The influence of layer-by-layer films of polyaniline and Ni-tetrasulfonated phthalocyanine (PANI/Ni-TS-Pc) on the electrical performance of polymeric light-emitting diodes (PLED) made from (poly[2-methoxy-5-(2`-ethyl-hexyloxy)-1,4-phenylene vinylene]) (MEH-PPV) is investigated by using current versus voltage measurements and impedance spectroscopy. The PLED is composed by a thin layer of MEH-PPV sandwiched between indium tin oxide (ITO) and aluminum electrodes, resulting in the device structure ITO/(PANI/Ni-TS-Pc)(n)/MEH-PPV/Al, where n stands for the number of PANI/Ni-TS-Pc bilayers. The deposition of PANI/Ni-TS-Pc leads to a decrease in the driving voltage of the PLEDs, which reaches a minimum when n = 5 bilayers. In addition, impedance spectroscopy data reveal that the PLED impedance decreases as more PANI/Ni-TS-Pc bilayers are deposited. The PLED structure is further described by an equivalent circuit composed by two R-C combinations, one for the bulk and other for the interface components, in series with a resistance originated in the ITO contact. From the impedance curves, the values for each circuit element is determined and it is found that both, bulk and interface resistances are decreased upon PANI/Ni-TS-Pc deposition. The results indicate that PANI/NiTS-Pc films reduce the contact resistance at ITO/MEH-PPV interface, and for that reason improve the hole-injection within the PLED structure. (c) 2007 Elsevier B.V. The search for bioactive molecules to be employed as recognition elements in biosensors has stimulated researchers to pore over the rich Brazilian biodiversity. In this sense, we introduce the use of natural cashew gum (Anacardium occidentale L) as an active biomaterial to be used in the form of layer-by-layer films, in conjunction with phthalocyanines, which were tested as electrochemical sensors for dopamine detection. We investigated the effects of chemical composition of cashew gum from two different regions of Brazil (Piaui and Ceara states) on the physico-chemical characteristics of these nanostructures. The morphology of the nanostructures containing cashew gum was studied by atomic force microscopy which indicates that smooth films punctuated by globular features were formed that showed low roughness values. The results indicate that, independent of the origin, cashew gum stands out as an excellent film forming material with potential application in nanobiomedical devices as electrochemical sensors. Magnetic nanoparticles are promising for a variety of applications, such as biomedical devices, spin electronics, magnetic data storage media, to name a few. However, these goals may only be reached if stable and organized structures are fabricated. In this article, we report on a single-step synthetic route with the coprecipitation method, in which iron oxide magnetic nanoparticles (Fe3O4 NPs) were stabilized in aqueous media using the poly(diallyldimethylammonium chloride) (PDAC) polyelectrolyte. The Fe3O4 NPs had a diameter of ca. 5 nm, according to transmission electron microscopy (TEM) images, being arranged in an inverse spinel structure typical of magnetite. An investigation with infrared spectroscopy indicated that the mechanisms of stabilization in the polymer matrix were based on the interaction between quaternary amide groups from PDAC and the nanoparticle surface. The Fe3O4-PDAC NPs exhibited considerable magnetic susceptibility, with a monotonic increase in the magnetization with decreasing temperature. These Fe3O4-PDAC NPs were immobilized in layer-by-layer (LbL) films, being alternated with layers of poly(vinylsulfonic acid) (PVS). The LbL films were much rougher than typical films made with polyelectrolytes, and Fe3O4-PDAC NPs have been responsible for the high electrocatalytic activity toward H2O2 reduction, with an overpotential shift of 0.69 V. Overall, the stability, magnetic properties and film-forming ability indicate that the Fe3O4-PDAC NPs may be used for nanoelectronics and bioelectrochemical devices requiring reversible and magnetic redox materials.

The immobilization of the glucose/mannose-binding lectin from Concanavalia ensiformis seeds (ConA) onto a monolayer made of a galactomannan extracted from Leucaena leucocephala seeds (GML), which was adsorbed onto - amino-terminated surfaces, was investigated by means of ellipsometry and atomic force microscopy. The mean thickness of GML monolayer, which polysaccharide consists of linear 1 -> 4-linked beta-D-mannopyranosil units partially substituted at C-6 by alpha-D-galactopyranosyl units, amounted to (1.5 +/- 0.2) nm. ConA molecules adsorbed onto GML surfaces forming (2.0 +/- 0.5) nm thick layers. However, in the presence of mannose the adsorption failed, indicating that ConA binding sites were blocked by mannose and were no longer available for mannose units present in the GML backbone. The GML film was also used as support for the adsorption of three serotypes of dengue virus particles (DENV-1, DENV-2 and DENV-3), where DENV-2 formed the thickest film (4 +/- 2) nm. The adsorbed layer of DENV-2 onto ConA-covered GML surfaces presented mean thickness values similar to that determined for DENV-2 onto bare GML surfaces. The addition of free mannose units prevented DENV-2 adsorption onto ConA-covered GML films by similar to 50%, suggesting competition between virus and mannose for ConA binding sites. This finding suggests that if ConA is also adsorbed to GML surface and its binding site is blocked by free mannose, virus particles are able to recognized GML mannose unities substituted by galactose. interactions between polysaccharides thin films, proteins, and viruses are of great relevance since they can provide basis for the development of biotechnological devices. These results indicate that GML is a potential polysaccharide for biomaterials development, as those could involve interactions between ConA in immune system and viruses. (C) 2011 Elsevier B.V. In this thesis we focussed on the characterization of the reaction center (RC) protein purified from the photosynthetic bacterium Rhodobacter sphaeroides. In particular, we discussed the effects of native and artificial environment on the light-induced electron transfer processes. The native environment consist of the inner antenna LH1 complex that copurifies with the RC forming the so called core complex, and the lipid phase tightly associated with it. In parallel, we analyzed the role of saccharidic glassy matrices on the interplay between electron transfer processes and internal protein dynamics. As a different artificial matrix, we incorporated the RC protein in a layer-by-layer structure with a twofold aim: to check the behaviour of the protein in such an unusual environment and to test the response of the system to herbicides. By examining the RC in its native environment, we found that the light-induced charge separated state P+QB - is markedly stabilized (by about 40 meV) in the core complex as compared to the RC-only system over a physiological pH range. We also verified that, as compared to the average composition of the membrane, the core complex copurifies with a tightly bound lipid complement of about 90 phospholipid molecules per RC, which is strongly enriched in cardiolipin. In parallel, a large ubiquinone pool was found in association with the core complex, giving rise to a quinone concentration about ten times larger than the average one in the membrane. Moreover, this quinone pool is fully functional, i.e. it is promptly available at the QB site during multiple turnover excitation of the RC. The latter two observations suggest important heterogeneities and anisotropies in the native membranes which can in principle account for the stabilization of the charge separated state in the core complex. The thermodynamic and kinetic parameters obtained in the RC-LH1 complex are very close to those measured in intact membranes, indicating that the electron transfer properties of the RC in vivo are essentially determined by its local environment. The studies performed by incorporating the RC into saccharidic matrices evidenced the relevance of solvent-protein interactions and dynamical coupling in determining the kinetics of electron transfer processes. The usual approach when studying the interplay between internal motions and protein function consists in freezing the degrees of freedom of the protein at cryogenic temperature. We proved that the “trehalose approach” offers distinct advantages with respect to this traditional methodology. We showed, in fact, that the RC conformational dynamics, coupled to specific electron transfer processes, can be modulated by varying the hydration level of the trehalose matrix at room temperature, thus allowing to disentangle solvent from temperature effects. The comparison between different saccharidic matrices has revealed that the structural and dynamical protein-matrix coupling depends strongly upon the sugar. The analyses performed in RCs embedded in polyelectrolyte multilayers (PEM) structures have shown that the electron transfer from QA - to QB, a conformationally gated process extremely sensitive to the RC environment, can be strongly modulated by the hydration level of the matrix, confirming analogous results obtained for this electron transfer reaction in sugar matrices. We found that PEM-RCs are a very stable system, particularly suitable to study the thermodynamics and kinetics of herbicide binding to the QB site. These features make PEM-RC structures quite promising in the development of herbicide biosensors. The studies discussed in the present thesis have shown that, although the effects on electron transfer induced by the native and artificial environments tested are markedly different, they can be described on the basis of a common kinetic model which takes into account the static conformational heterogeneity of the RC and the interconversion between conformational substates. Interestingly, the same distribution of rate constants (i.e. a Gamma distribution function) can describe charge recombination processes in solutions of purified RC, in RC-LH1 complexes, in wet and dry RC-PEM structures and in glassy saccharidic matrices over a wide range of hydration levels. In conclusion, the results obtained for RCs in different physico-chemical environments emphasize the relevance of the structure/dynamics solvent/protein coupling in determining the energetics and the kinetics of electron transfer processes in a membrane protein complex.

Aim of this thesis was the production of porosity-graded piezoelectric ceramics for ultrasonic applications by tape casting and screen printing. The study and optimization of each single step of the tape casting process allowed to produce flat and crack-free multilayers of Pb0.988(Zr0.52Ti0.48)0.976Nb0.024O3 (PZTN) of uniform and graded porosity. The multilayers of thickness ranging between 400 and 800 µm were produced stacking optimized green layers with different amount of pore former. The functionally graded materials showed porosity ranging between 10 and 30% with piezoelectric properties suitable for the specific ultrasonic applications. Screen printing inks of PZTN for deposition onto four different substrates were studied and optimized. Thick films with thickness ranging between 4 and 20 µm were produced tailoring the screen printing parameters and number of depositions.

Im ersten Teil der Arbeit wurden Ionomere durch aufeinanderfolgende polymeranaloge Reaktionen eines Reaktivesterpolymers mit verschiedenen primären bzw. sekundären Aminen synthetisiert. Polarisationsmikroskopische Untersuchungen zeigten, dass diese Ionomere sowohl über thermotrope als auch über lyotrope flüssigkristalline Eigenschaften verfügen. Anschließend wurden mit der 'self-assembly'-Methode Multischichtfilme aus den Ionomeren und hochgeladenen Polyelektrolyten sowie aus entgegengesetzt geladenen Ionomeren aufgebaut. Bei Röntgen-reflexionsmessungen an den Multischichtfilmen konnten winkelabhängige Modulationen der Strahlungsintensität beobachtet werden. Es ist zu vermuten, dass es sich bei diesen Modulationen um Überlagerungen von Kiessig-Ringen und Bragg-Peaks handelt, was ein Indiz dafür ist, dass regel-mäßige Subschichten in den Filmen existieren. Das Konzept der polymeranalogen Reaktionen eines Reaktivesterpolymers mit verschiedenen primären Aminen wurde außerdem dazu benutzt, Polymere zur Modifikation von Cadmiumselenid-Nanokristallen herzustellen. Hierbei konnte zum einen gezeigt werden, dass Nanokristalle direkt mit aminogruppenhaltigen Polymeren modifiziert werden können, zum anderen ist es auch möglich kationische Ionomere auf Nanokristallen mit Mercaptoessigsäure-Liganden zu adsorbieren. Im letzten Teil der Arbeit wurden ionische triazinhaltige Polymere hergestellt und elektrochemisch charakterisiert. Die hergestellten Polymere sind sehr leicht reduzierbar. Anschließend wurden aus den triazinhaltigen Polymeren und entgegengesetzt geladenen Polyelektrolyten ebenfalls Multischichtfilme hergestellt.

Die Dissertation 'Azobenzol- und Perylendiimid-funktionalisierte Polyphenylen-Dendrimere - Synthese, Charakterisierung und Eigenschaften' gliedert sich in vier Themengebiete. Der erste Abschnitt beschäftigt sich mit der Synthese unterschiedlich dichter Dendrimere um einen Azobenzol-Kern. Einkristallstrukturen und Molekülvisualisierungen verdeutlichen die dreidimensionale Gestalt der Dendrimere. Die Dendrimere zeigen erstmalig eine Abhängigkeit des Isomerisationsverhaltens von der das Chromophor umgebenden Struktur. Der zweite Abschnitt hat Interaktionen von Chromophoren, deren Distanz und Orientierung zueinander gezielt durch einen äußeren Impuls geändert werden können, zum Thema. Die Verbindung von Azobenzol und PMI führt durch deren gegenseitige Beeinflussung zu einem Verlust der charakteristischen Eigenschaften der Chromophore. Eine Oligo-L-Lysinkette, deren Enden mit NMI und PMI funktionalisiert sind, stellt ein FRET-System dar. Distanz und Orientierung der Chromophore zueinander werden durch den mittels TFE induzierten Übergang des Peptids vom Knäuel zur Helix verändert. Der dritte Abschnitt führt die Synthese von PDI-gekernten Dendrimeren durch Substitution in der bay-Region des Chromophors ein. Die Eignenschaften der Verbindungen wurden mittels optischer Methoden und cyclovoltammetrischen Studien untersucht. Weiter wurde die Oberflächenfunktionalisierung mit Aminosäuren und Oligopeptiden zu wasserlöslichen Dendrimeren mit hoher Oberflächenladung verfolgt. Das letzte Kapitel stellt Untersuchungen zur Organisation von Polyphenylen-Dendrimeren auf HOPG vor. Es lassen sich einerseits Nanofasern formieren, andererseits können auch geordnete Mono- und Multilagen erzeugt werden.

Aufgaben der vorliegenden Untersuchungen waren die Etablierung von planaren Multilayern aus menschlichen Tumorzellen (WiDr und SiHa) und die Testung dieses Zellsystems als Bestrahlungsmodell solider Tumoren. Neben der konventionellen Röntgenbestrahlung (250 kV) wurde auch das Überleben nach Schwerionenbestrahlung (12C6+) und nach Behandlung mit dem Chemotherapeutikum Etoposid untersucht. Multilayer aus beiden Zelllinien zeigten ein geringeres Überleben nach Röntgen- und Schwerionenbestrahlung als die entsprechenden Monolayer. Die hier beschriebene multizelluläre Sensitivierung steht allerdings im Gegensatz zu der in der Literatur beschriebenen multizellulären Resistenz der Sphäroide, dem sog. Kontakteffekt. Nach durchflußzytometrischen Messungen arretierten die bestrahlten SiHa-Zellen in der G2/M-Phase. Im Gegensatz zum transienten Block der Monolayer verweilten die Multilayer in einem permanenten Arrest. Im Vergleich zur Röntgenbestrahlung verlängerte sich die Arrestzeit der Monolayer nach Schwerionenbestrahlung im Bragg-Peak um 12-24 h. Auch waren mehr Zellen betroffen. Im Gegensatz dazu war kein Unterschied zwischen beiden Bestrahlungsmodalitäten bei den Multilayern bis zum Ende des Beobachtungszeitraumes zu verzeichnen. Nach Etoposid-Behandlung verhielten sich die Multilayer deutlich resistenter als die Monolayer. Somit zeigten Multilayer interessanterweise nach Bestrahlung eine Sensitivierung und nach Etoposid-Behandlung eine Resistenz. Die Unterschiede im Überleben der beiden Kultivierungsformen beruhen zum Großteil auf den Differenzen in der Zellzyklusverteilung. Besonders deutlich wurde dieser Zusammenhang zwischen Überleben und Zellzyklusverteilung durch Wiederaussaat- und Synchronisations-Experimente.

Ziel der vorliegenden Arbeit war die Untersuchung von Struktur und Dynamik in Polymer-Ton-Nanokompositen mittels EPR-Spektroskopie; damit sollten ein Beitrag zur Analyse der Tensidschicht in solchen Systemen geleistet und die Ergebnisse anderer Messmethoden ergänzt werden. Die Tensidschicht in Polymer-Ton-Nanokompositen nimmt großen Einfluss auf das System, denn sie bestimmt die Wechselwirkung zwischen Ton und Polymer: Damit hydrophiler Ton gut mit hydrophobem Polymer (hier Polystyrol) mischbar ist, muss das Schichtsilikat zunächst mit Tensiden organisch-modifiziert werden; dies geschieht durch Kationenaustausch der Natriumionen im Ton gegen Tenside. Um mit Hilfe der EPR einen Einblick in die Tensidschicht zu gewinnen, muss etwa 1% der zur Tonmodifizierung eingesetzten Amphiphile spinmarkiert sein. So gelang es im Rahmen dieser Arbeit, Tenside mit verschiedenen Kopfgruppen, nämlich Trimethylammonium- bzw. Trimethylphosphoniumtenside, zu synthetisieren und sie an verschiedenen Positionen ihrer hydrophoben Alkylkette mit einem Nitroxidradikal zu markieren. Das Nitroxidradikal diente als Spinsonde für die EPR-Experimente. Neben der Synthese verschiedener, spinmarkierter Amphiphile, der anschließenden Darstellung organisch-modifizierten Tons (Kationenaustausch) und verschiedener Polymer-Ton-Nanokomposite (Schmelzinterkalation) wurden alle Proben mittels EPR-Spektroskopie untersucht; dabei wurden sowohl cw- als auch gepulste Messtechniken eingesetzt. Aus cw-Experimenten ging hervor, dass die Dynamik der gesamten Tensidschicht mit der Temperatur zunimmt und die Mobilität der hydrophoben Tensidalkylkette mit wachsendem Abstand zu ihrer Kopfgruppe wächst. Zugabe von Polymer behindert bei steigender Temperatur das Anschwellen des Tons bei Aufschmelzen der Tensidschicht; die Dynamik des Systems ist eingeschränkt. Mit Hilfe gepulster EPR-Messungen (ENDOR und ESEEM), die Informationen über Abstände bzw. Kontakt in den untersuchten Systemen lieferten, ließ sich ein Strukturmodell der Polymer-Ton-Nanokomposite skizzieren, das Vorstellungen anderer, älterer Methoden unterstützt: Hierbei richten sich die Tenside in Multischichten unterschiedlicher Mobilität parallel zur Tonoberfläche aus.

Nanoscience aims at manipulating atoms, molecules and nano-size particles in a precise and controlled manner. Nano-scale control of the thin film structures of organic/polymeric materials is a prerequisite to the fabrication of sophisticated functional devices. The work presented in this thesis is a compilation of various polymer thin films with newly synthesized functional polymers. Cationic and anionic LC amphotropic polymers, p-type and n-type semiconducting polymers with triarylamine, oxadiazole, thiadiazole and triazine moieties are suitable materials to fabricate multilayers by layer-by-layer (LBL) self-assembly with a well defined internal structure. The LBL assembly is the ideal processing technique to prepare thin polymer film composites with fine control over morphology and composition at nano-scale thickness, which may have applications in photo-detectors, light-emitting diodes (LEDs), displays and sensors, as well as in solar cells. The multilayer build-up was investigated with amphotropic LC polymers individually by solution-dipping and spin-coating methods; they showed different internal orders with respect to layering and orientation of the mesogens, as a result of the liquid crystalline phase. The synthesized p-type and n-type semiconducting polymers were examined optically and electrochemically, suggesting that they are favorably promising as hole-(p-type) or electron-(n-type) transport materials in electronic and optoelectronic devices. In addition, we report a successful film deposition of polymers by the vacuum deposition method. The vapor deposition method provides a clean environment; it is solvent free and well suited to sequential depositions in hetero-structured multilayer system. As the potential applications, the fabricated polymer thin films were used as simple electrochromic films and also used as hole transporting layers in LEDs. Electrochemical and electrochromic characterizations of assembled films reveal that the newly synthesized polymers give rise to high contrast ratio and fast switching electrochromic films. The LEDs with vacuum deposited films show dramatic improvements in device characteristics, indicating that the films are promising as hole transporting layers. These are the result of not only the thin nano-scale film structures but also the combination with the high charge carrier mobility of synthesized semiconducting polymers.

In this thesis foliation boudinage and related structures have been studied based on field observations and numerical modeling. Foliation boudinage occurs in foliated rocks independent of lithology contrast. The developing structures are called ‘Foliation boudinage structures (FBSs)’ and show evidence for both ductile and brittle deformation. They are recognized in rocks by perturbations in monotonous foliation adjacent to a central discontinuity, mostly filled with vein material. Foliation boudinage structures have been studied in the Çine Massif in SW-Turkey and the Furka Pass-Urseren Zone in central Switzerland. Four common types have been distinguished in the field, named after vein geometries in their boudin necks in sections normal to the boudin axis: lozenge-, crescent-, X- and double crescent- type FBSs. Lozengetype FBSs are symmetric and characterized by lozenge-shaped veins in their boudin neck with two cusps facing opposite sides. A symmetrical pair of flanking folds occurs on the two sides of the vein. Crescent-type FBSs are asymmetric with a single smoothly curved vein in the boudin neck, with vein contacts facing to one side. X- and double crescent- type FBSs are asymmetric. The geometry of the neck veins resembles that of cuspate-lobate structures. The geometry of flanking structures is related to the shape of the veins. The veins are mostly filled with massive quartz in large single crystals, commonly associated with tourmaline, feldspar and biotite and in some cases with chlorite. The dominance of large facetted single quartz crystals and spherulitic chlorite in the veins suggest that the minerals grew into open fluidfilled space. FLAC experiments show that fracture propagation during ductile deformation strongly influences the geometry of developing veins. The cusps of the veins are better developed in the case of propagating fractures. The shape of the boudin neck veins in foliation boudinage depends on the initial orientation and shape of the fracture, the propagation behaviour of the fracture, the geometry of bulk flow, and the stage at which mineral filling takes place. A two dimensional discrete element model was used to study the progressive development of foliation boudinage structures and the behavior of visco-elastic material deformed under pure shear conditions. Discrete elements are defined by particles that are connected by visco-elastic springs. Springs can break. A number of simulations was Abstract vii performed to investigate the effect of material properties (Young’s modulus, viscosity and breaking strength) and anisotropy on the developing structures. The models show the development of boudinage in single layers, multilayers and in anisotropic materials with random mica distribution. During progressive deformation different types of fractures develop from mode I, mode II to the combination of both. Voids develop along extension fractures, at intersections of conjugate shear fractures and in small pull-apart structures along shear fractures. These patterns look similar to the natural examples. Fractures are more localized in the models where the elastic constants are low and the competence contrast is high between the layers. They propagate through layers where the constants are high and the competence contrast is relatively low. Flow localize around these fractures and voids. The patterns similar to symmetric boudinage structures and extensional neck veins (e.g. lozenge type) more commonly develop in the models with lower elastic constants and anisotropy. The patterns similar to asymmetric foliation boudinage structures (e.g. X-type) develop associated with shear fractures in the models where elastic constants and anisotropy of the materials are relatively high. In these models boudin neck veins form commonly at pull-aparts along the shear fractures and at the intersection of fractures.
