Introduction: The aim of this study was to assess the effect of nitrogen ion implantation on the flexibility of rotary nickel-titanium (NiTi) instruments as measured by the load required to bend implanted and nonimplanted instruments at a 30 degrees angle. Methods: Thirty K3 files, size #40, 0.02 taper and 25-mm length, were allocated into 2 groups as follows: group A, 15 files exposed to nitrogen ion implantation at a dose of 2.5 x 10(17) ions/cm(2), voltage 200 KeV, current density 1 mu A/cm(2), temperature 130 degrees C, and vacuum conditions of 10 x 10(-6) mm Hg for 6 hours; and group B, 15 nonimplanted files. One extra file was used for process control. All instruments were subjected to bend testing on a modified troptometer, with measurement of the load required for flexure to an angle of 30 degrees. The Mann-Whitney U test was used for statistical analysis. Findings with P <.05 were considered significant. Results: The mean load required to bend instruments at a 30 degrees angle was 376.26 g for implanted instruments and 383.78 g for nonimplanted instruments. The difference was not statistically significant. Conclusions: Our findings show that nitrogen ion implantation has no appreciable effect on the flexibility of NiTi instruments. The affinity of the d-galactose-binding lectin from Artocarpus heterophyllus lectin, known as jacalin, with immonuglobulins (Igs) was determined by biofunctionalization of a piezoelectric transducer. This piezoelectric biofunctionalized transducer was used as a mass-sensitive analytical tool, allowing the real-time binding analysis of jacalin-human immunoglobulin A1 (IgA(1)) and jacalin-bovine IgG(1) interactions from which the apparent affinity constant was calculated. The strategy was centered in immobilizing jacalin on the gold electrode's surface of the piezoelectric crystal resonator using appropriate procedures based on self-assembling of 11-mercaptoundecanoic acid and 2-mercaptoethanol thiol's mixture, a particular immobilization strategy by which it was possible to avoid cross-interaction between the proteins over electrode's surface. The apparent affinity constants obtained between jacalin-human IgA(1) and jacalin-bovine IgG(1) differed by 1 order of magnitude [(8.0 +/- 0.9) x 10(5) vs (8.3 +/- 0.1) x 10(6) L mol(-1)]. On the other hand, the difference found between human IgA(1) and human IgA(2) interaction with jacalin, eight times higher for IgA(1), was attributed to the presence of O-linked glycans in the IgA(1) hinge region, which is absent in IgA(2). Specific interaction of jacalin with O-glycans, proved to be present in the human IgA(1) and hypothetically present in bovine IgG(1) structures, is discussed as responsible for the obtained affinity values.

Xylanases (EC 3.2.1.8 endo-1,4-glycosyl hydrolase) catalyze the hydrolysis of xylan, an abundant hemicellulose of plant cell walls. Access to the catalytic site of GH11 xylanases is regulated by movement of a short beta-hairpin, the so-called thumb region, which can adopt open or closed conformations. A crystallographic study has shown that the D11F/R122D mutant of the GH11 xylanase A from Bacillus subtilis (BsXA) displays a stable "open" conformation, and here we report a molecular dynamics simulation study comparing this mutant with the native enzyme over a range of temperatures. The mutant open conformation was stable at 300 and 328 K, however it showed a transition to the closed state at 338 K. Analysis of dihedral angles identified thumb region residues Y113 and T123 as key hinge points which determine the open-closed transition at 338 K. Although the D11F/R122D mutations result in a reduction in local inter-intramolecular hydrogen bonding, the global energies of the open and closed conformations in the native enzyme are equivalent, suggesting that the two conformations are equally accessible. These results indicate that the thumb region shows a broader degree of energetically permissible conformations which regulate the access to the active site region. The R122D mutation contributes to the stability of the open conformation, but is not essential for thumb dynamics, i.e., the wild type enzyme can also adapt to the open conformation.

Habitat structure is known to influence the abundance of fishes on temperate reefs. Biotic interactions play a major role in determining the distribution and abundance of species. The significance of these forces in affecting the abundance of fishes may hinge on the presence of organisms that either create or alter habitat. On temperate reefs, for example, macroalgae are considered autogenic ecosystem engineers because they control resource availability to other species through their physical structure and provide much of the structure used by fish. On both coral and temperate reefs, small cryptic reef fishes may comprise up to half of the fish numbers and constitute a diverse community containing many specialized species. Small cryptic fishes (<100 mm total length) may be responsible for the passage of 57% of the energy flow and constitute ca. 35% of the overall reef fish biomass on coral reefs. These benthic fish exploit restricted habitats where food and shelter are obtained in, or in relation to, conditions of substrate complexity and/or restricted living space. A range of mechanisms has been proposed to account for the diversity and the abundance of small fishes: (1) lifehistory strategies that promote short generation times, (2) habitat associations and behaviour that reduce predation and (3) resource partitioning that allows small species to coexist with larger competitors. Despite their abundance and potential importance within reef systems, little is known of the community ecology of cryptic fishes. Specifically on habitat associations many theories suggested a not clear direction on this subject. My research contributes to the development of marine fish ecology by addressing the effects of habitat characteristics upon distribution of cryptobenthic fish assemblages. My focus was on the important shallow, coastal ecosystems that often serve as nursery habitat for many fish and where different type of habitat is likely to both play important roles in organism distribution and survival. My research included three related studies: (1) identification of structuring forces on cryptic fish assemblages, such as physical and biological forcing; (2) macroalgae as potential tools for cryptic fish and identification of different habitat feature that could explain cryptic fish assemblages distribution; (3) canopy formers loss: consequences on cryptic fish and relationship with benthos modifications. I found that: (1) cryptic fish assemblages differ between landward and seaward sides of coastal breakwaters in Adriatic Sea. These differences are explained by 50% of the habitat characteristics on two sides, mainly due to presence of the Codium fragile, sand and oyster assemblages. Microhabitat structure influence cryptic fish assemblages. (2) Different habitat support different cryptic fish assemblages. High heterogeneity on benthic assemblages reflect different fish assemblages. Biogenic components that explain different and diverse cryptic fish assemblages are: anemonia bed, mussel bed, macroalgal stands and Cystoseira barbata, as canopy formers. (3) Canopy forming loss is not relevant in structuring directly cryptic fish assemblages. A removal of canopy forming algae did not affect the structure of cryptic fish assemblages. Canopy formers algae on Conero cliff, does not seem to act as structuring force, probably due to its regressive status. In conclusion, cryptic fish have been shown to have species-specific associations with habitat features relating to the biological and non biological components afforded by fish. Canopy formers algae do not explain cryptic fish assemblages distribution and the results of this study and information from the literature (both from the Mediterranean Sea and elsewhere) show that there are no univocal responses of fish assemblages. Further exanimations on an non regressive status of Cystoseira canopy habitat are needed to define and evaluate the relationship between canopy formers and fish on Mediterranean sea.

Reinforced concrete columns might fail because of buckling of the longitudinal reinforcing bar when exposed to earthquake motions. Depending on the hoop stiffness and the length-over-diameter ratio, the instability can be local (in between two subsequent hoops) or global (the buckling length comprises several hoop spacings). To get insight into the topic, an extensive literary research of 19 existing models has been carried out including different approaches and assumptions which yield different results. Finite element fiberanalysis was carried out to study the local buckling behavior with varying length-over-diameter and initial imperfection-over-diameter ratios. The comparison of the analytical results with some experimental results shows good agreement before the post buckling behavior undergoes large deformation. Furthermore, different global buckling analysis cases were run considering the influence of different parameters; for certain hoop stiffnesses and length-over-diameter ratios local buckling was encountered. A parametric study yields an adimensional critical stress in function of a stiffness ratio characterized by the reinforcement configuration. Colonne in cemento armato possono collassare per via dell’instabilità dell’armatura longitudinale se sottoposte all’azione di un sisma. In funzione della rigidezza dei ferri trasversali e del rapporto lunghezza d’inflessione-diametro, l’instabilità può essere locale (fra due staffe adiacenti) o globale (la lunghezza d’instabilità comprende alcune staffe). Per introdurre alla materia, è proposta un’esauriente ricerca bibliografica di 19 modelli esistenti che include approcci e ipotesi differenti che portano a risultati distinti. Tramite un’analisi a fibre e elementi finiti si è studiata l’instabilità locale con vari rapporti lunghezza d’inflessione-diametro e imperfezione iniziale-diametro. Il confronto dei risultati analitici con quelli sperimentali mostra una buona coincidenza fino al raggiungimento di grandi spostamenti. Inoltre, il caso d’instabilità globale è stato simulato valutando l’influenza di vari parametri; per certe configurazioni di rigidezza delle staffe e lunghezza d’inflessione-diametro si hanno ottenuto casi di instabilità locale. Uno studio parametrico ha permesso di ottenere un carico critico adimensionale in funzione del rapporto di rigidezza dato dalle caratteristiche dell’armatura.

Mögliche Verformungsmechanismen, die zu den verschiedenen Glimmer- und Mineralfischen führen, sind: intrakristalline Verformung, Kristallrotation, Biegung und Faltung, Drucklösung in Kombination mit Ausfällung und dynamische Rekristallisation oder Mechanismen, die ein großes Mineral in mehrere kleine, fischförmige Kristalle aufspalten.Experimente mit ein neues Verformungsgerät und Objekten in zwei verschiedenen Matrixmaterialien werden beschrieben. Das eine ist PDMS, (Newtonianisch viskoses Polymer), und das andere Tapioca Perlen (Mohr-Couloumb Verhalten). Die Rotation von fischförmigen Objekten in PDMS stimmt mit der theoretischen Rotationsrate für ellipsenförmige Objekte in einem Newtonianischen Material überein. In einer Matrix von Tapioca Perlen nehmen die Objekte eine stabile Lage ein. Diese Orientierung ist vergleichbar mit der von Glimmerfischen. Die Verformung in der Matrix von Tapioca Perlen ist konzentriert auf dünne Scherzonen. Diese Ergebnisse implizieren, daß die Verformung in natürlichen Gesteinen auch in dünnen Scherzonen konzentriert ist.Computersimulationen werden beschrieben, mit denen der Einfluß der Eigenschaften einer Matrix auf die Rotation von Objekten und Verteilung von Deformation untersucht wird.Mit diesen Experimenten wird gezeigt, daß die Orientierung von Glimmerfischen nicht mit Verformung in einem nicht-linearen viskosen Material erklärt werden kann. Eine solche nicht-lineare Rheologie wird im Allgemeinen für die Erdkurste angenommen. Die stabile Orientierung eines Objektes kann mit weicheren Lagen in der Matrix erklärt werden.

E’ mostrata l’analisi e la modellazione di dati termocronologici di bassa temperatura da due regioni Alpine: il Sempione ed il Brennero. Le faglie distensive presenti bordano settori crostali profondi appartenenti al dominio penninico: il duomo metamorfico Lepontino al Sempione e la finestra dei Tauri al Brennero. I dati utilizzati sono FT e (U-Th)/He su apatite. Per il Sempione i dati provengono dalla bibliografia; per il Brennero si è provveduto ad un nuovo campionamento, sia in superficie che in sotterraneo. Gli attuali lavori per la galleria di base del Brennero (BBT), hanno consentito, per la prima volta, di raccogliere dati di FT e (U-Th)/He in apatite in sottosuolo per la finestra dei Tauri occidentale. Le analisi sono state effettuate tramite un codice a elementi finiti, Pecube, risolvente l’equazione di diffusione del calore per una topografia evolvente nel tempo. Il codice è stato modificato per tener conto dei dati sotterranei. L’inversione dei dati è stata effettuata usando il Neighbourhood Algorithm (NA), per ottenere il più plausibile scenario di evoluzione morfotettonico. I risultati ottenuti per il Sempione mostrano: ipotetica evoluzione dello stile tettonico della faglia del Sempione da rolling hinge a low angle detachment a 6.5 Ma e la cessazione dell’attività a 3 Ma; costruzione del rilievo fino a 5.5 Ma, smantellamento da 5.5 Ma ad oggi, in coincidenza dei cambiamenti climatici Messiniani e relativi all’inizio delle maggiori glaciazioni; incremento dell’esumazione da 0–0.6 mm/anno a 0.6–1.2 mm/anno a 2.4 Ma nell’emisfero settentrionale. I risultati al Brennero mostrano: maggiore attività tettonica della faglia del Brennero (1.3 mm/anno), maggiore attività esumativa (1–2 mm/anno) prima dei 10 Ma; crollo dell’attività della faglia del Brennero fra 10 Ma e oggi (0.1 mm/anno) e dell’attività esumativa nello stesso periodo (0.1–0.3 mm/anno); nessun aumento del tasso esumativo o variazioni topografiche negli ultimi 5 Ma.

This research has focused on the study of the behavior and of the collapse of masonry arch bridges. The latest decades have seen an increasing interest in this structural type, that is still present and in use, despite the passage of time and the variation of the transport means. Several strategies have been developed during the time to simulate the response of this type of structures, although even today there is no generally accepted standard one for assessment of masonry arch bridges. The aim of this thesis is to compare the principal analytical and numerical methods existing in literature on case studies, trying to highlight values and weaknesses. The methods taken in exam are mainly three: i) the Thrust Line Analysis Method; ii) the Mechanism Method; iii) the Finite Element Methods. The Thrust Line Analysis Method and the Mechanism Method are analytical methods and derived from two of the fundamental theorems of the Plastic Analysis, while the Finite Element Method is a numerical method, that uses different strategies of discretization to analyze the structure. Every method is applied to the case study through computer-based representations, that allow a friendly-use application of the principles explained. A particular closed-form approach based on an elasto-plastic material model and developed by some Belgian researchers is also studied. To compare the three methods, two different case study have been analyzed: i) a generic masonry arch bridge with a single span; ii) a real masonry arch bridge, the Clemente Bridge, built on Savio River in Cesena. In the analyses performed, all the models are two-dimensional in order to have results comparable between the different methods taken in exam. The different methods have been compared with each other in terms of collapse load and of hinge positions.

The aim of this study was to investigate the influence of the diaphragm flexibility on the behavior of out-of-plane walls in masonry buildings. Simplified models have been developed to perform kinematic and dynamic analyses in order to compare the response of walls with different restraint conditions. Kinematic non linear analyses of assemblages of rigid blocks have been performed to obtain the acceleration-displacement curves for walls with different restraint conditions at the top. A simplified 2DOF model has been developed to analyse the dynamic response of the wall with an elastic spring at the top, following the Housner rigid behaviour hypothesis. The dissipation of energy is concentrated at every impact at the base of the wall and is modelled through the introduction of the coefficient of restitution. The sets of equations of the possible configurations of the wall, depending on the different positions of the centre of rotation at the base and at the intermediate hinge have been obtained. An algorithm for the numerical integration of the sets of the equations of motion in the time domain has been developed. Dynamic analyses of a set of walls with Gaussian impulses and recorded accelerograms inputs have been performed in order to compare the response of the simply supported wall with the one of the wall with elastic spring at the top. The influence of diaphragm stiffness Kd has been investigated determining the variation of maximum displacement demand with the value of Kd. A more regular trend has been obtained for the Gaussian input than for the recorded accelerograms.

The aim of the present thesis was to investigate the influence of lower-limb joint models on musculoskeletal model predictions during gait. We started our analysis by using a baseline model, i.e., the state-of-the-art lower-limb model (spherical joint at the hip and hinge joints at the knee and ankle) created from MRI of a healthy subject in the Medical Technology Laboratory of the Rizzoli Orthopaedic Institute. We varied the models of knee and ankle joints, including: knee- and ankle joints with mean instantaneous axis of rotation, universal joint at the ankle, scaled-generic-derived planar knee, subject-specific planar knee model, subject-specific planar ankle model, spherical knee, spherical ankle. The joint model combinations corresponding to 10 musculoskeletal models were implemented into a typical inverse dynamics problem, including inverse kinematics, inverse dynamics, static optimization and joint reaction analysis algorithms solved using the OpenSim software to calculate joint angles, joint moments, muscle forces and activations, joint reaction forces during 5 walking trials. The predicted muscle activations were qualitatively compared to experimental EMG, to evaluate the accuracy of model predictions. Planar joint at the knee, universal joint at the ankle and spherical joints at the knee and at the ankle produced appreciable variations in model predictions during gait trials. The planar knee joint model reduced the discrepancy between the predicted activation of the Rectus Femoris and the EMG (with respect to the baseline model), and the reduced peak knee reaction force was considered more accurate. The use of the universal joint, with the introduction of the subtalar joint, worsened the muscle activation agreement with the EMG, and increased ankle and knee reaction forces were predicted. The spherical joints, in particular at the knee, worsened the muscle activation agreement with the EMG. A substantial increase of joint reaction forces at all joints was predicted despite of the good agreement in joint kinematics with those of the baseline model. The introduction of the universal joint had a negative effect on the model predictions. The cause of this discrepancy is likely to be found in the definition of the subtalar joint and thus, in the particular subject’s anthropometry, used to create the model and define the joint pose. We concluded that the implementation of complex joint models do not have marked effects on the joint reaction forces during gait. Computed results were similar in magnitude and in pattern to those reported in literature. Nonetheless, the introduction of planar joint model at the knee had positive effect upon the predictions, while the use of spherical joint at the knee and/or at the ankle is absolutely unadvisable, because it predicted unrealistic joint reaction forces.

In dieser Arbeit werden Synthesen und Eigenschaften von Verbindungen mit einer oder mehreren Ferrocen- bzw. Biferroceneinheiten beschrieben, die über Amid-, Anhydrid- oder Harnstoff-Funktionen verknüpft oder mittels Amidfunktion an α-Aminosäurederivate gebunden sind. Als Zentralbausteine dienen die künstlichen Aminosäuren 1’-Aminoferrocen-1-carbonsäure (Fca) bzw. 1’-Aminobiferrocen-1-carbonsäure (Bfca). Die Ferroceneinheit agiert als redoxschaltbares Gelenk, die Amidfunktion ermöglicht die Ausbildung von Sekundärstrukturen und die Bindung von Anionen. Das redoxschaltbare „Multiwellenlängen“-Sensorpaar [Dansyl-Ala-Fca-Ala-CH2-Naphthyl]0/+ ist in der Lage, insgesamt sieben Anionen aufgrund von sechs einfach zu erhaltenden optischen Messwerten eindeutig zu diskriminieren. Die Vorzugskonformation des neutralen Rezeptors mit intramolekularen Wasserstoffbrücken wird mittels X-Ray, NMR- und DFT-Methoden im Festkörper, in Lösung und in der Gasphase bestimmt. Die oligomeren Fca-Verbindungen SG-Fcan-HN-Fc (SG = Boc, Fmoc; n = 1, 2) und SG-Fca2-OMe (SG = Boc, Fmoc) werden mittels Peptidkupplung in Lösung hergestellt, Fmoc-Fca3-Gly-OMe, Fmoc-Fcan-OMe (n = 3-5) und Fmoc-Fca4-NH2 dagegen durch ein neu entwickeltes Festphasensynthese-Protokoll. Die amidverknüpften Verbindungen bilden eine „Zick-Zack“-Struktur mit 1,2’-Konformation der Fca-Einheiten und achtgliedrigen intramolekularen Wasserstoffbrücken-Ringen, wie durch X-Ray, 2D-NMR-, DFT-Methoden und Dipolmoment-Bestimmung gezeigt wird. Elektrochemische Experimente belegen eine elektronische Wechselwirkung der Eisenzentren. Die gemischt-valenten Verbindungen zeichnen sich durch IVCT-Banden im nahen Infrarot aus. Die elektronische Kopplungskonstante beträgt Hab ≈ 145-215 cm–1 für einen einzelnen FeII/FeIII-Übergang und belegt die Zugehörigkeit der Verbindungen zur Robin-Day-Klasse II. Im Festkörper sind die Valenzen gemäß Mößbauerspektren lokalisiert. Die vollständig oxidierten Verbindungen liegen nach DFT-Rechnungen nicht mehr in einer „Zick-Zack“-Struktur, sondern in einer gestreckten Konformation vor. Als Nebenprodukte bei der Amidkupplung werden die Anhydride SG-(Fca)2O (SG = Ac, Boc, Fmoc) isoliert. Diese zählen aufgrund des Fehlens einer IVCT-Bande zur Klasse I-II. Die ferrocenyloge Bfca wird in Form der N- und C-geschützten Bfca auf zwei Wegen synthetisiert. Schlüsselschritte stellen die Cu(II)-vermittelte Homokupplung bzw. die Pd-katalysierte Stille-Kupplung dar. Bfca und die amid- und harnstoffverknüpften Bis-Bfca-Verbindungen besitzen keine nachweisbare Vorzugskonformation in Lösung. Die gemischt-valenten Bfca-Kationen zeigen eine IVCT-Bande (Hab ≈ 300-600 cm–1) und gehören eher zur Klasse II-III. Die gemischt-valenten Verbindungen des als Nebenprodukt isolierten Tetraferrocenylstannans Sn[Fn(COOMe)4] (Fn = 1,1’-Ferrocenylen) mit einatomiger σ-Brücke zwischen den Ferroceneinheiten, zeigen IVCT-Banden im NIR-Spektrum und gehören somit zur Klasse II. Die elektronischen Kopplungen in Sn[Fn(COOMe)4]+/2+ betragen Hab ≈ 145 und 220 cm–1.

Oggetto di questa tesi è l’analisi delle modalità di rappresentazione del trauma nel romanzo del Novecento e, in particolare, nelle opere di Samuel Beckett, Georges Perec e Agota Kristof. Fondamento dello studio sarà una disamina dei procedimenti linguistici e narrativi di rappresentazione del trauma nelle prose degli autori citati, al fine tracciare le linee di un’estetica in grado di descrivere le caratteristiche peculiari delle narrazioni in cui la dimensione antinarrativa della memoria traumatica assume il ruolo di principio estetico guida. L’analisi si soffermerà sulla cruciale relazione esistente, in tutti e tre gli autori, tra rappresentazione del trauma e sviluppo di strategie narrativi definibili come “denegative”. L’analisi dei testi letterari è condotta sulla base del corpus critico dei Trauma Studies, dell’ermeneutica della narrazione di stampo ricœuriano e della teoria del linguaggio psicoanalitica e affiancata, ove possibile, da uno studio filologico-genetico dei materiali d’autore. Alla luce di tali premesse, intendo rivalutare il carattere rappresentativo e testimoniale della letteratura del secolo scorso, in contrasto con la consuetudine a vedere nel romanzo novecentesco il trionfo dell’antimimesi e il declino del racconto. Dal momento che le narrazioni traumatiche si costruiscono intorno e attraverso i vuoti di linguaggio, la tesi è che siano proprio questi vuoti linguistici e narrativi (amnesie, acronie, afasie, lapsus, omissioni e mancanze ancora più sofisticate come nel caso di Perec) a rappresentare, in modo mimetico, la realtà apparentemente inaccessibile del trauma. Si tenterà di dimostrare come questi nuovi canoni di rappresentazione non denuncino l’impossibilità del racconto, bensì una sfida al silenzio, celata in più sottili e complesse convenzioni narrative, le quali mantengono un rapporto di filiazione indiretto − per una via che potremmo definire denegativa − con quelle del romanzo tradizionale.

Escherichia coli kann unter aeroben und anaeroben Bedingungen mit C4-Dicarboxylaten wachsen, die Regulation des Stoffwechsels erfolgt durch das Zwei-Komponenten-System DcuSR. Die C4-Dicarboxylattransporter DctA (aerob) bzw. DcuB (anaerob) agieren als Co-Regulatoren und bilden gemeinsam mit der Sensor-Histidinkinase DcuS einen Sensorkomplex, in dem DcuS den Sensor darstellt und DctA bzw. DcuB diesen in seine rezeptive Form überführen. DcuS ist membranständig und verknüpft die Bindung von C4-Dicarboxylaten im Periplasma mit der Autophosphorylierung seiner Kinasedomäne im Cytoplasma. Dies stellt den Beginn einer Signalkaskade vom extrazellulären Reiz zum cytoplasmatischen Responseregulator DcuR dar.rnIn dieser Arbeit wurde die intramolekulare Signaltransduktion in DcuS und über die Membran untersucht. Der Fokus lag auf der Funktion der beiden Transmembranhelices TM1 und TM2 und der cytoplasmatischen PAS-Domäne, die die sensorische PASp- mit der effektorischen Kinasedomäne verbinden. Konformationsänderungen dieser Signalweiterleitung wurden durch Cysteinzugänglichkeitsstudien, oxidatives Cystein-Crosslinking und Mutageneseexperimente analysiert. rnTM2 wurde als der Überträger eines transmembranen Signals identifiziert, während TM1 als Membrananker fungiert. Der aktive Signalzustand von TM2 wird unabhängig von der Art der DcuS-Aktivierung (Effektorbindung, Deletion des Co-Regulators DctA oder PASc-ON-Mutationen) eingenommen. Der Signaltransduktion liegt eine Verschiebung von TM2 entlang ihrer Längsachse (Kolbenhub) in Richtung Periplasma zu Grunde. Cystein-Crosslinking offenbarte eine durchgehende Helix aus PASp-α6 und TM2, die im Dimer parallel mit ihrem Pendant verschoben wird. Die Amplitude des Kolbenhubs wurde anhand von Zugänglichkeitsveränderungen, der Lage verankernder Tryptophanreste, Strukturvergleichen und energetischen Berechnungen auf max. 4 - 6 Å festgelegt. Sie ist von der Effektorstärke abhängig und koppelt so die metabolische Bevorzugung einzelner Substrate an das Ausmaß des Kolbenhubs und der Genexpression. Für die cytoplasmatische PAS-Domäne wurde ein Zusammenhang zwischen lokaler Dimerisierung und Kontrolle der Sensorfunktion nachgewiesen. Schwächung der Dimerisierung führt zu einer Aktivierung der Sensorkinase. Es wurde eine hydrophobe Region identifiziert, deren strukturelle Integrität für diese Dimerisierung essentiell ist. Mit N248 wurde ein funktionell bedeutender Rest beschrieben, der auf Grund seiner Lage und seiner Eigenschaft mehrere Sekundärstrukturelemente zu verknüpfen, als Scharnier innerhalb der Domäne an der Umsetzung des Kolbenhubs in eine veränderte Quartärstruktur von PASc beteiligt sein könnte.

Analysis of the collapse of a precast r.c. industrial building during the 2012 Emilia earthquake, focus on the failure mechanisms in particular on the flexure-shear interactions. Analysis performed by a time history analysis using a FEM model with the software SAP2000. Finally a reconstruction of the collapse on the basis of the numerical data coming from the strength capacity of the elements failed, using formulation for lightly reinforced columns with high shear and bending moment.

Erneute Untersuchungen der mesozoischen Faltenstruktur des Otago Schiefergürtels, Südinsel, Neuseeland, zeigen, dass diese aus zwei aufeinander folgenden, ähnlichen, asymmetrischen, offenen bis mäßig engen Großfaltengenerationen im km- Größenbereich besteht anstatt aus den vorher angenommenen Decken- oder Halbfalten. Hauptproblem der Großfaltenstruktur sind Zonen von durchgreifender Boudinage, die in der Nähe der Großfaltenscharniere entstanden sind. Vorherige Bearbeiter deuteten diese Zonen als 'starke Verformungszonen' oder Überschiebungszonen. Diese Arbeit zeigt, dass in diesen Zonen nur durch die asymmetrische Faltung die unteren liegenden Schenkel der Großfalten boudiniert und somit häufig die ansonsten typischen Faltenstrukturen des liegenden Schenkels einer symmetrischen Faltung überprägt wurden. Ein weiteres Problem dieser mesozoischen Großfaltenstruktur ist die Überprägung einer Faltengeneration auf eine frühere. Weil die Verkürzungsrichtung der überprägenden Faltengeneration nicht subparallel zur älteren Faltenachse ist, sondern einen Winkel von rund 30 Grad einschließt, ist ein Wechsel von orthogonalen zu koaxialen Interferenzmustern der Kleinfalten beobachtbar. Folglich ist die Orientierung der Scheitellinie einer überprägenden und überprägten Kleinfalte nicht unbedingt subparallel zur Orientierung der Faltenachse der Großfalte trotz zylindrischer Faltung. Im letzten Teil dieser Arbeit wird die Überprägung der mesozoischen Großfaltenstruktur durch das känozoisch entstandene, transpressionale Alpine Störungssystem, das einen zweiseitigen Falten- und Überschiebungsgürtel im Otago und im Nordwesten anschließenden Alpinen Schiefergürtel bildet, beschrieben.
