We studied the morphometry of the sting apparatus of Acromyrmex subterraneus brunneus workers by dissection and mounting of the structures on slides, and present here a brief description and illustrations. Sting apparatus examinations showed differences in size, but without morphological differences. The seven morphometric variables studied permitted grouping of the Ac. subterraneus brunneus workers into the following three groups: cluster 1 - consisting of 85, 39 large, 45 medium and 1 small workers; cluster 2 - consisting of 44 small workers, and cluster 3 - consisting of 6 large workers. This study demonstrated that all worker castes follow the same morphological pattern, although functional characteristics of the sting apparatus should be highlighted.

Standard Test Methods (e.g. ASTM, DIN) for materials characterization in general, and for fatigue in particular, do not contemplate specimens with complex geometries, as well as the combination of axial and in-plane bending loads in their methodologies. The present study refers to some patents and the new configuration or configurations of specimens (non-standardized by the status quo of test methods) and a device developed to induce axial and bending combined forces resultants from axial loads applied by any one test equipment (dynamic or monotonic) which possesses such limitation, towards obtaining more realistic results on the fatigue behavior, or even basic mechanical properties, from geometrically complex structures. Motivated by a specific and geometrically complex aeronautic structure (motor-cradle), non-standardized welded tubular specimens made from AISI 4130 steel were fatigue-tested at room temperature, by using a constant amplitude sinusoidal load of 20 Hz frequency, load ratio R = 0.1 with and without the above referred auxiliary fatigue apparatus. The results showed the fatigue apparatus was efficient for introducing higher stress concentration factor at the welded specimen joints, consequently reducing the fatigue strength when compared to other conditions. From the obtained results it is possible to infer that with small modifications the proposed apparatus will be capable to test a great variety of specimen configurations such as: squared tubes and plates with welded or melted junctions, as well as other materials such as aluminum, titanium, composites, polymeric, plastics, etc. © 2009 Bentham Science Publishers Ltd.

This paper analyses the static and dynamic behavior of the railroad track model in laboratory. Measurements of stresses and strains on a large-scale railroad track apparatus were studied. The model includes: compacted soil, representing the final layers of platform, ballast layer, and ties (steel, wooden, and pre-stressed concrete). The soil and soil ballast interface were instrumented with pneumatic stress gauge. Settlement measurement device were positioned at the same levels as the load cells. Loads were applied by hydraulic actuators, statically and dynamically. After the prescribed number of load cycles, in pre-determined intervals, stresses and strains were measured. Observations indicate that stress and strain distributions, transmitted by wooden or steel ties, behave similarly. A more favorable behavior was observed with pre-stressed concrete mono block ties. Non-linear response was observed after a threshold numbers of cycles were surpassed, showing that the strain modulus increases with the numbers of cycles. © 2009 IOS Press.

This work focuses on applying fuzzy control embedded in microcontrollers in an experimental apparatus using magnetorheological fluid damper. The non-linear behavior of the magnetorheological dampers associated with the parametric variations on vehicle suspension models corroborate the use of the fuzzy controllers. The fundamental formulation of this controller is discussed and its performance is shown through numeric simulations. An experimental apparatus representing a two degree of freedom system containing a magnetorheological damper is used to identify the main parameters and to evaluate the performance of the closed-loop system with the embedded low-cost microcontroller-based fuzzy controller. © 2013 Brazilian Society for Automatics - SBA.

Disfunções imunes podem surgir pela combinação entre susceptibilidade genética e fatores ambientais. Existem evidências, em humanos expostos ao mercúrio (Hg), de alterações da resposta imunológica por auto-anticorpos induzidos por Hg. Este trabalho investigou a ocorrência de auto-imunidade induzida por Hgtotal entre indivíduos ribeirinhos da região do Tapajós (Brasília Legal, São Luiz do Tapajós e Barreiras), expostos ao Hgtotal, e da comunidade ribeirinha da região do Tocantins (Panacauera) não exposta ao Hgtotal. No período de junho de 2004 a dezembro de 2006 foram coletadas 236 pares de amostras de cabelo e sangue, nas quais a concentração de Hgtotal no cabelo foi determinada por espectrofotometria de absorção atômica, e no soro, os auto-anticorpos foram analisados por microscopia de imunofluorescência (IF) usando substrato de células epiteliais humanas (Hep-2). Os mais altos níveis de Hgtotal no cabelo foram os de São Luiz do Tapajós (11,24 ± 2,23 μg/g), seguido por Brasília Legal (10,00 ± 0,99) e Barreiras (8,64 ± 1,13), e os mais baixos foram os de Panacauera (2,98 ± 0,20). Em relação à variável sexo, foi observada associação somente em Brasília Legal, com níveis de Hgtotal mais altos no cabelo dos homens. Cerca de 79,65% ribeirinhos do Tapajós e 31,25% da região do Tocantins apresentaram no soro autoanticorpos induzidos por Hg. Os padrões de auto-anticorpos identificados por IF foram: misto (50,96%), nuclear (31,21%), nucleolar (14,65%) e aparelho mitótico/citoplasmático (3,18%), observando-se maior prevalência dos padrões misto e nuclear nas comunidades expostas (p<0,01). Os auto-anticorpos mais freqüentes foram, por ordem de prevalência: NuMa1, PM/Scl, Ssa-Ro, rRNP/Sm, golgi/Ssa/Ro, PCNA, rRNP, Ku, além de outros auto-anticorpos com especificidade ainda não definida. A intensidade de IF (p< 0,0001) foi mais reativa nos ribeirinhos do Tapajós. Análise por regressão logística múltipla indicou que o risco de apresentar auto-anticorpos foi aproximadamente duas vezes maior nos expostos ao mercúrio com faixa etária acima de 50 anos (p>0,01). Finalmente, estudos adicionais são indispensáveis para confirmar a especificidade destes auto-anticorpos induzidos pela exposição mercurial, bem como elucidar os mecanismos imunotoxicológicos da ação do mercúrio sobre o sistema imune humano.

Associations among feeding habit, beak type, and food source in birds have been widely studied and are well known to exist. The relationship between feeding habit and jaw apparatus in birds has not attracted attention from ornithologists, perhaps because of the complexity of the skeletal morphology of the feeding system of birds. The goal of this study was to compare the jaw apparatus and foraging strategies of various Oriental species of the Picidae (Meiglyptini and Picini tribes) using a morphofunctional analysis of the skeletal structure of the jaw apparatus. This study showed that there are at least three types of jaw apparatus in these woodpeckers, as follows: 1) robust, developed, and complex; 2) complexity and development intermediate, as observed in Meiglyptes tristis and Dinopium spp., whose main foraging method involves gleaning, probing, and tapping; and 3) poorly developed, as observed in Picus miniaceus and Hemicircus concretus. The success of woodpeckers as a natural group is due not only to their feeding diversity, but also their ability to explore a wide range of different resources, as appropriate to their jaw apparatus.

We describe the male reproductive apparatus of the giant hermit crab Petrochirus diogenes, with morphological and biometric analyses of the spermatophore, the gonopore and the ultrastructure of the spermatozoa. Specimens were collected from the southern coast of Sao Paulo, Brazil. Morphological analyses were done using stereoscopic, light, transmission and scanning electron microscopy. The reproductive system of this hermit crab is composed of elongate and lobular testes followed by vasa deferentia that connect to the exterior via gonopores. The gonopores are ovoid and surrounded by setae, and each gonopore is composed of a membranous operculum that forms a depression constituting the gonopore opening. The gonopore constitutes a unique structure among the Diogenidae due to its number of setae. The spermatophores are tripartite, composed of a sperm-containing ampulla, a peduncle and a proximal foot. The spermatozoon has 3 main regions (acrosomal vesicle, nucleus and cytoplasm). The structure of the spermatophore indicates that this species can be considered an exception within Diogenidae with regard to spermatophore morphology and can therefore be used for phylogenetic inferences.

The Neotropical genus Anacroneuria (Perlidae) is represented by more than 300 species. Among the Brazilian Anacroneuria, only the nymphs of two species have been described. In this paper, we described the nymph of A. ofaye Froehlich and an apparatus for rearing this genus. This is the first record of A. ofaye from Parana State, Brazil.

E-mail: marcio_medvet@hotmail.com Rheas are birds belonging to the ratites group and, among ostriches and emus, are the largest birds currently alive. In this work we studied the macroscopic aspects of rheas' digestive tract in order to provide important information to a better understanding of these birds' eating habits as well their anatomy. Twenty young animals aging between two and six months from the Centre for Wild Animals Multiplication (Cemas, scientific breeding license form Ibama no.1478912) were used. After dissection it was observed that their tongue was small and presented a rhomboid form, being disposed on the oral cavity floor, and inserted in its base by a frenulum. The esophagus was a rectilinear tube with elastic aspect and longitudinal elastic fibers, without dilation, which gives it an absence of crop. The proventriculus presented a fusiform form and the gastric ventricle showed and slightly oval form when filled, and was internally coated with a thick gastric cuticle. The small intestine was composed of three distinct regions: duodenum, jejunum and ileum. The duodenum had a light gray color and showed a "U" curved shaped. The jejunum was dark green, long and composed of several short loops arranged above each other. The ileum had a gray color and was connected with the jejunum. In ventral line to the rectum and cloaca, the ileum extended cranially, dorsally to the ascending duodenum. The large intestine was composed of two caeca, one right and one left, and colon-rectum and ileum were continuous with the cloaca. The structures of the rhea digestive tract resemble those described in the literature regarding to its shape and topography, even though rhea's caeca are well developed and relatively long.

Photosynthetic organisms have sought out the delicate balance between efficient light harvesting under limited irradiance and regulated energy dissipation under excess irradiance. One of the protective mechanisms is the thermal energy dissipation through the xanthophyll cycle that may transform harmlessly the excitation energy into heat and thereby prevent the formation of damaging active oxygen species (AOS). Violaxanthin deepoxidase (VDE) converts violaxanthin (V) to antheraxanthin (A) and zeaxanthin (Z) defending the photosynthetic apparatus from excess of light. Another important biological pathway is the chloroplast water-water cycle, which is referred to the electrons from water generated in PSII reducing atmospheric O2 to water in PSI. This mechanism is active in the scavenging of AOS, when electron transport is slowed down by the over-reduction of NADPH pool. The control of the VDE gene and the variations of a set of physiological parameters, such as chlorophyll florescence and AOS content, have been investigated in response to excess of light and drought condition using Arabidopsis thaliana and Arbutus unedo.. Pigment analysis showed an unambiguous relationship between xanthophyll de-epoxidation state ((A+Z)/(V+A+Z)) and VDE mRNA amount in not-irrigated plants. Unexpectedly, gene expression is higher during the night when xanthophylls are mostly epoxidated and VDE activity is supposed to be very low than during the day. The importance of the water-water cycle in protecting the chloroplasts from light stress has been examined through Arabidopsis plant with a suppressed expression of the key enzyme of the cycle: the thylakoid-attached copper/zinc superoxide dismutase. The analysis revealed changes in transcript expression during leaf development consistent with a signalling role of AOS in plant defence responses but no difference was found any in photosynthesis efficiency or in AOS concentration after short-term exposure to excess of light. Environmental stresses such as drought may render previously optimal light levels excessive. In these circumstances the intrinsic regulations of photosynthetic electron transport like xanthophyll and water-water cycles might modify metabolism and gene expression in order to deal with increasing AOS.

Seit der Entdeckung des Golgi-Apparates im Jahre 1898 wurden seine Struktur, seine enzymatische Zusammensetzung und die Dynamik des in ihm stattfindenden Proteintransports intensiv untersucht. Dennoch blieben bis heute wesentliche Fragen zu seiner Funktionsweise unbeantwortet. So existieren nach wie vor mehrer konkurrierende Modelle zur Organisation der hoch komplexen räumlichen Verteilung seiner Enzyme sowie zum grundlegenden Mechanismus des Intra-Golgi Transports. Die Beantwortung dieser und weiterer Fragen ist für das Verständnis des Golgi-Apparates essentiell, aber aus methodischen Gründen höchst schwierig, da es bisher nicht möglich war die Struktur und Dynamik des Golgi-Apparates lebender Zellen mit der hierfür notwendigen Auflösung und Geschwindigkeit zu untersuchen. Bis heute gibt es für die funktionsmorphologischen Untersuchungen des Golgi-Apparates lebender Zellen keine echte Alternative zur Fernfeld- Fluoreszenzmikroskopie. Das MMM-4Pi-Mikroskop ermöglicht als erstes Fluoreszenzmikroskop, aufgrund seiner Auflösung von ~200 nm in der Fokalebene und 100-150 nm entlang der optischen Achse, die Untersuchung der Subkompartimente des Golgi-Apparates und kann, aufgrund seiner hohen Aufnahmegeschwindigkeit von 0.5 Hz, die Dynamik des Intra-Golgi Transports zeitlich auflösen. Ziel dieser Arbeit war es daher, den Golgi-Apparate lebender Zellen in zwei Farben sowie mit einer bisher nicht möglichen räumlichen und zeitlichen Auflösung zu untersuchen. Um die Leistungsfähigkeit der dreidimensionalen Bildgebung dieser Methode zu überprüfen, wurde erstmals der Golgi-Apparat fixierter Säugerzellen korrelativ mit dem Transmissionselektronenmikroskop und dem MMM-4Pi-Mikroskop aufgenommen. Die rekonstruierten Strukturen korrelierten in allen drei Raumrichtungen zu über 80%, was die Validität beider Methoden eindrucksvoll unter Beweis stellt. Zudem konnten mit dem MMM-4Pi-Mikroskop Aussackungen von Golgi-Cisternen aufgelöst werden, was die Eignung dieser Methode zur strukturellen Analyse der Subkompartimente des Golgi-Apparates unterstreicht. Des Weiteren wurde, in einer Reihe zweifarbiger Aufnahmeserien, die Verteilung dreier Golgi-Enzyme in lebenden Säugerzellen untersucht, und ihre mittlere relative Distanz bestimmt. Ihre aus der Literatur bekannten Lokalisationen konnten in zwei Fällen bestätigt (GalT, MannII) und in einem Fall korrigiert werden (2-OST). Im Gegensatz zu der konfokal bestimmten Cis-/Mid-Lokalisation von 2-OST zeigen die Ergebnisse der hoch aufgelösten Distanzanalyse deutlich, dass eine Mid-/Trans-Lokalisation vorliegt. Dieses Ergebnis wurde elektronenmikroskopisch überprüft und bestätigt. Da die räumliche Anordnung der Golgi-Enzyme die Reihenfolge ihrer Akitvität wiederspiegelt, ist eine möglichst präzise Bestimmung ihrer Konzentrationsverteilungen essentiell, um die Funktion des Golgi-Apparates zu verstehen. Insbesondere zeigt dieses Resultat, dass die Bestimmung der Lokalisation von Golgi-Enzymen über konfokale Kolokalisationsstudien zu falschen Ergebnissen führen kann. Die Kombination hoher räumlicher Auflösung mit einer schnellen Datenaufnahme erlaubte die Analyse der Transportdynamik innerhalb des Golgi-Apparates von Säugerzellen. In mehreren Zeitserien zweifarbiger Aufnahmen wurde der Transport des Frachtproteins VSVG relativ zum Trans-Golgi-Marker GalT untersucht. Dabei zeigte sich, dass das Trans-Golgi-Kompartiment in einigen Fällen durch eine deutliche Formänderung auf die Ankunft eines VSVG-Transportpulses reagierte und sich insgesamt wesentlich dynamischer verhielt als der Transportpuls selbst. Diese Beobachtung bestätigt tendenziell Transportmodelle, die den Golgi-Apparat nicht als statisches, sondern als dynamisches, aktiv am Transport beteiligtes Organell beschreiben. Die hier vorgestellten Experimente stellen die ersten Untersuchungen zur Verteilung von Golgi-Enzymen sowie zur Transportdynamik des Golgi-Apparates lebender Zellen mit einer dreidimensionalen Auflösung im Bereich von 100-200 nm dar. Wie am Beispiel von 2 OST gezeigt, ist es mit dem MMM-4Pi-Mikroskop allgemein möglich, die Lokalisation von Golgi-Enzymen wesentlich präziser als bisher zu bestimmen. Bei der Untersuchung dynamischer Prozesse ist in naher Zukunft eine Steigerung der Leistungsfähigkeit der Methode zu erwarten. Zum einen werden CCD-Kameras mit kürzeren Auslesezeiten und einer elektronischen Verstärkung des Signals die Datenaufnahme weiter beschleunigen. Zum anderen könnte durch die Entwicklung eines parallelisierten Mikroskops mit Einphotonen-Anregung das Bleichen konsekutiver Aufnahmen verringert werden, wodurch längere Aufnahmeserien möglich sein werden.

Intraflagellar transport (IFT) is required for the assembly and maintenance of cilia. In this study we analyzed the subcellular localization of IFT proteins in retinal cells by correlative high-resolution immunofluorescence and immunoelectron microscopy. The rod photoreceptor cell was used as a model system to analyze protein distribution in cilia. To date the expression of IFT proteins has been described in the ciliary region without deciphering the precise spatial and temporal subcellular localization of IFT proteins, which was the focus of my work. rnThe establishment of the pre-embedding immunoelectron method was an important first step for the present doctoral thesis. Results of this work reveal the differential localization of IFT20, IFT52, IFT57, IFT88, IFT140 in sub-ciliary compartments and also their presence in non-ciliary compartments of retinal photoreceptor cells. Furthermore, the localization of IFT20, IFT52 and IFT57 in dendritic processes of non-ciliated neurons indicates that IFT protein complexes also operate in non-ciliated cells and may participate in intracellular vesicle trafficking in eukaryotic cells in general.rnIn addition, we have investigated the involvement of IFT proteins in the ciliogenesis of vertebrate photoreceptor cilia. Electron microscopy analyses revealed six morphologically distinct stages. The first stages are characterized by electron dense centriolar satellites and a ciliary vesicle, while the formation of a ciliary shaft and of the light sensitive outer segment disks are features of the later stages. IFT proteins were expressed during all stages of photoreceptor cell development and found to be associated with the ciliary apparatus. In addition to the centriole and basal body IFT proteins are present in the photoreceptor cytoplasm, associated with centriolar satellites, post-Golgi vesicles and with the ciliary vesicle. Therewith the data provide an evidence for the involvement of IFT proteins during ciliogenesis, including the formation of the ciliary vesicle and the elongation of the primary cilium of photoreceptor cells. Moreover, the cytoplasmic localization of IFT proteins in the absence of a ciliary shaft in early stages of ciliogenesis indicates roles of IFT proteins beyond their well-established function for IFT in mature cilia and flagella. rn
Experimental characterization and modelling of a servo-pneumatic system for a knee loading apparatus

The new knee test rig developed in University of Bologna used pneumatic cylinder as actuator system. Specific characterization and modelling about the pneumatic cylinder and the related devices are needed in better controlling the test rig. In this thesis, an experimental environment for the related device is set up with data acquisition system using Real-time Windows Target, Simulink, MatLab. Based on the experimental data, a fitted model for the pneumatic cylinder friction is found.
