The Centropomidae family consists of three genera, Centropomus, Lates and Psammoperca. Centropomus is the most diverse group, with six Centropomus species occur in the Western Atlantic Ocean C. poeyi Chávez, 1961, C. parallelus Poey, 1860, C. mexicanus Bocourt, 1868, C. pectinatus Poey, 1860 and C. ensiferus Poey, 1860. Some of these species are considered cryptic, because of its morphological traits showed low resolution for identification purposes. Despite showing great interest as a natural resource and fish culture, aspects of their diversity and karyotypic patterns are poorly understood. In this work morphological identification and comparison of mitochondrial 16S gene sequence were used to identify the species of the genus Centropomus occurring in Rio Grande do Norte, northeastern Brazil. Two sepecies were identified, C. undecimalis and C. mexicanus, which had the chromosomal aspects analyzed, through Classical cytogenetic method analyzes (conventional staining, C-banding, Ag-NORs), fluorochrome staining AT- and GC-specific, replication bands by incorporating of the base analog 5-Bromo-2’-deoxyuridine (5-BrdU), in situ chromosomal mapping of (TTAGGG)n sequences and in situ chromosome mapping 18S and 5S rRNA genes. Both species show 2n=48 acrocentric chromosomes, with ribosomal sites (Ag-NOR/18S rDNA/ Mitramycin+) in second chromosomal pair, in telomeric position on the long arm in C. mexicanus and interstitial in C. undecimalis. The nuclear organization pair (pair 2) shown a resolutive cytotaxonomic marker for these two species. The generated data reveal a lower species diversity than previously believed, suggesting that greater attention should be paid in taxonomic identification of the species, in view of optimize commercial actions exploitation, biological conservation and cultivation.

The fracturing in carbonate rocks has been attracting increasingly attention due to new oil discoveries in carbonate reservoirs. This study investigates how the fractures (faults and joints) behave when subjected to different stress fields and how their behavior may be associated with the generation of karst and consequently to increased secondary porosity in these rocks. In this study I used satellite imagery and unmanned aerial vehicle UAV images and field data to identify and map faults and joints in a carbonate outcrop, which I consider a good analogue of carbonate reservoir. The outcrop comprises rocks of the Jandaíra Formation, Potiguar Basin. Field data were modeled using the TECTOS software, which uses finite element analysis for 2D fracture modeling. I identified three sets of fractures were identified: NS, EW and NW-SE. They correspond to faults that reactivate joint sets. The Ratio of Failure by Stress (RFS) represents stress concentration and how close the rock is to failure and reach the Mohr-Coulomb envelopment. The results indicate that the tectonic stresses are concentrated in preferred structural zones, which are ideal places for carbonate dissolution. Dissolution was observed along sedimentary bedding and fractures throughout the outcrop. However, I observed that the highest values of RFS occur in fracture intersections and terminations. These are site of karst concentration. I finally suggest that there is a relationship between stress concentration and location of karst dissolution in carbonate rocks.

Brazil is among the largest cashew nut producers of the world. However, the roasting process is still carried out artisanally, especially in the Brazilian semiarid region. In face of this occupational problem, the aim of this study was to perform a physical-chemical characterization of the particulate matter (PM) emitted by the roasting of cashew nuts, as well as to determine the occupational risk and molecular mechanisms associated. The most evident PM characteristics were the prevalence of fine particles, typical biomass burning morphologies such as tar ball and the presence of the elements K, Cl, S, Ca and Fe. In addition, atmospheric modeling analyses suggest that these particles can reach neighboring regions of the emission source. Polycyclic aromatic hydrocarbons (PAHs) with carcinogenic potential, such as benzo[a]pyrene, dibenz[a,h]anthracene, benzo[a]anthracene, benzo[b]fluoranthene, chrysene, benzo[k]fluoranthene, indeno[1,2,3-c,d]pyrene and benzo[j]fluoranthene were the most abundant PAHs found in the two air monitoring campaigns. Among the identified oxy-PAH the benzanthrone (7H-benz[d,e]anthracen-7-one) had the highest concentration and the evaluation of lifetime cancer risk showed an increase of 12 to 37 cases of cancer for every 10,000 exposed people. Chemical analysis of roasted cashew nuts identified the PAHs: phenanthrene, benzo[g,h,i]perylene, pyrene and benzo[a]pyrene, besides the 3-pentadecilfenol allergen (urushiol analogue) as prevalent. Occupational exposure to PAHs was confirmed by the increase of urinary 1-hydroxypyrene levels and genotoxic effects were evidenced by the increase on micronuclei and nuclear bud frequency in exfoliated buccal mucosa cells among the exposed workers. Other biomarkers of effects such as karyorrhexis, pyknotic, karyolytic, condensed chromatin and binucleated cells also have their frequencies increased when compared to an unexposed control group. The investigation of the molecular mechanisms associated with the PM organic extract showed cytotoxicity in human lung cell lines (A549) at concentrations ≥ 4 nM BaPeq. Using non-cytotoxic doses the extract was able to activate proteins involved in the DNA damage response pathway (Chk1 and p53). Moreover, the specific contribution of the four most representative PAHs in the cashew nut roasting sample showed that benzo[a]pyrene was the most efficient to activate Chk1 and p53. Finally, the organic extract was able to increase persistently the mRNA expression involved in the PAHs metabolism (CYP1A1 and CYP1B1), inflammatory response (IL-8 and TNF-α) and cell cycle arrest (CDKN1A) for DNA repair (DDB2). The high PM concentrations and its biological effects associated warn of the serious harmful effects of artisanal cashew nut roasting and urgent actions should be taken to the sustainable development of this activity.

En todas aquellas partes de El Salvador que están cubiertas por espesas capaz de cenizas volcánicas jovenes, la acción erosiva del agua produjo profundos barrancos. Durante la temporada lluviosa corre por ellos gran parte de las aguas meteóricas, mientras que en la época seca son quebradas secas. Estos barrancos están caracterizados por sus paredes altamente inclinadas a veces verticales o aun desplomadas. Estos barrancos se prestan especialmente para estudios ecológicos, porque poseen una flora y fauna muy característica la primera se presenta especialmente en aquellos lugares donde ambos paredones tienen inclinación vertical y donde la vegetación natural está protegida de los efectos del sol por el follaje de árboles, observando el desarrollo de estas plantas en el intervalo de la estación lluviosa a la seca, podemos distinguirlas fácilmente en dos grupos diferentes: el comportamiento del primer grupo es análogo a aquel de los efímeros de primera en la región del mediterráneo sus esporas o semillas germinan al comienzo de la época lluviosa, desarrollándose durante su duración y fructificando hasta el comienzo de la siguiente época seca, esto es debido a que saben aprovechar las reservas de agua que todavía han quedado en el suelo, por poseer un raigambre más profundo.

The development of scaffolds based on biomaterials is a promising strategy for Tissue Engineering and cellular regeneration. This work focuses on Bone Tissue Engineering, the aim is to develop electrically tailored biomaterials with different crystalline and electric features, and study their impacts onto cell biological behavior, so as to predict the materials output in the enhancement of bone tissue regeneration. It is accepted that bone exhibits piezoelectricity, a property that has been proved to be involved in bone growth/repair mechanism regulation. In addition electrical stimulations have been proved to influence bone growth and repair. Piezoelectric materials are therefore widely investigated for a potential use in bone tissue engineering. The main goal is the development of novel strategies to produce and employ piezoelectric biomaterials, with detailed knowledge of mechanisms involved in cell-material interaction. In the current work, poly (L-lactic) acid (PLLA), a synthetic semi-crystalline polymer, exhibiting biodegradibility, biocompatibility and piezoelectricity is studied and proposed as a promoter of enhanced tissue regeneration. PLLA has already been approved for implantation in human body by the Food and Drug Administration (FDA), and at the moment it is being used in several clinical strategies. The present study consists of first preparing films with different degrees of crystallinity and characterizing these PLLA films, in terms of surface and structural properties, and subsequently assessing the behavior of cells in terms of viability, proliferation, morphology and mineralization for each PLLA configuration. PLLA films were prepared using the solvent cast technique and submitted to different thermal treatments in order to obtain different degrees of crystallinity. Those platforms were then electrically poled, positively and negatively, by corona discharge in order to tailor their electrical properties. The cellular assays were conducted by using two different osteoblast cell lines grown directly onto the PLLA films:Human osteoblast Hob, a primary cell culture and Human osteosarcoma MG-63 cell line. This thesis gives also a comprehensive introduction to the area of Bone Tissue Engineering and provides a review of the work done in this field in the past until today, in that same field, including the one related with bone’s piezoelectricity. Then the experimental part deals with the effects of the crystallinity degrees and of the polarization in terms of surface properties and cellular bio assays. Three different degrees of crystallinity, and three different polarization conditions were prepared; which results in 9 different configurations under investigation.

A percepção e intervenção no espaço histórico, enquanto arquétipo cultural, constituíram o mote enquadrador do presente trabalho, merecendo amplo tratamento no primeiro capítulo. O segundo capítulo é dedicado ao estudo do palacete dos condes de Sampayo, elemento patrimonial de elevado valor significativo e que aqui se apresenta como objecto de tratamento central. O espólio consultado relativo ao arquivo da casa de Sampayo, permitiu-nos claramente fazer recuar no tempo a construção original do palacete, num percurso temporal análogo ao contíguo moinho de maré, ambos implementados no núcleo histórico de Alhos Vedros. Igualmente clarificou como, quando e de que forma a casa da Cova, dos Mendonça Furtado, convergirá com a casa dos Sampayo, num contexto socioeconómico específico em que a estruturação interna da nobreza se escorava nos sistemas de parentesco para consolidação do seu poder e para o fortalecimento e manutenção das casas titulares. Os modelos comparativos são tratados no capítulo três e surgem na figura das pousadas, valorizadoras dos edifícios com valor histórico e indutoras de sinergias locais. Por último, o capítulo quatro é totalmente dedicado à conceptualização de um projecto para a funcionalização do palacete como pousada. A perspectiva da sustentabilidade na fundamentação patrimonial do projeto, inscreve-se no programa de requalificação da frente ribeirinha do concelho da Moita e ancora numa perspetiva de âmbito mais alargado, inerente às políticas da administração do território e turismo da Grande Área Metropolitana de Lisboa. ABSTRACT; The perception and action in space history, as cultural archetype, constituted the framed motto of this work deserves wider treatment in the first chapter. The second chapter is devoted to the study of the palace of the Counts of Sampayo, of its assets in high-value significant and presented here is treated as central. The stock found on the file's house Sampayo, allowed us to clearly back in time the original construction of the palace, a temporal course similar to the adjacent tidal mill, both implemented in the historical center of Plantsville. He also clarified how, when and how the pit house, the Mendonca Furtado, converge with the house of Sampayo, specific socio-economic context in which the internal structure of the nobility was underpinned in kinship systems to consolidate its power and the strengthening and maintaining the homes owners. Comparative models are treated in chapter three, appearing in the picture of the holiday, not value of buildings of historic and inducing local synergies. Finally, chapter four is devoted entirely to the conceptualization of a project to re-functionalization of the mansion and guesthouse. The perspective of sustainability in the grounds of equity of the project is part of the program of the riverfront redevelopment of Moita and grounded on a broader scope, inherent in the policies of the planning and tourism Greater Metropolitan Area of Lisbon.

A Medicina Nuclear (MN) permite investigar o estado fisiológico dos tecidos de forma minimamente invasiva, usando radiofármacos (rf’s), moléculas compostas por um análogo biológico específico desses processos fisiológicos e um marcador radioativo (radionuclídeo). PET/CT (do acrónimo inglês Positron Emission Tomography/Computed Tomography), uma das modalidades de imagem em MN, está a expandir-se rapidamente em muitos Países. As imagens obtidas revelam a biodistribuição dos rf’s usados e permitem conhecer a sua distribuição precisa no organismo. 18F-Fluorodesoxiglicose (FDG), um análogo da glicose, é o rf mais comumente utilizado, isto porque em neoplasias as células são geralmente caracterizadas pelo aumento do metabolismo da glicose. A quantificação realizada em imagens de PET, tem por base uma estimativa quantitativa do metabolismo da glicose no tumor, utilizando o índice de captação estandardizado, SUV (do acrónimo inglês Standard Uptake Value). A realização de estudos dinâmicos em PET/CT, isto é, realizados em sequência temporal imediatamente após a administração endovenosa do rf e, durante um período de tempo pré-determinado (por exemplo, 15 minutos) permite que o registo da cinética inicial dos rf’s seja estudado. A análise dos dados obtidos com o estudo dinâmico permite compreender o grau e a perfusão tumoral. Habitualmente, quanto maior a captação de 18F-FDG num tumor, maior é a sua atividade metabólica glicolítica, o que tem sido traduzido em maior agressividade tumoral. Nesta investigação, realizaram-se estudos dinâmicos num grupo restrito de patologias oncológicas, nomeadamente: carcinoma da bexiga, carcinoma do colo do útero, carcinoma colorretal, carcinoma do endométrio, metástases hepáticas e adenocarcinoma pancreático. Realizaram-se estudos dinâmicos durante cerca de 10/15 minutos, com 1minuto por frame. O objetivo desta Investigação é tentar compreender se, tumores com maior perfusão respondem melhor à Radioterapia (RT), ou se, a resposta é independente da perfusão. Para avaliar os valores de SUV’s ao longo tempo, realizaram-se ROI’s (do acrónimo inglês Region of Interest), nas artérias femorais ou aorta e na lesão tumoral. Com estes dados, criaram-se gráficos de atividade/tempo onde, no eixo das abcissas é representado o tempo e no eixo das ordenadas os valores de SUV. A partir destes gráficos e dos dados neles contidos, calculou-se o Índice de Perfusão Tumoral através de 2 métodos: A, Método Trapezoidal de Aproximação que relaciona a razão entre a área perfusional do tumor e a área de fluxo arterial, até ao momento do cruzamento das curvas; B, mais simples, calculando o Índice de Perfusão do Tumor através da razão entre o valor de SUV máximo da curva tumoral e da curva arterial até ao momento do cruzamento das curvas. O Método de Comparação de Métodos de Altman&Bland, revelou que tanto o método A como o método B são semelhantes para o cálculo do Índice de Perfusão Tumoral. Em conclusão, apesar do número reduzido de indivíduos estudados, os dados apresentados indicam que existe uma tendência para que haja melhor resposta à RT por parte dos tumores com maior índice metabólico e maior índice de perfusão. Os tumores com menor índice metabólico e menor grau perfusional parece que respondem pior à RT.

Objetivo: Analizar las características asociadas al riesgo de feminicidio en España entre mujeres expuestas a la violencia de pareja o análogo y su posible asociación con las denuncias a los agresores. Métodos: Se realizó un estudio de casos y controles para el periodo 2010-2011. Los casos, 135 mujeres mayores de edad, asesinadas por su pareja o análogo durante dicho periodo, se identificaron a través de la página web de la Federación de Asociaciones de Mujeres Separadas y Divorciadas, y de los informes del Consejo General del Poder Judicial. Los controles, 185 mujeres expuestas a la violencia de pareja el último año, proceden de la Macroencuesta de Violencia de Género 2011. La asociación entre la denuncia y el riesgo de feminicidio se estimó mediante modelos de regresión logística multivariada. Resultados: No se encontró asociación entre denunciar al agresor y el riesgo de ser asesinada (odds ratio [OR]: 1,38; intervalo de confianza del 95% [IC95%]: 0,68-2,79). Las mujeres inmigrantes expuestas a la violencia de pareja registraron una mayor probabilidad de ser asesinadas (ref.: mujeres españolas; OR: 5,38; IC95%: 2,41-11,99). Esta asociación también se observó en las mujeres que vivían en zonas rurales (ref: zonas urbanas; OR: 2,94; IC95%: 1,36-6,38). Conclusiones: La denuncia judicial al agresor no parece modificar el riesgo de asesinato entre las mujeres expuestas a la violencia de pareja. Las medidas de protección a las mujeres deberían extremarse en las mujeres inmigrantes y las que viven en el medio rural.
