A controlled study was undertaken to assess the effect of gamma irradiation on post-traumatic intraocular cellular proliferation. A standard perforating injury in the posterior segment of the rabbit eye was used to induce intraocular cellular proliferation and vitreo-retinal membrane formation. The site of injury was irradiated with an ophthalmic Cobalt60 applicator which provided a continuous source of gamma rays. Non-irradiated eyes developed traction retinal detachments associated with post-traumatic vitreo-retinal membranes. Irradiated eyes developed attenuated membranes or atrophic retinal scars, with the retina remaining attached. The membranes in non-irradiated eyes were highly cellular with abundant collagen, while irradiated membranes had fewer cells within a sparse collagen matrix. The episcleral fibroblasts, on autoradiographic studies appeared to be the main source of the cells that formed the proliferating tissue in both non-irradiated and irradiated eyes. In irradiated eyes both the inflammatory response and division of fibroblasts were delayed and reduced.

A robust multiscale scheme referred to as micro–macro method has been developed for the prediction of localized damage in fiber reinforced composites and implemented in a finite element framework. The micro–macro method is based on the idea of partial homogenization of a structure. In this method, the microstructural details are included in a small region of interest in the structure and the rest is modeled as a homogeneous continuum. The solution to the microstructural fields is then obtained on solving the two different domains, simultaneously. This method accurately predicts local stress fields in stress concentration regions and is computationally efficient as compared with the solution of a full scale microstructural model. This scheme has been applied to obtain localized damage at high and low stress zones of a V-notched rail shear specimen. The prominent damage mechanisms under shear loading, namely, matrix cracking and interfacial debonding, have been modeled using Mohr–Coulomb plasticity and traction separation law, respectively. The average stress at the notch has been found to be 44% higher than the average stresses away from the notch for a 90 N shear load. This stress rise is a direct outcome of the geometry of the notch.

Cyathostomins comprise a group of 50 species of parasitic nematodes that infect equids. Ribosomal DNA sequences, in particular the intergenic spacer (IGS) region, have been utilized via several methodologies to identify pre-parasitic stages of the commonest species that affect horses. These methods rely on the availability of accurate sequence information for each species, as well as detailed knowledge of the levels of intra- and inter-specific variation. Here, the IGS DNA region was amplified and sequenced from 10 cyathostomin species for which sequence was not previously available. Also, additional IGS DNA sequences were generated from individual worms of 8 species already studied. Comparative analysis of these sequences revealed a greater range of intra-specific variation than previously reported (up to 23%); whilst the level of inter-specific variation (3-62%) was similar to that identified in earlier studies. The reverse line blot (RLB) method has been used to exploit the cyathostomin IGS DNA region for species identification. Here, we report validation of novel and existing DNA probes for identification of cyathostomins using this method and highlight their application in differentiating life-cycle stages such as third-stage larvae that cannot be identified to species by morphological means.

Electric vehicles (EVs) and hybrid electric vehicles (HEVs) can reduce greenhouse gas emissions while switched reluctance motor (SRM) is one of the promising motor for such applications. This paper presents a novel SRM fault-diagnosis and fault-tolerance operation solution. Based on the traditional asymmetric half-bridge topology for the SRM driving, the central tapped winding of the SRM in modular half-bridge configuration are introduced to provide fault-diagnosis and fault-tolerance functions, which are set idle in normal conditions. The fault diagnosis can be achieved by detecting the characteristic of the excitation and demagnetization currents. An SRM fault-tolerance operation strategy is also realized by the proposed topology, which compensates for the missing phase torque under the open-circuit fault, and reduces the unbalanced phase current under the short-circuit fault due to the uncontrolled faulty phase. Furthermore, the current sensor placement strategy is also discussed to give two placement methods for low cost or modular structure. Simulation results in MATLAB/Simulink and experiments on a 750-W SRM validate the effectiveness of the proposed strategy, which may have significant implications and improve the reliability of EVs/HEVs.

Numerous experimental studies of damage in composite laminates have shown that intralaminar (in-plane) matrix cracks lead to interlaminar delamination (out-of-plane) at ply interfaces. The smearing of in-plane cracks over a volume, as a consequence of the use of continuum damage mechanics, does not always effectively capture the full extent of the interaction between the two failure mechanisms. A more accurate representation is obtained by adopting a discrete crack approach via the use of cohesive elements, for both in-plane and out-of-plane damage. The difficulty with cohesive elements is that their location must be determined a priori in order to generate the model; while ideally the position of the crack migration, and more generally the propagation path, should be obtained as part of the problem’s solution. With the aim of enhancing current modelling capabilities with truly predictive capabilities, a concept of automatic insertion of interface elements is utilized. The consideration of a simple traction criterion in relation to material strength, evaluated at each node of the model (or of the regions of the model where it is estimated cracks might form), allows for the determination of initial crack location and subsequent propagation by the insertion of cohesive elements during the course of the analysis. Several experimental results are modelled using the commercial package ABAQUS/Standard with an automatic insertion subroutine developed in this work, and the results are presented to demonstrate the capabilities of this technique.

Cancer clinical trials have been one of the key foundations for significant advances in oncology. However, there is a clear recognition within the academic, care delivery and pharmaceutical/biotech communities that our current model of clinical trial discovery and development is no longer fit for purpose. Delivering transformative cancer care should increasingly be our mantra, rather than maintaining the status quo of, at best, the often miniscule incremental benefits that are observed with many current clinical trials. As we enter the era of precision medicine for personalised cancer care (precision and personalised medicine), it is important that we capture and utilise our greater understanding of the biology of disease to drive innovative approaches in clinical trial design and implementation that can lead to a step change in cancer care delivery. A number of advances have been practice changing (e.g. imatinib mesylate in chronic myeloid leukaemia, Herceptin in erb-B2-positive breast cancer), and increasingly we are seeing the promise of a number of newer approaches, particularly in diseases like lung cancer and melanoma. Targeting immune checkpoints has recently yielded some highly promising results. New algorithms that maximise the effectiveness of clinical trials, through for example a multi-stage, multi-arm type design are increasingly gaining traction. However, our enthusiasm for the undoubted advances that have been achieved are being tempered by a realisation that these new approaches may have significant cost implications. This article will address these competing issues, mainly from a European perspective, highlight the problems and challenges to healthcare systems and suggest potential solutions that will ensure that the cost/value rubicon is addressed in a way that allows stakeholders to work together to deliver optimal cost-effective cancer care, the benefits of which can be transferred directly to our patients.

Accurately encoding the duration and temporal order of events is essential for survival and important to everyday activities, from holding conversations to driving in fast flowing traffic. Although there is a growing body of evidence that the timing of brief events (< 1s) is encoded by modality-specific mechanisms, it is not clear how such mechanisms register event duration. One approach gaining traction is a channel-based model; this envisages narrowly-tuned, overlapping timing mechanisms that respond preferentially to different durations. The channel-based model predicts that adapting to a given event duration will result in overestimating and underestimating the duration of longer and shorter events, respectively. We tested the model by having observers judge the duration of a brief (600ms) visual test stimulus following adaptation to longer (860ms) and shorter (340ms) stimulus durations. The channel-based model predicts perceived duration compression of the test stimulus in the former condition and perceived duration expansion in the latter condition. Duration compression occurred in both conditions, suggesting that the channel-based model does not adequately account for perceived duration of visual events.

In this work we develop a methodology for the economic evaluation of soil tillage technologies, in a risky environment, and to capture the influence of farmer behaviour on his technology choice. The model has short-term activities, that change with the type of year, and long-term activities, in which sets of traction investment activities are included. Although these activities do not change with the type of year, they lead to different availability of resources for each type of year, since the same tractor has different available fieldwork days under different weather conditions. We prove that the model is sensitive to the greater income variability resulting from the use of alternative technologies and to the balance between income and risk, accounting for the probability of occurrence of each state of nature and giving an investment solution that considers the best production plan for each type of year. Over the last few decades, China has seen a steep rise in diverse eco city and low carbon city policies. Recently, attention has begun to focus on the perceived shortcomings in the practical delivery of related initiatives, with several publications suggesting a gap between ambitious policy goals and the emerging realities of the newly built environment. To probe this further, in this article we examine – based on the policy network approach – how the gap between high-level national policies and local practice implementation can be explained in the current Chinese context. We develop a four-pronged typology of eco city projects based on differential involvement of key (policy) actor groups, followed by a mapping of what are salient policy network relations among these actors in each type. Our analysis suggests that, within the overall framework of national policy, a core axis in the network relations is that between local government and land developers. In some cases, central government agencies– often with buy-in from international architecture, engineering and consulting firms – seek to influence local government planning through various incentives aimed at rendering sustainability a serious consideration. However, this is mostly done in a top-down manner, which overemphasizes a rational, technocratic planning mode while underemphasizing interrelationships among actors. This makes the emergence of a substantial implementation gap in eco city practice an almost predictable outcome. Consequently, we argue that special attention be paid in particular to the close interdependency between the interests of local government actors and those of land and real estate developers. Factoring in this aspect of the policy network is essential if eco city implementation is to gain proper traction on the ground.

Este trabalho é realizado no domínio das obras de engenharia, área onde o desmonte de rocha com recurso a explosivos em obras rodoviárias é uma actividade específica e consistiu no acompanhamento e execução de três obras rodoviárias de média e grande dimensão. A necessidade de executar escavações, recorrendo a técnicas de desmonte cuidadoso de contorno, onde o plano de corte do talude final deve obedecer a requisitos de localização, alinhamento, inclinação, estabilidade e também estéticos, acrescendo a isto a necessidade de optimizar os meios envolvidos, obriga a que esta actividade seja encarada de uma forma sistematizada, visando o racional aproveitamento de recursos. A execução desta actividade requer conhecimentos no domínio das técnicas de desmonte de contorno, dos explosivos, do mecanismo de rotura de rochas, da operação de perfuração e da geomecânica dos maciços. A abordagem deste trabalho incide sobre a técnica denominada de pré‐corte e tem como objectivo encontrar uma equação característica que permita relacionar diferentes parâmetros envolvidos nesta actividade. Este objectivo é alcançado recorrendo à correlação entre equações relativas à pressão de detonação, à pressão no furo e ao espaçamento entre furos consecutivos, desenvolvidas por outros autores. Desta forma obteve‐se uma equação que relaciona parâmetros relativos ao maciço rochoso (resistência à tracção), ao explosivo (velocidade de detonação e densidade) e ao diagrama de fogo (concentração de carga – volume de explosivo e comprimento do furo – volume do furo). A comparação entre os valores destes parâmetros obtidos na produção e os obtidos com recurso à equação característica permite concluir que a sua aplicação para execução de futuras obras possibilita uma optimização dos meios envolvidos.

A presente tese descreve diferentes soluções que permitem a reutilização da energia recuperada em ascensores eléctricos de roda de aderência dotados de conversores electrónicos de frequência e dessa forma contribuir para a melhoria da eficiência energética nos ascensores. Nos ascensores, a energia potencial é constantemente transferida enquanto a cabina está em movimento. Se a cabina se estiver a movimentar em sentido descendente com plena carga, ou em sentido ascendente, mas vazia, o motor estará em modo gerador. Quando a cabina se movimenta em sentido descendente, e o peso na cabina é superior ao peso do contrapeso, então o binário do motor encontra-se em sentido contrário à velocidade, isto é, o motor está a travar, havendo lugar à recuperação de energia. Igualmente, se a cabina subir vazia, também se poderá recuperar energia eléctrica. A energia acumulada em forma de energia potencial nas pessoas e no contrapeso pode ser recuperada, dado que o motor estará a funcionar como um gerador. De modo a estudar a viabilidade técnica e económica das diferentes soluções foram realizadas medições a uma amostra representativa de ascensores eléctricos de roda de aderência. Esta amostra é constituída por 39 ascensores que estão instalados em diferentes tipos de edifícios e que pertencem a diferentes categorias de utilização, de acordo com a norma VDI 4707:2009. Para cada ascensor foi medida a energia consumida e a energia gerada para uma manobra completa – a descida e a subida da cabina sem carga. A partir das medições, e com base na norma VDI 4707:2009 foram calculados os valores anualizados de energia eléctrica consumidos e produzidos por cada ascensor. A partir das 5 hipóteses identificadas para a utilização da energia recuperada (carregamento de bateria para alimentação dos circuitos em stand-by; carregamento de supercondensador para alimentação dos circuitos em stand-by; carregamento de supercondensador para alimentar o barramento DC; reinjecção da energia no barramento DC de um conjunto de ascensores em grupo; reinjecção da energia na rede eléctrica do edifício onde o ascensor está instalado) foi realizada a avaliação técnica e a avaliação económico-financeira para cada um dos ascensores. Por último, foi desenvolvido um simulador que permite definir a solução de recuperação de energia que seja técnica e economicamente mais viável, para um dado ascensor eléctrico de roda de aderência instalado, mediante a introdução dos parâmetros técnicos do ascensor em avaliação.

O uso de ligações adesivas aumentou significativamente nos últimos anos e é hoje em dia uma técnica de ligação dominante na indústria aeronáutica e automóvel. As ligações adesivas visam substituir os métodos tradicionais de fixação mecânicos na união de estruturas. A melhoria ao longo dos anos de vários modelos de previsão de dano, nomeadamente através do Método de Elementos Finitos (MEF), tem ajudado ao desenvolvimento desta técnica de ligação. Os Modelos de Dano coesivo (MDC), usados em conjunto com MEF, são uma ferramenta viável para a previsão de resistência de juntas adesivas. Os MDC combinam critérios da resistência dos materiais para a iniciação do dano e conceitos da mecânica da fratura para a propagação da fenda. Existem diversas formas de leis coesivas possíveis de aplicar em simulações por MDC, em função do comportamento expectável dos materiais que estão a ser simulados. Neste trabalho, estudou-se numericamente o efeito de diversas formas de leis coesivas na previsão no comportamento de juntas adesivas, nomeadamente nas curvas forçadeslocamento (P-) de ensaios Double-Cantilever Beam para caracterização à tração e ensaios End-Notched Flexure para caraterização ao corte. Também se estudou a influência dos parâmetros coesivos à tração e corte nas curvas P- dos referidos ensaios. Para o Araldite®AV138 à tração e ao corte, a lei triangular é a que melhor prevê o comportamento do adesivo. Para a previsão da resistência de ambos os adesivos Araldite® 2015 e SikaForce® 7752, a lei trapezoidal é a que melhor se adequa, confirmando assim que esta lei é a que melhor caracteriza o comportamento de dano de adesivos tipicamente dúcteis. O estudo dos parâmetros revelou influência distinta na previsão do comportamento das juntas, embora com bastantes semelhanças entre os diferentes tipos de adesivos.
