to strong bonding to living tissue. This particular trait along with good sintering ability and high mechanical strength make them ideal materials for scaffold fabrication. The work presented in this thesis is directed towards understanding the composition-structure-property relationships in potentially bioactive glasses designed in CaOMgOP2O5SiO2F system, in some cases with added Na2O. The main emphasis has been on unearthing the influence of glass composition on molecular structure, sintering ability and bioactivity of phosphosilicate glasses. The parent glass compositions have been designed in the primary crystallization field of the pseudo-ternary system of diopside (CaO•MgO•2SiO2) – fluorapatite (9CaO•3P2O5•CaF2) – wollastonite (CaO•SiO2), followed by studying the impact of compositional variations on the structure-property relationships and sintering ability of these glasses. All the glasses investigated in this work have been synthesized via melt-quenching route and have been characterized for their molecular structure, sintering ability, chemical degradation and bioactivity using wide array of experimental tools and techniques. It has been shown that in all investigated glass compositions the silicate network was mainly dominated by Q2 units while phosphate in all the glasses was found to be coordinated in orthophosphate environment. The glass compositions designed in alkali-free region of diopside – fluorapatite system demonstrated excellent sintering ability and good bioactivity in order to qualify them as potential materials for scaffold fabrication while alkali-rich bioactive glasses not only hinder the densification during sintering but also induce cytotoxicity in vitro, thus, are not ideal candidates for in vitro tissue engineering. One of our bioglass compositions with low sodium content has been tested successfully both in vivo and in preliminary clinical trials. But this work needs to be continued and deepened. The dispersing of fine glass particles in aqueous media or in other suitable solvents, and the study of the most important factors that affect the rheology of the suspensions are essential steps to enable the manufacture of porous structures with tailor-made hierarchical pores by advanced processing techniques such as Robocasting.

Estuaries are poles of attraction for human settlement which is a source of pressures to surface water bodies. The implementation of the European Water Framework Directive (WDF, 2000/60/EC) has increased the investigation in order to develop methodologies to assess the Ecological Quality Status (EQS) of aquatic ecosystems. Transitional systems are naturally stressed and characterized by highly dynamic physical, chemical and hydro-morphologic conditions and by species with a higher level of tolerance to change, being more difficult to develop suitable quality indicators for these systems. The general purpose of this study is to test the ability of synthesis descriptors, including primary (S, taxa richness) and derived biological variable (H’, Shannon-Wiener diversity), biotic indices (AMBI and M-AMBI), body size properties (abundance distribution by body size classes, length, weight and length-weight relationships) and non-taxonomic indices (ISS), as well as functional indicators related to the decomposition rates of various experimental substrates, a macrophyte (Phragmites australis) and an alga (Fucus vesiculosus), to evaluate the environmental quality in transitional systems. This study was carried out in one of the most pristine channels of the Ria the Aveiro, Mira Channel, along a full salinity gradient and in a metals and metalloid sediment contamination area, the Estarreja Channel, and two reference channels (Canelas and Salreu). In this study were used different sampling techniques, the leaf-bag technique and a hand-held corer. In Mira Channel, the alga and the macrophyte presented an opposite trend in the decomposition rate along the salinity gradient, with the decomposition rates of the alga always higher than those of the macrophyte. The decomposition rates of the macrophyte and the alga were higher in the mid estuary and in higher salinity areas, respectively, corresponding to the preferencial distribution areas of each species. The macrobenthic fauna associated with the decaying and an artificial substrate (control) showed equally well the benthic succession from the marine to the freshwater areas and, despite the strong differences in the decay rates, no significant differences were found between the benthic communities associated with the alga and the macrophyte. The body size properties of the macrobenthic fauna associated with the P. australis leaf-bag (1mm and 5mm) and corer samples were studied along the full salinity gradient. The dominant species of the sub-set of measured specimens were not the same of the original macrobenthic fauna sampled but, despite that, the sub-set of measured specimens was also able to show the benthic succession from the marine to the freshwater areas. The body size abundance distribution of the benthic macroinvertebrates according to the ISS size classes did not show a particular trend in any sampler along the salinity gradient. Significant differences were found in the length, weight and length-weight relationships of Annelids, , Molluscs and even some species along the salinity gradient. No significant differences were found in the AMBI, M-AMBI and ISS values along the salinity gradient for all the samplers. The EQS of the corer samples obtained using the M-AMBI was lower than that of the leaf-bags. The EQS obtained with the ISS was higher than that obtained with the M-AMBI in the leaf-bags but not in the corer samples. The ecological effects of contaminated sediments associated with the industrial chemical effluents discharged in the Estarreja Channel were studied a decade after ceasing the emissions, using the Sediment Quality Triad approach and two reference channels. The results showed that despite the emissions ceased in 2004, the sediment remains polluted with high levels of metals and metalloid, available to bioaccumulation and with severe consequences at the community level. The sediment contamination problem was also studied using the leaf-bag technique with a macrophyte, an alga and a control substrate. The results showed that the decay rates, the associated macrofauna and the application of the AMBI, M-AMBI and ISS indices to the mesh-bag samples were not able to identify the sediment contamination. Contrarily to the AMBI, the M-AMBI and the ISS showed significant differences between the contaminated and the reference channels for the corer samples. Although such statistical significance, the interest of using these complex biotic indices could be questioned, when much simple ones, like the S and H’ allow to reach the same conclusions.

During the last century mean global temperatures have been increasing. According to the predictions, the temperature change is expected to exceed 1.5ºC in this century and the warming is likely to continue. Freshwater ecosystems are among the most sensitive mainly due to changes in the hydrologic cycle and consequently changes in several physico-chemical parameters (e.g. pH, dissolved oxygen). Alterations in environmental parameters of freshwater systems are likely to affect distribution, morphology, physiology and richness of a wide range of species leading to important changes in ecosystem biodiversity and function. Moreover, they can also work as co-stressors in environments where organisms have already to cope with chemical contamination (such as pesticides), increasing the environmental risk due to potential interactions. Therefore, the objective of this work was to evaluate the effects of climate change related environmental parameters on the toxicity of pesticides to zebrafish embryos. The following environmental factors were studied: pH (3.0-12.0), dissolved oxygen level (0-8 mg/L) and UV radiation (0-500 mW/m2). The pesticides studied were the carbamate insecticide carbaryl and the benzimidazole fungicide carbendazim. Stressors were firstly tested separately in order to derive concentration- or intensity-response curves to further study the effects of binary combinations (environmental factors x pesticides) by applying mixture models. Characterization of zebrafish embryos response to environmental stress revealed that pH effects were fully established after 24 h of exposure and survival was only affected at pH values below 5 and above 10. Low oxygen levels also affected embryos development at concentrations below 4 mg/L (delay, heart rate decrease and edema), and at concentrations below 0.5 mg/L the survival was drastically reduced. Continuous exposure to UV radiation showed a strong time-dependent impact on embryos survival leading to 100% of mortality after 72 hours of exposure. The toxicity of pesticides carbaryl and carbendazim was characterized at several levels of biological organization including developmental, biochemical and behavioural allowing a mechanistic understanding of the effects and highlighting the usefulness of behavioural responses (locomotion) as a sensitive endpoint in ecotoxicology. Once the individual concentration response relationship of each stressor was established, a combined toxicity study was conducted to evaluate the effects of pH on the toxicity of carbaryl. We have shown that pH can modify the toxicity of the pesticide carbaryl. The conceptual model concentration addition allowed a precise prediction of the toxicity of the jointeffects of acid pH and carbaryl. Nevertheless, for alkaline condition both concepts failed in predicting the effects. Deviations to the model were however easy to explain as high pH values favour the hydrolysis of carbaryl with the consequent formation of the more toxic degradation product 1- naphtol. Although in the present study such explanatory process was easy to establish, for many other combinations the “interactive” nature is not so evident. In the context of the climate change few scenarios predict such increase in the pH of aquatic systems, however this was a first approach focused in the lethal effects only. In a second tier assessment effects at sublethal level would be sought and it is expectable that more subtle pH changes (more realistic in terms of climate changes scenarios) may have an effect at physiological and biochemical levels with possible long term consequences for the population fitness.

As algas do género Nannochloropsis são microalgas marinhas que apresentam um perfil bioquímico único, principalmente no que é respeitante a lípidos, e uma vasta gama de compostos bioativos que possibilitam a sua aplicabilidade comercial em várias áreas biotecnológicas, destacando-se a alimentação e nutrição humana, indústria cosmética e farmacêutica, produção de biocombustíveis e a sua utilização em aquacultura. Em aquacultura, são usadas maioritariamente microalgas vivas, cuja produção representa elevados custos. Tem havido assim uma pesquisa de dietas alternativas, entre as quais os concentrados de microalgas se apresentam promissores. Os desafios atuais das empresas produtoras de concentrados de microalgas prendem-se com a conservação e armazenamento destes concentrados. Assim, neste trabalho foi proposto o estudo da influência da refrigeração, congelação e adição de conservantes a PhytoBloom Green Formula®, concentrado de Nannochloropsis sp. comercializado pela empresa Necton S.A., com o objetivo de averiguar a variação de parâmetros bioquímicos e organoléticos com a exposição do concentrado aos diferentes métodos de conservação. Pretendia-se assim observar se estes processos podem ser usados para aumentar o tempo de prateleira do concentrado em estudo. Para tal, foram avaliadas amostras recolhidas em três pontos temporais e analisados os seguintes parâmetros: perfil de ácidos gordos, quantificação de hidroperóxidos lipídicos, quantificação espectrofotométrica de clorofila a e carotenóides, bem como parâmetros organoléticos. Inicialmente, foi efetuada uma avaliação de diferentes parâmetros organoléticos, não se observando variações relevantes entre amostras das diferentes condições. Assim, foi posteriormente realizada a avaliação bioquímica. Primeiramente, foi efetuada a quantificação de ácidos gordos por GC-FID das diferentes amostras, nas quais não se observou diferenças significativas entre as condições experimentais. Foi também efetuado um ensaio de FOX II, que permitiu avaliar o grau de peroxidação lipídica de cada amostra por quantificação de hidroperóxidos lipídicos formados. As amostras nas quais houve adição de conservantes apresentaram um teor menor de hidropéroxidos lipídicos, permitindo inferir que a ação dos conservantes com propriedades antioxidantes permitiu uma melhor conservação da amostra. Quando se determinou a concentração de clorofila a e de carotenóides verificou-se que, em ambos os casos, a congelação conduziu a uma estabilização da concentração destes pigmentos. No entanto, os melhores resultados foram obtidos usando a combinação de congelação com adição de conservantes. Estes resultados, embora promissores, carecem de uma confirmação por um novo estudo, completando com análises com maior rigor e sensibilidade associados, no sentido de se verificar qual o método mais vantajoso para a extensão do tempo de prateleira de PhytoBloom Green Formula®.

Estudo da variação da abundância e diversidade de procariotas em sedimentos subsuperficiais marinhos

Os sedimentos marinhos subsuperficiais profundos são, atualmente, um ambiente ainda pouco conhecido do ponto de vista microbiológico, nomeadamente quanto aos processos metabólicos que nele têm lugar e quanto à sua possível influência nos ciclos biogeoquímicos. O acesso a amostras colhidas em sedimentos profundos, particularmente no âmbito dos programas IODP (International Ocean Discovery Program) e ECORD (European Consortium for Ocean Research Drilling) tem permitido recolher informação sobre a estrutura das comunidades de procariotas bem como sobre alguns dos fatores que regulam a sua distribuição e atividade. Este estudo teve como objetivo caracterizar a distribuição e a diversidade estrutural das comunidades de procariotas em sedimentos subsuperficiais profundos colhidos no Arco Izu-Bonin-Mariana, no mar das Filipinas, com recurso a métodos independentes de cultivo (PCR-DGGE) e à contagem de células por microscopia de epifluorescência. Os resultados apontam para a existência de comunidades de Bacteria e Archaea diversas. Os valores do índice de diversidade de Shannon-Weaver (H’) calculados com base nos perfis de DGGE (Bacteria) foram significativamente mais elevados (3,035 – 1,971) nas camadas superficiais (< 140 mafm) do que nos sedimentos (2,519 - 1,049) correspondentes a profundidades superiores entre 163 e 879 mafm. A abundância máxima (8,66 x 106 células.gps-1) foi registada à profundidade de 67 mafm e valor mínimo (2,26 x 106 células.gps-1) foi observado em amostras colhidas a 879 mafm de profundidade. Abundância e diversidade apresentaram correlação negativa com a profundidade e com o teor de sulfato. Os resultados indicam que ao longo da coluna de sedimento se estabelecem comunidades de procariotas estruturalmente diferentes e adaptadas ao ambiente geoquímico prevalecente, nomeadamente em termos dos aceitadores de eletrões disponíveis. O estudo do microbioma destas amostras representativas do ambiente sedimentar subsuperficial profundo será continuado e detalhado, com recurso a técnicas de sequenciação avançada.

Diplodia corticola is regarded as the most virulent fungus involved in cork oak decline, being able to infect not only Quercus species (mainly Q. suber and Q. ilex), but also grapevines (Vitis vinifera) and eucalypts (Eucalyptus sp.). This endophytic fungus is also a pathogen whose virulence usually manifests with the onset of plant stress. Considering that the infection normally culminates in host death, there is a growing ecologic and socio-economic concern about D. corticola propagation. The molecular mechanisms of infection are hitherto largely unknown. Accordingly, the aim of this study was to unveil potential virulence effectors implicated in D. corticola infection. This knowledge is fundamental to outline the molecular framework that permits the fungal invasion and proliferation in plant hosts, causing disease. Since the effectors deployed are mostly proteins, we adopted a proteomic approach. We performed in planta pathogenicity tests to select two D. corticola strains with distinct virulence degrees for our studies. Like other filamentous fungi D. corticola secretes protein at low concentrations in vitro in the presence of high levels of polysaccharides, two characteristics that hamper the fungal secretome analysis. Therefore, we first compared several methods of extracellular protein extraction to assess their performance and compatibility with 1D and 2D electrophoretic separation. TCA-Acetone and TCA-phenol protein precipitation were the most efficient methods and the former was adopted for further studies. The proteins were extracted and separated by 2D-PAGE, proteins were digested with trypsin and the resulting peptides were further analysed by MS/MS. Their identification was performed by de novo sequencing and/or MASCOT search. We were able to identify 80 extracellular and 162 intracellular proteins, a milestone for the Botryosphaeriaceae family that contains only one member with the proteome characterized. We also performed an extensive comparative 2D gel analysis to highlight the differentially expressed proteins during the host mimicry. Moreover, we compared the protein profiles of the two strains with different degrees of virulence. In short, we characterized for the first time the secretome and proteome of D. corticola. The obtained results contribute to the elucidation of some aspects of the biology of the fungus. The avirulent strain contains an assortment of proteins that facilitate the adaptation to diverse substrates and the identified proteins suggest that the fungus degrades the host tissues through Fenton reactions. On the other hand, the virulent strain seems to have adapted its secretome to the host characteristics. Furthermore, the results indicate that this strain metabolizes aminobutyric acid, a molecule that might be the triggering factor of the transition from a latent to a pathogenic state. Lastly, the secretome includes potential pathogenicity effectors, such as deuterolysin (peptidase M35) and cerato-platanin, proteins that might play an active role in the phytopathogenic lifestyle of the fungus. Overall, our results suggest that D. corticola has a hemibiotrophic lifestyle, switching from a biotrophic to a necrotrophic interaction after plant physiologic disturbances.This understanding is essential for further development of effective plant protection measures.
