Resumo: A produção de leite de cabra no Brasil destaca-se no Nordeste, onde as condições de criação e os programas governamentais favorecem a produção. A qualidade higiênico-sanitária do leite de cabra, além de ser uma característica indispensável ao produto, exigida pela legislação Brasileira, contribui para diminuição nas perdas econômicas decorrentes da mastite e para a manutenção dos produtores na atividade. O Kit Embrapa de Ordenha Manual® para caprinos leiteiros é um conjunto de utensílios e práticas indispensáveis para a obtenção higiênica do leite de cabra, o qual apresenta-se como uma alternativa viável aos agricultores familiares devido à sua facilidade na utilização e baixo custo de aquisição. O Kit foi implantado e validado em 41 propriedades familiares, localizadas nos estados do Rio Grande do Norte, Paraíba e Ceará. Procedeu-se a coleta de leite de cabra 3 dias antes e 3 dias após a utilização do Kit, para a realização da Contagem Total de Bactérias (CTB). Observou-se uma redução média de 72% da CTB no leite produzido, e, 95% das propriedades adequaram-se à CTB exigida pela legislação Brasileira. [Effect of the use of the Milking Manual® EMBRAPA Kit for dairy goats total count in milk bacteria]. Abstract: The goat milk production in Brazil stands out in the Northeast, where the breeding conditions and government programs favor the production. The sanitary hygienic quality of goat milk as well as being an indispensable feature to the product, required by Brazilian law, contributes to decrease in economic losses resulting from mastitis and to the maintenance of the producers in the activity. The Embrapa manual milking kit® for dairy goats is an essential set of tools and practices for obtaining hygienic goat milk, which presents itself as a viable alternative to farmers due to its ease of use and low cost. The Kit has been deployed and validated in 41 family farms in the states of Rio Grande do Norte, Paraíba and Ceará. The goat milking practice was carried out both three days before and three days after using the kit for the realization of Total Bacteria Count (CTB). There was an average reduction of 72 % CTB on the milk, and 95 % of the properties have adapted to the CTB required by Brazilian law.

Com o objetivo de avaliar as características de crescimento de mangueiras cultivadas em sistema orgânico foram avaliados cinco compostos orgânicos elaborados a partir da combinação entre os insumos capim elefante, bagaço de coco, restos de poda de videira, esterco de caprino e torta de mamona. O ensaio foi disposto em blocos ao acaso, sendo constituído de seis tratamentos (cinco compostos e uma testemunha) e quatro repetições. Não houve efeito dos tratamentos sobre a matéria orgânica e a CTC do solo, nem sobre as características de crescimento da mangueira (número de fluxos por ramo e comprimento do primeiro fluxo).

A atividade física sempre que praticada conscientemente é um meio benéfico para qualquer indivíduo na promoção da sua saúde e do seu bem estar físico (Livro Verde da Atividade Física, 2011). Esta pode ser praticada independentemente da sua idade, género, saúde ou mesma da sua situação económica. Alguns dos seus benefícios são observados na diminuição do stress, na prevenção de doenças, na prevenção da perda de autonomia, autoestima, bem-estar, qualidade de vida e isto acaba por ser uma ponte para um bom ambiente familiar, profissional e social. Face ao exposto, foi definido como objetivo geral: Estudar os níveis de AF, a vulnerabilidade ao stress e os estilos de vida dos elementos da PSP; foram propostos como objetivos específcos: (1) Verificar o impacto da AF nos estilos de vida e no stress nos elementos policiais; (2) Avaliar o grau de relação entre os diferentes níveis AF, a vulnerabilidade ao stress e os estilos de vida; (3) Analisar o efeito da idade nos níveis de AF, no stress e no estilo de vida dos elementos. Neste estudo de caso participaram 245 elementos pertencentes à Esquadra do Funchal, Esquadra de Benfica, Esquadra de Cascais e EIR. Para realizar a investigação foram utilizados os seguintes instrumentos: International Physical Activity Questionaire (IPAQ) para medir a AF; 23 Questionário de Vulnerabilidade ao Stress (23QVS); Questionário de Estilo de Vida Fantástico (QEVF). Através destes instrumentos conseguiu-se obter os seguintes resultados: (1) Existe um impacto positivo da atividade física no estilo de vida e na diminuição do stress; (2) Uma relação positiva ente a AF e os estilos de vida, por outro lado não há relação entre os níveis AF e a vulnerabilidade ao stress. (3) A idade não surte qualquer efeito nos níveis de AF, no entanto o mesmo não se verifica na vulnerabilidade ao stress e no estilo de vida.

The main problem faced by the shrimp industry are the infectious diseases. The hypodermal and hematopoietic necrosis infection (IHHN) is one of the major cause of disease in the cultured shrimp, Litopenaeus vannamei. Environmental changes involving water quality, oxygen concentration, salinity, temperature, stocking density, presence of pathogens, among others, triggering a stressing condition for the cultured shrimp, weakening them and allowing the outbreak of diseases. The stress on the animal leads to a change in the molecules immune response components, which can be used as indicators of shrimp health. Thus, the objective of the present study was to evaluate the effect of salinity, stocking density and IHHNV infection on the L. vannamei shrimp. The immune parameters used to check the shrimp health were the total hemocytes counts (THC), the agglutinating activity (AA) and the clotting time (CT) of the serum of shrimp. These parameters were analyzed in healthy and IHHNV-infected shrimp, grown in low (0-0.5 ), medium (19-24 ) and high (> 38 ) salinity, and extensive (7-12 cam.m-2), semi-intensive (15-25 cam.m-2) and intensive (33-45 cam.m -2) stocking density. The IHHNV infection rate was significantly higher in low salinity (P<0.005) and intensive density (P<0.005), both stressful conditions for L. vannamei. Low salinity significantly increased THC (P<0.05) and decreased and CT (P<0.05) in healthy and infected shrimp, but AA (P<0.05) significantly decreased in healthy shrimp at medium salinity. Culture intensification did not affect the THC, AA and CT of healthy and infected shrimp (P>0.05). The IHHNV infection did not affect any immune parameters of shrimp cultured at different salinities and stocking densities. It is necessary to emphasize that this study was conducted in shrimp grown in ponds, where several environmental factors are acting simultaneously. CONTEXTO: O enfermeiro deve interagir com as pessoas em situação de saúde ou doença, melhorando o seu contexto sociocultural e o seu processo transição. Estas interações são organizadas com um propósito em que o enfermeiro utiliza ações terapêuticas para promover a saúde. A massagem terapêutica consiste em pressionar e esfregar o corpo ou partes deste com fins terapêuticos. OBJETIVO(S): Avaliar o efeito da intervenção da massagem terapêutica na saúde mental das pessoas com patologia oncológica. METODOLOGIA: Revisão sistemática da literatura, através dos descritores “pacientes”, “oncologia” e “massagem”, estudos de 2000 a 2012, em bases de dados e motores de busca online. RESULTADOS: O corpus do estudo foi composto por 15 artigos e demonstra que a massagem terapêutica reduz, quer de imediato, quer a longo prazo, a ansiedade e a depressão. Contudo, há estudos em que não existiram diferenças nestas variáveis. Esta intervenção melhora, de imediato, o relaxamento, o bem-estar emocional e o padrão de sono, a dor, o desconforto físico-emocional e a fadiga. Não houveram diferenças estatísticas significativas que corroborassem a melhoria da qualidade de vida, stress e sofrimento. CONCLUSÕES: A massagem terapêutica tem efeitos benéficos a curto prazo na saúde mental das pessoas com patologia oncológica. No entanto, relativamente à ansiedade e depressão os resultados analisados são contraditórios, reportando-se para investigações futuras. Sugerimos novas pesquisas para maior consistência empírica sobre os efeitos da intervenção da massagem terapêutica na saúde mental das pessoas com patologia oncológica e assim contribuir para a prática de uma enfermagem de excelência baseada na evidência.

Introdução: o stress pode influenciar negativamente o bem-estar dos estudantes de enfermagem, o seu estado de saúde e os resultados académicos. Apesar de serem conhecidas as principais variáveis intervenientes neste processo não existe evidência empírica sistematizada que oriente para a tomada de decisão sobre a inclusão de programas para a redução de stress nos cursos de enfermagem. Objetivo: identificar os estudos disponíveis acerca do efeito de intervenções para redução de stress em estudantes de enfermagem e analisar a consistência dos resultados obtidos. Metodologia: revisão sistemática de literatura na B-on, TRIP e Cochrane, com os seguintes descritores, nas línguas português, espanhol e inglês: Estudantes de enfermagem + Stress, para busca em título; e, Intervenção ou programa + Impacto ou Efeito ou Eficácia ou Influencia, em resumo. Resultados: foram analisados 14 artigos que obedeceram aos critérios de inclusão. A maioria são estudos quasi-experimentais (11), com uma grande variedade de programas de intervenção, mas encontrando diminuição estatisticamente significativa para ansiedade e stress. O resultado para depressão não é consistente. Discusao: não existem estudos com follow-up em número suficiente para compreender os efeitos a longo prazo. Conclusão: as intervenções para reduzir o stress em estudantes de enfermagem parecem ser eficazes a curto prazo numa grande variedade de programas. São necessários estudos que analisem os efeitos a longo prazo.

Introdução: Os profissionais de saúde estão sujeitos a Burnout. O Reiki é uma terapia complementar em que é transmitida a “Energia Universal” para as zonas mais necessitadas da pessoa, podendo apresentar benefícios nos profissionais de saúde. Objetivo: Efetuar uma revisão sistemática da literatura de forma a analisar a literatura mais recente sobre os efeitos do Reiki nos profissionais de saúde sujeitos a stress/burnout. Métodos: Efetuadas pesquisas nas bases de dados electrónicas Cinahl Plus, Medline, Lilacs e Cochrane e Library no motor de busca da EBSCO, na Biblioteca do Conhecimento Online (B-On) sendo selecionados 7 artigos. A pesquisa ocorreu durante o mês de Abril de 2016. Em artigos em português, inglês ou espanhol com publicação entre Janeiro de 2011 e Abril de 2016, fazendo referência a Reiki e profissionais de saúde. Resultados: Da análise efetuada, emergem duas grandes categorias relativamente aos benefícios do Reiki nos profissionais de saúde: benefícios a nível pessoal e profissional. A nível pessoal, os benefícios relacionados com a saúde física e com a saúde mental. Aspetos físicos como a tensão arterial e a imunidade são influenciados de forma positiva pelo Reiki; na saúde mental diminui o stress relacionado com o trabalho e aumenta a autoconfiança. Conclusões: O Reiki é benéfico para os profissionais de saúde sujeitos a stress/burnout, melhorando a saúde, diminuindo o stress, proporcionando autoconfiança e bem-estar.

The purpose of this bibliography review was to approach the thermal comfort rates on milk production of goats from Alpine and Saanen breeds in Brazil. The caloric stress caused by weather changes to which the animals are submitted, influence on the mechanisms of normal physiological processes of the body. Thus, the effect on the lactation process in goats can be mentioned, where it decreases the amount of water in the body with the consequent decrease in synthesis and milk ejection interfering in the production of hormone prolactin and growth hormone. The animal?s interaction with the environment must be considered when the aim in livestock farming is welfare, because the different responses of the animal to the peculiarities of each region are crucial for the success of the animal adaptation. So, the correct identification of the factors that influence the productive life of the animal, such as the stress caused by the seasonal fluctuations of the environment, allow production systems management, making it possible to make them sustainable and viable. The maintenance of these parameters in normal levels is very important, to the point of being used in the evaluation of climate adaptability of breeds to a certain environmental condition. In this way, the concerns about animal welfare and environmental comfort are due to the climatic variables and the behavioral, physiological and productive responses are prevailing when implementing the suitability of certain production systems.

Although various abutment connections and materials have recently been introduced, insufficient data exist regarding the effect of stress distribution on their mechanical performance. The purpose of this study was to investigate the effect of different abutment materials and platform connections on stress distribution in single anterior implant-supported restorations with the finite element method. Nine experimental groups were modeled from the combination of 3 platform connections (external hexagon, internal hexagon, and Morse tapered) and 3 abutment materials (titanium, zirconia, and hybrid) as follows: external hexagon-titanium, external hexagon-zirconia, external hexagon-hybrid, internal hexagon-titanium, internal hexagon-zirconia, internal hexagon-hybrid, Morse tapered-titanium, Morse tapered-zirconia, and Morse tapered-hybrid. Finite element models consisted of a 4×13-mm implant, anatomic abutment, and lithium disilicate central incisor crown cemented over the abutment. The 49 N occlusal loading was applied in 6 steps to simulate the incisal guidance. Equivalent von Mises stress (σvM) was used for both the qualitative and quantitative evaluation of the implant and abutment in all the groups and the maximum (σmax) and minimum (σmin) principal stresses for the numerical comparison of the zirconia parts. The highest abutment σvM occurred in the Morse-tapered groups and the lowest in the external hexagon-hybrid, internal hexagon-titanium, and internal hexagon-hybrid groups. The σmax and σmin values were lower in the hybrid groups than in the zirconia groups. The stress distribution concentrated in the abutment-implant interface in all the groups, regardless of the platform connection or abutment material. The platform connection influenced the stress on abutments more than the abutment material. The stress values for implants were similar among different platform connections, but greater stress concentrations were observed in internal connections.

Lutein (LT) is a carotenoid obtained by diet and despite its antioxidant activity had been biochemically reported, few studies are available concerning its influence on the expression of antioxidant genes. The expression of 84 genes implicated in antioxidant defense was quantified using quantitative reverse transcription polymerase chain reaction array. DNA damage was measured by comet assay and glutathione (GSH) and thiobarbituric acid reactive substances (TBARS) were quantified as biochemical parameters of oxidative stress in mouse kidney and liver. cDDP treatment reduced concentration of GSH and increased TBARS, parameters that were ameliorated in treatment associated with LT. cDDP altered the expression of 32 genes, increasing the expression of GPx2, APC, Nqo1 and CCs. LT changed the expression of 37 genes with an induction of 13 mainly oxygen transporters. In treatments associating cDDP and LT, 30 genes had their expression changed with a increase of the same genes of the cDDP treatment alone. These results suggest that LT might act scavenging reactive species and also inducing the expression of genes related to a better antioxidant response, highlighting the improvement of oxygen transport. This improved redox state of the cell through LT treatment could be related to the antigenotoxic and antioxidant effects observed.

Uncoupling protein one (UCP1) is a mitochondrial inner membrane protein capable of uncoupling the electrochemical gradient from adenosine-5'-triphosphate (ATP) synthesis, dissipating energy as heat. UCP1 plays a central role in nonshivering thermogenesis in the brown adipose tissue (BAT) of hibernating animals and small rodents. A UCP1 ortholog also occurs in plants, and aside from its role in uncoupling respiration from ATP synthesis, thereby wasting energy, it plays a beneficial role in the plant response to several abiotic stresses, possibly by decreasing the production of reactive oxygen species (ROS) and regulating cellular redox homeostasis. However, the molecular mechanisms by which UCP1 is associated with stress tolerance remain unknown. Here, we report that the overexpression of UCP1 increases mitochondrial biogenesis, increases the uncoupled respiration of isolated mitochondria, and decreases cellular ATP concentration. We observed that the overexpression of UCP1 alters mitochondrial bioenergetics and modulates mitochondrial-nuclear communication, inducing the upregulation of hundreds of nuclear- and mitochondrial-encoded mitochondrial proteins. Electron microscopy analysis showed that these metabolic changes were associated with alterations in mitochondrial number, area and morphology. Surprisingly, UCP1 overexpression also induces the upregulation of hundreds of stress-responsive genes, including some involved in the antioxidant defense system, such as superoxide dismutase (SOD), glutathione peroxidase (GPX) and glutathione-S-transferase (GST). As a consequence of the increased UCP1 activity and increased expression of oxidative stress-responsive genes, the UCP1-overexpressing plants showed reduced ROS accumulation. These beneficial metabolic effects may be responsible for the better performance of UCP1-overexpressing lines in low pH, high salt, high osmolarity, low temperature, and oxidative stress conditions. Overexpression of UCP1 in the mitochondrial inner membrane induced increased uncoupling respiration, decreased ROS accumulation under abiotic stresses, and diminished cellular ATP content. These events may have triggered the expression of mitochondrial and stress-responsive genes in a coordinated manner. Because these metabolic alterations did not impair plant growth and development, UCP1 overexpression can potentially be used to create crops better adapted to abiotic stress conditions.

Taurine is a sulfur-containing amino acid that exerts protective effects on vascular function and structure in several models of cardiovascular diseases through its antioxidant and anti-inflammatory properties. Early protein malnutrition reprograms the cardiovascular system and is linked to hypertension in adulthood. This study assessed the effects of taurine supplementation in vascular alterations induced by protein restriction in post-weaning rats. Weaned male Wistar rats were fed normal- (12%, NP) or low-protein (6%, LP) diets for 90 days. Half of the NP and LP rats concomitantly received 2.5% taurine supplementation in the drinking water (NPT and LPT, respectively). LP rats showed elevated systolic, diastolic and mean arterial blood pressure versus NP rats; taurine supplementation partially prevented this increase. There was a reduced relaxation response to acetylcholine in isolated thoracic aortic rings from the LP group that was reversed by superoxide dismutase (SOD) or apocynin incubation. Protein expression of p47phox NADPH oxidase subunit was enhanced, whereas extracellular (EC)-SOD and endothelial nitric oxide synthase phosphorylation at Ser 1177 (p-eNOS) were reduced in aortas from LP rats. Furthermore, ROS production was enhanced while acetylcholine-induced NO release was reduced in aortas from the LP group. Taurine supplementation improved the relaxation response to acetylcholine and eNOS-derived NO production, increased EC-SOD and p-eNOS protein expression, as well as reduced ROS generation and p47phox expression in the aortas from LPT rats. LP rats showed an increased aortic wall/lumen ratio and taurine prevented this remodeling through a reduction in wall media thickness. Our data indicate a protective role of taurine supplementation on the high blood pressure, endothelial dysfunction and vascular remodeling induced by post-weaning protein restriction. The beneficial vascular effect of taurine was associated with restoration of vascular redox homeostasis and improvement of NO bioavailability.

Ki-1/57 (HABP4) and CGI-55 (SERBP1) are regulatory proteins and paralogs with 40.7% amino acid sequence identity and 67.4% similarity. Functionally, they have been implicated in the regulation of gene expression on both the transcriptional and mRNA metabolism levels. A link with tumorigenesis is suggested, since both paralogs show altered expression levels in tumor cells and the Ki-1/57 gene is found in a region of chromosome 9q that represents a haplotype for familiar colon cancer. However, the target genes regulated by Ki-1/57 and CGI-55 are unknown. Here, we analyzed the alterations of the global transcriptome profile after Ki-1/57 or CGI-55 overexpression in HEK293T cells by DNA microchip technology. We were able to identify 363 or 190 down-regulated and 50 or 27 up-regulated genes for Ki-1/57 and CGI-55, respectively, of which 20 were shared between both proteins. Expression levels of selected genes were confirmed by qRT-PCR both after protein overexpression and siRNA knockdown. The majority of the genes with altered expression were associated to proliferation, apoptosis and cell cycle control processes, prompting us to further explore these contexts experimentally. We observed that overexpression of Ki-1/57 or CGI-55 results in reduced cell proliferation, mainly due to a G1 phase arrest, whereas siRNA knockdown of CGI-55 caused an increase in proliferation. In the case of Ki-1/57 overexpression, we found protection from apoptosis after treatment with the ER-stress inducer thapsigargin. Together, our data give important new insights that may help to explain these proteins putative involvement in tumorigenic events.

Beta cell destruction in type 1 diabetes (TID) is associated with cellular oxidative stress and mitochondrial pathway of cell death. The aim of this study was to determine whether oxidative stress and mitochondrial dysfunction are present in T1D model (non-obese diabetic mouse, NOD) and if they are related to the stages of disease development. NOD mice were studied at three stages: non-diabetic, pre-diabetic, and diabetic and compared with age-matched Balb/c mice. Mitochondria respiration rates measured at phosphorylating and resting states in liver and soleus biopsies and in isolated liver mitochondria were similar in NOD and Balb/c mice at the three disease stages. However, NOD liver mitochondria were more susceptible to calcium-induced mitochondrial permeability transition as determined by cyclosporine-A-sensitive swelling and by decreased calcium retention capacity in all three stages of diabetes development. Mitochondria H2O2 production rate was higher in non-diabetic, but unaltered in pre-diabetic and diabetic NOD mice. The global cell reactive oxygen species (ROS), but not specific mitochondria ROS production, was significantly increased in NOD lymphomononuclear and stem cells in all disease stages. In addition, marked elevated rates of 2',7'-dichlorodihydrofluorescein (H2DCF) oxidation were observed in pancreatic islets from non-diabetic NOD mice. Using matrix-assisted laser desorption/ionization (MALDI) mass spectrometry (MS) and lipidomic approach, we identified oxidized lipid markers in NOD liver mitochondria for each disease stage, most of them being derivatives of diacylglycerols and phospholipids. These results suggest that the cellular oxidative stress precedes the establishment of diabetes and may be the cause of mitochondrial dysfunction that is involved in beta cell death.
