This study evaluates the influence of depth and environmental parameters on the development of Gracilaria birdiae Plastino & Oliveira (Gracilariaceae Rhodophyta) in an organic shrimp pound (Litopenaeus vannamei) under euthrophical conditions. PVC structures (module) witch four ropes laden with 150 g of macroalgae each, were kept during 35 days at three different depths (surface, 10 and 20 cm depth). Wet biomass weighing and environmental parameters (temperature, salinity, turbidity, pH, transparence, precipitation, evaporation, insolation, accumulated solar radiation, nitrite, nitrate, ammonium and orthophosphate) were measured weekly. At all three proposed depths, the macroalgae displayed a higher biomass at the end of experiment than at the initial inoculations. The module kept at a 10 cm depth presented the greatest average biomass (186,3), followed by that kept at 20 cm (180,4 g) and the surface module (169,9 g). Biomass variations showed algae to suffer the direct effects of depths. Biomass loss was associated with the factors that influence light penetration, such as sediment deposits above the thallus, rate of evaporation and precipitation. The smallest loses occurred in the algae kept on surface (0,16%), followed by the algae kept at 20 cm (0,20%) and 10 cm (0,22%). The specific growth rate (SGR) of G. birdiae showed no significant difference between the three depths nor the sample periods. Nevertheless, the modules kept at 10 and 20 cm depths presented similar growth evolution, both growing 0,38%·per day-1, while the module kept on surface had an average SGR of 0,36%·day-1. The models related to growth rate demonstrated temperature, salinity, pH, orthophosphate, ammonium, precipitation and turbidity as the principal environmental parameters influencing the development of G. birdiae

The mushrooms have been object of intense research in view of its potential raising of application in different sectors of the pharmacology and alimentary industry. Among diverse bioactive composites of polyssacharides nature that exist in the fungus the glucans are much searched. These are polymers of glucose and classified as the type of glicosidic linking [α, β]. Peroxisome proliferator-activated receptors (PPARs), ranscription factors belonging to the family of nuclear receptors that bind themselves o specific agonists, have shown their importance in controlling the inflammatory process. The aim of this study was to perform a chemical characterization of extract rom the mushroom Caripia montagnei, assess its antiinflammatory and antibacterial effect and determine if this effect occurs via PPAR. This mushroom is composed of carbohydrates (63.3±4.1%), lipids (21.4l±0.9%) and proteins (2.2± 0.3%). The aqueous solution resulting from the fractionation contained carbohydrates (98.7±3.3%) and protein (1.3±0.25%). Analyses of infrared spectrophotometry and of nuclear magnetic esonance demonstrated that the extract of mushroom C. montagnei is rich in β-glucans. In hioglycolate-induced peritonitis, the C. montagnei glucans (50 mg/kg) educed the inflammatory process in 65.5±5.2% and agonists, pharmacological igands, for PPAR: Wy-14643 (49.3±6.1%), PFOA (48.9±3.8%) and clofibrate in 45.2±3.2%. Sodium diclofenac showed a reduction of 81.65±0.6%. In the plantar edema, the glucans from C. montagnei (50 mg/kg) and L-NAME reduced the edema to a similar degree 91.4±0.3% and 92.8±0,5 %, respectively. In all the groups tested, nitric oxide (NO), an inflammation mediator, showed a significant reduction in the nitrate/nitrite levels when compared to the positive control (P<0.001). The C. montagnei glucans did not show cytotoxicity in the concentrations tested (2.5, 5.0, 10.0, 20.0 and 40.0 µg/100 µL). Antibacterial activity demonstrated that, unlike total extract, there was no inhibition of bacterial growth. The C. montagnei glucans show great potential for antiinflammatory applications. This effect suggests that it is mediated by PPAR activation and by COX and iNOS inhibition

O cultivo do arroz no RS é fortemente relacionado com recursos hídricos, pois este se faz sob irrigação e durante o beneficiamento, especialmente no caso da parboilização, são necessários volumes consideráveis de água. Em vista disso, os solutos presentes nestas águas permitem estimar o impacto desta cultura e sua industrialização na qualidade do ambiente hídrico no RS. Foi desenvolvida uma rotina analítica para caracterização físico-química das águas da cadeia produtiva do arroz, onde foram analisados nitrogênio total, nitrogênio amoniacal, nitrogênio orgânico, açucares redutores, açucares redutores totais, sólidos totais, sólidos fixos, sólidos voláteis, ácidos voláteis, alcalinidade, pH, nitrato, nitrito e fósforo. Também um método multimicotoxinas foi desenvolvido avaliando a eficiência de cinco sistemas de extração-partição para aflatoxina B1 (AFA B1), aflatoxina B2 (AFA B2), ocratoxina A (OTA), zearalenona (ZEA) e deoxinivalenol (DON). A partição liquido-liquido com clorofórmio foi a mais eficiente recuperando 89; 96; 89; 97 e 72%, respectivamente de cada micotoxina. A comparação entre volumes de solventes, onde o número de etapas envolvidas e o tempo necessário para efetuar a determinação confirmaram a simplicidade, a economia e a rapidez deste sistema de partição a ser adotado para extrair micotoxinas de água da cadeia produtiva de arroz. O método foi aplicado para determinação de micotoxinas em amostras de águas de irrigação e parboilização de arroz de diferentes procedências verificando-se que as últimas aparecem contaminadas com micotoxinas, AFA B1 9,0 ng mL-1 e DON 110 ng mL-1 . Foi realizado também um estudo de migração de micotoxinas AFA B1, OTA, ZEA e DON e outros solutos sob condições de encharcamento (4 e 6 h) em dois níveis de fortificação, durante a parboilização do grão ficando demonstrado que durante o processo ocorria a lixiviação de solutos e micotoxinas para água de encharcamento do arroz.

A tecnologia do sistema de cultivo com bioflocos (BFT) permite a intensificação da densidade de estocagem, bem como o aumento da produtividade no qual exige esforços no manejo para a manutenção de qualidade de água. No entanto, um dos problemas é a liberação de nitrogênio através de alimento não consumidos, juntamente com as excretas dos organismos cultivados, principalmente na forma de amônia total. A adição de fontes de carbono orgânico é uma alternativa na redução de amônia tornando possível a conversão deste composto em proteína bacteriana, sendo disponível como alimento suplementar no ambiente de cultivo. Portanto foram testadas fontes de carbono como o melaço de cana-de-açúcar, dextrose e farelo de arroz no cultivo do camarão L. vannamei em sistemas BFT, avaliando a redução da concentração do nitrogênio amoniacal total em experimentos nas fases de berçário e engorda, bem como a análise de desempenho zootécnico dos animais. Os experimentos foram realizados utilizando caixas com volume útil de 800 L. O experimento berçário teve a duração de 35 dias, com densidade de estocagem de 1200 camarões m- ² e peso médio de 0,024±0,01 g. As fontes de carbono melaço de cana-de-açúcar (M) e farelo de arroz (F) foram combinadas em diferentes percentuais. No experimento engorda foi utilizada densidade de estocagem de 300 camarões m- ², peso médio de 4,09±0,51 g, durante 70 dias. Os tratamentos foram distinguidos pelas fontes de carbono dextrose e farelo de arroz. No experimento berçário a concentração da amônia foi significativamente menor (p<0,05) com o uso do melaço. O nitrito acumulou até o final do experimento, mas devido ao curto período experimental não interferiu nos índices de desempenho zootécnico, nos quais não apresentaram diferenças pelo uso das fontes de carbono. No experimento engorda, com o uso da dextrose, a concentração de amônia foi significativamente menor (p<0,05). Apesar de elevadas concentrações de nitrito, as sobrevivências foram similares e acima de 80%. Dados zootécnicos como peso final, ganho de peso semanal, conversão alimentar aparente e produtividade foram significativamente melhores (p<0,05) no tratamento farelo de arroz. Em ambos os experimentos as fontes de carbono de degradação rápida, como o melaço e a dextrose foram mais eficientes na redução de amônia. A degradação rápida destas fontes podem ter disponibilizado maiores teores de carbono como substrato para as bactérias heterotróficas metabolizarem a amônia, melhorando a qualidade da água.

The Iota, Kappa and Lambda commercial carrageenans are rarely pure and normally contain varying amounts of the other types of carrageenans. The exact amount of impurity depends on the seaweed source and extraction procedure. Then, different analysis methods have been applied for determination of the main constituents of carrageenans because these three carrageenans are extensively used in food, cosmetic and pharmaceutical industry. The electrophoresis of these compounds proved that the carrageenans are constituted by sulfated polysaccharides. These compounds were characterized by colorimetric methods and was observed that the Lambda carrageenan shown the greater value (33.38%) of sulfate. These polymers were examined by means of 13C NMR spectroscopy and infrared spectra. The polysaccharides consisted mainly of units alternating of sulfated galactoses and anhydrogalactoses. The aim of the study was also to test the inflammatory action of these different polysaccharides. A suitable model of inflammation is acute sterile inflammation of the rat hind limb induced by carrageenan. Paw edema was induced by injecting carrageenans (κ, ι and λ) in saline into the hind paw of a male Wistar rats (175–200 g). The pathway to acute inflammation by carrageenan (kappa, iota and lambda) were expressed as time-edema dependence and measured by paw edema volume. For this purpose, was used an apparatus (pakymeter), which makes it possible to measure the inflammation (swelling of the rat foot) with sufficient accuracy. The results showed that κ-carrageenan (1%) have an edema of 3.7 mm and the paw edema increase was time and dose dependent; the ι-carrageenan (0.2%) caused an edema of 4 mm and the λ-carrageenan (1%) caused an edema of 3.6 mm. Other model was used in this study based in the inflammation of pleura for comparatives studies. Injection of carrageenans into the pleural cavity of rat induced an acute inflammatory response characterized by fluid accumulation in the pleural cavity, a large number of neutrophils and raised NO production. The levels of NO were measured by Griess reactive. The ι-carrageenan caused the greater inflammation, because it has high concentration of nitrite/nitrate (63.478 nmoles/rat), exudato volume (1.52 ml) and PMNs (4902 x 103 cells). Quantitative evaluation of inflammations of rats is a useful and important parameter for the evaluation of the efficacy of anti-inflammatory drugs

Rheumatoid arthritis (RA) is systemic auto imune disorder. It is caracterized by chronic inflammation of joints leading to progressive erosion of cartilage and bone. We investigated the effect of the administration of fucoidan, sulfated polysaccharides, from algae Fucus vesiculosus in the acute (6h) in zymosan-induced arthritis (AZy). Wistar rats (180-230 g) were used for all groups experimental. Non-treated animals received just intraarticular injection of 1 mg the zymosan, control group received intraarticular injection of 50 µL the saline, groups received either fucoidan of Fucus vesiculosus (15, 30, 50 or 70 mg/Kg) or parecoxib (1 mg/Kg) 1 hour after injection of zymosan. After 6 h, the articular exudates were collected for evaluation of the cell influx and nitrite (Griess reaction) release. The sinovial membranes and articular cartilages were excised for histopathological analysis and by determination of the glycosaminoglycan (GAG), respectively. ZyA led to increased NO and cell influx into the joints. Therapeutic administration of the fucoidan or parecoxib did significantly inhibited the cell influx and the synovitis, as compared to non-treated rats (p<0,05), though being able to reduced NO release. Representative agarose gel electrophoresis of the GAGs, the content of condroitin-sulphate was observed during the process. These findings suggest that the fucoidan from Fucus vesiculosus has potential anti-inflammatory activity

The fucoidan from Fucus vesiculosus is known for having diverse biological properties. This study analyzed the therapeutic action of populations of commercial fucoidan (F. vesiculosus) on zymosan-induced arthritis. Three populations of fucoidan were obtained after acetone fractionation; these were denominated F1 (52.3%), F2 (36.7%) and F3 (10.7%). Chemical analyses showed that F1 contained the largest amount of sulfate ion. The electrophoretic profile shows that the commercial or total fucoidan (TF), different from the other fucoidans and from glycosaminoglycan patterns, is quite polydisperse, which indicates that it is composed of a mixture of sulfate polysaccharides. On the other hand, the fucoidans obtained from TF showed only an electrophoretic band with much lower polydispersion than that observed for TF. Fucoidan F2 showed a migration between fucoidans F1 and F3. Owing to the small amount of mass obtained from F3, we used only fucoidans F1 and F2 in the induced arthritis tests. After 1 hour of induction, we administered F1 or F2 (10, 25 and 50 mg/kg i.p.) or diclofenac sodium (10 mg/kg i.p.) or lumiracoxib (5 mg/kg o.a.) or L-NAME (30 mg/kg i.p.). After 6 hours, we performed analyses of cell influx and nitrite levels in addition to histopathological analysis. Fucoidans F1 and F2 were more potent both in decreasing the number of leukocytes and the amount of nitric oxide found in the synovial fluid. This indicates that the anti-inflammatory mechanism of these fucoidans is not only related to selectin block, but also to nitric oxide synthesis inhibition

This thesis has as objective presents a methodology to evaluate the behavior of the corrosion inhibitors sodium nitrite, sodium dichromate and sodium molybdate, as well as your mixture, the corrosion process for the built-in steel in the reinforced concrete, through different techniques electrochemical, as well as the mechanical properties of that concrete non conventional. The addition of the inhibitors was studied in the concrete in the proportions from 0.5 to 3.5 % regarding the cement mass, isolated or in the mixture, with concrete mixture proportions of 1.0:1.5:2.5 (cement, fine aggregate and coarse aggregate), superplasticizers 2.0 % and 0.40 water/cement ratio. In the modified concrete resistance rehearsals they were accomplished to the compression, consistence and the absorption of water, while to analyze the built-in steel in the concrete the rehearsals of polarization curves they were made. They were also execute, rehearsals of corrosion potential and polarization resistance with intention of diagnose the beginning of the corrosion of the armors inserted in body-of-proof submitted to an accelerated exhibition in immersion cycle and drying to the air. It was concluded, that among the studied inhibitors sodium nitrite , in the proportion of 2.0 % in relation to the mass of the cement, presented the best capacity of protection of the steel through all the studied techniques and that the methodology and the monitoring techniques used in this work, they were shown appropriate to evaluate the behavior and the efficiency of the inhibitors

The construction industry is one of the largest consumers of natural raw materials, and concrete is considered today the most used material wide. This accentuated consumption of natural resources has generated concern with the preservation of the environment, and has motivated various studies related to the use of resid ues, which can partially or entirely substitute, with satisfactory performance, some materials such as the aggregate, and in so doing, decrease the impact on the environment caused by the produced residues. Research has been done to better understand and improve the microstructure of concrete, as well as to understand the mechanism of corrosion in reinforced steel. In this context, this work was developed aiming at discovering the influence of the substitution of natural sand by artificial sand, with rega rd to mechanical resistance, microstructure, and durability. To obtain the electrochemical parameters, an adaptation was made to the galvanostatic electrochemical method to study the corrosion in reinforced steel. Concretes of categories 20 MPa and 40 MPa were produced, containing natural sand, and concretes of the same categories were produced with artificial sand substituting the natural sand, and with the addition of sodium nitrate and sodium chloride. Due to the use of rock dust reject (artificial sand), an evaluation was made of its environmental risk. The results indicate that the concretes of category 20 MPa present a better performance than the concrete made with natural sand, thus making it a viable substitute. For the category 40 MPa, the better performance is from the concrete containing natural sand. The adaptation of the galvanostatic electrochemical technique to the study of the corrosion of reinforced steel within concrete proved to be valid for obtaining electrochemical parameters with a high degree of reliability, considering the number of degrees of freedom

Devido ao crescente aumento do uso de aeradores artificiais em sistema de cultivo de peixes e à carência de estudos nesta área, este trabalho avaliou a influência de um aerador tipo propeller diffuser (AR 120) nas variáveis bióticas e abióticas em um viveiro de engorda de peixe. Foram demarcados três pontos de coleta localizados a 5, 10 e 23 m de distância do aerador, durante 15 dias consecutivos, sendo 5 dias antes do uso do aerador, 5 dias com o aerador ligado e 5 dias com o aerador desligado. As variáveis limnológicas estudadas não apresentaram diferenças significativas (P > 0,05) entre os três pontos de coleta, porém, em relação ao uso do aerador, variáveis como temperatura, transparência, pH, oxigênio dissolvido, bicarbonato, CO2 livre, fósforo total, ortofosfato, amônia, nitrato e nitrito diferiram significativamente (P < 0,05) com a agitação mecânica da água. Já a condutividade, alcalinidade, CO2 total e clorofila a não apresentaram diferenças significativas (P > 0,05) com o uso do aerador. A comunidade fitoplanctônica sofreu influência direta do aerador (P < 0,05), dominada pelas Chlorophyta, representando mais de 70% do total de organismos presentes, seguidas das Cyanophyta e Chrysophyta, porém estas últimas tenderam a aumentar após o uso do aerador, uma vez que estes grupos se adaptam rapidamente às mudanças do ambiente.

Devido à importância das condições das variáveis da água e do alimento disponível no desenvolvimento e sobrevivência das larvas de peixes, o objetivo deste estudo foi avaliar o efeito do uso de dois tratamentos alimentares (ração + zooplâncton e somente zooplâncton) e a qualidade da água, em tanques de larvicultura de Brycon orbignyanus. A transparência total da água (45 cm) esteve associada principalmente ao curto tempo de residência, fluxo contínuo e baixa profundidade. As concentrações de oxigênio dissolvido variaram de 1,32 a 7,00 mg.L-1 no tratamento contendo ração + zooplâncton, e de 1,82 a 7,60 mg.L-1 no tratamento contendo somente zooplâncton. Os nutrientes foram diretamente influenciados pela adição de ração no meio com exceção do nitrito. Rotifera apresentou a maior densidade, variando ao longo do período experimental de 8,7 x 10(5) a 1,3 x 10(6) org.m-3 entre os quatro tanques estudados, num total de dez espécies. A menor densidade foi observada para Cladocera, variando de 4,7 x 10(4) a 2,1 x 10(5) org.m-3 num total de seis espécies encontradas, sendo Diaphanosoma birgei a única espécie classificada como muito freqüente. O uso de ração junto com zooplâncton proporcionou melhor rendimento para larvas de B. orbignyanus.

The integrated culture of seaweed and aquatic animals is an ancient practice in Asian countries. The expansion of this practice to western countries is consequence of the recognition of this system as a sustainable alternative that allows economical diversification and mitigation of environmental impacts generated by effluents of aquaculture. This study evaluated the growth of the seaweed Gracilaria caudata and of the shrimp Litopenaeus vannamei in monoculture (shrimps) and integrated culture (shrimps and algae) systems, and accessed the effect of the seaweed in the water quality. There were two treatments in the experiment: monoculture (shrimps) and integrated culture (shrimps/ algae). The organisms were cultured in 6 aquaria (10L) filled with seawater (35.0±0.0 PSU and 28.1±0.4°C) for 28 days. The nutrients of water (PO43-, NH4+, NO2-, NO3- and DIN), the biomass and the relative growth rate (RGR, % day-1) of seaweed and shrimps were measured weekly. The parameters pH, temperature, salinity and dissolved oxygen were measured daily. The concentration of NH4+ in integrated culture (62.8±25.2µM) was lower (Mann-Whitney p<0.001) than in monoculture (85.6±24.3µM). The mean of PO4- in monoculture (10.4±4.6µM) was markedly higher (Mann-Whitney; p=0.024) than that in integrated culture (8.7±4.1µM). The level of dissolved oxygen in integrated culture (6.0±0.6mg/L) was higher (t-Student; P=0.014) than that in shrimp monoculture (5.8±0.6mg/L). The mean values of the parameters pH, NO2-, NO3- and DIN were 7.5±0.2, 10.1±12.2µM, 24.5±3.2µM and 120.17±30.76µM in monoculture, and 7.5±0.2, 10.5±13.2µM, 27.4±3.5µM and 100.76±49.59µM in integrated culture. There were not differences in these parameters between treatments. The biomass and RGR of seaweed reached 15.0±1.9g and 7.4±2.8% day-1 at the end of the experiment. The performance of shrimp was favorable in monoculture (1.5±0.8g; 5.7±1.6% dia-1) and in integrated culture (1.5±0.7g; 5.2±1.2% dia-1), and the rate of survival was 100% in both treatments. The tolerance and favorable performance of Gracilaria caudata suggest that this seaweed might be integrated into shrimp (Litopenaeus vannamei) culture systems
